Abstract

Solar vehicles, which simultaneously produce and consume energy, require meticulous energy management. However, potential users often feel uncertain about their operation compared to conventional vehicles. This study presents a simulator designed to help users understand long-distance travel in solar vehicles and recognize the importance of proper energy management. By utilizing Google Maps data and weather information, the simulator replicates real-world driving conditions and provides a dashboard displaying vehicle status, updated hourly based on user-inputted speed. Users can explore various speed policy scenarios and receive recommendations for optimal driving strategies. The simulator's effectiveness was validated using the route of the World Solar Challenge (WSC). This research enables users to monitor energy dynamics before a journey, enhancing their understanding of energy management and informing appropriate speed decisions.

Abstract

The ability to spatially and temporally map nanoscale environments in situ over extended timescales would be transformative for biology, biomedicine, and bioengineering. All nanometer objects, from nanoparticles down to single proteins, scatter light. Interferometric scattering stands as a powerful tool, offering ultrasensitivity and resolution vital for visualizing nanoscale entities. Interferometric scattering from an individual nanoparticle down to an individual protein has been detected; however, resolving adjacent nanometer objects with interferometric scattering has not yet been demonstrated. In this work, we present interferometric phase intensity nanoscopy to resolve adjacent nanometer objects with interferometric scattering. We demonstrate that multiphase and sensitivity of interferometric phase intensity nanoscopy reveals ellipse Airy patterns correlated with nanostructural features. We show that eliminating background fluctuation by employing circular polarized illumination in interferometric phase intensity nanoscopy is essential to separate proximal nanometer objects below the diffraction limit. We envision interferometric phase intensity nanoscopy for resolving a variety of adjacent nanometer objects from nanoparticles down to proximal proteins in situ over extended time periods for wide range of applications in biology, biomedicine and bioengineering.

Abstract

Fundamental to all living organisms and living soft matter are emergent processes in which the reorganization of individual constituents at the nanoscale drives group-level movements and shape changes at the macroscale over time. However, light-induced degradation of fluorophores, photobleaching, is a significant problem in extended live cell imaging in life science, and excellent prior arts of STED, MINSTED, or MINFLUX intentionally apply photobleaching optics for super-resolution, yet lack extended live-cell imaging capability as fluorophores reach an irreversible photobleaching limit. Here, we report an innovative way of opening a long-time investigation window of living matter by a nonbleaching phase intensity nanoscope: PINE. We accomplish phase-intensity separation to obtain phase differences between electric field components, such that nanoprobes within a diffraction limited region are distinguished from one another by an integrated phase-intensity multilayer thin film (polyvinyl alcohol/liquid crystal). We obtain distributional patterns of precisely localized nanoprobes to overcome a physical limit to resolve sub-10 nm cellular architectures, and achieve the first dynamic imaging of nanoscopic reorganization over 250 hours using nonbleaching PINE. Since PINE allows a long-time investigation window, we discover nanoscopic rearrangements synchronized with the emergence of group-level movements and shape changes at the macroscale according to a set of interaction rules. We believe that PINE and its application presented here provide a mechanistic understanding of emergent dynamics important in cellular and soft matter reorganization, self-organization, and pattern formation.

Methods We conducted a retrospective analysis of children aged < 3 years who were evaluated for UTI with urinalysis, urine culture, and uNGAL measurements during a 5-year period. Sensitivity, specificity, likelihood ratios (LRs), predictive values (PVs), area under the curves (AUCs) of uNGAL cut-off levels, and various microscopic pyuria thresholds for detecting UTI were calculated for dilute (SG < 1.015) and concentrated urine (SG [greater than or equal to] 1.015). Results Of 456 children included, 218 had UTI. The diagnostic value of urine white blood cell (WBC) concentration to define UTI changed with urine SG. For detecting UTI, uNGAL cut-off of 68.4 ng/mL had higher AUC values than pyuria [greater than or equal to] 5 WBCs/high power field (HPF) for dilute and concentrated urine samples (both P 0.05). Positive LR and PV and specificity of uNGAL were all greater than those of pyuria [greater than or equal to] 5 WBCs/HPF regardless of urine SG, although the sensitivity of pyuria [greater than or equal to] 5 WBCs/HPF was higher than that of uNGAL cut-off for dilute urine (93.8% vs. 83.5%) (P < 0.05). At uNGAL [greater than or equal to] 68.4 ng/mL and [greater than or equal to] 5 WBCs/HPF, posttest probabilities of UTI were 68.8% and 57.5% for dilute urine and 73.4% and 57.3% for concentrated urine, respectively. Conclusions Urine SG can affect the diagnostic performance of pyuria for detecting UTI and uNGAL might be helpful for identifying UTI regardless of urine SG in young children. Graphical abstract

Abstract

Introduction Pemphigus is a rare and life-threatening autoimmune disease that manifests as blisters and erosions on skin and/or mucosa. It is mediated by autoreactive B cells targeting desmoglein 1 (DSG1) [...], BACKGROUND. Pemphigus, a rare autoimmune bullous disease mediated by antidesmoglein autoantibodies, can be controlled with systemic medication like rituximab and high-dose systemic corticosteroids combined with immunosuppressants. However, some patients continue to experience chronically recurrent blisters in a specific area and require long-term maintenance systemic therapy. METHODS. Skin with chronic blisters was obtained from patients with pemphigus. Immunologic properties of the skin were analyzed by immunofluorescence staining, bulk and single-cell RNA and TCR sequencing, and a highly multiplex imaging technique known as CO-Detection by indEXing (CODEX). Functional analyses were performed by flow cytometry and bulk RNA-Seq using peripheral blood from healthy donors. Intralesional corticosteroid was injected into patient skin, and changes in chronically recurrent blisters were observed. RESULTS. We demonstrated the presence of skin tertiary lymphoid structures (TLSs) with desmoglein-specific B cells in chronic blisters from patients with pemphigus. In the skin TLSs, [CD4.sup.+] T cells predominantly produced CXCL13. These clonally expanded [CXCL13.sup.+][CD4.sup.+] T cells exhibited features of activated Th1-like cells and downregulated genes associated with T cell receptor-mediated signaling. Tregs are in direct contact with [CXCL13.sup.+][CD4.sup.+] memory T cells and increased CXCL13 production of [CD4.sup.+] T cells through IL-2 consumption and TGF-p stimulation. Finally, intralesional corticosteroid injection improved chronic blisters and reduced skin TLSs in patients with pemphigus. CONCLUSION. Through this study we conclude that skin TLSs are associated with the persistence of chronically recurrent blisters in patients with pemphigus, and the microenvironmental network involving [CXCL13.sup.+][CD4.sup.+] T cells and Tregs within these structures plays an important role in CXCL13 production. Abstract

We apply our tetrad-based approach for constructing spherically-symmetric solutions in general relativity to modelling a void, and compare it with the standard Lema\^itre-Tolman-Bondi (LTB) formalism. In particular, we construct models for the void observed in the direction of Draco in the WISE-2MASS galaxy survey, and a corresponding cosmic microwave background (CMB) temperature decrement in the Planck data in the same direction. We find that the present-day density and velocity profiles of the void are not well constrained by the existing data, so that void models produced from the two approaches can differ substantially while remaining broadly consistent with the observations.
