Gray sufu is a traditional Chinese fermented soybean curd, and its flavor is significantly influenced by bacterial community. In this study, volatile flavor compounds (VFCs) were examined by headspace solid‐phase microextraction (HS-SPME) in conjunction with gas chromatography and mass spectrometry (GC‐MS). Additionally, bacterial community was analyzed using high-throughput sequencing. The results showed that, a total of 156 VFCs were tentatively identified, in which 8 esters, 6 S/N-containing compounds, 3 alcohols, 3 aldehydes, 1 ketone and 3 other compounds were recognized as the main characteristic VFCs. The abundant S/N-containing compounds are the most important flavor characteristics of gray sufu. A total of 4 phyla and 12 genera were revealed as dominant bacteria. Firmicutes and Proteobacteria were the most abundant phylum, whereas Bacillus, Tetragenococcus and Lactococcus were the prevailing genera. According to the KEGG pathway annotation, amino acid metabolism (26.98%) was the highest metabolic pathway. Among them, aromatic amino acids metabolism accounted for the highest proportion, up to 34.94%. Correlation analysis showed that Lactobacillus, Tetragenococcus, Bacillus, Pseudomonas and Halanaerobium were the top five positive flavor-producing bacteria. The results will be helpful for understanding the fermentation mechanism of gray sufu and improving flavor quality by developing microbial regulation strategies.

IntroductionDue to its remarkable anti-inflammatory pharmacological activity, Farfarae Flos has gained extensive usage in the treatment of various inflammatory diseases such as bronchitis, pneumonia, prostatitis and colitis. And Farfarae Flos come in two color types depending on the color of the flowers: yellowish-white (YW), and purplish-red (PR). However, the difference in anti-inflammatory activity and metabolic profiles between the two flower colors remains unexplored.MethodsThis study aims to explore the difference in the anti-inflammatory potential between YW and PR variants of Farfarae Flos and unravel the mechanisms responsible for the observed differences in anti-inflammatory activity through an integrated approach encompassing untargeted metabolomics and in vivo/vitro experimental studies. Initially, we verified the contrasting effects of YW and PR on the inhibition of the inflammatory factors interleukin-6 (IL-6) and nitric oxide (NO) by utilizing an in vitro RAW 264.7 cell inflammation model. Subsequently, a comprehensive evaluation of the systemic inhibitory capacity of YW and PR on IL-6, Interleukin-10 (IL-10), and tumor necrosis factor-α (TNF-α) was conducted using a validated whole-body mouse model, followed by the analysis of inflammatory factors and histological examination of collected serum, liver, and spleen after 7 days. Furthermore, non-targeted metabolomics profiling was employed to analyze the metabolite profiles of Farfarae Flos with different colors, and quantitative analysis was conducted to identify differential metabolites between YW and PR. The correlation between the anti-inflammatory activities of differentially accumulated metabolites (DAMs) and Farfarae Flos was investigated, resulting in the identification of 48 compounds exhibiting significant anti-inflammatory activity. Additionally, KEGG pathway enrichment analysis was performed to elucidate the underlying mechanisms.ResultsOur findings demonstrate that both YW and PR possess anti-inflammatory abilities, with PR exhibiting significantly superior efficacy. The integration of in vivo/vitro experiments and non-targeted metabolomics confirmed the exceptional anti-inflammatory potential of PR and solidified its classification as the “purplish-red better” of Farfarae Flos.DiscussionThis study provides valuable insights into the breeding and medical transformation of Farfarae Flos varieties, along with a scientific basis for the establishment of quality standards and the development of new drugs utilizing Farfarae Flos.

ABSTRACT A total of 334 Salmonella isolates were recovered from 6,223 pet rectal samples collected at 50 pet clinics, 42 pet shops, 7 residential areas, and 4 plazas. Forty serovars were identified that included all strains except for one isolate that did not cluster via self-agglutination, with Salmonella Typhimurium monophasic variant, Salmonella Kentucky, Salmonella Enteritidis, Salmonella Pomona, and Salmonella Give being the predominant serovars. Fifty-one sequence types were identified among the isolates, and ST198, ST11, ST19, ST451, ST34, and ST155 were the most common. The top four dominant antimicrobials to which isolates were resistant were sulfisoxazole, ampicillin, doxycycline, and tetracycline, and 217 isolates exhibited multidrug resistance. The prevalence of β-lactamase genes in Salmonella isolates was 59.6%, and among these isolates, 185 harbored blaTEM, followed by blaCTX-M (66) and blaOXA (10). Moreover, six PMQR genes, namely, including qnrA (4.8%), qnrB (4.2%), qnrD (0.9%), qnrS (18.9%), aac(6')-Ib-cr (16.5%), and oqxB (1.5%), were detected. QRDR mutations (76.6%) were very common in Salmonella isolates, with the most frequent mutation in parC (T57S) (47.3%). Furthermore, we detected six tetracycline resistance genes in 176 isolates, namely, tet(A) (39.5%), tet(B) (8.1%), tet(M) (7.7%), tet(D) (5.4%), tet(J) (3.3%), and tet(C) (1.8%), and three sulfonamide resistance genes in 303 isolates, namely, sul1 (84.4%), sul2 (31.1%), and sul3 (4.2%). Finally, we found 86 isolates simultaneously harboring four types of resistance genes that cotransferred 2–7 resistance genes to recipient bacteria. The frequent occurrence of antimicrobial resistance, particularly in dogs and cats, suggests that antibiotic misuse may be driving multidrug-resistant Salmonella among pets.IMPORTANCEPet-associated human salmonellosis has been reported for many years, and antimicrobial resistance in pet-associated Salmonella has become a serious public health problem and has attracted increasing attention. There are no reports of Salmonella from pets and their antimicrobial resistance in Chongqing, China. In this study, we investigated the prevalence, serovar diversity, sequence types, and antimicrobial resistance of Salmonella strains isolated from pet fecal samples in Chongqing. In addition, β-lactamase, QRDR, PMQR, tetracycline and sulfonamide resistance genes, and mutations in QRDRs in Salmonella isolates were examined. Our findings demonstrated the diversity of serovars and sequence types of Salmonella isolates. The isolates were widely resistant to antimicrobials, notably with a high proportion of multidrug-resistant strains, which highlights the potential direct or indirect transmission of multidrug-resistant Salmonella from pets to humans. Furthermore, resistance genes were widely prevalent in the isolates, and most of the resistance genes were spread horizontally between strains.

This study focuses on the land transfer intentions of migrants and surrounding villagers in the SZ resettlement area of BS City, Guangxi. It systematically analyzes the coupling coordination relationship between migrants’ land transfer-in intentions and the land transfer-out intentions of surrounding villagers, verifying the practical value of the “Shared Land Resource Model” in the resettlement area and its surroundings. The study yields the following key conclusions: (1) there is a strong coupling between the land demand intentions of migrants and the land supply intentions of surrounding villagers, yet the actual coordination in the transfer process is limited, which constrains resource allocation efficiency and prevents land transfer from fully utilizing shared resources; (2) in the evaluation of migrants’ land transfer-in intentions, external environmental factors have the greatest influence (with a weight coefficient of 0.7877), while individual characteristics (0.0486) and psychological characteristics (0.0593) have relatively low weight coefficients, indicating that migrants primarily rely on government policy support and lack internal motivation; (3) the land transfer-out intentions of surrounding villagers are most affected by farmland resource endowment (weight coefficient of 0.3284), indicating that the quality and quantity of land resources are key factors affecting villagers’ transfer-out willingness, while individual endowment factors have the smallest impact (weight coefficient of 0.1220). Three recommendations are proposed: stimulating migrants’ intrinsic motivation to enhance livelihood autonomy, protecting villagers’ land rights to increase transfer participation, and building a systematic land resource sharing model to promote sustainable resource allocation. This study provides theoretical support for optimizing the land transfer mechanism in resettlement areas, aiming to improve land use efficiency, support the livelihood transition of migrants, and offer practical insights for land management planning in poverty alleviation and resettlement projects in other countries.

Within the context of agricultural green development, the construction of high-standard farmland plays a crucial role in enhancing agricultural productivity, ensuring food security, and achieving sustainable development. However, as the direct participants, farmers’ willingness and enthusiasm to engage in high-standard farmland construction largely determines the success of these initiatives. Therefore, based on data from 326 samples in Qinghai Province, this study employs a structural equation model to analyze the impact of farmers’ cognition and the current condition of farmland on the willingness to participate in high-standard farmland construction and verify the mediating effect of information trust. The results indicate the following: (1) the impact pathways of farmers’ cognition on participation willingness vary across different dimensions, with subjective norms and behavioral attitudes having a direct positive influence, while perceived behavioral control shows no direct effect; (2) the current condition of farmland has a direct and significant negative effect on participation willingness; (3) information trust exhibits varying mediating effects across different pathways, showing a partial mediating effect on behavioral attitudes and subjective norms, a full mediating effect on perceived behavioral control, and a suppressive effect on farmland condition. This study, grounded in the context of China’s green agricultural development goals, provides new insights into the relationship between government policy implementation and farmers’ willingness to participate. It establishes a scientific foundation for policymakers to formulate strategic green development policies.

Tandem alkali-catalyzed hydrolysis and alkaline electrolysis have gradually become appealing avenues for the reformation of polyester plastics into high-value-added chemicals and green hydrogen with remarkable environmental and economic benefits. In this study, an electrochemical upcycling strategy was developed for the electrocatalytic oxidation of polylactic acid (PLA) hydrolysate into valued C2 chemicals (i.e., acetate) and hydrogen fuel using N, P-doped CuOx nanowires (NW) supported on nickel foam (NF) as the electrocatalyst. This 3D well-integrated catalyst was easily prepared from a Cu(OH)2 NW/NF precursor with Saccharomycetes as a green and safe P and N source. The electrocatalyst can efficiently catalyze the lactate monomer derived from the hydrolysis of PLA waste to acetate with high selectivity and exhibits a lower onset potential for the lactate oxidation reaction (LOR) than for water oxidation, saving 224 mV to deliver a current density of 30 mA/cm2. The experimental results reveal that the plausible pathway of the LOR on these CuOx NW involves oxidation and subsequent decarboxylation. Divalent copper species have been verified to be active sites for LOR via in situ Raman spectroscopy.

Abstract Background To explore the value of magnetic resonance angiography (MRA) and high resolution magnetic resonance vessel wall imaging (HRMR-VWI) in cervicocranial artery dissection (CCAD) for the disease diagnosis, course staging and treatment. On the basis of qualitative evaluation, this study also extract the changes of different stages in vessel wall in different vessel segments to identify imaging indicators for the quantitative evaluation of CCAD. Methods We retrospectively enrolled 34 patients with CCAD (38branches) with conventional MRA and HRMR-VWI examinations. Two radiologists independently analyzed imaging features of vessel wall and lumen in the different stages, and the typical sign detection of artery dissection were compared between MRA and HRMR-VWI. Then the parameters of vessel wall was quantitatively evaluated by the post-processing software (Vesselmass, Leiden University Medical Center, Leiden, The Netherlands. Results HRMR-VWI revealed typical sign detection of artery dissection in all patients in the acute and subacute stage. Among them, the intimal flap/double lumen sign ditection were more common than the MRA, there was significant difference (P = 0.012). MRA revealed typical sign detection of artery dissection in more than half the patients, and the detection was no significant difference at the chronic stage between MRA and HRMR-VWI (P = 1.000/1.000/0.761). In the acute and subacute stage, the typical sign detection of intramural hematoma and Grade II enhancement revealed by HR-MRI was higher than the observations in the chronic stage (P = 0.000/0.000/0.016), while there was no significant difference by MRA (P = 0.902). The values of wall thickness, relative signal intensity of vessel wall enhancement, relative signal intensity of intramural hematoma (IMH), and percentage of stenosis in CCAD decreased from acute to subacute and then to chronic stages. Each quantitative parameter in patients with CCAD in the early stages (i.e., acute and subacute stages) was significantly different from that in patients with CCAD in the recovered group at chronic stage (P 0.05). Conclusions As the only noninvasive imaging technology, HRMR-VWI displays the structure of the vessel wall in vivo, showing not only excellent performance in the early diagnosis of CCAD, but also describing the changes of different stages in the qualitative and quantitative characteristics of vessel wall. It also helps to guide the diseasediagnosis, course staging and treatment of CCAD. Although the diagnostic efficacy of MRA was not as good as HRMR-VWI, it should be the first choice of method for routine examination in evaluating CCAD, especially at the chronic stage of CCAD.

Nocardia seriolae is the primary pathogen causing nocardiosis in various fish species, leads to significant economic losses in the aquaculture industry. In this study, 10 bacterial strains isolated from Micropterus salmoides and Channa argus infected with nocardiosis, were identified as N. seriolae by physiological and biochemical identification, as well as 16S rDNA sequencing. Moreover, the key virulence-related genes such as ESX-1, T7SS-2, T7SS-3, EspG1, sodC, sod2 and ESAT6 were all positive, and showing high homology among different strains. Pathogenicity testing revealed mortality rates ranging from 70 to 100%, accompanied by the presence of white nodules in the viscera of deceased fish. The drug sensitivity test demonstrated that LY21811, the most lethal strain, exhibited high sensitivity to nine types of antibiotics, including azithromycin, doxycycline, florfenicol and compound sulfamethoxazole, yet showed complete resistance to β-lactam antibiotics. Additionally, the tannic acid also demonstrated potent inhibitory effects against LY21811, with a minimum inhibitory concentration of 0.0625 mg/mL. These results showed that N. seriolae originated from M. salmoides and C. argus in Zhejiang Province were highly conserved, demonstrating a high homogeneity in genetic characteristics, pathogenicity and antimicrobial susceptibilities. These results provide a foundation for further research on the pathogenic characteristics and disease prevention of N. seriolae infections.

Although hydropower serves as a clean alternative energy source, the global challenge of resettlement due to dam construction underscores the importance of sustainable livelihoods in achieving the targets of the Sustainable Development Goals (SDGs). In recent years, the sustainable livelihood theory has offered a new perspective for examine the development of livelihoods of reservoir migrants after relocation. However, in-depth relationship between social capital and sustainable livelihoods has not been explored. In this study, three hydropower resettlement areas were selected in the G Autonomous Prefecture of China as an example. Our investigation delved into the influence of social networks, social norms, and social trust on the livelihoods of reservoir migrants. Study employed semi-structured interviews, participant observation, and qualitative analysis to explore this impact. The results show that the traditional social capital stock of reservoir migrants, dominated by family, neighborhood and religious networks, has declined. Therefore, the structural function of modern social capital should be strengthened by increasing the social trust of reservoir migrants in institutions and civil society organizations. The contribution of livelihood diversification in poverty reduction by 2030 (SDG-1) was realized by focusing on the role of social capital to promote infrastructure, improve living conditions and increase employment opportunities for migrants. The findings further point to the need to fully respect the customs and lifestyles of ethnic minorities during the resettlement process of reservoir migrants in order to preserve the role played by their traditional social capital, thereby promoting the sustainable development of the livelihoods of reservoir migrants.

The existing object detection methods are facing significant challenges when applied to marine environments, such as underwater image degradation caused by absorption and scattering of light, and domain transfer in water bodies with different water qualities. In this letter, we propose a marine organism detection framework to improve the detection performance and the domain generalization performance. First, a double domains data augmentation is proposed. This method combines underwater image enhancement and water quality transfer to improve the domain diversity of the original dataset. Second, we utilize the self-attention operations and the convolution to improve the detection performance of the YOLOv7, fully utilizing the advantages of self-attention and convolutional computation. Meanwhile, this model uses SIoU loss to accelerate convergence speed and improve the regression. Experiments on the URPC2019 and URPC2020 datasets show that the proposed object detection method achieves a mean average precision of 82.3% and 83.6%, respectively, which is superior to all other methods used for comparison.

Abstract Background Professional calling (PC) is crucial for ascertaining their professional goals and fulfilling career choices in nursing students. Thus, understanding its antecedents and helping schools improve PC among nursing students is critical. This study aims to explore whether professional identity (PI), as a crucial antecedent of PC, acts as an intermediary between career self-efficacy (CSE) and professional calling during the COVID-19 pandemic. Methods A sample of 565 nursing students were selected by a web-based survey through convenience sampling. The study was conducted from October to November 2020. Measures of CSE, PI, and PC were assessed during the COVID-19 pandemic. We analyzed demographic data and the correlation of the research variables. As urbanization has accelerated, China has started to build smart cities, which have formed smart-city clusters. It is critical to coordinate development within smart-city clusters to enhance the efficiency of city-cluster construction. From the perspective of demographic economics, this study innovatively constructed an evaluation system for the coordinated development of smart-city clusters and utilized the coupled coordination degree model to conduct an in-depth study of smart-city clusters in Jiangsu Province. The results show that there are clear differences in the development between the three regions of Jiangsu Province: Southern Jiangsu, Central Jiangsu, and Northern Jiangsu. The development within Jiangsu Province is imbalanced, where the overall development trend is high in the southern region and low in the northern region. The main driving factors include geography, the Matthew effect, game thinking, and industrial structure. Accordingly, the results suggest the following recommendations for the coordinated development of smart-city clusters: strengthening cross-regional cooperation, promoting data sharing and interoperability, deepening synergistic industrial development, and expanding innovation capacity.

The construction of reservoirs has led to difficult livelihood transitions of resettled households after relocation and resettlement, resulting in a series of socioeconomic problems. How to scientifically integrate regional resource advantages and improve the livelihood resilience of resettled households has become an important problem to be solved to prevent and resolve social equity and justice risks and sustainable livelihood development. Taking Xiangjiaba Hydropower Station (Yunnan Reservoir area) as an example, the evaluation index system was constructed from the three dimensions of buffer capacity, self-organization capacity and learning capacity, the gray correlation degree method was used to evaluate index system, and the clustering method was selected to screen the indices that have a great impact on livelihood resilience. On this basis, the comprehensive index method was used to calculate the livelihood resilience, and the contribution degree model was used to identify the main contribution factors of livelihood resilience and to explore the path of livelihood construction to improve the livelihood resilience of resettled households. The results showed that (1) the level of livelihood resilience of resettled households was not high as a whole, in which the self-organization capacity was the strongest, the buffer capacity was the second strongest, and the learning capacity was the weakest; (2) there were differences in the livelihood resilience of resettled households who choose different livelihood modes. Among them, buffer capacity, learning capacity and livelihood resilience were characterized by wage operation type > part-time balanced type > agricultural operation type > subsidy dependent type, while self-organization capacity was characterized by agricultural operation type > part-time balanced type > wage operation type > subsidy dependent type. (3) The main contribution factors of the livelihood resilience of resettled households who choose different livelihood modes are similar. The number of laborers, participation in social organizations, skills training opportunities and 11 other main indices promote the formation and development of livelihood resilience from different dimensions. (4) The livelihood of resettled households should follow the construction path of “identifying transition capacity, clarifying resource advantages, strengthening livelihood support, and promoting livelihood reorganization” to promote the comprehensive improvement of livelihood resilience, ultimately achieving sustainable livelihood and high-level welfare. The livelihood resilience of resettled households is a continuous and dynamic development process, therefore, it is necessary to further improve the dynamic monitoring and evolution process of livelihood resilience, incorporate the impact of diverse data. This is also an important breakthrough in the study of livelihood resilience.

ObjectivesPrevious studies primarily explored the unidirectional impact of cognition on physical function. However, the interplay between physical function and cognition and the temporal precedence in their predictive relationships have not been elucidated. We explored the bidirectional mechanism between physical function and cognition in a longitudinal dataset.Materials and methodsA total of 1,365 participants in the Chinese Longitudinal Healthy Longevity Survey assessed physical function and cognition in 2011 (T1), 2014 (T2), and 2018 (T3) by the Katz scale and the Chinese version of the Mini-Mental State Examination scale, respectively. Changes in the trajectories of physical function and cognition were examined using the latent growth model. The correlational and reciprocal relationships between physical function and cognition were examined using the parallel process latent growth model and autoregressive cross-lagged (ARCL) models.ResultsCognition and physical function decreased by an average of 0.096 and 0.017 points per year, respectively. Higher physical function was associated with better cognition at baseline (r = 0.237, p < 0.05), and longitudinal changes in physical function and cognition were positively correlated (r = 0.756, p < 0.05). ARCL analysis indicated that physical function at T1 positively predicted T2 cognitive function. However, this predictive relationship reversed between T2 and T3, whereby cognitive function at T2 predicted physical function at T3.ConclusionBoth physical function and cognition declined over time. Early identification and intervention in physical dysfunction among older adults could be critical to prevent further cognitive impairment and maintain functional independence. Hence, regular functional assessment and individualized care plans are required to achieve healthy aging.

Material identification is playing an increasingly important role in various sectors such as industry, petrochemical, mining, and in our daily lives. In recent years, material identification has been utilized for security checks, waste sorting, etc. However, current methods for identifying materials require direct contact with the target and specialized equipment that can be costly, bulky, and not easily portable. Past proposals for addressing this limitation relied on non-contact material identification methods, such as Wi-Fi-based and radar-based material identification methods, which can identify materials with high accuracy without physical contact; however, they are not easily integrated into portable devices. This paper introduces a novel non-contact material identification based on acoustic signals. Different from previous work, our design leverages the built-in microphone and speaker of smartphones as the transceiver to identify target materials. The fundamental idea of our design is that acoustic signals, when propagated through different materials, reach the receiver via multiple paths, producing distinct multipath profiles. These profiles can serve as fingerprints for material identification. We captured and extracted them using acoustic signals, calculated channel impulse response (CIR) measurements, and then extracted image features from the time–frequency domain feature graphs, including histogram of oriented gradient (HOG) and gray-level co-occurrence matrix (GLCM) image features. Furthermore, we adopted the error-correcting output code (ECOC) learning method combined with the majority voting method to identify target materials. We built a prototype for this paper using three mobile phones based on the Android platform. The results from three different solid and liquid materials in varied multipath environments reveal that our design can achieve average identification accuracies of 90% and 97%.

Objectives: Although the evidence from numerous longitudinal studies has indicated a remarkable change in cognitive function (CF) and depressive symptoms (DS) over time, the parallel latent growth curve model (LGCM) has seldom been used to simultaneously investigate the relationship between their change trajectories. This study aimed to examine whether a change in DS was associated with CF over time using an LGCM.Methods: Data were collected from the Chinese Longitudinal Healthy Longevity Survey’s 2011, 2014, and 2018 waves. A parallel LGCM examined the association between CF and DS.Results: The multivariate conditioned model’s goodness of fit supported the validity of the longitudinal model (Tucker-Lewis index [TLI] = 0.90, comparative fit index [CFI] = 0.96, root mean square error of approximation [RMSEA] = 0.04). The results showed that the CF intercept was positively to the DS slope (β = 0.42, p = 0.004). The CF and DS slopes were significantly linked (β = −0.65, p = 0.002).Conclusion: The findings expand the knowledge about CF’s effect on DS in older adults.

Hydrolyzed chicken meat extract (HCE) has previously been shown to mitigate inflammation and oxidative stress, and enhance cognitive function in humans. However, it is still not clear whether HCE and its bioactive components (BC) could affect immune function. Herein, we found that both HCE and BC administration promoted the phagocytic activity and attenuated the oxidative stress in RAW 264.7 cells. Moreover, HCE and BC treatment caused a significant decrease in M1 macrophage activation and an increase in M2 macrophage activation in RAW264.7 cells and peritoneal macrophages, resulting in an M2 dominant shift in macrophages. These effects of HCE and BC might be partly regulated by suppressing the MAPK and NF-κB pathways and enhancing the STAT6 and Akt pathways. Cyclo(Val-Pro) isolated from BC exerted clear anti-inflammatory and antioxidative effects, which might be partly responsible for the impacts of HCE and BC. Therefore, HCE and BC might serve as potential immunoregulatory agents.

Dietary intervention with a low glycemic index and full nutritional support is emerging as an effective strategy for diabetes management. Here, we found that the treatment of a novel compound dietary fiber and high-grade protein diet (CFP) improved glycemic control and insulin resistance in streptozotocin-induced diabetic mice, with a similar effect to liraglutide. In addition, CFP treatment ameliorated diabetes-related metabolic syndromes, such as hyperlipidemia, hepatic lipid accumulation and adipogenesis, systemic inflammation, and diabetes-related kidney damage. These results were greatly associated with enhanced gut barrier function and altered gut microbiota composition and function, especially those bacteria, microbial functions, and metabolites related to amino acid metabolism. Importantly, no adverse effect of CFP was found in our study, and CFP exerted a wider arrange of protection against diabetes than liraglutide. Thereby, fortification with balanced dietary fiber and high-grade protein, like CFP, might be an effective strategy for the management and treatment of diabetes.

To date, general-purpose object-detection methods have achieved a great deal. However, challenges such as degraded image quality, complex backgrounds, and the detection of marine organisms at different scales arise when identifying underwater organisms. To solve such problems and further improve the accuracy of relevant models, this study proposes a marine biological object-detection architecture based on an improved YOLOv5 framework. First, the backbone framework of Real-Time Models for object Detection (RTMDet) is introduced. The core module, Cross-Stage Partial Layer (CSPLayer), includes a large convolution kernel, which allows the detection network to precisely capture contextual information more comprehensively. Furthermore, a common convolution layer is added to the stem layer, to extract more valuable information from the images efficiently. Then, the BoT3 module with the multi-head self-attention (MHSA) mechanism is added into the neck module of YOLOv5, such that the detection network has a better effect in scenes with dense targets and the detection accuracy is further improved. The introduction of the BoT3 module represents a key innovation of this paper. Finally, union dataset augmentation (UDA) is performed on the training set using the Minimal Color Loss and Locally Adaptive Contrast Enhancement (MLLE) image augmentation method, and the result is used as the input to the improved YOLOv5 framework. Experiments on the underwater datasets URPC2019 and URPC2020 show that the proposed framework not only alleviates the interference of underwater image degradation, but also makes the mAP@0.5 reach 79.8% and 79.4% and improves the mAP@0.5 by 3.8% and 1.1%, respectively, when compared with the original YOLOv8 on URPC2019 and URPC2020, demonstrating that the proposed framework presents superior performance for the high-precision detection of marine organisms.

Timely sowing and harvesting play important roles in agricultural production. The appropriate management decisions are necessary to cope with climate change and ensure high and stable crop yields. This study analyzed the effects of sowing date on the growth process of winter wheat and quantified the effects of climate resources and photothermal potential yield on theoretical yield at different stages of winter wheat. The analysis was based on the data from winter wheat interval sowing experiments conducted at the Hebei Gucheng Agricultural Meteorology National Observation and Research Station (Gucheng station) in north China (115°40′ E, 39°08′ N) during 2017–2019. The results showed that: (1) with the delay in sowing date, the growth process of winter wheat significantly advanced, the proportion of vegetative growth period significantly reduced (0.19% for per day delay), the proportion of reproductive growth period (RGP) significantly increased (0.12% for per day delay), and the prewintering light and temperature resources significantly reduced (12.2 °C·d accumulated temperature and 19.0 MJ·m−2 solar radiation for per day delay); (2) the theoretical yield of winter wheat showed a significant exponential relationship with the photothermal potential yield of the whole growth period: the minimum photothermal potential for yield formation was 26.6 t·ha−1, and the maximum theoretical yield was 12.6 t·ha−1; and (3) the wheat yield and yield stability were highest when the RGP photothermal potential yield was 16.0 t·ha−1 and the prewintering active accumulated temperature was 400 °C·d. This study also proposed a method to estimate the suitable sowing and harvesting dates to achieve high and stable yield of winter wheat, showing that the suitable sowing dates of winter wheat at Gucheng station from 1997 to 2021 ranged from 1 to 15 October, with no significant interannual variation; the suitable harvesting period ranged from 5 June to 10 July and showed a trend of gradual advance with the delay of the year. The results of the study provide a reference for sowing date adjustment of crops to adapt to climate change.

In this work, we characterized a novel chromosome-encoded AmpC β-lactamase gene, blaPRC–1, in an isolate of a newly classified Pseudomonas species designated Pseudomonas wenzhouensis A20, which was isolated from sewage discharged from an animal farm in Wenzhou, China. Susceptibility testing, molecular cloning, and enzyme kinetic parameter analysis were performed to determine the function and enzymatic properties of the β-lactamase. Sequencing and comparative genomic analysis were conducted to clarify the phylogenetic relationship and genetic context of the blaPRC–1 gene. PRC-1 is a 379-amino acid AmpC β-lactamase with a molecular weight of 41.48 kDa and a predicted pI of 6.44, sharing the highest amino acid identity (57.7%) with the functionally characterized AmpC β-lactamase PDC-211 (ARX71249). blaPRC–1 confers resistance to many β-lactam antibiotics, including penicillins (penicillin G, amoxicillin, and amoxicillin-clavulanic acid) and cephalosporins (cefazolin, ceftriaxone, and cefotaxime). The kinetic properties of PRC-1 were compatible with those of a typical class C β-lactamase showing hydrolytic activities against β-lactam antibiotics, and the hydrolytic activity was strongly inhibited by avibactam. The genetic context of blaPRC–1 was relatively conserved, and no mobile genetic element was predicted in its surrounding region. Identification of a novel β-lactamase gene in an unusual environmental bacterium reveals that there might be numerous unknown resistance mechanisms in bacterial populations, which may pose potential risks to human health due to universal horizontal gene transfer between microorganisms. It is therefore of great value to carry out extensive research on the mechanism of antibiotic resistance.

Multidrug-resistant bacteria from different sources have been steadily emerging, and an increasing number of resistance mechanisms are being uncovered. In this work, we characterized a novel resistance gene named aac(2′)-If from an isolate of a novel Providencia species, Providencia wenzhouensis R33 (CCTCC AB 2021339). Susceptibility testing and enzyme kinetic parameter analysis were conducted to determine the function of the aminoglycoside 2′-N-acetyltransferase. Whole-genome sequencing and comparative genomic analysis were performed to elucidate the molecular characteristics of the genome and the genetic context of the resistance gene-related sequences. Among the functionally characterized resistance genes, AAC(2′)-If shares the highest amino acid sequence identity of 70.79% with AAC(2′)-Ia. AAC(2′)-If confers resistance to several aminoglycoside antibiotics, showing the highest resistance activity against ribostamycin and neomycin. The recombinant strain harboring aac(2′)-If (pUCP20-aac(2′)-If/DH5α) showed 256- and 128-fold increases in the minimum inhibitory concentration (MIC) levels to ribostamycin and neomycin, respectively, compared with those of the control strains (DH5α and pUCP20/DH5α). The results of the kinetic analysis of AAC(2′)-If were consistent with the MIC results of the cloned aac(2′)-If with the highest catalytic efficiency for ribostamycin (kcat/Km ratio = [3.72 ± 0.52] × 104 M–1⋅s–1). Whole-genome sequencing demonstrated that the aac(2′)-If gene was located on the chromosome with a relatively unique genetic environment. Identification of a novel aminoglycoside resistance gene in a strain of a novel Providencia species will help us find ways to elucidate the complexity of resistance mechanisms in the microbial population.

With the continuous improvement of people’s health awareness and the continuous progress of scientific research, consumers have higher requirements for the quality of drinking. Compared with high-sugar-concentrated juice, consumers are more willing to accept healthy and original Not From Concentrated (NFC) juice and packaged drinking water. At the same time, drinking category detection can be used for vending machine self-checkout. However, the current drinking category systems rely on special equipment, which require professional operation, and also rely on signals that are not widely used, such as radar. This paper introduces a novel drinking category detection method based on wireless signals and artificial neural network (ANN). Unlike past work, our design relies on WiFi signals that are widely used in life. The intuition is that when the wireless signals propagate through the detected target, the signals arrive at the receiver through multiple paths and different drinking categories will result in distinct multipath propagation, which can be leveraged to detect the drinking category. We capture the WiFi signals of detected drinking using wireless devices; then, we calculate channel state information (CSI), perform noise removal and feature extraction, and apply ANN for drinking category detection. Results demonstrate that our design has high accuracy in detecting drinking category.

Laser cladding technology is a reliable and efficient surface modification technology, which has been widely used in surface alloying and composite processing of steel materials. Firstly, the characteristics of laser cladding technology were introduced, and the effects of process control and the material system on the geometric shape, size, microstructure, and properties of cladding coating were analyzed by summarizing the research results of laser cladding on steel surfaces. The results show that with the increase of laser power, the dilution rate and width of the cladding coating increase, and the grain becomes coarse. Thus, the wear resistance deteriorates. Compared with alloy cladding coating, composite cladding coating exhibits better wear and corrosion resistance, but the plastic toughness is worse than alloy cladding coating. The research progress of surface alloying and composite processing of steel worldwide was analyzed from various aspects. Current results suggest that laser cladding alloying and compounding can enhance the wear resistance and corrosion resistance of steel materials. Based on the summary of the current research results, the development prospect and planning of laser cladding technology in the field of surface alloying and composite processing of steel are further pointed out.

A novel plasmid-encoded aminoglycoside 3''-nucleotidyltransferase ANT(3")-IId, was discovered in Acinetobacter lwoffi strain H7 isolated from a chick on an animal farm in Wenzhou, China. The whole-genome of A. lwoffii H7 consisted of one chromosome and five plasmids (pH7-250, pH7-108, pH7-68, pH7-48, and pH7-11). ant(3")-IId was identified as being encoded on pH7-250, sharing the highest amino acid identity of 50.64% with a function-known resistance gene, ant(3")-IIb (KB849358.1). Susceptibility testing and enzyme kinetic parameter analysis were conducted to determine the function of the aminoglycoside 3"-nucleotidyltransferase. The ant(3")-IId gene conferred resistance to spectinomycin and streptomycin [the minimum inhibitory concentration (MIC) levels of both increased 16-fold compared with the control strain]. Consistent with the MIC data, kinetic analysis revealed a narrow substrate profile including spectinomycin and streptomycin, with Kcat/Km ratios of 4.99 and 4.45×103M−1 S−1, respectively. Sequencing analysis revealed that the ant(3")-IId gene was associated with insertion sequences (IS) element [ΔISAba14-ΔISAba14-hp-orf-orf-orf1-ant(3")-IId], and ant(3")-IId were identified in plasmids from various Acinetobacter species. This study of the novel aminoglycoside 3"-nucleotidyltranferase ANT(3")-IId helps us further understand the functional and sequence characteristics of aminoglycoside 3"-nucleotidyltranferases, highlights the risk of resistance gene transfer among Acinetobacter species and suggests that attention should be given to the emergence of new aminoglycoside 3"-nucleotidyltranferase genes.

Coagulase-negative Staphylococcus warneri is an opportunistic pathogen that is capable of causing several infections, especially in patients with indwelling medical devices. Here, we determined the complete genome sequence of a clinical S. warneri strain isolated from the blood culture of a 1-year-old nursling patient with acute upper respiratory infection. Genome-wide phylogenetic analysis confirmed the phylogenetic relationships between S. warneri and other Staphylococcus species. Using comparative genomics, we identified three cell wall-anchored (CWA) proteins at the same locus (sdr), named SdrJ, SdrK, and SdrL, on the chromosome sequences of different S. warneri strains. Structural predictions showed that SdrJ/K/L have structural features characteristic of Sdr proteins but exceptionally contained an unusual N-terminal repeat region. However, the C-terminal repetitive (R) region of SdrJ contains a significantly larger proportion of alanine (142/338, 42.01%) than the previously reported SdrI (37.00%). Investigation of the genetic organization revealed that the sdrJ/K/L genes were always followed by one or two glycosyltransferase genes, gtfA and gtfB and were present in an ∼56 kb region bordered by a pair of 8 bp identical direct repeats, named Sw-Sdr. This region was further found to be located on a 160-kb region subtended by a pair of 160-bp direct repeats along with other virulence genes and resistance genes. Sw-Sdr contained a putative integrase that was probably a remnant of a functional integrase. Evidence suggests that Sw-Sdr is improbably an efficient pathogenicity island. A large-scale investigation of Staphylococcus genomes showed that sdr loci were a potential hotspot of insertion sequences (ISs), which could lead to intraspecific diversity at these loci. Our work expanded the repository of Staphylococcus Sdr proteins, and for the first time, we established the connection between sdr loci and phylogenetic relationships and compared the sdr loci in different Staphylococcus species, which provided large insights into the genetic environment of CWA genes in Staphylococcus.

Delftia tsuruhatensis has become an emerging pathogen in humans. There is scant information on the genomic characteristics of this microorganism. In this study, we determined the complete genome sequence of a clinical D. tsuruhatensis strain, TR1180, isolated from a sputum specimen of a female patient in China in 2019. Phylogenetic and average nucleotide identity analysis demonstrated that TR1180 is a member of D. tsuruhatensis. TR1180 exhibited resistance to β-lactam, aminoglycoside, tetracycline and sulphonamide antibiotics, but was susceptible to phenicols, fluoroquinolones and macrolides. Its genome is a single, circular chromosome measuring 6,711,018 bp in size. Whole-genome analysis identified 17 antibiotic resistance-related genes, which match the antimicrobial susceptibility profile of this strain, as well as 24 potential virulence factors and a number of metal resistance genes. Our data showed that Delftia possessed an open pan-genome and the genes in the core genome contributed to the pathogenicity and resistance of Delftia strains. Comparative genomics analysis of TR1180 with other publicly available genomes of Delftia showed diverse genomic features among these strains. D. tsuruhatensis TR1180 harbored a unique 38-kb genomic island flanked by a pair of 29-bp direct repeats with the insertion of a novel In4-like integron containing most of the specific antibiotic resistance genes within the genome. This study reports the findings of a fully sequenced genome from clinical D. tsuruhatensis, which provide researchers and clinicians with valuable insights into this uncommon species.

The novel pleuromutilin derivative, which showed excellent in vitro antibacterial activity against MRSA, 22-(2-(2-(4-((4-(4-nitrophenyl)piperazin-1-yl)methyl)-1H-1,2,3-triazol-1-yl)acetamido)phenyl)thioacety-l-yl-22-deoxypleuromutilin (Z33), was synthesized and characterized in our previous work. In this study, the preliminary pharmacodynamics and safety of Z33 were further evaluated. In in vitro antibacterial activity assays, Z33 was found to be a potent bactericidal antibiotic against MRSA that induced dose-dependent growth inhibition and long-term post-antibiotic effect (PAE). The drug-resistance test demonstrated that Z33 possessed a narrow mutant selection window and lower propensities to select resistance than that of tiamulin. Cytochrome P450 (CYP450) inhibition assay determined that the inhibitory effect of Z33 was similar to that of tiamulin against the activity of CYP3A4, and was lower than that of tiamulin on the activity of CYP2E1. Toxicity determination showed that both Z33 and tiamulin displayed low cytotoxicity of RAW264.7 cells. Furthermore, Z33 was found to be a high-security compound with a 50% lethal dose (LD50) above 5000 mg/kg in the acute oral toxicity test in mice. In an in vivo antibacterial activity test, Z33 displayed better therapeutic effectiveness than tiamulin in the neutropenic mouse thigh infection model. In summary, Z33 was worthy of further development as a highly effective and safe antibiotic agent against MRSA infection.

Rodents constitute the largest and most successful group of mammals worldwide. Brown rats (Rattus norvegicus) are one of the most common rodent species, and they serve as intermediate hosts of Hydatigera taeniaeformis. Although there have been a few studies reporting on the presence of the larval form of H. taeniaeformis (strobilocercus fasciolaris) in brown rats worldwide, little information is available on the genetic characterization of this parasite, with no molecular data from China. Therefore, from April 2014 to March 2016, this study was carried out to understand the prevalence and genetic characters of strobilocercus fasciolaris in brown rats captured in Heilongjiang Province in northeastern China. The livers of brown rats were collected and examined for the presence of cysts. Each cyst was identified based on morphological observation: the larvae with the naked eye and the scolexes under a microscope. The results were confirmed by polymerase chain reaction (PCR) and sequencing of the cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase subunit 4 (nad4) genes. At the investigated sites, 11.8% (13/110) of the brown rats were infected with strobilocercus fasciolaris. Based on sequence analysis, there were 10 and six haplotypes regarding the cox1 and the nad4 loci, with 24 and 42 polymorphic sites, respectively (degree of intraspecific variation: 0.3%–4.4% and 0.6%–4.7%, respectively). Twelve nucleotide sequences (six of the 10 at the cox1 locus and all six at the nad4 locus) have not previously been described. Base differences in three of the six novel cox1 gene sequences and five of the six novel nad4 gene sequences caused amino acid changes. Phylogenetic analyses of the cox1 and nad4 gene sequences based on neighbor-joining and Bayesian inference trees indicated that all the strobilocercus fasciolaris isolates belonged to Hydatigera taeniaeformis sensu stricto (s.s.). This is the first report on the genetic characterization of strobilocercus fasciolaris in brown rats in China. The findings of novel cox1 and nad4 nucleotide and amino acid sequences may reflect the region-specific genetic characterization of the parasite. The data will be useful to explore the biological and epidemiological significance of the intraspecific variation within H. taeniaeformis s.s.

Enterocytozoon bieneusi is one of the most common intestinal pathogens in humans and animals. E. bieneusi has been confirmed to be complex microsporidian species. Approximately 500 ITS genotypes of E. bieneusi have been defined. With the establishment and application of multilocus sequencing typing and population genetic tools in E. bieneusi, the studies on these aspects have been carried out worldwide, but little information is available. To understand genetic variation of mini-/micro-satellites and the population structure and substructure of E. bieneusi in northeastern China, 305 E. bieneusi DNA specimens composed of 28 ITS genotypes were from 13 mammal species and five bird species in the investigated areas. They were characterized by nested-PCR amplification and sequencing at four mini-/micro-satellite loci (MS1, MS3, MS4, and MS7). At the MS1, MS3, MS4, and MS7 loci, 153 (50.16%), 131 (42.95%), 133 (43.61%), and 128 (41.97%) DNA specimens were amplified and sequenced successfully with 44, 17, 26, and 24 genotypes being identified, respectively. Multilocus genotypes (MLGs) showed a higher genetic diversity than ITS genotypes. 48 MLGs were produced out of 90 ITS-positive DNA specimens based on concatenated sequences of all the five genetic loci including ITS. Linkage disequilibrium (LD) and limited genetic recombination were observed by measuring LD using both multilocus sequences and allelic profile data, indicating an overall clonal population structure of E. bieneusi in the investigated areas. These data will aid in the longitudinal tracking of the attribution of source of infection/contamination and in elucidating transmission dynamics, and will provide valuable information for making efficient control strategies to intervene with and prevent occurrence of microsporidiosis caused by E. bieneusi among animals and transmission of E. bieneusi from animals to humans in the investigated areas. Phylogenetic and network analyses identified three different subgroups, revealing the presence of host-shaped segregation and the absence of geographical segregation in E. bieneusi population. Meanwhile, the MLGs from zoonotic ITS genotypes were observed to be basically separated from the MLGs from host-adapted ones. Assessment of substructure will have a reference effect on understanding of zoonotic or interspecies transmission of E. bieneusi and evolution direction from zoonotic genotypes to host-adapted genotypes.

Abstract Background Fatigue is highly prevalent among doctors worldwide. However, no research has been done to examine the associations of emotional intelligence (EI) and emotional labor strategy with fatigue among Chinese doctors. This study aimed to examine whether or not emotional labor strategy mediates the association between EI and fatigue in this occupational group. Methods A cross-sectional study was conducted in Shenyang from March to April 2014. A set of self-administered questionnaires was distributed to 950 doctors, including Chalder Fatigue Scale (CFS), Wong and Law Emotional Intelligence Scale (WLEIS) and a 14-item emotional labor scale. Complete responses were obtained from 740 (77.9%) participants. Hierarchical linear regression was performed to examine the associations of EI and emotional labor strategies (surface acting, SA; deep acting, DA; natural acting, NA) with fatigue. Asymptotic and resampling strategies were used to examine the mediating roles of emotional labor strategies. Results The mean score of fatigue was 8.02 (SD = 3.39). The Ni-Ti-Cr-carbon nanotubes (CNTs) composite coatings was successfully fabricated by laser cladding. The optical microscope was used to observe the metallographic structure of the composite coating, the phase composition of the composite layer was detected by XRD, the microstructures of the composite coatings were observed by SEM, and the point distribution and line distribution of elements were analyzed by EDS. With the increase of laser specific energy (Es) and CNTs content, the TiC enhanced particles in the composite coatings evolves into coarse dendrites. Compared with the mild steel substrate, the microhardness and wear resistance of the composite coatings are obviously improved. The maximum microhardness obtained by the composite coating is approximately 5 times that of the mild steel substrate. The increase of Es and the excessive content of CNTs will reduce the microhardness and wear resistance of the composite coatings. The in situ synthesis of TiC particles not only enhance the microhardness of the composite coatings, but also improve the wear resistance of the coatings.

Background. Health-promoting behaviors and positive lifestyle changes are crucial for effective stroke prevention. However, individuals at high risk of stroke exhibit poor health behavior due to a deficiency of individual motivation. Moreover, there are only a few studies on health-promoting behaviors that have applied behavior change theories in individuals at high risk of stroke. Objective. This study aimed to use the theory of the planned behavior (TPB) model to investigate determinants of health-promoting behaviors for stroke prevention and control. Method. In this cross-sectional study, 263 participants were recruited from five community health centers in Qingdao. Confirmatory factor analysis was performed to assess the reliability and validity of the constructs, and structural equation modeling was used to analyze the proposed relationships between the TPB-related variables. Results. The attitudes, subjective norms, and perceptions of behavioral control positively influenced behavioral intention. The behavioral intention had a positive effect on health-promoting behaviors. Attitudes, subjective norms, and perceived behavioral control were influenced primarily by the mediating variable behavioral intention to affect health-promoting behaviors. Stroke knowledge was an influential facilitator of behavioral attitudes, subjective norms, and perceived behavior control. Conclusion. The TPB-based model is suitable for explaining health-promoting behaviors in individuals at risk of stroke and for guiding the development of effective health management programs. A comprehensive person-centered motivation behavior strategy that is based on health education and complemented by social support and health resource optimization is critical in promoting health behavior motivation and health promotion behaviors in stroke high-risk groups.

The willingness to take action against climate change may be shaped by cultural orientations. The present study investigated individualist–collectivist differences in climate change inaction as well as the mediating role of perceived intractability. In Study 1, a survey of 182 undergraduates showed that greater perceived intractability of climate change was significantly related to a lower frequency of climate-friendly actions in the preceding 6 months. In Study 2, participants who were exposed to information concerning the intractability of climate change (experimental group, n = 98) reported a significantly greater perceived intractability of climate change and lower intention to assume a low-carbon lifestyle than those presented with neutral information (control group, n = 83). Based on Studies 1 and 2, participants with collectivist or individualist orientations were recruited from a pool of Chinese undergraduate students in Study 3. We found that participants with a more individualist orientation (n = 62) are more subject to perceived intractability, and less likely to take climate-friendly action than those with a more collectivist orientation (n = 94), and individualist/collectivist status affects climate change inaction through perceived intractability as mediator. The implications of these findings are discussed in relation to the promotion of public engagement with climate change by mitigating perceived intractability.

The influence of environmental attitudes on environmental behaviors has long been discussed. However, few studies have addressed the foundation of such attitudes. In the present study, we explored primitive belief underlying environmental attitudes, i.e., connections with nature, and its relationship with pro-environmental behaviors. Specifically, we used scales, a computerized Implicit Association Test, and a situational simulation experiment to examine both explicit and implicit connections with nature, both deliberate and spontaneous environmental behaviors, and to find correlations between environmental connectedness and environmental behaviors. Results showed that explicit connectedness was positively correlated with deliberate environmental behaviors, while implicit connectedness was positively correlated with spontaneous environmental behaviors. Additionally, explicit and implicit connectedness was independent of each other. In conclusion, the current study confirms the positive role played by connections with nature in promoting environmental behavior, and accordingly suggests means to encourage pro-environmental behavior by enhancing people's connectedness to nature.
