Plasma arc additive manufacturing (PAAM) was applied to fabricate TC4 and Ti48Al (gamma titanium aluminide) bimetals in this work. The impact of deposition sequence on the microstructure and mechanical properties of two groups of bimetals (TC4-Ti48Al, first deposited TC4, then Ti48Al; Ti48Al-TC4, first deposited Ti48Al, then TC4) was investigated. The results show that the ultimate tensile strengths of two groups of bimetals exhibit significant differences, with 158.4 MPa for TC4-Ti48Al and 232.3 MPa for Ti48Al-TC4, the reason for this disparity is the difference in microstructure at the transition zone, which is attributed to heat input and Marangoni convection. The phase transformation evolution of TC4-Ti48Al transition zone is (major α2 + minor α)→ (major α2 + minor γ) → (major γ + minor α2), and (major α2 + minor α) → (major α + minor α2) for Ti48Al-TC4. The formation mechanism of transition zones and relationship between the microstructure characteristics and mechanical properties of two groups of bimetals were further discussed. Overall, the property of Ti48Al-TC4 was superior, and this work provides a promising new method for the preparation of TC4 and Ti48Al bimetals.

IntroductionGlobal mining activities have significant impacts on ecosystems, but most studies have focused only on the relationship between soil physicochemical properties and microbial diversity in soils. The present study provides an insight into the effects of mining activities on soil physico-chemical properties and endophytic bacterial community composition in the rhizosphere of three different crops.MethodsMusa basjoo Siebold L., Amygdalus persica L., and Triticum aestivum L. were collected from the inter-root soils and plant roots to determine the soil physicochemical properties and endophytic bacterial communities in the root system.ResultsThe results showed that mining resulted in soil acidification, altered trace element content and increased organic carbon. Abstract Background Curcubita ficifolia Bouché (Cucurbitaceae) has high value as a food crop and medicinal plant, and also has horticultural value as rootstock for other melon species. China is home to many different cultivars, but the genetic diversity of these resources and the evolutionary relationships among them, as well as the differences between C. ficifolia and other Cucurbita species, remain unclear. Results We investigated the chloroplast (cp) genomes of 160 C. ficifolia individuals from 31 populations in Yunnan, a major C. ficifolia production area in China. We found that the cp genome of C. ficifolia is ~151 kb and contains 128 genes, of which 86 are protein coding genes, 34 encode tRNA, and eight encode rRNAs. We also identified 64 SSRs, mainly AT repeats. The cp genome was found to contain a total of 204 SNP and 57 indels, and a total of 21 haplotypes were found in the 160 study individuals. The reverse repeat (IR) region of C. ficifolia contained a few differences compared with this region in the six other Cucurbita species. Sequence difference analysis demonstrated that most of the variable regions were concentrated in the single copy (SC) region. Moreover, the sequences of the coding regions were found to be more similar among species than those of the non-coding regions. The phylogenies reconstructed from the cp genomes of 61 representative species of Cucurbitaceae reflected the currently accepted classification, in which C. ficifolia is sister to the other Cucurbita species, however, different interspecific relationships were found between Cucurbita species. Conclusions These results will be valuable in the classification of C. ficifolia genetic resources and will contribute to our understanding of evolutionary relationships within the genus Cucurbita.

With the increasing demand for thermal management in electronic devices and industrial systems, nanofluids have emerged as a research hotspot due to their superior thermal conductivity and heat transfer efficiency. Among them, CuO-H2O demonstrates excellent heat transfer performance due to its high thermal conductivity, Fe3O4-H2O offers potential for further optimization by combining thermal and magnetic properties, and Al2O3-H2O exhibits strong chemical stability, making it suitable for a wide range of applications. These three nanofluids are representative in terms of particle dispersibility, thermal conductivity, and physical properties, providing a comprehensive perspective on the impact of nanofluids on microchannel heat exchangers. This study investigates the heat transfer performance and flow characteristics of various types and volume fractions of nanofluids in microchannel heat exchangers. The results reveal that with increasing flow rates, the convective heat transfer coefficient and Nusselt number of nanofluids exhibit an approximately linear growth trend, primarily attributed to the turbulence enhancement effect caused by higher flow rates. Among the tested nanofluids, CuO-H2O demonstrates the best performance, achieving a 4.89% improvement in the heat transfer coefficient and a 1.64% increase in the Nusselt number compared to pure water. Moreover, CuO-H2O nanofluid significantly reduces wall temperatures, showcasing its superior thermal management capabilities. In comparison, the performance of Al2O3-H2O and Fe3O4-H2O nanofluids is slightly inferior. In terms of flow characteristics, the pressure drop and friction factor of nanofluids exhibit nonlinear variations with increasing flow rates. High-concentration CuO-H2O nanofluid shows a substantial pressure drop, with an increase of 7.33% compared to pure water, but its friction factor remains relatively low and stabilizes at higher flow rates. Additionally, increasing the nanoparticle volume fraction enhances the convective heat transfer performance; however, excessively high concentrations may suppress heat transfer efficiency due to increased viscosity, leading to a decrease in the Nusselt number. Overall, CuO-H2O nanofluid exhibits excellent thermal conductivity and flow optimization potential, making it a promising candidate for efficient thermal management in MCHEs. However, its application at high concentrations may face challenges related to increased flow resistance. These findings provide valuable theoretical support and optimization directions for the development of advanced thermal management technologies.

ObjectiveRhizosphere microorganisms play crucial roles in the growth and development of plants, disease resistance, and environmental adaptability. As the only wild pepper variety resource in China, domesticated Capsicum frutescens Linn. (Xiaomila) exhibits varying beneficial traits and affects rhizosphere microbial composition compared with its wild counterparts. In this study, we aimed to identify specific rhizosphere microbiome and metabolism patterns established during the domestication process.MethodsThe rhizosphere microbial diversity and composition of domesticated and wild C. frutescens were detected and analyzed by metagenomics. Non-targeted metabolomics were used to explore the differences of metabolites in rhizosphere soil between wild and domesticated C. frutescens.ResultsWe found that the rhizosphere microbial diversity of domesticated variety was significantly different from that of the wild variety, with Massilia being its dominant bacteria. However, the abundance of certain beneficial microbes such as Gemmatimonas, Streptomyces, Rambibacter, and Lysobacter decreased significantly. The main metabolites identified in the wild variety included serylthreonine, deoxyloganic acid, vitamin C, among others. In contrast, those identified in the domesticated group were 4-hydroxy-l-glutamic acid and benzoic acid. Furthermore, the differentially enriched pathways were concentrated in tyrosine and tryptophan biosynthesis, histidine and purine-derived alkaloids biosynthesis, benzoic acid family, two-component system, etc.ConclusionThis study revealed that C. frutescens established specific rhizosphere microbiota and metabolites during domestication, which has important significance for the efficient utilization of beneficial microorganisms in breeding and cultivation practices.

Cucumber (Cucumis sativus L.) belongs to the cucumber genus of the Cucurbitaceae family, and the selection of cultivars with minimal or no lateral branches can enhance the cultivation management efficiency. The growth of lateral branches is inhibited by strigolactone. To investigate the regulatory mechanism of strigolactone on the lateral branch development in cucumber, the cultivar LZ1 exhibiting multiple lateral branches was selected as the experimental material. The axillae of the plants were infiltrated with 1, 5, and 10 μmol·L−1 germination releaser 24 (GR24) at the four- to five-leaf stage. It was identified that 1 μmol·L−1 GR24 exhibited the most potent inhibitory effect on cucumber lateral branches. Additionally, exogenous strigolactone decreased the auxin content in the apical bud and axillae and increased the auxin content in the stem. This inhibited polar auxin transport in the axillary bud and promoted polar auxin transport in the apical bud. The content of strigolactone in the axilla region of cucumbers was elevated, whereas the synthesis and expression of cytokinin in the same area were reduced. A low concentration of GR24 induced the expression of cucumber branched 1 (csbrc1), whereas a high concentration of GR24 downregulated the expression of cucumber lateral suppressor (cscls) and blind (csblind), which inhibited the growth of cucumber lateral branches.

Tomato spotted wilt virus (TSWV) is an important virus that has rapidly spread throughout the world. TSWV seriously hinders the production of tomato (Solanum lycopersicum) and other plants. In order to discover more new genes and metabolites related to TSWV resistance in tomato plants, the genes and metabolites related to the resistance of tomato plants inoculated with TSWV were identified and studied herein. The tomato TSWV-resistance line YNAU335 (335) and TSWV-susceptible lines NO5 and 96172I (961) were used as the transcriptome and metabolome research materials. Transcriptomic and metabolomic techniques were used to analyze the gene and metabolite response mechanisms to TSWV inoculation. A total of 3 566, 2 951, and 2 674 differentially expressed genes (DEGs) were identified in lines 335, NO5, and 961, respectively. Meanwhile, 208, 228, and 273 differentially accumulated metabolites (DAMs) were identified in lines 335, NO5, and 961, respectively. In line 335, the number of DEGs was the highest, but the number of DAMs was lowest. Furthermore, 903 DEGs and 94 DAMs were common to the response to TSWV in the three inbred lines. The 903 DEGs and 94 DAMs were mainly enriched in the plant hormone signal transduction and flavonoid synthesis pathways. In addition, many nucleotide-binding site-leucine-rich repeat genes and transcription factors were found that might be involved in the TSWV response. These results provide new insights into TSWV resistance mechanisms.

In the post-genomic era, virus-induced gene silencing (VIGS) has played an important role in research on reverse genetics in plants. Commonly used Agrobacterium-mediated VIGS inoculation methods include stem scratching, leaf infiltration, use of agrodrench, and air-brush spraying. In this study, we developed a root wounding–immersion method in which 1/3 of the plant root (length) was cut and immersed in a tobacco rattle virus (TRV)1:TRV2 mixed solution for 30 min. We optimized the procedure in Nicotiana benthamiana and successfully silenced N. benthamiana, tomato (Solanum lycopersicum), pepper (Capsicum annuum L.), eggplant (Solanum melongena), and Arabidopsis thaliana phytoene desaturase (PDS), and we observed the movement of green fluorescent protein (GFP) from the roots to the stem and leaves. The silencing rate of PDS in N. benthamiana and tomato was 95–100%. In addition, we successfully silenced two disease-resistance genes, SITL5 and SITL6, to decrease disease resistance in tomatoes (CLN2037E). The root wounding–immersion method can be used to inoculate large batches of plants in a short time and with high efficiency, and fresh bacterial infusions can be reused several times. The most important aspect of the root wounding–immersion method is its application to plant species susceptible to root inoculation, as well as its ability to inoculate seedlings from early growth stages. This method offers a means to conduct large-scale functional genome screening in plants.

The cellular structure formed in additive manufactured 316L stainless steel (316L SS) plays a crucial role in achieving strength–ductility synergy. The cellular structure was also found in the 316L SS fabricated by electron beam freeform fabrication process (EBF3), and its microstructural characterization was analyzed. Interrupted tensile tests were carried out to demonstrate the effect of the cellular structure on the mechanical properties. The results showed that the microstructure of the as-deposited 316L SS consisted of the single austenite phase, and the enrichment of Cr and Mo elements was found in the cellular boundary. The orientation of the cellular structure and boundary was similar, and the obvious {100} preferred orientation was parallel to the building direction. The yield strength and fracture elongation of the as-deposited 316L SS parts were 316 MPa and 37.2%, respectively. Its ultimate tensile strength with a value of 632 MPa, was higher than that manufactured by conventional methods. The hardness and elastic modulus in the cellular interior were 2.88 ± 0.11 GPa and 227.45 ± 10.66 GPa, respectively. The cellular boundary had a higher hardness and a lower elastic modulus than the cellular interior. At the initial stage of tensile deformation, the cellular boundary deforms first due to its lower elastic modulus, causing the dislocation pileup and orientation change. The high dislocation density improved the slip resistance leading to the appearance of deformation twins after necking. The micron-dimples and nano-dimples formed in the fracture surface with the deformation of the cellular boundary and interior. Different from 316L SS fabricated by other additive manufacturing processes, the cellular structure improved the strain hardening capacity of the 316L SS fabricated by the EBF3 process.

Global concern has been raised by the emergence and rapid transmission of the heavily mutated SARS-CoV-2 Omicron variant (B.1.1.529). So far, the infection features and immune escape ability of the Omicron variant have not been extensively studied. Here, we produced the Omicron pseudovirus and compared its entry, membrane fusion, and immune escape efficiency with the original strain and the dominating Delta variant. We found the Omicron variant showed slightly higher infectivity than the Delta variant and a similar ability to compete with the Delta variant in using Angiotensin-converting enzyme 2 (ACE2) in a BHK21-ACE2 cell line. However, the Omicron showed a significantly reduced fusogenicity than the original strain and the Delta variant in both BHK21-ACE2 and Vero-E6 cells. The neutralization assay testing the Wuhan convalescents’ sera one-year post-infection showed a more dramatic reduction (10.15 fold) of neutralization against the Omicron variant than the Delta variant (1.79 fold) compared with the original strain with D614G. Notably, immune-boosting through three vaccine shots significantly improved the convalescents’ immunity against the Omicron variants. Our results reveal a reduced fusogenicity and a striking immune escape ability of the Omicron variant, highlighting the importance of booster shots against the challenge of the SARS-CoV-2 antigenic drift.

Water dimer radical cations are regarded as key intermediates in many aqueous reactions and biochemical processes. However, the structure of the water dimer radical cations, and particularly the inter-conversion between their isomers, remain difficult to investigate experimentally due to their short lifetime and low abundance under ambient conditions. Furthermore, the isomers cannot be distinguished in a full mass spectra. In this study, we report the experimental evidence for the hemi-bonded and proton-transferred isomers of gas-phase water dimer radical cations, and the inter-conversion process between them in a linear ion trap at low pressure and near room temperature. Multiple collisions of isolated water dimer radical cations with He inside the ion trap were systematically investigated; first, under different trapping times (i.e., reaction times) ranging from 0.03 to 800 ms, and then at a very low collision energies ranging from 0.1% to 10% normalized collision energy. The proton-transferred isomers were dominant at shorter trapping times (≤250 ms), while the hemi-bonded isomers were dominant at longer trapping times (250−800 ms). Moreover, the difference in symmetry of the shapes of the H2O•+ signal profiles and the H3O+ signal profiles implied the existence of two kinds of isomers and there were small potential differences between them. Our results also suggested that by tuning the experimental parameters the hemi-bonded isomers would become dominant, which could allow the study of novel chemical reactions involving the hemi-bonded two-center-three-electron (2c-3e) structure in a linear ion trap.

The COVID-19 pandemic, which was caused by SARS-CoV-2, has had a significant effect on global public health, economies, and societies worldwide. Serum antibody testing is a critical method for the diagnosis of COVID-19 and can complement RT-PCR in the diagnosis of COVID-19 patients; however, the performance of rapid antibody assays in the clinical setting has not been established. Rapid antibody assays were evaluated by investigating 296 COVID-19-positive individuals and 542 negative individuals confirmed by clinical diagnosis. The clinical diagnostic results were used as controls to evaluate the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), kappa, and 95% confidence interval (CI) of the rapid tests. IgM-positivity had a sensitivity of 86.1% and specificity of 99.1%. IgG-positivity had a sensitivity of 86.5% and specificity of 98.7%. The sensitivity of combined IgM- and IgG-positivity in clinically confirmed patients was 73.1% in the early stage (1-7 days after symptom onset) and reached 99% 15 days after symptom onset. The concordance between rapid antibody-positive tests and clinical diagnosis-positivity had a kappa value of 0.93. In addition, the false-positive rate of IgM and IgG combined nucleic acid detection was 30% in the early stage. The combined use of IgM and IgG could serve as a more suitable alternative detection method for patients with COVID-19. The rapid antibody test can be considered as an excellent supplementary approach for detecting SARS-CoV-2 in clinical application.

The quick and accurate characterization of commercial electrochemical double-layer capacitor (EDLC) cells, especially their capacitance and direct-current equivalent series internal resistance (DCESR), is of great significance for the design, maintenance, and monitoring of EDLCs used in areas of energy, sensors, electric power, construction machinery, rail transit, automobile transportation, and military. In this study, the capacitance and DCESR of three commercial EDLC cells with similar performance were determined and compared by following the three commonly-used standards of IEC 62391, Maxwell, and QC/T741-2014, which are significantly different in test procedures and calculation methods. The analysis of the test procedures and results demonstrated that the IEC 62391 standard has the disadvantages of a large testing current, long testing time, and a complex and inaccurate DCESR calculation, whereas the Maxwell standard has the disadvantages of a large testing current, a small capacitance, and large DCESR testing results, and furthermore the QC/T 741 standard has the disadvantages of a high resolution requirement for the equipment and small DCESR results. Therefore, an improved method was proposed to determine the capacitance and DCESR of EDLC cells by short-time constant voltage charging and discharging interruption methods, respectively, with the advantages of high accuracy, low equipment requirements, short testing time, and the easy calculation of DCESR over the original three standards.

Abstract Background Over the last decade, the rapid development of high-throughput sequencing platforms has accelerated species description and assisted morphological classification through DNA barcoding. However, the current high-throughput DNA barcoding methods cannot obtain full-length barcode sequences due to read length limitations (e.g. a maximum read length of 300 bp for the Illumina’s MiSeq system), or are hindered by a relatively high cost or low sequencing output (e.g. a maximum number of eight million reads per cell for the PacBio’s SEQUEL II system). Results Pooled cytochrome c oxidase subunit I (COI) barcodes from individual specimens were sequenced on the MGISEQ-2000 platform using the single-end 400 bp (SE400) module. We present a bioinformatic pipeline, HIFI-SE, that takes reads generated from the 5′ and 3′ ends of the COI barcode region and assembles them into full-length barcodes. HIFI-SE is written in Python and includes four function modules of filter, assign, assembly and taxonomy. We applied the HIFI-SE to a set of 845 samples (30 marine invertebrates, 815 insects) and delivered a total of 747 fully assembled COI barcodes as well as 70 Wolbachia and fungi symbionts. Compared to their corresponding Sanger sequences (72 sequences available), nearly all samples (71/72) were correctly and accurately assembled, including 46 samples that had a similarity score of 100% and 25 of ca. 99%. Conclusions The HIFI-SE pipeline represents an efficient way to produce standard full-length barcodes, while the reasonable cost and high sensitivity of our method can contribute considerably more DNA barcodes under the same budget. Our method thereby advances DNA-based species identification from diverse ecosystems and increases the number of relevant applications.

Afferent loop (A-loop) obstruction presenting as acute pancreatitis is a rare clinical entity. We report a case of A-loop obstruction that occurred 15 years after Billroth II gastrectomy, leading to acute pancreatitis and accompanied by duodenal perforation and peritonitis. A 63-year-old man complaining of upper abdominal pain, distention, and nausea was referred to our hospital. The patient was previously treated with antibiotics and gastrointestinal decompression at the primary healthcare institute after being diagnosed with acute pancreatitis. However, the symptoms did not improve. Upon inter-hospital transportation, he experienced a period of relief from the pain but soon developed signs of diffuse peritonitis. Laboratory examination showed elevated serum amylase and lipase. A computed tomography scan revealed slight edema of the pancreas, a dilated and fluid-filled bowel loop across the mid-abdomen, and fluid accumulation in the abdominal cavity and pelvis. An emergency laparotomy was conducted, followed by symptomatic treatments. The patient had an uneventful recovery and was discharged in 4 weeks.

Most COVID-19 convalescents can build effective anti-SARS-CoV-2 humoral immunity, but it remains unclear how long it can maintain and how efficiently it can prevent the reinfection of the emerging SARS-CoV-2 variants. Here, we tested the sera from 248 COVID-19 convalescents around 1 year post-infection in Wuhan, the earliest known epicenter. SARS-CoV-2 immunoglobulin G (IgG) was well maintained in most patients and potently neutralizes the infection of the original strain and the B.1.1.7 variant. However, varying degrees of immune escape was observed on the other tested variants in a patient-specific manner, with individuals showing remarkably broad neutralization potency. The immune escape can be largely attributed to several critical spike mutations. These results suggest that SARS-CoV-2 can elicit long-lasting immunity but this is escaped by the emerging variants.

Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of coronavirus disease 2019 (COVID-19), which has resulted in a global pandemic.Methodology: We used a two-step polymerase chain reaction to detect the ACE genotype and ELISA kits to detect the cytokine factor. We also used proteomics to identify the immune pathway related to the ACE protein expression.Result: In this study, we found that the angiotensin-converting enzyme (ACE) deletion polymorphism was associated with the susceptibility to COVID-19 in a risk-dependent manner among the Chinese population. D/D genotype distributions were higher in the COVID-19 disease group than in the control group (D/D odds ratio is 3.87 for mild (p value < 0.0001), 2.59 for moderate (p value = 0.0002), and 4.05 for severe symptoms (p value < 0.0001), logic regression analysis. Moreover, genotype-specific cytokine storms and immune responses were found enriched in patients with the ACE deletion polymorphism, suggesting the contribution to the susceptibility to COVID-19. Finally, we identified the immune pathway such as the complement system related to the ACE protein expression of patients by lung and plasma proteomics.Conclusion: Our results demonstrated that it is very important to consider gene polymorphisms in the population to discover a host-based COVID-19 vaccine and drug design for preventive and precision medicine.

The highly heavy metal resistant strain Cupriavidus metallidurans BS1 was isolated from the Zijin gold–copper mine in China. This was of particular interest since the extensively studied, closely related strain, C. metallidurans CH34 was shown to not be only highly heavy metal resistant but also able to reduce metal complexes and biomineralizing them into metallic nanoparticles including gold nanoparticles. After isolation, C. metallidurans BS1 was characterized and complete genome sequenced using PacBio and compared to CH34. Many heavy metal resistance determinants were identified and shown to have wide-ranging similarities to those of CH34. However, both BS1 and CH34 displayed extensive genome plasticity, probably responsible for significant differences between those strains. BS1 was shown to contain three prophages, not present in CH34, that appear intact and might be responsible for shifting major heavy metal resistance determinants from plasmid to chromid (CHR2) in C. metallidurans BS1. Surprisingly, the single plasmid – pBS1 (364.4 kbp) of BS1 contains only a single heavy metal resistance determinant, the czc determinant representing RND-type efflux system conferring resistance to cobalt, zinc and cadmium, shown here to be highly similar to that determinant located on pMOL30 in C. metallidurans CH34. However, in BS1 another homologous czc determinant was identified on the chromid, most similar to the czc determinant from pMOL30 in CH34. Other heavy metal resistance determinants such as cnr and chr determinants, located on megaplasmid pMOL28 in CH34, were shown to be adjacent to the czc determinant on chromid (CHR2) in BS1. Additionally, other heavy metal resistance determinants such as pbr, cop, sil, and ars were located on the chromid (CHR2) and not on pBS1 in BS1. A diverse range of genomic rearrangements occurred in this strain, isolated from a habitat of constant exposure to high concentrations of copper, gold and other heavy metals. In contrast, the megaplasmid in BS1 contains mostly genes encoding unknown functions, thus might be more of an evolutionary playground where useful genes could be acquired by horizontal gene transfer and possibly reshuffled to help C. metallidurans BS1 withstand the intense pressure of extreme concentrations of heavy metals in its environment.

This study aimed to evaluate enterovirus 71 (EV-A71) vaccine candidate strains, including their genotypes, immunogenicity and cross-neutralization capacity. From clinical samples, EV-A71 strains were separated by using Vero cells. Six strains were chosen for vaccine candidates, and the sequences were analyzed. To detect the immunogenicity of the strains, we used them to immunize NIH mice at 0 and 14 days. Cytopathic effects (CPE) were examined to determine the EV-A71 neutralizing antibody (NTAb) titer 14 d after the first and second inoculations. To evaluate the cross-neutralizing capacity of the EV-A71 vaccine candidate strains, we tested serum immunized mice with ten EV-A71 genotype strains. Six EV-A71 vaccine candidate strains were identified, all belonging to sub-genotype C4, the prevalent genotype in China. The sequence similarity of the VP1 regions of the six candidate vaccine strains and three approved inactivated vaccines was 97.58%–97.77%, and the VP1 amino acid similarity was 98.65%–99.33%. Experiments were performed to evaluate the immunogenicity and cross-neutralizing activity of the EV-A71 vaccine candidate strains. The strains had good immunogenicity 14 d after two immunizations, inducing an NTAb titer ranging from 1:94 to 1:346. The NTAb seroconversion rates 14 d after one immunization were above 80% (except HB0007), and significantly increased immunogenicity of EV-A71 strains was observed post-inoculation. Furthermore, our candidate vaccine strains had broad cross-neutralizing activity after challenge with ten sub-genotypes of EV-A71. The highest NTAb titer/lowest NTAb titer ratios of sera against EV-A71 sub-genotypes were 8.0 (JS0002), 8.0 (JS0005), 21.3 (HB0005), 21.3 (HB0007), 10.7 (HB0040) and 8.0 (GD0002), respectively. Our EV-A71 strains had good immunogenicity and cross-neutralization activity, and have the potential to serve as vaccine strains for multivalent hand, foot and mouth disease vaccines.

Background: Severe fever with thrombocytopenia syndrome (SFTS), an emerging infectious disease caused by a novel bunyavirus, was discovered in rural areas of Central China in 2009. Methods: A case-control study based on hospital data was applied to detect the potential risk sources for SFTS in SFTS-endemic counties in Hubei Province. Cases were defined as hospitalized SFTSV confirmed patients. Controls were randomly selected from non-SFTSV patients in the same hospital ward within 2 weeks of inclusion of the cases, and they were matched by age (+/− 5 years) and gender according to 1:2 matching condition. Results: 68 cases and 136 controls participated in this study. In multivariate analysis, “Contact with cattle tick” was the major risk source (Conditional Logistic Regression OR-MH = 8.62, 95% CI = 1.79-41.51), outdoor activities and working in weeds or hillside fields could increase risk of cattle tick contact and SFTS infection (Conditional Logistic Regression OR-MH = 8.82, 95% CI = 1.69-46.05, P value = 0.01). Conclusion: Our results suggested cattle might be dominant hosts in SFTS-endemic regions in Hubei Province, which provided clues to transmission mechanism of “vectors, host animals, and humans”, thus more effectively preventing and controlling the disease.

Liver cirrhosis with splenomegaly and portal hypertension has been described in the medical literature as increasing the risk of splenic rupture. We report a case of atraumatic splenic rupture in a male with liver cirrhosis associated with splenomegaly, which was further complicated by hypersplenism and intra-cystic haemorrhage in the spleen. The 56-year-old man was hospitalized because of sudden onset of intermittent pain in the left quadrant abdomen with no history of trauma. Upon admission, the patient presented no fever, palpable abdominal tenderness, splenomegaly, and hypersplenism. Enhanced computed tomography revealed a splenic subcapsular haematoma connected to a cystic mass located at the splenic hilum and free fluid in the abdomen, which is indicative of splenic rupture. The patient underwent immediate laparotomy and splenectomy followed by proper management. Post-surgery diagnostic microscopy indicated liver steatosis, perivenular fibrosis, and regenerative nodules, which are suggestive of cirrhosis. The patient was discharged from the hospital with an uneventful recovery.

The non-pathogenic Human Pegivirus (HPgV, formerly GBV-C/HGV), the most prevalent RNA virus worldwide, is known to be associated with reduced morbidity and mortality in HIV-infected individuals. Although previous studies documented its ubiquity and important role in HIV-infected individuals, little is known about the underlying genetic mechanisms that maintain high genetic diversity of HPgV within the HIV-infected individuals. To assess the within-host genetic diversity of HPgV and forces that maintain such diversity within the co-infected hosts, we performed phylogenetic analyses taking into account 229 HPgV partial E1-E2 clonal sequences representing 15 male and 8 female co-infected HIV patients from Hubei province of central China. Our results revealed the presence of eleven strongly supported clades. While nine clades belonged to genotype 3, two clades belonged to genotype 2. Additionally, four clades that belonged to genotype 3 exhibited inter-clade recombination events. The presence of clonal sequences representing multiple clades within the HIV-infected individual provided the evidence of co-circulation of HPgV strains across the region. Of the 23 patients, six patients (i.e., five males and one female) were detected to have HPgV recombinant sequences. Our results also revealed that while male patients shared the viral strains with other patients, viral strains from the female patients had restricted dispersal. Taken together, the present study revealed that multiple infections with divergent HPgV viral strains may have caused within-host genetic recombination, predominantly in male patients, and therefore, could be the major driver in shaping genetic diversity of HPgV.

Hybrid complexes with N,N′-bis(2-benzimidazolylmethyl)amine and cyclen moieties are novel enzyme mimics and controlled DNA release materials, which could interact with DNA through three models under different conditions. In this paper, the interactions between plasmid DNA and seven different complexes were investigated, and the methods to change the interaction patterns by graphene oxide (GO) or concentrations were also investigated. The cleavage of pUC19 DNA promoted by target complexes were via hydrolytic or oxidative mechanisms at low concentrations ranging from 3.13 × 10−7 to 6.25 × 10−5 mol/L. Dinuclear complexes 2a and 2b can promote the cleavage of plasmid pUC19 DNA to a linear form at pH values below 7.0. Furthermore, binuclear hybrid complexes could condense DNA as nanoparticles above 3.13 × 10−5 mol/L and partly release DNA by graphene oxide with π-π stacking. Meanwhile, the results also reflected that graphene oxide could prevent DNA from breaking down. Cell viability assays showed dinuclear complexes were safe to normal human hepatic cells at relative high concentrations. The present work might help to develop novel strategies for the design and synthesis of DNA controllable releasing agents, which may be applied to gene delivery and also to exploit the new application for GO.

GB virus C (GBV-C), which is highly prevalent among HIV/AIDS, seemed to slow the HIV disease progression. The HIV/GBV-C co-infected individuals may represent an interesting model for the investigation of the role played by HIV infection and/or the immune system in driving the evolution of the GBV-C viral populations. The present study investigated the prevalence and population dynamics of GB virus C in HIV infected individuals representing 13 geographic regions of Hubei Province of China. Approximately 37% of HIV-1 infected individuals were infected with GBV-C and genotype 3 is appeared to be predominant. Utilizing the 196 complete E2 nucleotide sequence data from 10 HIV/GBV-C infected individuals and employing coalescence based phylogenetic approaches; the present study has investigated the intra-host dynamics of GBV-C. The results revealed patient-specific unique GBV-C viral lineages and each viral lineage showed the evidence of rapid population expansion in respective HIV-1 infected patients, thus suggesting HIV-1 was unlikely to have been inhibiting effect on the GBV-C viral replication. GBV-C in all patients has experienced intense purifying selection, suggesting the GBV-C viral invasion and subsequent expansion within the HIV-1 infected hosts without any modification of the functional epitopes at their membrane protein. The finding of within host GBV-C recombinant sequences indicated recombination was one of the significant forces in the evolution and divergence of GBV-C.

Apoptosis repressor with caspase recruitment domain (ARC) is a highly potent and multifunctional suppressor of various types of programmed cell death (PCD) (e.g. apoptosis, necroptosis, and pyroptosis) and plays a key role in determining cell fate. Under physiological conditions, ARC is predominantly expressed in terminally differentiated cells, such as cardiomyocytes and skeletal muscle cells. Its expression and activity are tightly controlled by a complicated system consisting of transcription factor (TF), non-coding RNA (ncRNA), and post-translational modification (PTM). ARC dysregulation has been shown to be closely associated with many chronic diseases, including cardiovascular disease, cancer, diabetes, and neurodegenerative disease. However, the detailed mechanisms of ARC involved in the progression of these diseases remain unclear to a large extent. In this review, we mainly focus on the regulatory mechanisms of ARC expression and activity and its role in PCD. We also discuss the underlying mechanisms of ARC in health and disease and highlight the potential implications of ARC in the clinical treatment of patients with chronic diseases. This information may assist in developing ARC-based therapeutic strategies for patients with chronic diseases and expand researchers’ understanding of ARC.

Seroma formation and poor wound healing are common complications of many surgeries that create anatomical dead space (i.e., mastectomy), often causing tissue infection and even necrosis. Although negative pressure drainage and tissue adhesives are investigated to alleviate fluid accumulation post-surgery, however, their therapeutic efficacy remains unsatisfactory in most cases. Herein, the catechol-rich chemically crosslinked gelatin microspheres (ca-CGMSs) have been developed as biodegradable reconstructive implants for preventing seroma formation and concurrently promoting subcutaneous wound healing. Compared with the most representative hydrogel adhesive, i.e. commercial porcine fibrin sealant (PFS), the loosely packed ca-CGMSs with diameters range from 50 to 350 μm, provide numerous cell-adhesive interfaces and interconnected macro-pores for enhanced cell adhesion, proliferation and migration. Subcutaneous embedding trials show the in situ swelling aggregation and wet tissue adhesion of ca-CGMSs as well as their capacity in recruiting autologous cells in rat mastectomy models. The trials in rabbit mastectomy models demonstrate that, compared with PFS gluing, the implanted dried ca-CGMSs not only significantly inhibit seroma formation, but also achieve enhanced wound healing by inducing the formation of vascularized neo-tissue. The ca-CGMSs show a great potential to be the next-generation of restorative materials for both preventing seroma formation and healing subcutaneous wounds.

Abstract Background The development of neuropathic pain (NP) is one of the reasons why the pain is difficult to treat, and microglial activation plays an important role in NP. Recently, platelet-rich plasma (PRP) has emerged as a novel therapeutic method for knee osteoarthritis (KOA). However, it’s unclarified whether PRP has analgesic effects on NP induced by KOA and the underlying mechanisms unknown. Purpose To observe the analgesic effects of PRP on NP induced by KOA and explore the potential mechanisms of PRP in alleviating NP. Methods KOA was induced in male rats with intra-articular injections of monosodium iodoacetate (MIA) on day 0. The rats received PRP or NS (normal saline) treatment at days 15, 17, and 19 after modeling. The Von Frey and Hargreaves tests were applied to assess the pain-related behaviors at different time points. After euthanizing the rats with deep anesthesia at days 28 and 42, the corresponding tissues were taken for subsequent experiments. The expression of activating transcription factor 3 (ATF3) in dorsal root ganglia (DRG) and ionized-calcium-binding adapter molecule-1(Iba-1) in the spinal dorsal horn (SDH) was detected by immunohistochemical staining. In addition, the knee histological assessment was performed by hematoxylin-eosin (HE) staining. Results The results indicated that injection of MIA induced mechanical allodynia and thermal hyperalgesia, which could be reversed by PRP treatment. PRP downregulated the expression of ATF3 within the DRG and Iba-1 within the SDH. Furthermore, an inhibitory effect on cartilage degeneration was observed in the MIA + PRP group only on day 28. Conclusion These results indicate that PRP intra-articular injection therapy may be a potential therapeutic agent for relieving NP induced by KOA. This effect could be attributed to downregulation of microglial activation and reduction in nerve injury.
