Objective: To study the effect of different triggering thresholds on the quality of pulmonary artery CT angiography (CTA) images. Materials and Methods: A prospective study included 112 patients with suspected pulmonary embolism admitted to Tinglin Hospital in the Jinshan District of Shanghai between December 2021 to April 2023. Among them, there were 49 males and 63 females aged between 37 and 93 years, with an average age of 64.28 years. Patients were randomly assigned to three groups based on trigger thresholds. Group A included 38 cases with a trigger threshold of 120 HU, Group B included 37 cases with a trigger threshold of 200 HU, and Group C included 37 cases with a trigger threshold of 250 HU. There were no statistically significant differences in gender, age, height, or weight among the three groups. One-way ANOVA was used to compare the CT values and subjective image quality scores of the superior vena cava, main pulmonary artery, left and right pulmonary arteries, and right pulmonary vein among the three groups. Result: There were no statistical differences in the CT values of the main pulmonary artery and left and right pulmonary arteries among the three groups, but there were statistical differences in the CT values of the superior vena cava and right pulmonary vein. There was a statistical difference in the subjective score of image quality among the three groups; the subjective evaluation of the obtained image quality between the two physicians was highly consistent (κ=0.78). Conclusion: When the triggering threshold of pulmonary artery CTA is 200 HU, it can not only ensure the concentration of pulmonary artery trunk CT value meets the clinical diagnosis, but also ensures that the contrast agent is fully injected into the 5~6 grade branches, leading to less retention of superior vena cava, weak pulmonary vein development, and the highest image quality of pulmonary artery CTA.

Abstract Retrogressive thaw slumps (RTSs), which frequently occur in permafrost regions of the Qinghai-Tibet Plateau (QTP), China, can cause significant damage to the local surface, resulting in material losses and posing a threat to infrastructure and ecosystems in the region. However, quantitative assessment of ground ice ablation and hydrological ecosystem response was limited due to a lack of understanding of the complex hydro-thermal process during RTS development. In this study, we developed a three-dimensional hydro-thermal coupled numerical model of a RTS in the permafrost terrain at the Beilu River Basin of the QTP, including ice–water phase transitions, heat exchange, mass transport, and the parameterized exchange of heat between the active layer and air. Based on the calibrated hydro-thermal model and combined with the electrical resistivity tomography survey and sample analysis results, a method for estimating the melting of ground ice was proposed. Simulation results indicate that the model effectively reflects the factual hydro-thermal regime of the RTS and can evaluate the ground ice ablation and total suspended sediment variation, represented by turbidity. Between 2011 and 2021, the maximum simulated ground ice ablation was in 2016 within the slump region, amounting to a total of 492 m3, and it induced the reciprocal evolution, especially in the headwall of the RTS. High ponding depression water turbidity values of 28 and 49 occurred in the thawing season in 2021. The simulated ground ice ablation and turbidity events were highly correlated with climatic warming and wetting. The results offer a valuable approach to assessing the effects of RTS on infrastructure and the environment, especially in the context of a changing climate.

8-17 DNAzymes (8-17, 17E, Mg5, and 17EV1) are in vitro-selected catalytic DNA molecules that are capable of cleaving complementary RNAs. The conserved residues in their similar catalytic cores, together with the metal ions, were suggested to contribute to the catalytic reaction. Based on the contribution of the less conserved residues in the bulge loop residues (W12, A15, A15.0) and the internal stem, new catalytic cores of 8-17 DNAzymes were programmed. The internal stem CTC-GAG seems to be more favorable for the DNAzymes than CCG-GGC, while an extra W12.0 led to a significant loss of activity of DNAzymes, which is contrary to the positive effect of A15.0, by which a new active DNAzyme 17EM was derived. It conducts a faster reaction than 17E. It is most active in the presence of Pb2+, with the metal ion preference of Pb2+ >> Zn2+ > Mn2+ > Ca2+ ≈ Mg2+. In the Pb2+ and Zn2+-mediated reactions of 17EM and 17E, the same Na+- and pH dependence were also observed as what was observed for 17E and other 8-17 DNAzymes. Therefore, 17EM is another member of the 8-17 DNAzymes, and it could be applied as a potential biosensor for RNA and metal ions.

Innovative production in high-tech industries is seen as a promoter of corporate profitability and a driver of China's economic growth. However, some scholars point out that high-tech industry is in its infancy and has insufficient innovative production efficiency, which severely restricts regional economic development. To explore this further, we studied the innovation production efficiency of China's high-tech industry during the 13th Five-Year Plan period (2016-2020). The three-stage Data Envelopment Analysis model was utilized to calculate the efficiency of the innovation production in this industry, and we initially employed the DEA-BCC model to calculate the efficiency for 31 provinces and applied similar-stochastic frontier analysis regression to eliminate the potential influence of external environmental factors. The empirical results findings reveal significant inter-regional differences in the efficiency of innovation production, with the Eastern region is the most efficient in innovation production, the Western region has greater growth potential, and the Central region requires to improve its overall efficiency by increasing technological inputs. In addition, we attempt to provide recommendations to policy makers based on our conclusions.

Abstract Background The biological effects of cerium dioxide nanoparticles (CeO2NPs), a novel material in the biomedical field, have attracted widespread attention. Our previous study confirmed that exposure to CeO2NPs during pregnancy led to abnormal trophoblast invasion during early placental development, thereby impairing placental development. The potential mechanisms may be related to low-quality decidualization triggered by CeO2NPs exposure, such as an imbalance in trophoblast invasion regulators secreted by decidual cells. However, the intermediate link mediating the “dialogue” between decidual cells and trophoblasts during this process remains unclear. As an important connection between cells, exosomes participate in the “dialogue” between endometrial cells and trophoblasts. Exosomes transfer bioactive microRNA into target cells, which can target and regulate the level of mRNA in target cells. Results Here, we constructed a mice primary uterine stromal cell-induced decidualization model in vitro, and detected the effect of CeO2NPs exposure on the expression of decidual-derived exosomal miRNAs by high-throughput sequencing. Bioinformatics analysis and dual-luciferase reporter assays were performed to identify target genes of the screened key miRNAs in regulating trophoblast invasion. Finally, the role of the screened miRNAs and their target genes in regulating trophoblast (HTR-8/SVneo cells) invasion was confirmed. The results showed that CeO2NPs exposure inhibited trophoblast invasion by promoting miR-99a-5p expression in decidual-derived exosomes, and Ppp2r5a is a potential target gene for miR-99a-5p to inhibit trophoblast invasion. Conclusions This study revealed the molecular mechanism by which CeO2NPs exposure inhibits trophoblast invasion from the perspective of decidual derived exosomal miRNAs. These results will provide an experimental basis for screening potential therapeutic targets for the negative biological effects of CeO2NPs exposure and new ideas for studying the mechanism of damage to trophoblast cells at the decidual-foetal interface by harmful environmental or occupational factors.

Abstract Background Trauma in the elderly is gradually growing more prevalent as the aging population increases over time. The purpose of this study is to assess hospitalization costs of the elderly trauma population and analyze the association between those costs and the features of the elderly trauma population. Methods In a retrospective analysis, data on trauma patients over 65 who were admitted to the hospital for the first time due to trauma between January 2017 and March 2022 was collected from a tertiary comprehensive hospital in Baotou. We calculated and analyzed the hospitalization cost components. According to various therapeutic approaches, trauma patients were divided into two subgroups: non-surgical patients (1320 cases) and surgical patients (387 cases). Quantile regression was used to evaluate the relationship between trauma patients and hospitalization costs. Results This study comprised 1707 trauma patients in total. Mean total hospitalization costs per patient were ¥20,741. Patients with transportation accidents incurred the highest expenditures among those with external causes of trauma, with a mean hospitalization cost of ¥24,918, followed by patients with falls at ¥19,809 on average. Hospitalization costs were dominated by medicine costs (¥7,182 per capita). According to the quantile regression results, all trauma patients' hospitalization costs were considerably increased by length of stay, surgery, the injury severity score (16–24), multimorbidity, thorax injury, and blood transfusion. For non-surgical patients, length of stay, multimorbidity, and the injury severity score (16–24) were all substantially linked to higher hospitalization costs. For surgical patients, length of stay, injury severity score (16–24), and hip and thigh injuries were significantly associated with greater hospitalization costs. Conclusions Using quantile regression to identify factors associated with hospitalization costs could be helpful for addressing the burden of injury in the elderly population. Policymakers may find these findings to be insightful in lowering hospitalization costs related to injury in the elderly population.

Objective: To explore the influence on the image quality of head and neck CTA under different tube voltages. Materials and methods: A total of 67 patients with clinically suspected head and neck vascular diseases in Tinglin hospital from September 2021 to January 2022 were collected as the research objects, including 37 males and 30 females, with an average age of 68.94. According to the applied tube voltage, the patients were randomly assigned into groups A, B and C. There were 23 cases of 120 kV in group A, 24 cases of 100 kV in group B and 20 cases of 80 kV in group C. Single factor ANOVA test was used to compare the CT value, SD value, SNR, CNR of aortic arch, internal carotid artery, M1 segment of middle cerebral artery and sternocleidomastoid muscle among the three groups, and furthermore the subjective score of image quality, TDLP and ED. Kappa test was used to evaluate the consistency of subjective scores of the two physicians. Results: There were statistical differences in CT and SD values of aortic arch, internal carotid artery and M1 segment of middle cerebral artery among the three groups; The CT and SD values of aortic arch, internal carotid artery and M1 segment of middle cerebral artery increased with the decrease of tube voltage, with an increase range of 15%～55%. There was no statistical difference in SNR and CNR among the three groups; There was statistical difference in subjective score of image quality among the three groups; Group B > group A > group C, and group B showed the highest score; the kappa value of the subjective score of the two phsicianss=0. 80, which was highly consistent; There were statistical differences in TDLP and ED among the three groups, TDLP and ED decreased with the decrease of tube voltage, with the decrease ranges of 25% and 39%. Conclusion: According to the requirements of CT radiation dose diagnosis reference level, 100 kV is the best tube voltage condition for head and neck CTA.

Abstract Background The increasing use of cerium dioxide nanoparticles (CeO2NPs) in biomedical field has attracted substantial attention about their potential risks to human health. Recent studies have shown that nanoparticles can induce placental dysfunction and even fetal abortion, but a more detailed mechanism of nanoparticles affecting placental development remains elusive. Results Here, we constructed a mouse exposure model with different doses of CeO2NPs (2.5, 4, 5, 7.5, and 10 mg kg−1 day−1, average particle size 3–5 nm), finding that intravenous exposure to pregnant mice with CeO2NPs could cause abnormal placental development. Deposited nanoparticles were able to be observed in the placental trophoblast at doses of 5 and 7.5 mg kg−1 day−1. Diving into molecular mechanisms indicated that CeO2NPs exposure could lead to autophagy activation in placental trophoblast. At the cellular level, exposure to CeO2NPs inhibited the migration and invasion of HTR-8/SVneo and activated the autophagy through mammalian target of rapamycin complex1 (mTORC1) signaling pathway. Furthermore, inhibition of autophagy initiation by 3-Methyladenine (3-MA) partially restored the function of HTR-8/SVneo, while blocking autophagic flow by Chloroquine (CQ) aggravated the functional damage. Conclusions Maternal exposure to CeO2NPs impairs placental development through trophoblast dysfunction mediated by excessive autophagy activation. These results suggested that autophagy dysfunction may be a potential mechanism for the impairment of trophoblast by CeO2NPs exposure. As above, our findings provide insights into the toxicity mechanism to the reproductive system induced by rare-earth nanoparticles exposure. Graphical Abstract

The environmental pollutant Benzo(a)pyrene (BaP) has an adverse effect on the reproductive performance of mammals. We previously showed that BaP treatment during early pregnancy damages endometrial morphology and impairs embryo implantation. Endometrial decidualization at the implantation site (IS) after embryo implantation is crucial for pregnancy maintenance and placental development. The balance between proliferation and differentiation in endometrial stromal cells (ESCs) is a crucial event of decidualization, which is regulated by the cell cycle. Here, we report that abnormal decidualization caused by BaP is associated with cell cycle disturbance of stromal cells. The mice in the treatment group were gavaged with 0.2 mg/kg/day BaP from day 1–8 of pregnancy, while those in control were gavaged with corn oil in parallel. BaP damaged the decidualization of ESCs and reduced the number of polyploid cells. Meanwhile, BaP up-regulated the expression of Ki67 and PCNA, affecting the differentiation of stromal cells. The cell cycle progression analysis during decidualization in vivo and in vitro showed that BaP induced polyploid cells deficiency with enhanced expressions of CyclinA(E)/CDK2, CyclinD/CDK4 and CyclinB/CDK1, which promote the transformation of cells from G1 to S phase and simultaneously activate the G2/M phase. The above results indicated that BaP exposure accelerates cell cycle progression, promotes ESC proliferation, inhibits differentiation, and impedes proper decidualization and polyploidy development. Thus, the imbalance of ESC proliferation and differentiation would be an important mechanism for BaP-induced defective decidualization.

Background and Aim The relationship between the oral microbiota and type 1 diabetes (T1D) remains unclear. We aimed to evaluate the variations in the oral microbiome in T1D and identify potentially associated bacterial factors.Methods We performed high-throughput sequencing of the V3-V4 area of the 16S rRNA gene to profile the oral bacterial composition of 47 healthy children (CON group), 46 children with new-onset T1D in the acute phase (NT1D group), and 10 children with T1D in the chronic phase receiving insulin treatment (CT1D group). Multivariate statistical analysis of sequencing data was performed.Results Compared to the CON group, the NT1D group was characterized by decreased diversity and increased abundance of genera harboring opportunistic pathogens, while this trend was partially reversed in the CT1D group. Differential genera between groups could distinguish the NT1D group from the CON group (AUC = 0.933) and CT1D group (AUC = 0.846), respectively. Moreover, T1D-enriched genera were closely correlated with HbA1c, FBG and WBCs levels.Conclusion Our results showed that the acute phase of T1D was characterized by oral microbiota dysbiosis, which could be partially ameliorated via glycemic control. The possible role of oral microbiota dysbiosis on oral health and systemic metabolic status in T1D warrants further mechanistic investigation.

Numerous studies have suggested the possibility of explaining the etiology of metabolic syndrome through DNA methylation. DNA methyltransferase 3B (DNMT3B) plays an important role in de novo DNA methylation. There was an alteration in maternal (F0) endometrial function, which might lead to growth and developmental disorder in offspring (F1). In this study, we investigated the effect of maternal endometrial DNMT3B deficiency on the metabolism in offspring. We constructed endometrial DNMT3B conditional knockout female mice (cKO) which were mated with normal C57BL/6 male mice to obtain the F1 generation. Further, to study the development of these offspring, we observed them at three different life stages which included the 6-week-old juvenile, 9-week-old sub-adult and 12-week-old adult. Follow the detection of a range of metabolism-related indicators, we found that in the cKO F1 generation, liver triglyceride level was significantly elevated in 9-week-old female mice, lipid droplet deposition was significantly increased in 9-week-old and 12-week-old mice, and the expression of lipid metabolism key factors in the liver was markedly decreased except of 6-week-old male mice. These results indicate that maternal endometrial DNMT3B conditional knockout leads to imbalance in hepatic metabolism in F1 generation, the mechanism of which requires further discussion.

ABSTRACT An increasing amount of sunlight is available to sunken greenhouses located in northern China, but it remains unclear how the indoor temperature changes at different heights and at the north and south ends of the structure. To study how the temperature in a single indoor section, at multiple points, varies in a sunken solar greenhouse, 10 measurement lines were arranged in a section 30 m from the west wall. Each line was equipped with a corresponding WT-59 wireless sensor. Temperature and humidity data were collected from 8 March 2019 to 26 December 2019. The following results were obtained. (1) The indoor temperature variation in a day could be divided into four stages: from 0:00 to 7:30 h, the temperature at all measurement points decreased linearly; from 7:30 to 14:30 h, the temperature at all measurement points increased curvilinearly; from 14:30 to 18:00, the temperature at all measurement points decreased curvilinearly; and from 18:30 to 24:00, the temperature at all measurements points decreased linearly. (2) The maximum temperature on sunny days in the greenhouse was higher than that on cloudy days and snowy days, but the maximum temperature on cloudy days and snowy days in the greenhouse occurred 3 h earlier than on sunny days. (3) The lowest average temperature of a sunken solar greenhouse over 3–6 months was 7.5ଌ, without heating. The research results provide theoretical and reference data on how the temperature changes in the interior of a solar greenhouse and inform about the three-dimensional planting height and ventilation times.

Cationic polymeric materials and cell-penetrating peptides (CPPs) were often used as the delivery vectors in the evaluation of nucleic acid therapeutics. 10-23 DNAzyme is a kind of potential antisense therapeutics by catalytic cleavage of the disease-related RNAs. Here, lipofectamine 2000 and Tat peptide were evaluated for their effect on the catalytic activity of 10-23 DNAzyme, with the observed rate constant, thermal stability, CD spectra, and PAGE analysis, with a duplex DNA mimicking DNAzyme-substrate as a control. It was shown that the cationic carriers had a negative effect on the catalytic performance of the 10-23 DNAzyme. Significantly, the destabilizing effect of the cationic carriers on the duplex formation was noteworthy, as a duplex formation is an essential prerequisite in the silencing mechanisms of antisense and RNAi.

AimRecurrent miscarriage (RM) is associated with numerous clinical factors. However, some RM occurred without specific factors. It has been revealed that some molecules such as hormones, miRNAs, and transcription factors are involved in RM by regulating proliferation, apoptosis, etc. However, the mechanism of RM has yet to be identified clearly. Circular RNAs (circRNAs) are a class of endogenous non-coding RNAs that often act as sponges for miRNAs or binds to proteins involved in biological processes. However, the functional role of circRNAs in the uterine decidua of patients with early RM is still unclear. In this study, we aimed to investigate the mechanisms of circ-CYP24A1 in RM.MethodsThe Dual-Luciferase Activity Assay was designed to analyze the bonding between circ-CYP24A1 and miR-224, and miR-224 and prolactin receptor (PRLR) mRNA 3′UTR. In situ hybridization (ISH) and immunohistochemistry (IHC) were used to observe the expression of circ-CYP24A1 and PRLR in the decidua. Rescue experiments were performed to investigate the regulating effects of circ-CYP24A1, miR-224, and PRLR. Western blotting was conducted to test the expression level of PRLR. The proliferation and apoptosis-related markers in Ishikawa cells were analyzed using CCK8, immunofluorescence staining, and the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) assay.ResultsIn this study, based on the microarray analysis data, we identified a high level of circ-CYP24A1 and PRLR in the decidua of patients with early RM. Based on the bioinformatics prediction, the binding relationship between circ-CYP24A1 and miR-224, as well as miR-224 and PRLR, were verified. Functional experiments demonstrated that circ-CYP24A1 regulated proliferation and apoptosis by binding to and inhibiting miR-224, resulting in increased PRLR expression. Taken together, this study provides new insights into the mechanism of RM.ConclusionIn this study, we found that circ-CYP24A1 plays a role in RM by impairing the balance of cell proliferation and apoptosis by sponging miR-224, thereby regulating PRLR.

Circular RNAs (circRNAs) are a novel class of endogenous noncoding RNAs that play important roles in gene expression regulation. This study aimed to evaluate the potential role of circRNAs in decidual tissue of patients with early recurrent miscarriage (RM). We constructed circRNA expression profiles in decidual tissue using microarray data. Exposure to ethephon (ETH), a plant growth regulator commonly used for several purposes, can potentially decrease sperm numbers and viability. Occasional findings regarding ETH effects on female reproduction during early pregnancy have also been reported. During early pregnancy, endometrial decidualization is a critical event for embryo implantation and pregnancy maintenance. Thus, we aimed to explore the effect and mechanism of ETH on endometrial decidualization both in vivo and in vitro. Mice were gavaged with 0 and 285 mg/kg b.w. ETH from gestational days (GD)1 until sacrifice, whereas pseudopregnant mice from pseudopregnant day 1 (PPD-1) until PPD-8. Primary mouse endometrial stromal cells (mESCs) received 640 ug/ml ETH and added E2 and P4 to induce decidualization. Results indicated female albino CD1 mice exposed to high dose of ETH (285 mg/kg b.w.) by oral gavage, the number of embryo implantation sites on GD6 and GD8 were significantly decreased, the levels of serum E2 and P4 on GD8 were significantly decreased. Compared with the control group, the decidualization response artificially induced by corn oil in pseudopregnant mice and by E2 and P4 in primary mouse endometrial stromal cells (mESCs) was weakened in the high dose of ETH treated group. The high dose, 285 mg/kg b.w ETH treated group altered the expression of endometrial decidual markers on GD6 and GD8. The triglyceride and fatty acid metabolism-related genes were significantly increased after female albino CD1 mice exposed to high does, 285 mg/kg b.w ETH on GD6 and GD8. GPR120 was substantially reduced after ETH treatment. When overexpression of GPR120, the compromised decidualization induced by ETH treatment was rescued. Furthermore, molecular docking presented Thr234 and His251 of GPR120 as preferred binding sites for ETH. Mutation of these two sites rescued the compromised decidualization induced by ETH. In conclusion, we demonstrated that ETH exposure could impair decidualization during early pregnancy. GPR120 expression and binding between GPR120 and ETH are crucial for impaired decidualization mediated via ETH.

Background: The role of sodium bicarbonate therapy (SBT) remains controversial. This study aimed to investigate whether hemodynamic status before SBT contributed to the heterogeneous outcomes associated with SBT in acute critically ill patients.Methods: We obtained data from patients with metabolic acidosis from the Medical Information Mart for Intensive Care (MIMIC)-III database. Propensity score matching (PSM) was applied to match the SBT group with the control group. Logistic regression and Cox regression were used to analyze a composite of newly “developed or exacerbated organ dysfunction” (d/eOD) within 7 days of ICU admission and 28-day mortality associated with SBT for metabolic acidosis.Results: A total of 1,765 patients with metabolic acidosis were enrolled, and 332 pairs obtained by PSM were applied to the final analyses in the study. An increased incidence of newly d/eOD was observed in the SB group compared with the control group (54.8 vs. 44.6%, p < 0.01). Multivariable logistic regression indicated that the adjusted OR of SBT for this composite outcome was no longer significant [OR (95% CI): 1.39 (0.9, 1.85); p = 0.164]. This effect of SBT did not change with the quintiles stratified by pH. Interestingly, SBT was associated with an increased risk of the composite of newly d/eOD in the subgroup of patients with worsening hemodynamics before SBT [adjusted OR (95% CI): 3.6 (1.84, 7.22), p < 0.001]. Moreover, the risk potential for this composite of outcomes was significantly increased in patients characterized by both worsening [adjusted OR (95% CI): 2.91 (1.54, 5.47), p < 0.001] and unchanged hemodynamics [adjusted OR (95% CI): 1.94 (1.01, 3.72), p = 0.046] compared to patients with improved hemodynamics before SBT. Our study failed to demonstrate an association between SBT and 28-day mortality in acute critically ill patients with metabolic acidosis.Conclusions: Our findings did not demonstrate an association between SBT and outcomes in critically ill patients with metabolic acidosis. However, patients with either worsening or unchanged hemodynamic status in initial resuscitation had a significantly higher risk potential of newly d/eOD subsequent to SBT.

The association between the accumulation of synthetic chemicals with estrogenic activity and risks to oogenesis has become a growing concern. This study indicates that in utero estrogen exposure can affect homologous recombination in early oogenesis and influence the reproductive potential and lifespan of female offspring. We conducted this study in developing mouse ovaries using two different models: oral doses administered to the mother, and fetal ovary cultures. Our analyses of meiotic fetal oocytes suggest that 17-β-estradiol induces gross aberrations in prophase I events, including delayed meiotic progression, increased unrepaired DNA damage, and altered homologous recombination levels. These effects were mainly mediated by estrogen receptor 2 (ESR2) activation. Mid-gestation exposure to estrogen also led to delayed primordial folliculogenesis after birth, impaired follicle development after prepuberty, and ultimately reduced the total litter size of the offspring. This raises the concern that maternal exposures to substances activating ESR2 may compromise the fertility of the exposed female fetus.

Sparganosis is a zoonotic parasitic disease caused by the larvae (spargana) of the genus Spirometra, which is widely distributed globally and threatens human health. More than 60 species of Spirometra have already been identified, and over 2000 cases have been reported. This review summarizes the prevalence of humans, frogs, snakes, and other animals with spargana. Furthermore, the infection mode, distribution, and site are summarized and analyzed. We also describe the epidemiology, molecular diagnosis, and other aspects which are of considerable significance to preventing sparganum.

Polymer materials, such as extruded polystyrene (XPS), polyurethane (PU) and polystyrene (EPS), are extensively used as thermal insulators in cold regions to protect the engineering from frost damage. However, the performance degradation caused by freeze-thaw cycles (FTC) of polymer material has not been fully quantified. To fill this gap, a series of experiments were carried out by taking XPS board as the test material. After the XPS board was subjected to hundreds of FTC, the volumetric water absorption, thermal conductivity, uniaxial compressive strength and Young's modulus as well as creep strain were measured and analyzed. The results demonstrate that the volumetric water absorption and thermal conductivity change slightly with the FTC. After 400 times of FTC, the volumetric water absorption increased within only 0.1% and the thermal conductivity decreased within 9 × 10−4 W⋅(m⋅K)−1. Additionally, the uniaxial compressive strength and Young's modulus decrease with the increasing of FTC, and the effect of FTC on Young's modulus is mainly concentrated in the first 50 times. The creep behavior of XPS board belongs to decay creep and the exponential function can describe the creep process well. The results obtained in this study may help us to understand the degradation of XPS board and provide references for the design of controlling engineering damages caused by FTC in cold regions.

Sparganosis, caused by the plerocercoid larvae of Spirometra tapeworms, is a public health hazard worldwide. The prevalence and genetics of sparganum from snakes remain unclear. In this study, we investigated the prevalence of sparganum infection in wild snakes in Hunan province and compared the prevalence of Spirometra tapeworms in snakes worldwide. Furthermore, the genetic diversity of collected isolates was analyzed using mitochondrial cytb and cox1 genes. The result shows that the sparganum infection rate in wild snakes (89.50%, 402/449) was higher in Hunan than in other regions. Genetic diversity analysis based on concatenated sequences revealed high genetic diversity but no distinct genetic structure among Spirometra populations. Phylogenetic analysis supported the division of European and Chinese Spirometra isolates and a single species in Chinese Spirometra isolates. The prevalence of Spirometra tapeworms in snakes is serious, and the risk of sparganosis should be further publicized.

Benzo(a)pyrene (B(a)P) is a widespread persistent organic pollutant (POP) and a well-known endocrine disruptor. Exposure to BaP is known to disrupt the steroid balance and impair embryo implantation, but the mechanism under it remains unclear. The corpus luteum (CL), the primary source of progesterone during early pregnancy, plays a pivotal role in embryo implantation and pregnancy maintenance. The inappropriate luteal function may result in implantation failure and spontaneous abortions. Therefore, this study was conducted to assess the effects and potential mechanisms of B(a)P on the CL function. Our results showed that pregnant mice received B(a)P displayed impaired embryo implantation and dysfunction of ovarian CL. The estrogen and progesterone levels decreased by B(a)P. In vitro, exposure to BPDE, which is the metabolite of B(a)P, affected the luteinization of granular cell KK-1. Additionally, melatonin and its receptors, which are important for ovarian function and anti-oxidative damage, were affected by B(a)P or BPDE. B(a)P or BPDE-treated alone impaired antioxidant capacity of ovarian granulosa cells, caused an increasing of ROS and cell apoptosis, and disrupted the PI3K/AKT/GSK3β signaling pathway in vivo and in vitro. Co-treatment with melatonin alleviated B(a)P or BPDE-induced CL dysfunction by ameliorating oxidative stress, counteracting phosphorylation of PI3K/AKT/GSK3β signaling pathway, decreasing the apoptosis of the ovarian cells. Moreover, activation of the melatonin receptor by ramelteon in KK-1 cells exhibits an analogous protective effect as melatonin. In conclusion, our findings not only firstly clarify the potential mechanisms of BaP-induced CL dysfunction, but also extend the understanding about the ovarian protection of melatonin and its receptors against B(a)P exposure.

Background: Cervical lymph node (LN) metastasis of papillary thyroid carcinoma (PTC) is critical for treatment and prognosis. To examine the feasibility of MRI radiomics to preoperatively predict cervical LN metastasis in patients with PTC. Methods: Between January 2015 and March 2018, a total of 61 patients with pathologically confirmed PTC were analyzed retrospectively. The patients were divided into cervical LN metastasis group (n = 37) and no cervical LN metastasis (n = 24). T2WI and T2WI-fat-suppression (T2WI-FS) images were collected. A number of radiomic features were automatically extracted from the largest section of tumor. Three types of classifier (the random forests, the support vector machine classifier and the generalized linear model) based on T2WI and T2WI-FS images of cervical LN metastasis and no cervical LN metastasis were constructed and evaluated with a nested cross-validation scheme. Results: Radiomic features extracted from T2WI images were more discriminative than T2WI-FS images. The random forests model showed the best discriminate performance with the highest area under the curve (0.85, CI:0.76 -1), accuracy (0.87), sensitivity (0.83), specificity (1.00), positive predictive value (PPV = 1.00) and negative predictive value (NPV = 0.88). Conclusion: MRI radiomics analysis based on conventional T2WI and T2WI-FS can predict cervical LN metastasis in patients with PTC, and the radiomics is shown to be an assistant diagnosis tool for radiologists.

Objective: To investigate whether diffusion-weighted imaging (DWI) with multi b values can be used as a quantitative assessment tool to predict central lymph node metastasis (CLNM) in papillary thyroid carcinoma (PTC).Method: A total of 214 PTC patients were enrolled from January 2015 to April 2018. Each patient underwent multi b value DWI (300, 500, and 800 s/mm2) preoperatively and then clinical treatment of central LN dissection at the Thyroid Surgery Department. These patients were divided as two groups based on with and without CLNM. The corresponding apparent diffusion coefficients (ADCs) were evaluated with separated b value, i.e., 300, 500, or 800 s/mm2. Clinicopathological variables and ADC values were analyzed retrospectively by using univariate and binary logistic regression. The corresponding obtained variables with statistical significance were further applied to create a nomogram in which the bootstrap resampling method was used for correction.Results: PTCs with CLNM had significantly lower ADC300, ADC500, and ADC800 values compared with PTCs without CLNM. Using receiver operating characteristic (ROC) analysis, the ADC500 value (0.817) showed a higher area under the curve (AUC) than those of the ADC300 and ADC800 values (0.610 and 0.641, respectively) in differentiating patients with CLNM and without CLNM. The corresponding cutoff value for ADC500 was also determined (1.444 × 10−3 mm2/s), with respective sensitivity and specificity of 88.6 and 66%. The nomogram was generated by binary logistic regression results, incorporating four variables: gender, primary tumor size, extrathyroidal extension (ETE), and ADC500 value. The AUC of the nomogram was 0.894 in predicting CLNM. Moreover, as shown in the calibration curve between nomogram and clinical findings, a strong agreement was observed in the prediction of CLNM.Conclusions: In summary, the ADC value is a valuable noninvasive imaging biomarker for evaluating CLNM in PTCs. The nomogram, as a clinical predictive model, is able to provide an effective evaluation of CLNM risk in PTC patients preoperatively.

Paris polyphylla. is a traditional medicinal herb that has long been used to prevent cancer in many Asian countries. Polyphyllin I (PPI), an important bioactive constituent of Paris polyphylla, has been found to exhibit a wide variety of anticancer activities in many types of cancer cells. However, the effects of PPI on human gastric carcinoma cells and its mechanism of action remain unclear. In this study, we examined the effective anti-gastric carcinoma activity of PPI and its underlying mechanism of action in HGC-27 cells. In vitro, sub-micromolar concentrations of PPI inhibited HGC-27 cell proliferation with an IC50 of 0.34 ± 0.06 μM after a 72-h treatment. In vivo, 3 mg/kg PPI significantly inhibited proliferation of HGC-27 tumor cells, with a 78.8% inhibition rate compared to paclitaxel, and demonstrated higher safety. Analysis of MDC and mGFP-LC3 fluorescence, Western blotting and flow cytometry indicated that PPI induced cell cycle arrest in HGC-27 cells by promoting the conversion of LC3-I to LC3-II and by downregulating cyclin B1. Furthermore, Western blotting showed that PPI inhibited the autophagy-regulating PDK1/Akt/mTOR signaling pathway in vitro and in vivo. In addition, immunohistochemistry and TUNEL staining revealed that PPI decreased Ki67 expression and increased the percentage of apoptotic cells in HGC-27 xenograft tumors. These data indicate that PPI is an PDK1/Akt/mTOR signaling inhibitor and of therapeutic relevance for gastric cancer treatment and that the rhizome of Paris polyphylla deserves further clinical investigation as an alternative therapy for gastric cancer.

Farm marketing has been recognized as an important factor for a successful farm business. Due to the increasing interest of consumers in food safety, direct marketing of farm products to consumers in the local farmers' markets has become very popular. Compared to traditional farm marketing channels, relatively little is known about the use of direct-to-consumer marketing strategies by farmers. This paper aims to provide a more comprehensive picture of farmers' choices among available farm marketing channels using the case study of Taiwan. Using a population-based survey of 5600 family farms in Taiwan in 2014, in this study we quantify the extent to which demographic characteristics of farm operators, farm production and family conditions may influence the decision of farms to sell farm products to the government, wholesale markets, and in direct-to-consumer sales. We develop a trivariate probit model, and our results indicate that education level and engagement in the off-farm labour market of farm operators, the number of household members, farm size, land ownership, and the type of farm are the key factors in determining farmers' choice of marketing channels. The findings of this study may have important implications for the design of more effective farm marketing programs by agricultural authorities.

Novel α-aminoamide derivatives containing different benzoheterocyclics moiety were synthesized and evaluated as voltage-gated sodium ion channels blocks the treatment of pain. Compounds 6a, 6e, and 6f containing the benzofuran group displayed more potent in vivo analgesic activity than ralfinamide in both the formalin test and the writhing assay. Interestingly, they also exhibited potent in vitro anti-Nav1.7 and anti-Nav1.8 activity in the patch-clamp electrophysiology assay. Therefore, compounds 6a, 6e, and 6f, which have inhibitory potency for two pain-related Nav targets, could serve as new leads for the development of analgesic medicines.

The stress-strain relationship of frozen soil is a hot research topic in the field of frozen soil mechanics. In order to study the effect of particle crushing on the stress-strain relationship, a series of triaxial compression tests for frozen sandy soil are performed under confining pressures from 1 to 8 MPa at the temperatures of −3 and −5°C, and the energy consumption caused by particle breakage is analyzed during the triaxial shear process based on the energy principle. It is found that the energy consumption caused by the particle breakage presents a hyperbolic trend with axial strain. In view of the obvious advantages of the double yield surface elastoplastic model in describing soil dilatancy, stress path effect, and stress history influence, a modified double yield surface elastoplastic model for frozen sandy soil is proposed based on the energy principle. The validity of the model is verified by comparing its modeling results with test results. As a result, it is found that the stress-strain curves predicted by this model agree well with the corresponding experimental results under different confining pressures and temperatures.

The mechanism of birth defects induced by folate deficiency was focused on mainly in fetal development. Little is known about the effect of folate deficiency on the maternal uterus, especially on decidual angiogenesis after implantation which establishes vessel networks to support embryo development. The aim of this study was to investigate the effects of folate deficiency on decidual angiogenesis. Serum folate levels were measured by electrochemiluminescence. The status of decidual angiogenesis was examined by cluster designation 34 (CD34) immunohistochemistry and the expression of angiogenic factors, including vascular endothelial growth factor A (VEGFA), placental growth factor (PLGF), and VEGF receptor 2 (VEGFR2) were also tested. Serum levels of homocysteine (Hcy), follicle stimulating hormone (FSH), luteinizing hormone (LH), prolactin (PRL), progesterone (P4), and estradiol (E2) were detected by Enzyme-linked immunosorbent assay. The folate-deficient mice had a lower folate level and a higher Hcy level. Folate deficiency restrained decidual angiogenesis with significant abnormalities in vascular density and the enlargement and elongation of the vascular sinus. It also showed a reduction in the expressions of VEGFA, VEGFR2, and PLGF. In addition, the serum levels of P4, E2, LH, and PRL were reduced in folate-deficient mice, and the expression of progesterone receptor (PR) and estrogen receptor α (ERα) were abnormal. These results indicated that folate deficiency could impaire decidual angiogenesis and it may be related to the vasculotoxic properties of Hcy and the imbalance of the reproductive hormone.

Functionalization of the catalytic loop of 10–23 DNAzyme with an amino group was performed by incorporation of 7-(3-aminopropyl)-8-aza-7-deaza-2′-deoxyadenosine in different single positions. Among the nine modified positions in the catalytic loop, A9 is the unique position with positive contribution by such modification. These results indicated that more efficient deoxyribozymes remain to be explored by introduction of exogenous functional groups in an appropriate position in the catalytic loop of 10–23 DNAzyme, such as the combination of 7-functional group substituted 8-aza-7-deaza-2′-deoxyadenosine analogs and A9 position.

Members of the genera Prevotella, Veillonella and Fusobacterium are the predominant culturable obligate anaerobic bacteria isolated from periodontal abscesses. When determining the cumulative number of clinical anaerobic isolates from periodontal abscesses, ambiguous or overlapping signals were frequently encountered in 16S rRNA gene sequencing chromatograms, resulting in ambiguous identifications. With the exception of the genus Veillonella, the high intra-chromosomal heterogeneity of rrs genes has not been reported.The 16S rRNA genes of 138 clinical, strictly anaerobic isolates and one reference strain were directly sequenced, and the chromatograms were carefully examined. Gene cloning was performed for 22 typical isolates with doublet sequencing signals for the 16S rRNA genes, and four copies of the rrs-ITS genes of 9 Prevotella intermedia isolates were separately amplified by PCR, sequenced and compared. Five conserved housekeeping genes, hsp60, recA, dnaJ, gyrB1 and rpoB from 89 clinical isolates of Prevotella were also amplified by PCR and sequenced for identification and phylogenetic analysis along with 18 Prevotella reference strains.Heterogeneity of 16S rRNA genes was apparent in clinical, strictly anaerobic oral bacteria, particularly in the genera Prevotella and Veillonella. One hundred out of 138 anaerobic strains (72%) had intragenomic nucleotide polymorphisms (SNPs) in multiple locations, and 13 strains (9.4%) had intragenomic insertions or deletions in the 16S rRNA gene. In the genera Prevotella and Veillonella, 75% (67/89) and 100% (19/19) of the strains had SNPs in the 16S rRNA gene, respectively. Gene cloning and separate amplifications of four copies of the rrs-ITS genes confirmed that 2 to 4 heterogeneous 16S rRNA copies existed.Sequence alignment of five housekeeping genes revealed that intra-species nucleotide similarities were very high in the genera Prevotella, ranging from 94.3-100%. However, the inter-species similarities were relatively low, ranging from 68.7-97.9%. The housekeeping genes rpoB and gyrB1 were demonstrated to be alternative classification markers to the species level based on intra- and inter-species comparisons, whereas based on phylogenetic tree rpoB proved to be reliable phylogenetic marker for the genus Prevotella.

In the 15-mer catalytic core of 10-23 DNAzyme, each residue contributes to the catalytic conformation differently. Here, the critically conserved T4 and the least conserved T8 were modified on their 5-position with hydroxyl, imidazolyl, and amino groups with a hydrogen-bonding ability. These external functional groups induced new interactions within the catalytic core, resulting in both negative and positive effects on the catalytic activity of 10-23 DNAzyme, and the different linkages could be used to modulate the effect of the functional groups. The conservation of T4 and T8 could be recognized at the level of the nucleobase, but at the level of the functional group, T4 is not completely conserved. Their 5-methyl groups could be modified for a better performance in terms of the DNAzyme.

OBJECTIVE: Embryo implantation is directly affected by genes related to uterine receptivity. Studies have demonstrated the important roles of miRNAs in the regulation of gene expression. Our early miRNA chip analyses revealed that the mmu-miR-141 expression in endometrial tissue is lower after embryo implantation than before it. However, the possible roles of miR-141 in embryo implantation have not yet been elucidated. Here, mmu-miR-141 was designed to detect the expression and role of miR-141 in the endometria of mice in early pregnancy following embryo implantation. METHODS: Real-time PCR and in-situ hybridization were used to study mmu-miR-141 expression in mouse uterus. Cell proliferation was detected by tetrazolium dye (MTT) assay and flow cytometry. Real-time PCR and Western blot analysis were used to confirm the mRNA and protein levels of phosphatase and tensin homolog (PTEN) to determine whether it was the target gene of mmu-miR-141. Enhanced green fluorescent protein (EGFP) fluorescence reporter vector analysis was also performed. A functional study was performed by injecting mice uteri with mmu-miR-141 inhibitor or mimic vectors. RESULTS: mmu-miR-141 expression was lower on day 6 (D6) than day 4 (D4) and could be increased by progesterone. Reduced mmu-miR-141 could decrease the proliferation activity of stromal cells and promote apoptosis. Upregulation of mmu-miR-141 inhibited PTEN protein expression but downregulation of mmu-miR-141 increased it, while the mRNA level remained unchanged. EGFP fluorescence reporter vector analysis showed that miR-141 targets the 3'-untranslated region of the PTEN mRNA. In addition, when the physiological mmu-miR-141 level was altered on D2 by injecting with inhibitor or mimic, the embryo implantation sites were significantly decreased on D7. CONCLUSIONS: This study demonstrated that mmu-miR-141 might influence cell proliferation and apoptosis in the endometrium by negatively regulating PTEN expression, and could also influence the number of embryo implantation sites. mmu-miR-141 plays an essential role in embryo implantation.

This paper addresses the problem of the lack of intrinsic and contact parameters when applying the discrete element method to simulate and analyze the key aspects of white kidney bean seed sowing, harvesting and clearing. The experiment took white kidney bean seeds as the research object, measured the intrinsic parameters of white kidney bean seeds by using the universal testing machine, and measured the collision recovery coefficient of 0.445, static friction coefficient of 0.452 and rolling friction coefficient of 0.091 between white kidney bean seeds and Q235 steel plate by physical test and EDEM discrete element simulation parameter calibration. The stacking experiment was carried out by the cylinder lifting method, numerical processing was performed with MATLAB, and the actual stacking angle of white kidney bean seeds was 31.28°. The steepest climbing test was designed with the interspecific contact parameters as factors and the relative error between the actual and simulated stacking angles as indicators. The optimal combination of the interspecific contact parameters of white kidney bean was determined by response surface optimization analysis, and the interspecific collision recovery coefficient of white kidney bean was obtained as 0.39, static friction coefficient was 0.53, rolling friction coefficient was 0.092. Using the optimal parameters for the simulation test, the relative error between the actual stacking angle of white kidney bean seeds and the simulated stacking angle was 1.63%, indicating that the calibrated simulated contact parameters were reliable and could provide reference for the discrete element simulation of white kidney bean seeds. Cold regional tunnels have been encountering numerous frost damages as a result of dynamic changes in hydrothermal conditions of tunnel structural layers. The climate change is recognized as a major contributor for the problems. In this study, the hydrothermal conditions of a high speed railway tunnel are evaluated under different climate scenarios based on in-situ data and numerical analysis. Subsequently, the effect of different thicknesses of insulation board on hydrothermal conditions inside the tunnel is compared and the reasonable thickness is obtained. The main findings are: The temperature and unfrozen water content gradually decreased and the ice content gradually increased with the service time of the tunnel in the early 15 years; the maximum frozen depth occurs at the tunnel sidewall and it with a depth of 1.64 m within 30 years after the construction. The hydrothermal conditions inside the tunnel are significantly affected by annual mean air temperature (AMAT) and annual range of air temperature (ARAT); the maximum frozen depth decreases with AMAT, but increases with ARAT. After the XPS board with a thickness of 5 cm is laid in tunnel structural layers, the temperature at the most unfavorable position is 0.55 °C in the cold season, which suggests that frost damages disappeared and this method can be used to protect the tunnel against the frost damages.
