Many healthcare workers who handle anticancer drugs are at risk for occupational exposure. However, there are no established permissible limits for occupational exposure to anticancer drugs; thus, in this study, we aimed to search for and improve procedures that have a greater impact on the amount of spatter for handling anticancer drugs in vials, which are frequently used, based on the quantitative evaluation of the amount of exposure. We used sodium riboflavin phosphate (FMN) as a simulated anticancer drug and measured the amount of FMN dispersed to the handling area by the wiping method and the amount of FMN dispersed in both gloves using high-performance liquid chromatography with fluorescence detection (HPLC-FL). In this study, it was suggested that the overall amount of dispersal in the preparation process was affected by the different methods of injecting the drug solution into the infusion bottles and whether recapping. It was also found that the variation in the amount of dispersal differed depending on the selected preparation technique. It was suggested that the amount of dispersal could be reduced by selecting an appropriate dissolution method for multiple vials, recapping, an appropriate method for injecting the drug into the infusion bottle, and properly preparing the internal pressure of the infusion bottle. The results of this study suggest that there are some techniques and procedures in the preparation process of vials that have a significant effect on the amount of dispersal, and that proper implementation of these techniques can contribute to the reduction of dispersal.

The effect of nanoparticle type, shape, as well as primary and secondary particle size on toxicity remains poorly characterized. In this study, suspensions of nickel oxide (NiO) nanoparticles with the same primary particle size (50 nm) but different secondary particle sizes were prepared, and their cytotoxicity was investigated. A planetary ball mill wet nanopulverizer with zirconium milling balls of decreasing sizes (φ: 0.5, 0.1, and 0.05 mm) yielded NiO nanoparticles of decreasing mean particle size (310.4 ± 6.7, 172.0 ± 2.8, and 102.0 ± 0.5 nm). The aim of this investigation is to clarify the types and concentrations of VOCs present in various commercial household water-based hand pump spray products used in Japan, and to estimate their average concentrations in indoor air when the spray product is used. We selected glycol and glycol ethers as the main target compounds, as these chemicals were detected at high frequencies and concentrations in a national survey of Japanese indoor air pollution. The extraction of these chemicals using graphite carbon cartridges was examined, with good recoveries and reproducibilities being obtained. Eighteen chemicals were analyzed in 54 commercial products and 8 chemicals were detected. More specifically, dipropylene glycol (DPG) was present in 44 samples (1.1 × 101-1.8 × 104 μg/mL); propylene glycol (PG) was present in 22 samples (1.5 × 101-2.9 × 104 μg/mL); diethylene glycol monoethyl ether (DGMEE) was found in 15 samples (trace amount-1.9 × 103 μg/mL); diethylene glycol (DEG) was present in 9 samples (1.0 × 101-2.4 × 103 μg/mL); 1,3-butandiol (13BG) was found in 5 samples (trace amount-7.4 × 103 μg/mL); 2-ethyl-1-hexanol (2E1H) was detected in 5 samples (3.2 × 10-1-4.4 × 101 μg/mL); diethylene glycol monobutyl ether (DGMBE) was present in 4 samples (2.1 × 101-7.1 × 101 μg/mL); and 3-methoxy-3-methylbutanol (MMB) was found in 2 samples (2.4 × 101-4.7 × 102 μg/mL). In addition, the average concentrations of these chemicals in indoor air were estimated using their maximum concentrations observed in the spray product. The estimated average concentrations of the chemicals in indoor air were determined to range between 1.0 × 10-2 and 1.0 mg/m3, with the exception of 2E1H and DGMBE. Furthermore, the estimated average concentrations of PG, 13BG, and DGMEE in indoor air were comparable to or higher than those reported in a national survey of Japanese indoor air pollution. It therefore appeared that household water-based hand pump sprays may contribute to the presence of these chemicals in indoor air. In contrast, estimated average concentrations of 2E1H in indoor air were low, its concentrations observed in a national survey of Japanese indoor air pollution are likely due to the use of plasticizers and paints.

This study aimed to optimize the ratio of dioctyl 4-cyclohexene-1,2-dicarboxylate (DOTH) and di-isononyl-cyclohexane-1,2-dicarboxylate (DINCH®) for use as plasticizers in poly(vinyl chloride) (PVC) sheets. We also evaluated the biological safety of DOTH for its potential to be part of a safe PVC-based blood container. The suppression of hemolysis in mannitol-adenine-phosphate / red cell concentrates (MAP/RCC) with DOTH/(DINCH®-PVC) sheets and the elution of plasticizers from the sheets increased with higher DOTH compositions. The properties of the PVC sheet containing DOTH and DINCH® in the ratio of 25:33 parts against PVC 100 parts as a weight were almost identical to the PVC sheet made of di(2-ethylhexyl) phthalate. From a subchronic toxicity test, DOTH did not show any adverse effects on all organs, including the testes, epididymis, liver, and kidneys. The no-observed-adverse-effect level was 300 mg/kg body weight/day in a rat. These results suggest that DOTH/DINCH® (25:33) is a promising candidate for the replacement of di(2-ethylhexyl) phthalate in blood containers. J. VINYL ADDIT. This study deals with the development and performance evaluation of a positive reference material for hemolysis testing, which is used for evaluating the biological safety of medical devices. Genapol X-080, a nonionic detergent, was selected as a candidate hemolytic substance in a survey of 23 chemical compounds; it showed significant hemolytic activity against rabbit defibrinated blood at concentrations more than 20 µg/mL. A polyvinyl chloride (PVC) sheet spiked with 0.6% (w/w) of the compound exhibited weak hemolytic activity in direct contact and/or extract-based assays after 4 h incubation at 37°C. A PVC sheet containing 5.8% (w/w) Genapol X-080 induced complete hemolysis in both assays. The amount of Genapol X-080 eluted from each PVC sheet during hemolysis testing using the direct contact method increased time-dependently and reached 25.6 (former sheet) or 1154 (later sheet) µg/mL after 4 h incubation, which was similar to or much higher than the critical micelle concentration, respectively. Similar elution behavior was observed using the extract-based method, and the Genapol X-080 content in test solutions prepared by autoclave extraction of both sheets was 22.5 and 358 µg/mL, respectively, indicating a clear relationship between the degree of hemolytic activity and the eluted amount of Genapol X-080. Thus, a PVC sheet spiked with a compound exhibiting different hemolytic activity depending on its concentration may be useful as a positive reference material to validate the hemolysis tests.

Although nanomaterials are already being used for various applications in the industry, the safety of nanomaterials has not yet been sufficiently elucidated. An in vitro cellular toxicological study using well-characterized nanomaterials is conducted for the evaluation of the biological effects of nanomaterials. In this study, the effects of copper oxide nanoparticles on the global gene expression of human lung epithelial A549 cells were analyzed, and the molecular responses of A549 cells to the toxicity of the copper oxide (CuO) nanoparticles were inferred. Furthermore, the cytotoxic effects of silicon dioxide (SiO2) nanoparticles and titanium dioxide (TiO2) nanoparticles coexisting with some metal salts in Chinese hamster lung fibroblast V79 cells were also examined, and SiO2 nanoparticles increased the cytotoxicity of some of the coexisting metal salts as a result. Finally, the importance of in vitro studies in the safety evaluation of nanomaterials was discussed.

The aim of this study is to identify a plasticizer that is effective in the suppression of the autohemolysis of the stored blood and can be used to replace di(2-ethylhexyl) phthalate (DEHP) in blood containers. The results of hemolysis test using mannitol-adenine-phosphate/red cell concentrates (MAP/RCC) spiked with plasticizers included phthalate, phthalate-like, trimeliate, citrate, and adipate derivatives revealed that di-isononyl-cyclohexane-1,2-dicarboxylate (Hexamoll(®) DINCH), di(2-ethylhexyl)-1,2,3,6-tetrahydro-phthalate (DOTP), and diisodecyl phthalate (DIDP) exhibited a hemolysis suppression effect almost equal to that of DEHP, but not other plasticizers. This finding suggested that the presence of 2 carboxy-ester groups at the ortho position on a 6-membered ring of carbon atoms may be required to exhibit such an effect. The hemolytic ratios of MAP/RCC-soaked polyvinyl chloride (PVC) sheets containing DEHP or different amounts of DINCH or DOTP were reduced to 10.9%, 9.2-12.4%, and 5.2-7.8%, respectively (MAP/RCC alone, 28.2%) after 10 weeks of incubation. The amount of plasticizer eluted from the PVC sheet was 53.1, 26.1-36.5, and 78.4-150 µg/mL for DEHP, DINCH, and DOTP, respectively. PVC sheets spiked with DIDP did not suppress the hemolysis induced by MAP/RCC because of low leachability (4.8-6.0 µg/mL). These results suggested that a specific structure of the plasticizer and the concentrations of least more than ∼10 µg/mL were required to suppress hemolysis due to MAP/RCC.

In this study, a titanium surface was chemically modified with calcium ions and assessed for its influence on osteogenic differentiation and molecular responses of human mesenchymal stem cells (hMSCs). Titanium disks were treated with NaOH (NaOH treatment), NaOH + CaCl2 (CaCl2 treatment), or NaOH + Ca(OH)2 (Ca(OH)2 treatment). Ca(OH)2 treatment caused significantly greater calcium incorporation onto the titanium surface and apatite formation than CaCl2 treatment. The morphology of hMSCs differed on CaCl2- and Ca(OH)2-treated disks. The osteopontin (OPN) expression in hMSCs cultured on CaCl2-treated titanium was significantly higher than that in cells cultured on NaOH-treated disks; OPN expression was significantly higher in cells cultured on Ca(OH)2-treated disks than on un-, NaOH-, and CaCl2-treated disks. Osteocalcin (OCN) protein expression in hMSCs cultured on Ca(OH)2-treated disks was significantly higher than that on all the other disks. Comparative expression profiling by DNA microarray and pathway analyses revealed that calcium modification of the titanium surface induced integrin β3 after OPN upregulation and promoted Wnt/β-catenin signaling in hMSCs. In addition, Ca(OH)2 treatment upregulated the expression of bone morphogenetic protein 2, cyclooxygenase 2, and parathyroid hormone-like hormone in comparison to CaCl2 treatment. These observations suggest that calcium-modified titanium surfaces affect osteogenic differentiation in hMSCs and that Ca(OH)2 treatment induced osteogenic differentiation in hMSCs, whereas CaCl2 treatment had a limited effect. © 2013 Wiley Periodicals, Inc. This study examines the chemical, physicochemical, and biological properties of PVC sheets treated with UV irradiation on their surfaces to suppress the elution of a plasticizer, di-(2-ethylhexyl) phthalate (DEHP), for developing novel polyvinyl chloride (PVC) medical devices. The PVC sheets irradiated under conditions 1 (52.5 μW/cm(2), 136 J/cm(2)) and 2 (0.45 mW/cm(2), 972 J/cm(2)) exhibited considerable toxicity in cytotoxicity tests and chromosome aberration tests due to the generation of DEHP oxidants, but no toxicity was detected in the PVC sheet irradiated under condition 3 (8.3 mW/cm(2), 134 J/cm(2)). The release of DEHP from the surface irradiated under condition 3 was significantly suppressed, and mono-(2-ethylhexyl) phthalate (MEHP) converted from a portion of DEHP could be easily removed from the surface by washing with methanol. The physicochemical properties of the surface regarding the suppression of DEHP elution remained stable through all sterilizations tested, but MEHP elution was partially recrudesced by the sterilizations except for gamma irradiation. These results indicated that UV irradiation using a strong UV-source over a short time (condition 3) followed by methanol washing and gamma sterilization may be useful for preparing novel PVC products that did not elute plasticizers and do not exhibit toxicity originating from UV irradiation.

Nanomaterials are relatively new and unconventional materials with many useful properties, but their effects on biological systems are poorly understood. Nanoclay is a general term for layered mineral silicate nanoparticles that are ideally suited for use in clay-based nanocomposites. The potential biological hazards of nanoclays have not been addressed, however. Therefore, we investigated the in vivo effects and drug interactions of nanoclays. In mice, administration of nanoclay particles via the tail vein led to acute liver injury. Co-administration of nanoclay and carbon tetrachloride, paraquat, or cisplatin resulted in both liver and kidney injury. Our findings thus indicate that nanoclay particles are potentially hepato- and nephrotoxic. The international standard ISO 8124-3: 1997 "Safety of toys -Part 3: Migration of certain elements" and "Interim Enforcement Policy for Children's Metal Jewelry Containing Lead- 2/3/2005" by the U.S. Consumer Product Safety Commission (CPSC) to control the amount of eluted lead from metal accessories cannot be simply compared, because the acid extraction methods and the limit values are different from each other. Therefore acid extraction tests based on the ISO standard and the CPSC policy were conducted and the amounts of eluted lead from small metal products were compared between both tests. There was less eluted lead in the ISO method than in the CPSC method. Moreover, the amount of eluted lead in the ISO method did not even reach that of the first elution in the CPSC method. It became clear that the acid extraction test of the ISO standard was not as good as that of the CPSC policy, because of the difference in test conditions. In 16 small metal products, seven products were unsuitable for the ISO standard and 14 products were unsuitable for the CPSC policy; however, all these products were originally inapplicable to the ISO standard and the CPSC policy. The calculation grounds of the limit values were also different between the ISO standard and the CPSC policy. The standardization of an acid extraction test that simulates the lead elution to gastric juice is required so as to prevent adverse health effects in children due to their accidental ingestion of small metal products containing lead.

The use of tributyltin (TBT) and triphenyltin (TPT) in some household products are prohibited by "Act on the Control of Household Products Containing Harmful Substances" in Japan. In this study, methods for determination of TBT and TPT in water soluble paints and adhesives were developed by GC-MS. These compounds in paints and adhesives, which were mainly composed of vinyl acetate, urethane and acryl resins, and chloroprene rubber, were firstly extracted with HCl-acetone, and then extracted with hexane. On the other hand, the adhesive composed of natural rubber was firstly dispersed in water before acidification. The organotins were extracted with hexane from this solution and then these compounds were extracted with acetonitrile from hexane extract. These extracts were purified by a florisil cartridge column after ethyl-derivation with sodium tetraethylborate, and analyzed by GC-MS. The quantifications using deuterated compound of both organotins as surrogate standard were conducted, and good results were obtained. The recoveries were 81 to 118% and the coefficients of variation were 0.83 to 4.3% (TBT and TPT added; 5 microg/g). The method quantification limits were 0.0090 to 0.025 microg/g, which were lower than those of an official method. These methods were applied to monobutyltin (MBT), dibutyltin (DBT), monophenyltin (MPT), and diphenyltin (DPT). DBT and DPT in paints and adhesives were quantified, except for DPT in natural rubber. These methods were applied to commercial products. DBT was detected at low concentrations (t.r.-0.19 microg/g) in some paint samples, while TBT and TPT were not detected in all samples.

To investigate relationships between particle (as a model of aggregates) size in a nanomaterial test suspension and its cytotoxicity, a series of eleven sizes of polystyrene (PS) particles were tested in the cytotoxicity test and the chromosome aberration test by using a Chinese hamster cell line CHL. The PS particles were spheres with defined diameters ranging from 0.1 to 9.2 µm. A series of eight sizes of particles with diameters ranging from 0.92 to 4.45 µm showed stronger cytotoxicity than the others. There was a marked difference in cytotoxicity between the 4.45- and 5.26-µm particles. The 0.92- to 4.45-µm particles did not induce structural chromosome aberrations but induced a high frequency of polyploidy in the chromosome aberration test. The 5.26-µm particles showed very weak induction of polyploidy. The incorporation of the 4.45-µm particles into CHL cells was observed by scanning electron microscopy (SEM). Some cells incorporated more than 10 particles. The semi-quantitative measurement of incorporation of particles into cells was performed by flow cytometry with a parameter of side scattered light (SSC) intensity. It showed that CHL cells preferably incorporated the 4.45-µm particles to the 5.26-µm particles. These findings suggest that CHL cells may have a kind of size-recognition ability and incorporate a particular size of particles. The particles may prevent a normal cytokinesis resulting in polyploidy induction. Nanomaterials also may show size-dependent toxicity. Data on particle (or aggregate) size distribution in the test suspension should be provided to evaluate properly the results of toxicity tests of nanomaterials.

Japan has published safety guideline on waterproof aerosol sprays. Furthermore, the Aerosol Industry Association of Japan has adopted voluntary regulations on waterproof aerosol sprays. Aerosol particles of diameter less than 10 µm are considered as "fine particles". In order to avoid acute lung injury, this size fraction should account for less than 0.6% of the sprayed aerosol particles. In contrast, the particle size distribution of aerosols released by hand-pump sprays containing fluorine-based or silicone-based compounds have not been investigated in Japan. Thus, the present study investigated the aerosol particle size distribution of 16 household hand-pump sprays. In 4 samples, the ratio of fine particles in aerosols exceeded 0.6%. This study confirmed that several hand-pump sprays available in the Japanese market can spray fine particles. Since the hand-pump sprays use water as a solvent and their ingredients may be more hydrophilic than those of aerosol sprays, the concepts related to the safety of aerosol-sprays do not apply to the hand pump sprays. Therefore, it may be required for the hand-pump spray to develop a suitable method for evaluating the toxicity and to establish the safety guideline.

Diethyl-4,4'-methylenebis(N-phenylcarbamate) (MDU) is a urethane compound that we originally synthesized, along with three other compounds, to investigate how polyurethane is hydrolysed. We tested the four compounds for cytotoxicity in two Chinese hamster cell lines (CHL and V79) and a human cancer cell line (HeLa S3). MDU showed the strongest cytotoxicity in all the cell lines with an IC50 of around 0.1 microg/ml. We further investigated MDU for its ability to induce chromosome aberrations (CAs) and micronuclei (MN) in CHL cells. MDU induced around 100% polyploid cells at 0.5 microg/ml after 24- and 48-h treatment in the CA test and a significantly increased frequency of micronuclei, polynuclear cells, and mitotic cells in the MN test, suggesting that it may induce numerical CAs. MDU's ability to cause mitotic arrest in CHL cells was greater than that of taxol and colchicine. Based on a COMPARE analysis using JFCR39, a panel of cancer cell lines, we predicted MDU to be a tubulin inhibitor. We confirmed this possibility in nerve growth factor-stimulated PC12 cells as well as in HT1080 cells, in which MDU exhibited the activity to inhibit tubulin polymerization. MDU is simpler in structure than existing anticancer drugs taxol and vincristine and can be synthesized relatively easily. Here we offer MDU as a potential new type of anticancer drug, stable even at room temperature, and inexpensive.

To promote the osteogenic properties of osteoblasts, we synthesized a hydroxyapatite (HAp) with beta-tricalcium phosphate (beta-TCP) biphasic calcium phosphate containing Nb ions (NbTCP/HAp). NbTCP/HAp was prepared by annealing precipitates obtained by coprecipitation of an aqueous solution of Ca(NO(3))(2) and a mixture of (NH(4))(2)HPO(4) and aqueous Nb solution. The precipitates can be regarded as a calcium-deficient HAp, the PO(4) sites of which are partly occupied by Nb ions. NbTCP/HAp was successfully synthesized by thermal decomposition of the precipitates. NbTCP/HAp enhanced the calcification of normal human osteoblasts (NHOst), and the amount of calcified tissue increased in proportion to the Nb ion concentration in the NbTCP/HAp. The alkaline phosphatase (ALP) activity of NHOst was also enhanced by NbTCP/HAp. Because Nb ions significantly enhance the ALP activity of NHOst, calcification by NbTCP/HAp is considered to be due to enhancement of ALP activity induced by Nb ions dissolved from NbTCP/HAp. These results indicate that NbTCP/HAp can be an effective bone repair material.

To promote the activity of normal human osteoblasts (NHOst), the novel HAp ceramics containing Nb ions (NbHAp) were synthesized by wet chemical process, which reacting aqueous solution containing a mixture of Ca(NO3)2, (NH4)2HPO4, and the Nb aqueous solution. X-ray diffraction patterns indicated that NbHAp had a monolithic apatitic structure, although crystallite decreased as Nb content increased. We attempted to determine the processing conditions for decreasing the migration of phthalate esters, particularly di-2-ethylhexyl phthalate (DEHP), from polyvinyl chloride (PVC) products using a drug solvent after dilution based on the package insert. PVC sheets and PVC tubing were subjected to optical irradiation (ultraviolet (UV), visible light irradiation) and heat treatment to determine whether they are deteriorated by these treatments. UV irradiation to one side of the PVC sheet decreased the levels of DEHP migration from the sheets by almost 50%, although the amount of DEHP content in PVC sheet was observed no significant change. On the other hand, the levels of DEHP migrating from the inner surface of PVC tubing UV-irradiated from the outer surface were not decreased compared with the control. Therefore, the surface structure was examined by conducting Fourier transform infrared spectroscopy (FT-IR), electron spectroscopy for chemical analysis (ESCA) and static angle of contact measurement. In FT-IR analysis, we found that the UV-irradiated PVC sheets were exhibited broadened absorption bands with time. In ESCA analysis, the chlorine content was decreased and the oxygen content was increased with time in UV-irradiated PVC sheets. Moreover, the other treated PVC sheets shows no significant change compared with the non-UV-irradiated PVC sheet. Therefore, the surface structure of the UV-irradiated PVC sheet was changed. As a result, the migration of DEHP from PVC products can be decreased with simple treatment, such as UV-irradiation. This could be a useful method to develop novel PVC products.

The effects of the γ-irradiatted PLLA on the osteoblasts and apatite formation were investigated in vitro. The PLLA sheet was γ-ray irradiated at the dose of 10, 25 or 50 kGy. The mouse osteoblast-like MC3T3-E1 cells and normal human osteoblast NHOst cells were micromass cultured on the PLLA sheet for 2 weeks, and then the proliferation and differentiation of the cells were determined. The proliferations of MC3T3-E1 and NHOst cells hardly changed with increasing irradiation dose. However, the differentiations of MC3T3-E1 and NHOst cells increased with irradiation dose. On the other hand, the surface of the PLLA sheet after soaking in the medium without the cells was characterized by SEM, EDX, FT-IR and XPS. The hydroxyapatite was formed on the surface of the PLLA sheet after soaking, and the amount of hydroxyapatite increased with irradiation dose. In summary, the γ-irradiated PLLA increased the differentiation of osteoblasts and also increased apatite-forming ability even without the osteoblasts. The osteoblast differentiation was enhanced well in the apatite formation on the surface of PLLA after the γ-irradiation.

We tested extracts of custom-made natural rubber samples for cytotoxicity using V79 cells and for chromosome aberration (CA) induction using CHL cells in compliance with the Japanese guidelines for basic biological tests of medical materials and devices. The samples were formulated with a high level of 2-mercaptobenzothiazole (MBT) (A); a low level of MBT (B); or zinc dibutyldithiocarbamate (ZDBC) (C). In the CA test, MBT induced mainly polyploidy, including endoreduplication, and ZDBC induced structural CAs. In the cytotoxicity test, culture medium extracts of A, B, and C suppressed colony formation to 50% of the control value at 53.1%, 94.3%, and >100%, respectively. Culture medium extracts of sample A induced polyploidy and structural CAs in the absence of an exogenous metabolic activation system (S9 mix), but at lower concentrations in its presence, indicating the existence of other leachable promutagens. The extracts of sample B induced structural CAs at the highest concentration and only with S9 mix. Sample C was negative. The facts suggest that sample A may be a candidate for a positive control for genotoxicity tests. The high frequency of polyploidy induced by sample A was not predicted by MBT, suggesting the usefulness of the test for safety evaluation of medical devices. Numerical CAs induced by MBT and sample A are discussed.

Azo colorants that generate primary aromatic amines (PAAs) have been recently deliberated as a controlled harmful substance by the "Act on the Control of Household Products Containing Harmful Substances" in Japan. Therefore, we examined an identification test for 22 kinds of PAAs originating from the azo colorants in commercial textile products and leather products using high performance liquid chromatography (HPLC). When a PAAs standard solution containing 2,4-xylidine and 2,6-xylidine was analyzed using the condition according to EN14362-1:2012 at 240 nm as a basic condition, we observed enough separation for all the PAAs to identify. However, in the some sample solutions, the peaks of several PAAs were overlapped with the interference peaks, and their identifications were difficult. In these cases, some PAAs were able to identify by alteration to suitable wavelength. Furthermore, the retention time of almost PAAs and interference peaks were changed by using acetonitrile as the organic solvent in eluent or phenyl type column. These modifications were helpful for identification of PAA which was overlapped to interference substances by the basic condition. Thus, we suggest the HPLC condition for an identification test is in accordance to that described in EN14362-1:2013. And we propose that the HPLC condition can be modified as necessary.

Normal human osteoblast NHOst cells were cultured on the dental Au–Ag–Pd casting alloy using the micromass culture, and the proliferation and differentiation of NHOst cells were determined. 13Au–58Ag–21Pd which met JIS T 6106 and 10Au–62Ag–13Pd which did not meet JIS T 6106 were used in this experiment. 10Au–62Ag–13Pd contained Cu more than 13Au–58Ag–21Pd. The proliferation and differentiation of NHOst cells cultured on 10Au–62Ag–13Pd were significantly decreased more than those on 13Au–58Ag–21Pd, respectively. It was suggested that the content of Cu caused the difference in the proliferation and differentiation between NHOst cells cultured on 13Au–58Ag–21Pd and that on 10Au–62Ag–13Pd. Moreover, the cytotoxicity of the Au–Ag–Pd alloy was evaluated by the MEM elution assay and the colony assay using fibroblast L929 and V79 cells, in order to compare with the cytological effects of the alloy on NHOst cells. 13Au–58Ag–21Pd showed no cytotoxicity to L929 and V79 cells in the MEM elution assay and the colony assay. However, 10Au–62Ag–13Pd showed extremely weak cytotoxicity to L929 cells and weak cytotoxicity to V79 cells in the colony assay, though 10Au–62Ag–13Pd showed no cytotoxicity to L929 cells in the MEM elution assay. NHOst cells expressed the toxicity to the proliferation and differentiation on 10Au–62Ag–13Pd clearly. The in vitro toxicity test based on the proliferation and differentiation of NHOst cells was useful to evaluate the toxicity of medical materials.

Recently, cases of contact dermatitis that were related to the use of polyvinyl alcohol (PVA) cooling towels containing isothiazolinone preservatives were reported in Japan. The aim of this investigation was to analyze the concentrations of five different isothiazolinone compounds present in PVA towels and to assess the effectiveness of washing in removing the preservatives from new towels prior to being used for the first time. Twenty-seven PVA towels were used in this study. Two groups (i.e., laboratory-simulation and volunteer) of washing experiments were conducted to evaluate the effect of washing procedures. Qualitative and quantitative analyses were performed by LC/MS/MS, which detected 2-methyl-4-isothiazolin-3-one (MI) and 5-chloro-2-methyl-4-isothaizolin-3-one (CMI) in 23 samples (MI: 0.29-154 μg g-wet(-1), CMI: 2.2-467 μg g-wet(-1)), 2-n-octyl-4-isothiazolin-3-one (OIT) in one sample (478 μg g-wet(-1)). The compounds 4,5-Dichloro-2-n-octyl-4-isothiazolin-3-one (2Cl-OIT) and 1,2-benzisothiazolin-3-one (BIT) were not detected in all samples. We confirmed the presence of residual MI, CMI, and OIT in the washed towels, and the residual-to-original content ratio of OIT was higher than that of MI and CMI in PVA towels, due to the higher hydrophobicity of OIT than MI and CMI. A concern has been raised about the occurrence of contact dermatitis being caused by the use of PVA towels. It is suggested that a detailed description of isothiazolinone preservatives in PVA towels and an effective washing procedure for the removal of these preservatives should be provided by the manufacturer. Further, alternative non-sensitizing preservatives might be considered for the manufacture of PVA cooling towels in the future.

A method for the determination of 2-mercaptobenzimidazole (MBI) type antioxidants in rubber products by HPLC was developed. The analytical conditions to separate 2-mercapto-4-methylbenzimidazole (4MMBI) from 2-mercapto-5-methylbenzimidazole (5MMBI) by HPLC and to extract MBI type antioxidants from rubber products were examined in detail. The recommended method was as follows. MBI type antioxidants was extracted with methanol five times. The evaporation residue of the extract was dissolved in water and injected to HPLC. The HPLC conditions were as follows : column, CAPCELL PAK C18 (4.6 i. d.×250mm) ; column temperature, 40°C ; mobile phase, 0.1 M phosphate buffer (pH 6.5)-acetonitrile (87 : 13) ; flow rate, 1.0ml/min ; detection wavelength, 304nm (MBI, 4MMBI) and 307 nm (5MMBI). This method was applied to determine MBI type antioxidants in 12 commercial rubber boots. Consequently, 11.5-67.7 μg/g of MBI was detected in 4 samples, but 4MMBI and 5MMBI were not detected in any samples.

In order to evaluate the safety of antimicrobial/deodorant agents, a research on totally 1113 kinds of commercially available products in 47 stores has been performed in cooperation with the public health institutes and the administrative departments of Osaka prefecture for 6 years since 1991. For the classification of these research products, a new classification on household products was made. Furthermore, the results of this research were gathered in"the data base (DB) on commercially available products research, "which is a component of"the DB system on antimicrobial/deodorant agents."The DB system includes the DB on active ingredients (AIs) and so on. As a result of this research, AIs were directly shown in the labellings of 226 out of 1113 products investigated (20.3%). And then, AIs could be estimated from the names of processing in the labellings of 210 out of 1113 products (18.9%). Namely, consumers may be informed of AIs in only 39.2% of processed products. In other words, the results of our research revealed that AIs of antimicrobial/deodorant agents in commercially available products were inadequately informed of package labellings. Therefore, consumers cannot obtain enough information on antimicrobial/deodorant agents in commercially available products. Moreover, the results of this research have been utilized in the beginning of other investigations such as analytical investigation and so on, and have been used for the maintenance of the safety management of household products.

The chemical structure of 2-mercaptobenzimidazole (2-MBI), which is widely used as a rubber antioxidant, is partially similar to those of thiourea (TU) and ethylenethiourea (ETU), both potent thyrotoxic compounds. In order to determine the oral toxicity of 2-MBI, a 28-day repeated dose toxicity study in Wistar rats followed by observation over a 14-day recovery period was conducted at dose levels of 2, 10 and 50 mg/kg 2-MBI administered by gavage. No toxic deaths occurred due to 2-MBI treatment. Decreases of body weight gain and food consumption in the 50 mg/kg dose group were observed during the second half of the treatment period. In addition, hematological examination and serum biochemical tests revealed decreased white blood cells and hemoglobin and increased serum urea nitrogen, cholesterol, phospholipid, gamma-glutamyl transpeptidase and the Na+/K+ ratio in the 50 mg/kg dose group. Marked thyroid enlargement (to 10 fold the control weight), histopathologically associated with diffuse hyperplasia of follicles with decreased colloid and thickening of the fibrous capsule, was found. Reduction in thymus weight was also observed in a dose-dependent manner, without significant histopathological alteration. The non-observed effect level (NOEL) of 2-MBI in this gavage study was found to be less than 2 mg/kg/day based on the significant decrease in thymus weight in the 2 mg/kg 2-MBI treatment group. In an ancillary study, measurement of serum levels of T3, T4 and TSH, and thyroid weight after gavage treatment with 0.15 and 0.3 mmol/kg of three antithyroid compounds for 14 days revealed a more potent antithyroid effect for 2-MBI than for TU or ETU.

The official analytical method for tris(2,3-dibromopropyl)phosphate (TDBPP), which is banned from use in textile products by the "Act on Control of Household Products Containing Harmful Substances", requires revision. This study examined an analytical method for TDBPP by GC/MS using a capillary column. Thermal decomposition of TDBPP was observed by GC/MS measurement using capillary column, unlike in the case of gas chromatography/flame photometric detector (GC/FPD) measurement based on a direct injection method using a capillary megabore column. A quadratic curve, Y=2572X(1.416), was obtained for the calibration curve of GC/FPD in the concentration range 2.0-100 μg/mL. The detection limit was 1.0 μg/mL under S/N=3. The reproducibility for repetitive injections was satisfactory. A pretreatment method was established using methanol extraction, followed by liquid-liquid partition and purification with a florisil cartridge column. The recovery rate of this method was ～100%. TDBPP was not detected in any of the five commercial products that this study analyzed. To understand the cause of TDBPP decomposition during GC/MS (electron ionization; EI) measurement using capillary column, GC/MS (chemical ionization; CI), GC/FPD, and gas chromatography/flame ionization detector (GC/FID) measurements were conducted. It was suggested that TDBPP might thermally decompose both during GC injection, especially through a splitless injection method, and in the column or ion sources. To attempt GC/MS measurement, an injection part comprising quartz liner was used and the column length was halved (15 m); thus, only one peak could be obtained.

Contraventions of regulations regarding primary aromatic amines (PAAs) originating from azo dyes in commercial textile products and leather products in European Union (EU), notified in the period between 2006 and 2012 were collected from the Rapid Alert System for non-food consumer products (RAPEX), were characterized. Various types of products (clothes, footwear, bedding, etc.) and their raw materials (cotton, silk, viscose, leather, etc.) were reported to have contravened the regulations. The contravention frequencies for products made in China and India were higher than those for other countries. Ten percentage of the country in which the reported products were produced was unknown. The notification frequencies for benzidine and 4-aminoazobenzene were higher than those for other PAAs. Contravention of regulations regarding benzidine, 4-aminoazobenzene, and 3,3'-dimethoxybenzidine were notified every year. Contraventions of regulations regarding five PAAs--classified as IARC group 1--were notified one or several times. Since the scale of the survey conducted in Japan were small compared with RAPEX, it is necessary that many kinds and number of products should be surveyed in Japan. In addition, it is also necessary to pay attention to 4-aminoazobenzene, while it has not been detected in the previous studies conducted in Japan.

This study deals with the development and performance evaluation of a positive reference material for hemolysis testing, which is used for evaluating the biological safety of medical devices. Genapol X-080, a nonionic detergent, was selected as a candidate hemolytic substance in a survey of 23 chemical compounds; it showed significant hemolytic activity against rabbit defibrinated blood at concentrations more than 20 µg/mL. A polyvinyl chloride (PVC) sheet spiked with 0.6% (w/w) of the compound exhibited weak hemolytic activity in direct contact and/or extract-based assays after 4 h incubation at 37°C. A PVC sheet containing 5.8% (w/w) Genapol X-080 induced complete hemolysis in both assays. The amount of Genapol X-080 eluted from each PVC sheet during hemolysis testing using the direct contact method increased time-dependently and reached 25.6 (former sheet) or 1154 (later sheet) µg/mL after 4 h incubation, which was similar to or much higher than the critical micelle concentration, respectively. Similar elution behavior was observed using the extract-based method, and the Genapol X-080 content in test solutions prepared by autoclave extraction of both sheets was 22.5 and 358 µg/mL, respectively, indicating a clear relationship between the degree of hemolytic activity and the eluted amount of Genapol X-080. Thus, a PVC sheet spiked with a compound exhibiting different hemolytic activity depending on its concentration may be useful as a positive reference material to validate the hemolysis tests.

The aim of this study is to identify a plasticizer that is effective in the suppression of the autohemolysis of the stored blood and can be used to replace di(2-ethylhexyl) phthalate (DEHP) in blood containers. The results of hemolysis test using mannitol-adenine-phosphate/red cell concentrates (MAP/RCC) spiked with plasticizers included phthalate, phthalate-like, trimeliate, citrate, and adipate derivatives revealed that di-isononyl-cyclohexane-1,2-dicarboxylate (Hexamoll(®) DINCH), di(2-ethylhexyl)-1,2,3,6-tetrahydro-phthalate (DOTP), and diisodecyl phthalate (DIDP) exhibited a hemolysis suppression effect almost equal to that of DEHP, but not other plasticizers. This finding suggested that the presence of 2 carboxy-ester groups at the ortho position on a 6-membered ring of carbon atoms may be required to exhibit such an effect. The hemolytic ratios of MAP/RCC-soaked polyvinyl chloride (PVC) sheets containing DEHP or different amounts of DINCH or DOTP were reduced to 10.9%, 9.2-12.4%, and 5.2-7.8%, respectively (MAP/RCC alone, 28.2%) after 10 weeks of incubation. The amount of plasticizer eluted from the PVC sheet was 53.1, 26.1-36.5, and 78.4-150 µg/mL for DEHP, DINCH, and DOTP, respectively. PVC sheets spiked with DIDP did not suppress the hemolysis induced by MAP/RCC because of low leachability (4.8-6.0 µg/mL). These results suggested that a specific structure of the plasticizer and the concentrations of least more than ∼10 µg/mL were required to suppress hemolysis due to MAP/RCC.

The use of triphenyltin (TPT) and tributyltin (TBT) in some household products is banned by "Act on the Control of Household Products Containing Harmful Substances" in Japan. To revise the official analytical method, the method for detecting these organotin compounds was examined in six laboratories using a textile product, water-based adhesive, oil-based paint, which contained known amounts of TPT and TBT (0.1, 1.0, 10 μg/g). TPT and TBT were measured by GC-MS after ethyl-derivation with sodium tetraethylborate. The TBT recoveries in the samples were 70-120%. The TPT recoveries in the water-based adhesive samples were 80-110%, while its concentrations in the textile product and oil-based paint samples decreased because of dephenylation during storage. However, the precision of the method examined was satisfactory because most coefficients of variation for TPT and TBT in the samples were less than 10%. Furthermore, the revised method was able to detect concentrations lower than the officially regulated value. However, the sample matrix and the condition of analytical instrument might affect the estimated TPT and TBT concentrations. Therefore, the revised method may not be suitable for quantitative tests; rather, it can be employed to judge the acceptable levels of these organotin compounds by comparing the values of control sample containing regulated amounts of TPT and TBT with those for an unknown sample, with deuterated TPT and TBT as surrogate substances. It is desirable that TPT in textile and oil-based paint samples are analyzed immediately after the samples obtained because of the decomposition of TPT.

This study examines the chemical, physicochemical, and biological properties of PVC sheets treated with UV irradiation on their surfaces to suppress the elution of a plasticizer, di-(2-ethylhexyl) phthalate (DEHP), for developing novel polyvinyl chloride (PVC) medical devices. The PVC sheets irradiated under conditions 1 (52.5 μW/cm(2), 136 J/cm(2)) and 2 (0.45 mW/cm(2), 972 J/cm(2)) exhibited considerable toxicity in cytotoxicity tests and chromosome aberration tests due to the generation of DEHP oxidants, but no toxicity was detected in the PVC sheet irradiated under condition 3 (8.3 mW/cm(2), 134 J/cm(2)). The release of DEHP from the surface irradiated under condition 3 was significantly suppressed, and mono-(2-ethylhexyl) phthalate (MEHP) converted from a portion of DEHP could be easily removed from the surface by washing with methanol. The physicochemical properties of the surface regarding the suppression of DEHP elution remained stable through all sterilizations tested, but MEHP elution was partially recrudesced by the sterilizations except for gamma irradiation. These results indicated that UV irradiation using a strong UV-source over a short time (condition 3) followed by methanol washing and gamma sterilization may be useful for preparing novel PVC products that did not elute plasticizers and do not exhibit toxicity originating from UV irradiation.

The relationships between cell attachment, cytotoxicity and in vivo tissue response such as muscle inflammation and eye irritation were investigated using lens materials, radiation vulcanized natural rubber latex (RVNRL) and standard reference materials (SRM). Five RVNRL materials, which induced muscle tissue inflammation, were cytotoxic and also significantly decreased cell attachment activities. The cytotoxic potential of a SRM made of segmented polyurethane correlated well with the decrease in cell attachment activity. However, among lens materials, cytotoxicity was detected in fluorosilicone acrylate but not in poly(methylmethacrylate) (PMMA), although the cell attachment activity of PMMA was the lowest. Eye irritation induced by wearing various kinds of lenses correlated with cytotoxic potential (r = −0·90) but not with cell attachment activities. In conclusion, in vivo toxic tissue responses correlated well with cytotoxic potentials but not always with cell attachment activity. Our experimental results also showed that direct contact colony formation was mainly inhibited by cytotoxicity, but not the low activity of cell attachment.

5 cases of allergic contact dermatitis from rubber footwear were investigated by a combination of patch testing in patients and chemical analysis of causative rubber products. Our studies revealed 2-mercaptobenzothiazole (MBT) and benzothiazyl disulfide (MBTS) (typical allergenic accelerators) as causative chemicals in 3 cases from children's rubber shoes, ladies' rubber boots and ladies' canvas shoes. These 3 patients reacted to mercaptobenzothiazole-type accelerators including MBT and MBTS. MBT and MBTS were determined in each item of causative footwear by chemical analysis, including extraction by shaking with acetone-chloroform (1:1) mixture at room temperature and determination using reversed-phase high-performance liquid chromatography (RP-HPLC). Subsequently, we identified styrenated phenol (SP), a newly found allergenic antioxidant, as a causative chemical in a case from ladies' canvas shoes. The patient reacted to SP but not to MBT and MBTS, though SP, MBT and MBTS were determined in the causative shoes by gas chromatography (GC), GC-mass spectrometry (GC-MS) and HPLC. We also identified p-tert-butylphenol-formaldehyde resin (PTBP-F-R), (a known allergenic adhesive ingredient) as a causative chemical in a case from ladies' sneakers. The patient reacted to PTBP-F-R but not to p-tert-butylphenol (PTBP), MBT and MBTS. These 4 compounds were determined in the causative sneakers by GC, GC-MS and HPLC. Thus, our studies revealed that not only known allergens, such as MBT, MBTS and PTBP-F-R, but also a newly found one, such as SP, were important causes of allergic contact dermatitis from rubber footwear.

The indications on packages of paper bags for electric vacuum cleaners had antimicrobial agents changed. Benzimidazole and biguanide compounds were newly indicated as antimicrobial agents, and pyrethroid compounds were as insecticides. Thiabendazole and methyl benzimidazolylcarbamate were identified as benzimidazole compounds. In 3 or 2 out of 7 paper bags, these two compounds were determined in the ranges of concentration of 3-513 μg/g and 1-160 μg/g by high performance liquid chromatography (HPLC), respectively. In 1 out of 7 paper bags, as a biguanide compound, chlorhexidine was identified and determined in the concentration of 1300 μg/g by HPLC. On the other hand, in 5 out of 7 paper bags, as a pyrethroid compound, permethrin was identified and determined in the concentration of 82-6310 μg/g by gas chromatography. In most of the paper bags analyzed, larger amounts of these 4 compounds were present in the inner bags than in the outer bags.

A compositional gradient structure in hyaluronic acid (HA) and poly(N-isopropylacrylamide) (PIPAAm) blend film was self-organized from a homogeneous aqueous solution in a plasma-treated polystyrene dish (PTPSD), and the formation mechanisms of the gradient structure were studied by casting the same solution on PTPSD and a non-treated polystyrene dish (NTPSD) under ambient and vacuum conditions. The formation of a compositional gradient structure in HA/PIPAAm blend film was confirmed by scanning electron microscopy, energy dispersive X-ray (EDX) mapping analysis and step-scan photoacoustic Fourier transformed infrared spectroscopy (PAS-FT-IR) measurements. The EDX mapping measurements for Na element revealed that the HA component gradually decreases from the dish-side to the air-side of the film cast on PTPSD, while for the film cast on NTPSD no such obvious change was observed on the cross-section. Further studies on the films prepared on PTPSD and NPTPSD under ambient and vacuum conditions demonstrated that the hydrophilic interaction and the solvent evaporation rate were the most significant factors leading to the formation of a compositional gradient structure in the HA/PIPAAm blend system.

The purpose of this study was to clarify the actual condition of 26 types of carcinogenic primary aromatic amines (PAAs) originated from azo dyes in commercial textile products in Japan. In the case of textiles made of various fibers of various colors, the fibers were separated by color and analyzed. A total of 86 textile products (117 samples) were analyzed. Twenty-one kinds of PAAs were detected in the samples and almost all the PAAs were detected at low concentrations. However, the concentrations of benzidine, 3,3'-dimethoxybenzidine, and 2,4-diaminotoluene (56-440 μg g⁻¹) in placemats made of cotton were found to exceed EU regulation limits of 30 μg g⁻¹. Although such placemats do not always come into contact with the user's skin, it is thought that they should be handled more carefully. Finally, 7 products (8 samples) contained PAAs at concentrations that exceeded the regulation limits. Two sample preparation methods (with and without solvent extraction) were performed on the same sample in order to compare the PAAs in samples in which it is difficult to separate the component materials, such as a cotton fabric that contained polyester fibers. In a comparison of the results obtained from the two methods, it was observed that the concentrations and/or kinds of PAAs detected in the samples were different. It was therefore thought that textile products that present this particular challenge should be analyzed by both methods.

In preparing for the revision of the authorized analytical method for tributyltin (TBT) and triphenyltin (TPT), which are banned from using according to the "Act on the Control of Household Products Containing Harmful Substances", an examination was conducted on the detection method of these substances using gas chromatography/mass spectrometry (GC/MS), after derivatizing them (ethyl-derivatizing method and hydrogen-derivatizing method). Ethyl-derivatized compounds had stability, which enabled the detection of TPT with a higher sensitivity. In addition, a preparation suitable for the following analytical objects was established: (1) textile products, (2) water-based products (such as water-based paint), (3) oil-based products (such as wax), and (4) adhesives. Addition-recovery experiments were conducted using the prescribed pretreatment method, when each surrogate substances (TBT-d27, TPT-d15) were added and the data were corrected, good recovery rates (94.5-118.6% in TBT, and 86.6-110.1% in TPT) were obtained. When TBT and TPT in 31 commercially available products were analyzed based on the developed analytical method, an adhesive showed 13.2 microg/g of TBT content, which exceeded the regulatory criterion (1 microg/g as tin). Next, when the same products with different manufacturing date were analyzed, TBT (10.2-10.8 microg/g), which exceeded the regulatory criterion, was detected in 4 products among 8 products, and simultaneously, a high concentration (over 1000 microg/g) of dibutyltin (DBT) was detected. It was suggested that TBT as an impurity of DBT remained, and the manufacturer chose the voluntary recall of the product. The new method is considered sufficiently applicable as a revised method for the conventionally authorized method.
