This research aimed at evaluating the effects of capping on the mitigation of impacts generated by a closed unlined landfill in São Carlos, SP, Brazil. Physicochemical and microbiological analyses (16S rRNA sequencing) of buried solid waste samples were performed, in capped and uncapped areas. Even though leachate pockets could still be encountered in capped areas, the capping construction reduced oxygen availability and created more reducing conditions, propitiating the development of sulfate-reducing bacteria and possibly contributing to the precipitation of the metals Pb, Cd, Ni, Co, As, and Zn as metal sulfides, causing their immobilization. The microbial populations adapted to the anaerobic conditions created under capped zones belonged to the phyla Firmicutes, Chloroflexi and Euryarchaeota and the genera Methanosaeta, Hydrogenispora, Smithella,and Gelria. Differently, the phyla Acidobacteria, Proteobacteria, Bacteroidetes, and Actinobacteria were more abundant in samples from the uncapped zones, in which the abundance of different genera varied homogeneously. Methanogenic activity was not impaired by the intervention measure, as assessed by the Specific Methanogenic Activity (SMA). Capping of old unlined landfills brings benefits to the immobilization of metals and does not impair microbial degradation, being effective for the mitigation of impacts on soils and water resources.

Carbonaceous chondrites are considered to have originated from C-type asteroids and represent some of the most primitive material in our solar system. Furthermore, since carbonaceous chondrites can contain significant quantities of volatile elements, they may have played a crucial role in supplying volatiles and organic material to Earth and other inner solar system bodies. However, a major challenge of unravelling the volatile composition of chondritic meteorites is distinguishing between which features were inherited from the parent body, and what may be a secondary feature attributable to terrestrial weathering. In December 2020, the Hayabusa2 mission of the Japan Aerospace Exploration Agency (JAXA) successfully returned surface material from the C-type asteroid (162173) Ryugu to Earth. This material has now been classified as closely resembling CI-type chondrites, which are the most chemically pristine meteorites. The analysis of material from the surface of Ryugu therefore provides a unique opportunity to analyse the volatile composition of material that originated from a CI-type asteroid without the complications arising from terrestrial contamination. Given their highly volatile nature, the noble gas and nitrogen inventories of chondrites are highly sensitive to different alteration processes on the asteroid parent body, and to terrestrial contamination. Here, we investigate the nitrogen and noble gas signature of two pelletized grains collected from the first and second touchdown sites (Okazaki et al., 2022a), to provide an insight into the formation and alteration history of Ryugu. The concentration of trapped noble gas in the Ryugu samples is greater than the average composition of previously measured CI chondrites and are primarily derived from phase Q, although a significant contribution of presolar nanodiamond Xe-HL is noted. The large noble gas concentrations coupled with a significant contribution of presolar nanodiamonds suggests that the Ryugu samples may represent some of the most primitive unprocessed material from the early solar system. In contrast to the noble gases, the abundance of nitrogen and δ15N composition of the two Ryugu pellets are lower than the average CI chondrite value. We attribute the lower nitrogen abundances and δ15N measured in this study to the preferential loss of a 15N-rich phase from our samples during aqueous alteration on the parent planetesimal. The analyses of other grains returned from Ryugu have shown large variations in nitrogen concentrations and δ15N indicating that alteration fluids heterogeneously interacted with material now present on the surface of Ryugu. Finally, the ratio of trapped noble gases to nitrogen is higher than CI chondrites, and is closer to refractory phase Q and nanodiamonds. This indicates that Ryugu experienced aqueous alteration that led to the significant and variable loss of nitrogen, likely from soluble organic matter, without modification of the noble gas budget, which is primarily hosted in insoluble organic matter and presolar diamonds and is therefore more resistant to aqueous alteration., Geochimica et Cosmochimica Acta, 345, ISSN:0016-7037, ISSN:1872-9533

Using the Response Surface Methodology (RSM) and Rotational Central Composite Design (RCCD), this study evaluated the removal of DCF under denitrifying conditions, with ethanol as cosubstrate, in batch reactors, being 1 L Erlenmeyer flasks (330 mL of reactional volume) containing Dofing medium and kept under agitation at 130 rpm and incubated at mesophilic temperature (30 °C). It considered the individual and multiple effects of the variables: nitrate (130 - 230 mg NO3− L−1), DCF (60–100 µg DCF L−1) and ethanol (130 - 230 mg EtOH L−1). The highest drug removal efficiency (17.5%) and total nitrate removal were obtained at 176.6 ± 4.3 mg NO3 −L−1, 76.8 ± 3.7 µg DCF L−1, and 180.0 ± 2.5 mg EtOH L−1. Under such conditions, the addition of ethanol and nitrate was significant for the additional removal of diclofenac (p > 0.05). The prevalence of Rhodanobacter, Haliangium and Terrimonas in the inoculum biomass (activated sludge systems) was identified through the 16S rRNA gene sequencing. The potential of these genera to remove nitrate and degrade diclofenac was inferred, and the main enzymes potentially involved in this process were α-methylacyl-CoA racemase, long-chain fatty acid-CoA ligase, catalases and pseudoperoxidases.

SUMMARYDue to the post-mitotic nature of skeletal muscle fibers, adult muscle maintenance relies on dedicated muscle stem cells (MuSCs). In most physiological contexts, MuSCs support myofiber homeostasis by contributing to myonuclear accretion, which requires a coordination of cell-type specific events between the myofiber and MuSCs. Here, we addressed the role of the kinase AMPKα2 in the coordination of these events supporting myonuclear accretion. We demonstrate that AMPKα2 deletion impairs skeletal muscle regeneration. Throughin vitroassessments of MuSC myogenic fate and EdU-based cell tracing, we reveal a MuSC-specific role of AMPKα2 in the regulation of myonuclear accretion, which is mediated by phosphorylation of the non-metabolic substrate BAIAP2. Similar cell tracingin vivoshows that AMPKα2 knockout mice have a lower rate of myonuclear accretion during regeneration, and that MuSC-specific AMPKα2 deletion decreases myonuclear accretion in response to myofiber contraction. Together, this demonstrates that AMPKα2 is a MuSC-intrinsic regulator of myonuclear accretion.

Background Rapid reduction of heart rate (HR) is the most evident physiological response of vagal nerve stimulation (VNS). Since HR reduction is the most potent factor to decrease myocardial oxygen consumption rate (MVO2), the appropriate VNS can exert cardio-protective effects. It is also known that VNS reduces inflammation, oxidative stress, and sympathetic overload. In addition, the VNS during ischemia-reperfusion is known to attenuate myocardial damage by studies in various animal species. Despite the presence of preclinical evidence of VNS benefits, the lack of the device has limited the translation of this technology to clinical practice. We have recently developed an intravenous VNS catheter (JOHAKU, Neuroceuticals Inc.) that can stimulate the right vagal nerve via superior vena cava (SVC) (Figure 1) on temporary basis. Purpose We aimed to confirm the feasibility of JOHAKU as a device to modulate heart rate and MVO2 rapidly by a canine experiment. Methods In eight beagle dogs, JOHAKU was inserted from the right femoral vein and placed at the SVC level. The stimulation intensity was adjusted to 10–20 V (20 Hz). We simultaneously recorded electrocardiogram and intraarterial blood pressure (BP). In three of eight dogs, we measured the left anterior descending coronary artery flow and oxygen saturations of arterial and coronary sinus blood to calculate MVO2. We compared HR, BP, and MVO2 during JOHAKU stimulation to ones at baseline. Results As shown in Figure 2, JOHAKU attenuated HR immediately after stimulation. Compared with baseline, JOHAKU significantly reduced HR (baseline: 135±13 vs. 5 min on stimulation: 107±13 bpm, p Conclusion JOHAKU rapidly attenuated cardiac metabolism burden via the rapid HR reduction. The controllability of HR by JOHAKU without affecting BP enables us to apply the VNS even for patients with hemodynamic instability, such as heart failure and acute myocardial infarction. Background Epidemiology of non-pulmonary vein (PV) triggers of atrial fibrillation (AF) is not fully known. Purpose This study aimed to clarify the prevalence and distribution of non-PV triggers in real-world clinical settings of AF catheter ablation. Methods One-thousand and twenty patients undergoing AF ablations were retrospectively analyzed (mean age 65 years old, 702 males (69%), 506 paroxysmal and 514 non-paroxysmal). Induction and observation of AF triggers were attempted using intravenous isoproterenol/adenosine triphosphate and repeated direct current cardioversion during AF in each session. Documentable non-PV AF triggers were characterized in the studied population. Premature atrial contractions that did not initiate AF were excluded. Results A hundred and twenty-six non-PV triggers were documented in 108 patients (10.6%). Non-PV trigger was documented in 6.3% of 1st session cases, whereas 30.9% of recurrent cases undergoing multiple sessions (p Conclusions Prevalence of non-PV trigger in cases undergoing multiple sessions is extremely high, suggesting a particular importance of non-PV trigger targeting in patients with recurrent AF undergoing 2nd or 3rd ablation sessions. Progressive nature of AF with newly generated AF triggers should be under consideration. Distributions of non-PV triggers are largely different between paroxysmal and non-paroxysmal AF. Samples of the carbonaceous asteroid Ryugu were brought to Earth by the Hayabusa2 spacecraft. We analyzed 17 Ryugu samples measuring 1 to 8 millimeters. Carbon dioxide–bearing water inclusions are present within a pyrrhotite crystal, indicating that Ryugu’s parent asteroid formed in the outer Solar System. The samples contain low abundances of materials that formed at high temperatures, such as chondrules and calcium- and aluminum-rich inclusions. [Context] Methanol masers of Class I (collisionally pumped) and Class II (radiatively pumped) have been studied in great detail in our Galaxy in a variety of astrophysical environments such as shocks and star-forming regions and are they are helpful to analyze the properties of the dense interstellar medium. However, the study of methanol masers in external galaxies is still in its infancy., [Aims] Our main goal is to search for methanol masers in the central molecular zone (CMZ; inner 500 pc) of the nearby starburst galaxy NGC 253., [Methods] Covering a frequency range between 84 and 373 GHz (λ = 3.6–0.8 mm) at high angular (1.″6 ∼ 27 pc) and spectral (∼8–9 km s−1) resolution with ALCHEMI (ALMA Comprehensive High-resolution Extragalactic Molecular Inventory), we have probed different regions across the CMZ of NGC 253. In order to look for methanol maser candidates, we employed the rotation diagram method and a set of radiative transfer models., [Results] We detect for the first time masers above 84 GHz in NGC 253, covering an ample portion of the J−1 → (J − 1)0 − E line series (at 84, 132, 229, and 278 GHz) and the J0 → (J − 1)1 − A series (at 95, 146, and 198 GHz). This confirms the presence of the Class I maser line at 84 GHz, which was already reported, but now being detected in more than one location. For the J−1 → (J− 1)0 − E line series, we observe a lack of Class I maser candidates in the central star-forming disk., [Conclusions] The physical conditions for maser excitation in the J−1 → (J − 1)0 − E line series can be weak shocks and cloud-cloud collisions as suggested by shock tracers (SiO and HNCO) in bi-symmetric shock regions located in the outskirts of the CMZ. Phosphorus (P) is a crucial element for life given its central role in several biomolecules. P-bearing molecules have been discovered in different regions of the Milky Way, but not yet towards an extragalactic environment. We have searched for P-bearing molecules towards the nearby starburst Galaxy NGC 253. Using observations from the ALMA Comprehensive High-resolution Extragalactic Molecular Inventory (ALCHEMI) project, we used the MADCUBA package to model the emission of P-bearing molecules assuming Local Thermodynamic Equilibrium (LTE) conditions. We have also performed a non-LTE analysis using SpectralRadex. We report the detection of a P-bearing molecule, phosphorus nitride (PN), for the first time in an extragalactic environment, towards two giant molecular clouds (GMCs) of NGC 253. The LTE analysis yields total PN beam-averaged column densities $N$=(1.20$\pm$0.09)$\times$10$^{13}$ cm$^{-2}$ and $N$=(6.5$\pm$1.6)$\times$10$^{12}$ cm$^{-2}$, which translate into abundances with respect to H$_2$ of $\chi$=(8.0$\pm$1.0)$\times$10$^{-12}$ and $\chi$=(4.4$\pm$1.2)$\times$10$^{-12}$. We derived a low excitation temperature of $T_{\rm ex}$=(4.4$\pm$1.3) K towards the GMC with the brightest PN emission, which indicates that PN is sub-thermally excited. The non-LTE analysis results in column densities consistent with the LTE values. We have also searched for other P-bearing molecules (PO, PH$_{3}$, CP and CCP), and upper limits were derived. Electrical current-voltage (I x V) analysis found the percolation threshold through the sample surface to be between 0.6 and 0.8 vol%. Below the percolation threshold concentration (p(c)), the effect of the Coulomb potential is accentuated on the CB dispersion, while in the region over p(c), the London-van der Waals potential is important. The repeatability in the piezoresistive behavior was observed under the application of several loading cycles. The gauge factors obtained were 4.9 and 2.0 for samples with 0.8 and 1.0 vol% of CB, respectively. The results indicate that the material can be used as a piezoresistive sensor. Chrysanthemum is one of the most important ornamental plants. Mutation breeding is an important tool in breeding of chrysanthemum, and many cultivars have been produced through mutation breeding by using radiation such as gamma ray, etc. Although ion-beam irradiation is now widely used as a mutagen in the mutation breeding of plants, synchrotron light, another form of radiation, has not been used as a mutagen. Therefore, this study was conducted to clarify whether synchrotron light can be used as a new mutagen in chrysanthemum breeding. Five cultivars of spray chrysanthemum were subjected to synchrotron-light irradiation. As a result of the irradiation, the survival rate decreased as the absorbed dose increased. Flower-colour mutations were observed in three cultivars. There were varietal differences in survival rates and the frequencies of flower-colour mutation. Flowers of the mutants showed various colour patterns such as a deeper and paler colour than the original flowers. In the mutant of a cultivar with a marginal picotee, the proportions of picotee changed. A moderate absorbed dose of synchrotron-light irradiation for flower-colour mutation was found to be 11 to 23 Gy. These results indicate that synchrotron light can be used as a new mutagen in the mutation breeding of chrysanthemum.

Martín, S., et al., [Context] The interstellar medium is the locus of physical processes affecting the evolution of galaxies which drive or are the result of star formation activity, supermassive black hole growth, and feedback. The resulting physical conditions determine the observable chemical abundances that can be explored through molecular emission observations at millimeter and submillimeter wavelengths. [Aims] Our goal is to unveiling the molecular richness of the central region of the prototypical nearby starburst galaxy NGC 253 at an unprecedented combination of sensitivity, spatial resolution, and frequency coverage. [Methods] We used the Atacama Large Millimeter/submillimeter Array (ALMA), covering a nearly contiguous 289 GHz frequency range between 84.2 and 373.2 GHz, to image the continuum and spectral line emission at 1.6″(∼28 pc) resolution down to a sensitivity of 30 - 50 mK. This article describes the ALMA Comprehensive High-resolution Extragalactic Molecular Inventory (ALCHEMI) large program. We focus on the analysis of the spectra extracted from the 15″ (∼255 pc) resolution ALMA Compact Array data. [Results] We modeled the molecular emission assuming local thermodynamic equilibrium with 78 species being detected. Additionally, multiple hydrogen and helium recombination lines are identified. Spectral lines contribute 5 to 36% of the total emission in frequency bins of 50 GHz. We report the first extragalactic detections of C2H5OH, HOCN, HC3HO, and several rare isotopologues. Isotopic ratios of carbon, oxygen, sulfur, nitrogen, and silicon were measured with multiple species. [Concluison] Infrared pumped vibrationaly excited HCN, HNC, and HC3N emission, originating in massive star formation locations, is clearly detected at low resolution, while we do not detect it for HCO+. We suggest high temperature conditions in these regions driving a seemingly "carbon-rich"chemistry which may also explain the observed high abundance of organic species close to those in Galactic hot cores. The Lvib/LIR ratio was used as a proxy to estimate a 3% contribution from the proto super star cluster to the global infrared emission. Measured isotopic ratios with high dipole moment species agree with those within the central kiloparsec of the Galaxy, while those derived from 13C/18O are a factor of five larger, confirming the existence of multiple interstellar medium components within NGC 253 with different degrees of nucleosynthesis enrichment. The ALCHEMI data set provides a unique template for studies of star-forming galaxies in the early Universe.
