Cardiovascular diseases are the leading cause of mortality, morbidity, and hospitalization around the world. Recent technological advances have facilitated analyzing, visualizing, and monitoring cardiovascular diseases using emerging computational fluid dynamics, blood flow imaging, and wearable sensing technologies. Yet, computational cost, limited spatiotemporal resolution, and obstacles for thorough data analysis have hindered the utility of such techniques to curb cardiovascular diseases. We herein discuss how leveraging machine learning techniques, and in particular deep learning methods, could overcome these limitations and offer promise for translation. We discuss the remarkable capacity of recently developed machine learning techniques to accelerate flow modeling, enhance the resolution while reduce the noise and scanning time of current blood flow imaging techniques, and accurate detection of cardiovascular diseases using a plethora of data collected by wearable sensors.

Microfluidic systems are widely used for studying the mechanotransduction of flow-induced shear stress in mechanosensitive cells. However, these studies are generally performed under constant flow rates, mainly, due to the deficiency of existing pumps for generating transient flows. To address this limitation, we have developed a unique dynamic gravity pump to generate transient flows in microfluidics. The pump utilises a motorised 3D-printed cam-lever mechanism to change the inlet pressure of the system in repeated cycles. 3D printing technology facilitates the rapid and low-cost prototyping of the pump. Customised transient flow patterns can be generated by modulating the profile, size, and rotational speed of the cam, location of the hinge along the lever, and the height of the syringe. Using this unique dynamic gravity pump, we investigated the mechanotransduction of shear stress in HEK293 cells stably expressing Piezo1 mechanosensitive ion channel under transient flows. The controllable, simple, low-cost, compact, and modular design of the pump makes it suitable for studying the mechanobiology of shear sensitive cells under transient flows.

High dielectric constant (high-k) ultrathin films are required as insulating gate materials. The well-known high-k dielectrics, including HfO2, ZrO2, and SrTiO3, feature three-dimensional lattice structures and are thus not easily obtained in the form of distinct ultrathin sheets. Therefore, their deposition as ultrathin layers still imposes challenges for electronic industries. Consequently, new high-k nanomaterials with k in the range of 40 to 100 and a band gap exceeding 4 eV are highly sought after. Antimony oxide nanosheets appear as a potential candidate that could fulfill these characteristics. Here, we report on the stoichiometric cubic polymorph of 2D antimony oxide (Sb2O3) as an ideal high-k dielectric sheet that can be synthesized via a low-temperature, substrate-independent, and silicon-industry-compatible liquid metal synthesis technique. A bismuth-antimony alloy was produced during the growth process. Preferential oxidation caused the surface of the melt to be dominated by α-Sb2O3. This ultrathin α-Sb2O3 was then deposited onto desired surfaces via a liquid metal print transfer. A tunable sheet thickness between ∼1.5 and ∼3 nm was achieved, while the lateral dimensions were within the millimeter range. The obtained α-Sb2O3 exhibited high crystallinity and a wide band gap of ∼4.4 eV. The relative permittivity assessment revealed a maximum k of 84, while a breakdown electric field of ∼10 MV/cm was observed. The isolated 2D α-Sb2O3 nanosheets were utilized in top-gated field-effect transistors that featured low leakage currents, highlighting that the obtained material is a promising gate oxide for conventional and van der Waals heterostructure-based electronics.

Synthesizing two dimensional (2D) nanomaterials with controlled sub-nanometer thicknesses from non-layered crystals presents both significant challenges and vast opportunities. However, mechanical exfoliation techniques and physical/wet chemical deposition processes are widely disadvantageous for applicability to non-layered structures. Here we have utilized a simple self-limiting approach to prepare large sheets of 2D zinc oxide (ZnO) at the metal-melt/air interface. These ultra-thin sheets demonstrated highly crystalline hexagonal structures. The specific ZnO hexagonal sheet thickness and its interaction with the substrate were found to have a critical impact on d33. This unusual structure resulted in an exceptionally high out of plane piezoelectricity, yielding a giant value of 80 ± 0.8 pm/V at 2.5 unit-cell thickness for d33, which is 5 Zn-O layers in the wurtzite crystal. This out of plane piezoelectricity value is approximately 8 times larger than that of the value for bulk ZnO. Theoretical studies were carried out to elucidate the impact of the thickness and the substrate’s role on the polarization of the layers. The existence of a large piezoelectricity offered by the synergy of the substrate and specific thickness of ultrathin films offers the opportunity for other groups of potentially piezoelectric materials to be explored.

Leidenfrost droplets can be considered as soft engines capable of directly transforming heat into mechanical energy. Despite remarkable advancements in understanding the propulsion of Leidenfrost droplets on asymmetric structures, the complex dynamics of droplets in enclosed structures is not fully understood. To address this fundamental gap, we investigated the dynamics of Leidenfrost droplets restricted by metal disks. The disk alters the accumulation and release of the vapour generated by the droplet, and substantially changes its dynamic characteristics. Our experiments reveal the formation of oscillating multi-lobed structures when restricting the droplet within a disk. In comparison, patterning offset radial grooves on the surface of the disk rectifies the vapour flow and facilitates the self-propulsion of the droplet along the edge of the disk. Our work offers opportunities for developing soft and short-living actuators, which can operate at high temperatures.

Here, we demonstrate a modular, reconfigurable, and self-sufficient convective heat exchanger for regulation of temperature in microfluidic systems. The heat exchanger consists of polymer tubes wrapped around a plastic pole and fully embedded in an elastomer block, which can be easily mounted onto the microfluidic structure. It is compatible with various microfluidic geometries and materials. Miniaturized, battery-powered piezoelectric pumps are utilized to drive the heat carrying liquid through the heat exchanger at desired flow rates and temperatures. Customized temperature profiles can be generated by changing the configuration of the heat exchanger with respect to the microfluidic structure. Tailored dynamic temperature profiles can be generated by changing the temperature of the heat carrying liquid in successive cycles. This feature is used to study the calcium signaling of endothelial cells under successive temperature cycles of 24 to 37 °C. The versatility, simplicity, and self-sufficiency of the heat exchanger makes it suitable for various microfluidic based cellular assays.

We describe a piezoelectric pumping system for studying the mechanobiology of human aortic endothelial cells (HAECs) under pulsatile flow in microfluidic structures. The system takes advantage of commercially available components, including pumps, flow sensors, and microfluidic channels, which can be easily integrated, programmed, and operated by cellular biologists. Proof-of-concept experiments were performed to elucidate the complex mechanotransduction processes of endothelial cells to pulsatile flow. In particular, we investigated the effect of atheroprone and atheroprotective pulsatile shear stress on endothelial cytoskeleton remodeling and distribution of β-catenin, as well as nuclear shape and size. The system is simple to operate, relatively inexpensive, portable, and controllable, providing opportunities for studying the mechanobiology of endothelial cells using microfluidic technologies.

Biodevices are crucial for monitoring vital physiological signals, managing chronic health conditions, developing artificial organs for assisting people with disabilities, and conducting various clinical and surgical procedures. However, existing biodevices are mostly composed of rigid components, which can cause discomfort to the user, whereas the high stiffness of implants is known to be the major cause of inflammation and scarring. Gallium‐based liquid metals are intrinsically soft and possess desirable properties, including low toxicity, high conductivity, and deformability, which make them ideally suited for developing soft, deformable, reconfigurable, and healable biodevices. Herein, recent advancements in the emerging field of liquid‐metal‐based biodevices are discussed. This includes a description of the properties of gallium‐based liquid metals which make them so distinct from conventional materials, a brief outline of various techniques devised for fabrication of liquid‐metal‐based devices/structures, and an overview of the diverse range of wearable or implantable liquid‐metal‐enabled biodevices. The outlook and challenges are also discussed.

The human circulatory system is a marvelous fluidic system, which is very sensitive to biophysical and biochemical cues. The current animal and cell culture models do not recapitulate the functional properties of the human circulatory system, limiting our ability to fully understand the complex biological processes underlying the dysfunction of this multifaceted system. In this review, we discuss the unique ability of microfluidic systems to recapitulate the biophysical, biochemical, and functional properties of the human circulatory system. We also describe the remarkable capacity of microfluidic technologies for exploring the complex mechanobiology of the cardiovascular system, mechanistic studying of cardiovascular diseases, and screening cardiovascular drugs with the additional benefit of reducing the need for animal models. We also discuss opportunities for further advancement in this exciting field.

Background: Aortic valve stenosis is an increasingly prevalent degenerative and inflammatory disease. Transcatheter aortic valve implantation (TAVI) has revolutionized its treatment, thereby avoiding its life-threatening/disabling consequences. Whether aortic valve stenosis is accelerated by inflammation and whether it is itself a cause of inflammation are unclear. We hypothesized that the large shear forces exerted on circulating cells, particularly on the largest circulating cells, monocytes, while passing through stenotic aortic valves result in proinflammatory effects that are resolved with TAVI. Methods: TAVI provides a unique opportunity to compare the activation status of monocytes under high shear stress (before TAVI) and under low shear stress (after TAVI). The activation status of monocytes was determined with a single-chain antibody, MAN-1, which is specific for the activated β 2 -integrin Mac-1. Monocyte function was further characterized by the adhesion of myocytes to stimulated endothelial cells, phagocytic activity, uptake of oxidized low-density lipoprotein, and cytokine expression. In addition, we designed a microfluidic system to recapitulate the shear rate conditions before and after TAVI. We used this tool in combination with functional assays, Ca 2+ imaging, siRNA gene silencing, and pharmacological agonists and antagonists to identify the key mechanoreceptor mediating the shear stress sensitivity of monocytes. Last, we stained for monocytes in explanted stenotic aortic human valves. Results: The resolution of high shear stress through TAVI reduces Mac-1 activation, cellular adhesion, phagocytosis, oxidized low-density lipoprotein uptake, and expression of inflammatory markers in monocytes and plasma. Using microfluidics and pharmacological and genetic studies, we could recapitulate high shear stress effects on isolated human monocytes under highly controlled conditions, showing that shear stress–dependent calcium influx and monocyte adhesion are mediated by the mechanosensitive ion channel Piezo-1. We also demonstrate that the expression of this receptor is shear stress dependent and downregulated in patients receiving TAVI. Last, we show monocyte accumulation at the aortic side of leaflets of explanted aortic valves. Conclusions: We demonstrate that high shear stress, as present in patients with aortic valve stenosis, activates multiple monocyte functions, and we identify Piezo-1 as the mainly responsible mechanoreceptor, representing a potentially druggable target. We demonstrate an anti-inflammatory effect and therefore a novel therapeutic benefit of TAVI.

Atomically thin two-dimensional (2D) nanosheets of metal oxides with structural anisotropy, rich surface chemistry and unique electronic structures are technologically intriguing. Importantly their chemical versatility makes them different from their counterparts and potential candidates for number of applications, specifically in the field of energy. This review article will summarize the recent progress in the synthesis of atomically thin 2D metal oxides and their applications for energy storage. It will first explain the structural fundamentals at atomic level thickness and change in surface chemistry due to defective structure and/or unsatisfied surface atoms. Further, it will highlight how different synthesis routes bring variety of 2D materials with tuneable electronic structures. In progress, it will describe the advantages of atomically thin materials over their bulk equals for applications in energy storage devices such as batteries and supercapacitors. Finally, conclusive remarks will be listed with future perspectives towards designing heterostructured metal oxides in 2D nanosheets to overcome their limitations for various energy storage applications.

The role of skin in the human body is indispensable, serving as a barrier, moderating homeostatic balance, and representing a pronounced endpoint for cosmetics and pharmaceuticals. Despite the extensive achievements of in vitro skin models, they do not recapitulate the complexity of human skin; thus, there remains a dependence on animal models during preclinical drug trials, resulting in expensive drug development with high failure rates. By imparting a fine control over the microenvironment and inducing relevant mechanical cues, skin-on-a-chip (SoC) models have circumvented the limitations of conventional cell studies. Enhanced barrier properties, vascularization, and improved phenotypic differentiation have been achieved by SoC models; however, the successful inclusion of appendages such as hair follicles and sweat glands and pigmentation relevance have yet to be realized. The present Review collates the progress of SoC platforms with a focus on their fabrication and the incorporation of mechanical cues, sensors, and blood vessels.

Here, we describe the magnetic actuation of soft shuttles for open-top microfluidic applications. The system is comprised of two immiscible liquids, including glycerol as the soft shuttle and a suspension of iron powder in sucrose solution as the magnetic drop. Permanent magnets assembled on 3D printed motorized actuators were used for the actuation of the magnetic drop, enabling the glycerol shuttle to be propelled along customized linear, circular, and sinusoidal paths. The dynamics of the hybrid shuttle-magnetic drop system was governed by the magnetic force, the friction at the interface of the shuttle and the substrate, and the surface tension at the interface of the shuttle and the magnetic drop. Increasing the magnetic force leads to the localized deformation of the shuttle and eventually the full extraction of the magnetic drop. The versatility of the system was demonstrated through the propelling of the shuttle across a rough surface patterned with microfabricated barriers as well as taking advantage of the optical properties of the shuttle for the magnification and translation of microscale characters patterned on a planar surface. The integration of the system with current electrowetting actuation mechanisms enables the highly controlled motion of the magnetic drop on the surface of a moving shuttle. The simplicity, versatility, and controllability of the system provide opportunities for various fluid manipulation, sample preparation, and analysis for a range of chemical, biochemical, and biological applications.

Here, we demonstrate a multilayered microfluidic system integrated with commercially available polymer tubes for controlling the temperature of the sample under various static and dynamic conditions. Highly controllable temperature profiles can be produced by modulating the flow rate or inlet temperature of the water passing through the tubes. Customised temperature gradients can be created across the length or width of a channel by mismatching the inlet temperature of the tubes. Temperature cycles can also be produced by repeatedly switching the tubes between hot and cold flasks. Proof-of-concept experiments demonstrate the utility of this system for studying the drug-induced calcium signaling of human monocytes under dynamic thermal conditions. The versatility and simplicity of our system provides opportunities for studying temperature-sensitive chemical, biochemical, and biological samples under various operating conditions.

Here, we present a self-sufficient droplet generation system using a highly porous elastomeric sponge. Upon manual compression, aqueous solutions can be loaded into or unloaded from the sponge. The surface of the sponge accommodates hundreds of small orifices, which facilitate generation of microscale droplets when compressed inside an oil container. The physics underlying the generation and size distribution of droplets is explored. Proof-of-concept experiments demonstrate the capability of this method for encapsulation of human monocytes inside droplets. The droplets are chemically isolated, mechanically stable, and do not evaporate due to the presence of oil in the microwell. The cells settle at the lowest surface of the droplets without using any secondary immobilisation mechanisms. These features facilitate chemical stimulation and microscopic analysis of encapsulated cell clusters in a quick and parallel manner. The simplicity and self-sufficiency of this method makes it suitable for conducting various droplet-based cellular assays.

The detection of cancer cells at the single-cell level enables many novel functionalities such as next-generation cancer prognosis and accurate cellular analysis. While surface-enhanced Raman spectroscopy (SERS) has been widely considered as an effective tool in a low-cost and label-free manner, however, it is challenging to discriminate single cancer cells with an accuracy above 90% mainly due to the poor biocompatibility of the noble-metal-based SERS agents. Here, we report a dual-functional nanoprobe based on dopant-driven plasmonic oxides, demonstrating a maximum accuracy above 90% in distinguishing single THP-1 cell from peripheral blood mononuclear cell (PBMC) and human embryonic kidney (HEK) 293 from human macrophage cell line U937 based on their SERS patterns. Furthermore, this nanoprobe can be triggered by the bio-redox response from individual cells towards stimuli, empowering another complementary colorimetric cell detection, approximately achieving the unity discrimination accuracy at a single-cell level. Our strategy could potentially enable the future accurate and low-cost detection of cancer cells from mixed cell samples.

Atomically thin, semiconducting transition and post transition metal oxides are emerging as a promising category of materials for high-performance oxide optoelectronic applications. However, the wafer-scale synthesis of crystalline atomically thin samples has been a challenge, particularly for oxides that do not present layered crystal structures. Herein we use a facile, scalable method to synthesise ultrathin bismuth oxide nanosheets using a liquid metal facilitated synthesis approach. Monolayers of α-Bi2O3 featuring sub-nanometre thickness, high crystallinity and large lateral dimensions could be isolated from the liquid bismuth surface. The nanosheets were found to be n-type semiconductors with a direct band gap of ∼3.5 eV and were suited for developing ultra violet (UV) photodetectors. The developed devices featured a high responsivity of ∼400 AW-1 when illuminated with 365 nm UV light and fast response times of ∼70 μs. The developed methods and obtained nanosheets can likely be developed further towards the synthesis of other bismuth based atomically thin chalcogenides that hold promise for electronic, optical and catalytic applications.

Post-transition elements, together with zinc-group metals and their alloys belong to an emerging class of materials with fascinating characteristics originating from their simultaneous metallic and liquid natures. These metals and alloys are characterised by having low melting points (i.e. between room temperature and 300 °C), making their liquid state accessible to practical applications in various fields of physical chemistry and synthesis. These materials can offer extraordinary capabilities in the synthesis of new materials, catalysis and can also enable novel applications including microfluidics, flexible electronics and drug delivery. However, surprisingly liquid metals have been somewhat neglected by the wider research community. In this review, we provide a comprehensive overview of the fundamentals underlying liquid metal research, including liquid metal synthesis, surface functionalisation and liquid metal enabled chemistry. Furthermore, we discuss phenomena that warrant further investigations in relevant fields and outline how liquid metals can contribute to exciting future applications.

Here, we demonstrate a self-sufficient, inexpensive and disposable pressure pump using commercially available latex balloons. The versatility of the pump is demonstrated against various microfluidic structures, liquid viscosities, and ambient temperatures. The flow rate of the pump can be controlled by varying the size and thickness of the balloon. Importantly, the soft structure of the balloon allows for almost instantaneous change of the flow rate upon manual squeezing of the balloon. This feature has been used for dynamically changing the flow ratio of parallel streams in a T-shaped channel or varying the size of droplets in a droplet generation system. The self-sufficiency, simplicity of fabrication and operation, along with the low-cost of the balloon pump facilitate the widespread application of microfluidic technologies for various research, education, and in situ monitoring purposes.

Microfluidic platforms enable the influence of a variety of chemical and physical gradients on single or multiple cells to be examined and monitored in real-time. Research into chemical gradients has been more prevalent in literature; however, with the interest in mechanotransduction, research investigating the influence of physical gradients, such as force from shear stress, are appearing. Shear stress studies, to date have been focused on using parallel plate flow chambers, or shear stress gradients spanning the length of the microchannel. In this paper, we designed a Trapezoid microchannel, which enables production of a unique lateral spatial shear stress gradient, in order to examine the responses it evokes in cells, using microscopy. We investigated the effect of a lateral spatial shear stress gradient on intracellular calcium signalling in HEK-293-TRPV4 cells. The Trapezoid microchannel was designed to produce a spatial shear stress gradient, spanning the width of the microchannel. Additionally, we investigated the effect of temporal shear stress using the microfluidic platform, to further examine the complexity of the transduction of shear stress forces, which mimic pathological and physiological conditions into cellular signalling. We show that the temporal pattern of shear stress, modulated intracellular calcium signalling, with slower response times (responding to stimulus and reaching maximum Calcium influx), compared to those observed in the absence of temporal shear stress. Our experiments highlight the importance of considering the nature of shear stress under physiological and pathological conditions – with microfluidic platforms providing a useful means to do so.

Mechanical stress from blood flow has a significant effect on endothelial physiology, with a key role in initiating vasoregulatory signals. Disturbances in blood flow, such as in regions of disease-associated stenosis, arterial branch points, and sharp turns, can induce proatherogenic phenotypes in endothelial cells. The disruption of vascular homeostasis as a result of endothelial dysfunction may contribute to early and late stages of atherosclerosis, the underlying cause of coronary artery disease. In-depth knowledge of the mechanobiology of endothelial cells is essential to identifying mechanosensory complexes involved in the pathogenesis of atherosclerosis. In this review, we describe different blood flow patterns and summarize current knowledge on mechanosensory molecules regulating endothelial vasoregulatory functions, with clinical implications. Such information may help in the search for novel therapeutic approaches.
