5. ROLE OF RGS14 PROTEIN IN VISUALMEMORYAND THE REGULATION OF SYNAPTIC PLASTICITY IN PERIRHINAL CORTEX

Though the concept of participation of perirhinal cortex and frontal cortex in the processing of object memory has long been appreciated, but recently our laboratory extended this to area V2 of visual cortex. We found that activation of area V2 neurons by overexpression of RGS14 protein led to an enhancement of object recognition memory. The memory enhancement was of such extent that it converted the short term memory of 45 minutes into long lasting long-term memory that could be traced even after many months. Here, we have tested the memory enhancer effect of RGS14 in perirhinal cortex (PRh), an area known to be involved in object memory processing, and further explored the relationship of behavioral memory performance with synaptic plasticity within this area. Stimulation of PRh with RGS14 protein produced an equally robust increase in object memory as was observed in area V2. In addition, we found that RGS14-mediated activation of PRh, (i) blocked the depotentiation induced by 1Hz stimulation during 10min; (ii) blocked the LTP induced by 20Hz stimulation while showed no effect at 100Hz stimulation; and (iii) reduced the LTD induced by the application of 20 µM of carbachol, a cholinergic agonist, during 10min, however no effect was observed at a higher concentration (50 µM). Furthermore, we also observed that phosphorylated isoforms of AMPA receptor 1 (iGluR1) were significantly reduced. Thus, our results indicate that iGluR1 reflects the level of synaptic plasticity (LTP and LTD) observed in RGS-animals but lack this correlation in behavioral outcome. - Abstract
A sunflower (Helianthus annuus) seed husk agar medium has been developed and evaluated for differentiation of Candida dubliniensisfrom Candida albicanson the basis of colony morphology and chlamydospore production. All C. dubliniensisisolates (n=40) produced rough colonies with hyphal fringes and abundant chlamydospores whereas 101 of 105 (96.2%) C. albicansisolates produced smooth colonies with no evidence of chlamydospore production. Since this medium is free from oil droplets, chlamydospores can be examined with greater clarity by Dalmau plate technique. This medium provides a simple and cost-effective tool for the presumptive differentiation of C. dubliniensisfrom C. albicansand is particularly suited for clinical microbiology laboratories where biochemical or molecular methods for the differentiation of these two species are not available.

11. TRICHOSPORON ASAHIIAS AN EMERGING ETIOLOGIC AGENT OF DISSEMINATED TRICHOSPORONOSIS: A CASE REPORT AND AN UPDATE

- Abstract
Purpose: To report a fatal case of disseminated trichosporonosis caused by Trichosporon asahiiin a patient with acute myeloblastic leukemia (AML) and to present an update on systemic trichosporonosis with special reference to India. Methods: The diagnosis was based on repeated demonstration of budding yeast cells and arthroconidia by direct microscopic examination of sputum and by isolation of T. asahiiin culture of sputum and blood. The update is largely based upon literature search in Medline and Review of Medical and Veterinary Mycology. Results: A 41-year-old male presented with acute myeloblastic leukemia, cough and fever. He had received cytotoxic drug therapy, broad spectrum antibiotics and was neutropenic. Trichosporon asahiiwas repeatedly demonstrated in his sputum by direct microscopy and culture, and also isolated from blood. It was identified by appropriate morphological and physiological characteristics viz., arthroconidium formation, diazonium blue B reaction, urease activity and assimilation of carbon and nitrogen compounds. The identification was confirmed by PCR amplification and direct DNA sequencing of internally transcribed spacer (ITS) 1 and ITS2 of rDNA. Conclusion: With greater awareness of etiologic significance of T.asahii, trichosporonosis is likely to be recognised more frequently than apparent from the available published reports.

12. Poor perinatal care practices in urban slums: Possible role of social mobilization networks

Background: Making perinatal care accessible to women in marginalized periurban areas poses a public health problem. Many women do not utilize institutional care in spite of physical accessibility. Home-based care by traditional birth attendants (TBA) is hazardous. Inappropriate early neonatal feeding practices are common. Many barriers to perinatal care can be overcome by social mobilization and capacity building at the community level. Objectives: To determine the existing perinatal practices in an urban slum and to identify barriers to utilization of health services by mothers. Study Design: This is a cross-sectional descriptive study. Setting and Participants: The high-risk periurban areas of Nabi Nagar, Aligarh has a population of 40,000 living in 5,480 households. Mothers delivering babies in September 2007 were identified from records of social mobilization workers (Community Mobilization Coordinators or CMCs) already working in an NGO in the area. A total of 92 mothers were interviewed at home. Current perinatal practices and reasons for utilizing or not utilizing health services were the topics of inquiry. Statistical Analysis: Data was tabulated and analyzed using SPSS 12. Results: Analyses revealed that 80.4% of mothers had received antenatal care. However, this did not translate into safe delivery practices as more than 60% of the women had home deliveries conducted by traditional untrained or trained birth attendants. Reasons for preferring home deliveries were mostly tradition (41.9%) or related to economics (30.7%). A total of 56% of the deliveries were conducted in the squatting position and in 25% of the cases, the umbilical cord was cut using the edge of a broken cup. Although breast-feeding was universal, inappropriate early neonatal feeding practices were common. Prelacteal feeds were given to nearly 50% of the babies and feeding was delayed beyond 24 hours in 8% of the cases. Several mothers had breastfeeding problems. Conclusion: Barriers to utilization of available services leads to hazardous perinatal practices in urban slums.

Aims and Objectives: To determine the prevalence of common ocular morbidities (cataract, refractive errors, glaucoma, and corneal opacities) and their demographic and sociocultural correlates. Settings and Design: The present cross-sectional study was conducted in the field practice areas of the Department of Community Medicine, JNMC, AMU, Aligarh, for a period of one year, from September 2005 to August 2006. Materials and Methods: Systematic random sampling was done to select the required sample size. All adults aged 20 years and above in the selected households were interviewed and screened using a 6/9 illiterate ′E′ chart. Those who could not read the ′E′ chart were referred to the respective health training center for a complete eye examination by an ophthalmologist. Statistical Analysis: Chi- square test. Results: The prevalence of visual impairment, low vision, and blindness, based on presenting visual acuity was 13.0, 7.8, and 5.3%, respectively. The prevalence of cataract was 21.7%. Bilateral cataract was present in 16.9% of the population. Cataract was significantly associated with age, education, and fuel use. The prevalence of myopia, hypermetropia, and astigmatism was 11.5, 9.8, and 3.7%, respectively. Glaucoma was diagnosed in six patients, giving a prevalence rate of 0.9%. All the six patients of glaucoma were aged above 40 years. The prevalence of corneal opacity was 4.2%. Conclusion: There is a high prevalence of treatable or preventable morbidities such as cataract, refractive errors, and corneal opacity.

Research question: Is there an association between common socio â€" cultural variables and congenital malformation? Objectives: To determine the rate of congenital malformation at birth in hospital deliveries and its association with socio- cultural factors. Study design : Cross- sectional. Setting: J.N. Medical College Hospital and Mohanlal Gautam Rajkiya Hahila Chikitsalaya (District hospital), Aligarh. Participants: All newborn babies (including still births) delivered in the two hospitals and their mothers. Study variables: Social class, religion, consanguinity of marriage, age of mother, parity, urban rural status, history of viral illness, drug intake and tobacco use during pregnancy. Outcome variables: Congenital malformations at birth. Statistical analysis: Chi- square test. Results: The overall prevalence of congenital malformations was 2.8%, being 1.6% in live births and 15.6% in stillbirths. Social class, consanguinity of marriage, parity, urban or rural status and history of viral illness during pregnancy were significantly associated with the date of congenital malformation. Conclusion: Certain socio- cultural factors are associated with congenital malformations and can be used in screening during ante- natal period.

In the present study, a door follow up study of mothers and infants was done one year after the mothers had been registered in the Ante-Natal Clinic of the Rural Health Training Center. Home delivery by locally available dai was present in a almost all cases. The choice of dai depended on the family tradition rather than training of the dai. Where mothers had not attended Ante-Natal Clinic, the reason given by a majority (55.8%) was ignorance of services available. The health status and immunization status was significantly better in infants where mothers had regularly attended Ante-Natal clinics.

- Abstract
Ethnopharmacological Relevance: The safety assessment of herbal products is critical for their appropriate pharmacological applications. Garcinia cowa Roxb., commonly known as Cha-muang in Thai, has ethnopharmacological relevance for inflammation, infectious diseases, and diabetes. The leaf extracts of G. cowa have been extensively reported for their anticancer, anti-inflammatory, antimicrobial, and antioxidant effects. Notably, chamuangone is their major active constituent that contributes to various pharmacological properties., Aim of the Study: The current study aims to establish a standardized chamuangone enriched extract (CEE) and assess its acute and sub-acute toxicities in animal models., Methodology: CEE was established from G. cowa leaves using a microwave-assisted extraction (MAE), followed by fractionation and enrichment through silica gel vacuum and column chromatography. The concentration of chamuangone in the extract was quantified using a validated quantitative high-performance liquid chromatography (HPLC) method. The safety profiles of CEE were thoroughly evaluated in rodents according to the Organization for Economic Cooperation and Development (OECD) 425 and 407 guidelines. The effects on oxidative stress markers such as superoxide dismutase (SOD), reduced glutathione (GSH), catalase (CAT), and malondialdehyde (MDA) levels were also evaluated in various organs., Results: Based on the quantitative HPLC analysis, the CEE contained 73.0 ± 2.0% w/w of chamuangone. In the acute toxicity study, following up and down procedure the female rats were dosed with CEE at 1750 and 550 mg/kg body weight (b.w.), with CEE 1750 mg/kg b.w. was toxic, causing mortality, while CEE 550 mg/kg b.w. was deemed safe. An LD
50 value was calculated according to the standard protocols, resulting in 970 mg/kg b.w. In histopathological examination, 550 mg/kg b.w. of CEE was safe in all the selected organs, while the 1750 mg/kg b.w. CEE treated rats exhibited toxic effects in histological tissues sections in the form of necrosis in the brain, cardiac muscle hypertrophy, liver inflammation, mild untoward effect in the spleen, fibrosis in the lungs, pancreatitis, pyelonephritis, and ovarian cyst. Administration of CEE at doses of 550 mg/kg b.w. (single dose) in the acute and 100 mg/kg b.w. (regularly 28-days) in the sub-acute toxicity studies significantly (p < 0.05) decreased levels of uric acid, triglycerides, and cholesterol. Importantly, the CEE (550 and 100 mg/kg b.w.) also significantly increased the levels of antioxidant enzymes (SOD, GSH, and CAT) and decreased MDA levels. Normal histopathology was observed in the sub-acute toxicity study in all treated groups., Conclusion: This study successfully concludes that CEE at a dose of 100 mg/kg b.w. is safe for therapeutic application or use as a chemopreventive functional food utilizing green extraction methods. Medical image machines serve as a valuable tool to monitor and diagnose a variety of diseases. However, manual and centralized interpretation are both error-prone and time-consuming due to malicious attacks. Numerous diagnostic algorithms have been developed to improve precision and prevent poisoning attacks by integrating symptoms, test methods, and imaging data. But in today's digital technology world, it is necessary to have a global cloud-based diagnostic artificial intelligence model that is efficient in diagnosis and preventing poisoning attacks and might be used for multiple purposes. We propose the Healthcare Federated Ensemble Internet of Learning Cloud Doctor System (FDEIoL) model, which integrates different Internet of Things (IoT) devices to provide precise and accurate interpretation without poisoning attack problems, thereby facilitating IoT-enabled remote patient monitoring for smart healthcare systems. Furthermore, the FDEIoL system model uses a federated ensemble learning strategy to provide an automatic, up-to-date global prediction model based on input local models from the medical specialist. This assures biomedical security by safeguarding patient data and preserving the integrity of diagnostic processes. The FDEIoL system model utilizes local model feature selection to discriminate between malicious and non-malicious local models, and ensemble strategies use positive and negative samples to optimize the performance of the test dataset, enhancing its capability for remote patient monitoring. The FDEIoL system model achieved an exceptional accuracy rate of 99.24% on the Chest X-ray dataset and 99.0% on the MRI dataset of brain tumors compared to centralized models, demonstrating its ability for precision diagnosis in IoT-enabled healthcare systems., (© 2024. 21. GroupFormer for hyperspectral image classification through group attention.

- Abstract
Hyperspectral image (HSI) data has a wide range of valuable spectral information for numerous tasks. HSI data encounters challenges such as small training samples, scarcity, and redundant information. Researchers have introduced various research works to address these challenges. Convolution Neural Network (CNN) has gained significant success in the field of HSI classification. CNN's primary focus is to extract low-level features from HSI data, and it has a limited ability to detect long-range dependencies due to the confined filter size. In contrast, vision transformers exhibit great success in the HSI classification field due to the use of attention mechanisms to learn the long-range dependencies. As mentioned earlier, the primary issue with these models is that they require sufficient labeled training data. To address this challenge, we proposed a spectral-spatial feature extractor group attention transformer that consists of a multiscale feature extractor to extract low-level or shallow features. For high-level semantic feature extraction, we proposed a group attention mechanism. Our proposed model is evaluated using four publicly available HSI datasets, which are Indian Pines, Pavia University, Salinas, and the KSC dataset. Our proposed approach achieved the best classification results in terms of overall accuracy (OA), average accuracy (AA), and Kappa coefficient. - Abstract
Superficial skin injuries especially burn injuries and unhealed diabetic foot open wounds remain troubling for public health. The healing process is often interrupted by the invasion of resistant pathogens that results in the failure of conventional procedures outside the clinical settings. Herein, we designed nanofibers dressing with intrinsic antibacterial potential of poly(vinyl-pyrrolidone)-iodine/ poly (vinyl)-alcohol by electrospinning with chitosan encapsulating ceftriaxone (CPC/NFs). The optimized electrospun CPC/NFs exhibited smooth surface morphology with average diameter of 165 ± 7.1 nm, drug entrapment and loading efficiencies of 76.97 ± 4.7 % and 8.32 ± 1.73 %, respectively. The results displayed smooth and uniformed fibers with adequate thermal stability and ensured chemical doping. The enhanced in vitro antibacterial efficacy of CPC/NFs against resistant E. coli isolates and biosafety studies encourage the use of designed nanofibers dressing for burn injuries and diabetic foot injuries. In vivo studies proved the healing power of dressing for burn wounds model and diabetic infected wounds model. Immunofluorescence investigation of the wound tissue also suggested promising healing ability of CPC/NFs. 23. A comparison of the accuracy of Tzanakis and Alvarado Score in the diagnosis of acute appendicitis: A systematic review and meta-analysis.

- Abstract
Background and Objectives: Acute appendicitis is one of the most commonly encountered surgical emergencies on a global level. Due to the requirement of an immediate clinical diagnosis and the presence of limited resources, clinicians and diagnosticians refer to scoring systems to diagnose this condition, among which Alvarado and Tzanakis scoring systems are widely used. This meta-analysis aims to compare the diagnostic accuracy of these two systems., Methods: We searched PubMed, Google Scholar, and SCOPUS databases. All studies that reported diagnostic parameters of Alvarado and Tzanakis scores in patients with suspected acute appendicitis were selected. Diagnostic values such as sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and diagnostic accuracy were extracted from the selected studies and statistical analysis was performed with Meta Disc 1.4 software. Quality assessment of the selected studies was performed using the QUADAS-2 and QUADAS-C tools. Fourteen studies were included in our meta-analysis which enrolled 2235 patients., Results: The overall sensitivity of the Tzanakis score was calculated as 0.86 (95% CI; 0.84-00.87) while the specificity was 0.73 (95% CI; 0.69-0.78). In addition, the area under the curve (AUC) was 0.9261 (SE; 0.0169) and the diagnostic Odds Ratio (OR) was 22.52 (95% CI; 9.47-53.56). The pooled sensitivity of Alvarado score was 0.67 (95% CI; 0.65-0.69) and the specificity was 0.74 (95% CI; 0.69-0.79). Introduction and Aims: Adhesion to buccal epithelial cells (BEC) and denture acrylic surfaces (DAS), germ tube (GT) formation, cell surface hydrophobicity (CSH), and haemolysin production are attributes associated with pathogenicity of Candida. Candida albicans and Candida dubliniensis are allied in causing oral candidosis. Lysozyme and lactoferrin exert antimicrobial activity on a range of oral microorganisms, including Candida. There is no information on the impact of brief exposure to lysozyme and lactoferrin on adhesion-related attributes and haemolysin production of aforementioned oral Candida isolates. Thus, we investigated the impact of lysozyme and lactoferrin on adhesion to BEC and DAS, GT formation, CSH, and haemolysin production of these isolates., Methods: After exposure to lysozyme and lactoferrin for 1 hour, susceptibility to lysozyme and lactoferrin of 20 isolates each of C albicans and C dubliniensis isolates was determined following a 48-hour period of incubation. Candida cell suspensions, obtained from colony-forming units after this period, were assessed for adhesion to BEC and DAS, GT formation, CSH, and haemolysin production using in vitro assays., Results: Exposure to lysozyme and lactoferrin significantly suppressed the ability of C albicans and C dubliniensis isolates to adhere to BEC and DAS, GT formation, CSH, and haemolysin production (P < 0.01 for all virulent attributes tested)., Conclusions: These data provide a tantalising glimpse into the possibility that exposure to either lysozyme or lactoferrin, even for a brief period, would induce a sustainable antifungal effect by suppressing adhesion-related attributes and haemolysin production of these oral Candida species in vitro. Resistance to conventional antifungal agents has been reported in clinical isolates of Candida. The presence of such resistance indicates the need for possible alternative therapies to facilitate the management of oral candidosis. - Abstract
Interruption of wound healing by multi-drug resistant-bacterial infection is a harmful issue for the worldwide health care system, and conventional treatment approaches may not resolve this issue due to antimicrobial resistance. So, there is an unmet need to develop scaffolds with intrinsic wound healing properties to combat bacterial-infected wounds. Inspired by the α-lactalbumin's (Lalb's) ability to promote collagen synthesis, we herein electrospun Lalb with cephalexin (CPL) and epigallocatechin (EP) to produce nanofibers (CE-Lalb NFs) to solve this issue. The CE-Lalb NFs were prepared using the electrospinning technique and subjected to physicochemical characterizations, in vitro, and in vivo assessments. The CE-Lalb NFs promoted fibroblast migration, proliferation, and collagen synthesis, while CPL/EP annihilated MRSA and E. coli infections. Physicochemical characterizations proved the successful fabrication and doping of CE-Lalb NFs. Antimicrobial assays and fractional inhibitory concentration index (FICI) declared synergistic antibacterial activity of CE-Lalb NFs against MRSA and E. coli. The in vivo and immunohistochemical data evidenced its exceptional potential for wound healing, promoting growth factor, collagen synthesis, and reduced scar formation. The presence of mature collagen, fewer inflammatory cytokines, increased expression of blood vessels, and low expression of IL-6 at the wound site support in vitro and in vivo results. 26. The development of anti-freezing and anti-evaporating conductive organohydrogel for flexible strain-sensing electronic devices.

- Abstract
The hydrogels got hot attention from the researchers due to their excellent flexibility, mechanical properties, conductivity, and response to external stimuli, however, their applications are hampered due to a lack of water retention properties and anti-freezing ability. In this study, the organohydrogel was designed from the two solvents system (DMSO and water) in which acrylamide (Amm) and acrylic acid (AA) work as monomers and Arabic gum (AG) works as biopolymers. The functional groups, surface morphology, mechanical properties, rheological properties, anti-freezing properties, and strain-sensing properties of the fabricated organohydrogel were studied. The DMGA6 organohydrogel showed good mechanical properties with fracture stress of 197 kPa, fracture strain of 593%, cyclic stability to five cycles at 400% strain without relaxation time, excellent rheological performance (elastic behavior), anti-freezing properties (-54.6 °C), the response time of 65 ms with a gauge factor of 8.86 at 500% strain. The organohydrogel can detect different human motions like bending of the finger, elbow, neck, and wrist at normal and harsh temperatures. 27. Comparative Microstructural Evaluation of Wood in Three Dominant Ziziphus Species of Desert Ecosystem (Cholistan), Pakistan.

The present microstructural evaluation was carried out on the woods of three ethnobotanically important local fruit trees, namely, Ziziphus mauritiana Lam., Z. spina-christi (L.) Willd., and Z. nummularia (Burm.f.) Wight and Arn., of family Rhamnaceae from Cholistan Desert of Pakistan. Wood samples were sectioned with sliding sledge microtome to make permanent slides for observing different anatomical parameters under the light microscope. All selected species were observed to have diffuse-porous wood with indistinct growth rings. The vessels were rounded in outline in all the species studied and found mostly solitary or in radial multiples of 2 in Ziziphus mauritiana and Z. nummularia , while in radial multiples of 2 to 5 in Z. spina-christi . The intervessel pits were scalariform to opposite. The rays were uniseriate in Ziziphus mauritiana , while mostly were biseriate in Ziziphus spina-christi . Simple perforation plates and diffuse, confluent, and vasicentric types of axial parenchyma were present in all the selected species. The fibers were thin-walled and nonseptate. One-way ANOVA followed by the Tukey test was conducted to observe different anatomical variations within selected species. Principal component analysis revealed correlations among studied anatomical parameters. The number of rays per mm was comparatively larger in Ziziphus nummularia, showing its greater susceptibility to wood-deteriorating agents than in other selected species. 28. Promotion of structural plasticity in area V2 of visual cortex prevents against object recognition memory deficits in aging and Alzheimer's disease rodents.

JOURNAL/nrgr/04.03/01300535-202408000-00038/figure1/v/2023-12-16T180322Z/r/image-tiff Memory deficit, which is often associated with aging and many psychiatric, neurological, and neurodegenerative diseases, has been a challenging issue for treatment. Up till now, all potential drug candidates have failed to produce satisfactory effects. Therefore, in the search for a solution, we found that a treatment with the gene corresponding to the RGS14414 protein in visual area V2, a brain area connected with brain circuits of the ventral stream and the medial temporal lobe, which is crucial for object recognition memory (ORM), can induce enhancement of ORM. In this study, we demonstrated that the same treatment with RGS14414 in visual area V2, which is relatively unaffected in neurodegenerative diseases such as Alzheimer's disease, produced long-lasting enhancement of ORM in young animals and prevent ORM deficits in rodent models of aging and Alzheimer's disease. Furthermore, we found that the prevention of memory deficits was mediated through the upregulation of neuronal arborization and spine density, as well as an increase in brain-derived neurotrophic factor (BDNF). A knockdown of BDNF gene in RGS14414-treated aging rats and Alzheimer's disease model mice caused complete loss in the upregulation of neuronal structural plasticity and in the prevention of ORM deficits. These findings suggest that BDNF-mediated neuronal structural plasticity in area V2 is crucial in the prevention of memory deficits in RGS14414-treated rodent models of aging and Alzheimer's disease. - Abstract
Background: Dentin hypersensitivity (DH) is one of the most challenging and persistent dental complaints characterized by transient, intense pain triggered by various stimuli. It affects a significant portion of the global population, predominantly those aged 20-40. This study aims to evaluate the desensitizing efficacy of seventh-generation dentin bonding agents (Single Bond Universal by 3 M ESPE and Xeno-V + by Dentsply) against a control group using Bifluorid 12 by Voco in mitigating DH within a month of the follow-up period., Methods: This was a single-center, parallel-group, double-blind, controlled randomized clinical trial conducted at Dow University of Health Sciences, Karachi, Pakistan. A total of 105 patients with DH were allocated into three groups for this study. The patients were divided into three groups (Single Bond Universal by 3 M ESPE and Xeno-V + by Dentsply) and the control group containing fluoride varnish (Bifluorid 12 by Voco). Discomfort Interval Scale scores and Schiff Cold Air Sensitivity Scale scores were recorded at baseline, immediately after the intervention, after 01 weeks, and after 01 month., Results: All the materials demonstrated a statistically significant reduction in discomfort and sensitivity (DIS scores p-value 0.01) immediately after 01 week and over a period of 01 month after treatment compared with the baseline scores before application, with no single material proving superior over the one-month observation period. The study also provided insights into dental hygiene practices, with a significant majority using a toothbrush and sensitivity patterns, with cold stimuli being the most common cause of sensitivity., Conclusion: The study demonstrates that Single Bond Universal, Xeno V+, and Bifluorid 12 are equally effective in reducing dentin hypersensitivity, with no distinct superiority observed over a one-month period. 30. Stimuli-sensitive biomimetic nanoparticles for the inhibition of breast cancer recurrence and pulmonary metastasis.

Biomimetic nanoparticles represent a promising avenue for mitigating rapid clearance by the reticuloendothelial system (RES); however, current challenges include insufficient tumour targeting, suboptimal adhesion, and inadequate localized drug release within tumour regions. These shortcomings contribute to persistent contests, such as recurrence and pulmonary metastasis, even with advanced breast cancer therapies. Stimuli-sensitive drug release can furbish the membrane coated nanoparticles for their efficiency against the stated problems. To enhance the efficacy of biomimetic nanoparticles in addressing these issues, we proposed a versatile, stimuli-responsive drug delivery system by encapsulating doxorubicin (Dox) and perfluorohexane (PFH) within poly (lactic- co -glycolic acid) (PLGA) nanoparticles, subsequently coated with macrophage-derived cell membranes. Within this framework, PFH serves as the mediator for ultrasonic (US)-irradiation-triggered drug release specifically within tumour microenvironment, while the macrophage-derived cell membrane coating enhances cell adhesion, enables immune evasion, and natural tumour-homing ability. The characterization assays and in vitro evaluations yielded encouraging results, indicating enhanced targeting and release efficiencies. In vivo studies demonstrated marked inhibitory effects on both breast cancer recurrence and pulmonary metastasis. The resulting data indicate that these engineered nanoparticles have notable potential for targeted delivery and controlled release upon US irradiation, thereby offering significant therapeutic efficacy against primary breast cancer, pulmonary metastasis, and recurrent malignancies. 31. Nitrogen-Doped Graphene-Supported Nickel Nanoparticles Reveal Low Dehydrogenation Temperature and Long Cyclic Life of Magnesium Hydrides.

Magnesium hydride (MgH
2 ) is a promising hydrogen storage candidate due to its large capacity; however, high dehydrogenation temperature and slow kinetic rates are the main bottlenecks. Herein, we proposed a strategy for designing nitrogen-doped graphene-supported Ni nanoparticles (NPs) (Ni@NC) to tackle these problems. The results showed that the MgH2 + 15 wt % Ni@NC nanocomposite reduced the on-set dehydrogenation temperature to 195 °C, which was 175 °C lower than pristine MgH2 . In addition, MgH2 + 15 wt % Ni@NC achieved 1.7 and 6.5 wt % desorption capacities at 225 and 300 °C, respectively, while absorbing 5.5 wt % hydrogen at 100 °C. The MgH2 + 15 wt % Ni@NC nanocomposite showed high cyclic stability, achieving 98.0% capacity retention after 100 cycles at 270 °C with negligible loss in capacity. This remarkable hydrogen storage performance can be attributed to the homogeneous distribution of Ni NPs on N-doped graphene layers, in situ formed Mg2 NiH2 NPs, and multiphasic regions, promoting the nucleation and growth process during hydrogenation/dehydrogenation, which stabilized and improved the cyclic stability. Aims: Ventral pathway circuits are constituted by the interconnected brain areas that are distributed throughout the brain. These brain circuits are primarily involved in processing of object related information in brain. However, their role in object recognition memory (ORM) enhancement remains unknown. Here, we have studied on the implication of these circuits in ORM enhancement and in reversal of ORM deficit in aging., Methods: The brain areas interconnected to ventral pathway circuits in rat brain were activated by an expression of a protein called regulator of G-protein signaling 14 of 414 amino acids (RGS14
414 ). RGS14414 is an ORM enhancer and therefore used here as a gain-in-function tool. ORM test and immunohistochemistry, lesions, neuronal arborization, and knockdown studies were performed to uncover the novel function of ventral pathway circuits., Results: An activation of each of the brain areas interconnected to ventral pathway circuits individually induced enhancement in ORM; however, same treatment in brain areas not interconnected to ventral pathway circuits produced no effect. Further study in perirhinal cortex (PRh), area V2 of visual cortex and frontal cortex (FrC), which are brain areas that have been shown to be involved in ORM and are interconnected to ventral pathway circuits, revealed that ORM enhancement seen after the activation of any one of the three brain areas was unaffected by the lesions in other two brain areas either individually in each area or even concurrently in both areas. This ORM enhancement in all three brain areas was associated to increase in structural plasticity of pyramidal neurons where more than 2-fold higher dendritic spines were observed. Additionally, we found that an activation of either PRh, area V2, or FrC not only was adequate but also was sufficient for the reversal of ORM deficit in aging rats, and the blockade of RGS14414 activity led to loss in increase in dendritic spine density and failure in reversal of ORM deficit., Conclusions: These results suggest that brain areas interconnected to ventral pathway circuits facilitate ORM enhancement by an increase in synaptic connectivity between the local brain area circuits and the passing by ventral pathway circuits and an upregulation in activity of ventral pathway circuits. In addition, the finding of the reversal of ORM deficit through activation of an interconnected brain area might serve as a platform for developing not only therapy against memory deficits but also strategies for other brain diseases in which neuronal circuits are compromised., (© 2024 The Authors. Perirhinal cortex is a brain area that has been considered crucial for the object recognition memory (ORM). However, with the use of an ORM enhancer named RGS14
414 as gain-in-function tool, we show here that frontal association cortex and not the Perirhinal cortex is essential for the ORM of objects with complex features that consisted of detailed drawing on the object surface (complex ORM). An expression of RGS14414 , in rat brain frontal association cortex, induced the formation of long-term complex ORM, whereas the expression of the same memory enhancer in Perirhinal cortex failed to produce this effect. Instead, RGS14414 expression in Perirhinal cortex caused the formation of ORM of objects with simple features that consisted of the shape of object (simple ORM). Further, a selective elimination of frontal association cortex neurons by treatment with an immunotoxin Ox7-SAP completely abrogated the formation of complex ORM. - Abstract
Gastric ulcers are worrying, and their worsening conditions may result in bleeding in the internal lining of the stomach. The problem is annoying, and both patients and professionals are still not satisfied with the available treatment options. Hesperidin, a flavonoid molecule with potent anti-inflammatory and antioxidant effects, can work like witchcraft to repair gastric ulcers and preserve the stomach lining. Here, we employed a strategy that involved covering the surface of the nano-lipid carriers (NLCs) with sericin before encasing the hesperidin within (Se-He-NLC). Sericin, a biodegradable polymer increases the muco-adhesion with stomach lining and deployment of hesperidin in controlled manner. Se-He-NLCs were physico-chemically characterized for drug loading, encapsulation, particle size, morphology, drug release, chemical stability, and chemical bonding. The nanocarriers showed first order drug release in a controlled manner. Se-He-NLCs showed better in vitro permeation and ex vivo mucoadhesion, thereby by promoting the in vivo bioavailability. Se-He-NLCs also promoted the reduced glutathione (GSH) and glutathione-S-transferase (GST) levels by 2.24- and 1.61-folds, respectively in the stomach lining, and also the regulation of superoxide dismutase (SOD) and catalase (CAT) activities parallel to the control group. In addition, tissues lipid hydroperoxides (LOOH) and myeloperoxidase (MPO) activity were reduced significantly with Se-He-NLCs administration. Se-He-NLC therapy of stomach ulcers in vivo demonstrated better binding ratio and ulcer healing potential. 36. Relationship Between Cardiometabolic Index in Early Pregnancy and Hypertensive Disorder Complicating Pregnancy.

- Abstract
Background This study aimed to examine the cardiometabolic index during early pregnancy in individuals with hypertension-complicating pregnancy, especially preeclampsia. Additionally, this study sought to determine the relationship between cardiometabolic index and the incidence of varying degrees of preeclampsia. Methodology This study included 289 pregnant women diagnosed with preeclampsia who were registered and delivered at our hospital. These women were assigned to the preeclampsia group. Additionally, a group of 289 healthy pregnant women of identical gestational ages within the same time frame was included for comparison. Clinical data on pregnancy, including body mass index (BMI), blood pressure, waistline, triglyceride levels, and cardiometabolic index, were compared between the two groups. An analysis was conducted to examine the association between early pregnancy cardiometabolic index and the occurrence of preeclampsia. Results There was a significant association between the quartile of cardiometabolic index and the proportion of preeclampsia patients (p < 0.001). Furthermore, after controlling for age and BMI, the risk of preeclampsia remained significantly elevated and was associated with the cardiometabolic index. 37. Design and optimization of a transparent and flexible MIMO antenna for compact IoT and 5G applications.

- Abstract
This work presents an optically transparent and flexible MIMO antenna that features two square patch elements placed in close proximity, aiming to meet the demands of compactness, flexibility, optical transparency, and visual appeal for IoT applications and future 5G wireless communication. The design includes a simple offset fed configuration to achieve the required isolation and impedance matching. It simplifies the process of creating closely spaced transparent MIMO antenna configurations. By optimizing and analyzing this structure, the antenna achieves better isolation and diversity gain performance, even when the patch elements are positioned very close to each other. To achieve optical transparency and flexibility, the antenna uses thin polyethylene terephthalate (PET) material as a substrate, which is a thermoplastic polymer resin from the polyester family. The wired metal mesh parameters for conducting parts of the MIMO antenna and offset position of the feed are carefully optimized to achieve required optical transparency, isolation, impedance matching and radiation performance without any complex decoupling or impedance matching network., (© 2023. - Abstract
Cutaneous extranodal non-Hodgkin lymphoma is a rare cancer and chemotherapeutic agents like etoposide, vincristine and cyclophosphamide are the drug of choice, which rarely cause hyperpigmentation in skin and nails. However, herein we present a case of hyperpigmentation that was seen in tongue and even in teeth. The hyperpigmentation of the tongue and teeth occurred shortly after the initiation of chemotherapy. Hyperpigmentation was self-limiting and rectified in a week without need of any pharmacological, surgical, or lifestyle interventions.

- Abstract
Loratadine (LRD) belongs to second-generation tricyclic H1 antihistamine class, known for its non-sedating properties in allergic reactions. H1 antihistamines avoid and block the responses to allergens or histamine in nose and conjunctivae, thereby abolishing itching, congestion and sneezing. LRD is a Biopharmaceutical Class System (BCS) class II drug with dissolution or solubility limited absorption which limited the oral bioavailability and therapeutic efficacy of LRD. To improve the oral bioavailability of LRD for allergic disease (urticaria) treatment, LRD solid dispersions (LRD-SDs) were integrating into oro-dispersible films (ODFs). LRD-SDs were prepared through hot-melt extrusion method (HME) using d-alpha-tocopherol polyethylene glycol 1000 succinate (TPGS-1000), and polyvinyl caprolactam-polyvinyl acetate-polyethylene glycol graft copolymer (SP). Subsequently, LRD-SDs were incorporated in ODFs by solvent casting method. The physicochemical and mechanical properties of LRD solid dispersions-loaded oro-dispersible films (LRD-SDs-ODFs), were evaluated. The in-vitro dissolution, ex-vivo permeation, oral bioavailability, and pharmacodynamics studies were conducted to evaluate LRD-SDs-ODFs efficiency. LRD-SDs-ODFs showed superior solubility and in-vitro dissolution results compared to that of pure LRD (p < 0.05). The solubility of the LRD-SD coded as LTS-4 was 190 times higher than the pure drug in aqueous media. The average hydrodynamic particle size (PS), polydispersity index (PDI), and zeta potential (ZP) of SD particles were 76 ± 2.1 nm, 0.20 ± 0.08 and - 19.16 ± 1.4 mV, respectively. Moreover, differential scanning calorimetry (DSC) and X-ray diffraction (XRD) results confirmed the amorphousness of LRD in LRD-SDs-ODFs. The permeability flux of LRD was 44.6 ± 3.1 μg/cm
2 /h from DPF-5 formulation. Likewise, in vivo oral bioavailability of DPF-5 in Sprague-Dawley rats was significantly increased (p < 0.05) compared to free LRD. Further, wheal area was reduced 20 % higher than LRD in 8 h (p < 0.05). Overall, LRD-SDs-ODFs considerably enhanced LRD solubility, dissolution rate, bioavailability, and antihistaminic efficacy. 40. Dual-sense slot-based CP MIMO antenna with polarization bandwidth reconfigurability.

- Abstract
In this letter, a compact, planar circularly polarized (CP) sub-GHz slot-based multiple-input-multiple-output (MIMO) antenna with dual sense CP along with polarization bandwidth reconfigurability is presented. The pentagonal reactively loaded slot is fed by two folded tapered feedlines to achieve CP. The antenna offers left-hand-circular polarization (RHCP) with the as well as right hand circular polarization (LHCP). The antenna exhibit linearly polarization (LP) by exciting two ports simultaneously. Moreover, the antenna CP resonance can be reconfigured by varying the capacitance of the varactor diode. The antenna has a wide -10 dB operating frequency band from 578-929 MHz. while the axial ratio (AR) bandwidth ranges from 490-810 MHz. Moreover, the two elements MIMO are optimized and placed on compact dimensions 100 × 100 × 0.76 mm
3 to realize pattern diversity. The antenna's key characteristics are compact size, wide-band sub-GHz operation, dual sense CP, polarization bandwidth reconfigurability and good MIMO performance. Singlet molecular oxygen (
1 O2 ) has been reported in wide arrays of applications ranging from optoelectronic to photooxygenation reactions and therapy in biomedical proposals. It is also considered a major determinant of photodynamic therapy (PDT) efficacy. Since the direct excitation from the triplet ground state (3 O2 ) of oxygen to the singlet excited state1 O2 is spin forbidden; therefore, a rational design and development of heterogeneous sensitizers is remarkably important for the efficient production of1 O2 . For this purpose, quantum dots (QDs) have emerged as versatile candidates either by acting individually as sensitizers for1 O2 generation or by working in conjunction with other inorganic materials or organic sensitizers by providing them a vast platform. Thus, conjoining the photophysical properties of QDs with other materials, e.g., coupling/combining with other inorganic materials, doping with the transition metal ions or lanthanide ions, and conjugation with a molecular sensitizer provide the opportunity to achieve high-efficiency quantum yields of1 O2 which is not possible with either component separately. Objectives: To explore the awarenesslevel of literate mothersregarding newborn screening programmes, and to evaluate the associated factors., Methods: The descriptive, cross-sectional study was conducted at the Section of Chemical Pathology, Department of Pathology andLaboratoryMedicine,AgaKhanUniversity,Karachi,fromJanuary toSeptember 2021, andcomprisedmothers aged 18 years or more. Data was collected using a structured questionnaire about newborn screening, and the subjects were compared in terms of age, residential background, education and parity. Data was analysed using SPSS 23., Results: Of the 1016 responses, 896(88.2%) were analysed. The mean age of the sample was 37.7±10.87 years. There were 470(52.4%) mothers aged 31-45 years, 859(95.87%) were from urban areas, 751(84%) had a graduate degree, 652(72.7%) weremultiparous andhad824(91.9%)hadhealthy children.Overall, 386 (43%)mothershadawarenessofnewbornscreening programmes. The main factors associated with awareness were age, education, primiparity, having healthy children, and province ofresidencebeing Sindh andPunjab(p<0.05),while the urban-ruraldividedwas not a significantfactor(p=0.737). Cost of healthcare 417(46.5%) and lack of awareness among physicians 356(39.7%) were identified asthe main challenges in establishing newborn screening servicesin the country., Conclusions: The awareness among mothers about new born screening programmes was generally low among the subjects studied.

Water pollution due to organic waste and various microorganisms cause severe health problems. Numbers of techniques are used to eliminate organic waste and microorganisms from water because water pollution is a substantial issue in the current era. In the present study, sustainable and effective CuO/SnO
2 @g-C3 N4 nanocomposites were prepared via green and chemical approach. The photo degradation of ciprofloxacin (CIP) and methylene blue (MB) by the green synthesized nanocomposite were tested. Visible and dark conditions both were used to conduct this test. The results showed that the nanocomposite is much more effective in light than in dark conditions. The synthesized nanocomposite was also tested both in light and dark against highly drug resistant microorganisms' Bacillus subtilis (B.subtilis) and Escherichia coli (E.coli). As a result, the antibacterial evaluation revealed substantial antibacterial activity in the presence of light, with a zone of inhibition covering an area of 19 (±0.5) mm and 20 (±0.1) mm, respectively, against gram negative and gram positive bacteria such as E. coli and B. subtilis. 44. Eco-Friendly Sustainable Nanocarriers to Treat Oxidative Stresses and Skin Aging-Related Ailments, Valorization of a By-Product.

- Abstract
The peel from Citrus-sinensis L. is a medicinally significant food waste, and its extract (O-Ext) could be significant against oxidative stresses and skin aging, However, the penetration barriers, instability in formulation, undefined toxicities, and enzymatic activities make the O-Ext difficult to formulate and commercialize. The goal of this study was to evaluate O-Ext against oxidative stress, prepare O-Ext-loaded nano-lipid carriers (O-NLCs), and load them into topical O/W-emulsion (O-NLC-E) to improve O-Ext permeation and its in vivo antiaging effects. TPC, TFC, DPPH activity, and mineral/metal contents of O-Ext were determined via atomic-absorption spectroscopy. For bioactive compounds profiling, GC-MS analysis was carried out. O-NLCs were prepared and tested for physicochemical attributes, while HaCaT and fibroblast cells were used to study permeation and cytotoxicity. The kinetic characteristics of ex vivo permeation through rat skin were established, following the Higuchi model. Following written consent, safety investigations were conducted on human volunteers for three months, where optimized O-NLC-E and B-NLC-E were regularly applied on cheeks. Non-invasive procedures were used to assess the volunteer's skin erythema, TEWL, sebum level, melanin, hydration, pH, elasticity, and pore sizes after specified intervals. The results demonstrated that applying O-NLC-E formulation to the skin of volunteers directed significant antiaging benefits. The study offers nanotechnology-based sustainability approach against skin ageing.

- Abstract
A miniaturized folded dipole patch antenna (FDPA) design for biomedical applications operating at sub 1 GHz (434 MHz) band is presented. Antenna is fabricated on FR-4 substrate material having dimensions of 16.40 mm [Formula: see text] 8.60 mm [Formula: see text] 1.52 mm (0.023[Formula: see text] [Formula: see text] 0.012[Formula: see text] [Formula: see text] 0.002[Formula: see text]). Indirect feed coupling is applied through two parallel strips at bottom layer of the substrate. The antenna size is reduced by 83% through lumped inductor placed at the center path of the radiating FDPA, suitable for biomedical (implantable) applications and hyperthermia. Moreover, Impedance matching is achieved without using any Balun transformer or any other complex matching network. The proposed antenna provides an impedance bandwidth of 6 MHz (431-437 MHz) below - 10 dB and a gain of - 31 dB at 434 MHz. The designed antenna is also placed on a human body model to evaluate its performance for hyperthermia through Specific Absorption Rate (SAR), Effective Field Size (EFS), and penetration depth (PD)., (© 2023. 46. A High Performance All-Textile Wearable Antenna for Wristband Application.

- Abstract
A compact, conformal, all-textile wearable antenna is proposed in this paper for the 2.45 GHz ISM (Industrial, Scientific and Medical) band. The integrated design consists of a monopole radiator backed by a 2 × 1 Electromagnetic Band Gap (EBG) array, resulting in a small form factor suitable for wristband applications. An EBG unit cell is optimized to work in the desired operating band, the results of which are further explored to achieve bandwidth maximization via floating EBG ground. A monopole radiator is made to work in association with the EBG layer to produce the resonance in the ISM band with plausible radiation characteristics. The fabricated design is tested for free space performance analysis and subjected to human body loading. The proposed antenna design achieves bandwidth of 2.39 GHz to 2.54 GHz with a compact footprint of 35.4 × 82.4 mm
2 . The experimental investigations reveal that the reported design adequately retains its performance while operating in close proximity to human beings. The presented Specific Absorption Rate (SAR) analysis reveals 0.297 W/kg calculated at 0.5 W input power, which certifies that the proposed antenna is safe for use in wearable devices.- Published

Our brain is continuously challenged by daily experiences. Thus, how to avoid systematic erasing of previously encoded memories? While it has been proposed that a dual-learning system with 'slow' learning in the cortex and 'fast' learning in the hippocampus could protect previous knowledge from interference, this has never been observed in the living organism. Here, we report that increasing plasticity via the viral-induced overexpression of RGS14414 in the prelimbic cortex leads to better one-trial memory, but that this comes at the price of increased interference in semantic-like memory. Indeed, electrophysiological recordings showed that this manipulation also resulted in shorter NonREM-sleep bouts, smaller delta-waves and decreased neuronal firing rates. In contrast, hippocampal-cortical interactions in form of theta coherence during wake and REM-sleep as well as oscillatory coupling during NonREM-sleep were enhanced. 48. Monoclonal antibodies for the treatment of Graves' ophthalmopathy: A systematic review and meta-analysis.

Purpose: The traditional standard of care for Graves' ophthalmopathy (GO) is glucocorticoid therapy, which is associated with many long-term side effects. The aim of this systematic review and meta-analysis was to compare the traditional therapy to novel monoclonal antibodies (e.g. rituximab [RTX], teprotumumab, and tocilizumab [TCZ])., Methods: We searched the Medline, Embase, and Cochrane Central Register of Controlled Trials databases. We included randomized controlled trials (RCTs) that compared different monoclonal antibodies (e.g. RTX, teprotumumab, and TCZ) with glucocorticoids or placebo in patients with GO. We evaluated the clinical activity score (CAS), proptosis, subjective diplopia using the Gorman score, quality of life (QoT), adverse events, change in lid fissure, NOSPECS score, and TSH receptor antibody (TRAb) levels. The odds ratio (OR) was used to represent dichotomous outcomes. The continuous outcomes were represented as standardized mean difference (SMD). Data were pooled using the inverse variance weighting method. Risk of bias was assessed using the revised Cochrane risk-of-bias tool for randomized trials., Results: Six ( n = 571) RCTs were deemed eligible. The different monoclonal antibodies were significantly more efficacious than glucocorticoid/placebo in terms of reduction in CAS (SMD = -1.44, 95% confidence interval (CI): -1.91--0.97, P < 0.00001, I
2 = 74%), change in proptosis (SMD = -4.96, 95% CI: -8.02--1.89, P = 0.002, I2 = 99%), QoL (SMD = 2.64, 95% CI: 0.50-4.79, P = 0.02, I2 = 97%), and Gorman score for diplopia (OR = 3.42, 95% CI: 1.62-7.22, P = 0.001, I2 = 8%). However, monoclonal antibodies have shown higher rates of adverse events (OR = 2.91, 95% CI: 1.12-7.56, P = 0.03, I2 = 62%). No significant difference was found with respect to lid fissure, NOSPECS, and TRAb levels., Conclusion: This meta-analysis demonstrated that monoclonal antibodies were associated with more favorable clinical outcomes than standard steroid therapy or placebo, especially with regard to CAS, change in proptosis, diplopia, and QoL, with teprotumumab being superior. The recent advancement of nanoparticles (NPs) holds significant potential for treating various ailments. NPs are employed as drug carriers for diseases like cancer because of their small size and increased stability. In addition, they have several desirable properties that make them ideal for treating bone cancer, including high stability, specificity, higher sensitivity, and efficacy. Furthermore, they might be taken into account to permit the precise drug release from the matrix. Drug delivery systems for cancer treatment have progressed to include nanocomposites, metallic NPs, dendrimers, and liposomes. Materials' mechanical strength, hardness, electrical and thermal conductivity, and electrochemical sensors are significantly improved using nanoparticles (NPs). New sensing devices, drug delivery systems, electrochemical sensors, and biosensors can all benefit considerably from the NPs' exceptional physical and chemical capabilities. Nanotechnology is discussed in this article from a variety of angles, including its recent applications in the medical sciences for the effective treatment of bone cancers and its potential as a promising option for treating other complex health anomalies via the use of anti-tumour therapy, radiotherapy, the delivery of proteins, antibiotics, and vaccines, and other methods. This also brings to light the role that model simulations can play in diagnosing and treating bone cancer, an area where Nanomedicine has recently been formulated. There has been a recent uptick in using nanotechnology to treat conditions affecting the skeleton.
