1. Habitat loss and fragmentation potentially affect the performance of bees that forage nectar and pollen of plants in their habitats. In forest landscapes, silvicultural conifer plantations often have reduced and fragmented natural broadleaf forests, which seem to provide more floral resources for bees than do the plantations. 2. This study evaluated the effects of forest characteristics (i.e. elevation, area, edge length, and tree size of natural forests) on pollen diets (plant taxa assemblages of pollen grains in provisions) and total provision mass in oviposited chambers in nests made by a standardised number of Osmia cornifrons bees at 14 sites in a forestry area in central Japan. 3. From April to May, the numbers of nests and chambers per nest increased, and the provision mass per chamber decreased. Main pollen sources were Prunus at higher elevations in April and Wisteria at lower elevations in May, foraging on which increased the numbers of nests and chambers per nest. The provision mass per chamber was smaller at higher elevations in more fragmented natural forests. Decreases in the area of natural forests within the foraging range (400‐m radii) of O. cornifrons increased the utilisation of Rubus pollen and decreased the total provision mass. 4. These findings suggest that the loss and fragmentation of natural broadleaf forests change pollen diets and reduce the provision mass of mason bees, which may reduce the number and size of their offspring.

Seed transfer may affect not only the performance of trees growing from transferred seeds but also the reproduction of the trees and the performance of their progeny. We investigated Quercus mongolica var. crispula trees transplanted from multiple provenances and their seedlings naturally regenerated in northeastern and southwestern test sites in Japan. The test sites and provenances were grouped into seven regions based on their geographic and climatic profiles. We defined the local population as trees from provenances in a region of either test site and the foreign population as trees from provenances in the other regions. Tree size was larger in the local population than in the foreign population in both test sites. We estimated foreign population ancestry from the allele frequencies in the local and foreign populations in each test site using microsatellite genotypes. We discriminated the local and foreign populations using foreign population ancestry although genetic differentiation between these populations were relatively low (0.022 ≤ FST ≤ 0.034). Different frequency distributions of foreign population ancestry between transplanted trees and regenerated seedlings indicated crossing between the local and foreign populations. Tree size decreased as foreign population ancestry increased in both test sites. Seedling mass increased in the northeastern test site but decreased in the southwestern test site as foreign population ancestry increased, indicating larger mass of seedlings with genetic properties of southwestern provenances. The findings suggest that seed transfer between regions results in reduced performance of trees growing from transferred seeds and admixed progeny of trees from local and foreign regions.

Numerous cultivars of Japanese flowering cherry (Prunus subgenus Cerasus) are recognized, but in many cases they are difficult to distinguish morphologically. Therefore, we evaluated the clonal status of 215 designated cultivars using 17 SSR markers. More than half the cultivars were morphologically distinct and had unique genotypes. However, 22 cultivars were found to consist of multiple clones, which probably originate from the chance seedlings, suggesting that their unique characteristics have not been maintained through propagation by grafting alone. We also identified 23 groups consisting of two or more cultivars with identical genotypes. Most members of these groups were putatively synonymously related and morphologically identical. However, some of them were probably derived from bud sport mutants and had distinct morphologies. SSR marker analysis provided useful insights into the clonal status of the examined Japanese flowering cherry cultivars and proved to be a useful tool for cultivar characterization.

Mating processes of local demes and spatial genetic structure of island populations at the self-incompatibility (S-) locus under negative frequency-dependent selection (NFDS) were evaluated in Prunus lannesiana var. speciosa in comparison with nuclear simple sequence repeat (SSR) loci that seemed to be evolutionarily neutral. Our observations of local mating patterns indicated that male-female pair fecundity was influenced by not only self-incompatibility, but also various factors, such as kinship, pollen production and flowering synchrony. In spite of the mating bias caused by these factors, the NFDS effect on changes in allele frequencies from potential mates to mating pollen was detected at the S-locus but not at the SSR loci, although the changes from adult to juvenile cohorts were not apparent at any loci. Genetic differentiation and isolation-by-distance over various spatial scales were smaller at the S-locus than at the SSR loci, as expected under the NFDS. Allele-sharing distributions among the populations also had a unimodal pattern at the S-locus, indicating the NFDS effect except for alleles unique to individual populations probably due to isolation among islands, although this pattern was not exhibited by the SSR loci. Our results suggest that the NFDS at the S-locus has an impact on both the mating patterns and the genetic structure in the P. lannesiana populations studied.

Landscape disturbances can affect reproductive performance of animal-pollinated trees. We verified the effects of the loss and fragmentation of natural forests caused by the creation of coniferous plantations on fruit and seed production as well as mating patterns of animal-pollinated trees. We investigated 146 and 134 individual flowering trees of Prunus verecunda (Koidz.) Koehne in 2012 and 2013, respectively, at 12 sites. These sites were at least 1.2 km apart from each other in an 8 × 15-km forestry region, and the composition and configuration of natural forests varied in the study area. The mean outcrossing rate was >0.99 across the sites. Among trees within the sites, the number of fruits per inflorescence was positively correlated with the basal stem area and leaf chlorophyll density of the trees. Among the sites, the mean number of fruits per inflorescence was positively correlated with the site elevation, and the correlated paternity was positively correlated with the mean distance between trees. The sound-seed rate was positively correlated with the natural-forest area among the sites. These results suggest that environments and resources of trees influence their fruit production, that a loss of natural forests increases in embryo mortality, and that a reduction in tree density decreases pollen donor diversity in P. verecunda. Thus, a landscape disturbance may decrease seed production, whereas outbreeding is maintained, and fruit production is not likely to be dependent on the landscape disturbance in this species.
