Nowadays, Li-S batteries are facing many thorny challenges like volume expansion and lithium dendrites on the road to commercialization. Due to the peculiarity of complete lithiation and the capability to match non-lithium anodes, Li2S-based Li-S batteries have attracted more and more attention. Nevertheless, the same notorious shuttle effect of polysulfides as in traditional Li-S batteries and the poor conductivity of Li2S lead to sluggish conversion reaction kinetics, poor Coulombic efficiency, and cycling performance. Herein, we propose the interconnected porous carbon skeleton as the host, which is modified by an atomically dispersed Mn catalyst as well as O, N atoms (named as ON-MnPC) via the melt salt method, and introduce the Li2S nanosheet into the carbon host with poly(vinyl pyrrolidone) ethanol solution. It has been found that the introduction of O, N to bind with Mn atoms can endow the nonpolar carbon surface with ample unsaturated coordination active sites, restrain the shuttle effect, and enhance the diffusion of Li+ and accelerate the conversion reaction kinetics. Besides, due to the ultra-high catalyst activity of atomically dispersed Mn catalysts, the Li2S/ON-MnPC cathode shows good electrochemical performance, e.g., an initial capacity of 534 mAh g-1, a capacity of 514.18 mAh g-1 after 100 cycles, a high retention rate of 96.23%, and a decay rate of 0.04% per cycle. Hence, use of atomically dispersed Mn catalysts to catalyze the chemical conversion reactions of polysulfides from multiple dimensions is a significant exploration, and it can provide a brand-new train of thought for the development and commercialization of the economical, high-performance Li2S-based Li-S batteries.

Digital holography has been frequently used to measure the micro-deformation in mechanical tests due to its full-field measurement with high resolution and accuracy. To measure dynamic three-dimensional absolute displacements without a known reference displacement, a new technique based on the combination of off-axis multiplexed digital holography and stereo photogrammetry is proposed. Under the illumination of two different wavelength lasers along various directions, two off-axis multiplexed holograms recorded by the dual-camera system are used to extract four phase maps with different sensitivity vectors simultaneously. Meanwhile, the variation of sensitivity vectors and registration of phase maps are carried out by the object shape measured by the dual-camera system. By the four registered phases with four varying sensitivity vectors, three-dimensional absolute displacements can be determined. The feasibility of our method is well demonstrated by a quantitative experiment and finite element analysis, and the dynamic measurement of a resistor undergoing thermal expansion is presented.

The critical role of circular RNAs (circRNAs) in osteoporosis (OP) has been highlighted. We tried to explore the role of circPVT1 in OP in relation to microRNA-30d-5p (miR-30d-5p) and ITGB3.After bone marrow collection, bone marrow mesenchymal stem cells (BMSCs) were isolated and identified. Then, Pearson coefficient was used to analyze the correlation among circPVT1, miR-30d-5p and ITGB3, and the binding sites were predicted and verified. Gain- and loss-of function assays in circPVT1, miR-30d-5p and ITGB3 were performed to analyze their effect on osteogenic differentiation of BMSCs.The osteogenic differentiation of BMSCs from OP patients was significantly decreased, and reduced circPVT1 expression was found in the BMSCs from OP patients. Overexpression of circPVT1 stimulated the formation of calcified nodules, increased alkaline phosphatase activity, and enhanced the expression of osteogenic marker genes in the BMSCs from OP patients. Additionally, circPVT1 expression was negatively correlated with miR-30d-5p, and miR-30d-5p was negatively correlated with ITGB3 in OP patients. Mechanically, circPVT1 regulated the osteogenic differentiation potential of BMSCs by relieving the inhibition of miR-30d-5p on ITGB3 through the competitive endogenous RNA mechanism.Our study highlighted a circPVT1/miR-30d-5p/ITGB3 axis in regulating osteogenic differentiation potential of BMSCs from OP patients.

In this paper, a new method, Moment-Stiffness Method (MSM), is proposed for measuring the edge and depth of internal defects which can be applied to any structures with known stiffness. MSM is based on the relation of the differences of displacements between flawed and flawless areas caused by the change of stiffness. Theoretical, numerical and experimental results are presented. In the method, digital speckle pattern interferometry (DSPI) is used to measure the out-of-plane displacement of a plate with an internal defect. In the detection of the first specimen, the distributions of first- and second-order radial derivatives of the out-of-plane displacement are obtained by the radial numerical differential. Then the full edge and depth of the defect is accurately determined using MSM. All the results indicate that the proposed method is an effective tool to measure internal defects precisely.

To explore the effect and mechanism of Jiaotai Pill (交泰丸, JTW) on intestinal mucosal damage in rats with chronic partial sleep deprivation (PSD). Obesity resistant (OR) rats were selected, and underwent 4 h PSD by being exposed to environmental noise for 4 weeks. During the whole PSD period, JTW and estazolam were orally given to the rats respectively in the treating groups. Plasma concentration of lipopolysaccharide (LPS) which is the marker of gut-origin endotoxemia was examined. Intestinal morphology changes were observed by optical microscopy. The protein expression of occludin (Ocln) in the intestine was measured by immunofluorescence technique and Western blot. Digital speckle pattern interferometry (DSPI) is a well-established and widely used optical measurement technique for obtaining qualitative as well as quantitative measurements of objects deformation. The simultaneous measurement of an object's surface displacements in three dimensions using DSPI is of great interest. This paper presents a triple-optical-path DSPI based method for the simultaneous and independent measurement of three-dimensional (3D) displacement fields. In the proposed method, in-plane speckle interferometers with dual-observation geometry and an out-of-plane interferometer are optimally combined to construct an integrated triple-optical-path DSPI system employing the phase shift technique, which uses only a single laser source and three cameras. These cameras are placed along a single line to synchronously capture real-time visible speckle fringe patterns in three dimensions. In addition, a pre-filtering method based on the fast discrete curvelet transform (FDCT) is utilized for denoising the obtained wrapped phase patterns to improve measurement accuracy. Finally, the simultaneous measurement of the 3D displacement fields of a simple beam and a composite laminated plate respectively subjected to three-point and single-point bend loading are investigated to validate the feasibility and effectiveness of the proposed method.

Berberine (BBR) is an isoquinoline alkaloid extracted from Rhizoma coptidis and has been used for treating type 2 diabetes mellitus (T2DM) in China. The development of T2DM is often associated with insulin resistance and impaired glucose uptake in peripheral tissues. In this study, we examined whether BBR attenuated glucose uptake dysfunction through the cholinergic anti-inflammatory pathway in HepG2 cells. Cellular glucose uptake, quantified by the 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)-amino]-2-deoxy-D-glucose (2-NBDG), was inhibited by 21% after HepG2 cells were incubated with insulin (10(-6) mol/L) for 36 h. Meanwhile, the expression of alpha7 nicotinic acetylcholine receptor (α7nAChR) protein was reduced without the change of acetylcholinesterase (AChE) activity. The level of interleukin-6 (IL-6) in the culture supernatant, the ratio of phosphorylated I-kappa-B kinase-β (IKκβ) Ser181/IKKβ and the expression of nuclear factor-kappa B (NF-κB) p65 protein were also increased. However, the treatment with BBR enhanced the glucose uptake, increased the expression of α7nAChR protein and inhibited AChE activity. These changes were also accompanied with the decrease of the ratio of pIKKβ Ser181/IKKβ, NF-κB p65 expression and IL-6 level. Taken together, these results suggest that BBR could enhance glucose uptake, and relieve insulin resistance and inflammation in HepG2 cells. The mechanism may be related to the cholinergic anti-inflammatory pathway and the inhibition of AChE activity.

Due to limits in the properties of digital cameras, in-line digital holography is commonly used to take full advantage of the sampling space of the camera. To realize the dynamic high-resolution measurement of in-line digital holography, dual-camera dynamic in-line digital holography is proposed. By means of a two-step phase-shifting cepstrum algorithm and a dual-camera parallel phase-shifting recording optical path, the complex amplitude of the object wave can be reconstructed without its zero-order and conjugate terms. Meanwhile, a novel spherical wave interference calibration method is also developed for the dual-camera recording system, and image correction is carried out via rotation, translation, and diffraction, with an average error of phase correction of 0.1107 rad. Finally, the feasibility and effectiveness of the proposed technique is well demonstrated by a practical application of dynamic temperature field measurement in a transparent medium.

A new, efficient switching median–mean filter is proposed to remove high density impulse noise from digital images. The proposed method consists of detection and filtering stages. The pixels that are labeled noise-free in the detection stage remain unchanged and the noisy-pixels are replaced by the reference image based the proposed filter with 3 × 3 window. The simulation results show that the proposed filter outperforms some existing methods, both in vision and quantitative measurements. Moreover, our method is not only efficient to remove high density impulse noise and preserve the details in the image, but also possess a short processing time.

In this paper, an effective switching morphology-mean filter is proposed to remove impulse noise from digital images. The proposed method consists of two stages. The first stage is to detect the impulse noise in an image. The second stage is to construct adaptive structuring elements and remove the impulse noise. In this stage, the “noise-pixels” are filtered out by using the morphology-mean filter, and the “noise-free pixels” are copied directly to the output image. The proposed filter uses the window of size 3 × 3 pixels. Simulation results show that the proposed method outperforms some existing algorithms for high-density impulse noise, both in vision and quantitative measurements. Moreover, our method can preserve effectively the details in an image, and has less processing time.

Background Obese subjects have been considered to be in a state of chronic, low-grade systemic inflammation. Excess fat accumulation and persistent inflammation may promote renal dysfunction, to cause chronic kidney disease (CKD) and even end-stage kidney failure. Coptidis Rhizoma is a classical traditional Chinese herb well known for its hypoglycemic and hypolipidemic properties. The mechanism is partially associated with its anti-inflammatory effect. However, this effect is rarely investigated in obesity and obesity-related glomerulopathy (ORG). Purpose The current study was designed to evaluate the effect of Coptidis Rhizoma on ORG. It also aimed to determine whether this renal protection effect of Coptidis Rhizoma was related to the inhibition of NLRP3 inflammasome in ORG. Methods Coptidis Rhizoma concentrated granules were prepared and the main components were identified by 3D-High Performance Liquid Chromatography (3D-HPLC) assay. The animal model of early stage ORG was established in obesity-prone (OP) rats by high protein and high fat diet feeding for 12 weeks. The treatment with Coptidis Rhizoma at different dosages was administered by intragastric infusion simultaneously. Then body weight, kidney weight, plasma lipid profiles, 24 h urine protein/albumin content and kidney histology were measured. Inflammatory biomarkers were examined both in the rat plasma and renal cortex. The gene expressions of NLRP3 inflammasome complex and NF-κB in renal tissues were also measured. Results Coptidis Rhizoma alleviated dyslipidemia and reduced the renal weight of the rats with ORG. Meanwhile, urinary albumin to creatinine ratio and creatinine clearance rate were significantly improved. Coptidis Rhizoma also attenuated glomerular hypertrophy, mesangial hyperplasia, and effacement of podocyte foot in renal tissues of ORG rats. In addition, Coptidis Rhizoma intervention decreased the levels of proinflammatory cytokines (TNF-α, IL-1β, IL-6, and IL-18) both in plasma and renal tissue. The gene expression of NLRP3 inflammasome was down-regulated and NF-κB activity was also inhibited by Coptidis Rhizoma in renal tissues of ORG rats. Conclusion Coptidis Rhizoma can ameliorate early renal damage in ORG rats and the mechanisms appear to be related to the inhibition of NLRP3 inflammasome complex.

This study aims to explore the effect and mechanism of Jiao-tai-wan (JTW) on systemic and tissue-specific inflammation and insulin resistance in obesity-resistant (OR) rats with chronic partial sleep deprivation (PSD). OR rats with PSD were orally given JTW and Estazolam for 4 weeks. The amount of food intake and metabolic parameters such as body weight increase rate, fasting plasma glucose (FPG), fasting insulin (FINS), homeostasis model assessment-insulin resistance (HOMA-IR) and plasma inflammatory markers were measured. The expression levels of circadian proteins cryptochrome 1 (Cryl) and cryptochrome 2 (Cry2) in hypothalamus, adipose and liver tissues were also determined. Meanwhile, the mRNA expression of inflammatory markers, activity of nuclear factor kappa B (NF-κB) p65 protein, as well as the expression levels of insulin signaling pathway proteins in hypothalamus, adipose and liver tissues were measured. Additionally, cyclic adenosine 3', 5'-monophosphate (cAMP) and activity of vasodilator-stimulated phosphoprotein (VASP) in hypothalamus tissue were measured. JTW significantly decreased the body weight increase rate and food intake, ameliorated systemic inflammation and insulin resistance. JTW effectively ameliorated inflammation and increased PI3K/AKT signaling activation in hypothalamus, adipose and liver. Interestingly, all these changes were associated with the up-regulation of circadian gene Cryl and Cry2 protein expression. We also found that in hypothalamus tissue of PSD rats, down-regulation of Cryl and Cry2 activated cAMP/PKA signaling and then led to inflammation, while JTW inhibited this signaling. These results suggested that JTW has the beneficial effect on ameliorating inflammation and insulin resistance in partially sleep-deprived rats by up-regulating Cry expression.

Background: Intestinal mucosal barrier dysfunction plays an important role in the development of diabetes mellitus (DM). Berberine (BBR), a kind of isoquinoline alkaloid, is widely known to be effective for both DM and diarrhea. Here, we explored whether the anti-diabetic effect of BBR was related to the intestine mucosal barrier. Methods and Results: The rat model of T2DM was established by high glucose and fat diet feeding and intravenous injection of streptozocin. Then, those diabetic rats were treated with BBR at different concentrations for 9 weeks. The results showed, in addition to hyperglycemia and hyperlipidemia, diabetic rats were also characterized by proinflammatory intestinal changes, altered gut-derived hormones, and 2.77-fold increase in intestinal permeability. However, the treatment with BBR significantly reversed the above changes in diabetic rats, presenting as the improvement of the high glucose and triglyceride levels, the relief of the inflammatory changes of intestinal immune system, and the attenuation of the intestinal barrier damage. BBR treatment at a high concentration also decreased the intestinal permeability by 27.5% in diabetic rats. Furthermore, BBR regulated the expressions of the molecules involved in TLR4/MyD88/NF-κB signaling pathways in intestinal tissue of diabetic rats. Conclusion: The hypoglycemic effects of BBR might be related to the improvement in gut-derived hormones and the attenuation of intestinal mucosal mechanic and immune barrier damages.

To investigate the effects of berberine (BBR) and cinnamic acid (CA), the main active components in Jiaotai Pill (, JTP), on palmitic acid (PA)-induced intracellular triglyceride (TG) accumulation in NIT-1 pancreatic β cells.Cells were incubated in culture medium containing PA (0.25 mmol/L) for 24 h. Then treatments with BBR (10 μmol/L), CA (100 μmol/L) and the combination of BBR and CA (BBR+CA) were performed respectively. Intracellular lipid accumulation was assessed by Oil Red O staining and TG content was measured by colorimetric assay. The expression of adenosine monophosphate-activated protein kinase (AMPK) protein and its downstream lipogenic and fatty acid oxidation genes, including fatty acid synthase (FAS), acetyl-coA carboxylase (ACC), phosphorylation acetyl-coA carboxylase (pACC), carnitine acyl transferase 1 (CPT-1) and sterol regulating element binding protein 1c (SREBP-1c) were determined by Western blot or real time polymerase chain reaction.PA induced an obvious lipid accumulation and a significant increase in intracellular TG content in NIT-1 cells. PA also induced a remarkable decrease in AMPK protein expression and its downstream targets such as pACC and CPT-1. Meanwhile, AMPK downstream lipogenic genes including SREBP-1c mRNA, FAS and ACC protein expressions were increased. Treatments with BBR and BBR+CA, superior to CA, significantly reversed the above genes changes in NIT-1 pancreatic β cells. However, the synergistic effect of BBR and CA on intracellular TG content was not observed in the present study.It can be concluded that in vitro, BBR and BBR+CA could inhibit PA-induced lipid accumulation by decreasing lipogenesis and increasing lipid oxidation in NIT-1 pancreatic β cells.

A new method for detecting the boundary of an internal defect is proposed in this paper. The proposed method can be used in phase-shifting digital speckle pattern interferometry to detect accurately the boundary of an internal defect based on discontinuity of the second-order gradient of out-of-plane displacement in the direction perpendicular to the defect boundary. Both theoretical analysis and experimental result are presented in this paper.

A method based on the measurement of out-of-plane displacement by the phase-shifting digital speckle pattern interferometry is proposed to detect the edge of internal defects. The out-of-plane displacement is scanned by the scanning method of gradient streamlines first, which can provide paths through the defect edge with the best signal-to-noise ratio everywhere. Then, continuous wavelet transform (CWT) combined with the weighted structural intensity of the wavelet maxima is applied to detect the hidden singularities of the gradient streamlines that form the defect edge later. Moreover, the weighted structural intensity method is optimized to adapt to the detection of the hidden singularity by CWT. The results of theory, simulation, and experiments agree well, and the proposed method is efficient in internal defect detection.

This paper studies the exponential and Butterworth low-pass filtering methods and proposes improved exponential and Butterworth low-pass filters. The improved low-pass filters proposed in the paper can be used effectively for filtering processing of digital speckle interference fringes obtained in digital speckle pattern interferometry. Both theoretical analysis and experimental results are presented in the paper.

Ideal low-pass filtering methods were studied in detail and adaptive elliptical ideal low-pass filters proposed for the first time. The proposed adaptive elliptical ideal low-pass filters can be used effectively for denoising of digital speckle interferograms obtained in phase-shifting digital speckle pattern interferometry. The shape of adaptive elliptical ideal low-pass filters is determined according to the spectrum distribution of interference fringes by using the adaptive method. Theoretical analysis, simulation calculation, and experimental results are presented, and are in good agreement.

Jiao Tai Wan (JTW), a Chinese herbal formula containing Rhizoma Coptidis and Cortex Cinnamomi, has been used for diabetic treatment for many years. The aim of this study was to determine the main components in JTW and to investigate the effects of JTW on hepatic lipid accumulation in diabetic rats and humans. JTW extract was prepared and the main components were assayed by HPLC. An animal model of diabetes mellitus was established and JTW was administered intragastrically. In the clinical study, diabetic patients with poor glycemic control were treated with JTW. Blood glucose and lipid parameters, liver histology, hepatic triglyceride content and lipogenic gene expression were examined. Our data demonstrated that JTW significantly improved hyperglycemia, hyperlipidemia and hepatic lipid accumulation in diabetic rats. This was accompanied by the down-regulation of acetyl coenzyme A carboxylase (ACC) and fatty acid synthase (FAS) protein expressions, and the up-regulation of AMP-activated protein kinase (AMPK) and phosphorylated-ACC (pACC) protein expressions in the liver tissues. Diabetic patients also exhibited decreases in their hepatic triglyceride content. The results suggest that JTW attenuates hepatic lipid accumulation in diabetic rats and humans. These beneficial effects are possibly associated with the inhibition of lipogenic gene expression in the liver.

Hu-Lu-Ba-Wan (HLBW) is a Chinese herbal prescription used to treat kidney deficiency. The aim of this study was to explore the effect and mechanism of HLBW on diabetic nephropathy (DN) in type 2 diabetic rats. The rat model of DN was established by being fed a high-fat diet and intravenous injection of streptozotocin. Then, HLBW decoction was administered for 16 weeks. Blood glucose level, lipid profile, renal function, 24-hour total urinary protein, and albumin content were examined. Renal morphology and superoxide anion levels were evaluated. The activity of nicotinamide-adenine dinucleotide phosphate (NADPH) and protein kinase C-alpha (PKC-α) related genes expression in renal tissue were also determined. Our data demonstrated that HLBW significantly improved hyperglycemia, hyperlipidemia, and proteinuria in diabetic rats compared with those of control group. HLBW also alleviated glomerular expansion and fibrosis, extracellular matrix accumulation and effacement of the foot processes. Additionally, HLBW reduced superoxide anion level, NADPH oxidase activity, the protein and mRNA expressions of p47phox, and the protein expression of phosphorylated PKC-αin renal tissue. These results suggest that HLBW is effective in the treatment of DN in rats. The underlying mechanism may be related to the attenuation of renal oxidative stress via PKC-α/NADPH oxidase signaling pathway.

This paper proposes a new method for detecting the size and centroid of an internal defect based on the Radon transform and morphological operation. This method proposed in the paper can be used in phase-shifting digital speckle pattern interferometry to measure quantitatively the size of the internal defect and to determine accurately the centroid of the internal defect. Theoretical analysis, experimental result, and error calculation are presented in this paper.

Volume-grating digital speckle pattern interferometry used for measuring dynamic out-of-plane displacement fields of a continuously deformed object is proposed in this paper. These continuous out-of-plane displacement fields can be successfully measured by using the method discussed in the paper. A comparison between theoretical and experimental results is presented in the text.

The modern digital speckle pattern interferometry (DSPI) technique is applied for the accurate measurement of the full-field deflection distribution of a bent composite laminates. Two kinds of powerful phase-shifting methods, phase of differences method (PDM) and difference of phases method (DPM), are described briefly and employed in DSPI to quantitatively extract the phase information, respectively. A comparison of the deflection distributions measured by DPM and PDM indicates that the former has a better measurement accuracy than the latter in laminate's static test experiment.

Shape measurement is a significant application of digital image correlation (DIC). An improved method that combines a rotatable plane mirror is proposed to measure the shape of an immovable object. In this method, two images, one before and the other after rotating the plane mirror, are obtained and then in-plane translation which related to the shape of the detected object can be calculated by the use of two-dimensional digital image correlation (2D DIC). The relationship between the in-plane translation and the shape of the object is described. Experimental results show that the proposed method is feasible for shape and distance measurement with high accuracy.

Ethnopharmacological relevance Jinlida granule (JLDG), composed of seventeen Chinese medical herbs, is a widely used Chinese herbal prescription for treating diabetes mellitus. However, the mechanism underlying this effect remains unclear. To determine the main components in JLDG and to explore the effect of JLDG on autophagy and lipid accumulation in NIT-1 pancreatic β cells exposed to politic acid (PA) through AMP activated protein kinase (AMPK) signaling pathway. Materials and methods JLDG was prepared and the main components contained in the granules were identified by ultra performance liquid chromatography (UPLC) fingerprint. Intracellular lipid accumulation in NIT-1 cells was induced by culturing with medium containing PA. Intracellular lipid droplets were observed by Oil Red O staining and triglyceride (TG) content was measured by colorimetric assay. The formation of autophagosomes was observed under transmission electron microscope. The expression of AMPK and phospho-AMPK (pAMPK) proteins as well as its downstream fatty acid metabolism-related proteins (fatty acid synthase, FAS; acetyl-coA carboxylase, ACC; carnitine acyltransferase 1, CPT-1) and autophagy-related genes (mammal target of rapamycin, mTOR; tuberous sclerosis complex 1, TSC1; microtubule-associated protein 1 light chain 3, LC3-II) were determined by Western blot. The expression of sterol regulating element binding protein 1c (SREBP-1c) mRNA was examined by real time PCR (RT-PCR). Results Our data showed that JLDG could significantly reduce PA-induced intracellular lipid accumulation in NIT-1 pancreatic β cells. This effect was associated with increased protein expression of pAMPK and AMPK in NIT-1 cells. Treatment with JLDG also decreased the expression of AMPK downstream lipogenic genes (SREBP-1c mRNA, FAS and ACC proteins) whereas increased the expression of fatty acid oxidation gene (CPT-1 protein). Additionally, JLDG-treated cells displayed a markedly increase in the number of autophagosomes which was accompanied by the down-regulation of mTOR and the up-regulation of TSC1 and LC3-II proteins expression. However, when AMPK phosphorylation was inhibited by Compound C, JLDG supplementation did not exhibit any effect on the expression of these AMPK downstream molecules in NIT-1 cells. Conclusions The results suggest that JLDG could reduce intracellular lipid accumulation and enhance the autophagy in NIT-1 pancreatic β cells cultured with PA. The mechanism is possibly mediated by AMPK activation.

A double whole-field filtering technique to measure pure twist fringe distribution of a deformed object is proposed. In this method, the slope fringes which are contained in the pattern of twist fringes can be completely eliminated from the twist fringes. Thus a pattern of pure twist fringes with a good contrast can be obtained. Theoretical analysis and experimental results are given in this paper.

A double whole-field filtering technique to measure pure curvature fringe distribution of a deformed object is proposed. In this method, the slope fringes which are contained in the pattern of curvature fringes can be completely eliminated from the curvature fringes. Thus the pattern of pure curvature fringes with a good contrast can be obtained. Theoretical analysis and experimental results are given in this paper.
