Abstract

Introduction: Following heart transplantation, a cascade of immunological responses is initiated influencing the clinical outcome and long-term survival of the transplanted patients. The anti-inflammatory cytokine interleukin-10 (IL-10) was shown to be elevated in the blood of heart transplant recipients directly after transplantation but the releasing cell populations and the composition of lymphocyte subsets following transplantation have not been thoroughly studied. Methods: We identified immune cells by immunophenotyping and analyzed intracellular IL-10 production in peripheral blood mononuclear cells (PBMC) of heart transplanted patients (n= 17) before, directly after and 24h post heart transplantation. The cells were stimulated with lipopolysaccharide or PMA/Ionomycin to enhance cytokine production within leukocytes in vitro. Results and discussion: We demonstrate that intermediate monocytes (CD14highCD16+), but not CD8+ T cells, CD4+ T cells, CD56+ NK cells or CD20+ B cells appeared to be the major IL-10 producers within patients PBMC following heart transplantation. Consequently, the absolute monocyte count and the ratio of intermediate monocytes to classical monocytes (CD14+CD16-) were specifically increased in comparison to pre transplant levels. Hence, this population of monocytes, which has not been in the focus of heart transplantation so far, may be an important modulator of clinical outcome and long-term survival of heart transplant recipients. Alteration of blood-circulating monocytes towards a CD14highCD16+ phenotype could therefore shift the pro-inflammatory immune response towards induction of graft tolerance, and may pave the way for the optimization of immunosuppression. Abstract

New ideas are important for companies to evolve and overcome problems. This dissertation focuses on utilizing the companies’ potential for these ideas: their employees. It examines in detail extra-role creativity as a subdimension of creativity. Extra-role creativity describes the discretionary generation of new and useful ideas beyond one’s job role expectations. This subdimension has, to our knowledge, only been theoretically proposed, but not empirically assessed. The dissertation further evaluates its relation to innovation—the implementation of ideas—and performance. In addition, it examines social factors relevant to fostering extra-role creativity, innovation, and performance. It specifically looks at the influences of the direct social environment of an employee exerted through leadership behavior and different team climate dimensions. Taken together, this dissertation addresses three research questions. Each manuscript relates to one or more of the following research questions: 1) How is extra-role creativity perceived by employees? 2) What is the connection between extra-role creativity and innovation as well as between extra-role creativity and performance? 3) Which leadership behaviors or team climate dimensions are relevant for extra-role creativity, innovation, and performance?, Neue Ideen sind wichtig für Unternehmen, um sich kontinuierlich weiter zu entwickeln und Probleme zu überwinden. Diese Dissertation befasst sich mit der Nutzung des internen Potenzials der Unternehmen für diese Ideen: die Ideen der Mitarbeiterinnen und Mitarbeiter. Sie untersucht im Detail Extra-Rollen Kreativität als Subdimension von Kreativität. Extra-Rollen Kreativität beschreibt die Entwicklung neuer und nützlicher Ideen jenseits der Berufsrolle. Diese Subdimension ist nach unserem Wissen bisher nur theoretisch dargelegt, aber noch nicht empirisch erfasst worden. Abstract

The role of endothelial cells in the pathophysiology of antibody‐mediated rejection after renal transplantation has been widely investigated. We expand this scenario to the impact of epithelial cells on the microenvironment during rejection. Primary proximal tubular epithelial cells were stimulated via HLAclass I, CD155 and CD166 based on their potential signal‐transducing capacity to mediate back signaling after encounter with either T/NKcells or donor‐specific antibodies. Upon crosslinking of these ligands with mAbs, PTECsecreted IL‐6, CXCL1,8,10, CCL2, and sICAM‐1. These proteins were also released by PTECas consequence of a direct interaction with T/NKcells. Downmodulation of the receptor CD226 on effector cells confirmed the involvement of this receptor/ligand pair in back signaling. In vivo,CD155 and CD166 expression was detectable in proximal and distal tubuli of renal transplant biopsies, respectively. The composition of the protein microenvironment in these biopsies showed a substantial overlap with the PTECresponse. Cluster and principal component analyses of the microenvironment separated unsuspicious from rejection biopsies and, furthermore, ABMR,TCMR, and borderline rejection. In conclusion, our results provide evidence that epithelial cells may contribute to the rejection process and pave the way to a better understanding of the pathomechanisms of kidney allograft rejection. Proximal tubular epithelial cells respond to crosslinking of HLA class I molecules or direct interaction with allogeneic T and NK cells by secreting cytokines and chemokines that contribute to antibody‐ and T cell–mediated rejection, thus highlighting the impact of parenchymal cells to rejection episodes after kidney transplantation. 36. Leading performance: A moderated mediation analysis at the team level.

Abstract

Fostering team performance is an important leadership function. The underlying processes, however, are not well understood. We approach this question by integrating innovation research principles. Our multi-source study was conducted in two-waves in different German organizations and departments with a total of 609 employees in 84 teams. Regression analyses revealed that two team climate for innovation dimensions, (1) vision and (2) task orientation, mediate the relationship between transformational leadership and one year later team performance. This effect was stronger for teams with low autonomy. In these teams, (3) participative safety was a significant mediator as well. (4) Support for innovation, one of the strongest predictors for innovation, was not significant. Ways to enhance transformational leadership and team climate for innovation dimensions especially under circumstances when team members have little autonomy are discussed. [ABSTRACT FROM AUTHOR]
