Abstract: Nuclear reactors are uniquely powerful, abundant, and flavor-pure sources of antineutrinos that continue to play a vital role in the US neutrino physics program. The US reactor antineutrino physics community is a diverse interest group encompassing many detection technologies and many particle physics topics, including Standard Model and short-baseline oscillations, BSM physics searches, and reactor flux and spectrum modeling. The community's aims offer strong complimentary with numerous aspects of the wider US neutrino program and have direct relevance to most of the topical sub-groups composing the Snowmass 2021 Neutrino Frontier. Reactor neutrino experiments also have a direct societal impact and have become a strong workforce and technology development pipeline for DOE National Laboratories and universities. Abstract: Candida auris is an enigmatic yeast that provides substantial global risk in health care facilities and intensive care units. A unique phenotype exhibited by certain isolates of C. auris is their ability to form small clusters of cells known as aggregates, which have been to a limited extent described in the context of pathogenic traits. In this study, we screened several nonaggregative and aggregative C. auris isolates for biofilm formation, where we observed a level of heterogeneity among the different phenotypes. Next, we utilized an RNA sequencing approach to investigate the transcriptional responses during biofilm formation of a nonaggregative and aggregative isolate of the initial pool. Observations from these analyses indicate unique transcriptional profiles in the two isolates, with several genes identified relating to proteins involved in adhesion and invasion of the host in other fungal species. From these findings, we investigated for the first time the fungal recognition and inflammatory responses of a three-dimensional skin epithelial model to these isolates. In these models, a wound was induced to mimic a portal of entry for C. auris We show that both phenotypes elicited minimal response in the model minus induction of the wound, yet in the wounded tissue, both phenotypes induced a greater response, with the aggregative isolate more proinflammatory. This capacity of aggregative C. auris biofilms to generate such responses in the wounded skin highlights how this opportunistic yeast is a high risk within the intensive care environment where susceptible patients have multiple indwelling lines. Abstract: Background: Existing standardized biofilm assays focus on simple mono-species or bacterial-only models. Incorporating Candida albicans into complex biofilm models can offer a more appropriate and relevant polymicrobial biofilm for the development of oral health products. Aims: This study aimed to assess the importance of interkingdom interactions in polymicrobial oral biofilm systems with or without C. albicans, and test how these models respond to oral therapeutic challenges in vitro. Materials and Methods: Polymicrobial biofilms (two models containing 5 and 10 bacterial species, respectively) were created in parallel in the presence and absence of C. albicans and challenged using clinically relevant antimicrobials. The metabolic profiles and biomasses of these complex biofilms were estimated using resazurin dye and crystal violet stain, respectively. Quantitative PCR was utilized to assess compositional changes in microbial load. Additional assays, for measurements of pH and lactate, were included to monitor fluctuations in virulence “biomarkers.” Results: An increased level of metabolic activity and biomass in the presence of C. albicans was observed. Bacterial load was increased by more than a factor of 10 in the presence of C. albicans. Assays showed inclusion of C. albicans impacted the biofilm virulence profiles. C. albicans did not affect the biofilms’ responses to the short-term incubations with different treatments. Conclusions: The interkingdom biofilms described herein are structurally robust and exhibit all the hallmarks of a reproducible model. Abstract: Understanding the role of biofilms in recurrent vulvovaginal candidiasis (RVVC) and their impact on therapyBackground: Vulvovaginal candidiasis (VVC) is a global health problem, affecting up to 75% of healthy women at least once during their lifetime. Although the majority of cases are treated effectively, ~10% of patients encounter treatment failure and complain of more than 4 episodes of VVC per year, defined clinically as recurrent VVC (RVVC). It has been hypothesised that RVVC proves difficult to treat due to the presence of biofilms within the vaginal mucosa, however definitive evidence for this is lacking. Therefore, the role of biofilms in RVVC needs to be evaluated to aid the management of this infection.Materials/Methods: An anonymised series of high vaginal swabs (HVS, n=300) were obtained from woman reporting VVC, for at least the second time. The causative organism was identified using MALDI-TOF and biofilm formation assessed using crystal violet and metabolic dyes. Antifungal sensitivity testing was performed on planktonic cells and mature (24 h) biofilms, using antifungals currently used clinically (fluconazole, miconazole and clotrimazole) as well as the alternative natural agent carbohydrate-derived fulvic acid (CHD-FA).Results: Only 71% of isolates were identified as C. albicans, with the remaining isolates including C. glabrata, C. dubliniensis, and C. parapsilosis. Biofilm assessment showed vaginal isolates were able to form differential biofilms regardless of species; with C. albicans isolates eliciting the greatest variation in biomass. Fluconazole, the first line antifungal used to treat VVC, was ineffective against most isolates, with planktonic MICu2019s ranging from 1-32uf06dg/mL however when this agent was tested against sessile populations the MICu2019s ranged from 16->32uf06dg/mL. In contrast, the natural agent CHD-FA was shown to be equally effective against both planktonic and sessile cells with an MIC of 0.25%. Conclusions: Biofilm heterogeneity may contribute to the management of VVC infections, as these communities are known to be notoriously recalcitrant to antifungal therapy, and has been shown to correlate with in vitro antifungal therapy. Furthermore, the effectiveness of the natural agent CHD-FA, highlights its potential role to be used in the clinical management of these infections.

Abstract: Polymicrobial inter-kingdom biofilm infections represent a clinical management conundrum. The presence of co-isolation of bacteria and fungi complicates the ability to routinely administer single antimicrobial regimens, and synergy between the microorganisms influences infection severity. We therefore investigated the nosocomial pathogens Staphylococcus aureus and Candida albicans with respect to antimicrobial intervention. We characterized the interaction using biofilm assays and evaluated the effect of miconazole treatment using in vitro and in vivo assays. Finally, we assessed the impact of biofilm extracellular matrix (ECM) on these interactions. Data indicated that the C. albicans mycofilms supported adhesion and colonization by S. aureus through close interactions with hyphal elements, significantly increasing S. aureus biofilm formation throughout biofilm maturation. Miconazole sensitivity was shown to be reduced in both mono- and dual-species biofilms compared to planktonic cells. Within a three-dimensional biofilm model sensitivity was also hindered. Galleria mellonella survival analysis showed both enhanced pathogenicity of the dual-species infection, which was concomitantly desensitized to miconazole treatment. Analysis of the ECM revealed the importance of extracellular DNA, which supported the adhesion of S. aureus and the development of the dual-species biofilm structures. Collectively, these data highlight the clinical importance of dual-species inter-kingdom biofilm infections, though also provides translational opportunities to manage them more effectively.

Abstract: Polylactide (PLA) is a biodegradable polyester formed by the ring-opening polymerization of lactide, the cyclic dimer of lactic acid. Many of the physical properties of PLA are influenced by the amount and distribution of the R and S stereocenters in the polymer chain. NMR spectroscopy is the most common technique used to determine the stereosequence distribution of the polymer. The correct determination of the stereosequence distribution is contingent upon the assignment of the peaks in the NMR spectrum to specific stereosequences. Recently, alternative assignments to the commonly accepted stereosequence assignments have been proposed. If any of these alternative assignments are found to be correct, it would invalidate the conclusions of much of the previous work in understanding the kinetics of lactide polymerization and determining the stereoselectivity of new catalysts. The most significant problem is reconciling the commonly accepted peak assignments, which were based upon statistical probabilities, with contradictory connectivity data observed in a HETCOR NMR experiment. To describe the directionality in PLA, we have modified the nomenclature used to describe directionality in peptides and proteins. For PLA, the end containing the carboxylic group is referred to as the C terminus, and the end with the hydroxyl group is the O terminus. We had previously proposed that the central pairwise relationship (isotactic or syndiotactic, denoted i or s) in the ¹H NMR spectrum is determined by the stereocenter in the lactic acid unit attached to the O terminus and that in the ¹³C NMR spectrum it is determined by the central pairwise relationship of the stereocenter in the lactic acid unit attached to the C terminus. One- and two-dimensional NMR techniques, in combination with selective isotopic labeling, were used to show that this relationship is correct and that the commonly accepted assignments are correct. In addition, all of the nondegenerate resonances in the ¹H and ¹³C NMR spectrum of polylactide at the tetrad stereosequence level have been assigned.

Abstract: Poly(lactide) is synthesized by ring-opening copolymerization of various combinations of l-lactide, d-lactide, and meso-lactide. The influence of the stereochemical differences of the three lactides on the kinetics of melt copolymerization was determined by monitoring the change in the polymer stereosequence distribution as a function of conversion. The living stereo-copolymerization catalyzed by Sn(II) bis2-ethylhexanoate (Sn(II) octoate) in 1:10 000 molar ratio at 180 °C was investigated by comparing to the values predicted by a reversible polymerization scheme that achieves equilibrium at ~95% conversion. The stereosequence distribution was measured by high-resolution 500 MHz ¹H NMR. A preference for syndiotactic addition was observed and is thought to be due to steric hindrance at the growing site of the polymer. Furthermore, the syndiotactic preference decreased as the polymerization proceeded in a batch process. Similar behavior was observed during lactide polymerization catalyzed by butyl Sn(IV) tris2-ethylhexanoate. The increasingly random addition during the polymerization process is due to the interplay of kinetic and equilibrium effects. Kinetic effects control the stereochemistry during the early stages of the polymerization whereas equilibrium effects dominate at later stages. Viscosity changes in the melt additionally influence the stereospecificity by increasing the residence time of the lactide at the active site. Monte Carlo calculations were used for stereosequence predictions because analytical equations are not available for these complicated kinetics.

The intensity distribution of the various stereosequence resonances in the NMR spectra indicates a preference for syndiotactic addition during the polymerization process. Minimal evidence of transesterification was observed for these polymerization conditions.

Abstract: Solid-state ¹³C CP-MAS NMR spectroscopy has been used to investigate highly crystalline, partially crystalline, and amorphous poly(l-lactide) (PLLA). The amorphous domains have broad resonances with Gaussian line shapes, while the crystalline domains have narrower resonances with a high degree of Lorentzian character. Spectra of highly crystalline PLLA have at least five distinct resonances for the carbonyl and methine functional groups in the polymer, indicating five or more crystallographically inequivalent sites in the crystalline domains of the polymer. For the amorphous polymer, broad Gaussian peaks are observed for each of the methyl, methine, and carbonyl functional groups. Spectra of partially crystalline PLLA contains spectral features of both domains, and furthermore, the spectra appear to be sensitive to both short range (nanometer) and long range (micrometer) order within the polymer. A method to extract the information of crystallinity and morphology from the solid-state CP-MAS NMR spectra of partially crystalline PLLA is discussed.

Abstract: Small-angle X-ray scattering (SAXS), optical microscopy and differential scanning calorimetry were used to establish relationships between crystallization, microstructure, and equilibrium melting temperature (Tm⁰) for random copolymers of poly(l-lactide-co-meso-lactide). Tm⁰s derived using the Gibbs−Thomson and data-fitting²³ approaches were found to be in excellent agreement and decreased significantly with increasing meso-lactide concentration. Analysis of these equilibrium melting temperatures using models for copolymer crystallization leads to the conclusion that the meso defects are rejected from the crystalline regions. From analysis of the SAXS data, it was concluded that the copolymers contained significant interfibrillar regions whose concentration increased with higher comonomer content. Spherulitic growth rates were strongly dependent on meso content and were analyzed using the Lauritzen−Hoffman kinetic theory of crystal growth.
