Background: An ancient family of arrestin-fold proteins, termed alpha-arrestins, may have conserved roles in regulating nutrient transporter trafficking and cellular metabolism as adaptor proteins. One alpha-arrestin, TXNIP (thioredoxin-interacting protein), is known to regulate myocardial glucose uptake. However, the in vivo role of the related alpha-arrestin, ARRDC4 (arrestin domain-containing protein 4), is unknown. Methods: We first tested whether interaction with GLUTs (glucose transporters) is a conserved function of the mammalian alpha-arrestins. To define the in vivo function of ARRDC4, we generated and characterized a novel Arrdc4 knockout (KO) mouse model. We then analyzed the molecular interaction between arrestin domains and the basal GLUT1. Results: ARRDC4 binds to GLUT1, induces its endocytosis, and blocks cellular glucose uptake in cardiomyocytes. Despite the closely shared protein structure, ARRDC4 and its homologue TXNIP operate by distinct molecular pathways. Unlike TXNIP, ARRDC4 does not increase oxidative stress. Instead, ARRDC4 uniquely mediates cardiomyocyte death through its effects on glucose deprivation and endoplasmic reticulum stress. At baseline, Arrdc4 -KO mice have mild fasting hypoglycemia. Arrdc4 -KO hearts exhibit a robust increase in myocardial glucose uptake and glycogen storage. Accordingly, deletion of Arrdc4 improves energy homeostasis during ischemia and protects cardiomyocytes against myocardial infarction. Furthermore, structure-function analyses of the interaction of ARRDC4 with GLUT1 using both scanning mutagenesis and deep-learning Artificial Intelligence identify specific residues in the C-terminal arrestin-fold domain as the interaction interface that regulates GLUT1 function, revealing a new molecular target for potential therapeutic intervention against myocardial ischemia. Conclusions: These results uncover a new mechanism of ischemic injury in which ARRDC4 drives glucose deprivation-induced endoplasmic reticulum stress leading to cardiomyocyte death. Our findings establish ARRDC4 as a new scaffold protein for GLUT1 that regulates cardiac metabolism in response to ischemia and provide insight into the therapeutic strategy for ischemic heart disease.

Underlying molecular mechanisms for the development of diabetic cardiomyopathy remain to be determined. Long-term exposure to hyperglycemia causes oxidative stress, which leads to cardiomyocyte dysfunction. Previous studies established the importance of thioredoxin-interacting protein (Txnip) in cellular redox homeostasis and glucose metabolism. Txnip is a highly glucose-responsive molecule that interacts with the catalytic center of reduced thioredoxin and inhibits the antioxidant function of thioredoxin. Here, we show that the molecular interaction between Txnip and thioredoxin plays a pivotal role in the regulation of redox balance in the diabetic myocardium. High glucose increased Txnip expression, decreased thioredoxin activities, and caused oxidative stress in cells. The Txnip-thioredoxin complex was detected in cells with overexpressing wild-type Txnip but not Txnip cysteine 247 to serine (C247S) mutant that disrupts the intermolecular disulfide bridge. Then, diabetes was induced in cardiomyocyte-specific Txnip C247S knock-in mice and their littermate control animals by injections of streptozotocin (STZ). Prolonged hyperglycemia upregulated myocardial Txnip expression in both genotypes. The absence of Txnip's inhibition of thioredoxin in Txnip C247S mutant hearts promoted mitochondrial antioxidative capacities in cardiomyocytes, thereby protecting the heart from oxidative damage by diabetes. Stress hemodynamic analysis uncovered that Txnip C247S knock-in hearts have a greater left ventricular contractile reserve than wild-type hearts under STZ-induced diabetic conditions. These results provide novel evidence that Txnip serves as a regulator of hyperglycemia-induced cardiomyocyte toxicities through direct inhibition of thioredoxin and identify the single cysteine residue in Txnip as a therapeutic target for diabetic injuries.

Rationale Thioredoxin-interacting protein (Txnip) is a novel molecular target with translational potential in diverse human diseases. Txnip has several established cellular actions including binding to thioredoxin, a scavenger of reactive oxygen species (ROS). It has been long recognized from in vitro evidence that Txnip forms a disulfide bridge through cysteine 247 (C247) with reduced thioredoxin to inhibit the anti-oxidative properties of thioredoxin. However, the physiological significance of the Txnip-thioredoxin interaction remains largely undefined in vivo. Objective A single mutation of Txnip, C247S, abolishes the binding of Txnip with thioredoxin. Using a conditional and inducible approach with a mouse model of a mutant Txnip that does not bind thioredoxin, we tested whether the interaction of thioredoxin with Txnip is required for Txnip's pro-oxidative or cytotoxic effects in the heart. Methods and results Overexpression of Txnip C247S in cells resulted in a reduction in ROS, due to an inability to inhibit thioredoxin. Hypoxia (1% O2, 24 h)-induced killing effects of Txnip were decreased by lower levels of cellular ROS in Txnip C247S-expressing cells compared with wild-type Txnip-expressing cells. Then, myocardial ischemic injuries were assessed in the animal model. Cardiomyocyte-specific Txnip C247S knock-in mice had better survival with smaller infarct size following myocardial infarction (MI) compared to control animals. The absence of Txnip's inhibition of thioredoxin promoted mitochondrial anti-oxidative capacities in cardiomyocytes, thereby protecting the heart from oxidative damage induced by MI. Furthermore, an unbiased RNA sequencing screen identified that hypoxia-inducible factor 1 signaling pathway was involved in Txnip C247S-mediated cardioprotective mechanisms. Conclusion Txnip is a cysteine-containing redox protein that robustly regulates the thioredoxin system via a disulfide bond-switching mechanism in adult cardiomyocytes. Our results provide the direct in vivo evidence that regulation of redox state by Txnip is a crucial component for myocardial homeostasis under ischemic stress.

Marangoni flow driven by a temperature gradient was observed near the isotropic-nematic phase transition point. By applying the gradient to a liquid crystalline material in sandwich cells, it was possible to measure the flow field near the air interface using the photobleaching method. In the isotropic phase, the direction of the observed flow was opposite to that in the nematic phase. Moreover, when the measurement was performed in the coexistence state of these phases, the flow direction depended on the coating materials of the cell substrates. These singular flow properties are explained well by the singular changes in surface tension and the shape of the air interface near the transition point.

Steady rotation is induced in cholesteric droplets dispersed in a specific liquid solvent under a temperature gradient. In this phenomenon, two rotational modes have been considered: (1) collective rotation of the local director field and (2) rigid-body rotation of the whole droplet structure. However, here we present another rotational mode induced in a pillar-shaped cholesteric droplet confined between substrates under a temperature gradient, that is, a differential rotation where the angular velocity varies as a function of the radial coordinate in the pillar. A detailed flow field analysis revealed that every pillar under a temperature gradient involves a double convection roll. These results suggested that the differential rotation in the cholesteric pillars was driven by the inhomogeneous material flow induced by a temperature gradient. The present experimental study indicates that the coupling between the flow and the director motion plays a key role in the rotation of the cholesteric droplets under the temperature gradient.

Using a photo-responsive dimer exhibiting the transition between nematic (N) and twist-bend nematic (NTB) phases, we prepared spherical cap-shaped droplets on solid substrates exposed to air. The internal director structures of these droplets vary depending on the phase and on the imposed boundary conditions. The structural switching between the N and NTB phases was successfully performed either by temperature control or by UV light-irradiation. The N phase is characterized by an extremely small bend elastic constant K3, and surprisingly, we found that the droplet-air interface induces a planar alignment, in contrast to that seen for typical calamitic liquid crystals. As a consequence, the director configuration was stabilized in a structure substantially different from that normally found in conventional nematic liquid crystalline droplets. In the twist-bend nematic droplets characteristic structures with macroscopic length scales were formed, and they were well controlled by the droplet size. These results indicated that a continuum theory is effective in describing the stabilization mechanism of the macroscopic structure even in the twist-bend nematic liquid crystal droplets exhibiting director modulations on a scale of several molecular lengths.

When heat flux is applied to a chiral liquid crystal, unidirectional rotation is induced around the flux axis, as first discovered by Otto Lehmann in 1900. In recent years, this heat-flux-induced phenomenon has been studied mostly in droplets of cholesteric liquid crystals undergoing phase transition from the isotropic to cholesteric phase, i.e., in the coexistence region, which occurs over a very narrow temperature range. Here, we report that the heat-flux-induced rotation can be stabilised by the use of a dispersion system, in which the cholesteric droplets are dispersed in a viscous and poorly miscible isotropic solvent. Interestingly, the phenomenon is found to be topology dependent. Moreover, the rotation is not only stable but also more efficient than that in the known systems. We describe in detail how the dynamics of the heat-flux-induced rotation are altered in the present dispersion system., The Lehmann effect describes the spontaneous rotation of cholesteric liquid crystals in response to heat input. Here, the authors stabilise it by dispersing cholesteric droplets into a poorly miscible solvent and show dependences of rotation speed and conversion efficiency on the topological states.

Background No-touch environmental disinfection using ultraviolet devices has been highlighted in the past several years to control the transmission of multidrug-resistant organisms (MDROs). However, its effectiveness in non-US healthcare settings is yet to be examined. This study aimed to evaluate the effectiveness of disinfection by portable pulsed xenon ultraviolet (PX-UV) devices in controlling transmission of MDROs in a non-US healthcare setting. Methods All patients admitted in the intensive care unit in a 629-bed tertiary referral hospital in Japan from August 2016 to February 2019 were enrolled. During the study period, PX-UV disinfection was added to manual terminal cleaning after every patient transfer/discharge. For microbiological evaluation, surfaces were selected for sampling by contact plates before/after manual cleaning and after PX-UV. After overnight incubation, colonies on the plates were counted. Results The incidence of newly acquired methicillin-resistant Staphylococcus aureus (MRSA) declined significantly (13.8 to 9.9 per 10,000 patient days, incidence rate ratio 0.71, p = 0.002), as well as that of newly acquired drug-resistant Acinetobacter (48.5 to 18.1, 0.37, p Conclusions PX-UV is effective in further reducing the microbial burden and controlling MDROs in a non-US healthcare setting.

Myocardial ischemia/reperfusion injury represents a major threat to human health and contributes to adverse cardiovascular outcomes worldwide. Despite the identification of numerous molecular mechanisms, understanding of the complex pathophysiology of this clinical syndrome remains incomplete. Thioredoxin-interacting protein (Txnip) has been of great interest in the past decade since it has been reported to be a critical regulator in human diseases with several important cellular functions. Thioredoxin-interacting protein binds to and inhibits thioredoxin, a redox protein that neutralizes reactive oxygen species (ROS), and through its interaction with thioredoxin, Txnip sensitizes cardiomyocytes to ROS-induced apoptosis. Interestingly, evidence from recent studies also suggests that some of the effects of Txnip may be unrelated to changes in thioredoxin activity. These pleiotropic effects of Txnip are mediated by interactions with other signaling molecules, such as nod-like receptor pyrin domain-containing 3 inflammasome and glucose transporter 1. Indeed, Txnip has been implicated in the regulation of inflammatory response and glucose homeostasis during myocardial ischemia/reperfusion injury. This review attempts to make the case that in addition to interacting with thioredoxin, Txnip contributes to some of the pathological consequences of myocardial ischemia and infarction through endogenous signals in multiple molecular mechanisms.

Although the precise pathogenesis of diabetic cardiac damage remains unclear, potential mechanisms include increased oxidative stress, autonomic nervous dysfunction, and altered cardiac metabolism. Thioredoxin-interacting protein (Txnip) was initially identified as an inhibitor of the antioxidant thioredoxin but is now recognized as a member of the arrestin superfamily of adaptor proteins that classically regulate G protein-coupled receptor signaling. Here we show that Txnip plays a key role in diabetic cardiomyopathy. High glucose levels induced Txnip expression in rat cardiomyocytes in vitro and in the myocardium of streptozotocin-induced diabetic mice in vivo. While hyperglycemia did not induce cardiac dysfunction at baseline, β-adrenergic challenge revealed a blunted myocardial inotropic response in diabetic animals (24-wk-old male and female C57BL/6;129Sv mice). Interestingly, diabetic mice with cardiomyocyte-specific deletion of Txnip retained a greater cardiac response to β-adrenergic stimulation than wild-type mice. This benefit in Txnip-knockout hearts was not related to the level of thioredoxin activity or oxidative stress. Unlike the β-arrestins, Txnip did not interact with β-adrenergic receptors to desensitize downstream signaling. However, our proteomic and functional analyses demonstrated that Txnip inhibits glucose transport through direct binding to glucose transporter 1 (GLUT1). An ex vivo analysis of perfused hearts further demonstrated that the enhanced functional reserve afforded by deletion of Txnip was associated with myocardial glucose utilization during β-adrenergic stimulation. These data provide novel evidence that hyperglycemia-induced Txnip is responsible for impaired cardiac inotropic reserve by direct regulation of insulin-independent glucose uptake through GLUT1 and plays a role in the development of diabetic cardiomyopathy.

In this study, we demonstrate a self-excited oscillation induced in cholesteric liquid crystalline droplets under a temperature gradient. At equilibrium, a winding Maltese cross pattern with a point defect was observed via polarised microscopy in the droplets dispersed in an isotropic solvent. When the temperature gradient was applied, the pattern was deformed owing to the Marangoni convection induced by the gradient. Here, when both the droplet size and temperature gradient were sufficiently large, the periodic movement of the defect together with the pattern deformation was observed, which demonstrated the self-excited oscillation of the director field. To describe this phenomenon, we theoretically analysed the flow and director fields by using Onsager's variational principle. This principle enabled the simplified description of the phenomenon; consequently, the time evolution of the director field could be expressed by the phenomenological equations for the two parameters characterising the field. These equations represented the van der Pol equation, which well expressed the mechanism of the self-excited oscillation.

Flow and director fields strongly couple with each other in liquid crystalline systems, and herein we discuss the coupling effect in cylindrical and spherical-cap droplets formed by nematic liquid crystal. Applying a temperature gradient to droplets dispersed in a liquid solvent, we observed a crosslike texture in the droplets moved toward the high-temperature side, indicating that the director field was deformed from equilibrium. Additionally, measurement of the flow field revealed that a convective flow was induced in the droplets under temperature gradient. These results suggested that the director deformation in the droplet was induced by convection. By designing a simplified model based on this, we theoretically analyzed the above phenomenon based on Onsager's variational principle. The results show that the phenomenon was well described by a balance of surface energy gradient with viscous and elastic forces.

Japanese clinical engineer (CE) is a significant and unique profession compared with other nations with its dual clinical and technology focus and national licensing. The CE system of licensing was established in May 1987 under the Clinical Engineers Act. CEs are required to complete 3 to 4 years in designated schools and pass a national examination. It is a professional medical position responsible for the operation and maintenance of life-support and non-life-support medical device systems under the direction of physicians. In Japan, CEs support and operate various life-support medical devices. Technology developments have led to significant improvements in performance, making devices easier to break and requiring specialized maintenance. Some of our healthcare technology management (HTM) initiatives include: 1. Rental equipment: oversee use and conduct in-house testing and repair, avoiding faulty units. 2. Ventilator maintenance: a multi-year track record of assessing and replacing defective parts in-house, contributing to prompt repairs and reduced costs. 3. Battery-equipped devices: created a more efficient system for charge management. 4. Intermittent pneumatic compression device dedicated tester: reducing the incidence of thromboses and embolism in patients.

Background Contact precautions do not necessarily control transmission of MDROs as other route of transmission such as patient environment may be significant. No-touch environmental disinfection has been highlighted in the past several years to control transmission of MDROs. The effectiveness of portable Pulsed Xenon Ultraviolet (PX-UV) device has been examined and demonstrated in US healthcare settings. However, its effectiveness outside the US healthcare setting is seldom reported. Methods This study was conducted in the intensive care unit (ICU) of Yamagata University Hospital, a 637-bed tertiary referral hospital. The ICU has six rooms and beds. In the baseline period (August 2016 to January 2018), all rooms were manually cleaned after every patient transfer/discharge. In the intervention period (February 2018 to February 2019), PX-UV disinfection was added after the manual cleaning. In both periods, all patients were screened for MRSA and two drug-resistant Acinetobacter baumannii (2DRA) to detect acquisition of those pathogens in the ICU. For microbiological evaluation, surfaces were selected for sampling by contact plates before/after manual cleaning and after PX-UV. After overnight incubation, colonies on the plates were counted. Results The incidence of newly acquired MRSA declined over time (1.40 per 1,000 patient-days in the baseline period to 0.95 in the intervention period, relative risk (RR): 0.68, 95% confidence interval (CI): 0.12–3.70). The incidence of newly acquired 2DRA further declined (4.91 to 1.90, RR: 0.39, 95% CI: 0.13–1.18). Notably, no new acquisition of 2DRA was observed since August 2018 for more than 7 months, not only in the ICU but also throughout the hospital. The total count of colonies in the sampling of 140 sites after 17 patient discharges were 3,540 (before manual cleaning), 669 (after manual cleaning, before PX-UV) and 261 (after PX-UV). The percent reduction of microbiological burden by manual cleaning was 81%, but a further 61% reduction was achieved by PX-UV. Conclusion PX-UV is effective in further reducing the microbial burden even after through manual cleaning, which presumably led to termination of transmission of 2DRA in our hospital. The effectiveness of PX-UV in controlling MDROs in the non-US healthcare settings is suggested. Disclosures All authors: No reported disclosures.

We found for the first time the stabilization of a double twisted structure in cholesteric liquid crystals confined to small spherical droplets under weak anchoring conditions. The direct observation of the droplets using a polarized microscope revealed the physical properties of the structure. The experimental results showed that the stability of the double twisted structure is determined by the relationship between the helical pitch length and the droplet size. We theoretically analyzed the structural stability by the calculation of the Frank elastic free energy including the surface elastic term, and succeeded in explaining the experimental results. In this paper, we concluded that the stability of the double twisted structure is determined by the competition between the surface and the bulk elasticity.

New chiral dimers consisting of a rod-like and cholesterol mesogenic units are reported to form a chiral twist-bend nematic phase (NTB *) with heliconical structure. The compressibility of the NTB phase made of bent dimers was found to be as large as in smectic phases, which is consistent with the nanoperiodic structure of the NTB phase. The atomic force microscopy observations in chiral bent dimers revealed a periodicity of about 50 nm, which is significantly larger than the one reported previously for non-chiral compounds (ca. 10 nm).

The endoplasmic reticulum (ER) is responsible for protein folding, modification, and trafficking. Accumulation of unfolded or misfolded proteins represents the condition of ER stress and triggers the unfolded protein response (UPR), a key mechanism linking supply of excess nutrients to insulin resistance and type 2 diabetes in obesity. The ER harbors proteins that participate in protein folding including protein disulfide isomerases (PDIs). Changes in PDI activity are associated with protein misfolding and ER stress. Here, we show that thioredoxin-interacting protein (Txnip), a member of the arrestin protein superfamily and one of the most strongly induced proteins in diabetic patients, regulates PDI activity and UPR signaling. We found that Txnip binds to PDIs and increases their enzymatic activity. Genetic deletion of Txnip in cells and mice led to increased protein ubiquitination and splicing of the UPR regulated transcription factor X-box-binding protein 1 (Xbp1s) at baseline as well as under ER stress. Our results reveal Txnip as a novel direct regulator of PDI activity and a feedback mechanism of UPR signaling to decrease ER stress.

In this study, we investigated the diffusion dynamics at the interface between deuterated poly(methyl methacrylate) (d-PMMA) and protonated poly(methyl methacrylate) (h-PMMA) in two-layered thin films of d- and h-PMMA layers via neutron reflectivity (NR) measurements during isothermal annealing above the glass transition temperature Tg. When Tg of d-PMMA was higher than that of h-PMMA, the d-PMMA layer thickness increased with increasing annealing time ta and, simultaneously, the h-PMMA layer thickness decreased. However, the opposite ta dependence of the layer thicknesses was observed, if the Tg of d-PMMA was decreased by the increase in the fraction of the low-molecular weight d-PMMA: With increasing ta, the d-PMMA layer thickness decreased and the h-PMMA layer thickness increased when Tg of d-PMMA was lower than that of h-PMMA. This change in the ta dependence of the layer thickness was related to the change in the mobility of the d-PMMA layer accompanied by the change in the Tg value of d-PMMA. With the decrease in the d-PMMA layer thickness from 49 nm to 13 nm, when the h-PMMA layer thickness was maintained, the ta dependence of the layer thickness changed and the mobility of the d-PMMA layer dramatically increased. These results suggest that the mobility of thin polymer films can be determined by the observation of interfacial dynamics via NR measurements.

Cellular metabolism and reactive oxygen species (ROS) formation are interrelated processes in mitochondria and are implicated in a variety of human diseases including ischemic heart disease. During ischemia, mitochondrial respiration rates fall. Though seemingly paradoxical, reduced respiration has been observed to be cardioprotective due in part to reduced generation of ROS. Enhanced myocardial glucose uptake is considered beneficial for the myocardium under stress, as glucose is the primary substrate to support anaerobic metabolism. Thus, inhibition of mitochondrial respiration and uncoupling oxidative phosphorylation can protect the myocardium from irreversible ischemic damage. Growing evidence now positions the TXNIP/thioredoxin system at a nodal point linking pathways of antioxidant defense, cell survival, and energy metabolism. This emerging picture reveals TXNIP's function as a regulator of glucose homeostasis and may prove central to regulation of mitochondrial function during ischemia. In this review, we summarize how TXNIP and its binding partner thioredoxin act as regulators of mitochondrial metabolism. While the precise mechanism remains incompletely defined, the TXNIP-thioredoxin interaction has the potential to affect signaling that regulates mitochondrial bioenergetics and respiratory function with potential cardioprotection against ischemic injury.

A change in basal glucose transport into skeletal muscle, heart, and adipose is a critical feature of insulin resistance and diabetes. Glucose transport is tightly regulated by a protein family of glucose transporters (GLUTs). Thus, defining how cardiomyocytes adapt to changes in extracellular and intracellular glucose concentrations by recruiting GLUTs is crucial to understanding cardiac metabolism under both normal and diabetic conditions. Thioredoxin-Interacting Protein (Txnip), originally characterized as a binding partner of antioxidant thioredoxin, is now known to be a member of the arrestin protein superfamily. We have previously shown that glucose uptake is robustly enhanced in Txnip-knockout hearts; however, the precise mechanism for this effect remains elusive. Here, we present a novel feedback mechanism by which Txnip controls glucose metabolism. High glucose levels induced Txnip expression in rat cardiomyocytes in vitro and in the myocardium of streptozotocin (STZ)-induced diabetic mice in vivo. Our proteomic and functional analyses found that Txnip directly interacts with GLUT1 and GLUT4 and strikingly inhibits cellular glucose uptake through direct interactions with GLUTs. Thus, a high extracellular glucose concentration represses glucose uptake into the cytoplasm via Txnip-mediated inhibition of GLUTs. The metabolic alterations affected by Txnip deletion promote myocardial glucose uptake, directing cardiomyocyte towards enhanced functional reserve under diabetic conditions. These results provide a novel link between GLUTs and Txnip and highlight a fundamental regulatory mechanism of glucose homeostasis in the heart.

Metabolic studies suggest that the absorptive capacity of the small intestine for fructose is limited, though the molecular mechanisms controlling this process remain unknown. Here we demonstrate that thioredoxin-interacting protein (Txnip), which regulates glucose homeostasis in mammals, binds to fructose transporters and promotes fructose absorption by the small intestine. Deletion of Txnip in mice reduced fructose transport into the peripheral bloodstream and liver, as well as the severity of adverse metabolic outcomes resulting from long-term fructose consumption. We also demonstrate that fructose consumption induces expression of Txnip in the small intestine. Diabetic mice had increased expression of Txnip in the small intestine as well as enhanced fructose uptake and transport into the hepatic portal circulation. The deletion of Txnip in mice abolished the diabetes-induced increase in fructose absorption. Our results indicate that Txnip is a critical regulator of fructose metabolism and suggest that a diabetic state can promote fructose uptake. The amount of fructose in our diet has increased dramatically in the last few decades. Growing evidence suggests that excessive amounts of fructose contribute to several metabolic diseases, including fatty liver disease and diabetes. Fructose is absorbed in the small intestine via transport proteins called GLUT2 and GLUT5 and then travels to the liver where it can stimulate the cells to make fats. However, it is not clear how fructose uptake is regulated in the small intestine. Glucose is taken into cells by a transport protein that is closely related to GLUT2 and GLUT5. Another protein called thioredoxin-interacting protein (Txnip) interacts with the glucose transporter and regulates glucose uptake. Here, Dotimas et al. investigated whether Txnip also regulates the activities of GLUT2 and GLUT5 to control how cells absorb fructose. Initial experiments in cells showed that Txnip binds to both GLUT2 and GLUT5 and increases the amount of fructose taken up by both mouse and human cells. Cells from mutant mice that do not produce Txnip absorbed less fructose than normal cells did. Furthermore, the mutant mice had lower levels of fructose in the blood and less severe metabolic disease after consuming fructose regularly for six months. Mice with diabetes absorbed more fructose through the small intestine than normal mice, and the loss of Txnip from these mice abolished this effect. Together the findings of Dotimas et al. suggest that Txnip plays an important role in regulating fructose absorption and indicate that, at least in some circumstances, diabetes may lead to more fructose being absorbed in the small intestine. The next steps following on from this work are to understand the molecular details of how Txnip regulates fructose uptake and to determine if other forms of diabetes also show increased fructose uptake. Classic therapeutics for ischemic heart disease are less effective in individuals with the metabolic syndrome. As the prevalence of the metabolic syndrome is increasing, better understanding of cardiac metabolism is needed to identify potential new targets for therapeutic intervention. Thioredoxin-interacting protein (Txnip) is a regulator of metabolism and an inhibitor of the antioxidant thioredoxins, but little is known about its roles in the myocardium. We examined hearts from Txnip-KO mice by polony multiplex analysis of gene expression and an independent proteomic approach; both methods indicated suppression of genes and proteins participating in mitochondrial metabolism. Consistently, Txnip-KO mitochondria were functionally and structurally altered, showing reduced oxygen consumption and ultrastructural derangements. Given the central role that mitochondria play during hypoxia, we hypothesized that Txnip deletion would enhance ischemia-reperfusion damage. Surprisingly, Txnip-KO hearts had greater recovery of cardiac function after an ischemia-reperfusion insult. Similarly, cardiomyocyte-specific Txnip deletion reduced infarct size after reversible coronary ligation. Coordinated with reduced mitochondrial function, deletion of Txnip enhanced anaerobic glycolysis. Whereas mitochondrial ATP synthesis was minimally decreased by Txnip deletion, cellular ATP content and lactate formation were higher in Txnip-KO hearts after ischemia-reperfusion injury. Pharmacologic inhibition of glycolytic metabolism completely abolished the protection afforded the heart by Txnip deficiency under hypoxic conditions. Thus, although Txnip deletion suppresses mitochondrial function, protection from myocardial ischemia is enhanced as a result of a coordinated shift to enhanced anaerobic metabolism, which provides an energy source outside of mitochondria.

Inherited mutations cause approximately 30% of all dilated cardiomyopathy cases, with autosomal dominant mutations in the LMNA gene accounting for more than one third of these. The LMNA gene encodes the nuclear envelope proteins lamins A and C, which provide structural support to the nucleus and also play critical roles in transcriptional regulation. Functional deletion of a single allele is sufficient to trigger dilated cardiomyopathy in humans and mice. However, whereas Lmna(-/-) mice develop severe muscular dystrophy and dilated cardiomyopathy and die by 8 weeks of age, heterozygous Lmna(+/-) mice have a much milder phenotype, with changes in ventricular function and morphology only becoming apparent at 1 year of age. Here, we studied 8- to 20-week-old Lmna(+/-) mice and wild-type littermates in a pressure overload model to examine whether increased mechanical load can accelerate or exacerbate myocardial dysfunction in the heterozygotes. While overall survival was similar between genotypes, Lmna(+/-) animals had a significantly attenuated hypertrophic response to pressure overload as evidenced by reduced ventricular mass and myocyte size. Analysis of pressure overload-induced transcriptional changes suggested that the reduced hypertrophy in the Lmna(+/-) mice was accompanied by impaired activation of the mechanosensitive gene Egr-1. In conclusion, our findings provide further support for a critical role of lamins A and C in regulating the cellular response to mechanical stress in cardiomyocytes and demonstrate that haploinsufficiency of lamins A and C alone is sufficient to alter hypertrophic responses and cardiac function in the face of pressure overload in the heart.

活性汚泥と処理水の分離に精密濾過中空糸膜モジュールを用いた浸漬型膜分離活性汚泥法において，膜の細孔が目詰まりを起こしたり膜表面に汚泥が付着すると，濾過抵抗が増大して吸引圧が上昇し効率的な運転が出来なくなる．本研究では，有機物除去が良好に行える低いBOD容積負荷で膜透過流束一定の条件で，反応槽内の初期汚泥濃度（MLSS）をできるだけ広い範囲で変化させて排水処理実験を行い，MLSSが各種濾過抵抗に与える影響について検討した．その結果，MLSSが12000 mg/Lを超えると吸引圧が上昇する傾向がみられた．このとき，膜モジュールに付着する汚泥量が増加する．この付着汚泥量とケークによる濾過抵抗は比例し，基本的にRuthの濾過方程式に従うことが分かった．これらのことより，浸漬型膜分離活性汚泥法の装置を長期間安定して運転するためには，吸引圧の上昇を抑えることができるMLSSが12000 mg/L以下の条件で操作することが望ましいことが明らかとなった．

Background Ubiquitous deletion of thioredoxin reductase 2 (Txnrd2) in mice is embryonically lethal and associated with abnormal heart development, while constitutive, heart-specific Txnrd2 inactivation leads to dilated cardiomyopathy and perinatal death. The significance of Txnrd2 in aging cardiomyocytes, however, has not yet been examined. Methods and Results The tamoxifen-inducible heart-specific αMHC-MerCreMer transgene was used to inactivate loxP-flanked Txnrd2 alleles in adult mice. Hearts and isolated mitochondria from aged knockout mice were morphologically and functionally analyzed. Echocardiography revealed a significant increase in left ventricular end-systolic diameters in knockouts. Fractional shortening and ejection fraction were decreased compared with controls. Ultrastructural analysis of cardiomyocytes of aged mice showed mitochondrial degeneration and accumulation of autophagic bodies. A dysregulated autophagic activity was supported by higher levels of lysosome-associated membrane protein 1 (LAMP1), microtubule-associated protein 1A/1B-light chain 3-I (LC3-I), and p62 in knockout hearts. Isolated Txnrd2-deficient mitochondria used less oxygen and tended to produce more reactive oxygen species. Chronic hypoxia inducible factor 1, α subunit stabilization and altered transcriptional and metabolic signatures indicated that energy metabolism is deregulated. Conclusions These results imply a novel role of Txnrd2 in sustaining heart function during aging and suggest that Txnrd2 may be a modifier of heart failure.

A precise control of oxidation/reduction of protein thiols is essential for intact cardiac physiology. Irreversible oxidative modifications have been proposed to play a role in the pathogenesis of cardiovascular diseases. An imbalance of redox homeostasis with diminution of antioxidant capacities predisposes the heart to oxidant injury. There is growing interest in endoplasmic reticulum (ER) stress in the cardiovascular field, since perturbation of redox homeostasis in the ER is sufficient to cause ER stress. Because a number of human diseases are related to altered redox homeostasis and defects in protein folding, many research efforts have been devoted in recent years to understanding the structure and enzymatic properties of the thioredoxin superfamily. The thioredoxin superfamily has been well documented as thiol oxidoreductases to exert a role in various cell signaling pathways. The redox properties of the thioredoxin motif account for the different functions of several members of the thioredoxin superfamily. While thioredoxin and glutaredoxin primarily act as antioxidants by reducing protein disulfides and mixed disulfide, another member of the superfamily, protein disulfide isomerase (PDI), can act as an oxidant by forming intrachain disulfide bonds that contribute to proper protein folding. Increasing evidence suggests a pivotal role of PDI in the survival pathway that promotes cardiomyocyte survival and leads to more favorable cardiac remodeling. Thus, the thiol redox state is important for cellular redox signaling and survival pathway in the heart. This review summarizes the key features of major members of the thioredoxin superfamily directly involved in cardiac physiology and pathology.

Objective— Cellular redox balance is regulated by enzymatic and nonenzymatic systems and freely diffusible nitric oxide (NO) promotes antioxidative mechanisms. We show the NO-dependent transcriptional regulation of the antioxidative thioredoxin system. Methods and Results— Incubation of rat pulmonary artery smooth muscle cells (RPaSMC) with the NO donor compound S -nitroso-glutathione (GSNO, 100 μmol/L) suppressed thioredoxin-interacting protein (Txnip), an inhibitor of thioredoxin function, by 71±18% and enhanced thioredoxin reductase 2.7±0.2 fold (n=6; both P P cis -regulatory elements between −1777 and −1127 bp upstream of the start codon. Hyperglycemia induced Txnip promoter activity (3.9±0.2-fold; P P Conclusions— NO can regulate cellular redox state by changing expression of Txnip and thioredoxin reductase. This represents a novel antioxidative mechanism of NO independent of posttranslational protein S -nitrosylation of thioredoxin.

The thioredoxin system helps maintain a reducing environment in cells, but thioredoxin functions as more than simply an antioxidant. Thioredoxin functions depend on the protein's redox state, as determined by two conserved cysteines. Key biologic activities of thioredoxin include antioxidant, growth control, and antiapoptotic properties, resulting from interaction with target molecules including transcription factors. Mechanisms by which thioredoxin regulates cell growth include binding to signaling molecules such as apoptosis signal-regulating kinase-1 (ASK-1) and thioredoxin-interacting protein (Txnip). The molecular interplay between thioredoxin, ASK-1, and Txnip potentially influences cell growth and survival in diverse human diseases such as cancer, diabetes, and heart disease. In this review, we focus on the structure of thioredoxin and its functional regulation of cell growth through the interactions with signaling molecules.
