Periungual warts are common warts that grow on the periungual or nail margin, and it's challenging to remove them due to their particular location, causing its high recurrence rate and brings difficulties to the treatments. We successfully cured two cases of stubborn periungual warts by local hyperthermia. A male with warts on his hands and knees and a girl with periungual warts on her fingers received local hyperthermia of 44 °C for 30 min a time. One month after the last treatment, their lesions disappeared entirely and did not relapse during the follow-up period. These two cases showed that local hyperthermia might be a safe and effective method for treating periungual warts in patients with poor traditional treatment.

Background: Atopic dermatitis (AD) is a disturbing, relapsing inflammatory skin disease characterized by recurrent eczema foci and intense itching. Due to the effectiveness and safety of Dupilumab, it is considered a savior for AD patients.Objective: To investigate the efficacy and safety of JAK inhibitors (JAKi) plus Dupilumab therapy in refractory AD patients.Method: An observational study analyzing the disease control status of refractory AD patients under the treatment of Dupilumab. Data on patients with moderate to severe AD treated with Dupliumab were obtained from the First Affiliated Hospital of Wenzhou Medical University. The clinical effectiveness and safety of Dupilumab combined with JAKi were evaluated and monitored.Results: Of the 198 patients enrolled, 56 (28.3%) had underreacted to Dupilumab to dupilumab at 16 weeks. Proportion of patients with an improvement from baseline in the Eczema Area and Severity Index (EASI-75) of at least 75% (65.7%;130/198). Five patients eventually discontinued treatment due to adverse reactions. Among uncontrolled patients (n=51) who did not achieve EASI≤7 or IGA 0/1 during the 16-week study period, 74.5% (38/51) achieved EASI-75 by week 52, including 71.1%(27/38) in combination with JAKi therapy. No significant systemic side effects in the combined JAKi group.Limitations: Including small sample size, short observation period.Conclusion: Given the tremendous burden of AD, the demand for new targeted therapies is very high. The combination of JAKi and Dupilumab in AD patients with low response rates may dramatically changing the treatment expectations of patients and doctors.

Psoriasis is a chronic inflammatory skin disease characterized by abnormal proliferation and differentiation of keratinocytes. Since curcumol exhibits anti-inflammatory properties in various diseases, we investigated its anti-inflammatory potential in stimulated human keratinocytes. Our data show that curcumol significantly inhibits proliferation and induces cell cycle arrest in NHEK cells stimulated with proinflammatory cytokines (IL-1α, IL-17A, IL-22, oncostatin M, and TNF-α; mix M5). In addition, curcumol markedly ameliorates inflammatory response and promotes differentiation of M5-stimulated NHEK cells. Curcumol inhibits activity of JAK1, resulting in the inhibition of STAT3, downregulation of cyclin D2, and cell cycle arrest in stimulated NHEK cells. Together, our data show that curcumol reduces proliferation and inflammatory gene expression in stimulated keratinocytes by inhibiting the JAK1/STAT3 signaling, suggesting that it might serve as a potential therapeutic option for the treatment of psoriasis.

The morbidity and mortality of melanoma which accounts for 90% of cutaneous neoplasm-related deaths is growing over the last few decades. Common treatments for melanoma are limited to poor tissue selectivity, high toxicity and drug resistance. Photodynamic therapy (PDT) is an effective adjuvant therapy and could be a promising therapy for melanoma. Multiple mechanisms are involved in PDT2 and programmed cell death (PCD) which comprises of autophagy and apoptosis is likely to be a critical one. Whereas, the molecular mechanism and subsequent effect of PDT-induced autophagy in melanoma are still unclear. In this study, we first analyzed gene expression data in the TCGA3 and GEO4 databases to clarify that PDT-induced-autophagy improved the prognosis of melanoma. The expression of FOS which generally defined as an immediate-early gene (IEG) and related to cell autophagy was found significantly elevated after PDT. To further investigate whether FOS played a key role in PDT-induced-autophagy of melanoma, we first determined the optimum concentration of ICG solution for autophagy observation. Then, relative FOS expression was detected at mRNA and protein level and cell autophagy was observed by western blot and flow cytometry. We found that ICG-PDT treatment could significantly elevate FOS expression in SKCM5 B16 cells, and FOS promoted ICG-PDT-induced cell autophagy. To sum up, our data indicated that FOS was involved in ICG-PDT-induced-autophagy in melanoma and furthermore improved the prognosis of melanoma.

Emerging edging computing services and applications bring an unprecedented demand for bulk transfers at the network edges. However, the expensive access cost makes it difficult to deliver bulk data between geo-distributed-edge-datacenter. In this paper, storage in edge-datacenter is introduced into the transmission scheme for temporarily storing delay-tolerant bulky flows. We formulate the upstream resource allocation as a stratified multi-objective optimization model, which can adjust the spectrum and storage allocation between latency-critical flows and delay-tolerant flows. Our studies reveal that 60% of the system cost can be saved by trading cost-effective storage for expensive spectrum resources.

Access links that connect cloud data centers or enterprise networks to public networks are important parts of the network infrastructure and make up considerable portions of overall network expenditures. When a customer network is connected to an overlaid elastic optical network, determining the capacity of the access link is difficult. Furthermore, access links carry mixed packet streams and circuit connections, each with different performance requirements. Additionally, traffic loads are time-varying and often exhibit diurnal patterns. In this study, we formulate the access link capacity dimensioning problem as an optimization problem with a nested dynamic constraint satisfaction subproblem. Instead of using hard performance measures, such as packet delays and blocking probabilities, we use the probability of fulfillment with regard to different traffic arrival rates as the performance measure and the performance–cost ratio as our optimization objective. Numerical results suggest that a 7.5% to 19.47% cost savings can be achieved if resources are shared between packet streams and circuit connections. And if resource preemptions are allowed, the delay performance for packet streams can be improved by 12% with more frequent preemptions. Our study provides useful insights into the planning and operation of access links.

Peroxisomal LON2 protease plays a pivotal role in protein quality control through its proteolysis and chaperone functions. However, little is known regarding its function in the regulation of cellular reactive oxygen species (ROS) homeostasis and in adversity tolerance in plants. To begin to assess the function of LON2, we isolated a VcLON2 cDNA from blueberry (Vaccinium corymbosum L.), which is predicted to encode a peroxisomal protein containing a long N-terminal domain, a central AAA+ module and a C-terminal proteolytic domain. Quantitative real-time RT-PCR analysis revealed that VcLON2 was expressed ubiquitously throughout the blueberry plant including vegetative and reproductive organs, and its expression increased in older tissues. Overexpression of VcLON2 in tobacco led to increased tolerance to NaCl, PEG and heat stresses, as indicated by the lower contents of H2O2, malondialdehyde (MDA) and carbonyl groups, compared with wild type (WT). In contrast, silencing of the VcLON2 homolog, NbLON2, in tobacco seedlings resulted in significantly higher sensitivity to various abiotic stresses. Furthermore, the activity of catalase (CAT), a peroxisomal enzyme involved in the detoxification of ROS, was found to be elevated in VcLON2-overexpressed seedlings and conversely reduced in NbLON2-silenced seedlings under non-stress and stress conditions, compared with WT. Our results suggest that VcLON2 plays an important role in the maintenance of the ROS-scavenging functions of peroxisomes, which is necessary for tolerance to abiotic stresses.

We propose a resource allocation strategy on IP-over-EONs access links. It realizes the dynamic self-adaptive spectrum resource adjustment applying to traffic fluctuations and handles the performance requirements under the circuit/packet hybrid architecture.

Free-tropospheric aerosol layers and their seasonal variation over Wuhan (30.5°N, 114.4°E), China, are presented based on a 532-nm polarization lidar measurements on 162 days from January through December 2013. Using the aerosol layer selection criterions, a total of 402 free-tropospheric aerosol layer events were identified. The bottom height of the aerosol layers below 2 km accounts for 68% of the total, while approximately 76% of the layer's top height ranges from 1 km to 4 km. Out of the 402 events, 269 (67%) are optically-thin layers with aerosol optical depth (AOD) less than 0.1. The free tropospheric AOD2-7 contribute ~13–31% to the AOD0-7 and the free-tropospheric aerosol layers show considerable moderate variation. The aerosol layers have the maximum mean geometrical thickness of 1.2 km in spring, while the minimum mean thickness is 0.7 km in autumn, and the mean thickness is 0.93 km and 1 km in summer and winter, respectively. The mean backscatter coefficient of aerosol layers during spring, summer, autumn and winter were 1.8 ± 1.4 Mm−1sr−1, 2.3 ± 2 Mm−1sr−1, 2.8 ± 2.7 Mm−1sr−1 and 2.3 ± 2.2 Mm−1sr−1, respectively. Aerosol layers in different seasonal are classified by particle depolarization ratio, there are a large amount of non-spherical particles and mixed particles present in spring, autumn and winter, and the mean particle polarization ratio of aerosol layers during spring, summer, autumn and winter were 0.22, 0.06, 0.15 and 0.14, respectively.

Vitiligo is a common depigmentary disease characterized as diagnosis simplicity and cure difficulty in view of the ambiguity of etiology, thus novel and effective treatments are urgently needed. Paeoniflorin, the major active compound extracted from the root of Paeonia lactiflora Pall, a traditional Chinese medicine, has been validated pharmacological properties such as antioxidant stress, a theory participating in the occurrence of vitiligo, but the effect on melanogenesis is still unclear. In this study, melanosythesis effect of paeoniflorin and the potential mechanism were evaluated. We found that treatment with paeoniflorin at the concentration of 10 μg/ml significantly increased melanin content and intracellular tyrosinase activity of human melanocytes, in accordance with the elevation of protein levels of microphthalmia-associated transcription factor (MITF), tyrosinase-related protein 1 (TRP-1). In addition, we also investigated that paeoniflorin promoted phosphorylation of cAMP-response element binding (CREB) and extracellular signal-regulated kinase (ERK) without affecting p38 and c-Jun N-terminal kinase (JNK). These results demonstrated that paeoniflorin had a synergistic effect on normal human melanocytes via ERK/CREB pathway with up-regulation of MITF and TRP-1, enhancing melanin synthesis. Meanwhile, the milder pathological changes in vitiligo mice treat with paeoniflorin also confirmed its potential in treating vitiligo. To sum up, we suggest that paeoniflorin may be a potential medicine of vitiligo treatment in clinical.

With the rapid development of new technologies and new building materials,new network floor as a type of new building materials has been widely used in ground projects, owing to its wiring flexible, simple and practical, easy maintenance, and environmental friendly features. However, there are problems in the traditional construction process of network floor, like unreasonable arrangement, poor flatness and the waste of side plate cutting. To address these issues, this paper firstly proposes three innovative technologies for the comprehensive control of the implementation of network floor project, including CAD secondary typesetting and layout technology, precision laser level levelling + shock pads fine tuning technology, and computer code nesting of side plate technology. Furthermore, a complete set of construction technology processes has been presented. Finally, the economic and social benefits of these innovative technologies have been proved by the engineering project.
