Abstract

Fusarium circinatum, the causal agent of pine pitch canker (PPC), is currently one of the most important threats of Pinus spp. globally. This pathogen is known in many pine-growing regions, including natural and planted forests, and can affect all life stages of trees, from emerging seedlings to mature trees. Despite the importance of PPC, the global distribution of F. circinatum is poorly documented, and this problem is also true of the hosts within countries that are affected. The aim of this study was to review the global distribution of F. circinatum, with a particular focus on Europe. We considered (1) the current and historical pathogen records, both positive and negative, based on confirmed reports from Europe and globally; (2) the genetic diversity and population structure of the pathogen; (3) the current distribution of PPC in Europe, comparing published models of predicted disease distribution; and (4) host susceptibility by reviewing literature and generating a comprehensive list of known hosts for the fungus. These data were collated from 41 countries and used to compile a specially constructed geo-database. A review of 6297 observation records showed that F. circinatum and the symptoms it causes on conifers occurred in 14 countries, including four in Europe, and is absent in 28 countries. Field observations and experimental data from 138 host species revealed 106 susceptible host species including 85 Pinus species, 6 non-pine tree species and 15 grass and herb species. Our data confirm that susceptibility to F. circinatum varies between different host species, tree ages and environmental characteristics. Knowledge on the geographic distribution, host range and the relative susceptibility of different hosts is essential for disease management, mitigation and containment strategies. Abstract

Downy mildews caused by obligate biotrophic oomycetes result in severe crop losses worldwide. Among these pathogens, Pseudoperonospora cubensis and P. humuli, two closely related oomycetes, adversely affect cucurbits and hop, respectively. Discordant hypotheses concerning their taxonomic relationships have been proposed based on host–pathogen interactions and specificity evidence and gene sequences of a few individuals, but population genetics evidence supporting these scenarios is missing. Furthermore, nuclear and mitochondrial regions of both pathogens have been analyzed using microsatellites and phylogenetically informative molecular markers, but extensive comparative population genetics research has not been done. Here, we genotyped 138 current and historical herbarium specimens of those two taxa using microsatellites (SSRs). Our goals were to assess genetic diversity and spatial distribution, to infer the evolutionary history of P. cubensis and P. humuli, and to visualize genome-scale organizational relationship between both pathogens. High genetic diversity, modest gene flow, and presence of population structure, particularly in P. cubensis, were observed. When tested for cross-amplification, 20 out of 27 P. cubensis-derived gSSRs cross-amplified DNA of P. humuli individuals, but few amplified DNA of downy mildew pathogens from related genera. Collectively, our analyses provided a definite argument for the hypothesis that both pathogens are distinct species, and suggested further speciation in the P. cubensis complex.

We evaluated spray coverage on grape leaves and bunches by a high-volume sprayer (HVS), an air-assisted, low volume knapsack sprayer (LVS), and a motorized knapsack dust applicator modified to spray liquid formulations (common sprayer – CS). At the full development of the vine canopy 1400, 250 and 150 L ha−1 were applied via HVS, CS and LVS, respectively. Sprayer type, leaf or bunch orientation, leaf position and leaf side significantly affected spray coverage (P < 0.001). Mean spray coverage on leaves or bunches was below 21% for CS, and above 40 and 55% for LVS and HVS, respectively. Downy mildew leaf infection was below 5% throughout the season, with a significant effect of sprayer, time and an interaction between sprayer and time (P < 0.001). Grape berry moth damage did not differ between sprayers and the untreated control. The current work suggests that LVS might be a viable alternative to HVS, but further work is needed to establish whether differences in spray coverage affect biological efficacy. Abstract

BACKGROUNDConsumer concerns regarding high-quality produce, free of pesticide residues, direct research towards disease management strategies that minimise or even exclude the use of synthetic chemistries in crop production. The efficacy of a chitosan-based edible coating combined with the acetonic extract of Salvia fruticosa Mill. (ASF) was assessed against the grey mould of table grapes. RESULTSHPLC-SPE-NMR and q-NMR analyses defined major constituents of ASF to be the flavonoids hispidulin, salvigenin and cirsimaritin and the diterpenes carnosic acid, carnosol and the 12-methoxycarnosic acid. The extract was found to be efficacious in reducing spore germination and mycelial growth of Botrytis cinerea in vitro at 10 and 25 degrees C. However, the combination of the ASF with chitosan 1% (w/v; CHIT) significantly improved fungal inhibition. Similarly, in fruit inoculation trials at 10 degrees C, the efficacy of the combined application of the ASF at 500 mg L-1 with CHIT against grey mould was statistically equal to the synthetic fungicide thiabendazole, ranging from 98.4% to 92.7% at 12 and 21 days post-inoculation, respectively. Furthermore, chitosan coating alone and in combination with ASF decreased the rate of fruit weight loss during cold storage, while preserved soluble solids content and titratable acidity. Chitosan-based coatings did not affect quality attributes and the bioactive compounds in table grapes. CONCLUSIONThe combined application of the ASF in the form of an edible coating with chitosan could effectively control B. cinerea without deteriorating quality and physico-chemical properties of grapes. Abstract

Kresoxim-methyl (KM) belongs to the strobilurins, one of the most important classes of agricultural fungicides, displaying a direct effect on physiological and developmental plant processes thus leading to greening of plants via induction of hormonal biosynthesis. The aim of this study was to examine the effect of KM pre-treatment on Medicago truncatula plants (ecotype Jemalong A17) subjected to drought and salinity conditions, evaluating key physiological, biochemical and molecular parameters. Foliar application with 10-8M KM resulted in increased stomatal conductance and chlorophyll fluorescence in leaves compared with stressed plants under both stress conditions, while maximum values for both parameters were obtained following control treatment with 10-8M KM but no subsequent stress imposition. The protective role of KM was further supported by attenuation of cellular damage indicated by lipid peroxidation and ROS content measurements following KM pre-treatment under both stress conditions. However, while KM pre-treatment resulted in lowering of NO content compared with drought-stressed plants, no significant alterations were observed in NO content under salt stress. Furthermore, molecular analysis of antioxidant gene expression (AOX, GST, cAPX, FeSOD) by real-time qRT-PCR revealed differential regulation under both drought and salinity conditions for all the genes examined. KM pre-treatment under salinity conditions resulted in massive induction of all genes compared with stressed samples, whereas suppression of expression levels was observed under drought conditions indicating activation or suppression of the antioxidant pathway following KM pre-treatment, respectively. Moreover, data from microarray analysis using Affymetrix technology indicated a large number of genes belonging to several families that appear to be regulated in response to KM pre-treatment in comparison with respective stressed samples.

Abstract

This article documents the addition of 512 microsatellite marker loci and nine pairs of Single Nucleotide Polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Alcippe morrisonia morrisonia, Bashania fangiana, Bashania fargesii, Chaetodon vagabundus, Colletes floralis, Coluber constrictor flaviventris, Coptotermes gestroi, Crotophaga major, Cyprinella lutrensis, Danaus plexippus, Fagus grandifolia, Falco tinnunculus, Fletcherimyia fletcheri, Hydrilla verticillata, Laterallus jamaicensis coturniculus, Leavenworthia alabamica, Marmosops incanus, Miichthys miiuy, Nasua nasua, Noturus exilis, Odontesthes bonariensis, Quadrula fragosa, Pinctada maxima, Pseudaletia separata, Pseudoperonospora cubensis, Podocarpus elatus, Portunus trituberculatus, Rhagoletis cerasi, Rhinella schneideri, Sarracenia alata, Skeletonema marinoi, Sminthurus viridis, Syngnathus abaster, Uroteuthis (Photololigo) chinensis, Verticillium dahliae, Wasmannia auropunctata, and Zygochlamys patagonica. This article also documents the addition of nine sequencing primer pairs and sixteen allele specific primers or probes for Oncorhynchus mykiss and Oncorhynchus tshawytscha; these primers and assays were cross-tested in both species.

Abstract

The highly heterogeneous nature of Botrytis cinerea provides adaptive benefits to variable environmental regimes. Disentangling pathogen population structure in anthropogenic agroecosystems is crucial to designing more effective management schemes. Herein, we studied how evolutionary forces exerted in different farming systems, in terms of agrochemicals-input, shape B. cinerea populations. In total, 360 B. cinerea isolates were collected from conventional and organic, strawberry and tomato farms in Cyprus and Greece. The occurrence and frequency of sensitivities to seven botryticides were estimated. Results highlighted widespread fungicide resistance in conventional farms since only 15.5% of the isolates were sensitive. A considerable frequency of fungicide-resistant isolates was also detected in the organic farms (14.9%). High resistance frequencies were observed for boscalid (67.7%), pyraclostrobin (67.3%), cyprodinil (65.9%), and thiophanate-methyl (61.4%) in conventional farms, while high levels of multiple fungicide resistance were also evident. Furthermore, B. cinerea isolates were genotyped using a set of seven microsatellite markers (simple sequence repeat [SSR] markers). Index of association analyses (Ia and rBarD) suggest asexual reproduction of the populations, even though the mating-type idiomorphs were equally distributed, indicating frequency-dependent selection. Fungicide resistance was correlated with farming systems across countries and crops, while SSRs were able to detect population structure associated with resistance to thiophanate-methyl, pyraclostrobin, boscalid, and cyprodinil. The expected heterozygosity in organic farms was significantly higher than in conventional, suggesting the absence of selective pressure that may change the allelic abundance in organic farms. However, genetic variance among strawberry and tomato populations was high, ranking host specificity higher than other selection forces studied. [ABSTRACT FROM AUTHOR]

Abstract

Aspergillus fungi are very important in our lives as they play a key role in the recycling of environmental carbon and nitrogen. We can find them in different environments such as plants, seeds and soil. This genus has been extensively studied because of its ability to infect and produce mycotoxins in many commercial crops. In recent years, the scientific community has shown interest in finding antifungal ingredients from natural sources, resulting in the identification of various plant-derived compounds with antifungal activity. In the present study, ten natural phenols were evaluated for their ability to inhibit the growth of Aspergillus niger fungus and the production of fumonisin B2. In particular, the antifungal effects of phenols were studied as the minimal inhibitory concentration, the mycelial growth, the conidia production and the germination of conidia of Aspergillus niger fungus were assessed. Results showed that the natural phenolic compounds in terms of antifungal activity with respect to the growth of the fungus Aspergillus niger showed the following order: carbarcol> thymol> isoeugenol > eugenol> 4-ethylguaicol> o-cresol > guaiacol> p-cresol> vanillin> vanilic acid. In addition, the inhibitory capacity of the phenolic compounds in the production of fumonisin B2 from said fungus was tested. The use of eugenol, isoeugenol, thymol, carvacrol and p-cresol decreased significantly (93-99%) the production of mycotoxin, called fumonisin B2. Overall, natural phenols can be considered as a pool of antifungal agents against Aspergillus niger. Οι μύκητες του γένους Aspergillus είναι πολύ σημαντικοί στην ζωή μας, καθώς διαδραματίζουν βασικό ρόλο στην ανακύκλωση του περιβαλλοντικού άνθρακα και του αζώτου. Τους συναντάμε σε διάφορα περιβάλλοντα όπως είναι τα φυτά, οι σπόροι αλλά και το έδαφος. Το γένος αυτό έχει μελετηθεί εκτενώς λόγω της ικανότητάς του να μολύνει και να παράγει μυκοτοξίνες σε πολλές εμπορικές καλλιέργειες. Τα τελευταία χρόνια, η επιστημονική κοινότητα παρουσίασε ενδιαφέρον για την εύρεση αντιμυκητιακών συστατικών από φυσικές πηγές, με αποτέλεσμα να εντοπισθούν διάφορες ενώσεις φυτικής προέλευσης με ικανότητα να παρεμποδίζουν την ανάπτυξη των μυκήτων. Στην παρούσα μελέτη, αξιολογήθηκαν δέκα φυσικές φαινόλες ως προς την ικανότητα τους να παρεμποδίζουν την ανάπτυξη του μύκητα Aspergillus niger και την παραγωγή φουμονισίνης Β2. Πιο αναλυτικά, μελετήθηκε η επίδρασή τους στην ελάχιστη ανασταλτική συγκέντρωση, στην μυκηλιακή ανάπτυξη, στην παραγωγή κονιδίων και στη βλαστικότητα των κονιδίων του μύκητα Aspergillus niger. Τα αποτελέσματα έδειξαν ότι οι φυσικές φαινολικές ενώσεις, ως προς την άποψη της αντιμυκητιακής δραστικότητας σε σχέση με την ανάπτυξη του μύκητα Aspergillus niger, παρουσίασαν την ακόλουθη σειρά: καρβακρόλη> θυμόλη> ισοεγενόλη> ευγενόλη> 4-εθυλ-γουαικόλη> ο-κρεσόλη> γουαϊακόλη> π -κρεζόλη> βανιλίνη> βανιλικό οξύ. Επιπλέον, ελέγχθηκε και η ανασταλτική ικανότητα των φαινολικών ενώσεων στη παραγωγή της φουμονισίνης Β2 από τον εν λόγω μύκητα. Η χρήση των ενώσεων ευγενόλης, ισοευγενόλης, θυμόλης, καρβακρόλης και π-κρεζόλης μείωσαν την παραγωγή φουμονισίνης Β2 από 93% έως 99%. Abstract

Background: Consumer concerns regarding high-quality produce, free of pesticide residues, direct research towards disease management strategies that minimise or even exclude the use of synthetic chemistries in crop production. The efficacy of a chitosan-based edible coating combined with the acetonic extract of Salvia fruticosa Mill. (ASF) was assessed against the grey mould of table grapes., Results: HPLC-SPE-NMR and q-NMR analyses defined major constituents of ASF to be the flavonoids hispidulin, salvigenin and cirsimaritin and the diterpenes carnosic acid, carnosol and the 12-methoxycarnosic acid. The extract was found to be efficacious in reducing spore germination and mycelial growth of Botrytis cinerea in vitro at 10 and 25 °C. However, the combination of the ASF with chitosan 1% (w/v; CHIT) significantly improved fungal inhibition. Similarly, in fruit inoculation trials at 10 °C, the efficacy of the combined application of the ASF at 500 mg L -1 with CHIT against grey mould was statistically equal to the synthetic fungicide thiabendazole, ranging from 98.4% to 92.7% at 12 and 21 days post-inoculation, respectively. Furthermore, chitosan coating alone and in combination with ASF decreased the rate of fruit weight loss during cold storage, while preserved soluble solids content and titratable acidity. Chitosan-based coatings did not affect quality attributes and the bioactive compounds in table grapes., Conclusion: The combined application of the ASF in the form of an edible coating with chitosan could effectively control B. cinerea without deteriorating quality and physico-chemical properties of grapes. © 2016 Society of Chemical Industry., (© 2016 Society of Chemical Industry.)
