Abstract

The Compact Linear Collider (CLIC) is a TeV-scale high-luminosity linear $e^+e^-$ collider under development at CERN. Following the CLIC conceptual design published in 2012, this report provides an overview of the CLIC project, its current status, and future developments. It presents the CLIC physics potential and reports on design, technology, and implementation aspects of the accelerator and the detector. CLIC is foreseen to be built and operated in stages, at centre-of-mass energies of 380 GeV, 1.5 TeV and 3 TeV, respectively. CLIC uses a two-beam acceleration scheme, in which 12 GHz accelerating structures are powered via a high-current drive beam. For the first stage, an alternative with X-band klystron powering is also considered. CLIC accelerator optimisation, technical developments and system tests have resulted in an increased energy efficiency (power around 170 MW) for the 380 GeV stage, together with a reduced cost estimate at the level of 6 billion CHF. The detector concept has been refined using improved software tools. Significant progress has been made on detector technology developments for the tracking and calorimetry systems. A wide range of CLIC physics studies has been conducted, both through full detector simulations and parametric studies, together providing a broad overview of the CLIC physics potential. Each of the three energy stages adds cornerstones of the full CLIC physics programme, such as Higgs width and couplings, top-quark properties, Higgs self-coupling, direct searches, and many precision electroweak measurements. The interpretation of the combined results gives crucial and accurate insight into new physics, largely complementary to LHC and HL-LHC. The construction of the first CLIC energy stage could start by 2026. 3. Variations in climate drive behavior and survival of small desert tortoises

Abstract

In the Mojave Desert, timing and amounts of precipitation profoundly affect the availability of water and annual plant foods necessary for the threatened Agassiz’s desert tortoise (Gopherus agassizii) to survive, especially during prolonged droughts. As part of recovery actions to increase declining populations, we translocated 83 juvenile and young desert tortoises raised in head-start pens for 4–10 years to a new location 15 km away during the fall of 2013 and 2014. We tracked them for 9 years during a megadrought, during multiple years of low rainfall, and a few years when precipitation neared or exceeded long-term norms. We evaluated behaviors and how precipitation and forage availability affected survival. At the end of the study, 21.6% of tortoises were alive, and six had grown to adulthood. Annual models of survival indicated that tortoise size was the driving variable in most years, followed by the number of repeatedly used burrows during periods of temperature extremes. Other variables affecting survival in ≥1 year were vegetation, movements during the first 2 years post-translocation, and condition index, a measure of health. Tortoises moved more, expanded home ranges, and grew rapidly in years when winter rainfall approached or exceeded long-term norms and annual plants were available to eat. During dry years, movements and growth were limited. Exceptions to this pattern occurred in the last year of study, a dry year: tortoises grew and moved more, and home ranges increased. The increase in size and approaching adulthood may have stimulated greater traveling. Some left the study area, indicating a need for large release areas. We may have aided survival by offering water twice yearly when handling because some tortoises drank and increased in mass up to 40%. Prolonged droughts and hotter temperatures can limit the recovery of populations, reduce the survival of young tortoises, and increase the time to maturity.

Abstract

Euclid is a space-based survey mission from the European Space Agency designed to understand the origin of the Universe's accelerating expansion. It will use cosmological probes to investigate the nature of dark energy, dark matter and gravity by tracking their observational signatures on the geometry of the universe and on the cosmic history of structure formation. The mission is optimised for two independent primary cosmological probes: Weak gravitational Lensing (WL) and Baryonic Acoustic Oscillations (BAO). The Euclid payload consists of a 1.2 m Korsch telescope designed to provide a large field of view. It carries two instruments with a common field-of-view of ~0.54 deg2: the visual imager (VIS) and the near infrared instrument (NISP) which contains a slitless spectrometer and a three bands photometer. The Euclid wide survey will cover 15,000 deg2 of the extragalactic sky and is complemented by two 20 deg2 deep fields. For WL, Euclid measures the shapes of 30-40 resolved galaxies per arcmin2 in one broad visible R+I+Z band (550-920 nm). The photometric redshifts for these galaxies reach a precision of dz/(1+z) < 0.05. They are derived from three additional Euclid NIR bands (Y, J, H in the range 0.92-2.0 micron), complemented by ground based photometry in visible bands derived from public data or through engaged collaborations. The BAO are determined from a spectroscopic survey with a redshift accuracy dz/(1+z) =0.001. The slitless spectrometer, with spectral resolution ~250, predominantly detects Ha emission line galaxies. Euclid is a Medium Class mission of the ESA Cosmic Vision 2015-2025 programme, with a foreseen launch date in 2019. The Farnesoid-X Receptor, FXR, is a nuclear bile acid receptor. Its originally described function is in bile acid synthesis and regulation within the liver. More recently, however, FXR has been increasingly appreciated for its breadth of function and expression across multiple organ systems, including the intestine. While FXR’s role within the liver continues to be investigated, increasing literature indicates that FXR has important roles in responding to inflammation, maintaining intestinal epithelial barrier function, and regulating immunity within the gastrointestinal (GI) tract. Given the complicated and multi-factorial nature of intestinal barrier dysfunction, it is not surprising that FXR’s role appears equally complicated and not without conflicting data in different model systems. Recent work has suggested translational applications of FXR modulation in GI pathology; however, a better understanding of FXR physiology is necessary for these treatments to gain widespread use in human disease. This review aims to discuss current scientific work on the role of FXR within the GI tract, specifically in its role in intestinal inflammation, barrier function, and immune response, while also exploring areas of controversy.

Abstract

In the wild, prereproductive Agassiz's Desert Tortoises rarely co-occur in burrows, which we determined by tracking 83 head-started and translocated juvenile and immature tortoises with radio transmitters at least once per month between 2013 and 2022. We observed only 2 instances of co-occurrence at burrows out of 4622 locations; 594 of these locations were at burrows often used repeatedly during temperature extremes in summer and late fall and winter. Abstract

The Asp233-Asp246 pair is highly conserved in Class A β-lactamases, which hydrolyze β-lactam antibiotics. Here, we characterize its function using CTX-M-14 β-lactamase. The D233N mutant displayed decreased activity that is substrate-dependent, ranging from k(cat)/K(m) reduction of 20% for nitrocefin, to 6-fold for cefotaxime. In comparison, the mutation reduced the binding of a known reversible inhibitor by 10-fold. The mutant structures showed movement of the 213-219 loop and the loss of the Thr216-Thr235 hydrogen bond, which was restored by inhibitor binding. Mutagenesis of Thr216 further highlighted its contribution to CTX-M activity. These results demonstrate the importance of the aspartate pair to CTX-M hydrolysis of substrates with bulky side chains, while suggesting increased protein flexibility as a means to evolve drug resistance.

Abstract

In dit rapport worden de resultaten van het onderzoek gepresenteerd dat Aeres Hogeschool Dronten voor de Stichting Natuurlijk Melken 2050 heeft uitgevoerd. Met het onderzoek wordt een bijdrage geleverd aan het project Melklab 2.0. In het project Melklab 2.0 wordt gezocht naar de meerwaarde van natuurinclusieve melkveehouderij voor de melkkwaliteit en de management keuzes die melkveehouders kunnen maken om die melkkwaliteit te beïnvloeden. Het onderzoek dat door Aeres Hogeschool is uitgevoerd bestaat uit twee delen. Een deel gaat over de praktijkvraag naar de relatie tussen kruidenrijk grasland en gezondere melk en een deel gaat over andere aspecten van het rantsoen en de relatie met de samenstelling van melk en kaas.
