Bu araştırmada, tavuk, martı dışkılarından ve alabalık barsak içeriklerinden hareketli aeromonaslar izole ve identifiye edilerek, bu suşlann protein profilleri SDS- PAGE yöntemiyle tespit edildi. Herbir susun protein profilinin diğer suşlarla olan benzerlik oranlan belirlenerek sonuçlar epidemiyolojik olarak değerlendirildi. Bu amaçla 50 adet tavuk, 50 adet martı dışkısı ve 50 adet alabalık barsak içeriğinden steril svaplarla alınan toplam 150 adet svap örneği Stuart taşıma ortamı içerisinde kısa sürede mikrobiyoloji laboratuvarma ulaştırıldı. Svap örneklerini hareketli aeromonaslar yönünden zenginleştirmek için Alkali Peptonlu Su (pH: 8.4) tüplerine ekim yapıldı.Daha sonra izolasyon amacıyla Aeromonas Medium'a ekim yapıldı, izole edilen bakterilerin biyokimyasal identifikasyonlan klasik yöntemlere göre yapıldı. Araştırma sonucunda tavuklara, martılara ve alabalıklara ait toplam 150 adet dışkı numunesinden 23 (%15.3) adet Aeromonas spp. izole edilerek, bunların 5 (21.7)'i A. caviae, 4 ( %17.4)'ü A. hydrophila ve 14 (%60.9)'ü A. sobria olarak identifiye edildi. İzole ve identifiye edilen hareketli aeromonas suşlarının protein profilleri SDS-PAGE yöntemiyle tespit edildi. Herbir susun protein profili diğer suşlann protein profilleri ile karşılaştırılarak benzerlik oranlan belirlendi. 23 adet susun karşılaştırılması sonucu oluşturulan 276 çiftin 241 (%87.3)'i farklı veya şüpheli ilişkili bulunurken, 35 (%12.7)'i identik bulundu. Aynı türe ait suşlar kendi içinde karşılaştırıldığında, A caviae 'nm 5 susu ile oluşturulan 10 çiftin 4 (%40)'ü şüpheli ilişkili, 6 (%60)'sı identik bulunurken, farklı çiftlere rastlanmadı. A. hydrophila' nın 4 susu ile oluşturulan 6 çiftin 1 (%16.6)'i farklı, 4 (%66.6)'ü şüpheli ilişkili bulunurken, 1 (%16.6)'i identik bulundu. A. sobria'mn 14 susu ile oluşturulan 91 çiftin 16 (%17.6)'sı farklı, 53 (%58.2)'ü şüpheli ilişkili bulunurken, 22 (%24.2)'si identik bulundu. Aynı hayvan türünden izole edilen, aynı türe ait hareketli Aeromonas suşlan karşılaştırıldı. Martılardan izole edilen A. caviae suşlan ile oluşturulan 5 çiftin tamamı (%100) identik bulundu. Alabalıklardan izole edilen A. hydrophila suşlan ile oluşturulan 3 çiftin 2 (%66.7)'si şüpheli ilişkili, 1 (%33.3)'i identik bulunurken, farklı çiftlere rastlanmadı. Tavuklardan izole edilen A. sobria suşlan ile oluşturulan 6 çiftin 5 (%83.3)'i şüpheli ilişkili, 1 (%16.7)'i identik bulunurken, farklı çiftlere rastlanmadı. Martılardan izole edilen A. sobria suşlan ile oluşturulan 6 çiftin tamamı (%100) identik bulundu. Alabalıklardan izole edilen A. sobria suşlan ile oluşturulan 15 çiftin 4 (%26.7)'ü şüpheli ilişkili, 11 (%73.3)'i identik bulunurken, farklı çiftlere rastlanmadı. İzole edilen her bir hareketli Aeromonas türüne ait suşlar, diğer türe ait suşlarla, izole edildikleri hayvan türleri dikkate alınmaksızın karşılaştırıldı. A. caviae suşlan ile, A. hydrophila suşlannın karşılaştırılması ile oluşturulan 20 çiftin 10 (%50)'u farklı, 5 (%25)'i şüpheli ilişkili bulunurken, 5 (%25)'i identik bulundu. A. caviae suşlan ile A. sobria suşlannın karşılaştırılması ile oluşturulan 70 çiftin 11 (%15.7)'i farklı, 59 (%84.2)ru şüpheli ilişkili bulunurken, identik çiftlere rastlanmadı. A. hydrophila suşlan ile A. sobria suşlannın karşılaştırılması ile oluşturulan 56 çiftin 26 (%46.2)'sı farklı, 30 (%53.6)'u şüpheli ilişkili bulunurken, identik çiftlere rastlanmadı. Farklı hayvan türlerinden izole edilen, aynı türe ait hareketli Aeromonas suşlan karşılaştınldı. Böyle bir karşılaştırma sonucu martılardan ve alabalıklardan izole edilen A. caviae suşlan ile oluşturulan 4 çiftin tamamı (%100) identik bulundu. Martıdan ve alabalıklardan izole 39edilen A. hydrophila suşları ile oluşturulan 3 çiftin 1 (%33.3)'i farklı, 2 (%66.7)'si şüpheli ilişkili bulunurken, identik çiftlere rastlanmadı. Tavuklardan ve martılardan izole edilen A. sobria suşları ile oluşturulan 16 çiftin tamamı (%100) şüpheli ilişkili bulundu. Tavuklardan ve alabalıklardan izole edilen A. sobria suşları ile oluşturulan 24 çiftin 16 (%66.6)'sı farklı, 8 (%33.3)'i şüpheli ilişkili bulunurken, identik çiftlere rastlanmadı. Martılardan ve alabalıklardan izole edilen A. sobria suşları ile oluşturulan 24 çiftin 20 (%83.3)'si şüpheli ilişkili, 4 (%16.7)'ü identik bulunurken, farklı çiftlere rastlanmadı Bu araştırmanın sonucunda, Türkiye'de martı dışkılarından hareketli aeromonaslann izolasyonu ile tavuk, martı ve gökkuşağı alabalıklarınım dışkılarından izole edilen hareketli aeromonaslann SDS-PAGE yöntemiyle protein profillerinin tespiti ilk kez gerçekleştirildi. `The isolation of Motile aeromonads strains from the feces of chicken, gull and rainbow trout (Oncorhyncus mykiss), and the determination of their protein profiles by SDS-PAGE` In this study, motile aeromonads taken from chicken and gull feces, and from the intestinal contents of rainbow trout were isolated and identified, and protein profiles of these strains were described by SDS-PAGE. In the study, a total of 150 samples, collected via sterile swabs from the feces of 50 chicken, 50 gull and 50 intestinal content of rainbow trout were used. The swabs were carried as soon as possible to microbiology laboratory in the Stuart Transport Medium. Samples were cultured onto Alkaline Peptone Water (pH:8.4) to enrich them in respect of motile aeromonads. Then, it was cultured on to Aeromonas Medium for isolation. The biochemical identification of isolated bacteria was performed according to classical methods. At the end of the study, 23 (%15.3) Aeromonas spp. were isolated from feces samples of chicken, gull and rainbow trout. Of them 5 (%21.7) were A. caviae, 4 (%17.4) A. hydrophila and 14 (%60.9) A. sobria. Protein profiles of the isolated and identified motile aeromonads strains were determined by SDS-PAGE. Protein profiles of each strain were compared with protein profiles of the other strains, and their similarity rate was recognised. 23 strains were compared and 276 pairs were obtained. Of them, 241 were either different or had uncertain relationship while 35 pairs were identical. The strains of the same species were compered among themselves. 5 strains of A. caviae were compared and 10 pairs were formed. Of them 4 (%40) pairs were found to have uncertain relationhip, 6 pairs (%60) were identical while different pairs were not determined. 4 strains of A. hydrophila were compared and 6 pairs were formed. Of them 1 pair (%16.6) was different, 4 pairs (%66.6) to have uncertain relationship while 1 pair (%16.6) was identical. 14 strains of A. sobria were compared and 91 pairs were formed. Of them 16 pairs (%17.6) were different, 53 pairs (%58.2) were found to have uncertain relationship while 22 pairs (%24.2) were found identical. The motile aeromonads strains isolated from the animals of the same kind and belonging to same species were compared. A. caviae strains isolated from gulls were compared and 5 pairs were formed. They all were identical. A. hydrophila strains isolated from rainbow trout were compared, and 3 pairs were formed. Of them 2 pairs (%66.7) were found to have uncertain relationship, 1 pair (%33.3) was identical while differend pairs were not determined. A. sobria strains isolated from chicken were compared and 6 pairs were formed. Of them 5 pairs (%83.3) were found to have uncertain relationship, 1 pair (%16.7) was identical while different pairs were not determined. A. sobria strains isolated from gulls were compared and 6 pairs were formed. They all were identical. A. sobria strains isolated from rainbow trout were compared, and 15 pairs were formed. Of them 4 pairs (%26.7) were found to have uncertain relationship, 1 1 pairs (%73.3) were identical while different pairs were not determined. The isolated strains belonging to each Motile aeromonad species were compared with the other strains without considering the kind of animal they were isolated from. A. caviae strains were compared with A. hydrophila strains, and 20 pairs were formed. Of them 10 pairs (%50) were different, 5 pairs (%25) were found to have uncertain relationship while 5 pairs (%25) were identical. A. caviae strains 41were compared with A. sobria, and 70 pairs were formed. Of them 11 pairs (%15.7) were diffrent, 59 pairs (%84.2) were found to have uncertain relationship while identical pairs were not determined. A. hydrophila strains were compared with A. soria strains and 56 pairs were formed. Of them 26 pairs (%46.2) were different, 30 pairs (%53.6) were found to have uncertain relationship while identical pairs were not determined. The strains isolated from different kinds of animals and belonging to the same species were compared. A. caviae strains isolated from gulls and rainbow trout were compared and 4 pairs were formed. Of them all were identical. A. hydrophila strains isolated from gulls and rainbow trout were compared, and 3 pairs were formed. Of them 1 pair (%33.3) was different, 2 pairs (%66.7) were found to have uncertain relationship while identical pairs were not determined. A. sobria strains isolated from chicken and gulls were compared and 16 pairs were formed. They all were found to have uncertain relationship. A. sobria strains isolated from chicken and rainbow trout were compared and 24 pairs were formed. Of them 16 pairs (%66.6) were different, 8 pairs (%33.3) were found to have uncertain relationship while identical pairs were not determined, sobria strains isolated from gulls and rainbow trout compared, and 24 pairs were formed. Of them 20 pairs (%83.3) were found to have uncertain relationship, 4 pairs (%16.7) were identical while different pairs were not determined. In conclusion, the identification of motile aeromonads from gull feces was done for the first time in Turkey as well as the determinaton of protein profiles of the isolated strains from feces of chicken, gull and rainbow and trout by SDS-PAGE. 56
