The purpose of this work was to determine the tablet content and carry out the dissolving test of expired and non-expired tablet as well as establish a simple, selective, linear, precise, accurate and sensitive RP-HPLC technique for the analysis of carvedilol tablets. The experiment was carried out using a mobile phase consisting of 20 mM phosphate buffer (PB) pH 2.5: acetonitrile (ACN) 55:45 (V/V) at a pH of 2.5 using a column (Oyster ODS 3,150-4.6 mm, 5m) and a L6 6AD pump. The sample had a volume of 20 µL, and it was injected at room temperature for a run period of 6 minutes at a flow rate of 1.0 mL min-1. Detection was carried out at 240 nm using an ultraviolet detector. The retention time of the sample was 2.92 minutes. At 37 °C ± 0.5 °C, the preliminary dissolution tests were performed on an Electro Lab dissolution tester. The results of this analysis revealed that there was no distinct shift in the amount of drug present. The percentage of non-expired tablets discovered in the test was 94.67 %, whereas the percentage of expired tablets was 91.71 %. [ABSTRACT FROM AUTHOR]

The present research work is focussed on the development of FT-IR Spectrophotometric method for estimation and validation of ranitidine (RANI) and dicyclomine (DICY). It is simple, fast, accurate, cost efficient and reproducible Spectrophotometric method, developed for the estimation of RANI and DICY. The wave numbers selected were in range of 1361 cm-1(Nitro) for RANI and 1716 cm-1 (Ester) for DICY. The linearity for these drug at the selected wave numbers lies between 3-18 (% w/w) for RANI and 0.2- 1.2 (% w/w) for DICY respectively, with correlation coefficient of 0.998 for RANI and 0.998 for DICY. The accuracy and precision of the method were determined and validated according to ICH guidelines. The method has good reproducibility with % RSD less than two. A simple, accurate and precise UV spectrometric method has been developed for the simultaneous determination of valsartan and hydrochlorothiazide in tablet dosage form. Spectra of valsartan and hydrochlorothiazide in methanol and water (50:50 V/V) show λ max at 250.0 nm and 271.4 nm, respectively. Valsartan and hydrochlorothiazide are subjected to various stress conditions like acid, alkali, thermal and photolytic degradation. Beer's law was obeyed in concentration range of 4-24 μg mL-1 for valsartan and 0.5-3 μg mL-1 for hydrochlorothiazide at their respective wavelengths. The proposed method was successfully applied to tablet dosage form for determination of both drugs. The percentage recovery of valsartan and hydrochlorothiazide were found to be 100.19 % and 99.51 %, respectively. A new method of analysis with reverse phase chemistry was designed and developed. Validation for method of analysis was performed for its intended use to calculate assay and content uniformity of drug substance sitagliptin, metformin and empagliflozin in the drug products. The method has a run time of 10 min on X-bridge C18 column having 250 mm length, 4.6 mm internal diameter and Particle Size of 5 µm, by the use of 0.1% Trifluoroacetic acid Buffer 40%: Methanol 40%: Acetonitrile 20% ratio as constituent composition in the proposed mobile phase and chromatography run at wavelength of 224 nm. The retention time of Metformin, Empagliflozin and Sitagliptin, were 3.383, 5.571 and 6.429 min respectively. International Conference on Harmonization guideline was referred for validation. The method showed adequate sensitivity for precision, linearity and accuracy parameter (between the range 25-75 µg/mL, 250-750 µg/mL and 2.5-7.5 µg/mL for sitagliptin, metformin and empagliflozin respectively). The percentage recoveries obtained for sitagliptin, metformin and empagliflozin are in the range of 98.0--102.0%. A simple, accurate, precise and validated FTIR method was developed for the determination of pregabalin (PRG) and methylcobalamin (MCA) in bulk as well as in tablet dosage form. The drug was analyzed by FTIR spectrophotometer with DRIFT sampling technique. A wavenumber range 1660-1600 cm-1 (-COOH) and 3400-3250 cm-1(-CONH2) was selected for pregabalin and methylcobalamin, respectively. The method was found to be linear over the range of PRG 1-6 % w/w and MCA 0.2-1.2 % w/w with a good regression coefficient (r²) of PRG 0.9960 and MCA 0.9976. The percent recovery of pregabalin and methylcobalamin in marketed tablet dosage form was in the range of 98.25 - 99.00 % and 98.62 - 99.60 %, respectively. The LOD & LOQ were found 1.4003 & 4.2434 of PRG and 0.2027& 0.6142 of MCA respectively.
