Abstract The current study aimed to investigate the status of genes with prognostic DNA methylation sites in bladder cancer (BLCA). We obtained bulk transcriptome sequencing data, methylation data, and single-cell sequencing data of BLCA from public databases. Initially, Cox survival analysis was conducted for each methylation site, and genes with more than 10 methylation sites demonstrating prognostic significance were identified to form the BLCA prognostic methylation gene set. Subsequently, the intersection of marker genes associated with epithelial cells in single-cell sequencing analysis was obtained to acquire epithelial cell prognostic methylation genes. Utilizing ten machine learning algorithms for multiple combinations, we selected key genes (METRNL, SYT8, COL18A1, TAP1, MEST, AHNAK, RPP21, AKAP13, RNH1) based on the C-index from multiple validation sets. Single-factor and multi-factor Cox analyses were conducted incorporating clinical characteristics and model genes to identify independent prognostic factors (AHNAK, RNH1, TAP1, Age, and Stage) for constructing a Nomogram model, which was validated for its good diagnostic efficacy, prognostic prediction ability, and clinical decision-making benefits. Expression patterns of model genes varied among different clinical features. Seven immune cell infiltration prediction algorithms were used to assess the correlation between immune cell scores and Nomogram scores. Finally, drug sensitivity analysis of Nomogram model genes was conducted based on the CMap database, followed by molecular docking experiments. Our research offers a reference and theoretical basis for prognostic evaluation, drug selection, and understanding the impact of DNA methylation changes on the prognosis of BLCA.

Abstract Optimizing resource allocation and routing to satisfy service needs is paramount in large-scale networks. Software-defined networking (SDN) is a new network paradigm that decouples forwarding and control, enabling dynamic management and configuration through programming, which provides the possibility for deploying intelligent control algorithms (such as deep reinforcement learning algorithms) to solve network routing optimization problems in the network. Although these intelligent-based network routing optimization schemes can capture network state characteristics, they are prone to falling into local optima, resulting in poor convergence performance. In order to address this issue, this paper proposes an African Vulture Routing Optimization (AVRO) algorithm for achieving SDN routing optimization. AVRO is based on the African Vulture Optimization Algorithm (AVOA), a population-based metaheuristic intelligent optimization algorithm with global optimization ability and fast convergence speed advantages. First, we improve the population initialization method of the AVOA algorithm according to the characteristics of the network routing problem to enhance the algorithm’s perception capability towards network topology. Subsequently, we add an optimization phase to strengthen the development of the AVOA algorithm and achieve stable convergence effects. Finally, we model the network environment, define the network optimization objective, and perform comparative experiments with the baseline algorithms. The experimental results demonstrate that the routing algorithm has better network awareness, with a performance improvement of 16.9% compared to deep reinforcement learning algorithms and 71.8% compared to traditional routing schemes.

Abstract Background Osteoarthritis (OA) is a prevalent disease plaguing the elderly. Recently, chondrocyte ferroptosis has been demonstrated to promote the progression of OA. Peroxisome proliferator-activated receptor-γ (PPARγ) is an important factor in maintaining cartilage health. However, the relationship between PPARγ and chondrocyte ferroptosis in OA and its mechanism is completely unclear. Methods We established a surgically induced knee OA rat model to investigate PPARγ and chondrocyte ferroptosis in OA. Rat knee specimens were collected for Safranin O/Fast Green staining and immunohistochemical staining after administered orally placebo or pioglitazone (PPARγ agonist) for 4 weeks. We used RSL3 to establish a chondrocyte ferroptosis model cultured in vitro to study the role of PPARγ activation toward ferroptosis, mitochondrial function, and PTEN-induced putative kinase 1 (Pink1)/Parkin-dependent mitophagy. GW9662 (PPARγ antagonist), Mdivi-1 (mitophagy inhibitor), and chloroquine (mitophagy inhibitor) were employed to investigate the mechanism of PPARγ-Pink1/Parkin-dependent mitophagy in the inhibition of ferroptosis. Results We found that PPARγ activation by pioglitazone attenuated not only OA but also inhibited the expression of the ferroptosis marker acyl-CoA synthetase long-chain family member 4 (ACSL4) at the same time in rats. Furthermore, in vivo and in vitro data indicated that PPARγ activation restored Pink1/Parkin-dependent mitophagy, improved mitochondrial function, inhibited chondrocyte ferroptosis, and delayed the progression of OA. Conclusions The present study demonstrated that PPARγ activation attenuates OA by inhibiting chondrocyte ferroptosis, and this chondroprotective effect was achieved by promoting the Pink1/Parkin-dependent mitophagy pathway.

Abstract As the main dust source area in East Asia, the Gobi Desert (GD) exerts critical impacts on the radiation budget in downstream regions. Dust radiative feedback in the GD on the Mongolian cyclone, however, remains poorly understood. Herein, the dynamic dust source is coupled with the Weather Research and Forecasting model with Chemistry (WRF-Chem) for better dust simulations. Results show that the Mongolian cyclone dominates the dust event in May 2019. Dust radiative feedback results in downward momentum transport and cools northeast of Mongolia by affecting zonal winds and temperature advection. Lower-troposphere cooling and upper-atmosphere warming change the atmospheric vertical structure and enhance baroclinicity. Further, cold air deeply descends into the bottom of the warm air in a wedge shape and promotes the ascension of warm air to enhance the Mongolian cyclone. The strong dust is maintained and continues to cause high dust concentrations in northern China via westerlies. This study explores how dust radiative feedback over the GD can intensify the Mongolian cyclone and provides a scientific reference for related studies.

Abstract Sterol carrier protein 2 (SCP2) is highly expressed in human osteoarthritis (OA) cartilage, accompanied by ferroptosis hallmarks, especially the accumulation of lipid hydroperoxides (LPO). However, the role of SCP2 in chondrocyte ferroptosis remains unexplored. Here, we identify that SCP2 transports cytoplasmic LPO to mitochondria in RSL3-induced chondrocyte ferroptosis, resulting in mitochondrial membrane damage and release of reactive oxygen species (ROS). The localization of SCP2 on mitochondria is associated with mitochondrial membrane potential, but independent of microtubules transport or voltage-dependent anion channel. Moreover, SCP2 promotes lysosomal LPO increase and lysosomal membrane damage through elevating ROS. However, SCP2 is not directly involved in the cell membrane rupture caused by RSL3. Inhibition of SCP2 markedly protects mitochondria and reduces LPO levels, attenuating chondrocyte ferroptosis in vitro and alleviating the progression of OA in rats. Our study demonstrates that SCP2 mediates the transport of cytoplasmic LPO to mitochondria and the spread of intracellular LPO, accelerating chondrocyte ferroptosis.

Deep reinforcement learning has strong abilities of decision-making and generalization and often applies to the quality of service (QoS) optimization in software defined network (SDN).However, traditional deep reinforcement learning algorithms have problems such as slow convergence and instability.An algorithm of quality of service optimization algorithm of based on deep reinforcement learning (AQSDRL) was proposed to solve the QoS problem of SDN in the data center network (DCN) applications.AQSDRL introduces the softmax deep double deterministic policy gradient (SD3) algorithm for model training, and a SumTree-based prioritized empirical replay mechanism was used to optimize the SD3 algorithm.The samples with more significant temporal-difference error (TD-error) were extracted with higher probability to train the neural network, effectively improving the convergence speed and stability of the algorithm.The experimental results show that the proposed AQSDRL effectively reduces the network transmission delay and improves the load balancing performance of the network than the existing deep reinforcement learning algorithms.

Abstract Background Kidney disease of children markedly affects their health and development. Limited clinical data of early‐stage kidney disease render a tremendous challenge for the accurate diagnosis. Trio whole‐exome sequencing (Trio‐WES) is emerging as a first‐line diagnostic strategy in pediatric kidney disease, and shows important implications for the precision medicine strategies of children with kidney disease. Methods Trio‐WES was performed in 133 Chinese children with kidney disease and their parents. The results for casual variants in genes known to cause kidney disease were analyzed. We further assessed the genetic diagnostic yield and the clinical implications of genetic testing. Results An overall diagnostic yield of 52.63% (70/133) was found, and the diagnostic rates ranged from 44.74% to 59.62% in different clinical phenotypes. The diagnostic yield of the three groups of simple proteinuria, renal insufficiency, and “other” was 50%, 50%, and 54.55%, respectively. Eight‐seven diagnostic variants were identified in 70 probands with variants spanning 30 genes. The top 7 genes with diagnostic variants were COL4A5 (23, 26.44%), COL4A4 (13, 14.94%), ADCK4 (7, 8.05%), CLCN5 (3, 3.45%), ACE (3, 3.45%), PKD1 (3, 3.45%), and SLC12A3 (3, 3.45%), accounting for 63.22% of all variations in the cohort. Conclusions The retrospective cohort study summarized the clinical utility of genetic testing in 133 probands, and expanded the phenotypic and genetic profiles of kidney disease in children. Trio‐WES is an efficient diagnostic tool for children with kidney disease, which facilitates the clinical diagnosis and treatment. Our findings have important implications for the precise diagnosis of childhood nephropathy and may provide clinical guideline for disease management.

The functionalization of noble metals is an effective approach to lowering the sensing temperature and improving the sensitivity of metal oxide semiconductor (MOS)-based gas sensors. However, there is a dearth of comparative analyses regarding the differences in sensitization mechanisms between the two functionalization modes of noble metal loading and doping. In this investigation, we synthesized Pt-doped CuO gas-sensing materials using a one-pot hydrothermal method. And for Pt-loaded CuO, Pt was deposited on the synthesized pristine CuO surface by using a dipping method. We found that both functionalization methods can considerably enhance the response and selectivity of CuO toward NO2 at low temperatures. However, we observed that CuO with Pt loading had superior sensing performance at 25 °C, while CuO with Pt doping showed more substantial response changes with an increase in the operating temperature. This is mainly due to the different dominant roles of electron sensitization and chemical sensitization resulting from the different forms of Pt present in different functionalization modes. For Pt doping, electron sensitization is stronger, and for Pt loading, chemical sensitization is stronger. The results of this study present innovative ideas for understanding the optimization of noble metal functionalization for the gas-sensing performance of metal oxide semiconductors.

The two-dimensional electron gas (2DEG) generated at the LaAlO3/SrTiO3 interface has been in the focus of oxides research since its first discovery. Although oxygen vacancies play an important role in the generation of the insulator-to-metal transition of the SrTiO3 bare surface, their contribution at the LaAlO3/SrTiO3 interface remains unclear. In this work, we investigated a LaAlO3/SrTiO3 heterostructure with regional distribution of defect-based localized polar sites at the interface. Using static and time-resolved threshold photoemission electron microscopy, we prove that oxygen vacancies are induced near those polar sites, resulting in the increase of carrier density of the 2DEG states. In addition, oxygen-related surface states were uncovered, which we attributed to the release of lattice oxygen during the formation of oxygen vacancies. Such effects are mainly found spatially located around the defect sites at the buried interface, while other regions remain unaffected. Our results confirm that the itinerant electrons induced by oxygen vacancies can coexist with the charge transfer mechanism in the LaAlO3/SrTiO3 heterostructure, together leading to the formation of the metallic interface. These observations provide fundamental insights into the nature of LaAlO3/SrTiO3 interface based 2DEG and unique perspectives for potential applications.

Neocardiochiles Szépligeti, 1908, is a rare Neotropical genus of the subfamily Cardiochilinae Ashmead, 1900. The genus was previously synonymized with Heteropteron Brullé, 1846 by Dangerfield et al. (1999). In this study, we examined multiple specimens of Heteropteron-related genera: Heteropteron, Neocardiochiles, and Wesmaelella Spinola, 1851, and resurrect Neocardiochiles as a valid genus based on morphological data. As a result, five new species, N. alexeyi Kang, sp. nov. from Ecuador, N. franki Kang, sp. nov. from Costa Rica, N. braeti Kang, sp. nov., N. chriscarltoni Kang, sp. nov., and N. victoriae Kang, sp. nov., from French Guiana are included as members of Neocardiochiles and described based on morphological and molecular data. Additionally, four species previously included in Heteropteron are transferred to Neocardiochiles: Neocardiochiles fasciipennis Szépligeti, 1908, comb. nov., Neocardiochiles hasegawai (Dabek & Whitfield, 2020) comb. nov., Neocardiochiles kidonoi (Dabek & Whitfield, 2020), comb. nov., and Neocardiochiles whitfieldi (Mercado, 2003), comb. nov.. Diagnosis of each taxon and both traditional and interactive identification keys to Neocardiochiles species are included. Molecular data of N. alexeyi sp. nov., N. chriscarltoni sp. nov., N. victoriae sp. nov., and N. hasegawaii (Dabek & Whitfield, 2020), are also provided.

Mounting evidence indicates that inhibition of microglial activation and neuronal pyroptosis plays important roles in brain function recovery after subarachnoid hemorrhage (SAH). LDC7559 is a newly discovered gasdermin D (GSDMD) inhibitor. Previous studies have demonstrated that LDC7559 could inhibit microglial proliferation and pyroptosis. However, the beneficial effects of LDC7559 on SAH remain obscure. Based on this background, we investigated the potential role and the mechanism of LDC7559 on SAH-induced brain damage both in vivo and in vitro. The findings revealed that microglial activation and neuronal pyroptosis were evidently increased after SAH, which could be markedly suppressed by LDC7559 both in vivo and in vitro. Meanwhile, LDC7559 treatment reduced neuronal apoptosis and improved behavior function. Mechanistically, LDC7559 decreased the levels of GSDMD and cleaved GSDMD after SAH. In contrast, nod-like receptor pyrin domain-containing 3 (NLRP3) inflammasome activation by nigericin increased GSDMD-mediated pyroptosis and abated the beneficial effects of LDC7559 on SAH-induced brain damage. However, LDC7559 treatment did not significantly affect the expression of NLRP3 after SAH. Taken together, LDC7559 might suppress neuronal pyroptosis and microglial activation after SAH by inhibiting GSDMD, thereby promoting brain functional recovery.

New colonies of Formosan subterranean termites are founded by monogamous pairs. During swarming season, alates (winged reproductives) leave their parental colony. After swarming, they drop to the ground, shed their wings, and male and female dealates find suitable nesting sites where they mate and become kings and queens of new colonies. The first generation of offspring is entirely dependent on the nutritional resources of the founder pair consisting of the fat and protein reserves of the dealates and their microbiota, which include the cellulose-digesting protozoa and diverse bacteria. Since termite kings and queens can live for decades, mate for life and colony success is linked to those initial resources, we hypothesized that gut microbiota of founders affect pair formation. To test this hypothesis, we collected pairs found in nest chambers and single male and female dealates from four swarm populations. The association of three factors (pairing status, sex of the dealates and population) with dealate weights, total protozoa, and protozoa Pseudotrichonympha grassii numbers in dealate hindguts was determined. In addition, Illumina 16S rRNA gene sequencing and the QIIME2 pipeline were used to determine the impact of those three factors on gut bacteria diversity of dealates. Here we report that pairing status was significantly affected by weight and total protozoa numbers, but not by P. grassii numbers and bacteria diversity. Weight and total protozoa numbers were higher in paired compared to single dealates. Males contained significantly higher P. grassii numbers and bacteria richness and marginally higher phylogenetic diversity despite having lower weights than females. In conclusion, this study showed that dealates with high body weight and protozoa numbers are more likely to pair and become colony founders, probably because of competitive advantage. The combined nutritional resources provided by body weight and protozoa symbionts of the parents are important for successful colony foundation and development.

In this study, a vertical twin-roll cast (TRC) thermal-fluid coupling model was established to determine a suitable roll-casting velocity. A slab of 0.9 mm thick roll-cast Al-50 wt.% Si alloy with a two-phase zone width of 473 °C was prepared at a velocity of 20 m/min for the first time. In addition, the finite element simulation showed that the average heat transfer rate had a positive correlation with the roll-casting velocity. When the roll-casting velocity increased to 20 m/min, the average heat transfer rate reached 1260 K/s, at which point, the average size of the primary Si was merely 16.5 μm. This was a decrease of approximately 90% when compared with the size of alloys cast with conventional methods. After carrying out semi-solid heating, an observation on the distribution of alloy elements identified a novel phenomenon of solute redistribution, i.e., inverse segregation. This inverse segregation process was highly significant in further controlling solute redistribution in the alloy. Additionally, it provided a new concept for improving roll-cast segregation.

Subterranean termites depend nutritionally on their gut microbiota, which includes protozoa as well as taxonomically and functionally diverse bacteria. Our previous metavirome study revealed a high diversity and novel families of bacteriophages in the guts of Coptotermes formosanus workers from New Orleans, Louisiana, United States. Two assembled bacteriophage genomes (Phages TG-crAlp-04 and 06, family Podoviridae) existed in all colonies and showed similarity to a prophage (ProJPt-Bp1) previously sequenced from a bacterial endosymbiont (Candidatus Azobacteroides pseudotrichonymphae, CAP) of protozoa in the gut of a termite species of the genus Prorhinotermes from Taiwan. In this study the genomes of Phage TG-crAlp-04 and 06 were subjected to detailed functional annotation. Both phage genomes contained conserved genes for DNA packaging, head and tail morphogenesis, and phage replication. Approximately 30% of the amino acid sequences derived from genes in both genomes matched to those of ProJPt-Bp1 phage or other phages from the crAss-like phage group. No integrase was identified; the lack of a lysogeny module is a characteristic of crAss-like phages. Primers were designed to sequence conserved genes of the two phages and their putative host bacterium (CAP) to detect their presence in different termite species from native and introduced distribution ranges. Related strains of the host bacterium were found across different termite genera and geographic regions. Different termite species had separate CAP strains, but intraspecific geographical variation was low. These results together with the fact that CAP is an important intracellular symbiont of obligate cellulose-digesting protozoa, suggest that CAP is a core gut bacterium and co-evolved across several subterranean termite species. Variants of both crAss-like phages were detected in different Coptotermes species from the native and introduced range, but they did not differentiate by species or geographic region. Since similar phages were detected in different termite species, we propose the existence of a core virome associated with core bacterial endosymbionts of protozoa in the guts of subterranean termites. This work provides a strong basis for further study of the quadripartite relationship of termites, protozoa, bacteria, and bacteriophages.

The gravitational deposition and coagulation of aerosols were studied experimentally based on the COSTTHES (Containment Source-Term and Thermal Hydraulics Experiment System) facility and numerically investigated based on a continuous general dynamic equation; the experimental and the numerical results were in good agreement. In the present study, the applicability of the zero-dimensional model for deposition and coagulation of aerosols was verified. A considerable distortion can occur due to the coagulation of the aerosol, which significantly influenced the removal rate of the aerosol with a diameter greater than 0.76 µm, and the dynamic shape factor value of the aerosol used in the present experiment was obtained as 4. In the experimental and numerical studies, it was found that the combined effect of the gravitational deposition and coagulation of the aerosol can increase the number density of the aerosol with specific diameters, and the concentration-increment size would increase as settling continued.

Resolving the photonic modes in real space is essential to understand the fundamental process and control of photonic behavior in optoelectric devices. However, the understanding of the photonic modes of semimetal Cd3As2 is still lacking. Herein, the quasicylindrical waves (QCW) on Cd3As2 nanoplates using photoemission electron microscopy (PEEM) are found out. The QCW is due to the optical field scattered by subwavelength indentations, with wavelength being the same as the incident light, and their amplitude decays as r−1/2 from the edges. The transverse magnetic (TM) mode dominates the observed QCW of Cd3As2 nanoplates, which is affected by the edge structure. In broadband from UV to visible, the QCWs on Cd3As2 nanoplates are observed. Further, nanostructures to achieve subwavelength focusing and interference lattice of the Cd3As2 optical QCW are achieved. These findings demonstrate the regulation of QCW, which benefits optimizing the optoelectronic performances of semimetal materials in the future.

Abstract Traffic and vehicle simulations are often developed individually. However, vehicle performance is heavily affected by traffic conditions. Cosimulations of traffic and vehicle under real‐road situations can reflect the semi‐real‐world performance of vehicles, with traffic conditions being taken into considerations. This paper proposed an approach to combine the traffic and vehicle simulations that are realized by simulation of urban mobility (SUMO) and GT‐Suite software, respectively. In this paper, the sensitivities of the road grade and vehicle speed to the fuel economy and exhaust emissions were investigated; vehicle fuel consumption and regular exhaust emissions on a real‐road were analyzed; the effect of the traffic accident and congestions on fuel consumption and exhaust emissions were quantified. The results indicated that nitrogen oxides (NOx) and soot emission were consistent with fuel consumption rate, which was dominated by vehicle acceleration whose effect was aggravated by road grade. The fuel penalties caused by accident were in the range of 0.015‐0.023 kg depending on the severity of the accidents. The fuel consumption increased from 1.199 to 1.312 kg and 1.559 kg for 900 and 1800 vehicles/h traffic flow cases compared with 180 vehicles/h traffic flow.

Age-related hearing loss (ARHL) may limit communication, which is closely associated with cognitive decline of the elderly and negatively affects their quality of life. In ARHL patients who suffer chronic otitis media (COM), hearing impairment may worsen and negatively affect the cognition and quality of life. It is currently unknown whether restoration of the conductive hearing in the mixed hearing loss through middle ear surgeries can improve both the cognitive function and quality of life of the ARHL patients. Therefore, in the present study, the ARHL patients were followed up for 6 months after middle ear surgeries for COM, and both the cognitive functions and quality of life of the patients were assessed using Montreal Cognitive Assessment and Glasgow Benefit Inventory. It was found that both the cognitive functions and quality of life were improved 6 months after middle ear surgeries. In conclusion, hearing recovery after middle ear surgeries could improve cognitive functions and quality of life of ARHL patients with COM, and surgical intervention is, hence, recommended for COM.

Anthropogenic dust (AD), as a crucial component of particulate matter, is defined as dust emitted through modifying or disturbing soil particles directly or indirectly associated with human activities in urban areas, croplands, pasturelands and dry lakes. The sources, characteristics, and impacts of AD remain poorly studied, in contrast to the large body of research on natural dust (ND). This review summarizes scientific findings published since the 1990s regarding the emissions, physical-chemical characteristics, and spatio-temporal distributions of AD from the micro to the global scale. AD accounts for 5%–60% of the global dust loading, with notable spread in existing estimates. Compared with ND, AD has more complex and variable compositions and physical-chemical properties. Influenced by human disturbances, AD exhibits small particle sizes, easily accessible critical friction velocity, and large emissions. Further research should improve the observations and simulations to investigate the complex interactions among AD, climate change, and human health.

Software-defined networking (SDN) has become one of the critical technologies for data center networks, as it can improve network performance from a global perspective using artificial intelligence algorithms. Due to the strong decision-making and generalization ability, deep reinforcement learning (DRL) has been used in SDN intelligent routing and scheduling mechanisms. However, traditional deep reinforcement learning algorithms present the problems of slow convergence rate and instability, resulting in poor network quality of service (QoS) for an extended period before convergence. Aiming at the above problems, we propose an automatic QoS architecture based on multistep DRL (AQMDRL) to optimize the QoS performance of SDN. AQMDRL uses a multistep approach to solve the overestimation and underestimation problems of the deep deterministic policy gradient (DDPG) algorithm. The multistep approach uses the maximum value of the n-step action currently estimated by the neural network instead of the one-step Q-value function, as it reduces the possibility of positive error generated by the Q-value function and can effectively improve convergence stability. In addition, we adapt a prioritized experience sampling based on SumTree binary trees to improve the convergence rate of the multistep DDPG algorithm. Our experiments show that the AQMDRL we proposed significantly improves the convergence performance and effectively reduces the network transmission delay of SDN over existing DRL algorithms.

Femtosecond filament-induced breakdown spectroscopy (FIBS) is an efficient approach in remote and in situ detection of a variety of trace elements, but it was recently discovered that the FIBS of water is strongly dependent on the large-bandgap semiconductor property of water, making the FIBS signals sensitive to laser ionization mechanisms. Here, we show that the sensitivity of the FIBS technique in monitoring metal elements in water can be efficiently improved by using chirped femtosecond laser pulses, but an asymmetric enhancement of the FIBS intensity is observed for the negatively and positively chirped pulses. We attribute the asymmetric enhancement to their different ionization rates of water, in which the energy of the photons participating in the ionization process in the front part of the negatively chirped pulse is higher than that in the positively chirped pulse. By optimizing the pulse chirp, we show that the limit of detection of the FIBS technique for metal elements in water, e.g., aluminum, can reach to the sub-ppm level, which is about one order of magnitude better than that by the transform-limited pulse. We further examine the FIBS spectra of several representative water samples including commercial mineral water, tap water, and lake water taken from two different environmental zones, i.e., a national park and a downtown business district (Changchun, China), from which remarkably different concentrations of Ca, Na, and K elements of these samples are obtained. Our results provide a possibility of using FIBS for direct and fast metal elemental analysis of water in different field environments.

Optical transport networks (OTNs) are widely used in backbone- and metro-area transmission networks to increase network transmission capacity. In the OTN, it is particularly crucial to rationally allocate routes and maximize network capacities. By employing deep reinforcement learning (DRL)- and software-defined networking (SDN)-based solutions, the capacity of optical networks can be effectively increased. However, because most DRL-based routing optimization methods have low sample usage and difficulty in coping with sudden network connectivity changes, converging in software-defined OTN scenarios is challenging. Additionally, the generalization ability of these methods is weak. This paper proposes an ensembles- and message-passing neural-network-based Deep Q-Network (EMDQN) method for optical network routing optimization to address this problem. To effectively explore the environment and improve agent performance, the multiple EMDQN agents select actions based on the highest upper-confidence bounds. Furthermore, the EMDQN agent captures the network’s spatial feature information using a message passing neural network (MPNN)-based DRL policy network, which enables the DRL agent to have generalization capability. The experimental results show that the EMDQN algorithm proposed in this paper performs better in terms of convergence. EMDQN effectively improves the throughput rate and link utilization of optical networks and has better generalization capabilities.

Against the background of the natural deposition of erosols in containment under severe accident conditions, a relevant experimental facility was designed and constructed to study the decay constant of the natural deposition of erosols. TiO2 was used as the erosol medium, and the decay constant of erosols was studied. The results show that when erosol concentration is < 15,000/cm3, the decay constant of erosols increases with the increase of the particle size. However, when it is ≥ 15,000/cm3, because the agglomeration of small particles into larger particles accelerates the deposition of particles with smaller size, the decay constant of erosols decreases first and then increases with the increase of the particle size. In addition, the effect of electrostatic on the decay constant of the natural deposition of erosols was studied. When the electrostatic effect is present, the decay constant of erosols increases significantly. Finally, the natural deposition characteristics of erosols were obtained.

Fabricating electrical double-layer capacitors (EDLCs) with high energy density for various applications has been of great interest in recent years. However, activated carbon (AC) electrodes are restricted to a lower operating voltage because they suffer from instability above a threshold potential window. Thus, they are limited in their energy storage. The deposition of inorganic compounds’ atomic layer deposition (ALD) aiming to enhance cycling performance of supercapacitors and battery electrodes can be applied to the AC electrode materials. Here, we report on the investigation of zinc oxide (ZnO) coating strategy in terms of different pulse times of precursors, ALD cycles, and deposition temperatures to ensure high electrical conductivity and capacitance retention without blocking the micropores of the AC electrode. Crystalline ZnO phase with its optimal forming condition is obtained preferably using a longer precursor pulse time. Supercapacitors comprising AC electrodes coated with 20 cycles of ALD ZnO at 70 °C and operated in TEABF4/acetonitrile organic electrolyte show a specific capacitance of 23.13 F g−1 at 5 mA cm−2 and enhanced capacitance retention at 3.2 V, which well exceeds the normal working voltage of a commercial EDLC product (2.7 V). This work delivers an additional feasible approach of using ZnO ALD modification of AC materials, enhancing and promoting stable EDLC cells under high working voltages.

Abstract Background Microglial activation and the subsequent inflammatory response in the central nervous system play important roles in secondary damage after traumatic brain injury (TBI). High-mobility group box 1 (HMGB1) protein, an important mediator in late inflammatory responses, interacts with transmembrane receptor for advanced glycation end products (RAGE) and toll-like receptors (TLRs) to activate downstream signaling pathways, such as the nuclear factor (NF)-κB signaling pathway, leading to a cascade amplification of inflammatory responses, which are related to neuronal damage after TBI. Omega-3 polyunsaturated fatty acid (ω-3 PUFA) is a commonly used clinical immunonutrient, which has antioxidative and anti-inflammatory effects. However, the effects of ω-3 PUFA on HMGB1 expression and HMGB1-mediated activation of the TLR4/NF-κB signaling pathway are not clear. Methods The Feeney DM TBI model was adopted to induce brain injury in rats. Modified neurological severity scores, brain water content, and Nissl staining were employed to determine the neuroprotective effects of ω-3 PUFA supplementation. Assessment of microglial activation in lesioned sites and protein markers for proinflammatory, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, interferon (IFN)-γ, and HMGB1 were used to evaluate neuroinflammatory responses and anti-inflammation effects of ω-3 PUFA supplementation. Immunofluorescent staining and western blot analysis were used to detect HMGB1 nuclear translocation, secretion, and HMGB1-mediated activation of the TLR4/NF-κB signaling pathway to evaluate the effects of ω-3 PUFA supplementation and gain further insight into the mechanisms underlying the development of the neuroinflammatory response after TBI. Results It was found that ω-3 PUFA supplementation inhibited TBI-induced microglial activation and expression of inflammatory factors (TNF-α, IL-1β, IL-6, and IFN-γ), reduced brain edema, decreased neuronal apoptosis, and improved neurological functions after TBI. We further demonstrated that ω-3 PUFA supplementation inhibited HMGB1 nuclear translocation and secretion and decreased expression of HMGB1 in neurons and microglia in the lesioned areas. Moreover, ω-3 PUFA supplementation inhibited microglial activation and the subsequent inflammatory response by regulating HMGB1 and the TLR4/NF-κB signaling pathway. Conclusions The results of this study suggest that microglial activation and the subsequent neuroinflammatory response as well as the related HMGB1/TLR4/NF-κB signaling pathway play essential roles in secondary injury after TBI. Furthermore, ω-3 PUFA supplementation inhibited TBI-induced microglial activation and the subsequent inflammatory response by regulating HMGB1 nuclear translocation and secretion and also HMGB1-mediated activation of the TLR4/NF-κB signaling pathway, leading to neuroprotective effects.

For the additive manufacturing (AM) of metal objects, the powder-based fusion (PBF) method is routinely utilized to fabricate macroscale parts. On the other hand, electrochemical additive manufacturing (ECAM), in which metallic structures are deposited through the electrochemical reduction of metal ions, is a promising technique for producing micro- and nanoscale objects. However, a gap exists in terms of fabricating mesoscale objects within the current AM techniques. The PBF method is limited by fabrication precision due to pronounced residual stresses, and most current ECAM systems are difficult to scale up to print mesoscale objects. In the present paper, the novel design of a low-cost ECAM 3D printer based on a microfluidic system is proposed for fabricating mesoscale metal parts. The meniscus-guided electrodeposition approach is utilized, in which a meniscus is formed between the print head and substrate, and electrodeposition is confined within the meniscus. A 3D object is fabricated by the meniscus moving with the print head according to the programmed pattern and the material subsequently being deposited at the designated locations. The key to the proposed design is to maintain a mesoscale meniscus, which normally cannot be sustained by the electrolyte surface tension with a print nozzle having a mesoscale diameter. Therefore, a microfluidic system, called the fountain pen feed system, constituting a semi-open main channel and comb structure, was designed to maintain a mesoscale meniscus throughout the printing process. Two materials, copper and nickel, with various geometric shapes were attempted to print by the proposed ECAM system, and, during the printing process, both fluid leaking and meniscus breaking were completely prevented. Free standing tilted copper pillars with controlled angles were printed to show the ability of the proposed design in fabricating 3D structures. A copper circuit was also printed on a non-conductive substrate to demonstrate a possible application of the proposed ECAM system in the fabrication of functional electronics.
