Abstract Background Thermostability is a fundamental property of proteins to maintain their biological functions. Predicting protein stability changes upon mutation is important for our understanding protein structure–function relationship, and is also of great interest in protein engineering and pharmaceutical design. Results Here we present mutDDG-SSM, a deep learning-based framework that uses the geometric representations encoded in protein structure to predict the mutation-induced protein stability changes. mutDDG-SSM consists of two parts: a graph attention network-based protein structural feature extractor that is trained with a self-supervised learning scheme using large-scale high-resolution protein structures, and an eXtreme Gradient Boosting model-based stability change predictor with an advantage of alleviating overfitting problem. The performance of mutDDG-SSM was tested on several widely-used independent datasets. Then, myoglobin and p53 were used as case studies to illustrate the effectiveness of the model in predicting protein stability changes upon mutations. Our results show that mutDDG-SSM achieved high performance in estimating the effects of mutations on protein stability. In addition, mutDDG-SSM exhibited good unbiasedness, where the prediction accuracy on the inverse mutations is as well as that on the direct mutations. Conclusion Meaningful features can be extracted from our pre-trained model to build downstream tasks and our model may serve as a valuable tool for protein engineering and drug design.

Abstract Pressing environmental challenges require focused research on sustainable solutions in the domains of energy, water, food, land, and climate. The pigment eumelanin has recently been positioned as a promising candidate for solving issues in health, sensors, and energy storage. However, the low solubility of eumelanin in aqueous solvents, difficult film processibility, and high cost have hindered the material from wide deployment. Here, we propose melanin extracted from the black soldier fly, Hermetia illucens (Mel-BSF), as a sustainable alternative for the preparation of organic electrodes in energy storage applications. Mel-BSF displays pseudocapacitive behaviour with a high potential window, good electrochemical stability, and higher maximum capacity (91.8 mAh g−1) compared to synthetic eumelanin (17.3 mAh g−1) as the working electrode material in zinc-ion hybrid capacitors using an ionic liquid electrolyte. Structural and surface investigations reveal that additional aliphatic compounds, potentially lipids present after Mel-BSF refinement, significantly increase the film stability and redox centre availability.

Abstract Background Graduate students exhibit vulnerability to problematic Internet use, which can result in adverse physical, psychological, and social consequences. However, limited studies have addressed this issue among graduate students, and even fewer have explored the unique factors contributing to their problematic Internet use. Therefore, to address this gap, the current study aims to probe the relationship between supervisor’s neuroticism and problematic Internet use among graduate students, the mediating effect of the supervisor-student relationship quality, as well as the moderating effect of fear of the supervisor’s negative evaluation. Methods A cross-sectional study was conducted in 2018 at three universities in Beijing, China. Anonymous data from 448 graduate students were collected regarding problematic Internet use, supervisor’s neuroticism, supervisor-student relationship quality, and the fear of the supervisor’s negative evaluation. A moderated mediation analysis was performed using Hayes’ PROCESS macro (Model 14). Results Supervisor’s neuroticism was positively linked to graduate students’ problematic Internet use, supervisor-student relationship quality mediated the linkage, and fear of the supervisor’s negative evaluation played a moderating role in the second stage. Specifically, for students lower in fear of the supervisor’s negative evaluation, supervisor-student relationship quality negatively predicted students’ problematic Internet use. While for the graduate students higher in fear of the supervisor’s negative evaluation, supervisor-student relationship quality could not significantly predict students’ problematic Internet use. The mediating effect was only significant for graduate students lower in fear of the supervisor’s negative evaluation. Conclusions This study established a theoretical model linking supervisor’s neuroticism to graduate students’ problematic Internet use, highlighting the potential roles of supervisor-student relationship quality and fear of the supervisor’s negative evaluation. Reducing the neuroticism level of the supervisor, enhancing the quality of the supervisor-student relationship, and mitigating students’ fear of the supervisor’s negative evaluation will contribute to the reduction of problematic Internet use among graduate students.

Abstract Background The experiences and challenges associated with breastfeeding multiple births can be considerably more complex than those of singletons. Multiple births refer to the delivery of more than one offspring in a single birth event. Emphasizing the needs and experiences of mothers with multiple births during breastfeeding can enable healthcare providers to design targeted interventions that enhance breastfeeding rates. However, existing breastfeeding and health education resources and practices do not fully meet the needs of women who breastfeed multiples. This review aimed to review and synthesize qualitative studies on the breastfeeding experiences of women with multiple births. Methods A systematic search was conducted in 10 electronic databases for papers published from the inception of the database to March 2024. The Joanna Briggs Institute Critical Appraisal Checklist for Qualitative Research was utilized to evaluate the methodological quality of the studies included. The thematic synthesis method of Thomas and Harden was employed to integrate and analyze the included literature to derive new categories and conclusions. Findings Eight studies met the inclusion criteria and quality assessment criteria for this study. Through the integration of their results, four themes were identified: the choice and willingness to breastfeed multiple births; the challenges of breastfeeding multiple births; stage management and individualised adaptation of breastfeeding; and the experience of support. Conclusion Throughout the feeding process from pregnancy to the postpartum period, mothers with multiple births often have predominantly negative experiences with breastfeeding. Consequently, hospitals should create a multidisciplinary follow-up team comprising obstetrics, neonatology, psychology, and community services to offer specialized and personalized support to these women at various stages. Abstract Conventional wireless communication schemes indiscriminately transmit information into the whole space and pose inherent security risks. Recently, directional information modulation (DIM) has attracted enormous attention as a promising technology. DIM generates correct constellation symbols in the desired directions and distorts them in undesired directions, thus ensuring the security of the transmitted information. Although several DIM schemes have been reported, they suffer from defects of bulkiness, energy consumption, high cost, and inability to support two-dimensional (2D) and high-order modulations. Here, we propose a DIM scheme based on a 2-bit programmable metasurface (PM) that overcomes these defects. A fast and efficient discrete optimization algorithm is developed to optimize the digital coding sequences, and the correct constellation symbols can be generated and transmitted in multi-directional beams. As a proof-of-concept, three sets of constellation diagrams (8 phase shift keying (PSK), 16 quadrature amplitude modulation (QAM), and 64QAM) are realized in the multi-channel modes. This work provides an important route of employing DIM for ensuring physical-layer security and serves as a stepping stone toward endogenous secure communications.

Abstract Background Acute kidney injury (AKI) in patients with acute myocardial infarction (AMI) often indicates a poor prognosis. Objective This study aimed to investigate the association between the TyG index and the risk of AKI in patients with AMI. Methods Data were taken from the Medical Information Mart for Intensive Care (MIMIC) database. A 1:3 propensity score (PS) was set to match patients in the AKI and non-AKI groups. Multivariate logistic regression analysis, restricted cubic spline (RCS) regression and subgroup analysis were performed to assess the association between TyG index and AKI. Abstract Rheumatoid arthritis (RA) is a progressive autoimmune disease accompanied by joint swelling, cartilage erosion and bone damage. Drug therapy for RA has been restricted due to poor therapeutic effect, recurrence and adverse effects. Macrophages and synovial fibroblasts both play important roles in the pathology of RA. Macrophages secrete large amount of pro-inflammatory cytokines, while synovial fibroblasts are tightly correlated with hypoxia synovium microenvironment, cytokine release, recruitment of pro-inflammatory cells, bone and cartilage erosion. Therefore, in this timely research, an injectable and pH-sensitive peptide hydrogel loading methotrexate (MTX) and bismuthene nanosheet/polyethyleneimine (BiNS/PEI) has been developed to reduce the activity of macrophages and eliminate over-proliferated synovial fibroblasts simultaneously. MTX can reduce the cytokine secretion of macrophages/anti-apoptosis property of synovial fibroblasts and BiNS/PEI can eliminate synovial fibroblasts via photodynamic therapy (PDT) and photothermal therapy (PTT) routes. The hydrogel was injected into the acidic inflammatory synovium for precise targeting and served as a drug reservoir for pH responsive and sustained drug release, while improving the bioavailability and reducing the toxicity of MTX. Excellent therapeutic efficacy has been achieved in both in vivo and in vitro studies, and this unique drug delivery system provides a new and robust strategy to eliminate synovial fibroblasts and modulate immune system for RA treatment in clinical. Various water-saving irrigation (WSI) practices (e.g., dry cultivation, intermittent irrigation, controlled irrigation, shallow-wet irrigation, and rain-gathering irrigation) have been applied to rice cultivation mitigate water scarcity in China. However, in previous studies, these WSI practices have shown different water savings and yield increases, mainly due to different application conditions. A meta-analysis was applied to investigate the responses of the actual evapotranspiration (ETact), irrigation water (IW), rice yield (Y), and water productivity (WP) to WSI practices in different conditions, and 956 data sets were selected from 108 published papers. The results showed that, compared to traditional flood irrigation, rain-gathering irrigation decreased ETact and IW by 25.41 % and 55.7 % respectively, and increased WP greatly by 14.26 % while having a slight decrease in Y. Except for dry cultivation, all WSI practices increased WP by 4.72–14.26 % compared to traditional flood irrigation. The effects of different soil qualities on rice water consumption and production vary; medium soils with high organic content and a pH below 6.5 are better for rice growth. As for rice seasons, WSI practices had the least impact on ETact in middle rice, with an average reduction of 5.84 %, followed by early rice (–12.66 %) and late rice (–18.81 %). Higher mean annual temperature and more precipitation led to more Y under WSI practices. Differences in the effects of mean annual temperature and mean annual precipitation on WP were not significant. Our meta-analysis provides more insight into the effects of water-saving irrigation practices on rice water consumption, yield, and water productivity at various experimental sites. In general, there is considerable variation in the responses of Y and WP to different water-saving irrigation practices, and more evaluation of aspects such as rice seasons, soil properties, and meteorological conditions is needed for optimizing WSI in practice.

Abstract Dual‐polarization programmable metasurfaces can flexibly manipulate electromagnetic (EM) waves while providing approximately twice the information capacity. Therefore, they hold significant applications in next‐generation communication systems. However, there are three challenges associated with the existing dual‐polarization programmable metasurfaces. This article aims to propose a novel design to address them. First, the design overcomes the challenge of element‐ and polarization‐independent controls, enabling more powerful manipulations of EM waves. Second, by using more energy‐efficient tunable components and reducing their number, the design can be nearly passive (maximum power consumption of 27.7 mW), leading to a significant decrease in the cost and power consumption of the system (at least two orders of magnitude lower than the power consumption of conventional programmable metasurfaces). Third, the design can operate in a broad bandwidth, which is attractive for practical engineering applications. Both the element and array of the metasurface are meticulously designed, and their performance has been carefully studied. The experiments demonstrate that 2D wide‐angle beam scanning can be realized. Moreover, secure communication based on directional information modulation can be implemented by exploiting the metasurface and an efficient discrete optimization algorithm, showing its programmable, multiplexing, broadband, green, and secure features.

Objective: This study aimed to develop and validate several artificial intelligence (AI) models to identify acute myocardial infarction (AMI) patients at an increased risk of acute kidney injury (AKI) during hospitalization. Methods: Included were patients diagnosed with AMI from the Medical Information Mart for Intensive Care (MIMIC) III and IV databases. Two cohorts of AMI patients from Changzhou Second People's Hospital and Xuzhou Center Hospital were used for external validation of the models. Patients' demographics, vital signs, clinical characteristics, laboratory results, and therapeutic measures were extracted. Totally, 12 AI models were developed. The area under the receiver operating characteristic curve (AUC) were calculated and compared. Results: AKI occurred during hospitalization in 1098 (28.3 %) of the 3882 final enrolled patients, split into training (3105) and test (777) sets randomly. Among them, Random Forest (RF), C5.0 and Bagged CART models outperformed the other models in both the training and test sets. The AUCs for the test set were 0.754, 0.734 and 0.730, respectively. The incidence of AKI was 9.8 % and 9.5 % in 2202 patients in the Changzhou cohort and 807 patients in the Xuzhou cohort with AMI, respectively. The AUCs for patients in the Changzhou cohort were RF, 0.761; C5.0, 0.733; and bagged CART, 0.725, respectively, and Xuzhou cohort were RF, 0.799; C5.0, 0.808; and bagged CART, 0.784, respectively. Conclusion: Several machines learning-based prediction models for AKI after AMI were developed and validated. The RF, C5.0 and Bagged CART model performed robustly in identifying high-risk patients earlier. Multi-functional triply periodic minimal surface (TPMS) structures have been widely studied and proven to have significant importance. Most studies are based on several typical TPMSs. However, some research suggests that the hybrid forms of the TPMSs can exhibit superior mechanical properties. The challenge lies in the unpredictability of the hybrid TPMS property. In this study, an approach is proposed for precisely identify the hybrid TPMS using fully connected neural network (FCNN) and particle swarm optimization (PSO) algorithm. A design space of four TPMSs hybridization is introduced and used to generate over 8000 samples. Stiffness constants obtained using the homogenization method is used in elastic characterization. The hybrid TPMSs is proved to lack common cubic symmetry but possess wide rotational symmetry. Indirect stiffness constant-based way exhibits better predictions for optimal stiffness and elastic isotropy than direct property parameter-based one. The approach successfully identified useful structures: quasi-elastic-isotropic ones and a ultra-stiff one that is ∼ 9 % stronger than the currently strongest TPMS. The method offers comprehensive guidance for stiffness constant-based TPMS design using DL technique.

Abstract Lithium metal batteries with inorganic solid‐state electrolytes have emerged as strong and attractive candidates for electrochemical energy storage devices because of their high‐energy content and safety. Nonetheless, inherent challenges of deleterious lithium dendrite growth and poor interfacial stability hinder their commercial application. Herein, we report a liquid metal‐coated lithium metal (LM@Li) anode strategy to improve the contact between lithium metal and a Li6PS5Cl inorganic electrolyte. The LM@Li symmetric cell shows over 1000 h of stable lithium plating/stripping cycles at 2 mA cm−2 and a significantly higher critical current density of 9.8 mA cm−2 at 25°C. In addition, a full battery assembled with a high‐capacity composite LiNbO3@LiNi0.7Co0.2Mn0.1O2 (LNO@NCM721) cathode shows stable cycling performance. Experimental and computational results have demonstrated that dendrite growth tolerance and physical contact in solid‐state batteries can be reinforced by using LM interlayers for interfacial modification.

Fruit and vegetable wastes are linked to the depletion of natural resources and can pose serious health and environmental risks (e.g. eutrophication, water and soil pollution, and GHG emissions) if improperly managed. Current waste management practices often fail to recover high-value compounds from fruit wastes. Among emerging valorization methods, the utilization of fruit wastes as a feedstock for microalgal biorefineries is a promising approach for achieving net zero waste and sustainable development goals. This is due to the ability of microalgae to efficiently sequester carbon dioxide through photosynthesis, utilize nutrients in wastewater, grow in facilities located on non-arable land, and produce several commercially valuable compounds with applications in food, biofuels, bioplastics, cosmetics, nutraceuticals, pharmaceutics, and various other industries. However, the application of microalgal biotechnology towards upcycling fruit wastes has yet to be implemented on the industrial scale due to several economic, technical, operational, and regulatory challenges. Here, we identify sources of fruit waste along the food supply chain, evaluate current and emerging fruit waste management practices, describe value-added compounds in fruit wastes, and review current methods of microalgal cultivation using fruit wastes as a fermentation medium. We also propose some novel strategies for the practical implementation of industrial microalgal biorefineries for upcycling fruit waste in the future.

Abstract Respiratory syncytial virus (RSV) is one of the most important pathogens causing respiratory tract infection in humans, especially in infants and the elderly. The identification and structural resolution of the potent neutralizing epitopes on RSV fusion (F) protein enable an “epitope-focused” vaccine design. However, the display of RSV F epitope II on the surface of the widely-used human hepatitis B virus core antigen (HBcAg) has failed to induce neutralizing antibody response in mice. Here, we used the hepadnavirus core protein (HcAg) from different mammalian hosts as scaffolds to construct chimeric virus-like particles (VLPs) presenting the RSV F epitope II. Mouse immunization showed that different HcAg-based chimeric VLPs elicited significantly different neutralizing antibody responses, among which the HcAg derived from roundleaf bat (RBHcAg) is the most immunogenic. Furthermore, RBHcAg was used as the scaffold platform to present multiple RSV F epitopes, and the immunogenicity was further improved in comparison to that displaying a single epitope II. The designed RBHcAg-based multiple-epitope-presenting VLP formulated with MF59-like adjuvant elicited a potent and balanced Th1/Th2 immune response, and offered substantial protection in mice against the challenge of live RSV A2 virus. The designed chimeric VLPs may serve as the potential starting point for developing epitope-focused vaccines against RSV. Our study also demonstrated that RBHcAg is an effective VLP carrier for presenting foreign epitopes, providing a promising platform for epitope-focused vaccine design.

Abstract Mitochondria are inherited exclusively from the mothers and are required for the proper development of embryos. Hence, germline mitochondrial quality is highly regulated during oogenesis to ensure oocyte viability. How nutrient availability influences germline mitochondrial quality control is unclear. Here we find that fasting leads to the accumulation of mitochondrial clumps and oogenesis arrest in Drosophila. Fasting induces the downregulation of the DIP1-Clueless pathway, leading to an increase in the expression of a stable intronic sequence RNA called sisR-1. Mechanistically, sisR-1 localizes to the mitochondrial clumps to inhibit the poly-ubiquitination of the outer mitochondrial protein Porin/VDAC1, thereby suppressing p62-mediated mitophagy. Alleviation of the fasting-induced high sisR-1 levels by either sisR-1 RNAi or refeeding leads to mitophagy, the resumption of oogenesis and an improvement in oocyte quality. Thus, our study provides a possible mechanism by which fasting can improve oocyte quality by modulating the mitochondrial quality control pathway. Of note, we uncover that the sisR-1 response also regulates mitochondrial clumping and oogenesis during protein deprivation, heat shock and aging, suggesting a broader role for this mechanism in germline mitochondrial quality control.

Spontaneous combustion of coal seams poses significant threats to ecology and human health, making it a global concern. Precise location identification of coal fire burning is vital for disaster management. Traditional methods of identifying fire zones focus on specific indicators, such as temperature anomalies. However, solely relying on one indicator can easily lead to “heterogeneous homogeneity,” and using multiple indicators complicates threshold settings. To address this, this article presents an enhanced ensemble learning model named AdaBoost_RF_StBP for coal fire zone identification. By integrating multimodal remote sensing data, we extracted diverse abnormal features of fire zones. Using a standard deviation ellipse, we categorized feature samples into three fire zone types, which were then analyzed using the proposed model. The combined approach of base and metalearners was applied in coal fire zones located in Jiangjun Gobi, Jiangjun Temple, Sandaoba, and Beishan coal mines. Results show that the multimodal remote sensing data coupled by our model can effectively addresses the “heterogeneous homogeneity” issue. The accuracy of the model in terms of accuracy, precision, recall rate, F1_score, etc., has been improved from 0.56 to 0.87 of the initial base-learners to 0.8–0.96 of the metalearners. Additionally, the recognition areas of AdaBoost_RF_StBP in Jiangjun Gobi, Jiangjun Temple, and Sandaoba fire areas account for 122.7%, 109.8%, and 115.8% of the actual fire areas, respectively. This underscores the promising potential of the AdaBoost_RF_StBP model for fire zone identification. This model offers valuable insights and guidance for large-scale fire zone monitoring and identification, proving beneficial for precise and efficient fire control.

Abstract Background The purpose of this study was to develop a Nomogram model to identify the risk of all-cause mortality during hospitalization in patients with heart failure (HF). Methods HF patients who had been registered in the Medical Information Mart for Intensive Care (MIMIC) III and IV databases were included. The primary outcome was the occurrence of all-cause mortality during hospitalization. Two Logistic Regression models (LR1 and LR2) were developed to predict in-hospital death for HF patients from the MIMIC-IV database. The MIMIC-III database were used for model validation. The area under the receiver operating characteristic curve (AUC) was used to compare the discrimination of each model. Calibration curve was used to assess the fit of each developed models. Decision curve analysis (DCA) was used to estimate the net benefit of the predictive model. Results A total of 16,908 HF patients were finally enrolled through screening, of whom 2,283 (13.5%) presented with in-hospital death. Totally, 48 variables were included and analyzed in the univariate and multifactorial regression analysis. The AUCs for the LR1 and LR2 models in the test cohort were 0.751 (95% CI: 0.735∼0.767) and 0.766 (95% CI: 0.751–0.781), respectively. Both LR models performed well in the calibration curve and DCA process. Nomogram and online risk assessment system were used as visualization of predictive models. Conclusion A new risk prediction tool and an online risk assessment system were developed to predict mortality in HF patients, which performed well and might be used to guide clinical practice.

Coal fire is a geological disaster that causes resource waste and environmental pollution globally. Accurate identification of the spatial location of coal fires is critical for effective coal fire governance. However, existing methods for identifying coal fire zones have problems, such as a high omission and misclassification ratio and insufficient consideration of the temporal variation in temperature. Therefore, this article proposed a temporal temperature anomaly extraction algorithm based on adaptive windows (TTAE-AW) to extract temporal temperature anomaly information. Moreover, the spatial coverage of deformation monitoring points was improved using distributed scatterer interferometric synthetic aperture radar (DS-InSAR), and then a double-threshold two-stage filter method (DTTF) was proposed to accurately identify the spatial location of coal fire zones. The Rujigou mining area in Ningxia (China) was chosen as the region of study. Results showed that the temperature anomalies extracted using the TTAE-AW method are more concentrated in coal fire zones and that the amount in different seasons is more stable. The average accuracy and Kappa coefficient were improved by 15.5% and 0.345, respectively, over those of the conventional method. Compared with the small baselines subset InSAR approach, the DS-InSAR technique has 158% higher spatial coverage for monitoring coal fire zones. Compared with in situ observations of coal fire points, the accuracy and Kappa coefficient of the spatial location of fire zones obtained using the DTTF method were 91% and 0.77, respectively, demonstrating that the proposed method can provide reliable technical support for coal fire monitoring and management.

Porous materials with excellent load-bearing capacity are essential for aerospace, shipbuilding, and biomedical devices where lightweight structures are required. In this study, a design strategy of mechanical metamaterials with a shell-based structure is proposed. By re-filling curved surfaces in the tetrahedral wireframe, closed cavities inside the structure are eliminated and the manufacturability is improved. The modified tetrahedron is mirrored, cut with specific proportions (25%, 50%, and 75%), and arrayed to form different structures. The compression performance of the structure is investigated using stereolithographic specimens and numerical methods. Compared with the mechanical metamaterials based on triply periodic minimal surfaces (TPMS) and strut-based structures, the new shell-based structures exhibit excellent strength and stiffness. The compressive strength and Young’s modulus of the structure with 50% cutting proportion are increased by 65% and 80%, respectively, compared with the Gyroid structure. This study offers a new design strategy for mechanical metamaterials with excellent compression-resistant capability.

ABSTRACTThe increasingly distressing problem of noise pollution prompts the rapid research and development of lattice-based acoustic materials for practical applications. These lattices boast excellent mechanical properties and unlimited design freedom made possible using additive manufacturing technologies. Currently, there is a lack of analytical mathematical models relating the lattice geometry to the acoustic models and the absence of software dedicated to the acoustic modelling of strut lattice-based sound absorbers. This work presents LattSAC, an acoustics calculator that takes in all the geometrical parameters of the lattice structure and outputs directly the sound absorption coefficients (SAC) and their statistics. Hidden within the code is the novel Multi-layered Micropore-Cavity (MMC) model with semi-empirical mathematical models well-trained using experimental data to output the SACs with excellent accuracies. This software aims to accelerate the research and development of lattice-based acoustic absorbers for actual applications. It also functions as a valuable tool for users looking to design sound-absorbing materials using lattice structures. AbstractIn China, the concept of Museum Night Opening, which involves keeping museums open to the public beyond regular daytime hours, has gained popularity since its inception in 2019. However, its implementation has posed both social benefits and operational challenges. This study aims to develop management strategies for Museum Night Opening in China, using a regional perspective centered on Shanghai. A systematic review of Shanghai cases spanning from 2007 to August 2023 was conducted for the development status. A SWOT-TWOS analysis was further employed for the development environment and management strategies. Crabapple is a valuable tree species in gardens due to its captivating array of flower and leaf colors, rendering it a favored choice in landscaping. The economic and ornamental values of Malus crabapple are closely associated with the biosynthesis of anthocyanin, a pigment responsible for its vibrant hues. The intricate regulation of anthocyanin biosynthesis involves the concerted activity of various genes. However, the specific mechanism governing this process in crabapple warrants in-depth exploration. In this study, we explored the inhibitory role of MsMYB62-like in anthocyanin biosynthesis. We identified MsDFR and MsANS as two downstream target genes of MsMYB62-like. These genes encode enzymes integral to the anthocyanin biosynthetic pathway. The findings demonstrate that MsMYB62-like directly binds to the promoters of MsDFR and MsANS, resulting in the downregulation of their expression levels. Additionally, our observations indicate that the plant hormone cytokinins exert a suppressive effect on the expression levels of MsMYB62-like, while concurrently upregulating MsDFR and MsANS. This study reveals that the MsMYB62-like-MsDFR/MsANS module plays an important role in governing anthocyanin levels in Malus crabapple. Notably, the regulatory interplay is modulated by the plant hormone cytokinins.

Microalgae, Chlorella vulgaris exhibits substantial potential as a sustainable food ingredient, but its robust cell wall and limited protein solubility hinder industrial scale protein recovery. A comprehensive solution was devised, integrating dry bead milling and alkaline solubilization. In this method, cells were initially disrupted with bead milling followed by alkali (NaOH) treatment. Without bead milling, protein extraction yield was very low (5.0 % with water, 16.8 % with 0.1 M NaOH) at a biomass loading of 20 g/L. However, the integrated approach significantly improved these results, achieving a maximum protein extraction yield of about 47.3 % at a biomass loading of 100 g/L. The optimized conditions for both dry bead milling (frequency 26 Hz, duration 1.0 h) and alkaline solubilization (biomass weight to NaOH molar ratio 200–250 (g/M), 37 °C, mixing time 1.0 h) played a pivotal role in realizing these improved results. This integrated approach effectively addresses the challenges and holds industrial relevance, offering a more efficient way to extract microalgal protein.

Microalgal alternative proteins are increasingly recognized as a sustainable and promising source for future food production. Despite the widespread use of alkaline solubilization in protein extraction, its influence on the functional properties of microalgal protein extracts has received limited attention. This study marks the pioneering effort to assess the impact of alkaline (NaOH) solubilization on both the extraction process and the functional properties of Chlorella vulgaris protein extract (CVPE) obtained through isoelectric precipitation. With the gradual escalation of alkaline concentration from 0 to 0.7 M, there was a notable improvement in protein extraction efficiency, increasing significantly from 6.22 % to 56.23 %. However, elevated NaOH concentrations resulted in protein degradation, as evidenced in SDS-PAGE studies. Alkaline solubilization exhibited a positive impact on various functional properties of CVPE. It enhanced solubility, water absorption capacity, foaming capacity and stability, as well as emulsifying activity and stability. However, there was a slight decrease in oil absorption capacity with increasing NaOH concentration. Notably, the maximum solubility, oil adsorption capacity, foaming capacity, and emulsion activity of CVPE were 2.3-, 1.8-, 2-, and 1.3-fold higher, respectively, than those of commercial soy protein isolate (SPI). This highlights the effectiveness of alkaline solubilization in modifying C. vulgaris protein and emphasizes the high potential of CVPE as a functional food ingredient.

Summary: Inter-regulation between related genes, such as ribosomal protein (RP) paralogs, has been observed to be important for genetic compensation and paralog-specific functions. However, how paralogs communicate to modulate their expression levels is unknown. Here, we report a circular RNA involved in the inter-regulation between RP paralogs RpL22 and RpL22-like during Drosophila spermatogenesis. Both paralogs are mutually regulated by the circular stable intronic sequence RNA (sisRNA) circRpL22(NE,3S) produced from the RpL22 locus. RpL22 represses itself and RpL22-like. Interestingly, circRpL22 binds to RpL22 to repress RpL22-like, but not RpL22, suggesting that circRpL22 modulates RpL22’s function. circRpL22 is in turn controlled by RpL22-like, which regulates RpL22 binding to circRpL22 to indirectly modulate RpL22. This circRpL22-centric inter-regulatory circuit enables the loss of RpL22-like to be genetically compensated by RpL22 upregulation to ensure robust male germline development. Thus, our study identifies sisRNA as a possible mechanism of genetic crosstalk between paralogous genes.

Background Approximately two-thirds of patients with relapsed or refractory large B-cell lymphoma (R/R LBCL) do not respond to or relapse after anti-CD19 chimeric antigen receptor T (CAR T)-cell therapy, leading to poor outcomes. Previous studies have suggested that intensified lymphodepletion and hematological stem cell infusion can promote adoptively transferred T-cell expansion, enhancing antitumor effects. Therefore, we conducted a phase I/II clinical trial in which CNCT19 (an anti-CD19 CAR T-cell) was administered after myeloablative high-dose chemotherapy and autologous stem cell transplantation (HDT/ASCT) in patients with R/R LBCL.Methods Transplant-eligible patients with LBCL who were refractory to first-line immunochemotherapy or experiencing R/R status after salvage chemotherapy were enrolled. The study aimed to evaluate the safety and efficacy of this combinational therapy. Additionally, frozen peripheral blood mononuclear cell samples from this trial and CNCT19 monotherapy studies for R/R LBCL were used to evaluate the impact of the combination therapy on the in vivo behavior of CNCT19 cells.Results A total of 25 patients with R/R LBCL were enrolled in this study. The overall response and complete response rates were 92.0% and 72.0%, respectively. The 2-year progression-free survival rate was 62.3%, and the overall survival was 68.5% after a median follow-up of 27.0 months. No unexpected toxicities were observed. All cases of cytokine release syndrome were of low grade. Two cases (8%) experienced grade 3 or higher CAR T-cell-related encephalopathy syndrome. The comparison of CNCT19 in vivo behavior showed that patients in the combinational therapy group exhibited enhanced in vivo expansion of CNCT19 cells and reduced long-term exhaustion formation, as opposed to those receiving CNCT19 monotherapy.Conclusions The combinational therapy of HDT/ASCT and CNCT19 demonstrates impressive efficacy, improved CNCT19 behavior, and a favorable safety profile.Trial registration numbers ChiCTR1900025419 and NCT04690192.

Abstract Aiming at the problem that the existing ankle rehabilitation robot is difficult to fully fit the complex motion of human ankle joint and has poor human-machine motion compatibility, an equivalent series mechanism model that is highly matched with the actual bone structure of the human ankle joint is proposed and mapped into a parallel rehabilitation mechanism. The parallel rehabilitation mechanism has two virtual motion centers (VMCs), which can simulate the complex motion of the ankle joint, adapt to the individual differences of various patients, and can meet the rehabilitation needs of both left and right feet of patients. Firstly, based on the motion properties and physiological structure of the human ankle joint, the mapping relationship between the rehabilitation mechanism and ankle joint is determined, and the series equivalent model of the ankle joint is established. According to the kinematic and constraint properties of the ankle equivalent model, the configuration design of the parallel ankle rehabilitation robot is carried out. Secondly, according to the intersecting motion planes theory, the full-cycle mobility of the mechanism is proved, and the continuous axis of the mechanism is judged based on the constraint power and its derivative. Then, the kinematics of the parallel ankle rehabilitation robot is analyzed. Finally, based on the OpenSim biomechanical software, a human-machine coupling rehabilitation simulation model is established to evaluate the rehabilitation effect, which lays the foundation for the formulation of a rehabilitation strategy for the later prototype.

Abstract Amyotrophic lateral sclerosis (ALS) is a devastating fatal neurodegenerative disease with no cure. Receptor-interacting protein kinase 1 (RIPK1) has been proposed to mediate pathogenesis of ALS. Primidone has been identified as an old drug that can also inhibit RIPK1 kinase. We conducted a drug-repurposing biomarker study of primidone as a RIPK1 inhibitor using SOD1G93A mice and ALS patients. SOD1G93A mice treated with primidone showed significant delay of symptomatic onset and improved motor performance. One-hundred-sixty-two ALS participants dosed daily with primidone (62.5 mg) completed 24-week follow-up. With the development of IoT monitoring equipment, an increasing number of monitoring indicators are employed to monitor the operational status of water pumps, thereby resulting in the challenge of data redundancy. This paper proposes an algorithm for predicting the health status of pumps that integrates multiple monitoring variables. Initially, the original dataset is classified using the maximum relevance minimum redundancy method. Next, principal component dimensionality reduction is used to reduce the dimensionality of the classified dataset. Finally, a long and short term memory neural network is employed to construct the association model between monitoring data and equipment health. The proposed algorithm takes into account the correlation between variables and the negative impacts of long-term dependence on the prediction results. It is capable of predicting abnormal working conditions, which has been experimentally verified in the Xiasha Pumping Station located in Hangzhou. The algorithm was compared with LR, SVM, and RNN algorithms, and it was found that the proposed algorithm achieved the highest prediction accuracy. Abstract The importance of psychological detachment during nonwork time has been emphasized, and its effect on depressive symptoms has been identified. However, the mechanism underlying this association remains to be elucidated. This study was conducted to identify the indirect role of emotional exhaustion in the association between psychological detachment and depressive symptoms and the moderating role of self-compassion in the relationship between emotional exhaustion and depressive symptoms on the basis of the recovery–engagement–exhaustion model and emotion regulation theory. Through an online survey, relevant data were collected from 727 university teachers (mean age, 37.65 ± 7.77 years; 38.5% men). Indirect and moderation effect were analyzed through structural equation modeling (Mplus). Psychological detachment was found to be associated with depressive symptoms through emotional exhaustion. Self-compassion moderated the association between emotional exhaustion and depressive symptoms. The association between emotional exhaustion and depressive symptoms was weaker among university teachers with high levels of self-compassion than among those with low levels of self-compassion. This study improves our understanding of the association between psychological detachment and depressive symptoms by identifying the emotional pathway and protective function of self-compassion. Interventions for improving mental health in work context should be designed considering psychological detachment and self-compassion to deal with work-induced emotional strain.

Thin-walled concrete-filled steel tubes are efficient and economical with promising applications in civil and light industrial buildings. However, their local buckling resistance and deformation capacity are low, which adversely affects the seismic safety of structures. There are relatively few studies on thin-walled concrete-filled steel tubular columns with ultra-large diameter-to-thickness ratios, and there is also a lack of relevant experimental research on them. In this study, horizontal hysteresis tests were conducted on concrete columns with a large diameter-to-thickness ratio. The seismic performances of regular and straight-ribbed specimens were analyzed and compared, including the analyses of load-displacement hysteresis curves, strain distribution, skeleton curves, ductility, and energy dissipation capacity. Using these results, a restoring force model for concrete columns with a large diameter-to-thickness ratio was established. The findings indicate that under horizontal loading, the ductility of concrete columns with a regular thin-walled steel tube is 3.9, with an equivalent viscous damping coefficient of 1.65. Meanwhile, the ultimate bearing capacity is 201 kN. After adding stiffening ribs, the ultimate bearing capacity reaches 266 kN and the ductility coefficient reaches 4.4, resulting in the stiffeners increasing the ultimate bearing capacity and ductility by >30% and 12.8%, respectively. However, they have a less pronounced effect on deformation and energy dissipation. Building on these research outcomes, we propose a dimensionless three-line skeleton curve model and a restoring force model. The calculation results from these models align well with the test results, offering valuable insights for the seismic safety analysis of real-world engineering structures.

Nickel-based catalysts are regarded as the most excellent urea oxidation reaction (UOR) catalysts in alkaline media. Whatever kind of nickel-based catalysts is utilized to catalyze UOR, it is widely believed that the in situ-formed Ni3+ moieties are the true active sites and the as-utilized nickel-based catalysts just serve as pre-catalysts. Digging the pre-catalyst effect on the activity of Ni3+ moieties helps to better design nickel-based catalysts. Herein, five different anions of OH−, CO32−, SiO32−, MoO42−, and WO42− were used to bond with Ni2+ to fabricate the pre-catalysts β-Ni(OH)2, Ni-CO3, Ni-SiO3, Ni-MoO4, and Ni-WO4. It is found that the true active sites of the five as-fabricated catalysts are the same in situ-formed Ni3+ moieties and the five as-fabricated catalysts demonstrate different UOR activity. Although the as-synthesized five catalysts just serve as the pre-catalysts, they determine the quantity of active sites and activity per active site, thus determining the catalytic activity of the catalysts. Among the five catalysts, the amorphous nickel tungstate exhibits the most superior activity per active site and can catalyze UOR to reach 158.10 mA·cm–2 at 1.6 V, exceeding the majority of catalysts. This work makes for a deeper understanding of the pre-catalyst effect on UOR activity and helps to better design nickel-based UOR catalysts.

Objective: Extracellular vesicles (EVs) have garnered considerable attention among researchers as candidates for natural drug delivery systems. This study aimed to investigate whether extracellular vesicle mediated targeting delivery of growth differentiation factor-15 (GDF15) improves myocardial repair by reprogramming macrophages post myocardial injury. Methods: EVs were isolated from macrophages transfected with GDF15 (EXO-GDF15) and control macrophages (EXO-NC). In vitro and vivo experiments, we compared their reprogram ability of macrophages and regeneration activity. Furthermore, proteomic analysis were employed to determine the specific mechanism by which GDF15 repairs the myocardium. Results: Compared with EXO-NC, EXO-GDF15 significantly regulated macrophage phenotypic shift, inhibited cardiomyocyte apoptosis, and enhanced endothelial cell angiogenesis. Moreover, EXO-GDF15 also significantly regulated macrophage heterogeneity and inflammatory cytokines, reduced fibrotic area, and enhanced cardiac function in infarcted rats. Proteomic analysis revealed a decrease in fatty acid-binding protein 4 (FABP4) protein expression following treatment with EXO-GDF15. Mechanistically, the reprogramming of macrophages by EXO-GDF15 is accomplished through the activation of Smad2/3 phosphorylation, which subsequently inhibits the production of FABP4. Conclusions: Extracellular vesicle mediated targeting delivery of growth differentiation factor-15 improves myocardial repair by reprogramming macrophages post myocardial injury via down-regulating the expression of FABP4. EXO-GDF15 may serve as a promising approach of immunotherapy.

Periprosthetic hip infection caused by Brucella abortus is rare and only a few cases have been reported. This current case report presents a case of a man in his early 50s who developed periprosthetic hip infection 2 years after right hip arthroplasty. There was no fever or pain, the usual cardinal signs of infection, except for a sinus tract at the previous surgical incision. Laboratory and arthrocentesis culture examinations (done twice) confirmed infection with B. abortus . Accordingly, a two-stage revision surgery was performed accompanied by antibiotic treatment with doxycycline and rifampicin after each stage. There was no recurrence at the 2-year follow-up, with good functional recovery of the hip joint. Clinically, this case serves to highlight the fact that periprosthetic hip infections caused by B. abortus might not present with the typical symptoms such as fever or hip pain. Furthermore, this current case involved a chronic sinus tract, so the diagnostic and therapeutic course of this case offers useful insights for managing similar cases in the future. In addition, a review of the literature on the diagnosis and treatment of Brucella -caused periprosthetic hip infection is presented.

IntroductionDiabetic nephropathy (DN), distinguished by detrimental changes in the renal glomeruli, is regarded as the leading cause of death from end-stage renal disease among diabetics. Cellular senescence plays a paramount role, profoundly affecting the onset and progression of chronic kidney disease (CKD) and acute kidney injuries. This study was designed to delve deeply into the pathological mechanisms between glomerulus-associated DN and cellular senescence.MethodsGlomerulus-associated DN datasets and cellular senescence-related genes were acquired from the Gene Expression Omnibus (GEO) and CellAge database respectively. By integrating bioinformatics and machine learning methodologies including the LASSO regression analysis and Random Forest, we screened out four signature genes. The receiver operating characteristic (ROC) curve was performed to evaluate the diagnostic performance of the selected genes. Rigorous experimental validations were subsequently conducted in the mouse model to corroborate the identification of three signature genes, namely LOX, FOXD1 and GJA1. Molecular docking with chlorogenic acids (CGA) was further established not only to validate LOX, FOXD1 and GJA1 as diagnostic markers but also reveal their potential therapeutic effects.Results and discussionIn conclusion, our findings pinpointed three diagnostic markers of glomerulus-associated DN on the basis of cellular senescence. These markers could not only predict an increased risk of DN progression but also present promising therapeutic targets, potentially ushering in innovative treatments for DN in the elderly population.

Abstract The reduction of noises, achieved through absorption, is of paramount importance to the well‐being of both humans and machines. Lattice structures, defined as architectured porous solids arranged in repeating patterns, are emerging as advanced sound‐absorbing materials. Their immense design freedom allows for customizable pore morphology and interconnectivity, enabling the design of specific absorption properties. Thus far, the sound absorption performance of various types of lattice structures are studied and they demonstrated favorable properties compared to conventional materials. Herein, this review gives a thorough overview on the current research status, and characterizations for lattice structures in terms of acoustics is proposed. Till date, there are four main sound absorption mechanisms associated with lattice structures. Despite their complexity, lattice structures can be accurately modelled using acoustical impedance models that focus on critical acoustical geometries. Four defining features: morphology, relative density, cell size, and number of cells, have significant influences on the acoustical geometries and hence sound wave dissipation within the lattice. Drawing upon their structural‐property relationships, a classification of lattice structures into three distinct types in terms of acoustics is proposed. It is proposed that future attentions can be placed on new design concepts, advanced materials selections, and multifunctionalities.

IntroductionBroken eggs are a byproduct of the poultry industry and a potential nitrogen source for mushroom cultivation. However, its feasibility needs to be evaluated experimentally.MethodsIn this study, a series of different addition amounts (0, 1.8, 3.6, 5.3 and 8.5%, w/w) of broken egg mixture (BEM) were applied in the composting cultivation process of oyster mushroom. The physicochemical properties and bacterial communities of composting substrate, and agronomic and nutritional properties of fruiting bodies were determined.Results and discussionThe results showed that the BEM addition significantly (P < 0.05) increased the total nitrogen content in the composted substrate, and the contents of crude protein, total amino acids and essential amino acids of mushrooms. The P3 treatment (initial C/N of 26:1) showed the highest biological efficiency (BE) of 100.19% and a low contamination rate (CR) of 7.00%, while the higher dosage of BEM (P4 and P5) led to a sharp decrease in BE and a sharp increase in CR. High throughput sequencing revealed that the addition of BEM significantly (P < 0.05) changed the bacterial communities in the substrate at the beginning of composting. Streptococcus and Lactococcus were predominant bacterial genera in BEM treatments at the beginning stage of composting, while Acinetobacter became predominant at the ending stage. The co-occurrence network analysis showed that the P3 treatment demonstrated a much more complex bacterial community. The structural equation model analysis indicated that the addition of BEM affected the bacterial communities and nitrogen metabolism during composting, which further affected agronomic and nutritional properties of oyster mushrooms. An appropriate amount of BEM combined with composting processes can significantly improve the yield and quality of oyster mushroom, providing a new way for efficient utilization of BEM.

Background Despite the increasing long‐term survival after out‐of‐hospital cardiac arrest (OHCA), the risk of subsequent acute myocardial infarction (AMI) remains poorly understood. We aimed to determine the incidence, predictors, and long‐term outcomes of AMI among survivors of OHCA. Methods and Results We assembled a retrospective cohort of 882 patients with OHCA who survived to 30 days or discharge from the hospital between 2010 and 2019. Survivors of OHCA had an increased risk of subsequent AMI, defined as AMI occurring 30 days after index OHCA or following discharge from the hospital after OHCA, compared with the general population when matched for age and sex (standardized incidence ratio, 4.64 [95% CI, 3.52–6.01]). Age‐specific risks of subsequent AMI for men (standardized incidence ratio, 3.29 [95% CI, 2.39–4.42]) and women (standardized incidence ratio, 6.15 [95% CI, 3.27–10.52]) were significantly increased. A total of 7.2%, 8.3%, and 14.3% of survivors of OHCA had a subsequent AMI at 3 years, 5 years, and end of follow‐up, respectively. Age at OHCA (hazard ratio [HR], 1.04 [95% CI, 1.02–1.06]) and past medical history of prior AMI, defined as any AMI preceding or during the index OHCA event (HR, 1.84 [95% CI, 1.05–3.22]), were associated with subsequent AMI, while an initial shockable rhythm was not (HR, 1.00 [95% CI, 0.52–1.94]). Survivors of OHCA with subsequent AMI had a higher risk of death (HR, 1.58 [95% CI, 1.12–2.22]) than those without. Conclusions Survivors of OHCA are at an increased risk of subsequent AMI compared with the general population. Prior AMI, but not an initial shockable rhythm, increases this risk, while subsequent AMI predicts death. Preventive measures for AMI including cardiovascular risk factor control and revascularization may thus improve outcomes in selected patients with cardiac pathogenesis.

ObjectiveOrganizational support has been identified as one of the causes for task performance, while previous studies have not adequately explored the underlying mechanisms. Thus, this study aims to reveal the potential mechanisms that linked organizational support to task performance.MethodsA questionnaire survey was conducted among the 720 participants from 12 audit firms in Beijing in December 2020. Participants completed anonymous questionnaires assessing their organizational support, task performance, organizational commitment, and job satisfaction. Data analysis was conducted with SPSS 26.0.Results(1) Organizational support was positively associated with task performance, (2) job satisfaction and organizational commitment both mediated the relationship between organizational support and task performance respectively, and (3) the relationship between organizational support and task performance was also serially mediated by job satisfaction and then organizational commitment.ConclusionJob satisfaction and organizational commitment played a serial multiple mediating role in the association between organizational support and task performance, which provides a potential path for improving task performance.

In order to explore the fracture zone characteristics and microseismic evolution within overburden rock above mining coal seam, based on the geological engineering conditions of a mine, a microseismic simulation method was constructed according to the moment tensor and particle flow theory, and the microseismic evolution characteristics of overburden rock above panels were simulated. The results show that the magnitude of microseismic events in mining-disturbed overburden is between −2.7 and −1, and the microseismic fracture strength conforms to the Gaussian distribution. The relationship between microseismic frequency and moment magnitude also satisfies the classical moment magnitude-frequency relationship. The microseismic events show prominent regional distribution characteristics in the horizontal direction of the panel. The microseismic events in the left and right coal walls of the goaf occur earlier than the roof above goaf, and the microseismic events develop to both sides around the mining space, and there is a certain advance and lag. When the panel is mined for a certain distance, the vertical stress of the overburden above the goaf shows a 'double peak' characteristic, and the microseismic events extend upward rapidly and gather and develop in the weak overburden so that the microseismic events also show prominent layered distribution characteristics in the vertical direction. According to the stress, displacement, and microseismic distribution characteristics within overburden, the overburden can be divided into ‘four horizontal zones’ and ‘three vertical zones’. The proportion of microseismic events and fracture types in each fracture area is different. The coal wall supported and bed separation areas are the large proportion areas of shear and tensile microseismic events, respectively. In addition, a microseismic monitoring system was established for the study area. The field monitoring and numerical simulation results are consistent, indicating that the constructed microseismic simulation method has a good applicability and can reproduce the microseismic evolution process of mining-disturbed overburden from the mesoscopic level. The research results provide a theoretical basis for revealing the evolution mechanism of mining-induced fractures.

Biomaterials are natural, synthetic, or hybrid materials, which are used in medical devices or implants that are placed in contact with the human biological system to compensate for or restore diminished functions of the body. The field of biomaterials has rapidly developed to meet the ever-expanding needs in healthcare and medicine practices. Advancements in science and technology have enabled the fabrication and reengineering of biomaterials into useful medical devices or implants, such as heart valves, bone plates, hip joints, and cardiac pacemakers. Because biomaterials are placed in continuous close contact with the recipient’s body fluids or tissues, the classification of available biomaterials is crucial for selecting safer and highly biocompatible materials. This review focuses on biomaterial classification, namely bioceramic, polymeric, and metallic biomaterials. Their medical applications, advantages, and disadvantages are discussed. Current trends in biomaterials involved in disease treatments, such as controlled drug delivery and cancer therapy, are additionally explored.

Abstract Background Glioblastoma (GBM) is the most malignant glioma, with poor survival rates and prognosis. Several studies have reported the abnormal expression of circular RNAs (circRNAs) and their functions in the malignant biological behavior of GBM. However, such research is still in the preliminary stages, and further study is needed to confirm the therapeutic potential of circRNAs in GBM. Methods RNA-seq was performed using four tumor tissues from patients with GBM and their adjacent non-tumor brain tissues to screen differentially expressed circRNAs. Fluorescence in situ hybridization assay was used to examine the location of circ_0021350 in glioma cells. In addition, a series of biological function assays were employed to verify the oncogenic role of circ_0021350 in GBM. Quantitative reverse transcription PCR was used to examine circular, micro- (miRNA), and messenger RNA (mRNA) levels. Furthermore, dual-luciferase reporter, RNA pull-down, and RNA binding protein immunoprecipitation assays were applied to verify the interaction between circ_0021350 and its downstream effectors. Results Circ_0021350 was significantly elevated in GBM tissues and glioma cells. Overexpression of circ_0021350 promoted glioma cell proliferation and metastatic ability; silencing of circ_0021350 had the opposite effect. Mechanistic analysis revealed that circ_0021350 sponged miR-1207-3p to regulate PIK3R3, whose overexpression reversed the reduction in the malignant biological behavior of glioma cells caused by silencing circ_0021350. Conclusion Our findings suggest that circ_0021350 is an oncogenic circRNA in GBM, and the circ_0021350/miR-1207-3p/PIK3R3 axis may serve as a potential therapeutic target in GBM treatment.

Laser powder bed fusion (L-PBF) has been proposed as a potential manufacturing process of high-entropy alloys involving rapid cooling rate and compositional element tuning. It, to some extent, compensates for the lack of strength of face-centered cubic (FCC) CoCrFeNi alloy by introducing high-density dislocations and generating hierarchical structures. In this study, the CoCrFeNi-based alloy was manufactured by L-PBF with the minor addition of B4C micro-particles and their properties and microstructure were evaluated. Nano-sized precipitates consisting of the boride and varying carbides were found in the form of a core-shell structure. An in-site transformation of carbides appeared around the core of the precipitate due to a large temperature gradient in the melt pool during manufacturing. The tensile yield and ultimate strength of CoCrFeNi–B4C alloy (1249.5 MPa and 1421.0 MPa) are double that of CoCrFeNi matrix (624.5 MPa and 691.1 MPa) at room temperature with competitive ductility. The exceptional mechanical properties can be mainly ascribed to precipitate hardening, dislocation hardening and grain refinement, as well as the particle-dislocation interaction.

Abstract Mesenchymal stem cells (MSCs) possess potent immunomodulatory activity and have been extensively investigated for their therapeutic potential in treating inflammatory disorders. However, the mechanisms underlying the immunosuppressive function of MSCs are not fully understood, hindering the development of standardized MSC-based therapies for clinical use. In this study, we profile the single-cell transcriptomes of MSCs isolated from adipose tissue (AD), bone marrow (BM), placental chorionic membrane (PM), and umbilical cord (UC). Our results demonstrate that MSCs undergo a progressive aging process and that the cellular senescence state influences their immunosuppressive activity by downregulating PD-L1 expression. Through integrated analysis of single-cell transcriptomic and proteomic data, we identify GATA2 as a regulator of MSC senescence and PD-L1 expression. Overall, our findings highlight the roles of cell aging and PD-L1 expression in modulating the immunosuppressive efficacy of MSCs and implicating perinatal MSC therapy for clinical applications in inflammatory disorders.

Abstract Background Blood–brain barrier (BBB) is a crucial but dynamic structure that functions as a gatekeeper for the central nervous system (CNS). Managing sufficient substances across the BBB is a major challenge, especially in the development of therapeutics for CNS disorders. Methods To achieve an efficient, fast and safe strategy for BBB opening, an acoustofluidic transwell (AFT) was developed for reversible disruption of the BBB. The proposed AFT was consisted of a transwell insert where the BBB model was established, and a surface acoustic wave (SAW) transducer realized using open-source electronics based on printed circuit board techniques. Results In the AFT device, the SAW produced acousto-mechanical stimulations to the BBB model resulting in decreased transendothelial electrical resistance in a dose dependent manner, indicating the disruption of the BBB. Moreover, SAW stimulation enhanced transendothelial permeability to sodium fluorescein and FITC-dextran with various molecular weight in the AFT device. Further study indicated BBB opening was mainly attributed to the apparent stretching of intercellular spaces. An in vivo study using a zebrafish model demonstrated SAW exposure promoted penetration of sodium fluorescein to the CNS. Conclusions In summary, AFT effectively disrupts the BBB under the SAW stimulation, which is promising as a new drug delivery methodology for neurodegenerative diseases. Graphical Abstract

Abstract Background As a life-threatening and serious condition, aortic dissection (AD) is divided into type A and B according to its association with the ascending or descending aorta. Type A AD is often accompanied by aortic regurgitation, while type B dissections are rarely accompanied by severe aortic regurgitation. Case presentation We present a 71 year-old Chinese man with a rare case of type B AD with severe aortic insufficiency, who self-healed after 1 year of an aortic valve replacement. He complained of chest tightness and abdominal pain. Due to poor cardiac function, he underwent aortic valve replacement before intervening on the dissection. The operation was successful, and the dissection was treated conservatively. During the 1-year follow-up, his chest tightness improved, and the type B dissection was healed. His general condition is considerably improved. Conclusions In type B AD combined with severe aortic insufficiency, aortic valve replacement should be prioritized. This is potentially explained by the aortic root activity and pulse pressure difference.

Phytoremediation is recognized as an environmentally friendly technique. However, the low biomass production, high time consumption, and exposure to combined toxic stress from contaminated media weaken the potential of phytoremediation. As a class of plant-beneficial microorganisms, arbuscular mycorrhizal fungi (AMF) can promote plant nutrient uptake, improve plant habitats, and regulate abiotic stresses, and the utilization of AMF to enhance phytoremediation is considered to be an effective way to enhance the remediation efficiency. In this paper, we searched 520 papers published during the period 2000–2023 on the topic of AMF-assisted phytoremediation from the Web of Science core collection database. We analyzed the author co-authorship, country, and keyword co-occurrence clustering by VOSviewer. We summarized the advances in research and proposed prospective studies on AMF-assisted phytoremediation. The bibliometric analyses showed that heavy metal, soil, stress tolerance, and growth promotion were the research hotspots. AMF–plant symbiosis has been used in water and soil in different scenarios for the remediation of heavy metal pollution and organic pollution, among others. The potential mechanisms of pollutant removal in which AMF are directly involved through hyphal exudate binding and stabilization, accumulation in their structures, and nutrient exchange with the host plant are highlighted. In addition, the tolerance strategies of AMF through influencing the subcellular distribution of contaminants as well as chemical form shifts, activation of plant defenses, and induction of differential gene expression in plants are presented. We proposed that future research should screen anaerobic-tolerant AMF strains, examine bacterial interactions with AMF, and utilize AMF for combined pollutant removal to accelerate practical applications.

Access to mental healthcare is undoubtedly of major importance for LGBT+ people worldwide, given the high prevalence of mental health difficulties due to minority stress exposures. This study drew mixed-method survey data from the community-based KAMI Survey (n = 696) to examine the enablers, barriers, and unmet needs experiences of LGBT+ individuals in accessing mental healthcare services in Malaysia. First, we present findings from a series of descriptive analyses for sociodemographic differences in unmet needs for mental healthcare, barriers, and satisfaction levels with different types of mental healthcare. Next, we conducted an inductive thematic analysis of open-text comments (n = 273), with relevance drawn to Andersen’s Behavioural Model of Healthcare. More than a quarter (29.5%) reported an unmet need for mental healthcare, and some groups (younger, asexual or queer, or participants living in non-major cities) reported higher unmet needs. More than three-fifths (60.5%) reported not knowing where to find culturally safe mental health professionals. The thematic analysis uncovered key contextual (e.g., mental health practitioners’ stance, stigma, collaborative client-care) and individual (e.g., positive expectation of mental health services and anticipated stigma) attributes that influence healthcare experiences. Participants also identified resources that facilitate healthcare utilisation, such as affordability, availability of suitable professionals, and geographical considerations. The implications of our findings for the mental healthcare practices in Malaysia were outlined.
