BACKGROUNDPrimary Sjögren's syndrome (pSS) is characterized by B cell hyperactivity and elevated B-lymphocyte stimulator (BLyS). Anti-BLyS treatment (e.g., belimumab) increases peripheral memory B cells; decreases naive, activated, and plasma B cell subsets; and increases stringency on B cell selection during reconstitution. Anti-CD20 therapeutics (e.g., rituximab) bind and deplete CD20-expressing B cells in circulation but are less effective in depleting tissue-resident CD20+ B cells. Combined, these 2 mechanisms may achieve synergistic effects.METHODSThis 68-week, phase II, double-blind study (GSK study 201842) randomized 86 adult patients with active pSS to 1 of 4 arms: placebo, s.c. belimumab, i.v. rituximab, or sequential belimumab + rituximab.RESULTSOverall, 60 patients completed treatment and follow-up until week 68. The incidence of adverse events (AEs) and drug-related AEs was similar across groups. Infections/infestations were the most common AEs, and no serious infections of special interest occurred. Near-complete depletion of minor salivary gland CD20+ B cells and a greater and more sustained depletion of peripheral CD19+ B cells were observed with belimumab + rituximab versus monotherapies. With belimumab + rituximab, reconstitution of peripheral B cells occurred, but it was delayed compared with rituximab. At week 68, mean (± standard error) total EULAR Sjögren's syndrome disease activity index scores decreased from 11.0 (1.17) at baseline to 5.0 (1.27) for belimumab + rituximab and 10.4 (1.36) to 8.6 (1.57) for placebo.CONCLUSIONThe safety profile of belimumab + rituximab in pSS was consistent with the monotherapies. Mixedwood forest composition, or co-dominance of hardwood and softwood species, has been interpreted as both stable and unstable. Through review of existing theory, we propose a conceptual model to understand mixedwood compositional stability in boreal and temperate forests of eastern North America. We first review the current theory that the strength of neighborhood effects (i.e., species ability to self-replace under their own canopy) is essential to understanding stability such that when self-replacement is strong for both dominant hardwood and softwood species, composition is stable except at extreme disturbance severities. In contrast, when mixedwood forests are dominated by negligible or weak affinities to self-replace, composition is unstable and sensitive to changes in disturbance. Our new concept further posits that changes in both the disturbance severity and its vertical direction are essential to understanding stability. For example, where moderate-severity surface fires (which impact forests from below) cease and are replaced by moderate-severity blowdowns (which impact forests from above), instability can occur even when disturbance severity is unchanged. We therefore pose and discuss an extension to current theory to provide a new unifying concept of stability for mixedwood forests and, more broadly, for mixed-species forests.

Temperate mixedwoods (hardwood–softwood mixtures) in central and eastern United States and Canada can be classified into two overarching categories: those with shade-tolerant softwoods maintained by light to moderate disturbances and those with shade-intolerant to mid-tolerant softwoods maintained by moderate to severe disturbances. The former includes red spruce (Picea rubens Sarg.), balsam fir (Abies balsamea (L.) Mill.), or eastern hemlock (Tsuga canadensis (L.) Carr.) in mixture with northern hardwood species; the latter includes pine (Pinus) – oak (Quercus) mixtures. Such forests have desirable socio-economic values, wildlife habitat potential, and (or) adaptive capacity, but management is challenging because one or more softwood species in each can be limited by depleted seed sources, narrow regeneration requirements, or poor competitive ability. Appropriate silvicultural systems vary among mixedwood compositions depending on shade tolerance and severity of disturbance associated with the limiting softwoods, site quality, and level of herbivory. Sustainability of mixedwood composition requires that stand structure and composition be managed at each entry to maintain vigorous trees of species with different growth rates and longevities and to encourage development of advance reproduction or seed-producing trees of desired species. Regardless of silvicultural system, maintaining seed sources of limiting softwoods, providing suitable germination substrates, and controlling competition are critical. Here, we describe commonalities among temperate mixedwoods in central and eastern North America and present a framework for managing them.

As interest in managing and maintaining mixedwood forests in the northern United States (US) grows, so does the importance of understanding their abundance and distribution. We analyzed Forest Inventory and Analysis data for insights into mixedwood forests spanning 24 northern US states from Maine south to Maryland and westward to Kansas and North Dakota. Mixedwoods, i.e., forests with both hardwoods and softwoods present but neither exceeding 75%–80% of composition, comprise more than 19 million hectares and more than one-quarter of the northern US forest. They are most common in the Adirondack – New England, Laurentian, and Northeast ecological provinces but also occur elsewhere in hardwood-dominated ecological provinces. These mixtures are common even within forest types nominally categorized as either hardwood or softwood. The most common hardwoods within those mixtures were species of Quercus and Acer, and the most common softwoods were species of Pinus, Tsuga, and Juniperus. Although mixedwoods exhibited stability in total area during our analysis period, hardwood saplings were prominent, suggesting widespread potential for eventual shifts to hardwood dominance in the absence of disturbances that favor regeneration of the softwood component. Our analyses suggest that while most mixedwood plots remained mixedwoods, harvesting commonly shifts mixedwoods to either hardwood- or softwood-dominated cover types, but more specific information is needed to understand the causes of these shifts.

Mixedwood stands are composed of hardwoods and softwoods, with neither comprising greater than 75%–80% of basal area or aboveground biomass. By conferring associational resistance and greater resilience to forests when stressed or disturbed, the more diverse composition of mixedwood stands contributes to forest health. We analyzed three examples where mixedwood stands are more resistant to insect infestations in eastern North America. In balsam fir (Abies balsamea L. Mill.) and spruce (Picea spp.) forests, susceptibility and vulnerability to spruce budworm infestations is reduced with increasing hardwood density. In mid-Atlantic oak-dominated forests, oak mortality following gypsy moth infestations was much greater in oak-dominated forests than in oak–pine mixedwoods, while pine mortality during southern pine beetle infestations was greater in pine-dominated forests than in oak–pine mixedwoods. Overall, mixedwood stands have greater resistance to infestations of defoliators and bark beetles and recover more rapidly from disturbances, reducing economic losses associated with tree mortality and mitigating short-term impacts to ecosystem functioning resulting from insect damage, especially carbon sequestration. Finally, we discuss challenges and opportunities for mixedwood management to minimize insect damage. Management strategies that incorporate mixedwood stands may provide better continuity in supply of forest products and ecosystem services in the face of projected increases in insect infestations associated with changing climate.

Currently, our ability to link wildland fire behaviour to fire effects is through the lens of fire severity assessments, because there are no ground-based post-fire metrics that are able to quantitively capture aspects of heat transfer to plants. This presents a particular challenge when considering tree mortality linked to cambial damage, which can occur in both low-intensity surface fires through to high-intensity crown fires. Recent research suggests that measuring the amount of light reflected from charcoals produced by wildland fires will provide information about the energy flux that created the char. We created an experimental forest fire in which we had instrumented individual trees to record the energy delivered to the bark close to the base of the trees. We then assessed the bark charcoal reflectance of the same trees. We found that bark charcoal reflectance showed a strong positive correlation (r2 > 0.86, P = 0.0031) with increasing duration of heating and the total energy delivered to the bark. We suggest that this may provide useful quantitative data that can be included in models or post-fire surveys to estimate tree mortality due to cambial kill.

ObjectivesThis ongoing Phase-2, randomised, placebo-controlled, double-blind study evaluated the efficacy, safety and pharmacokinetics of intravenous belimumab in childhood-onset systemic lupus erythematosus (cSLE).MethodsPatients (5 to 17 years) were randomised to belimumab 10 mg/kg intravenous or placebo every 4 weeks, plus standard SLE therapy. Primary endpoint: SLE Responder Index (SRI4) response rate (Week 52). Key major secondary endpoints: proportion of patients achieving the Paediatric Rheumatology International Trials Organisation/American College of Rheumatology (PRINTO/ACR) response using 50 and ‘30 alternative’ definitions (Week 52), and sustained response (Weeks 44 to 52) by SRI4 and Parent Global Assessment of well-being (Parent-global). Safety and pharmacokinetics were assessed. Study not powered for statistical testing.ResultsNinety-three patients were randomised (belimumab, n=53; placebo, n=40). At Week 52, there were numerically more SRI4 responders with belimumab versus placebo (52.8% vs 43.6%; OR 1.49 (95% CI 0.64 to 3.46)). PRINTO/ACR 30 alternative (52.8% vs 27.5%; OR 2.92 (95% CI 1.19 to 7.17)) and PRINTO/ACR 50 (60.4% vs 35.0%; OR 2.74 (95% CI 1.15 to 6.54)) responses were more frequent with belimumab than placebo, as were sustained responses for SRI4 (belimumab, 43.4%; placebo, 41.0%; OR 1.08 (95% CI 0.46 to 2.52)) and Parent-global (belimumab, 59.1%; placebo, 33.3%; OR 3.49 (95% CI 1.23 to 9.91)). Serious adverse events were reported in 17.0% of belimumab patients and 35.0% of placebo patients; one death occurred (placebo). Week-52, geometric mean (95% CI) belimumab trough concentration was 56.2 (45.2 to 69.8) µg/mL.ConclusionThe belimumab intravenous pharmacokinetics and benefit–risk profile in cSLE are consistent with adult belimumab studies and the 10 mg/kg every 4 weeks dose is appropriate.Trial registration numberNCT01649765.

Mesoscale models are a class of atmospheric numerical model designed to simulate atmospheric phenomena with horizontal scales of about 2–200 km, although they are also applied to microscale phenomena with horizontal scales of less than about 2 km. Mesoscale models are capable of simulating wildland fire impacts on atmospheric flows if combustion byproducts (e.g., heat, smoke) are properly represented in the model. One of the primary challenges encountered in applying a mesoscale model to studies of fire-perturbed flows is the representation of the fire sensible heat source in the model. Two primary methods have been implemented previously: turbulent sensible heat flux, either in the form of an exponentially-decaying vertical heat flux profile or surface heat flux; and soil temperature perturbation. In this study, the ARPS-CANOPY model, a version of the Advanced Regional Prediction System (ARPS) model with a canopy submodel, is utilized to simulate the turbulent atmosphere during a low-intensity operational prescribed fire in the New Jersey Pine Barrens. The study takes place in two phases: model assessment and model sensitivity. In the model assessment phase, analysis is limited to a single control simulation in which the fire sensible heat source is represented as an exponentially decaying vertical profile of turbulent sensible heat flux. In the model sensitivity phase, a series of simulations are conducted to explore the sensitivity of model–observation agreement to (i) the method used to represent the fire sensible heat source in the model and (ii) parameters controlling the magnitude and vertical distribution of the sensible heat source. In both phases, momentum and scalar fields are compared between the model simulations and data obtained from six flux towers located within and adjacent to the burn unit. The multi-dimensional model assessment confirms that the model reproduces the background and fire-perturbed atmosphere as depicted by the tower observations, although the model underestimates the turbulent kinetic energy at the top of the canopy at several towers. The model sensitivity tests reveal that the best agreement with observations occurs when the fire sensible heat source is represented as a turbulent sensible heat flux profile, with surface heat flux magnitude corresponding to the peak 1 min mean observed heat flux averaged across the flux towers, and an e-folding extinction depth corresponding to the average canopy height in the burn unit. The study findings provide useful guidance for improving the representation of the sensible heat released from low-intensity prescribed fires in mesoscale models.

The hazards associated with firebrands have been well documented. However, there exist few studies that allow for the hazard from a given fire to be quantified. To develop predictive tools to evaluate this hazard, it is necessary to understand the conditions that govern firebrand generation and those that affect firebrand deposition. A method is presented that allows for time-resolved measurements of fire behavior to be related to the dynamics of firebrand deposition. Firebrand dynamics were recorded in three fires undertaken in two different ecosystems. Fire intensity is shown to drive firebrand generation and firebrand deposition—higher global fire intensities resulting in the deposition of more, larger firebrands at a given distance from the fire front. Local firebrand dynamics are also shown to dominate the temporal firebrand deposition with periods of high fire intensity within a fire resulting in firebrand shower at deposition sites at times commensurate with firebrand transport. For the range of conditions studied, firebrand deposition can be expected up to 200 m ahead of the fire line based on extrapolation from the measurements.

Damage from infestations of Lymantria dispar L. in oak-dominated stands and southern pine beetle (Dendroctonus frontalis Zimmermann) in pine-dominated stands have far exceeded impacts of other disturbances in forests of the mid-Atlantic Coastal Plain over the last two decades. We used forest census data collected in undisturbed and insect-impacted stands combined with eddy covariance measurements made pre- and post-disturbance in oak-, mixed and pine-dominated stands to quantify how these infestations altered forest composition, structure and carbon dynamics in the Pinelands National Reserve of southern New Jersey. In oak-dominated stands, multi-year defoliation during L. dispar infestations resulted in > 40% mortality of oak trees and the release of pine saplings and understory vegetation, while tree mortality was minimal in mixed and pine-dominated stands. In pine-dominated stands, southern pine beetle infestations resulted in > 85% mortality of pine trees but had minimal effect on oaks in upland stands or other hardwoods in lowland stands, and only rarely infested pines in hardwood-dominated stands. Because insect-driven disturbances are both delaying and accelerating succession in stands dominated by a single genus but having less effect in mixed-composition stands, long-term disturbance dynamics are favoring the formation and persistence of uneven age oak-pine mixedwood stands. Changes in forest composition may have little impact on forest productivity and evapotranspiration; although seasonal patterns differ, with highest daily rates of net ecosystem production (NEP) during the growing season occurring in an oak-dominated stand and lowest in a pine-dominated stand, integrated annual rates of NEP are similar among oak-, mixed and pine-dominated stands. Our research documents the formation of mixedwood stands as a consequence of insect infestations in the mid-Atlantic region and suggests that managing for mixedwood stands could reduce damage to forest products and provide greater continuity in ecosystem functioning.

Turbulent fluxes of heat and momentum in the vicinity of wildland fires contribute to the redistribution of heat and momentum in the fire environment, which in turn can affect the heating of fuels, fire behavior, and smoke dispersion. As an extension of previous observational studies of turbulence regimes in the vicinity of wildland fires in forested environments, this study examines the effects of spreading surface fires and forest overstory vegetation on turbulent heat and momentum fluxes from near the surface to near the top of the overstory vegetation. Profiles of high-frequency (10 Hz) wind velocity and temperature measurements during two prescribed fire experiments are used to assess the relative contributions of horizontal and vertical turbulent fluxes of heat and momentum to the total heat and momentum flux fields. The frequency-dependent temporal variability of the turbulent heat and momentum fluxes before, during, and after fire-front passage is also examined using cospectral analyses. The study results highlight the effects that surface wildland fires and forest overstory vegetation collectively can have on the temporal and vertical variability of turbulent heat and momentum fluxes in the vicinity of the fires and the substantial departures of heat and momentum cospectra from typical atmospheric surface-layer cospectra that can occur before, during, and after fire-front passage.

Atmospheric turbulent circulations in the vicinity of wildland fire fronts play an important role in the transfer of momentum into and out of combustion zones, which in turn can potentially affect the behavior and spread of wildland fires. The vertical turbulent transfer of momentum is accomplished via individual sweep, ejection, outward interaction, and inward interaction events, collectively known as sweep-ejection dynamics. This study examined the sweep-ejection dynamics that occurred before, during, and after the passage of a surface fire front during a prescribed fire experiment conducted in an open-canopied forest in the New Jersey Pine Barrens. High-frequency (10 Hz), tower-based, sonic anemometer measurements of horizontal and vertical wind velocity components in the vicinity of the fire front were used to assess the relative frequencies of occurrence of the different types of momentum-flux events, their contributions to the overall momentum fluxes, and their periodicity patterns. The observational results suggest that the presence of surface fire fronts in open-canopied forests can substantially change the sweep-ejection dynamics that typically occur when fires are not present. In particular, sweep events resulting in the downward transport of high horizontal momentum air from above were found to be more prominent during fire-front-passage periods.

Large datasets of greenhouse gas and energy surface-atmosphere fluxes measured with the eddy-covariance technique (e.g., FLUXNET2015, AmeriFlux BASE) are widely used to benchmark models and remote-sensing products. This study addresses one of the major challenges facing model-data integration: To what spatial extent do flux measurements taken at individual eddy-covariance sites reflect model- or satellite-based grid cells? We evaluate flux footprints—the temporally dynamic source areas that contribute to measured fluxes—and the representativeness of these footprints for target areas (e.g., within 250–3000 m radii around flux towers) that are often used in flux-data synthesis and modeling studies. We examine the land-cover composition and vegetation characteristics, represented here by the Enhanced Vegetation Index (EVI), in the flux footprints and target areas across 214 AmeriFlux sites, and evaluate potential biases as a consequence of the footprint-to-target-area mismatch. Monthly 80% footprint climatologies vary across sites and through time ranging four orders of magnitude from 103 to 107 m2 due to the measurement heights, underlying vegetation- and ground-surface characteristics, wind directions, and turbulent state of the atmosphere. Few eddy-covariance sites are located in a truly homogeneous landscape. Thus, the common model-data integration approaches that use a fixed-extent target area across sites introduce biases on the order of 4%–20% for EVI and 6%–20% for the dominant land cover percentage. These biases are site-specific functions of measurement heights, target area extents, and land-surface characteristics. We advocate that flux datasets need to be used with footprint awareness, especially in research and applications that benchmark against models and data products with explicit spatial information. We propose a simple representativeness index based on our evaluations that can be used as a guide to identify site-periods suitable for specific applications and to provide general guidance for data use.

The vertical turbulent transfer of heat and momentum in the lower atmospheric boundary layer is accomplished through intermittent sweep, ejection, outward interaction, and inward interaction events associated with turbulent updrafts and downdrafts. These events, collectively referred to as sweep–ejection dynamics, have been studied extensively in forested and nonforested environments and reported in the literature. However, little is known about the sweep–ejection dynamics that occur in response to turbulence regimes induced by wildland fires in forested and nonforested environments. This study attempts to fill some of that knowledge gap through analyses of turbulence data previously collected during three wildland (prescribed) fires that occurred in grassland and forested environments in Texas and New Jersey. Tower-based high-frequency (10 or 20 Hz) three-dimensional wind-velocity and temperature measurements are used to examine frequencies of occurrence of sweep, ejection, outward interaction, and inward interaction events and their actual contributions to the mean vertical turbulent fluxes of heat and momentum before, during, and after the passage of fire fronts. The observational results suggest that wildland fires in these environments can substantially change the sweep–ejection dynamics for turbulent heat and momentum fluxes that typically occur when no fires are present, especially the relative contributions of sweeps versus ejections in determining overall heat and momentum fluxes.

Significance Forests remove about 30% of anthropogenic CO2 emissions through photosynthesis and return almost 40% of incident precipitation back to the atmosphere via transpiration. The trade-off between photosynthesis and transpiration through stomata, the water-use efficiency (WUE), is an important driver of plant evolution and ecosystem functioning, and has profound effects on climate. Using stable carbon and oxygen isotope ratios in tree rings, we found that WUE has increased by a magnitude consistent with estimates from atmospheric measurements and model predictions. Enhanced photosynthesis was widespread, while reductions in stomatal conductance were modest and restricted to moisture-limited forests. This result points to smaller reductions in transpiration in response to increasing atmospheric CO2, with important implications for forest–climate interactions, which remain to be explored., Multiple lines of evidence suggest that plant water-use efficiency (WUE)—the ratio of carbon assimilation to water loss—has increased in recent decades. Although rising atmospheric CO2 has been proposed as the principal cause, the underlying physiological mechanisms are still being debated, and implications for the global water cycle remain uncertain. Here, we addressed this gap using 30-y tree ring records of carbon and oxygen isotope measurements and basal area increment from 12 species in 8 North American mature temperate forests. Our goal was to separate the contributions of enhanced photosynthesis and reduced stomatal conductance to WUE trends and to assess consistency between multiple commonly used methods for estimating WUE. Our results show that tree ring-derived estimates of increases in WUE are consistent with estimates from atmospheric measurements and predictions based on an optimal balancing of carbon gains and water costs, but are lower than those based on ecosystem-scale flux observations. Although both physiological mechanisms contributed to rising WUE, enhanced photosynthesis was widespread, while reductions in stomatal conductance were modest and restricted to species that experienced moisture limitations. This finding challenges the hypothesis that rising WUE in forests is primarily the result of widespread, CO2-induced reductions in stomatal conductance.

Monitoring drought impacts in forest ecosystems is a complex process, because forest ecosystems are composed of different species with heterogeneous structural compositions. Even though forest drought status is a key control on the carbon cycle, very few indices exist to monitor and predict forest drought stress. The Forest Drought Indicator (ForDRI) is a new monitoring tool developed by the National Drought Mitigation Center (NDMC) to identify forest drought stress. ForDRI integrates 12 types of data, including satellite, climate, evaporative demand, ground water, and soil moisture, into a single hybrid index to estimate tree stress. The model uses Principal Component Analysis (PCA) to determine the contribution of each input variable based on its covariance in the historical records (2003–2017). A 15-year time series of 780 ForDRI maps at a weekly interval were produced. The ForDRI values at a 12.5km spatial resolution were compared with normalized weekly Bowen ratio data, a biophysically based indicator of stress, from nine AmeriFlux sites. There were strong and significant correlations between Bowen ratio data and ForDRI at sites that had experienced intense drought. In addition, tree ring annual increment data at eight sites in four eastern U.S. national parks were compared with ForDRI values at the corresponding sites. The correlation between ForDRI and tree ring increments at the selected eight sites during the summer season ranged between 0.46 and 0.75. Generally, the correlation between the ForDRI and normalized Bowen ratio or tree ring increment are reasonably good and indicate the usefulness of the ForDRI model for estimating drought stress and providing decision support on forest drought management.

Aerodynamic canopy height (h(a)) is the effective height of vegetation canopy for its influence on atmospheric fluxes and is a key parameter of surface-atmosphere coupling. However, methods to estimate h(a) from data are limited. This synthesis evaluates the applicability and robustness of the calculation of h(a) from eddy covariance momentum-flux data. At 69 forest sites, annual h(a) robustly predicted site-to-site and year-to-year differences in canopy heights (R-2=0.88, 111site-years). At 23 cropland/grassland sites, weekly h(a) successfully captured the dynamics of vegetation canopies over growing seasons (R-2>0.70 in 74site-years). Our results demonstrate the potential of flux-derived h(a) determination for tracking the seasonal, interannual, and/or decadal dynamics of vegetation canopies including growth, harvest, land use change, and disturbance. The large-scale and time-varying h(a) derived from flux networks worldwide provides a new benchmark for regional and global Earth system models and satellite remote sensing of canopy structure. Plain Language Summary Vegetation canopy height is a key descriptor of the Earth surface and is in use by many modeling and conservation applications. However, large-scale and time-varying data of canopy heights are often unavailable. This synthesis evaluates the applicability and robustness of the calculation of canopy heights from the momentum flux data measured at eddy covariance flux tower sites (i.e., meteorological observation towers with high frequency measurements of wind speed and surface fluxes). We show that the aerodynamic estimation of annual canopy heights robustly predicts the site-to-site and year-to-year differences in canopy heights across a wide variety of forests. The weekly aerodynamic canopy heights successfully capture the dynamics of vegetation canopies over growing seasons at cropland and grassland sites. Our results demonstrate the potential of aerodynamic canopy heights for tracking the seasonal, interannual, and/or decadal dynamics of vegetation canopies including growth, harvest, land use change, and disturbance. Given the amount of data collected and the diversity of vegetation covered by the global networks of eddy covariance flux tower sites, the flux-derived canopy height has great potential for providing a new benchmark for regional and global Earth system models and satellite remote sensing of canopy structure. There are few whole-canopy or ecosystem scale assessments of the interplay between canopy temperature and photosynthesis across both spatial and temporal scales. The stable oxygen isotope ratio (δ18O) of plant cellulose can be used to resolve a photosynthesis-weighted estimate of canopy temperature, but the method requires independent confirmation. We compare isotope-resolved canopy temperatures derived from multi-year homogenization of tree cellulose δ18O to canopy-air temperatures weighted by gross primary productivity (GPP) at multiple sites, ranging from warm temperate to boreal and subalpine forests. We also perform a sensitivity analysis for isotope-resolved canopy temperatures that showed errors in plant source water δ18O lead to the largest errors in canopy temperature estimation. Ambient and fire-induced atmospheric turbulence in the vicinity of wildland fires can affect the behavior of those fires and the dispersion of smoke. The presence of forest overstory vegetation can further complicate the evolution of local turbulence regimes and their interaction with spreading fires and smoke plumes. Previous observational studies of wildland fire events in forested environments have shown that turbulence energy and anisotropy in the vicinity of spreading line fires exhibit temporal and spatial variability influenced by the presence of overstory vegetation. This study builds on those previous observational studies to further examine turbulence regimes during two wildland fires in forested environments, with an emphasis on the effects of forest canopies on turbulence energy budgets, the skewness in turbulent velocity distributions, and stability–anisotropy variations before, during, and after fire-front-passage periods. Analyses indicate that turbulence anisotropy tends to persist throughout the vertical extent of overstory vegetation layers even during highly buoyant fire-front-passage periods, with horizontal velocity perturbations dominating over vertical velocity perturbations. The analyses also suggest that the periods before and after fire-front passage in forested environments can be very different with respect to how diffusion and shear production concurrently affect the evolution of turbulence energy within the canopy layer. In addition, horizontal and vertical velocity distribution analyses carried out in this study suggest that spreading line fires can have a substantial effect on the skewness of daytime velocity distributions typically observed inside forest vegetation layers.

A series of potent, selective and long-acting quinoline-based sulfonamide human H1 histamine receptor antagonists, designed for once-daily intranasal administration for the treatment of rhinitis were developed. Sulfonamide 33b had a slightly lower affinity for the H1 receptor than azelastine, had low oral bioavailability in the rat and dog, and was turned over to five major metabolites. Furthermore, 33b had longer duration of action than azelastine in guinea pigs, lower rat brain-penetration, and did not cause time dependent inhibition of CYP2D6 or CYP3A4. The clinical dose in humans is expected to be low (approximately 0.5mg per day) based on the clinical dose used for azelastine and a comparison of efficacy data from animal models for 33b and azelastine.

Antisense oligonucleotide (ASO) gapmers downregulate gene expression by inducing enzyme-dependent degradation of targeted RNA and represent a promising therapeutic platform for addressing previously undruggable genes. Unfortunately, their therapeutic application, particularly that of the more potent chemistries (e.g., locked-nucleic-acid-containing gapmers), has been hampered by their frequent hepatoxicity, which could be driven by hybridization-mediated interactions. An early de-risking of this liability is a crucial component of developing safe, ASO-based drugs. To rank ASOs based on their effect on the liver, we have developed an acute screen in the mouse that can be applied early in the drug development cycle. A single-dose (3-day) screen with streamlined endpoints (i.e., plasma transaminase levels and liver weights) was observed to be predictive of ASO hepatotoxicity ranking established based on a repeat-dose (15 day) study. Furthermore, to study the underlying mechanisms of liver toxicity, we applied transcriptome profiling and pathway analyses and show that adverse in vivo liver phenotypes correlate with the number of potent, hybridization-mediated off-target effects (OTEs). We propose that a combination of in silico OTE predictions, streamlined in vivo hepatotoxicity screening, and a transcriptome-wide selectivity screen is a valid approach to identifying and progressing safer compounds. An experimental fire was conducted in 2016, in the Pinelands National Reserve of New Jersey, to assess the reliability of the fire pattern indicators used in wildland fire investigation. Objects were planted in the burn area to support the creation of the indicators. Fuel properties and environmental data were recorded. Video and infrared cameras were used to document the general fire behavior. This work represents the first step in the analysis by developing an experimental protocol suitable for field studies and describing how different fire indicators appeared in relation to fire behavior. Most of the micro- and macroscale indicators were assessed. The results show that some indicators are highly dependent on local fire conditions and may contradict the general fire spread. Overall, this study demonstrates that fire pattern indicators are a useful tool for fire investigators but that they must be interpreted through a general analysis of the fire behavior with a good understanding of fire dynamics.

An experimental approach has been developed to quantify the characteristics and flux of firebrands during a management-scale wildfire in a pine-dominated ecosystem. By characterizing the local fire behavior and measuring the temporal and spatial variation in firebrand collection, the flux of firebrands has been related to the fire behavior for the first time. This linkage is seen as the first step in risk mitigation at the wildland urban interface (WUI). Data analyses allowed the evaluation of firebrand flux with respect to observed fire intensities for this ecosystem. Typical firebrand fluxes of 0.82–1.36 pcs m −2 s −1 were observed for fire intensities ranging between 7.35±3.48 MW m −1 to 12.59±5.87 MW m −1 . The experimental approach is shown to provide consistent experimental data, with small variations within the firebrand collection area. Particle size distributions show that small particles of area 0.75–5×10 −5 m 2 are the most abundant (0.6–1 pcs m −2 s −1 ), with the total flux of particles >5×10 −5 m 2 equal to 0.2–0.3 pcs m −2 s −1 . The experimental method and the data gathered show substantial promise for future investigation and quantification of firebrand generation and consequently a better description of the firebrand risk at the WUI.

The synthesis of potent amide-containing phthalazinone H1 histamine receptor antagonists is described. Three analogues 3e, 3g, and 9g were equipotent with azelastine and were longer-acting in vitro. Amide 3g had low oral bioavailability, low brain-penetration, high metabolic clearance, and long duration of action in vivo, and it was suitable for once-daily dosing intranasally, with a predicted dose for humans of approximately 0.5 mg per day.

Background: Belimumab (BEL), a monoclonal antibody targeting the B-lymphocyte stimulator, is approved in adults with active systemic lupus erythematosus (SLE). This is the first clinical trial of belimumab in pediatric patients with childhood-onset SLE (cSLE). Objectives: PLUTO, a Phase 2, randomised, double-blind trial (BEL114055; NCT01649765), evaluated the efficacy, safety and pharmacokinetics (PK) of intravenous (IV) BEL vs placebo (PBO), plus standard of care (SoC), in cSLE. Methods: Patients with cSLE 5–17 years of age were randomised to BEL 10 mg/kg IV or PBO every 4 weeks, plus SoC. Primary endpoint: SRI4 at Week 52. Major secondary endpoints: PRINTO/ACR 30 and 50 cSLE evaluation criteria for improvement at Week 52; cSLE core response variables at Week 52; and sustained SRI4 and ParentGA (patient well-being) responses (Weeks 44–52). Other endpoints: components of SRI4 at Week 52; and frequency of severe flares using the modified SELENA-SLEDAI Flare Index. Safety and PK were assessed. Analyses were performed on the intent-to-treat population. The study was not powered to test for differences between groups; p-values were not calculated. Results: 93 patients were included (BEL, n=53; PBO, n=40). Groups (BEL vs PBO) were balanced at baseline for age (mean [standard deviation] 13.5 [2.59] vs 14.8 [2.17] years, respectively) and SELENA-SLEDAI score (10.3 [3.34] vs 10.4 [3.63], respectively). Compared with PBO, there were more SRI4 responders (including all 3 components of SRI4), and PRINTO/ACR 30 and 50 responders in the BEL group (Figure). Likewise, more BEL than PBO recipients had sustained improvement of SRI and patient well-being (ParentGA) (Figure). Changes in cSLE core response variables are shown in the Table. Severe flares were 62% less frequent with BEL vs PBO (hazard ratio 0.38 [95% CI 0.18, 0.82]). PK: BEL exposures in cSLE were similar to adult SLE studies. 9/53 (17%) BEL patients had ≥1 serious adverse event vs 14/40 (35%) PBO patients. One PBO patient died of acute pancreatitis. Conclusion: The benefit:risk profile of BEL IV plus SoC in cSLE is generally consistent with BEL in adult SLE. The 10 mg/kg IV dose used in adults may be an appropriate dose in cSLE. This paper represents a study on the characterization of firebrand production which was carried out, using experimental fires conducted as prescribed fires in the New Jersey Pine Barrens, USA in March of 2013–2015. Several preliminary techniques were tested to characterize the firebrand production. Firebrands were collected from three plots for each year and analyzed for mass and size distribution. Thermal imagery was used to measure the velocity, size and number of firebrands in 2014 and 2015. The distribution of firebrands was evaluated in a monitored volume during the experiment. It was found that not less than 70% of collected particles were bark fragments and the rest were pine and shrub branches. The number of firebrands decreases with increasing the cross section area of firebrands. The mass of the particles varied from 5 to 50 mg, and the maximum number of the particles was observed for the mass range of 10–20 mg. About 80% of firebrands were particles with the cross section area of (5–20) × 10−5 m2. These results are consistent with the available observations of real fires [1] , [2] . Processing of infrared video showed that starting from a distance of 13 m from fire front, an increasing number of firebrands were observed in a controlled volume, increasing in number up to 180 per second. Relationships describing the time-variation of the number of particles that dropped on a 1.4 m2 surface and the number of particles that flew through a 1 m3 volume were obtained. Comparing the experimental and calculated data, we can conclude that these relationships allow us to describe the two numbers with an acceptable accuracy (adj. R2 0.74 and 0.86, respectively). In addition, the velocity of the particles, which depended on the wind velocity, was in the 0.1–10.5 m/s range, with an average value of 2.5 m/s.

Activities of burrowing herbivores, including movement of soil and litter and deposition of waste material, can alter the distribution of labile carbon (C) and nitrogen (N) in soil, affecting spatial patterning of nutrient dynamics in ecosystems where they are abundant. Their role in ecosystem processes in surface soil has been studied extensively, but effects of burrowing species on processes in subsurface soil remain poorly known. We investigated the effects of burrowing and grazing by plains vizcachas (Lagostomus maximus, Chinchilidae), a large colonial burrowing rodent native to South America, on the distribution and dynamics of C and N in soil of a semi-arid scrub ecosystem in central Argentina. In situ N mineralization (Nmin), potential Nmin and CO2 emissions were measured in surface soil (0–10 cm) and soil at the mean depth of burrows (65 ± 10 cm; mean ± 1 SD) in five colonial burrow systems and adjacent grazed and ungrazed zones. Decomposition and N dynamics of vizcacha feces on the soil surface and in burrow soil was assessed using litterbags. Total C and N in soil in burrows were 1.6 and 5.5 times greater than in undisturbed soil at similar depths, and similar to amounts in surface soil. Inorganic N, particularly NO3−, was consistently highest in burrows, intermediate in surface soil on burrow systems, and relatively low in all other zones. Despite high C and N content in all burrows, in situ net Nmin was highly variable in burrow soil. Feces decomposed and released N more rapidly in burrow soil. Laboratory incubations indicated that soil moisture limited Nmin under conditions that typically characterize burrow microclimate, and that rates increased dramatically at soil moisture contents >25% field capacity, which likely occurs during pulsed rainfall events. Thus, the high and seasonally stable NO3− content in burrow soil likely originated from the accumulation of pulsed mineralization events over time. Burrowing and waste deposition by vizcachas produced “resource islands” at the landscape scale. At a measured density of 0.3 burrow systems per hectare, colonial burrow soil contained an amount of inorganic N equal to 21% and 30% of plant-available N in surface soil and subsurface soil, respectively, in an area that represented only 0.35% of the landscape. Our study indicates that burrowing and deposition of waste results in a highly active subsurface layer in which C and N dynamics function much like surface soil when soil moisture is not limiting. Ecosystem models often perform poorly in reproducing interannual variability in carbon and water fluxes, resulting in considerable uncertainty when estimating the land-carbon sink. While many aggregated variables (growing season length, seasonal precipitation, or temperature) have been suggested as predictors for interannual variability in carbon fluxes, their explanatory power is limited and uncertainties remain as to their relative contributions. Recent results show that the annual count of hours where evapotranspiration (ET) is larger than its 95th percentile is strongly correlated with the annual variability of ET and gross primary production (GPP) in an ecosystem model. This suggests that the occurrence of favorable conditions has a strong influence on the annual carbon budget. Here we analyzed data from eight forest sites of the AmeriFlux network with at least 7 years of continuous measurements. We show that for ET and the carbon fluxes GPP, ecosystem respiration (RE), and net ecosystem production, counting the "most active hours/days" (i.e., hours/days when the flux exceeds a high percentile) correlates well with the respective annual sums, with correlation coefficients generally larger than 0.8. Phenological transitions have much weaker explanatory power. By exploiting the relationship between most active hours and interannual variability, we classify hours as most active or less active and largely explain interannual variability in ecosystem fluxes, particularly for GPP and RE. Our results suggest that a better understanding and modeling of the occurrence of large values in high-frequency ecosystem fluxes will result in a better understanding of interannual variability of these fluxes.

The exchange of carbon between the Earth’s atmosphere and biosphere influences the atmospheric abundances of carbon dioxide (CO2) and methane (CH4). Airborne eddy covariance (EC) can quantify surface-atmosphere exchange from landscape-to-regional scales, offering a unique perspective on carbon cycle dynamics. We use extensive airborne measurements to quantify fluxes of sensible heat, latent heat, CO2, and CH4 across multiple ecosystems in the Mid-Atlantic region during September 2016 and May 2017. In conjunction with footprint analysis and land cover information, we use the airborne dataset to explore the effects of landscape heterogeneity on measured fluxes. Our results demonstrate large variability in CO2 uptake over mixed agricultural and forested sites, with fluxes ranging from −3.4 ± 0.7 to −11.5 ± 1.6 μmol m−2 s−1 for croplands and −9.1 ± 1.5 to −22.7 ± 3.2 μmol m−2 s−1 for forests. We also report substantial CH4 emissions of 32.3 ± 17.0 to 76.1 ± 29.4 nmol m−2 s−1 from a brackish herbaceous wetland and 58.4 ± 12.0 to 181.2 ± 36.8 nmol m−2 s−1 from a freshwater forested wetland. Comparison of ecosystem-specific aircraft observations with measurements from EC flux towers along the flight path demonstrate that towers capture ∼30%–75% of the regional variability in ecosystem fluxes. Diel patterns measured at the tower sites suggest that peak, midday flux measurements from aircraft accurately predict net daily CO2 exchange. We discuss next steps in applying airborne observations to evaluate bottom-up flux models and improve understanding of the biophysical processes that drive carbon exchange from landscape-to-regional scales.

Prescribed fires are conducted extensively in pine-dominated forests throughout the Eastern USA to reduce the risk of wildfires and maintain fire-adapted ecosystems. We asked how fire behavior and fuel consumption during prescribed fires are associated with turbulence and energy fluxes, which affect the dispersion of smoke and transport of firebrands, potentially impacting local communities and transportation corridors. We estimated fuel consumption and measured above-canopy turbulence and energy fluxes using eddy covariance during eight prescribed fires ranging in behavior from low-intensity backing fires to high-intensity head fires in pine-dominated forests of the New Jersey Pinelands, USA. Consumption was greatest for fine litter, intermediate for understory vegetation, and least for 1 + 10 hour wood, and was significantly correlated with pre-burn loading for all fuel types. Crown torching and canopy fuel consumption occurred only during high-intensity fires. Above-canopy air temperature, vertical wind velocity, and turbulent kinetic energy (TKE) in buoyant plumes above fires were enhanced up to 20.0, 3.9 and 4.1 times, respectively, compared to values measured simultaneously on control towers in unburned areas. When all prescribed fires were considered together, differences between above-canopy measurements in burn and control areas (&Delta, values) for maximum &Delta, air temperatures were significantly correlated with maximum &Delta, vertical wind velocities at all (10 Hz to 1 minute) integration times, and with &Delta, TKE. Maximum 10 minute averaged sensible heat fluxes measured above canopy were lower during low-intensity backing fires than for high-intensity head fires, averaging 1.8 MJ m&minus, 2 vs. 10.6 MJ m&minus, 2, respectively. Summed &Delta, sensible heat values averaged 70 &plusmn, 17%, and 112 &plusmn, 42% of convective heat flux estimated from fuel consumption for low-intensity and high-intensity fires, respectively. Surprisingly, there were only weak relationships between the consumption of surface and understory fuels and &Delta, air temperature, &Delta, wind velocities, or &Delta, TKE values in buoyant plumes. Overall, low-intensity fires were effective at reducing fuels on the forest floor, but less effective at consuming understory vegetation and ladder fuels, while high-intensity head fires resulted in greater consumption of ladder and canopy fuels but were also associated with large increases in turbulence and heat flux above the canopy. Our research quantifies some of the tradeoffs involved between fire behavior and turbulent transfer of smoke and firebrands during effective fuel reduction treatments and can assist wildland fire managers when planning and conducting prescribed fires.

Local point measurements of fire dynamics in field-scale experiments of wildland fires are highly useful. This is true both for understanding the mechanisms driving fire spread that result in the observed macroscopic behaviors, but also in terms of providing comparison points for numerical tools, such as detailed physics-based fire behavior models. This work describes measurements of temperature, velocity, and radiative heat flux that were made in a field-scale fire experiment in a pine forest, with the aim of providing both of the above benefits. Regions of both surface fire and crown fire were captured and are compared. The crown fire exhibited tall upright flames, compared to the shorter tilted flames of the surface fire. Crown fire resulted in a significant increase in integrated radiative preheating, by a factor of ∼1.75, as well as greater flow sheltering in the downstream region of the fire front. Further, a corrective factor is introduced for oblique sensor placement relative to the fire front, in order to improve the value of these and other measurements, particularly for model comparison. The presented methodology, while able to be improved, is shown to successfully characterize local differences in fire behavior.

Understanding processes underlying forest carbon dynamics is essential for accurately predicting the outcomes of non-stand-replacing disturbance in intermediate-age forests. We quantified net ecosystem production (NEP), aboveground net primary production (ANPP), and the dynamics of major carbon (C) pools before and during the decade following invasive insect defoliation and prescribed fires in oak- and pine-dominated stands in the New Jersey Pinelands National Reserve, USA. Gross ecosystem production (GEP) recovered during the year following defoliation at the oak stand, but tree mortality increased standing dead and coarse woody debris, and ecosystem respiration (Re) accounted for >97% of GEP. As a result, NEP averaged only 22% of pre-disturbance values during the decade following defoliation. At the pine stand, GEP also recovered to pre-disturbance values during the year following understory defoliation by gypsy moth and two prescribed fires, while Re was nearly unaffected. Overall, defoliation and tree mortality at the oak stand drove a decadal-scale reduction in NEP that was twofold greater in magnitude than C losses associated with prescribed fires at the pine stand. Our study documents the outcomes of different non-stand-replacing disturbances, and highlights the importance of detrital dynamics and increased Re in long-term measurements of forest C dynamics following disturbance in intermediate-age forests.

This study aims to develop a series of robust and efficient methodologies, which can be applied to understand and estimate firebrand generation and to evaluate firebrand showers close to a fire front. A field scale high intensity prescribed fire was conducted in the New Jersey Pine Barrens in March 2013. Vegetation was characterised with field and remotely sensed data, fire spread and intensity was characterised and meteorological conditions were monitored before and during the burn. Firebrands were collected from different locations in the forest and analysed for mass and size distribution. The majority were found to be bark slices (more than 70%) with substantial amounts of pine and shrub twigs. Shrub layer consumption was evaluated to supplement the firebrand generation study. Bark consumption was studied by measuring the circumference variation at several heights on each of three different pine trees. The variation was in the same order of magnitude as the bark thickness (1–5 mm). Testing and improving the protocol can facilitate the collection of compatible data in a wide range of ecosystems and fire environments, aiding in the development of solutions to prevent structural ignition at the Wildland Urban Interface.
