Introduction DSM-5 defines non-suicidal self-injury (NSSI) as socially unaccepted, direct, repeated and deliberate harm done to one’s own body. It is estimated that in a general population approximately 13-29% of adolescents present NSSI, and 70-80% among hospitalized youth. It seems that emotional dysregulation is the core characteristic of NSSI manifesting by self-harm behaviors, impulsiveness, lack of emotional awareness and experiencing high intensity of negative emotion. Emotional dysregulation is a pivotal characteristic of NSSI. Rationale of this theory is provided by the results of psychological and psychophysiological studies as well as those presenting brain activity. Neuroimaging data point to a variant pattern of brain activity of adolescents with NSSI during perception of emotionally negative stimuli i.e. hyperactivity in amygdala – a structure responsible for fear and automatic reaction to exciting stimuli and low activity of inferior frontal gyrus area – a structure responsible for inhibition and interpretation of social interactions. This activity pattern suggests a disorder of cortico-subcortical neuronal connections. Objectives The aim was to verify tDCS as a therapeutic aid for patients who exhibit NSSI despite implementation of pharmacotherapy and psychotherapy. Methods We investigated the modulation effect of tDCS treatment at the right inferior frontal gyrus (rIFG) in hospitalized adolescents with NSSI. Results Preliminary tDCS stimulation results indicate potential usefulness of this method in regulating emotions and improving executive functions. Conclusions Prefrontal cortex stimulation may restore balance in aforementioned connections and, as a result, positively influence an emotional regulation i.e. lower the impulsiveness, agitation and, by doing so, decrease NSSI frequency. Introduction Transsexual adolescents frequently present psychiatric comorbidities and psychopathology among which self-injurious behaviours are prominent (Modrego Pardo et al., 2021). GAHT seems to decrease the rate of mental health problems especially in terms of anxiety, depression and hostility (Heylens et al., 2014). However the impact of exogenous cross-sex hormones on non-suicidal and suicidal self-injurious behaviours is not thoroughly understood (Claes et al., 2015). Objectives We present the case of a transsexual boy who was first diagnosed with depression and benign self-injurious behaviours and subsequently - transsexualism. Methods He was prescribed a treatment with testosterone depot injections in fortnight intervals. The initiation of testosterone injections co-occurred with the switching of antidepressant drug. Self-injurious behaviours, substance abuse and suicidal attempts were observed regularly after GAHT onset – 10-14 days after a testosterone injection. The lethality and intensity of self-harm was greater than that observed before GAHT. After admission to the psychiatry unit pharmacotherapy was adjusted accordingly to presented symptoms. Remission of self-injurious behaviours followed. Results The incidence of self-injury 10-14 days after the injection of depot testosterone overlaps the peak of serum testosterone levels in treated patients (O’Connor et al., 2004). Moreover, a relative serotonin deficiency in a depressed patient may be insufficient to counteract testosterone believed to be driving aggressive tendencies (Batrinos, 2012). Conclusions Since psychiatric comorbidity and psychopharmacotherapy in transsexual young population is the rule rather than the exception, careful monitoring and therapy adjustment is crucial for maintaining safety and obtaining best results (Kaltiala et al., 2020). Talar avascular necrosis (AVN) is a devastating condition that frequently follows type III and IV talar neck fractures. As 60% of the talus is covered by hyaline cartilage, its vascular supply is limited and prone to trauma, which may eventually lead to AVN development. Early detection of AVN (Hawkins sign, MRI) is crucial, as it may prevent the development of the irreversible stages III and IV of AVN. Alertness is advised regarding non-obvious conditions that may cause this complication (sub chondroplasty, systemic lupus erythematosus, diabetes mellitus). Although, in stages I-II, AVN may be treated with non-surgical procedures (ESWT therapy, non-weight bearing) or joint-sparing techniques (core drilling, bone marrow aspirate injections), stages III-IV require more advanced procedures, such as joint-sacrificing procedures (hindfoot arthrodesis/ankle arthrodesis), or replacement surgery, including total talar replacement (TTR) or combined total ankle replacement (TAR). The advancement of 3D-printing technology and increased access to implant manufacturing are contributing to a rise in the production rates of third-generation total talar prostheses. As a result, there is a growing frequency of alloplasty procedures and combined total ankle replacement (TAR) surgeries. By performing TTR as opposed to deses, the operator avoids (i) delayed union, (ii) a shortening of the limb, (iii) a lack of mobility, and (iv) the stiffening of adjacent joints, which are the main disadvantages of joint-sacrificing procedures. Simultaneously, TTR and combined TAR offer (i) a brief period of weight-bearing restriction, (ii) quick pain relief, and (iii) preservation of the length of the limb. Here, we summarize the most up-to-date knowledge regarding AVN diagnosis and treatment, with a special focus on the role of TTR.

Purpose: Chondral and osteochondral lesions in the knee are common conditions that significantly impair individuals' well-being and can lead to osteoarthritis, imposing substantial burdens on healthcare systems. The limited natural healing capacity of articular cartilage necessitates innovative treatment strategies. Microfracture (MF) is a widely used technique for knee chondral defects, but its long-term efficacy is often inadequate. Although recent randomised controlled trials have compared microfractures with scaffold-enhanced therapies, a comprehensive systematic review and meta-analysis are lacking., Methods: An extensive literature search was conducted in PubMed and EMBASE databases following PRISMA guidelines. Inclusion criteria focused on randomised controlled trials (RCTs) comparing microfractures alone to matrix-induced chondrogenesis for knee chondral defects with at least a 12-month follow-up. Ten randomised controlled trials conducted between 2013 and 2024, enroling 378 patients, were included., Results: The meta-analysis showed no significant superiority of scaffolds over MF (p > 0.05) in International Knee Documentation Committee, Knee Injury and Osteoarthritis Outcome, Visual Analog Scale, and Magnetic Resonance Observation of Cartilage Repair Tissue scores at 12 and 24 months. However, individual studies suggested the potential benefits of scaffolds, especially in long-term outcomes. Clinical improvements from MF typically decline after 2-3 years, underscoring the need for long-term follow-up in future research., Conclusion: Our meta-analysis shows no significant difference between MF and MF with scaffold in treating knee cartilage defects, though some long-term RCTs demonstrate statistically significant differences. The absence of a universally accepted algorithm for analysing knee chondral defects limits this study. Dental composites are gaining great popularity in restorative dentistry because of their aesthetic appeal and capacity to replicate the natural color of teeth. Nevertheless, their lifespan and durability rely on various factors, such as the polishing technique and the environmental conditions they are exposed to. The study aimed to assess the influence of the method of final polishing of dental composite on the surface roughness and microhardness of materials also considering the environment of different pHs. Disc-shaped samples (5 mm diameter and 2 mm thickness) have been prepared for microhardness and roughness tests from two dental composites: A2 Clearfil Majesty ES 2 Classic and A2D Clearfil Majesty Premium. One-third of samples were polished with polishing discs, OptiDisc, another one-third of samples were polished with Eve Diacomp Twist rubbers and polishing brush with diamond particles, and rest of the samples were stored without any polishing (the control group). Tested materials were incubated in distilled water or acidic buffer (pH = 2) for 3 weeks at a temperature of 37 °C. No statistically significant differences were found for roughness for the two materials tested after incubation in liquids. A decrease in Vicker microhardness was found for Clearfil Majesty ES 2 Classic after soaking in a low pH liquid, and no such relationship was found for Clearfil Majesty Premium. The improved resistance of these materials to the negative oral environment may result in the longer survival of composite restorations in patients with poor diet or diseases, causing a decrease in oral pH.

Injuries to the articular cartilage of the human knee joint, commonly resulting from trauma, inflammation, or age- and activity-related wear and tear, have significant implications, primarily leading to osteoarthritis (OA). Conservative treatments for such injuries often yield suboptimal clinical outcomes. Surgical interventions using current methods may not consistently provide satisfactory results, largely due to the formation of low-quality scar tissue lacking the biomechanical properties of hyaline cartilage. In this retrospective study, we compared the results of two promising methods for regenerating cartilage defects in the knee joints using scaffolds soaked with stem cells of different origins: bone marrow aspirate concentrate mesenchymal stem cells (BMAC-MSCs) and human umbilical cord-derived mesenchymal stem cells (hUCB-MSCs). We evaluated 39 patients (39 knees, hUCB-MSCs: 20 knees, BMAC: 19 knees) at the 12-month follow-up using VAS, KOOS, Lysholm scales and radiologically with M-MOCART 2.0 score. The analysis demonstrated substantial overall improvement in both groups, notably reflected in enhanced quality of life for the patients. Interestingly, the final scores in the hUCB-MSCs group were comparable to those in the BMAC-MSCs group, with no statistically significant differences observed, despite variations in preoperative age and lesion size. Notably, the hUCB-MSCs group consisted of significantly older individuals with larger lesion sizes. Both procedures were found to be safe, and improvement was observed in both groups, which holds promise for future clinical investigations.

Whey protein isolate (WPI) hydrogels are attractive biomaterials for application in bone repair and regeneration. However, their main limitation is low mechanical strength. Therefore, to improve these properties, the incorporation of ceramic phases into hydrogel matrices is currently being performed. In this study, novel whey protein isolate/calcium silicate (WPI/CaSiO 3 ) hydrogel biomaterials were prepared with varying concentrations of a ceramic phase (CaSiO 3 ). The aim of this study was to investigate the effect of the introduction of CaSiO 3 to a WPI hydrogel matrix on its physicochemical, mechanical, and biological properties. Our Fourier Transform Infrared Spectroscopy results showed that CaSiO 3 was successfully incorporated into the WPI hydrogel matrix to create composite biomaterials. Swelling tests indicated that the addition of 5% ( w / v ) CaSiO 3 caused greater swelling compared to biomaterials without CaSiO 3 and ultimate compressive strength and strain at break. Cell culture experiments demonstrated that WPI hydrogel biomaterials enriched with CaSiO 3 demonstrated superior cytocompatibility in vitro compared to the control hydrogel biomaterials without CaSiO 3 . Thus, this study revealed that the addition of CaSiO 3 to WPI-based hydrogel biomaterials renders them more promising for bone tissue engineering applications.

Halogenated, labeled with deuterium, tritium or doubly labeled with deuterium and tritium in the 3S position of the side chain isotopomers of L-phenylalanine and phenylpyruvic acid were synthesized. Isotopomers of halogenated L-phenylalanine were obtained by addition of ammonia from isotopically enriched buffer solution to the halogenated derivative of (E)-cinnamic acid catalyzed by phenylalanine ammonia lyase. Isotopomers of halogenated phenylpyruvic acid were obtained enzymatically by conversion of the appropriate isotopomer of halogenated L-phenylalanine in the presence of phenylalanine dehydrogenase. As a source of deuterium was used deuterated water, as a source of tritium was used a solution of highly diluted tritiated water. In cat breeding, an important step is reproduction and obtaining healthy offspring. The main factor determining the survival of newborn kittens is the normal course and length of pregnancy. The purpose of this study was to determine the effect of gestation length on the early development of kittens. It was shown that kittens (premature kittens) later double their body weight (p < .01), have lower daily gains (p < .01) and higher body weight during eye-opening (p < .01), which occurs later than in kittens born on time. In addition, due to a shorter period of prenatal development, they require more time until the day of eye-opening, which, together with the length of pregnancy, was defined as development age., (© 2023 Wiley-VCH GmbH. Fracture toughness is one of the main factors influencing the durability of light-cured composites used for dental restorations and fillings. One of the methods of increasing the fracture toughness is the modification of the matrix with liquid acrylonitrile-free liquid rubber. This study aimed to assess the miscibility of acrylonitrile-free liquid rubber with a blend of resins and their stability over time, and to determine the optimal amount of liquid rubber (LR) in the blend due to mechanical properties. Two blends of dimethacrylate resins were used: resin "F" composed of BisGMA (60 wt.%), TEGDMA (20 wt.%), BisEMA (10 wt.%) and UDMA (10 wt.%), and "C" resin containing BisGMA (40 wt.%), TEGDMA (40 wt.%), BisEMA (10 wt.%) and UDMA (10 wt.%). The modifier Hypro ® 2000X168LC VTB liquid rubber was used in at 1%, 2%, 3%, 4%, 5%, 10%, 15% and 20% by weight in the resin blend. The miscibility was assessed by microscopy. The fracture toughness, flexural strength and Young's modulus were determined in the bending test. The results showed that the solubility of the liquid rubber depends on the ratio of BisGMA/TEGDMA in the resins. In resins with 40 wt.% TEGDMA, the LR solubility was as high as 5%, while resins with 20 wt.% TEGDMA, the liquid rubber did not dissolve. The LR-resin mixtures showed good time stability, and no changes in the size or morphology of the rubber domains were found after 24 h of mixing. The maximum fracture toughness (2.46 MPa m 1/2 ) was obtained for 5 wt.% LR in resin F and for 15 wt.% LR in resin C (2.53 MPa m 1/2 ). The modification with liquid rubber resulted in an exponential reduction in both flexural strength and Young's modulus. The analysis of the results of the mechanical tests allowed us to determine the optimal amount of LR for both resins. For resin F it was 5.4 wt.%, and for resin C it was 8.3 wt.%. It can be stated that the optimal amount of liquid rubber increases with its solubility in the resin.

Hydroxyapatites of high calcium and phosphate ions adsorption capacity are highly bioactive. However, they cause the removal of these ions from tissue liquids and cell culture media, thus reducing viability and proliferation potential of osteoblasts. Addition of small amount of gypsum (calcium sulfate dihydrate) to such hydroxyapatite-based composites may help to compensate the ions removal and stimulate the osteoblasts growth and proliferation. Therefore, the aim of this work was to enrich the highly porous hydroxyapatite-based composite with gypsum and verify its effect on ions adsorption as well as osteoblasts viability and proliferation. The results showed that addition of 1.5-1.75% gypsum caused short-term calcium ions compensation in media incubated with the composite and time-shifted increase of osteoblasts proliferation. Moreover, presence of gypsum in the composite increased the content of large pores in SBF-incubated biomaterials with no effect on their microstructure or mechanical parameters. The aim of this study was to evaluate the effect of modification with liquid rubber on the adhesion to tooth tissues (enamel, dentin), wettability and ability to inhibit bacterial biofilm formation of resin-based dental composites. Two commercial composites (Flow-Art-flow type with 60% ceramic filler and Boston-packable type with 78% ceramic filler; both from Arkona Laboratorium Farmakologii Stomatologicznej, Nasutów, Poland) were modified by addition of 5% by weight (of resin) of a liquid methacrylate-terminated polybutadiene. Results showed that modification of the flow type composite significantly ( p < 0.05) increased the shear bond strength values by 17% for enamel and by 33% for dentine. Addition of liquid rubber significantly ( p < 0.05) reduced also hydrophilicity of the dental materials since the water contact angle was increased from 81-83° to 87-89°. Interestingly, modified packable type material showed improved antibiofilm activity against Steptococcus mutans and Streptococcus sanguinis (quantitative assay with crystal violet), but also cytotoxicity against eukaryotic cells since cell viability was reduced to 37% as proven in a direct-contact WST-8 test. Introduction of the same modification to the flow type material significantly improved its antibiofilm properties (biofilm reduction by approximately 6% compared to the unmodified material, p < 0.05) without cytotoxic effects against human fibroblasts (cell viability near 100%). Thus, modified flow type composite may be considered as a candidate to be used as restorative material since it exhibits both nontoxicity and antibiofilm properties.

Curdlan hydrogel obtained after thermal gelling exhibits elasticity and high water-absorbing capacity. However, its modifications leading to the increase of biofunctionality usually alter its solubility and reduce mechanical parameters. Therefore, curdlan hydrogel was modified by deposition of polydopamine to improve its capacity to bind biologically active molecules with free amino groups. It exhibited the unchanged structure, mechanical properties and increased soaking capacity. Aminoglycoside antibiotic (gentamicin) as a model molecule was effectively immobilized to such modified curdlan via quinone moiety (but not amino groups) of polydopamine. Approximately 50 % of the immobilized drug was released following Fickian diffusion and inhibited the bacterial growth in matrix-surrounding medium in prolonged manner. The remaining drug amount was stably attached and prevented the hydrogel against bacterial adhesion even when all the mobile drug has been released. This study reports a novel impedimetric immunosensor for protein D detection in purified and bacterial (Haemophilus influenzae, Hi) samples. The detection was based on antigen recognition by anti-protein D antibodies (apD) immobilised at the maze-like boron-doped carbon nanowall electrodes (B:CNW). The B:CNW electrodes were synthesised, and their surface was characterised by scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS) methods. The sensor was prepared in a two-step procedure: apD were covalently linked on the previously modified B:CNW electrodes using diazonium salt. Modification steps were controlled by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) measurements. The immunosensor exhibited excellent electrochemical performance, stability, satisfactory sensitivities, and linear ranges for antigen detection. Protein D was detected down to 2.39 × 10 2 fg/mL with a linear range extending from 3.37 × 10 -11 to 3.37 × 10 -3 μg/mL (in purified sample). Next, Hi's LOD was 5.20 × 10 2 CFU/mL with a linear range of 8.39 × 10 1 -8.39 × 10 3 CFU/mL. Selectivity studies showed no reaction with negative samples as Streptococcus pyogenes, Streptococcus pneumoniae or Bordetella parapertussis bacteria. The ternary HAp/curdlan/nanomagnetite hybrids with ceramic and polymer phase incorporation of magnetite nanoparticles (MNPs) were fabricated to study their heating ability under action of the alternating magnetic field (AMF), 808 nm near infrared laser radiation (NIR) and their synergic stimulation. The energy conversion was evaluated in terms of the specific absorption rate (SAR) as a function of the MNPs concentration in composites and to estimate their potential in temperature-controlled regenerative processes and hyperthermia. Measurements were carried out on dry and Ringer's solution soaked composite materials in order to mimic in situ conditions. It was found that the MNPs release during prolonged experiment is limited and has no significant effect on energy conversion emphasizing stability of the hybrids. Incorporation of the MNPs in polymer phase of the hybrid can additionally limit particle leaking as well as plays a role as insulating layer for the heat dissipation lowering the risk of sample overheating. In general, it was shown that maximum temperature of hybrid can be achieved in a relatively short time of exposure to stimulating factors whereas its control can be done through optimization of experiment conditions. Background: The oral environment can negatively affect the physical properties of fiber-reinforced composite (FRC) materials, which can lead to the deterioration of mechanical stability and reduce the span of their clinical usefulness., Objectives: The aim of this study was to determine the influence of artificial saliva on the selected mechanical properties of FRC., Material and Methods: The core of the polymer material selected for the study was a bundle of ultrahighmolecular-weight polyethylene (UHMWPE) fibers. Fourteen samples were stored in an incubator at 37°C, in 20 mL of artificial saliva solution, and weighed on days 1 and 28. At the same time, mechanical tests were performed, including the measurements of Young's modulus, tensile stress, maximum tensile force, and tensile deformation., Results: The analysis of basic statistics together with the results of the Shapiro-Wilk test and the distribution of Spearman's rho coefficient showed a strong negative relationship between the pair of variables - tensile deformation and the sorption of synthetic saliva. The results related to Young's modulus of elasticity and tensile stress were not statistically significant., Conclusions: Water penetration into the space between the fibers does not adversely affect the mechanical properties of the material tested. In the static tensile test, high and desired mechanical strength was observed, which may justify the effective use of this type of material in clinical practice and may be a good alternative to prosthetic restorations, whose retention is obtained only through a mechanical connection with the abutment tooth.

There are many methods widely applied in the engineering of biomaterials to improve the mechanical properties of the dental composites. The aim of this study was to assess the effect of modification of dental composites with liquid rubber on their mechanical properties, degree of conversion, viscosity, and cytotoxicity. Both flow and packable composite consisted of a mixture of Bis-GMA, TEGDMA, UDMA, and EBADMA resins reinforced with 60 and 78 wt.% ceramic filler, respectively. It was demonstrated that liquid rubber addition significantly increased the fracture toughness by 9% for flow type and 8% for condensable composite. The influence of liquid rubber on flexural strength was not statistically significant. The addition of the toughening agent significantly reduced Young's modulus by 7% and 9%, respectively, while increasing deformation at breakage. Scanning electron microscopy (SEM) observations allowed to determine the mechanisms of toughening the composites reinforced with ceramic particles. These mechanisms included bridging the crack edges, blocking the crack tip by particles and dissipation of fracture energy by deflection of the cracks on larger particles. The degree of conversion increased after modification, mainly due to a decrease in the matrix resin viscosity. It was also shown that all dental materials were nontoxic according to ISO 10993-5, indicating that modified materials have great potential for commercialization and clinical applications.

The purpose of this study was to determine whether dialysis method allows for efficient protein entrapment in curdlan-based hydrogel. Thus, bovine serum albumin, a model of bioactive protein, was incorporated into curdlan matrix using ion-exchanging dialysis method against two concentrations of CaCl 2 solution - 2% and 10%, respectively. Then, physicochemical, mechanical, and biological properties of the bovine serum albumin-loaded curdlan hydrogels were evaluated. Received results show that neither the polymer nor the entrapment procedure change the bovine serum albumin conformation (as proven by Fourier transform infrared spectroscopy and circular dichroism spectroscopy) and the process guarantees high protein entrapment efficiency (above 95%). The curdlan-based carrier obtained against 2% of CaCl 2 solution was found to possess higher swelling ability, release greater amounts of bovine serum albumin (up to 4 weeks), and exhibit superior biocompatibility compared to curdlan-based carrier obtained against 10% of CaCl 2 solution. Thus, dialysis method enables efficient protein entrapment in curdlan hydrogel and obtained protein carrier via dialysis method into 2% of CaCl 2 solution may be considered as a promising protein delivery system especially for tissue engineering applications. Aim: The studies on tooth structure provided basis for nanotechnology-based dental treatment approaches known as nanodentistry which aims at detection and treatment of oral pathologies, such as dental caries and periodontal diseases, insufficiently being treated by conventional materials or drugs. This review aims at defining the role of nanodentistry in the medical area, its potential and hazards., Materials & Methods: To validate these issues, current literature on nanomaterials for dental applications was critically reviewed., Results: Nanomaterials for teeth restoration, bone regeneration and oral implantology exhibit better mechanical properties and provide more efficient esthetic outcome. However, still little is known about influence of long-term function of such biomaterials in the living organism., Conclusion: As application of nanomaterials in industry and medical-related sciences is still expanding, more information is needed on how such nano-dental materials may interfere with oral cavity, GI tract and general health.

This review compiles the combined chemical and enzymatic synthesis of aromatic l-amino acids (l-phenylalanine, l-tyrosine, l-DOPA, l-tryptophan, and their derivatives and precursors) specifically labeled with carbon and hydrogen isotopes, which were elaborated in our research group by the past 20 years. These compounds could be then employed to characterize the mechanisms of enzymatic reactions via kinetic and solvent isotope effects methods.

Natural polymer-based composites become very popular in design of biomaterials for bone tissue regeneration. Their rheological and mechanical properties are typically evaluated in vitro according to standard methods. However, in vivo such composites may behave completely different due to specific body conditions (e.g., inflammation-assisted acidification of tissue liquids). Such surprising phenomenon was observed for hydroxyapatite/β-1, 3-glucan composite during alveolus extraction socket augmentation in people. Implanted composite showed unexpected massive swelling, caused stitches loosening and wound reopening 5 days after implantation. Acidic pH was selected as a potential factor affecting this phenomenon, as all implantations are accompanied by transient local inflammation and acidification. Composite parameters were therefore studied after 5 days of soaking in acidic medium by weight and volume measurement, SEM, XRD, FTIR, microCT, and mercury intrusion techniques. Results showed significant volume increase, pore size remodelling and ceramic phase rearrangement in the composite, accompanied by change of mechanical properties. Simple quantitative correction of amount of implanted composite was sufficient to control in vivo appearance of side effects, confirming that pH-related volume increase of HAp/glucan composite is not a disqualifying factor. This strongly suggests the necessity of very detailed and individually designed tests concerning all new polymer-based composite biomaterials before clinical trials. © 2016 Wiley Periodicals, Inc. The addition of an antibacterial agent to dental implants may provide the opportunity to decrease the percentage of implant failures due to peri-implantitis. For this purpose, in this study, the potential efficacy of nanosilver-doped titanium biomaterials was determined. Titanium disks were incorporated with silver nanoparticles over different time periods by Tollens reaction, which is considered to be an eco-friendly, cheap, and easy-to-perform method. The surface roughness, wettability, and silver release profile of each disc were measured. In addition, the antibacterial activity was also evaluated by using disk diffusion tests for bacteria frequently isolated from the peri-implant biofilm: Streptococcus mutans, Streptococcus mitis, Streptococcus oralis, Streptococcus sanguis, Porphyromonas gingivalis, Staphylococcus aureus , and Escherichia coli . Cytotoxicity was evaluated in vitro in a natural human osteoblasts cell culture. The addition of nanosilver significantly increased the surface roughness and decreased the wettability in a dose-dependent manner. These surfaces were significantly toxic to all the tested bacteria following a 48-hour exposure, regardless of silver doping duration. A concentration of 0.05 ppm was sufficient to inhibit Gram-positive and Gram-negative species, with the latter being significantly more susceptible to silver ions. However, after the exposure of human osteoblasts to 0.1 ppm of silver ions, a significant decrease in cell viability was observed by using ToxiLight™ BioAssay Kit after 72 hours. Data from the present study indicated that the incorporation of nanosilver may influence the surface properties that are important in the implant healing process. The presence of nanosilver on the titanium provides an antibacterial activity related to the bacteria involved in peri-implantitis. Synthesis of 3-fluoro-[2- 2 H]-L-alanine (3-F-[ 2 H]-L-Ala) in reductive amination of 3-fluoropyruvic acid catalysed by L-alanine dehydrogenase (AlaDH) was described. Aromatic amino acids such as l-phenylalanine, l-tryptophan, 3',4'-dihydroxy-l-phenylalanine (l-DOPA), and their derivatives 3',4'-dihydroxyphenylacelaldehyde (DOPAL) and 3',4'-dihydroxyphenylethanol (DOPET), play an essential role in human metabolic processes. Incorrect or slow biotransformation of these compounds leads to some metabolic dysfunctions and in some cases to some neurodegenerative diseases. Therefore, studies of the biotransformation mechanisms of these metabolites draw biochemists' and medical researchers' attention. This study investigates the mechanisms of biotransformation of the aforementioned compounds using kinetic (KIE) and solvent (SIE) isotope effect methods. The overview presents the results and the numerical values of KIE and SIE methods, obtained in the study of biotransformation of l-phenylalanine, 5'-chloro-l-tryptophan, and l-DOPA, catalyzed by the enzymes from the lyases group (phenylalanine ammonia lyase, tryptophan indole-lyase, and tyrosine decarboxylase). Deuterium KIE was also determined during the deamination of 2'-chloro-l-phenylalanine in the presence of the enzyme l-phenylalanine dehydrogenase, as well as in the conversion of DOPAL into DOPET catalyzed by the enzyme alcohol dehydrogenase. Recent studies have shown that thermal method for β-1,3-glucan (curdlan) gelation performed at temperature above 80°C enables fabrication of biocompatible bone scaffolds. The aim of this study was to establish new method for fabrication of β-1,3-glucan/hydroxyapatite (glu/HA) scaffold using ion-exchanging dialysis for curdlan gelation that allows for the modifications of the glu/HA material with thermo-sensitive agents like growth factors or adhesive proteins. Obtained results reveal that fabricated scaffold appears to be highly osteoconductive as it is nontoxic, promotes osteoblast growth and proliferation as well as increases bone alkaline phosphatase level thereby enhancing cell differentiation. It was demonstrated that developed new method for the glu/HA scaffold fabrication allows to obtain material that not only can be modified with thermo-sensitive agents at the stage of production process but also is a promising candidate for bone tissue engineering applications to act as a framework for osteoblasts to spread and form new bone. Halogenated, labeled with tritium and doubly with deuterium and tritium, derivatives of L-tryptophan, i.e. 5'-bromo-[2-(3)H]-, 5'-bromo-[2-(2)H/(3)H]-, 5'-fluoro-[2-(3)H]-5'-fluoro-[2-(2)H/(3)H]-, 6'-fluoro-[2-(3)H]-, 6'-fluoro-[2-(2)H/(3)H]-L-tryptophan, as well as, L-tyrosine, i.e. 3'-fluoro-[2-(3)H]-, 3'-fluoro-[2-(2)H/(3)H]-, 3'-chloro-[2-(3)H]-, and 3'-chloro-[2-(2)H/(3)H]-L-tyrosine, and also L-phenylalanine, i.e. 2'-fluoro-[(3S)-(3)H]-, 2'-fluoro-[(3S)-(2)H/(3) H]-, 2'-chloro-[(3S)-(3)H]-, 2'-chloro-[(3S)-(2)H/(3)H]-, 4'-chloro-[(3S)-(3)H]-, and 4'-chloro-[(3S)-(2)H/(3)H]-L-phenylalanine were synthesized using enzymatic methods. Isotopomers of L-tryptophan were synthesized by coupling of halogenated indoles with S-methyl-L-cysteine carried out in deuteriated or tritiated incubation media. Labeled halogenated derivatives of L-tyrosine were obtained by the enzymatically supported exchange between halogenated L-tyrosine and isotopic water. Labeled halogenated isotopologues of L-Phe were synthesized by the enzymatic addition of ammonia to halogenated cinnamic acid. Bone implantable materials based on calcium sulfate dihydrate dissolve quickly in tissue liquids and release calcium ions at very high levels. This phenomenon induces temporary toxicity for osteoblasts, may cause local inflammation and delay the healing process. Reduction in the calcium ion release rate by gypsum could be therefore beneficial for the healing of gypsum-filled bone defects. The aim of this study concerned the potential use of calcium phosphate ceramics of various porosities for the reduction of high Ca(2+) ion release from gypsum-based materials. Highly porous ceramics failed to reduce the level of Ca(2+) ions released to the medium in a continuous flow system. However, it succeeded to shorten the period of high calcium level. It was not the phase composition but the high porosity of ceramics that was found crucial for both the shortening of the Ca(2+) release-related toxicity period and intensification of apatite deposition on the composite. Nonporous ceramics was completely ineffective for this purpose and did not show any ability to absorb calcium ions at a significant level. Unlabelled: Automatic implantable cardioverter-defibrillators (ICDs) are nowadays an essential tool for reducing mortality due to sudden cardiac death. Technological advances in the miniaturization of devices and lead fixation, and the development of surgical techniques has led to more frequent implantation of the defibrillation leads outside the right ventricular apex (RVA), especially in those patients requiring cardiac pacing, as data from large clinical trials showed that chronic RVA pacing is harmful, especially in heart failure subjects, who are an important target for the ICD. Very few studies have been published comparing the electrical characteristics of leads placed in the RVA versus those implanted outside the RVA, mainly to right ventricular outflow tract of the heart (RVA), hence any subsequent analysis of this issue seems to be a valuable addition to the available information in this topic. The aim of this study was to compare the electrical parameters of ICD leads implanted into the right ventricular apex (RVA), to those placed in one of the alternative sites: the right ventricular outflow tract (RVOT), or the area of the interventricular septum (RVS)., Material and Methods: Retrospective analysis of medical data from a single centre (teaching hospital), which included 132 patients with ICD implanted in 2010-2011, both in primary and secondary prevention of sudden cardiac death. We compared the most important electrical parameters of the ICD system, as the resistance of the pacing system, resistance of high-voltage coil, the amplitude of the sensed beats and pacing threshold. In addition, we compared the time of implantation, X-ray fluoroscopy time and X-ray exposure., Results: There were no statistically significant differences between the two analysed groups in terms of pacing-system resistance (601.012 vs. 602.7omega, p = 0,499), high-energy coil resistance (63.7omega vs. 67.22, p = 0,201), amplitude of sensed R-waves (14,6mV vs. 15.3mV, p = 0, 710) and the pacing threshold energy (0,368 microJ vs. 0.259 microJ, p = 0,803). Also the duration of implantation (123, 3 min vs. 123, 9 min, p = 0,940), fluoroscopy time (11,0 minutes vs. 8,6 minutes, p = 0,06) and dose exposure (1594, 5cGy/cm2 vs. 2094, 4cGy/cm2, p = 0,069) were comparable in both groups., Conclusions: Implantation of ICD leads to the RVOT/RVS is a safe procedure, and the basic electrical parameters of such systems are comparable to ICDs with lead implanted to the RVA.

The bioavailability of active compounds depends on their two main features: solubility and permeability. The experimental determination of these factors is rather cumbersome. The free enthalpies of salvation deltaG in water and chloroform, and the electrostatic potential surface around examined molecules were ab initio calculated by HF method for voriconazole, posaconazole and ravuconazole. These quantities are assumed to be the new determinants correctly describing both solubility and the affinity of biologically active compounds to lipophilic tendency to cross cellular membranes. The values of deltaG were compared to the theoretically and experimentally determined partition coefficients. The calculated values of deltaG and electrostatic potentials appeared to be consistent with these partition coefficients. It leads to conclusion that these theoretically derived parameters deltaG and electrostatic potential could be useful tools for fast and precise classification of chemical substances within the Biopharmaceutics Classification System (BCS).

Background: The definition of addiction is that it is an acquired, strong need to perform a specific activity or continued use of mood alerting substances. Increasing discussion about the development of Internet addiction, which like other addictions, have their roots in depression, impaired assessment esteem and social anxiety shows that it affects all users of the global network, regardless of gender or age. The aim of the study was to assess the impact of social networking on the ongoing behavior of respondents- the first step of a study on the possibility of dependence on social networks., Subjects and Methods: The study was based on an authors questionnaire placed on popular polish websites on February 2013. Questions related to the types and frequency of specific activities undertaken by the private profiles of users., Results: The study involved 221 respondents, 193 questionnaires were filled in completely and correctly, without missing any questions. 83.24% admitted to using social networking sites, 16.76% indicated that they never had their own profile. An overwhelming number of respondents are a member of Facebook (79.17%), specialized portals related to their profession or work were used by only 13.89%, Our-class (6.25%) and Twitter was a primary portal for one person only. Nobody marked a participation in dating services., Conclusion: There is a big difference between the addiction to the Internet and addictions existing within the Internet; the same pattern applies to social networking. There is a need to recognize the "social networking" for a particular activity, irrespective of Facebook, Twitter and Nasza-Klasa, which are commercial products.

In this study, the kinetic isotope effects and solvent isotope effects in the reaction of the deamination of [(1R)-(2)H ] putrescine--catalysed by enzyme diamine oxidase (EC 1.4.3.6)--were determined using a non-competitive spectroscopic method. Putrescine, stereospecifically labelled with deuterium, was obtained by enzymatic decarboxylation of l-ornithine that was carried out in a fully deuteriated incubation medium.

Background: Modern media have a huge impact on eating habits, which result in pathologies among young people, especially females. Fashion models have become a pattern for ordinary women, however the difference in a mean weight between these two groups is increasing. The aim of the study was to assess the correlation between school students' results in EAT-26 self-report questionnaire and their daily diet behavior and to analyze the occurrence of eating disorders among students in schools of the region of Silesia in Poland., Subjects and Methods: Our survey was based on the EAT-26 (Eating Attitude Test) and Behavioral Four Questions Test. 150 questionnaires were given to students of Secondary Schools, 116 were filled out correctly. The interviewed group consisted of 85 girls and 31 boys. Average age was 17.24±1.18. All of the data were analyzed statistically., Results: Average BMI was 21,06; half of the recruited subjects had correct BMI whereas over 40% of them presented underweight. Within 116 responses from the EAT-26 nearly 93% belonged to the group with a small risk of the development of future eating disorders. Nevertheless, over 6% of the survey participants were in the group at high risk. In the Behavioral Four Questions Test 10% participants turned out to be in the compulsive overeating group, other ones in the group of a probable development of anorexia (7%) or bulimia nervosa (3%)., Conclusions: There is a strong correlation between particular daily activities and the score achieved in EAT-26, however there is no significant correlation between the calculated BMI and EAT-26 results. Specific socio-cultural factors are faced by public services. They refer both to health promotion programs and the organization of spare time spent by teenagers outside schools.

Post transplant lymphoprolipherative disorder (PTLD) is a very severe, life threatening complication after organ transplantation. In adults after kidney transplantation the frequency of this complication is 0.3-3%; whereas in children 0.4-10%. Epstein-Barr virus infection discovered in 90% of patients, as a result of immunosuppression, plays an important role in the PTLD etiology. There is a coincidence between heavy immunosuppression, especially its cumulative doses, and the increased risk of lymphoprolipherative disorder. The clinical manifestation of PTLD may present as: mononucleosis like syndrome (fever, pharyngitis, tonsillar enlargement, lymphadenopathy), or as a rapid progressive course presenting with severe acidosis, lymphadenopathy, dysfunction of graft and other internal organs, where infiltration may mimic tumour like appearance. The present paper discusses methods and effectiveness of PTLD treatment, which aims to restore effectiveness of the immunological system, elimination of neoplastic cells and EBV.

Cyclomaltohexaose (α-cyclodextrin) and cyclomaltoheptaose (β-cyclodextrin) as well as their four methyl ether derivatives, that is, hexakis(2,3-di-O-methyl)cyclomaltohexaose, hexakis(2,3,6-tri-O-methyl)cyclomaltohexaose, heptakis(2,3-di-O-methyl)cyclomaltoheptaose, and heptakis(2,3,6-tri-O-methyl)cyclomaltoheptaose were investigated as the additives in the course of enzymatic decomposition of l-phenylalanine catalyzed by phenylalanine ammonia-lyase. Only a few of those additives behaved like classical inhibitors of the enzymatic reaction under investigation because the values of the Michaelis constants that were obtained, as well as the maximum velocity values depended mostly atypically on the concentrations of those additives. In most cases cyclodextrins caused mixed inhibition, both competitive and noncompetitive, but they also acted as activators for selected concentrations. This atypical behaviour of cyclodextrins is caused by three different and independent effects. The inhibitory effect of cyclodextrins is connected with the decrease of substrate concentration and unfavourable influence on the flexibility of the enzyme molecules. On the other hand, the activating effect is connected with the decrease of product concentration (the product is an inhibitor of the enzymatic reaction under investigation).
