Abstract

Die zentrale Fragestellung dieser Studie ist, wie sich gesellschaftliche und kulturelle Zugehörigkeit in transnationalen Lebensläufen herausbilden und wie sie sich unter dem Einfluss von globalen und nationalen Machtstrukturen entwickeln. Zur Beantwortung dieser Fragen werden erkenntnistheoretische Ansätze zum Zusammenspiel von gesellschaftlichen Zuschreibungen und individuellen Identitätskonstruktionen mit Strukturanalysen gesellschaftlicher Machtverhältnisse in Form von Rassismus und Kolonialismus verbunden. Die Arbeit zeigt auf, wie Migrant/innen die Probleme und Herausforderungen der Integration in verschiedenen Gesellschaften und sozio-kulturellen Bezugssystemen lösen und in ihre biografischen Selbstkonstruktionen einfließen lassen. Dabei konzentriert sie sich auf afrikanische, speziell sierra-leonische Migrant/innen in Deutschland. Empirically this paper identifies how immigrants deal with challenges of integration into the host society and incorporate these experiences into their biographical self-construction. It concentrates on African and particularly Sierra Leonean immigrants in Germany. The findings are generated from a reconstructive analysis of selected biographical narrations which were scrutinized from different perspectives. 17. Metabolism of cyclohexane carboxylic acid by the photosynthetic bacterium Rhodopseudomonas palustris.

Abstract

Cyclohexane carboxylate supported relatively rapid growth (doubling times 7-8 h) of Rhodopseudomonas palustris under oxic or photosynthetic conditions, but did not serve as a substrate for either of the known aromatic CoA ligases. A CoA ligase that thioesterifies cyclohexane carboxylate was partially purified and did not cross react immunologically with the two CoA ligases purified previously from this bacterium. Crude extracts of R. palustris cells grown with a range of aromatic or alicyclic acids contained a dehydrogenase that reacted with cyclohexane carboxyl-CoA or cyclohex-1-ene carboxyl-CoA, using 2,6-dichlorophenolindophenol or ferricenium ion as electron carrier. This activity was not detected in extracts of adipate-, glutamate-, or succinate-grown cells. No oxidation or reduction of nonesterified cyclohexane carboxylate or cyclohexene carbocylate was detected in extracts of cells grown with aromatic or aliphatic substrates, neither aerobically nor anaerobically. A constitutively expressed thioesterase that hydrolyzed cyclohexane carboxyl-CoA and also some alicyclic and aliphatic CoA derivatives was purified and characterized. The enzyme had little or no activity on benzoyl-CoA or 4-hydroxybenzoyl-CoA. The presence of a thioesterase that effectively hydrolyzes cyclohexane carboxyl-CoA suggests that transient production of cyclohexane carboxylate is a physiological response to temporary excess of reductant during metabolism of aromatic compounds. 19. Phylogeny of Thioplocaand Related Filamentous Sulfide-Oxidizing Bacteria

Abstract

The phylogenetic relationships of three Thioplocaspecies, T. araucae, T. chileaeand T. ingrica, as well as Beggiatoa alba, Beggiatoasp. str 1401-13, and Thiothrix niveawere determined by 16S rRNA sequence analysis. Fluorescent in situhybridization with Thioploca-specific oligonucleotide probes was used to substantiate the sequences of T. araucaeand T. chileae. All three Thioploca species form a monophyletic group. They are affiliated with Beggiatoa albaand Beggiatoasp. str 1401-13. Thioplocaand Beggiatoaconstitute a new phylogenetic lineage within the gamma-subdivision of the proteobacteria, showing similar genetic diversity as other major groups of gamma- or beta-subdivision sulfur oxidizers. Thiothrix niveais a member of the gamma proteobacteria, but does not form a monophyletic lineage with Beggiatoaand Thioploca.

20. Cryptoendolithic growth of the red alga <e1>Galdieria sulphuraria</e1> in volcanic areas

Abstract

The habitat of the acido- and thermophilic red algae Galdieria sulphuraria and Cyanidium caldarium was examined in acidic hot sulphur springs in the vicinity of Naples (Italy). These species grew on soil and rocks, but a large part of the populations was cryptoendolithic. The endolithic algal layer (1–3 mm in thickness) was covered by amorphous silica (1–2 mm in thickness) containing traces of hydrotroilite (FeS.nH2O) and elemental sulphur. Organotrophic bacteria and fungi were not found in the algal layer. Light penetration measurements showed that 0·1–1% of the sunlight reached the upper part of the algal layer. Thus, low-light-adapted algae should be able to perform some photosynthesis in this endolithic habitat. Under conditions where light is even more limited, e.g. in winter or after darkening of the covering layer, many of the cells might not survive. Aqueous extracts of these algae are excellent growth substrates for Galdieria sulphuraria. Therefore, we propose that compounds released from dead cells in the endolithic layer are used by the surrounding Galdieria cells for heterotrophic metabolism. This would increase their chance of surviving prolonged periods under detrimental conditions.

Abstract

Within this doctoral thesis, the distribution, phylogeny and structure of the dissimilatory APS reductase (AprBA) of sulfate-reducing and sulfur-oxidizing prokaryotes (SRP and SOP) has been analyzed. An assay for aprA-based functional gene analysis was developed for determination of the SRP and SOP diversity in environmental samples. In addition, the distribution of the sulfate thiol hydrolase (SoxB, a component of the thiosulfate-oxidizing multi-enzyme system Sox) in SOP was investigated.

Abstract

The guts of termites feature suitable conditions for methane oxidizing bacteria (MOB) with their permanent production of CH4 and constant supply of O2 via tracheae. In this study, we have isolated MOB from the gut contents of the termites Incisitermes marginipennis, Mastotermes darwiniensis, and Neotermes castaneus for the first time. The existence of MOB was indicated by detecting pmoA, the gene for the particulate methane monooxygenase, in the DNA of gut contents. Fluorescence in situ hybridization and quantitative real-time polymerase chain reaction supported those findings. The MOB cell titer was determined to be 10(2)-10(3) per gut. Analyses of the 16S rDNA from isolates indicated close similarity to the genus Methylocystis. After various physiological tests and fingerprinting methods, no exact match to a known species was obtained, indicating the isolation of new MOB species.
