Objective In this study, the Hub genes that are closely associated with spermatogenesis in klinefelter syndrome (KS) patients were identified, mainly through transcriptomic and single-cell RNA sequencing (scRNA-seq)data. Methods Transcriptome data were obtained from 3 adult 4 fetal KS patients and 8 normal individuals. ScRNA-seq data were obtained from 1 adult KS patient and 4 normal individuals. The differentially expressed genes (DEGs) were analysed using limma's method. The testicular tissue-specific protein-protein interaction (PPI) networks were analysed through the NetworkAnalyst website. The CytoHubba plugin was used to draw Veen Diagram. Integration of scRNA-seq data using the Seurat package. Developmental trajectories of sperm cells and hub genes were constructed by using the monocle3. Results The results of the GSE103905 and GSE103613 transcriptome datasets show a total of 700 DEGs. Gene Ontology (GO) analysis focused on memory, methylation, ATPase activity and mitochondrion. Kyoto Encyclopedia of Genes and Genomes (KEGG) focused on metabolic pathways, huntington's disease and nicotinamide metabolism. Four Hub genes (KIF2C, MRPS2, RPS15, TSFM) that have not been reported to be associated with KS were identified. Single-cell sequencing data showed that only KIF2C and RPS15 were expressed in sperm cells. hub genes' Pseudotime analysis showed a decreasing trend in RPS15 expression and an increasing trend in KIF2C expression throughout sperm development. Conclusion In this study, two RPS15 and KIF2C were identified that may be closely associated with spermatogenesis in KS patients, but there may be differences in the mechanisms by which they affect sperm development.

A novel monodispersed CaCO

ObjectivesMesenchymal stem cells (MSCs) play important roles in multiple myeloma (MM) pathogenesis. Previous studies have discovered a group of MM-associated potential biomarkers in MSCs derived from bone marrow (BM-MSCs). However, no study of the bioinformatics analysis was conducted to explore the key genes and pathways of MSCs derived from adipose (AD-MSCs) in MM. The aim of this study was to screen potential biomarkers or therapeutic targets of AD-MSCs and BM-MSCs in MM.MethodsThe gene expression profiles of AD-MSCs (GSE133346) and BM-MSCs (GSE36474) were downloaded from Gene Expression Omnibus (GEO) database. Gene Oncology (GO) enrichment, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis and protein-protein interaction (PPI) network of differentially expressed genes (DEGs) were performed.ResultsA total of 456 common downregulated DEGs in two datasets were identified and the remaining DEGs in GSE133346 were further identified as specific DEGs of AD-MSCs. Furthermore, a PPI network of common downregulated DEGs was constructed and seven hub genes were identified. Importantly, cell cycle was the most significantly enrichment pathway both in AD-MSCs and BM-MSCs from MM patients.ConclusionWe identified key genes and pathways closely related with MM progression, which may act as potential biomarkers or therapeutic targets of MM.

Background: Previous studies have reported that roux-en Y gastric bypass surgery (RYGBS) can benefit patients with type 2 diabetes mellitus (T2DM). However, their conclusions are still inconsistent. Thus, this study will aim to assess the effect of RYGBS for patients with T2DM. Methods: In this study, the electronic databases of MEDLINE, EMBASE, CENTRAL, CINAHL, AMED, and CNKI from inceptions to the present without any limitations to language and publication status. All randomized controlled trials on assessing the effect of RYGBS for patients with T2DM will be included in this study. Two independent authors will carry out study search and selection according to the previous designed inclusion and exclusion criteria. At the same time, 2 authors will independently evaluate the risk of bias assessment by Cochrane risk of bias tool. Any disagreements between 2 authors will be solved by a third author through discussion. RevMan 5.3 software will be utilized for statistical analysis. Results: This study will summarize the most recent studies and will provide a deeper understanding about using the effect of RYGBS for patients with T2DM. Conclusions: The findings of this study will present the existing evidence for the effect of RYGBS for patients with T2DM. Systematic review registration: INPLASY202040127.

Background: A numerous studies have reported that obese patients (OP) are easily to have type 2 diabetes mellitus (T2DM). Although a variety of managements are available to treat such disorder, their efficacy is still limited. Previous studies have reported that laparoscopic sleeve gastrectomy (LSGT) can benefit OP with T2DM. However, no study specifically and systematically explores this topic. Thus, this study will assess the efficacy and complications of LSGT for the management of OP with T2DM. Methods: The search strategy will be performed in the electronic databases from inception to the March 31, 2020 without limitations of language and publication time: PUBMED, EMBASE, Cochrane Library, Scopus, Web of Science, CINAHL, AMED, WANGFANG, VIP, and CNKI. Two authors will independently identify the articles, collect the data, and assess the risk of bias using Cochrane risk of bias tool. We will invite a third author to solve any differences between two authors. We will use RevMan 5.3 software to investigate the statistical analysis. Results: This study will supply a high-quality synthesis of randomized controlled trials (RCTs) on the analysis of LSGT for the management of OP with T2DM. Conclusions: This study will help to build proposals that aim at providing high quality RCTs in the management of LSGT in OP with T2DM. Systematic review registration: INPLASY202040128.

Objective To observe the effects of sevoflurane post-conditioning on the expression of Aquaporin 8 (AQP8) and intestinal fatty acid binding protein (I-FABP), in order to investigate the protective role of sevoflurane post-conditioning on intestinal injury and its underlying mechanism. Methods Eighteen bama miniature pigs were randomly divided into three groups (6 each) using a random number table: control group (S group), hemorrhagic shock group (HS group), and sevoflurane post-coditioning group (Post/ Sev group). Experimental animals were fasted for 8 hours before surgery, and propofol 3mg/kg was given viathe ear vein. Endotracheal intubation was done when the animal fell asleep. Bloodletting from the femoral artery after anesthesia was done to reproduce hemorrhagic shock. In Post/Sev group, 2% sevoflurane was given by inhalation for 30min (post-conditioning) after successful reproduction of the model. Blood samples were collected prior to anesthesia (T0) and 30min (T1), 1h (T2), 1.5h (T3), 2h (T4), 3h (T5), 4h (T6) after hemorrhagic shock. The quantity of blood I-FABP and intestinal AQP8 levels were determined with ELISA. Water content in the intestinal tissue was determined by wet and dry weight method. Histopathological changes in the intestinal tissue were observed with HE staining. The aim of the present study was to explore the roles and possible molecular mechanism of the alleviating effect of sevoflurane pre‑treatment on the extracorporeal circulation and to investigate the possible involvement of the Toll‑like receptor (TLR3) signaling pathway. A total of 64 male Sprague Dawley rats were randomly divided into three groups: The sham operation group (H group; n=8), cardiopulmonary bypass (CPB) group (C group; n=24) and sevoflurane pre‑conditioning group (S group; n=32). The C group was subjected to tracheal intubation and mechanical ventilation, vessel puncture and catheter placement in the right femoral artery and right internal jugular vein, while no CPB was performed in the H group. The S group was pre‑treated with 2.4% sevoflurane for 1 h prior to establishing the CPB model. The CPB in the C and S groups was performed for 1 h. Blood of the rats was analyzed and clinical parameters were detected prior to, during and at various time‑points after CPB. In addition, eight rats from the C and S groups each were sacrificed at these time‑points and brain tissue samples were analyzed. The levels of the brain damage‑specific protein S100‑β as well as IL‑6 and IFN‑β in the serum were detected by ELISA; furthermore, the expression levels of TLR3 and TIR‑domain‑containing adapter‑inducing interferon‑β (TRIF) in the left hippocampus were assessed by ELISA and/or western blot analysis. Ribosomal protein S6 (rpS6) has long been regarded as one of the primary r-proteins that functions in the early stage of 40S subunit assembly, but its actual role is still obscure. The correct forming of 18S rRNA is a key step in the nuclear synthesis of 40S subunit. In this study, we demonstrate that rpS6 participates in the processing of 30S pre-rRNA to 18S rRNA only when its C-terminal five serines are phosphorylated, however, the process of entering the nucleus and then targeting the nucleolus does not dependent its phosphorylation. Remarkably, we also find that the aggregation of rpS6 at the nucleolus correlates to the phasing of cell cycle, beginning to concentrate in the nucleolus at later S phase and disaggregate at M phase.
