9. Chaperonin 60 : molecular, cellular and structural studies to define its role as an information molecule

Abstract

Experimental evidence obtained in this thesis establishes that the purified, recombinant, E. coli chaperonin 60 protein, GroEL, is a potent stimulator of cytokine production by human peripheral blood mononuclear cells (PBMCs) and that this cytokine inducing activity is not due to LPS, or to the proteins that contaminate "purified' GroEL. Surprisingly, trypsinization of GroEL failed to inhibit the cytokine-inducing activity of this protein, and by use of reverse phase HPLC, MALDI-MS and N-terminal sequencing, the bioactive GroEL tryptic peptides were identified. The question as to whether physical interaction was required between GroEL and PBMCs in order to activate the cells was answered by flow cytometry, which demonstrated that leukocytes responding to GroEL, bind the protein whereas cells unresponsive to GroEL do not. Single- and double-labelling experiments revealed that only a proportion of CD-14 positive monocytes in PBMCs bind GroEL. GroEL was also shown to stimulate macrophage cells and cell lines, which suggests that the proportion of monocytes observed to bind GroEL may have a more macrophage-like morphology. Affinity chromatography of surface biotinylated PBMCs on GroEL affinity matrices indicates a 100-150kDa and a 35-40kDa band as possible receptors for GroEL, or as part of the GroEL receptor complex. It has also been demonstrated that GroEL does not stimulate cells via CD14 (a known LPS receptor) as this protein remains active in the presence of anti-CD14 antibodies and is unable to stimulate the CD14 positive cell line Mono Mac-6. These studies support the hypothesis that chaperonin 60 is a bacterial virulence factor able to interact with the host's immune system and induce inflammation.

16. Legitimizing State-sponsored Militants: The Role of Civil Institutions in Pakistan

Abstract

Using and sponsoring militant groups as proxies to meet geostrategic interests is an old political phenomenon. IR research indicates that state’s sponsor militant groups to increase their regional influence, destabilize regional rivals, export political ideology, maintain economic stability, preserve plausible deniability, and deter a more powerful rival. Yet, none of these motivations adequately explain the institutional foundations and the role of a state’s national identity in sponsorship. In this dissertation I present a theory for continued state-sponsorship, arguing that sponsorship of militant groups is linked to its ontological security, which refers to the security acquired by having a stable, consistent identity. I argue that sponsorship of militancy satisfies the state’s need of having a stable and consistent identity, and ultimately increases ontological security. How a state obtains and maintains its ontological security is deeply tied to its ability to achieve its geostrategic interests. To better understand how sponsorship of militant groups functions as a means to increase the state’s ontological security, I specify the institutional foundations and requirements for maintaining a state’s ontological security, and ask: Does a state’s sponsorship of militancy have the same or different effects on its territorial security versus its ontological security? What are the processes of ontological security? What is the role of civil institutions in the processes of ontological security? And how do civil institutions facilitate state-sponsorship of militant groups? I use Pakistan to illustrate the value of this approach and investigate how its civil institutions help legitimize the state’s policy of sponsorship as a means to enhance the state’s ontological security. Employing a reflexive approach and an ontological security framework to the study of state-sponsorship yields several important findings for IR theory, mainstream and critical security studies, and interdisciplinary approaches to studying militancy. First, my research emphasizes that while its external environment influences a state’s identity, it is nonetheless created internally by the interaction of its civil and military institutions. Second, disaggregating the state allows us to better understand how “security” is created and how closely “security” is intertwined with “identity” and “national interests.” Unpacking the state, therefore, adds more theoretical and empirical value to IR theory. Finally, by revealing the contributions of the Pakistani state’s civil institutions in the state’s policy of sponsoring militant groups, this dissertation disrupts the conventional understanding of Pakistan’s institutions, which has mainly considered the military as the primary institution involved in the sponsorship of militant groups. My dissertation highlights how this conventional view is incomplete and needs to be reexamined.

22. Organisational justice and organisational performance: mediation role of organisational citizenship behaviour in business institutes of Pakistan

Abstract

The purpose of this research was to examine the relationship of the factors of organisational justice (OJ), i.e., distributive, interactional, and procedural justice on organisational performance, through organisational citizenship behaviour (OCB), i.e., altruism and general compliance. The researchers have conducted quantitative research which utilised a close-ended questionnaire comprising of five-point Likert scale items for gathering data from 217 faculty members of business institutes in Pakistan. This research was conducted in the private higher educational institutions in Pakistan. The data were analysed by using the Smart-PLS and SEM is used for checking the multiple relationships among the variables. The findings of this research indicated that each dimension of OJ is important in the education sector of Pakistan for enhancing the organisational performance through OCB each dimension. This research responds to the most important variables contributing to the educational sector. Moreover, the educational institutes would get the benefit of this research to practice and apply policy implications. HR personnel and those concerned can understand which factors of OCB and OJ enhance organisational performance.

Abstract

Introduction: Computer usage has become an indispensable tool in the official set up of all the workplaces in the current era. Predominance of musculoskeletal disorders (MSDs) in relation to habitual posture during work is the utmost problem of modern society. Teaching staff stand out amongst a group of workers exposed to occupational MSDs. The objective of this study was to identify the prevalence and risk factors of musculoskeletal disorders in relation to posture and computer ergonomics at workplace among the college and university staff in Petaling Jaya, Malaysia. Methods: This cross-sectional quantitative study was conducted from August 2019-October 2019, among 419 volunteers by using a self-administered survey questionnaire. Descriptive and bivariate statistics were used for the analyses of multiple variables. The association between demographic characteristics, computer ergonomics and prevalence of musculoskeletal pains were analyzed through Chi-square test. Results: 55.8% respondents (n=234) reported neck pain (NP), (n=196) 46.8% shoulder pain (SP) and (n=308) 73.5% low back pain (LBP) respectively. A significant relationship between desktop computer usage and musculoskeletal pains in LBP (P=0.036) and SP (P=0.023) was observed. Significant association of head posture was found with NP (P=0.002), SP (P=0.042) and LBP (P=0.001), correspondingly. Discussion: Habitual postures were significantly associated with musculoskeletal pains while using computer. Conclusion: This study proved with precession that higher prevalence rate of musculoskeletal disorders was undoubtedly influenced by prolonged sitting, awkward postures at workstation, and repetitive movements of shoulders and hands. Abstract

Perovskites CaPd3B4O 1 2 (B = Ti, V) are studied theoretically using generalized gradient approximation (GGA), GGA-modified Becke–Johnson (GGA-mBJ), GGA with spin-orbit coupling (GGA + SOC) and hybrid functional (HF) in the domain of density functional theory (DFT). The estimated structural parameters are reliable with the experimentally reported data. Cohesive energy and enthalpy show that these compounds are stable thermodynamically. Bonding nature makes known that the chemical bond between Ca/Ti–O is ionic, Pd/V–O is covalent and Ti/V–Ti/V is metallic. The mechanical properties show that these compounds are stable, elastically anisotropic and ductile in nature. CaPd3Ti4O 1 2 is a 2.94 eV direct-wide bandgap semiconductor through GGA-mBJ and consistent with experiments. The optical properties show that CaPd3Ti4O 1 2 is a good dielectric material. The dense electronic states and the wide-gap semiconductor nature of CaPd3Ti4O 1 2 suggest that it can be used as a good thermoelectric material. [ABSTRACT FROM AUTHOR]

Abstract

Background: Tea (Camellia sinensis, Theaceae) is the second most consumed beverage in the world. Green tea is the least processed and thus contain rich antioxidant level, and believed to have most of the health benefits. Methods: We commenced to search bibliographic collection of peer reviewed research articles and review articles to meet the objective of this study. Results: From this study, we found that the tea beverage contains catechins are believed to have a wide range of health benefits which includes neuroprotective, anti-inflammatory, antiulcer, antiviral, antibacterial, and anti-parasitic effects. The four major catechin compounds of green tea are epigallocatechin (EGC), epicatechin (EC), epigallocatechin gallate (EGCG), and epicatechin gallate (ECG), of which EGCG is the major constituent and representing 50-80% of the total catechin content. And also contain xanthine derivatives such as caffeine, theophylline, and theobromine, and the glutamide derivative theanine. It also contains many nutritional components, such as vitamin E, vitamin C, fluoride, and potassium. We sum up the various green tea phytoconstituents, extraction methods, and its medicinal applications. Conclusion: In this review article, we have summarized the pharmacological importance of green tea catechin which includes antioxidant potential, anti-inflammatory, antimicrobial, anticancer, antidiabetic and cosmetic application.
