This paper describes design and logistical aspects of a decision analysis study to assess the performance of qualified firearms examiners working in accredited laboratories in the United States in terms of accuracy (error rate), repeatability, and reproducibility of decisions involving comparisons of fired bullets and cartridge cases. The purpose of the study was to validate current practice of the forensic discipline of firearms/toolmarks (F/T) examination. It elicited error rate data by counting the number of false positive and false negative conclusions. Preceded by the experimental design, decisions, and logistics described herein, testing was ultimately administered 173 qualified, practicing F/T examiners in public and private crime laboratories. The first round of testing evaluated accuracy, while two subsequent rounds evaluated repeatability and reproducibility of examiner conclusions. This project expands on previous studies by involving many F/T examiners in challenging comparisons and by executing the study in the recommended double-blind format.

This study addresses the permanence and persistence of friction ridges and the persistence of impressions made from these friction ridges over months and years. Permanence is the unchanging presence and appearance of friction ridge arrangements and their attributes between recurring observations of the skin. Permanence was evaluated from direct photographs of fingers collected over a period of 30–45 days (covering one or more skin regeneration cycles) as well as after 8 or more years had elapsed. Persistence embodies the operational concept of whether or not a pair of images displays sufficient similarity upon which to base an informed decision that they were made by the same finger, while acknowledging certain dissimilarities or distortions due to friction ridge physiology, image capture, matrix, substrate, and applied pressure. Persistence applies to both friction ridge skin and impressions made from these friction ridges. Permanence and persistence of skin were assessed from direct photographs of fingers taken two months apart and from finger photographs separated by an interval of at least 8 years. Permanence and persistence were also assessed from impressions taken over 4 months, as well as those separated by 8–53 years. Variability due to capture method was assessed by using four image capture methods over a four month period: direct photography of fingers, impressions captured by ink, holographic imaging, and live scan. Qualified latent fingerprint examiners assessed all changes observed over time, as well as any limitations imposed by capture method. The practice of comparison and identification of fingerprint impressions was upheld, as was the prevailing use of the word persistence to describe stability of friction ridges. All photographs and impressions of the same finger were identifiable as originating from the same source. Within all the periods of observation, level 1 detail was permanent and persistent. Persistence, but not permanence, was supported for level 2 detail. Notably, the small changes observed were only in appearance; there were no changes in the presence of new, or absence of existing, minutiae. Level 3 details of ridge edge shape and pore presence were neither permanent nor persistent. Ridge width was permanent and persistent. Incipient ridges were neither permanent nor persistent.

A study was undertaken using nonhuman mammal specimens to better understand environmental influences on postmortem hair root band (PMRB) formation and to see whether PMRBs would occur in nonhuman mammal hairs in a similar fashion to human hairs. Carcasses from surrounding roadways were the primary source of specimens for this study, augmented by donated deceased domestic pets. Sections of pelt from each specimen were placed in controlled environmental conditions while the remainder of the carcass was left in a secure outdoor setting. Hair samples were collected daily from outdoor and control specimens and examined for evidence of PMRBs. Several environmental factors were also recorded on a daily basis. Results demonstrate PMRBs can occur in nonhuman mammal hairs, and they have microscopic characteristics similar to human PMRBs. Factors found to correlate with PMRB formation include postmortem interval, temperature, pH, and the formation and subsequent volatilization of ammonia from the surrounding tissue.

This study employed an automated facial recognition system as a means of objectively evaluating biometric correspondence between a ReFace facial approximation and the computed tomography (CT) derived ground truth skin surface of the same individual. High rates of biometric correspondence were observed, irrespective of rank class (Rk) or demographic cohort examined. Overall, 48% of the test subjects' ReFace approximation probes (n=96) were matched to his or her corresponding ground truth skin surface image at R1, a rank indicating a high degree of biometric correspondence and a potential positive identification. Identification rates improved with each successively broader rank class (R10=85%, R25=96%, and R50=99%), with 100% identification by R57. A sharp increase (39% mean increase) in identification rates was observed between R1 and R10 across most rank classes and demographic cohorts. In contrast, significantly lower (p 0.05) performance differences were observed across demographic cohorts or CT scan protocols. Performance measures observed in this research suggest that ReFace approximations are biometrically similar to the actual faces of the approximated individuals and, therefore, may have potential operational utility in contexts in which computerized approximations are utilized as probes in automated facial recognition systems.

An extensive literature review and new post-irradiation experimental results are presented of genotyping blood stains and hair, and physical examinations of latent fingerprints, hairs, and fibers. Results indicate that successful development of nuclear short tandem repeat (STR) and mitochondrial DNA sequence profiles from human blood and hair evidence is possible-up to a point-following exposure to gamma, neutron, beta, and alpha radiation at several levels that would most likely be present at this type of crime scene (i.e., a "dirty bomb," etc.). Commencing at gamma radiation levels between 90 and 900kGy, DNA analysis using conventional DNA techniques was unsuccessful. In general, irradiation negatively affected the quality of latent fingerprints. All four radiation types degraded most fingerprint samples at all doses; nevertheless, many fingerprints remained of value for potential use in comparison. Although variable from one hair to another, microscopic changes observed for all types and levels of irradiation could potentially result in false exclusions. Negligible microscopic changes were observed in papers and fibers (used as substrates for fingerprints and bloodstains) up to 90kGy gamma, but fluorescence of fibers began to change above that dose. Paper and fibers, as well as plastic evidence enclosures, became extremely brittle leading to breakage after a gamma dose of 900kGy.

In addition to having blast mitigation properties, aqueous foam concentrate AFC-380 blast suppression foam is designed to capture aerosolized chemical, biological, and radioactive particles during render-safe procedures of explosive devices. Exposure to aqueous environments and surfactants may negatively affect forensic evidence found at the scene, but the effects of AFC-380 foam and aqueous gel on the preservation and subsequent analysis of forensic evidence have not previously been investigated. Sebaceous finger and palm prints and DNA samples on paper, cardboard, tape, and various metal and plastic items, along with hairs, carpet and yarn fibers, and inks and documents, were exposed to AFC-380 foam. Similar mock evidence was also exposed to a superabsorbent gel of the type found in aqueous gel blocks used for shrapnel containment. Exposure to foam or aqueous gel was associated with a dilution effect for recovered DNA samples, but quality of the samples was not substantially affected. In contrast, exposure to AFC-380 foam or gel was detrimental to development of latent finger and palm prints on any substrate. Neither the hair nor the fiber samples were affected by exposure to either the foam or gel. Indented writing on the document samples was detrimentally affected by foam or gel exposure, but not inks and toners. The results from this study indicate that most types of forensic evidence recovered after being exposed to aqueous gel or blast suppression foam can be reliably analyzed, but latent finger and palm prints may be adversely affected.

Hexamethylene triperoxide diamine (HMTD) is a peroxide-based explosive that is easily manufactured from household chemicals. This high-energy molecule is challenging to detect because it lacks the characteristic –NO2 functional group that is the primary target of modern detection devices. One tool that is well-suited for detection and characterization of HMTD and other peroxide explosives is Raman spectroscopy. Off-resonance Raman spectra with 514.5 nm excitation of HMTD and its 13C and 15N isotopologues are presented here. These experimental results are compared to calculations to reveal the vibrational structure and normal mode assignments of this strained molecule. Many characteristics of facial aging are common to all. The age of their onset and which characteristics tend to predominate varies among individuals depending on many factors including their genetic makeup, life experiences, environment in which they live, and the regional, ethnic, or socially perceived group to which they belong. Forensic artists are often asked to provide sketches, 2D or 3D digital renderings, or sculptures representative of how an individual may appear at an older age based on a provided photograph, victim or witness description, and/or cranial remains. The challenge escalates when the subject is a member of a regional, ethnic, or other socially perceived group to which the artist has had little or no exposure. We describe aspects of adult facial aging that are of particular relevance to the forensic artist, applicable software tools, and pertinent facial databases, especially those emphasizing non-white populations. We demonstrate that facial averaging offers two key advantages to the artistic portrayal of facial aging: first, the technique requires relatively small reference databases from groups that may present logistical challenges to collect and second, that a facial average provides a useful representation of the gestalt of the age and ethnicity cohort to which a subject belongs. The artist may use an average along with other available information such as photo reference books, eyewitness descriptions, photos of immediate family members, and cranial structure to guide production of a facial composite drawing, digital age progression, or sculpture of the subject in question.

Objective A geometric morphometric approach for testing the metric accuracy of a computerized facial approximation program is presented. Materials and methods This study uses a leave-one-out procedure to generate average approximations (not adjusted for weight/age) for each individual in the ReFace reference database (n = 388), consisting of eight sex/ancestry groups (both sexes for each: African, Asian, European, Hispanic). Quantitative comparisons of corresponding three-dimensional (3D) known faces and approximations were analyzed using 66 inter-landmark distances (ILDs) derived from twelve interior facial landmarks. Results and conclusions Average signed errors per ILD ranged from -0.67 mm to 1.92 mm, with most errors indicating smaller ILDs in approximations. For all individual ILD errors combined, 99.5% were within ± 10.0 mm, 90.8% were within ± 5.0 mm, 64.6% were within ± 2.5 mm, and 30.2% were within ± 1.0 mm. The largest errors were associated with cheilion and indicated underestimated mouth widths by an overall average of 1.92 mm (SD = 3.99 mm). A statistical matching model that modeled performance of automated facial recognition correctly identified 73–88% of known-approximation pairs as matches. The accuracy and matching results suggest that facial features are highly influenced by the underlying craniofacial skeleton regardless of weight/age variations, the matching model is predictive of automated facial recognition performance, and that ReFace approximations may be sufficiently accurate to biometrically match unidentified decedents to missing persons.

With continuing advancements in biomedical imaging technologies, anthropologists are increasingly making use of data derived from indirect measurement and analysis of skeletal material. To that end, the purpose of this study was to test the reliability of 26 standard craniometric measurements routinely utilized in forensic casework across several different imaging technologies. Measurements from five crania of known individuals were collected in duplicate by two anthropologists via computed tomography (CT) scans and three-dimensional (3D) laser scans of the known skulls. The laser scans were also used to create prototype models of the known skulls. These prototypes were, themselves, laser-scanned, and measurements were also collected from the prototypes and the laser scans of the prototypes. Measurement sets from each technology were then compared with one another using the previously collected osteometric measurements taken on the crania themselves as the ground truth. Result indicate that, while the majority of measurements showed no significant differences across data formats, a handful were found to be problematic for particular technologies. For instance, measurements taken in a supero-inferior direction (e.g., BBH, OBH) from CT scans were prone to greater deviation from direct measurements of the cranium than other technologies, especially for CT scans taken at 5 mm thickness and increment. Also, several measurements defined by Type 1 landmarks, particularly those occurring at complicated or indistinct suture junctures (e.g., ASB, ZMB), were found to have high variance across all technologies while measurements based on Type 3 landmarks proved to be highly reproducible. This is contrary to measurements taken directly on crania, in which measures defined by Type 1 landmarks are typically the most reliable, likely attributable to diminished or totally obscured suture definition in the scan data. If medical imaging data are to be increasingly utilized in anthropological studies, it may be prudent to bear in mind that the reliability of measurements taken on an actual skull may not be the same as for measurements taken from medical scans.

Facial approximations (whether clay sculptures, sketches, or computer-generated) can be presented to the public in a variety of layouts, but there are currently no clear indicators as to what style of presentation is most effective at eliciting recognition. The primary purpose of this study is to determine which of five presentation methods produces the most favorable recognition results. A secondary goal of the research is to evaluate a new method for assessing the accuracy of facial approximations. Previous studies have evaluated facial approximation effectiveness using standards similar to studies of eyewitness identification in which a single, definitive choice must be made by the research participant. These criteria seem inappropriate given that facial approximation is strictly an investigative tool to help narrow the search for potential matching candidates in the process of identification. Results from the study showed a higher performance for methods utilizing more than one image of the approximation, but which specific method performed best varied among approximation subjects. Also, results for all five presentation methods showed that, when given the opportunity to select more than one approximation, participants were consistently better at identifying the correct approximation as one of a few possible matches to the missing person than they were at singling out the correct approximation. This suggests that facial approximations have perhaps been undervalued as investigative tools in previous research.

Craniometric data from computed tomography (CT) head scans of 287 living Americans of three descent groups (African, Asian, European) and both sexes were analyzed for measurement precision. Classification accuracy was estimated by a leave-one-out cross validation, and group means were compared to the Forensic Data Bank (FDB). Landmarks were placed on 3D surface models of the skulls to approximate traditional cranial measurements utilized in sex and ancestry estimations. From repeat measurements by one observer on a subset of skulls (n = 14) reflecting 14 different CT protocols, the least precise landmark was euryon (SD ≤ 4.09 mm) and the least precise distances according to the coefficient of reliability (

ReFace (Reality Enhancement Facial Approximation by Computational Estimation) is a computer-automated facial approximation application jointly developed by the Federal Bureau of Investigation and GE Global Research. The application derives a statistically based approximation of a face from a unidentified skull using a dataset of ∼400 human head computer tomography (CT) scans of living adult American individuals from four ancestry groups: African, Asian, European and Hispanic (self-identified). To date only one unpublished subjective recognition study has been conducted using ReFace approximations. It indicated that approximations produced by ReFace were recognized above chance rates (10%). This preliminary study assesses: (i) the recognizability of five ReFace approximations; (ii) the recognizability of CT-derived skin surface replicas of the same individuals whose skulls were used to create the ReFace approximations; and (iii) the relationship between recognition performance and resemblance ratings of target individuals. All five skin surface replicas were recognized at rates statistically significant above chance (22–50%). Four of five ReFace approximations were recognized above chance (5–18%), although with statistical significance only at the higher rate. Such results suggest reconsideration of the usefulness of the type of output format utilized in this study, particularly in regard to facial approximations employed as a means of identifying unknown individuals.

Studies testing the effectiveness of facial approximations typically involve groups of participants who are unfamiliar with the approximated individual(s). This limitation requires the use of photograph arrays including a picture of the subject for comparison to the facial approximation. While this practice is often necessary due to the difficulty in obtaining a group of assessors who are familiar with the approximated subject, it may not accurately simulate the thought process of the target audience (friends and family members) in comparing a mental image of the approximated subject to the facial approximation. As part of a larger process to evaluate the effectiveness and best implementation of the ReFace facial approximation software program, the rare opportunity arose to conduct a recognition study using assessors who were personally acquainted with the subjects of the approximations. ReFace facial approximations were generated based on preexisting medical scans, and co-workers of the scan donors were tested on whether they could accurately pick out the approximation of their colleague from arrays of facial approximations. Results from the study demonstrated an overall poor recognition performance (i.e., where a single choice within a pool is not enforced) for individuals who were familiar with the approximated subjects. Out of 220 recognition tests only 10.5% resulted in the assessor selecting the correct approximation (or correctly choosing not to make a selection when the array consisted only of foils), an outcome that was not significantly different from the 9% random chance rate. When allowed to select multiple approximations the assessors felt resembled the target individual, the overall sensitivity for ReFace approximations was 16.0% and the overall specificity was 81.8%. These results differ markedly from the results of a previous study using assessors who were unfamiliar with the approximated subjects. Some possible explanations for this disparity in performance were examined, and it was ultimately concluded that ReFace facial approximations may have limited effectiveness if used in the traditional way. However, some promising alternative uses are explored that may expand the utility of facial approximations for aiding in the identification of unknown human remains.

This research examined how accurately 2D images (i.e., photographs) of 3D clay facial approximations were matched to corresponding photographs of the approximated individuals using an objective automated facial recognition system. Irrespective of search filter (i.e., blind, sex, or ancestry) or rank class (R1, R10, R25, and R50) employed, few operationally informative results were observed. In only a single instance of 48 potential match opportunities was a clay approximation matched to a corresponding life photograph within the top 50 images (R50) of a candidate list, even with relatively small gallery sizes created from the application of search filters (e.g., sex or ancestry search restrictions). Increasing the candidate lists to include the top 100 images (R100) resulted in only two additional instances of correct match. Although other untested variables (e.g., approximation method, 2D photographic process, and practitioner skill level) may have impacted the observed results, this study suggests that 2D images of manually generated clay approximations are not readily matched to life photos by automated facial recognition systems. Further investigation is necessary in order to identify the underlying cause(s), if any, of the poor recognition results observed in this study (e.g., potential inferior facial feature detection and extraction). Additional inquiry exploring prospective remedial measures (e.g., stronger feature differentiation) is also warranted, particularly given the prominent use of clay approximations in unidentified persons casework.

Facial approximations based on facial soft tissue depth measurement tables often utilize the arithmetic mean as a central tendency estimator. Stephan et al. (J Forensic Sci 2013;58:1439) suggest that the shorth and 75-shormax statistics are better suited to describe the central tendency of non-normal soft tissue depth data, while also accommodating normal distributions. The shorth, 75-shormax, arithmetic mean, and other central tendency estimators were evaluated using a CT-derived facial soft tissue depth dataset. Differences between arithmetic mean and shorth mean for the tissue depths examined ranged from 0 mm to +2.3 mm (average 0.6 mm). Differences between the arithmetic mean plus one standard deviation (to approximate the same data points covered by the 75-shormax) and 75-shormax values ranged from -0.8 mm to +0.7 mm (average 0.2 mm). The results of this research suggest that few practical differences exist across the central tendency point estimators for the evaluated soft tissue depth dataset.

The Federal Bureau of Investigation (FBI) Laboratory is currently exploring needs and protocols for the storage of evidentiary items contaminated with radioactive material. While a large body of knowledge on the behavior of storage polymers in radiation fields exists, this knowledge has not been applied to the field of forensics and maintaining evidentiary integrity. The focus of this research was to evaluate the behavior of several traditional evidentiary containment polymers when exposed to significant alpha, beta, gamma, neutron and mixed radiation sources. Doses were designed to simulate exposures possible during storage of materials. Several products were found to be poorly suited for use in this specific application based on standardized mechanical testing results. Remaining products were determined to warrant further investigation for the storage of radiologically contaminated evidence.

Currently in the United States, the remains of thousands of unidentified human decedents are housed in medical, law enforcement, and forensic facilities throughout the country. A number of digital data repositories have been established to curate and disseminate the details of these unidentified decedent cases; some repositories also maintain records of missing persons. Although a cross-reference for textual data similarity occurs between the missing persons and unidentified decedent records in some repositories, no repository is currently known to employ an image analysis technology for cross-referencing image data. Results suggest that the computer-generated facial approximations used in this research were consistently included in prioritized candidate lists when used in an automated facial recognition context. Two concurrent studies exploring the specific use-case discussed here were executed. The first employed an optimally-conditioned facial image gallery (g=6159) (i.e., a gallery comprised of highly consistent facial images), a research design intended to establish the ceiling performance of the combined use of the two software programs employed. The second employed a gallery (g=1816) compiled from a real-world dataset of missing persons' facial images, a research design intended to inform potential operational performance when using the highly varied facial images typically comprising public databases. Multiple types of facial approximations (reconstructions) with varying degrees of weight adjustments, age adjustments, or the presence (or absence) of visible eyes, and combinations of these variables, were evaluated. Overall, in the larger, optimally modeled study, 53% of the facial approximations for the t=159 test subjects examined were matched to his or her corresponding life photo within the top 50 images of a candidate list generated from a blind (unrestricted) search of the highly consistent gallery (g=6159). In the operationally modeled study, 31% of the test subjects' (t=16) facial approximations were matched to their corresponding life photos within the top 50 images of a candidate list generated from a blind search of the gallery populated with images from an operational dataset (g=1816). As anticipated, candidate list inclusion rates improved with the use of demographic filters. No significantly different inclusion rates were observed between the sex or age cohorts examined. Significant differences were, however, observed across population cohorts. Entities curating missing and unidentified decedent records may benefit from a paired implementation of facial recognition technology and computer-generated approximations as part of a comprehensive investigative strategy for the specific envisioned use-case discussed in this research.

The Scientific Working Group on DNA Analysis Methods (SWGDAM) Hispanic data set was analyzed to determine the diversity, phylogeny, and relevant single nucleotide polymorphisms (SNPs) that describe haplogroup patterns for Hispanic Americans (N = 686), and to assess the degree of admixture regarding mitochondrial DNA (mtDNA). The largest component of admixture based on mtDNA analysis derives from the four major haplogroups previously observed in Native American ancestry, including A (29.3%), B (15.7%), C (20.6%), and D (4.8%). European (17.8%) and African (11.8%) haplogroups also were observed within this data set. Hispanic SWGDAM samples from the southwest, compared with other SWGDAM Hispanic samples, were observed to have a greater percent of Native American haplogroups present (79.9%), and fewer African American haplogroups (4.5%). A total of 234 SNPs were observed in the data set, including 36 newly reported variable positions. These SWGDAM Hispanic data set SNPs ranged from having 1 to 31 changes (Length = L) on the phylogenetic tree, with site 16519 being the most variable. On average, there were 3.9 character changes for each variable position on the tree. The most variable sites (with 13 or more changes each listed from fastest to slowest) observed were 16519 (L = 31), 16189 (L = 23), 152 (L = 23), 16311 (L = 19), 146 (L = 17) 195 (L = 17), 16093 (L = 15), 16362 (L = 14), 16129 (L = 13), 150 (L = 13), and 153 (L = 13). These sites are consistent with other reports on highly variable positions. A total of 27 SNPs were chosen to identify all clusters containing 1% (N = 7) or more individuals in the SWGDAM Hispanic data set. The descriptive analyses revealed that the SWGDAM Hispanic data set is similar to published Native American and Hispanic data sets.

Human mitochondrial DNA (mtDNA) analysis is a valuable forensic tool, useful in cases where the amount of extracted DNA is low or highly degraded. Population databases are used to determine the relative rarity of a particular profile obtained in a forensic case. Rather than full DNA sequence information, sequence profiles are compared to a reference sequence, and the differences from the reference are recorded in forensic databases. A standard method is proposed for characterizing length variants, and examples are described using actual human control region mtDNA profiles. Consistency in alignment and nomenclature avoids inadvertently describing two sequences as different when in fact they are the same.

Most mtDNA studies on Native Americans have concentrated on hypervariable region I (HVI) sequence data. Mitochondrial DNA haplotype data from hypervariable regions I and II (HVI and HVII) have been compiled from Apaches (N=180) and Navajos (N=146). The inclusion of HVII data increases the amount of information that can be obtained from low diversity population groups. Less mtDNA variation was observed in the Apaches and Navajos than in major population groups. The majority of the mtDNA sequences were observed more than once; only 17.8% (32/180) of the Apache sequences and 25.8% of the Navajo sequences were observed once. Most of the haplotypes in Apaches and Navajos fall into the A and B haplogroups. Although a limited number of haplogroups were observed, both sample populations exhibit sufficient variation for forensic mtDNA typing. Genetic diversity was 0.930 in the Apache sample and 0.963 in the Navajo sample. The random match probability was 7.48% in the Apache sample and 4.40% in the Navajo sample. The average number of nucleotide differences between individuals in a database is 9.0 in the Navajo sample and 7.7 in the Apache sample. The data demonstrate that mtDNA sequencing can be informative in forensic cases where Native American population data are used.

Stature estimation is an important component of the biological profile. Human crania are sometimes recovered in the absence of other skeletal material in forensic casework, and stature estimation equations using cranial measurements have not been developed for populations in the United States. Both simple and multiple regression equations for estimating stature were developed from standard cranial measurements taken from both a cranial CT dataset and the Forensic Data Bank, and the resulting equations were tested using a separate dataset. A weak to moderate correlation with stature was found for some of the measurements tested. Tests of the sex- and ancestry-specific equations and pooled sex equations produced accurate estimated stature ranges for most of the individuals in the test dataset, but large 95% confidence intervals (±14-16cm) for these equations can produce only an imprecise estimated stature range for an unidentified individual. Pooled ancestry equations also produced accurate estimated stature ranges for many of the individuals in the test dataset, but with even larger 95% confidence intervals (±18-20cm). The results of this study indicate that stature can be estimated using cranial measurements, but the resulting 95% confidence intervals produce stature ranges that are too broad to use in most forensic casework.

Data from 1393 unrelated individuals have been compiled from eight population groups: African Americans, Africans (Sierra Leone), U.S. Caucasians, Austrians, French, Hispanics, Japanese, and Asian Americans. The majority of the mtDNA sequences were observed only once within each population group (i.e., ranging from a low of 60.3% (35/58) of the Asian American sequences to a high of 85.3% (93/109) of the French sequences). Genetic diversity ranged from 0.990 in the African sample to 0.998 in African Americans. Random match probability ranged from 2.50% in the Asian American sample to 0.52% in U.S. Caucasians. The average number of nucleotide differences between individuals in a database is greatest for the African American and African samples (14.1 and 13.1, respectively), and the least variable are the Caucasians (ranging from 7.2 to 8.4). Substitutions are the predominate polymorphism, and at least 92% of the substitutions are transitions. The most prevalent transversions are As substituted for Cs and Cs substituted for As. For most population groups these transversions occurred predominately in the HVI region; however, the African, African American, and Hispanic samples also demonstrated a large portion of their C to A and A to C transversions in the HVII region (at sites 186 and/or 189). Most insertions occur in the HVII region at sites 309.1 and 315.1, within a stretch of C's. Insertions of an additional C are common in all population groups. The sequence data were converted to SSO mtDNA types and compared with population data on Caucasians, Africans, Asians, Japanese, and Mexicans described by Stoneking et al. [M. Stoneking, D. Hedgecock, R.G. Higuchi, L. Vigilant, H.A. Erlich, Population variation of human mtDNA control region sequences detected by enzymatic amplification and sequence-specific oligonucleotide probes, Am. J. Hum. Genet. 48 (1991) 370-382] using an R x C contingency table test. Differences between major population groups (i.e., between African, Caucasian, and Asian) are quite evident, and similar ethnic population groups carried similar SSO polymorphism frequencies. There were only a few SSO types that showed significant differences between subpopulation groups. The SSO data alone can not be used to describe the population genetics with complete sequence data. However, the results of the SSO comparisons are similar to other analyses, and differences in sequence data in regions HVI and HVII are greater between major population groups than between subgroups.

The scientific working group on DNA analysis Methods (SWGDAM) mitochondrial DNA (mtDNA) population data set is used to infer the relative rarity of control region mtDNA profiles obtained from evidence samples and of profiles used for identification of missing persons. In this study, the African American haplogroup patterns in the SWGDAM data were analyzed in a phylogenetic context to determine relevant single nucleotide polymorphisms (SNPs) and to describe haplogroup distributions for Africans observed in these data sets. Over 200 SNPs (n=217) were observed in the African American data set (n=1148). These SNPs ranged from having 1-39 changes in the phylogenetic tree, with sites 152 and 16519 being the most variable. On average there were 5.8 changes for a character on the tree. The most variable sites (with 19 or more changes each) observed included 16093, 16129, 16189, 16311, 16362, 16519, 146, 150, 152, 189, and 195. These rapidly changing sites are consistent with other published analyses. Only 34 SNPs are needed to identify all clusters containing 10 or more individuals in the African American data set. The results show that the African American SWGDAM mtDNA data set contains variation consistent with that described in continental African populations. Thirteen of the 18 haplogroups previously observed in African populations were observed and include: L1a, L1b, L1c, L2a, L2b, L2c, L3b, L3d, L3e1, L3e2, L3e3, L3e4 and L3f. Haplogroup L2a is the most commonly observed cluster (18.8%) in the African American data set. The next most common haplogroups in the African American data set include the clusters L1c (11.0%), L1b (9.1%), L3e2 (9.0%) and L3b (8.1%). Approximately 8% of the haplogroups observed within African Americans were common in European Caucasians or East Asians; these were H (n=32), J (n=4), K (n=5), T (n=2), U5 (n=6), U6 (n=9 also known from North Africa), A (n=12), B (n=7), C (n=4), and M (n=16), respectively. The European Caucasian and East Asian haplogroups are expected due to admixture between individuals with recent ancestry in Western Eurasia and sub-Saharan Africa. The genetic characterization of these relevant data sets is fully consistent with other published mtDNA genetic variation. The sequence diversity observed in this data set makes it a valuable tool for forensic applications.

Facial approximation is the technique of developing a representation of the face from the skull of an unknown individual. Facial approximation relies heavily on average craniofacial soft tissue depths. For more than a century, researchers have employed a broad array of tissue depth collection methodologies, a practice which has resulted in a lack of standardization in craniofacial soft tissue depth research. To combat such methodological inconsistencies, Stephan and Simpson 2008 [15] examined and synthesized a large number of previously published soft tissue depth studies. Their comprehensive meta-analysis produced a pooled dataset of averaged tissue depths and a simplified methodology, which the researchers suggest be utilized as a minimum standard protocol for future craniofacial soft tissue depth research. The authors of the present paper collected craniofacial soft tissue depths using three-dimensional models generated from computed tomography scans of living males and females of four self-identified ancestry groups from the United States ranging in age from 18 to 62 years. This paper assesses the differences between: (i) the pooled mean tissue depth values from the sample utilized in this paper and those published by Stephan 2012 [21] and (ii) the mean tissue depth values of two demographically similar subsets of the sample utilized in this paper and those published by Rhine and Moore 1984 [16]. Statistical test results indicate that the tissue depths collected from the sample evaluated in this paper are significantly and consistently larger than those published by Stephan 2012 [21]. Although a lack of published variance data by Rhine and Moore 1984 [16] precluded a direct statistical assessment, a substantive difference was also concluded. Further, the dataset presented in this study is representative of modern American adults and is, therefore, appropriate for use in constructing contemporary facial approximations.

Representation of crime scenes as virtual reality 3D computer displays promises to become a useful and important tool for law enforcement evaluation and analysis, forensic identification and pathological study and archival presentation during court proceedings. Use of these methods for assessment of evidentiary materials demands complete accuracy of reproduction of the original scene, both in data collection and in its eventual virtual reality representation. The recording of spatially accurate information as soon as possible after first arrival of law enforcement personnel is advantageous for unstable or hazardous crime scenes and reduces the possibility that either inadvertent measurement error or deliberate falsification may occur or be alleged concerning processing of a scene. Detailed measurements and multimedia archiving of critical surface topographical details in a calibrated, uniform, consistent and standardized quantitative 3D coordinate method are needed. These methods would afford professional personnel in initial contact with a crime scene the means for remote, non-contacting, immediate, thorough and unequivocal documentation of the contents of the scene. Measurements of the relative and absolute global positions of object sand victims, and their dispositions within the scene before their relocation and detailed examination, could be made. Resolution must be sufficient to map both small and large objects. Equipment must be able to map regions at varied resolution as collected from different perspectives. Progress is presented in devising methods for collecting and archiving 3D spatial numerical data from crime scenes, sufficient for law enforcement needs, by remote laser structured light and video imagery. Two types of simulation studies were done. One study evaluated the potential of 3D topographic mapping and 3D telepresence using a robotic platform for explosive ordnance disassembly. The second study involved using the laser mapping system on a fixed optical bench with simulated crime scene models of the people and furniture to assess feasibility, requirements and utility of such a system for crime scene documentation and analysis.

In order that there can be confidence that DNA profile frequency estimates will not place undue bias against a defendant, 2 methods are described for estimating minimum allele frequency bounds for PCR-based loci. One approach estimates minimum allele frequencies for VNTR and STR loci using sample size and the observed heterozygosity at a locus, while the second approach, appropriate for loci typed with allele-specific oligonucleotide probes, is based only on sample size. The use of a minimum allele frequency enables compensation for sparse sampling of infrequent alleles in population databases.

Allele distributions were determined for several VNTR loci in the general French Caucasian population, a French regional population from Brittany and in three French Canadian populations, residing either in Montreal or in one of two other regions in Quebec. Allele distributions are highly polymorphic in all populations sampled. Despite a well-documented genetic founder effect in one of the populations, no disequilibrium was detected over all genotypes, within and between loci, for the data bases in this study. Moreover, there were no forensically significant differences observed between estimated frequencies of 1,964 Caucasian and non-Caucasian target DNA profiles estimated in any of the groups.

A National Research Council (NRC) report asserted that genetic diversity between subgroups within a race is greater than between races. While this is counterintuitive to the basic tenets of human population genetics, the forensic application of statistical estimates of DNA profile frequencies has been questioned. The general approach used for estimating DNA profile frequencies by the majority of North American forensic laboratories is the use of the product rule on fixed bin allele frequencies derived from general population group databases. An informal analysis of the differences in frequency estimates within Hinf I-generated RFLP data determined in three ethnically distinct population groups —Norwegians, Spanish, and Turks — and in Caucasians and Blacks from the United States is presented. The VNTR loci analyzed are D2S44, D7S21, and D12S11. The data demonstrate that the assertion by the NRC is unfounded and that major population group databases, in lieu of subpopulation databases, can be used to provide estimates of DNA profile frequencies without consequences of wrongful bias.

In 1990 the FBI began development of a national DNA identification index, CODIS. The concept driving this development is the rarity of a DNA profile obtained from the successful analysis of body fluid stains left at crime scenes. Given the recidivistic nature of rape and other violent crimes, and the fact that body fluids of the perpetrator are left at crime scenes, a national computer-based system of storing and comparing DNA records can result in the successful application of DNA profiling in the fight to prosecute and deter violent criminal behavior. There are now several examples of forensic DNA testing generating previously unavailable investigative leads. Also, this powerful forensic technique has been used to successfully prosecute/adjudicate cases involving the same subject after these cases have been linked together by DNA evidence.

The likelihood of occurence of 1,964-HaeIII-generated target DNA profiles was estimated using fixed bin VNTR frequencies from various Caucasian, Black, and Hispanic databases and the product rule. The data in this study demonstrate that for forensic purposes there are smaller differences in statistical estimates of DNA profile frequencies among subgroup databases than among estimates across major population databases. This observation does not support the premise asserted by the NCR Report (1992) that the differences among subgroups within a race would be greater than between races (at least for forensic purposes). Therefore, the data do not support the need for alternative procedures, such as the ceiling principle approach (NRC Reports, 1992), for deriving statistical estimates of DNA profile frequencies. Comparisons across major population groups provide reasonable, reliable, and meaningful estimates of DNA profile frequencies without forensically significant consequences.

Implementation of standard methods for the conduct of restriction fragment length polymorphism analysis into the protocols of United States crime laboratories offers an unprecedented opportunity for the establishment of a national computer database system to enable interchange of DNA typing information. The FBI Laboratory, in concert with crime laboratory representatives, has taken the initiative in planning and implementing such a database system. The Combined DNA Index System (CODIS) will be composed of three sub-indices: a statistical database, which will contain frequencies of DNA fragment alleles in various population groups; an investigative database which will enable linkage of violent crimes through a common subject; and a convicted felon database that will serve to maintain DNA typing profiles for comparison to profiles developed from violent crimes where the suspect may be unknown.

The detection of alleles of variable number of tandem repeats (VNTR) loci by restriction fragment length polymorphism analysis has become an important aspect of genetic characterization for identity testing. Some VNTR loci are so polymorphic that the analysis of three to five genetic markers could potentially provide unique identity. However, the more informative a genetic marker is ( i.e. , high degree of polymorphisms), the better it is as an exculpatory tool. This approach currently provides the best avenue for excluding a falsely associated individual with a particular sample. When an analysis fails to exclude an individual as the source of the questioned material, a value (frequency of occurrence) should be placed on the VNTR profiles to assess weight to the inclusion in identity testing. Arbitrarily defined fixed bins were designed to accommodate quasicontinuous data and to provide a result that would not place an underestimation of the frequency of occurrence of a set of alleles attributed to an individual.

Currently, the Scientific Working Group on DNA Analysis Methods (SWGDAM) mtDNA dataset is used to infer the relative rarity of mtDNA profiles (i.e., haplotypes) obtained from evidence samples and for identification of missing persons. The Caucasian haplogroup patterns in this forensic dataset have been characterized using phylogenetic methods. The assessment reveals that the dataset is relevant and representative of U.S. and European Caucasians. The comparisons carried out were both the observation of variable sites within the control region (CR) and the selection of a subset of these sites, which partition the variation within human mtDNA control region sequences into clusters (i.e., haplogroups). The aligned sequence matrix was analyzed to determine both single nucleotide polymorphisms (SNPs) in a phylogenetic context, as well as to check and standardize haplogroup designations with a focus on determining the characters that define these groups. To evaluate the dataset for forensic utility, the haplogroup identifications and frequencies were compared with those reported from other published studies.

A variety of general, regional, ancestral and ethnic databases is available for the polymerase chain reaction (PCR)-based loci LDLR, GYPA, HBGG, D7S8, Gc, DQA1, and D1S80. Generally, we observed greater differences in frequency estimations of DNA profiles between racial groups than between ethnic or geographic subgroups. Analysis revealed few forensically significant differences within ethnic subgroups, particularly within general United States groups, and multi-locus frequency estimates typically differ by less than a factor of ten. Using a database different from the one to which a target profile belongs tends to overestimate rarity. Implementation of the general correction of homozygote frequencies for a population substructure, advised by the 1996 National Research Council report, The Evaluation of Forensic DNA Evidence, has a minimal effect on profile frequencies. Even when it is known that both the suspect and all possible perpetrators must belong to the same isolated population, the special correction for inbreeding, which was proposed by the 1996 National Research Council report for this special case, has a relatively modest effect, typically a factor of two or less for 1% inbreeding. The effect becomes more substantial (exceeding a factor of ten) for inbreeding of 3% or more in multi-locus profiles rarer than about one in a million.

Concerns have been raised about hypothetical problems arising from the use of statistics for determining the likelihood of occurrence of DNA profiles for forensic purposes. A major contention is that reference databases based on subgroups of a major population category rather than on general (or major) population groups, might yield large differences in the estimated likelihood of occurrence of DNA profiles. This hypothetical issue is based on the assertion by some people that the differences among subgroups within a race would be greater than between races (at least for forensic purposes). To evaluate the effects of the above concern the likelihood of occurrence of 615 Hinf I-generated target DNA profiles was estimated using fixed bin frequencies from various ethnic databases and the multiplication rule. Based on the data in this study, differences in allele frequencies at a particular locus do not have substantial effects on VNTR profile frequency estimates when subgroup reference databases from within a major population group are compared. In contrast, the greatest variation in statistical estimates occurs across-major population groups. Therefore, the assertion, by some critics that the differences among subgroups within a race would be greater than between races (at least for forensic purposes), is unfounded. The data in the study support that comparisons across major population groups provide valid estimates of DNA profile frequencies without forensically significant consequences. The data do not support the need for alternate procedures, such as the ceiling principle approach, for deriving statistical estimates of DNA profile frequencies.

The likelihood of occurrence of 1964 Hae III-generated target DNA profiles was estimated using fixed bin frequencies from various regional and ethnic databases and the multiplication rule. The databases generally were from the following major categories: Black, Caucasian, Hispanic, Oriental, and American Indian. It was found that subdivision, either by ethnic group or by U.S. geographic region, within a major population group did not substantially affect forensic estimates of the likelihood of occurrence of a DNA profile. As expected, the greatest variation in estimates for within-group estimates was among American Indian databases. Because the greatest variation in statistical estimates occurs across-major population groups, in most cases, there will be no unfair bias applying general population database estimates. Therefore, based on empirical data, there is no demonstrable need for using alternate approaches, such as the ceiling approach, to derive statistical estimates. The current practice of using general population databases and the multiplication rule provides valid estimates of the likelihood of occurrence of a DNA profile.

When the results of a forensic comparison of highly polymorphic variable number tandem repeat (VNTR) loci fail to exclude a suspect as a possible contributor of biological evidence, it is desirable to convey to the trier of fact the significance of the match. Furthermore, in a forensic context, it is desirable that the estimated frequency of occurrence be conservative, that is, that any uncertainty in the estimate will favor the accused. Using an empirical approach with a data base of 2046 individuals belonging to one of four population groups, this study examined the effect of the method used to estimate frequency of occurrence of a VNTR profile from a reference data base, and the consequences of using a data base that may not represent the circumstances of the crime. The fixed bin method was at least as conservative as the floating bin and genotype counting (direct counting) methods. Secondly, for forensic purposes, profile frequency estimates from different reference populations do not deviate greatly. VNTR profiles are rare in any of the data bases.

Observation of “collapsed” coiled (steel) furniture springs has been utilized for several decades or more by arson investigators as an indicator of whether an accelerant or smoldering source (such as a cigarette) caused a fire. This paper cites the contradictory literature, synopsizes metallurgical phenomena operative when coiled steel springs are subjected to fires, and presents empirical data from U.S. Federal Bureau of Investigation (FBI) Laboratory testing. It is concluded that observation of the “collapsed” state of coiled furniture/bedding springs is not a reliable indicator of whether a fire was initiated by a smoldering cigarette or accelerated by the presence of a hydrocarbon.

Several laboratories have reported digital data acquisition and control systems for electron microscopes. The considerations involved in implementation of such a system have been reviewed. All of these systems have used a relatively small minicomputer, either as a stand-alone, or as a satellite processor linked to a larger computer, so as not to burden the large host with the mundane task of operating a microscope. In recent years however, hardware costs have decreased sufficiently that many laboratories have their own superminicomputer (e.g., one of the Digital VAX series), or share one within a small group of users.
