Food-derived opioid peptides include digestive products derived from cereal and dairy diets. If these opioid peptides breach the intestinal barrier, typically linked to permeability and constrained biosynthesis of dipeptidyl peptidase-4 (DPP4), they can attach to opioid receptors. The widespread presence of opioid receptors spanning gut, brain, and internal organs is fundamental to the diverse and systemic effects of food-derived opioids, with effects being evidential across many health conditions. However, manifestation delays following low-intensity long-term exposure create major challenges for clinical trials. Accordingly, it has been easiest to demonstrate causal relationships in digestion-based research where some impacts occur rapidly. Within this environment, the role of the microbiome is evidential but challenging to further elucidate, with microbiome effects ranging across gut-condition indicators and modulators, and potentially as systemic causal factors. Elucidation requires a systemic framework that acknowledges that public-health effects of food-derived opioids are complex with varying genetic susceptibility and confounding factors, together with system-wide interactions and feedbacks. The specific role of the microbiome within this puzzle remains a medical frontier. The easiest albeit challenging nutritional strategy to modify risk is reduced intake of foods containing embedded opioids. In future, constituent modification within specific foods to reduce embedded opioids may become feasible.

This is the first systematic review, to our knowledge, of published studies investigating the gastrointestinal effects of A1-type bovine β-casein (A1) compared with A2-type bovine β-casein (A2). The review is relevant to nutrition practice given the increasing availability and promotion in a range of countries of dairy products free of A1 for both infant and adult nutrition. In vitro and in vivo studies (all species) were included. In vivo studies were limited to oral consumption. Inclusion criteria encompassed all English-language primary research studies, but not reviews, involving milk, fresh-milk products, β-casein, and β-casomorphins published through 12 April 2017. Studies involving cheese and fermented milk products were excluded. Only studies with a specific gastrointestinal focus were included. However, inclusion was not delimited by specific gastrointestinal outcome nor by a specific mechanism. Inclusion criteria were satisfied by 39 studies. In vivo consumption of A1 relative to A2 delays intestinal transit in rodents via an opioid-mediated mechanism. Rodent models also link consumption of A1 to the initiation of inflammatory response markers plus enhanced Toll-like receptor expression relative to both A2 and nonmilk controls. Although most rodent responses are confirmed as opioid-mediated, there is evidence that dipeptidyl peptidase 4 stimulation in the jejunum of rodents is via a nonopioid mechanism. In humans, there is evidence from a limited number of studies that A1 consumption is also associated with delayed intestinal transit (1 clinical study) and looser stool consistency (2 clinical studies). In addition, digestive discomfort is correlated with inflammatory markers in humans for A1 but not A2. Further research is required in humans to investigate the digestive function effects of A1 relative to A2 in different populations and dietary settings.

This paper analyses the diversity of goals and values amongst 26 Brazilian commercial-scale beef farmers previously identified as exhibiting progressive technology adoption behaviours, and considers the implications for the agricultural innovation system (AIS). Following the Q-methodology guidelines, four main sets of goals and values were identified and labelled: the Professional Farmer (PF), the Committed Environmentalist (CE), the Profit Maximiser (PM) and the Aspirant Top Farmer (ATF). All farmer types believed in operating the farm as a business and agreed with the notion that cattle production and nature conservation are compatible. The PF aimed to run the farm professionally, based on sound technical and managerial practices. The CE put a particularly strong emphasis on the long-term sustainability of the farming system. The PM focused on technical issues to pursue economic returns and efficiency, thereby also creating space for lifestyle objectives. The ATF sought physical farm and livestock excellence, was growth oriented, and sought recognition by peers. These four typologies have implications for the farmers' technology adoption and its promotion, e.g. they are likely to influence the uptake of particular types of innovations, and reinforce the importance of considering AIS within a social systems framework. This is the first Q-methodology study to demonstrate considerable diversity of values and goals within a subset of technology-adopting progressive farmers, with implications within agricultural innovation systems for the integration of goal diversity and adoption theory.

We compared the gastrointestinal effects of milk-based diets in which the β-casein component was either the A1 or A2 type in male Wistar rats fed the experimental diets for 36 or 84 h. Gastrointestinal transit time was significantly greater in the A1 group, as measured by titanium dioxide recovery in the last 24 h of feeding. Co-administration of naloxone decreased gastrointestinal transit time in the A1 diet group but not in the A2 diet group. Colonic myeloperoxidase and jejunal dipeptidyl peptidase (DPP)-4 activities were greater in the A1 group than in the A2 group. Naloxone attenuated the increase in myeloperoxidase activity but not that in DPP-4 activity in the A1 group. Naloxone did not affect myeloperoxidase activity or DPP-4 activity in the A2 group. These results confirm that A1 β-casein consumption has direct effects on gastrointestinal function via opioid-dependent (gastrointestinal transit and myeloperoxidase activity) and opioid-independent (DPP-4 activity) pathways.

The area of shrimps farmed in the Tam Giang Cau Hai Lagoon expanded dramatically from 1995 to 2004, but then declined steadily. An exploratory investigation of the shrimp supply chain conducted in 2009 revealed that well- established relational contracts between shrimp farmers and wholesalers had succumbed to environmental risk following persistent and widespread outbreaks of disease in the Lagoon. These outbreaks have been attributed to water pollution, which - in turn - has been linked to the rapid expansion and intensification of shrimp production. While privatisation of the lagoon bed encouraged farmers to adopt more intensive production methods, over- intensification can be attributed to the open access nature of lagoon water, which allows farmers to externalise the cost of their water pollution. This study evaluates a range of pollution abatement instruments against well-known criteria and concludes that transferable quotas for shrimp seed would be the most effective policy instrument given prevailing social, institutional, organisational and political conditions. Even so, the introduction and administration of seed quotas pose major challenges that would require co-management with local user groups.

Globally type 1 diabetes incidence is increasing. It is widely accepted that the pathophysiology of type 1 diabetes is influenced by environmental factors in people with specific human leukocyte antigen haplotypes. We propose that a complex interplay between dietary triggers, permissive gut factors and potentially other influencing factors underpins disease progression. We present evidence that A1 β-casein cows’ milk protein is a primary causal trigger of type 1 diabetes in individuals with genetic risk factors. Permissive gut factors (for example, aberrant mucosal immunity), intervene by impacting the gut’s environment and the mucosal barrier. Various influencing factors (for example, breastfeeding duration, exposure to other dietary triggers and vitamin D) modify the impact of triggers and permissive gut factors on disease. The power of the dominant trigger and permissive gut factors on disease is influenced by timing, magnitude and/or duration of exposure. Within this framework, removal of a dominant dietary trigger may profoundly affect type 1 diabetes incidence. We present epidemiological, animal-based, in vitro and theoretical evidence for A1 β-casein and its β-casomorphin-7 derivative as dominant causal triggers of type 1 diabetes. The effects of ordinary milk containing A1 and A2 β-casein and milk containing only the A2 β-casein warrant comparison in prospective trials.

At present, there is debate about the gastrointestinal effects of A1-type beta-casein protein in cows' milk compared with the progenitor A2 type. In vitro and animal studies suggest that digestion of A1 but not A2 beta-casein affects gastrointestinal motility and inflammation through the release of beta-casomorphin-7. We aimed to evaluate differences in gastrointestinal effects in a human adult population between milk containing A1 versus A2 beta-casein.Forty-one females and males were recruited into this double-blinded, randomised 8-week cross-over study. Participants underwent a 2-week dairy washout (rice milk replaced dairy), followed by 2 weeks of milk (750 ml/day) that contained beta-casein of either A1 or A2 type before undergoing a second washout followed by a final 2 weeks of the alternative A1 or A2 type milk.The A1 beta-casein milk led to significantly higher stool consistency values (Bristol Stool Scale) compared with the A2 beta-casein milk. There was also a significant positive association between abdominal pain and stool consistency on the A1 diet (r=0.520, P=0.001), but not the A2 diet (r=-0.13, P=0.43). The difference between these two correlations (0.52 versus -0.13) was highly significant (P0.001). Furthermore, some individuals may be susceptible to A1 beta-casein, as evidenced by higher faecal calprotectin values and associated intolerance measures.These preliminary results suggest differences in gastrointestinal responses in some adult humans consuming milk containing beta-casein of either the A1 or the A2 beta-casein type, but require confirmation in a larger study of participants with perceived intolerance to ordinary A1 beta-casein-containing milk.

A herd of rusa deer (Cervus timorensis) was monitored for 4 years at University of Queensland, Gatton College. Reported characteristics include reproductive cycles and performance, antler cycles, growth rates, live weights, carcass parameters, animal health, and behavior. Additional results obtained from private herds in Queensland are presented. Breeding stags and hinds at Gatton College were run together throughout the year. However, of mature hinds 84% calved in autumn between 4 March and 15 May. Most primiparous hinds gave birth between June and October at 15 to 18 months of age. Intercalving intervals following autumn births were about 365 days but were shorter following winter and spring births. Mean gestation was 252.6 days. Pregnancy rates in mature hinds were 97% per annum. Weaning rates were 84% per annum. Mean birth weights were 5.1 kg in males and 4.7 kg in females. Highest growth rates occurred prior to 6 months of age. Most stags were slaughtered for venison at about 14 months and produced a 47 to 50 kg carcass. Live weight changes in males greater than 14 months were associated with the antler cycle and stags older than 2 years lost about 15% of their live weight during the breeding rut.

The role of milk and milk proteins in relation to human health remains controversial. However, there is a large body of evidence, reviewed in this paper, specifically linking A1 beta-casein to a range of illnesses. A1 beta‐casein is produced only by cattle of European origin. The proportion of European cattle producing A1 beta-casein varies by breed and country. High levels are found in many parts of northern Europe and also other parts of the world where particular breeds of European origin predominate. Cattle that produce A2 and not A1 beta-casein are known as A2 cattle. A2 beta-casein was the original variant. A1 beta-casein releases bovine beta-casomorphin7 (BCM7) on digestion. The amount and stability of BCM7 that is cleaved from A1 beta-casein is linked to the presence and levels of various enzymes, in particular dipeptidyl peptidase 4. BCM7 is a strong opioid. There are seven strands to the evidence linking A1 beta-casein and BCM7 to Type 1 diabetes.
