Streptococcal infections are becoming an increasing problem in aquaculture and have been reported worldwide in avariety of fish species. Here we describe the isolation and characterization of Streptococcus iniae from Asian sea bass (Latescalcarifer) and red tilapia (Oreochromis sp.) cultured in southern Thailand. Conventional and rapid identification systems,as well as the polymerase chain reaction (PCR), were used to determine that the isolate was S. iniae. The virulence of thisS. iniae was higher in Asian sea bass than in red tilapia, as shown by the 10 day-LD50 in Asian sea bass and red tilapia, being1.08x104 and 1.14x107 CFU, respectively. Histopathological changes in Asian sea bass are more severe than those observedin red tilapia. The changes can be found in several organs including liver, pancreas, heart, eye and brain. Histopathologicalfindings included cellular necrosis, infiltration of lymphocytes and granuloma formation in the infected organs.

Gonapodasmius epinepheli Abdul-Salam, Sreelatha & Farah, 1990, a didymozoid trematode, was found in orange spotted grouper (Epinephelus coioides, Hamilton, 1822) cultured in cages in Southern Thailand, both on the east coast (Gulf of Thailand) and the west coast (Indian Ocean). The parasite encysted on the primary lamellae of the gills. Histological sections revealed that larvae were primarily embedded underneath the gill epithelium and eggs of the parasite were distributed in some organs such as gill epithelium, heart and macrophage centers in the head kidney. Pathological changes and host response were shown mainly on gill lamellae, e.g. reduction and destruction of secondary lamellae. Prevalence of parasite and its life cycle are discussed in this report.

Ochratoxin A (OTA) and deoxynivalenol (DON), naturally occurring contaminants of animal feed, have been implicated in several mycotoxicoses in farm livestock but there is little information on their toxicity in aquatic invertebrates. Therefore, in the present study an 8-week feeding trial was conducted on black tiger shrimp (Penaeus monodon) to assess the effects of OTA and DON on growth performance, haemolymph parameters and histopathology of shrimp. Results showed that feed supplemented with DON caused no effect on growth or survival rate of the shrimp. However, shrimps fed DON feed diet with 1,000 ppb showed significantly higher growth performance. No significant difference in total haemocyte counts (THC) was found in shrimp fed mycotoxins-supplemented feed. Feeding high level of OTA (1,000 ppb) caused a decrease in phenoloxidase (PO) activity. Although no histopathological change was observed, decrease in serum alkaline phosphatase (ALP), serum glutamic-oxaloacetic transaminase (SGOT) and serum glutamic-pyruvic transaminase (SGPT) levels in shrimp fed OTA or DON indicated that mycotoxin may impair the functioning of hepatopancreatic cells. Since no adverse effect occurred with the highest levels of OTA and DON (1,000 and 2,000 ppb, respectively) on haemolymph parameters and no residue was detected at the completion of the 8-week feeding period, it can be concluded that shrimp feeds occasionally contaminated with OTA or DON have no negative impact on the shrimp culture industry.

In recent years, there has been a growing focus on using herbal extracts as immune enhancers for aquatic species, replacing antibiotics. In the present study, the effects of dietary supplementation of Hericium erinaceus extract (HE) on growth, feed utilization, hematology, expression of immunity-related genes, and immune responses in Nile tilapia infected by Streptococcus agalactiae were examined. A total of 240 Nile tilapia with an average body weight of 17.28 ± 0.01 g were fed diets enriched with different levels of HE: 0 (HE0), 0.1 (HE0.1), 1.0 (HE1.0), and 5.0 (HE5.0) g/kg. The results showed that growth parameters, feed conversion ratio, and organosomatic indexes were not linearly or quadratically affected by HE supplementation. Fish fed HE0.1 and HE1.0 increased protein efficiency ratio and protein productive values with significant linear and quadratic effects of HE enrichment. In addition, dietary supplementation of HE quadratically increased whole-body protein content. Red blood cell, white blood cell, and hematocrit were linearly and quadratically increased by HE supplementation. HE also linearly and quadratically decreased LDL cholesterol and linearly decreased the total cholesterol levels. Stress markers, serum glucose, and cortisol levels were linearly and/or quadratically decreased in HE-fed fish. The relative mRNA expression of tnf-α, il-1β, il-6, and il-10 were upregulated in the HE0.1 and HE1.0 groups, while dietary supplementation of HE significantly decreased hsp70cb1 mRNA expression in all groups. After feeding dietary HE supplementation for 10 weeks, fish were intraperitoneally injected with pathogenic S. agalactiae. A high survival after challenge was found in all HE supplementation groups with the highest percent survival observed in the HE1.0 and HE5.0 groups. Our findings represent that supplementation of 1 g/kg of HE (HE1.0) could obtain the greatest effects on immunity and survival of Nile tilapia. Microwaving has been used to qualitatively improve feed ingredients prior to including them in cattle feed. However, it is not known whether feed ingredients should be microwaved separately or in a mixture before being included in cattle feed concentrates. In the current study, the effects of the partial and full microwaving of high starch ingredients were investigated regarding their impact on the nutritional composition, physicochemical properties, and in vitro digestibility of feed concentrates. The cassava was microwave-irradiated before being mixed with other ingredients (MC, 35% of formulation). A microwave-cooked cassava-corn meal mixture (MCC, 45% of formulation) and a combination of all solid components (MSI, 96% of formulation) were also compared. A feed containing non-microwaved ingredients was formulated and served as the control (NM). Significantly altered proximate compositions and nutritive profiles were observed in all the microwaved components ( p < 0.05). The observed modifications in pHs, water absorption capacities, thermal properties (differential scanning calorimetry), diffraction patterns (X-ray diffractometry), and microstructures (scanning electron microscopy) indicated enhanced enzymatic hydrolysis in vitro. A higher cellulase organic matter solubility and digestible organic matter were observed in the MCC and MSIs feeds relative to the control NM ( p < 0.05). These findings indicate a combination of components could be added to feedstock before it is microwave processed to potentially include this pretreatment in the feed production process.

Physical pretreatment procedures can significantly influence the quality of food and feed raw materials. To increase the ability to be digested in animals, cassava chips were pretreated by four alternative methods (extrusion, microwave irradiation, gamma irradiation, or NaOH hydrolysis), and then the chemical composition, physicochemical properties, and in vitro digestibility of the pretreated samples were assessed and compared with unprocessed cassava chips (control). The chemical compositions (crude protein, ether extract, neutral detergent fiber, acid detergent fiber, ash, non-fiber carbohydrate, and gross energy) were significantly altered due to the pretreatment methods ( p < 0.05). The nutritive profile was qualitatively changed when assessed through Fourier-transform infrared spectroscopy. Some physicochemical properties in association with enzymatic hydrolysis, which include pH, water solubility, water absorption capacity, thermal properties (differential scanning calorimetry), diffraction pattern (X-ray diffractometry), and microstructure (scanning electron microscopy), were significantly changed. In vitro carbohydrate digestibility based on digestive enzyme extracts from Nile tilapia ( Oreochromis niloticus ) suggests the microwaving method for cassava chips preparation, while microwaving, followed by extrusion or gamma irradiation, was suggested for broiler ( Gallus gallus domesticus ). There were no differences in the pepsin-cellulase digestibility values tested for the ruminant model. These findings suggest the use of pretreated cassava chips in animal feeding.

The present study aimed to investigate the effect of thermal stress on growth, feed utilization, coloration, hematology, liver histology, and critical thermal maximum (CTmax) in goldfish (Carassius auratus) cultured at three different acclimation temperatures including 27 °C, 30 °C, and 34 °C for 10 weeks. Goldfish were assigned randomly to tanks with a quadruplicate setup, accommodating 20 fish per tank. The result showed that fish acclimated to different temperatures did not significantly differ in weight gain (WG) and specific growth rate (SGR). However, increasing temperature significantly decreased feed efficiency ratio (FER), protein efficiency ratio (PER), and protein productive value (PPV), but significantly increased feed conversion ratio (FCR) (P < 0.05). The coloration parameters significantly decreased by high temperature in the trunk region with increasing temperature (L* and a* at week 5; L*, a*, and b* at week 10; P < 0.05). Total carotenoid contents in serum, fin, muscle, and skin also significantly decreased with increasing temperature (P < 0.05). Total protein, albumin, and globulin levels exhibited a notable decrease, while the albumin: globulin ratio showed a slight insignificant increase, with increasing temperature. Aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), total cholesterol, and triglycerides significantly increased with increasing temperature (P < 0.05). While, high-density lipoprotein cholesterol (HDL-c) decreased linearly (P < 0.05). Glucose and cortisol levels linearly increased with increasing temperature, the highest levels being observed in the 34 °C group. Liver histology showed swollen hepatocytes, nuclei displacement, and infiltration of inflammation in fish cultured at 34 °C. Goldfish acclimated to 34 °C displayed a higher CTmax of 43.83 °C compared to other groups. The solid-state fermentation by effective microorganisms (containing photosynthetic bacteria, lactic acid bacteria, nitrogen-fixing bacteria, yeast and Bacillus sp.) improved the nutritive values of palm kernel meal (PKM). Increased crude protein (20.79%), nitrogen-free extract (40.07%) and gross energy (19.58%) were observed in fermented PKM (FPKM) relative to raw PKM while crude lipid (15.65%), crude fiber (36.45%) and ash (29.54%) were decreased. Replacement of soybean meal (SBM) with FPKM as a protein source was investigated for its effects in sex-reversed red tilapia ( Oreochromis niloticus × O. mossambicus ). The two-month-old fish (14.85 ± 0.28 g initial weight) were fed fish meal-SBM-based diets with replacement by FPKM at 25% (25FPKM), 50% (50FPKM), 75% (75FPKM) and 100% (100FPKM), while an FPKM-free diet (0FPKM) was used as a control. The five treatments, comprising triplicate cement ponds and forty fish each, were conducted in a recirculating system over 12 weeks. At the end of the feeding trial, fish fed the 50FPKM diet were superior in growth performance, while the feed utilization parameters were similar across all five treatments. Physiological adaptation of the protein-digesting (pepsin and trypsin) and lipid-digesting (lipase) enzymes was detected at all protein replacement levels (except for 25FPKM), as well as of the enzyme for cellulose digestion (cellulase), but not of the carbohydrate-digesting enzymes (amylase). Protein synthesis capacity in flesh was improved in fish fed the 50FPKM diet, while the quality of the main flesh proteins, actin and myosin, showed no significant differences across the five treatments. No differences in carcass composition and no negative effects on hematological parameters or liver histoarchitecture at the 50% replacement level of SBM by FPKM also support this alternative. Findings from the current study indicate the low-cost FPKM-containing diet for tilapia in comparison with control diet.

Streptococcus agalactiae (group B Streptococcus, GBS) infection remains a major problem associated with high mortality of cultured tilapia worldwide. The present study reports the serotype distribution and antimicrobial susceptibilities of GBS isolated from infected tilapia cultured in Thailand. One hundred and forty-four GBS isolates were identified by biochemical, serological and molecular analyses. Of these 144 GBS isolates, 126 were serotype Ia and 18 were serotype III. Antimicrobial susceptibilities of the 144 GBS isolates were determined by the disc diffusion method. Most GBS isolates were susceptible to lincomycin, norfloxacin, oxytetracycline, ampicillin, erythromycin and chloramphenicol, but resistant to oxolinic acid, gentamicin, sulfamethoxazole and trimethoprim. However, 17 isolates displayed an oxytetracycline-resistant phenotype and harboured the tet(M) gene. The broth microdilution method was used to determine the minimal inhibitory concentrations (MICs) of 17 oxytetracycline-resistant GBS isolates, and then minimal bactericidal concentrations (MBCs) of these isolates were evaluated. Oxytetracyline-resistant isolates were found to be susceptible to ampicillin, lincomycin, norfloxacin, erythromycin and chloramphenicol, with the MIC and MBC ranging from ≤ 0.125 to 0.5 μg ml- 1 and ≤ 0.125 to 2 μg ml- 1, respectively. Moreover, all 17 oxytetracycline-resistant isolates demonstrated resistance to trimethoprim, oxolinic acid, gentamicin, sulfamethoxazole and oxytetracycline, with the MIC and MBC ranging from 16 to ≥ 128 μg ml- 1 and ≥ 128 μg ml- 1, respectively. These findings are useful information for antibiotic usage in fish aquaculture.

Enteric septicaemia of catfish (ESC) caused by Edwardsiella ictaluri is becoming an increasing problem in aquaculture and has been reported worldwide in a variety of fish species. This study reports ESC in hybrid catfish, Clarias macrocephalus (Günther) × Clarias gariepinus (Burchell), cultured in southern Thailand. The bacteria were identified as E. ictaluri by conventional and rapid identification systems, as well as by genetic and phylogenetic characterization. Analysis of 16S rRNA indicated 100% homology to the 16S rRNA sequence of several E. ictaluri strains in GenBank. Plasmid profiles demonstrated 4.0- and 5.6-kb plasmids, compared with the 4.8- and 5.6-kb plasmids in the US isolates, and representative genes of three of the four known pathogenicity islands of US isolates were present. Serologically, lipopolysaccharide (LPS) purified from the Thai isolates was not recognized by a monoclonal antibody against the LPS of US isolates. Fish experimentally infected with E. ictaluri showed 23-100% mortality within 14 days with a 168-h LD50 of 6.92 × 10(7)  CFU mL(-1) by immersion and a 96-h LD50 of 1.58 × 10(6)  CFU fish(-1) by intraperitoneal injection. Examination of tissue sections obtained from both naturally and experimentally infected fish indicated that infection of hybrid catfish with E. ictaluri produced lesions in several organs including liver, kidney, spleen, heart and brain. During April to June 2009 and February 2010 to February 2011, numerous digenetic trematode metacercariae were observed embedded in the muscles of guppies Poecilia reticulata cultured at Kidchakan Supamattaya Aquatic Animal Health Research Center, Songkhla, southern Thailand. A total of 424 guppies were examined to identify the parasite and to study its pathogenicity. Based on conventional parasitological techniques, the parasitic digenean found in the guppies was identified as Euclinostomum heterostomum Rudolphi, 1809. Histological analysis revealed numerous metacercariae embedded in the fish muscle. A life cycle study indicated that the snail Indoplanorbis exustus was the first intermediate host, with the guppies serving as the second intermediate host. No E. heterostomum metacercariae were found in cohabited fish species, giant sailfin molly Poecilia velifera or platy Xiphophorus maculatus, which indicated that the guppy was the only suitable fish host present.
