İstihbarat yaşanan zamanın şartlarına uygun olarak geliştirilen bilinmezliğe karşı önceden tedbir almak, muhtemel sorunlarla mücadele etmek ve yön vermek için elde edilen verinin akıl, zekâ süzgecinden geçerek ortaya çıkarılmasıyla elde edilen üründür. Literatürde istihbarat akıl, zekâ, anlayış, malûmat, vukuf, işitilen, alınan, toplanan haberler ve bilgiler olarak tarif edilmektedir. Ancak teknik olarak incelediğimizde istihbarat kelimesinin kapsamı ham bilgilerin işlenmesi, tasnif, kıymetlendirme, yorum ve analiz sonucu üretilen ürün şeklindedir. Bu ürün plânlama, araştırma, deliller toplama, çeşitli aklî ve tecrübî ilmî metotlar ile onları değerlendirip bir sonuç istihsal edilmesi ve en uygun kullanma yöntemini içine alan faaliyetler bütününden oluşur. Soğuk Savaş döneminin bitmesi ve 11 Eylül saldırılarından sonra uluslararası arenada güvenlik ve istihbarat alanında önemli değişimler yaşanmıştır. Reel politik düşüncenin etkisi ile devletlerin hâkimiyet ve varlıklarını sürdürme mücadelesinde asli görevi olan güvenlik ve istihbarat alanında yaşanan teknolojik gelişmelerin hız kazanması ve hegemon devletlerin kamufle stratejileri istihbarat yönetiminde değişimleri ortaya çıkarmıştır. Bu değişimlerin en önemlilerinden biri devletlerin küresel ölçekte etkili olan özel istihbarat şirketlerini kullanmaya başlamalarıdır. Özel istihbarat şirketlerin kullanılmasındaki artış ve yaşanan durumlar uluslararası ilişkilerde yeni bir tartışmayı da beraberinde getirmiştir. İstihbarat yönetiminde özel şirketlerin kullanılmasının devletlere etkileri ve hizmet verdikleri bölgelerdeki uygulamaları sorgulanır hale gelmiştir. Sonuç olarak bu çalışmada Amerika Birleşik Devletleri (ABD) ve Rusya Federasyonun (RF) istihbarat sistemleri ve bu devletlerle ilintili uluslararası alanda ön plana çıkan özel şirketleri ele alınmak suretiyle, reel politik paradigma kapsamında özel istihbarat şirketlerinin istihbaratın geleceğindeki rolü analiz edilmiştir. Intelligence is a product that is developed in accordance with the conditions of the time, and obtained by revealing the data obtained in order to take precautionary measures against the unknown, to combat possible problems and to give direction, by passing through the intelligence filter. In the literature, intelligence is defined as mind, intelligence, understanding, knowledge, insight, heard, received, collected news and information. However, when we examine it technically, the scope of the word intelligence is the product produced as a result of processing raw information, classification, evaluation, interpretation and analysis. This product consists of all the activities that include planning, research, collecting evidence, evaluating them with various mental and experiential scientific methods, producing a result and using the most appropriate method. After the end of the Cold War period and the September 11 attacks, important changes have occurred in the field of security and intelligence in the international arena. With the effect of real political thought, the acceleration of technological developments in the field of security and intelligence, which is the main task of states in the struggle to dominate and maintain their existence, and the camouflage strategies of hegemonic states have revealed changes in intelligence management. One of the most important of these changes is that states have started to use private intelligence companies that are effective on a global scale. The increase in the use of private intelligence companies and the situations experienced have brought a new discussion in international relations. The effects of the use of private companies in intelligence management on the states and their practices in the regions they serve have become questionable. As a result, in this study, the role of private intelligence companies in the future of intelligence was analyzed within the scope of the realpolitik paradigm, by considering the intelligence systems of the United States (USA) and the Russian Federation (RF) and the internationally prominent private companies associated with these states.

In this study, low cost, sensitive and selective chronoamperometric determination of vanillin (VNL) is firstly achieved by using a disposable poly(Eriochrome Black T) modified pencil graphite electrode (poly(EBT)/PGE). Electro-oxidation behavior of 1.0 mM VNL was investigated at both poly(EBT)/PGE and PGE by the cyclic voltammetry (CV) method. CV measurements showed that oxidation current of VNL at poly (EBT)/PGE was much higher than the bare one. Furthermore, the pH effect on electro-oxidation current of VNL was tested at different pH values (2.0-8.0) of Britton Robinson Buffer solution (BRBS) for poly(EBT)/PGE and the highest current was obtained at pH 7.0 BRBS. Significant analytical parameters such as linear response range (LRR), limit of detection (LOD), and sensitivity were found as 0.050-10.0 µM, 0.013 µM and 5355 µA mM-1 cm-2, respectively. The fabricated sensor was tested on a VNL-containing sample (VNL content: 3.0%) and the result (3.04% ±0.01) obtained from the applicability study showed that the proposed sensor enables the determination of VNL with high accuracy and precision., Bu çalışmada, vanilinin (VNL) düşük maliyetli, duyarlı ve seçici kronoamperometrik tayini tek kullanımlık bir poli(Eriokrom Siyahı T) modifiye kalem grafit elektrodun (poly(EBT)/PGE) kullanılması ile ilk defa gerçekleştirilmiştir. 1.0 mM VNL’nin elektro-yükseltgenme davranışı, hem PGE’de hem de poly(EBT)/PGE’de döngüsel voltammetri (CV) metodu ile incelenmiştir. CV ölçümleri poly(EBT)/PGE’deki yükseltgenme akımının yalın elektroda göre daha yüksek olduğunu göstermiştir. Ayrıca, poly(EBT)/PGE’de VNL’nin yükseltgenme akımına pH’nın etkisi, farklı pH değerlerindeki (2.0-8.0) Britton Robinson tampon çözeltilerinde (BRBS) test edilmiş ve en yüksek akımın pH 7.0 BRBS’de alındığı gözlenmiştir. Doğrusal yanıt aralığı (LRR), belirtme alt sınırı (LOD) ve duyarlılık gibi önemli analitiksel parametreler sırasıyla 0.050-10.0 µM, 0.013 µM ve 5355 µA mM-1 cm-2 olarak bulunmuştur. Tasarlanan sensör, VNL içeren bir örnekte (VNL içeriği %3.0) başarıyla test edilmiş ve uygulanabilirlik çalışmasından elde edilen sonuç (%3.04 ±0.01), önerilen sensörün, VNL’nin yüksek doğrulukta ve kesinlikte tayinine imkân sağladığını göstermiştir.

One of the major problems in amperometric biosensors based on detection of H2O2 produced by enzymatic reaction between oxidase enzymes and substrate is the interference of redox active compounds such as ascorbic acid (AA), dopamine (DA) and uric acid (UA). To minimize these interferences, sodium bismuthate was used for the first time as an insoluble pre-oxidant in the flow injection (FI) amperometric glucose biosensor at a Glucose oxidase (GOx) immobilized Pt/Pd bimetallic modified pre-anodized pencil graphite electrode (p.PGE). In this context, these interfering compounds were injected into a flow injection analysis (FIA) system using an injector which was filled with NaBiO3. Thus, these interferents were converted into their redox inactive oxidized forms before reaching the electrode in the flow cell. While glucose was not influenced by the pre-oxidant in the injector, the huge oxidation peak currents of the interferents decreased significantly in the biosensor. FI amperometric current time curves showed that the AA, DA and UA were minimized by 96 %, 86 %, and 98 % respectively, in the presence of an equivalent concentration of interferences in a 1.0 mM glucose solution. The proposed FI amperometric glucose biosensor exhibits a wide linear range (0.01-10 mM, R-2=0.9994) with a detection limit of 2.4x10(-3) mM. Glucose levels in the artificial serum and two real samples were successfully determined using the fabricated FI amperometric biosensor.

Bu tez çalışmasında, (i) yüksek iletkenlik özelliğine ve katalitik etkinliğe sahip metal nanoparçacıklar (Au, Pt ve Pd), (ii) ucuz ve tek kullanımlık kalem grafit elektrot (pencil graphite electrode, PGE), (iii) hızlı ve rutin analizlere olanak sağlayan akışa enjeksiyon analiz (Flow Injection Analysis, FIA) metodu gibi üç faydalı bileşen üzerinden elektrokimyasal sensör ve biyosensör çalışmaları gerçekleştirilmiştir. Bu amaçla Au, Pt, Pd ve Pt-Pd nanoparçacıklar PGE yüzeyine elektrokimyasal olarak biriktirilmiş ve elde edilen elektrotların H2O2'ye yanıtı döngüsel voltammetri (CV) tekniği ile incelenmiştir. Elektrokimyasal H2O2 sensörü, sırasıyla Au ve Pt ile Pd ve Pt-Pd modifiye PGE'lerde H2O2'nin yükseltgenmesi ve indirgenmesi üzerine geliştirilmiştir. Her bir modifiye elektrot için çalışma potansiyeli ve akış hızı optimize edilmiş ve FIA sisteminde amperometrik H2O2 sensörünün analitiksel parametreleri belirlenmiş ve gerçek örneklere uygulanabilirlikleri başarıyla test edilmiştir. Tezin ikinci kısmında ise metal/bimetal nanoparçacık modifiye PGE'ler yüzeyine glukoz oksidaz (GOx) immobilize edilmiş ve glukoz biyosensör çalışmaları CV tekniğiyle gerçekleştirilmiştir. Glukoza en iyi elektrokatalitik etkinliğin ve yanıtın alındığı elektrotlar GOx immobilize Pt ve Pt-Pd modifiye PGE'ler olarak belirlenmiştir. Her iki elektrot için FIA sisteminde geliştirilen amperometrik glukoz biyosensörünün optimize edilen koşullar altında analitiksel performansları belirlenmiş ve gerçek örneklere uygulanabilirlikleri başarıyla test edilmiştir. Askorbik asit, dopamin ve ürik asitin geliştirilen glukoz biyosensörüne girişim etkileri, NaBiO3 önyükseltgeyicisi kullanılarak önemli derecede giderilmiştir. In this thesis, electrochemical sensor and biosensor studies were performed based on three useful components (i) metal nanoparticles (Au, Pt and Pd) with their high conductivity and catalytic activity, (ii) low-cost and disposable pencil graphite electrode (PGE) and (iii) flow injection analysis (FIA) which provides fast and routine analysis. For this purpose, Au, Pt, Pd and Pt-Pd nanoparticles were electrochemically deposited on the PGE surface and the response of the obtained electrodes to H2O2 was investigated by using cyclic voltammetry (CV) technique. The electrochemical H2O2 sensor was developed on the oxidation and reduction of H2O2 at the Au, Pt and Pd, Pt-Pd modified PGEs, respectively. The applied potential and flow rate for each modified electrode has been optimized and the analytical parameters of the amperometric H2O2 sensor in FIA have been determined and their applicability to the real samples has been successfully tested. In the second part of the thesis, glucose oxidase (GOx) was immobilized on the surface of metal/bimetal nanoparticle modified PGEs and glucose biosensor studies were performed by using CV technique. The electrodes which give the best electrocatalytic activity and response to glucose were identified as GOx immobilized Pt and Pt-Pd modified PGEs. Analytical performances of the amperometric glucose biosensor constructed in FIA system for both electrodes were determined under optimized conditions and their applicability to the real samples was successfully tested. The interference effects of ascorbic acid, dopamine and uric acid on the developed glucose biosensor were significantly reduced by using a pre-oxidant, NaBiO3. As a result, the design of cheap, fast, sensitive electrochemical sensor and biosensor using a combination of metal/bimetal nanoparticle, PGE and FIA system and the uses of the NaBiO3 as a pre-oxidant for the first time in FIA system for the elimination of interferents constitutes the original part of this study.Keywords: Electrochemical Sensor/Biosensor, Flow Injection Analysis, Glucose Oxidase, Sodium Bismuthate, Metal Nanoparticle, Pencil Graphite Electrode. 267

Yeni fonksiyonel organik bileşiklerin tasarımı ve sentezi, bu bileşiklerden elde edilen malzemelerden yararlanılarak moleküler elektronik ve fotonikler, algılayıcılar, makine ve aygıtlar, ışık yayan diyotlar (LED?ler), güneş pilleri, transistörler, elektrokromik cihazlar, veri işleme ve depolama aygıtları gibi ileri teknolojik uygulamalar nedeniyle oldukça önem arz etmektedir. Fonksiyonel organik bileşikler içerisinde lüminesans bileşikler, son zamanlarda hem biyo- ve nanoteknoloji ve çevre bilimleri, hem de malzeme bilimlerinde iyon ve/veya molekül tanıyan algılayıcılar, elektron ve/veya enerji aktarım sistemleri, görüntüleme ajanları, moleküler makina ve aygıtlar, aritmetik işlem yapabilen moleküler mantık kapıları vb. olarak kullanılabilir olmaları nedeniyle çok ilgi çekmektedir. Bu vesileyle dünyanın dört bir yanında başta sentetik organik kimya olmak üzere araştırma grupları uygulamaya açık yeni lüminesans bileşiklerin tasarımı, sentezi ve tanımlanmasına yönelik çalışmalara geniş bir yer ve önem vermektedirler. Bu çalışma, literatürde bilinmeyen, 4,4-Difloro-8-(1,4,7,10-tetraazasiklododekan)metil-1,3,5,7-tetrametil-2,6-dietil-4-bora-3a,4a-diaza-s-indasen (1) sentezlenmiş ve bu bileşiğin yapı ve özellikleri spektroskopik yöntemlerle (UV-vis, lüminesans -floresans, FTIR, NMR, elementel analiz, kütle) aydınlatılmıştır. Ayrıca elde edilen bu yeni bileşiğin iyonlara karşı duyarlılığı denenmiştir. Design and synthesis of novel functional organic compounds have attracted considerable attention since they lead to a variety of advanced technological applications in the field of molecular electronics and photonics, sensors, machines and devices, light emitting diodes, photovoltaics, transistors, electrochromics, data processing and storage media. Among functional compounds, luminescent organic materials are quite important due to their practical applications in bio- and nanotechnological as well as materials sciences as chemosensors, electron and/or energy transfer systems, imaging agents, molecular machines and devices, molecular logic gates and so on. For that reason, the design, synthesis and characterization of novel luminescent compounds are highly concerned by first the synthetic organic chemists and as well as other communities all around the world.In this study, a novel compound 4,4-Difloro-8-(1,4,7,10-tetraazasiklododekan)metil-1,3,5,7-tetrametil-2,6-dietil-4-bora-3a,4a-diaza-s-indasen (1) was designed, synthesized, and characterized spectroscopically (UV-Vis, luminescence-fluorescence, FTIR, NMR, mass spectroscopy, combustion analysis). Furthermore, this novel compound was tested as ion sensor.
