2. Conserving Human Creativity with Evolutionary Generative Algorithms: A Case Study in Music Generation

This study explores the application of evolutionary generative algorithms in music production to preserve and enhance human creativity. By integrating human feedback into Differential Evolution algorithms, we produced six songs that were submitted to international record labels, all of which received contract offers. In addition to testing the commercial viability of these methods, this paper examines the long-term implications of content generation using traditional machine learning methods compared with evolutionary algorithms. Specifically, as current generative techniques continue to scale, the potential for computer-generated content to outpace human creation becomes likely. This trend poses a risk of exhausting the pool of human-created training data, potentially forcing generative machine learning models to increasingly depend on their random input functions for generating novel content. In contrast to a future of content generation guided by aimless random functions, our approach allows for individualized creative exploration, ensuring that computer-assisted content generation methods are human-centric and culturally relevant through time., Comment: 7 pages, 3 figures

3. GABAergic integration of transient and persistent neurons in the developing mouse somatosensory cortex

GABA is an essential element in the function of neocortical circuits. The origin, migration and mechanisms of synaptogenesis of GABAergic neurons have been intensively studied. However, little information is available when GABAergic synapses are formed within the different cortical layers, neuronal cell types and subcellular compartments. To quantify the distribution of GABAergic synapses in the immature somatosensory mouse cortex, GABAergic synapses were identified by spatially coincident immunoprofiles for the pre- and postsynaptic markers vGAT and gephyrin at postnatal days (P)0-12. Between P0-5, GABAergic synapses are mainly restricted to the marginal zone, while at later developmental stages a more homogenous distribution is obtained. Cajal-Retzius neurons represent a major target of GABAergic synapses in the marginal zone with a homogeneous synapse distribution along the dendrite. The number of GABAergic synapses per pyramidal neuron increases substantially between P0 and P12, with a stable density and distribution in basal dendrites. In contrast, along apical dendrites synapses accumulate to more proximal positions after P8. Overall, the results of this study demonstrate that early GABAergic synaptogenesis is characterized by a consistent increase in the density of synapses with first a stringent overrepresentation in the marginal zone and a delayed establishment of perisomatic synapses in pyramidal neurons.

15. Lithospheric Foundering in Progress Imaged Under an Extinct Continental Arc

Abstract A long‐standing question is how felsic continental crust is differentiated from its mafic parent mantle magmas. One currently proposed fundamental mechanism is lithospheric foundering and loss of dense material into the mantle. A type locality is the young extinct arc forming the Sierra Nevada, California. Here, we image a distinct anisotropic shear layer below the crust‐mantle boundary using receiver functions. The sense of shear is consistent with west‐ to southwestward removal of lithosphere. The shear signal is strongest in the southern Sierra, where lithospheric foundering was proposed to have concluded several million years ago, and is deeper and less pronounced in the central Sierra, where ongoing lithospheric foundering is corroborated by a band of unusually deep (40+ km) seismicity along the western foothills. Our observations provide progressive snapshots of a lithospheric foundering process spanning several million years and hundreds of kilometers, illuminating a fundamental differentiation process by which continents are built.

16. Putting faults in the northern Chilean subduction margin into motion: evidence for remote dynamic earthquake triggering on the plate interface and within the forearc

Dynamic stresses on the order of ~1 kPa from passing waves of mainshock earthquakes can trigger aftershocks at remote distances. Here, we investigate the prevalence of remote earthquake triggering in northern Chile, where aseismic-slip triggering has been documented. Our twofold approach to quantify triggerability includes a statistical difference-of-means test to quantify seismicity-rate changes bracketing candidate mainshock times, and a waveform-based approach to look for triggered earthquakes missing from the local catalog. We find no persistent, statistically-significant seismicity-rate increases associated with any of the candidate mainshocks when considering the local catalog in aggregate. However, catalog statistics reveal evidence for localized triggering both on the subduction interface and within the shallower forearc faults. Waveforms reveal local, uncataloged earthquakes only visible using a high-pass filter that removes the mainshock signal that otherwise overprints the local signals. Based on Japan mainshocks, we cannot rule out antipodal triggering. Areas showing higher triggerability are consistent with regions of low locking inferred from GNSS models and regions of observed aseismic slip. The spatial coincidence of triggering and low-locking, combined with the absence of a stress-triggering threshold, requires non-linear triggering mechanisms, such as altered frictional strength or aseismic-slip triggering, to be consistent with the observations.

Background Online communities and social networking sites have great potential for supporting health behavior change. However, interventions vary greatly in participants’ engagement rates and, consequently, their effectiveness. Theory-based interventions in real-world contexts are needed to further increase engagement and effectiveness. Methods We experimentally tested whether a video intervention teaching Self-Determination-Theory-based communication strategies increases need-supportive communication strategy use over one week (Study 1, N = 76) and perceived need support, engagement, and goal attainment in a behavior change intervention supported by a forum-based online community (Study 2, N = 537). In Study 2, participants chose a goal (increasing either fruit or vegetable consumption or increasing moderate or vigorous physical activity) and joined an online community for 2 weeks. Data from both experiments were analyzed with mixed models and follow-up tests. Results In Study 1, participants in the intervention but not in the control group showed an increase in the number of need-supportive communication strategies used both immediately and one week after the intervention (condition × time interaction, partial η2 = 0.31). In Study 2, participants who watched the intervention video had a higher number of postings and reported a higher subjective forum use frequency (but not a higher number of logins) compared to participants who watched the control video. However, the effect on the subjective forum visit frequency was not robust. There were no intervention effects on perceived need support, goal attainment, or secondary outcomes. The results might be explained by low application of need-supportive communication strategies. Conclusion A brief video intervention may be a suitable, low-cost intervention to promote need-supportive communication strategy use, benefitting both engagement and behavior change. Future studies should incorporate additional means to further improve communication strategy uptake and engagement in online communities.

IntroductionShort-term synaptic plasticity (STP) is a widespread mechanism underlying activity-dependent modifications of cortical networks.MethodsTo investigate how STP influences excitatory and inhibitory synapses in layer 2/3 of mouse barrel cortex, we combined whole-cell patch-clamp recordings from visually identified pyramidal neurons (PyrN) and parvalbumin-positive interneurons (PV-IN) of cortical layer 2/3 in acute slices with electrical stimulation of afferent fibers in layer 4 and optogenetic activation of PV-IN.ResultsThese experiments revealed that electrical burst stimulation (10 pulses at 10 Hz) of layer 4 afferents to layer 2/3 neurons induced comparable short-term depression (STD) of glutamatergic postsynaptic currents (PSCs) in PyrN and in PV-IN, while disynaptic GABAergic PSCs in PyrN showed a stronger depression. Burst-induced depression of glutamatergic PSCs decayed within 11 s. Optogenetically-induced GABAergic PSCs in PyrN also demonstrated STD after burst stimulation, with a decay of >11 s. Excitatory postsynaptic potentials (EPSPs) in PyrN were unaffected after electrical burst stimulation, while a selective optogenetic STD of GABAergic synapses caused a transient increase of electrically evoked EPSPs in PyrN.DiscussionIn summary, these results demonstrate substantial short-term plasticity at all synapses investigated and suggest that the prominent STD observed in GABAergic synapses can moderate the functional efficacy of glutamatergic STD after repetitive synaptic stimulations. This mechanism may contribute to a reliable information flow toward the integrative layer 2/3 for complex time-varying sensory stimuli.

Abstract Background Habits drive many of our health behaviors in our daily lives. However, little is known about the relative contribution of different key factors for habit formation in real-world contexts. We examined the effects of behavioral performance, intrinsic reward value (operationalized as tastiness), and context stability on the formation of a higher-order nutrition habit. Methods Participants were recruited via mailing lists and posts on social media platforms. N = 199 participants (M age = 37.10 years, SD = 13.00, 86.93% female) received an online intervention for building the higher-order habit of filling half of their plates with vegetables at dinner and completed one daily online survey for up to 56 days, including the assessment of habit strength, behavioral performance, intrinsic reward value, and context stability, providing a total of N = 6352 daily measurements. N = 189 participants (N = 4175 measurements) could be included in the primary analysis. Utilizing multilevel modeling, we analyzed the impact of behavioral performance, intrinsic reward value, and context stability, as well as their interaction effects, on habit strength on the next day. Results Habit strength significantly increased over time. This effect was strengthened in persons with high mean levels of behavioral performance. Furthermore, mean levels of behavioral performance, intrinsic reward value, and context stability were all positively related to mean levels of habit strength. There were no positive effects of daily intraindividual variations in the three examined factors on habit strength at the next day. There was an unexpected negative effect of daily behavioral performance on habit strength at the next day. We found little to no evidence for our expected and pre-registered interaction effects. In an additional exploratory analysis, there were positive effects of daily intraindividual variations in the three factors on habit strength at the same day. Conclusions We found that behavioral performance, intrinsic reward value, and context stability were all independent predictors of habit strength of a higher-order habit at the between-person level. However, we did not find the expected associations at the within-person level. Habit interventions should promote the consistent performance of the target behaviors in stable contexts. BACKGROUND Recruitment and retention of Pulmonary and Critical Care Medicine (PCCM) trainees into academic research positions remain difficult. Factors influencing graduates, like salary and personal circumstances, remain unchangeable. However, some program-level factors, like research skill acquisition and mentorship, may be modifiable to encourage matriculation into academic research positions. OBJECTIVE We aim to identify proficiency in research-specific skills in PCCM trainees and barriers to careers as research-focused academic faculty. METHODS We surveyed PCCM fellows in a nationwide cross-sectional analysis including demographics, research intent, research skills self-assessment, and academic career barriers. The Association of Pulmonary and Critical Care Medicine Program Directors approved and disseminated the survey. Data were collected and stored using the REDCap database. Descriptive statistics were used to assess survey items. RESULTS 612 fellows received the primary survey with 112 completing the survey for a response rate of 18.3%. A majority were male (56.2%) and training at university-based medical centers (89.2%). Early fellowship trainees (first-/second-year fellows) comprised 66.9% of respondents with 33.1% being late fellowship trainees (third-/fourth-year fellows). Most early trainees (63.2%) indicated they intended to incorporate research into their careers. A chi-square testing of independence was performed to examine the relationship between training level and perceived proficiency. Significant relationships in perceived proficiency were identified between early and late fellowship trainees with an absolute difference of 25.3% (manuscript writing), 18.7% (grant writing), 21.6% (study design), and 19.5% (quantitative/qualitative methodology). The most prevalent barriers were unfamiliarity with grant writing (59.5%) and research funding uncertainty (56.8%). CONCLUSION With an ongoing need for academic research faculty, this study identifies self-perceived gaps in research skills including grant writing, data analytics, and study conception and design. These skills map to fellow-identified barriers to careers in academics. Mentorship and innovative curriculum focusing on the development of key research skills may enhance academic research faculty recruitment.

The development of functionally interconnected networks between primary (S1), secondary somatosensory (S2), and motor (M1) cortical areas requires coherent neuronal activity via corticocortical projections. However, the anatomical substrate of functional connections between S1 and M1 or S2 during early development remains elusive. In the present study, we used ex vivo carbocyanine dye (DiI) tracing in paraformaldehyde-fixed newborn mouse brain to investigate axonal projections of neurons in different layers of S1 barrel field (S1Bf), M1, and S2 toward the subplate (SP), a hub layer for sensory information transfer in the immature cortex. In addition, we performed extracellular recordings in neocortical slices to unravel the functional connectivity between these areas. Our experiments demonstrate that already at P0 neurons from the cortical plate (CP), layer 5/6 (L5/6), and the SP of both M1 and S2 send projections through the SP of S1Bf. Reciprocally, neurons from CP to SP of S1Bf send projections through the SP of M1 and S2. Electrophysiological recordings with multi-electrode arrays in cortical slices revealed weak, but functional synaptic connections between SP and L5/6 within and between S1 and M1. An even lower functional connectivity was observed between S1 and S2. In summary, our findings demonstrate that functional connections between SP and upper cortical layers are not confined to the same cortical area, but corticocortical connection between adjacent cortical areas exist already at the day of birth. Hereby, SP can integrate early cortical activity of M1, S1, and S2 and shape the development of sensorimotor integration at an early stage.

A long‐standing question is how felsic continental crust is differentiated from its mafic parent mantle magmas. One currently proposed fundamental mechanism is lithospheric foundering and loss of dense material into the mantle. A type locality is the young extinct arc forming the Sierra Nevada, California. Here, we image a distinct anisotropic shear layer below the crust‐mantle boundary using receiver functions. The sense of shear is consistent with west‐ to southwestward removal of lithosphere. The shear signal is strongest in the southern Sierra, where lithospheric foundering was proposed to have concluded several million years ago, and is deeper and less pronounced in the central Sierra, where ongoing lithospheric foundering is corroborated by a band of unusually deep (40+ km) seismicity along the western foothills. Our observations provide progressive snapshots of a lithospheric foundering process spanning several million years and hundreds of kilometers, illuminating a fundamental differentiation process by which continents are built. Plain Language Summary: The Earth's continents sit above sea level and thus make up the land surfaces we live on thanks to their large share of low‐density felsic minerals. The basaltic melts that continents are made from have a higher concentration of denser mafic minerals in comparison. To make continental crust, mafic portions have to be removed, a process called differentiation. A proposed mechanism for this is that dense portions of the lithosphere detach from more felsic portions above and are absorbed back into the Earth's mantle. We image the imprint on rocks at the current crust‐mantle boundary that recorded a dense portion below it peeling off under the Sierra Nevada in California, USA. The removal happened several million years ago in the southern part of the mountains and is still in progress under the central part, causing very deep small earthquakes, while the northern part still retains its dense layer. We have therefore captured snapshots of a fundamental continent‐building process. IntroductionThe SARS CoV-2 pandemic poses major challenges not only to patients but also to health care professionals and policy-makers, with rapidly changing, sometimes complex, recommendations, and guidelines to the population. Online forward triage tools (OFTT) got a major boost from the pandemic as they helped with the implementation and monitoring of recommendations.MethodsA multiphase mixed method sequential explanatory study design was employed. Quantitative data were collected first and informed the qualitative interview guides. Video interviews were held with key informants (health care providers and health authorities) between 2 September and 10 December 2020. Audio-recordings were transcribed verbatim, coded thematically and compared with patient perspectives (framework).ObjectivesTo explore the perspectives of health care providers and authorities in Canton Bern on the utility of a COVID-19 OFTT, as well as elicit recommendations for telehealth in future.ResultsThe following themes emerged; (i) accessibility (ii) health system burden reduction (iii) utility in preventing onward transmission (iv) utility in allaying fear and anxiety (v) medical decision-making utility (vi) utility as information source (vii) utility in planning and systems thinking. The health care providers and health authorities further provided insights on potential barriers and facilitators of telehealth in future.ConclusionSimilar to patients, health care providers acknowledge the potential and utility of the COVID-19 OFTT particularly as an information source and in reducing the health system burden. Data privacy, doctor-patient relationship, resistance to change, regulatory, and mandate issues, and lack of systems thinking were revealed as barriers to COVID-19 OFTT utility.

GABA (γ-amino butyric acid) is an inhibitory neurotransmitter in the adult brain that can mediate depolarizing responses during development or after neuropathological insults. Under which conditions GABAergic membrane depolarizations are sufficient to impose excitatory effects is hard to predict, as shunting inhibition and GABAergic effects on spatiotemporal filtering of excitatory inputs must be considered. To evaluate at which reversal potential a net excitatory effect was imposed by GABA (EGABAThr), we performed a detailed in-silico study using simple neuronal topologies and distinct spatiotemporal relations between GABAergic and glutamatergic inputs. These simulations revealed for GABAergic synapses located at the soma an EGABAThr close to action potential threshold (EAPThr), while with increasing dendritic distance EGABAThr shifted to positive values. The impact of GABA on AMPA-mediated inputs revealed a complex temporal and spatial dependency. EGABAThr depends on the temporal relation between GABA and AMPA inputs, with a striking negative shift in EGABAThr for AMPA inputs appearing after the GABA input. The spatial dependency between GABA and AMPA inputs revealed a complex profile, with EGABAThr being shifted to values negative to EAPThr for AMPA synapses located proximally to the GABA input, while for distally located AMPA synapses the dendritic distance had only a minor effect on EGABAThr. For tonic GABAergic conductances EGABAThr was negative to EAPThr over a wide range of gGABAtonic values. In summary, these results demonstrate that for several physiologically relevant situations EGABAThr is negative to EAPThr, suggesting that depolarizing GABAergic responses can mediate excitatory effects even if EGABA did not reach EAPThr.
