Abstract

We consider a parabolic-elliptic type of Keller-Segel equations with generalized diffusion and logistic source under homogeneous Neumann-Neumann boundary conditions. We construct bounded weak solutions globally in time in an unbalanced optimal transport framework, provided that the magnitude of the chemotactic sensitivity can be restricted depending on parameters. In the case of subquadratic degradation of the logistic source, we quantify the chemotactic sensitivity, in particular, in terms of the power of degradation and the pointwise bound of the initial density., Comment: 30 pages

Abstract

BackgroundHealth care technologies can help improve workers’ health and productivity by supporting workplace health promotion. A personal health record app is used to manage medical data such as results from medical checkups, which facilitates decision making for medical personnel. However, an analysis of users’ technology acceptance is required to provide appropriate services based on personal health record apps. ObjectiveThe purpose of this study was to analyze the factors influencing the behavioral intention of health experts and workers to use an app in workers’ health centers and to examine differences in their perception of the main variables. MethodsThe study involved health experts and workers who visited 21 workers’ health centers in Korea to verify a research model in which perceived risk was added to the unified theory of acceptance and use of technology, a representative theory of information technology acceptance. After receiving ethical approval from the Korea National Institute for Bioethics Policy, 1050 questionnaires were distributed over 7 weeks with cooperation of the Korea Occupational Safety and Health Agency. A multiple linear regression analysis and multigroup path analysis were performed to verify the hypotheses, and independent samples t tests were performed to analyze differences between workers’ and health experts’ perception of the main variables. Abstract

We study porous medium equations with a divergence form of drift terms in a bounded domain with no-flux lateral boundary conditions. We establish $L^q$-weak solutions for $ 1\leq q < \infty$ in Wasserstein space under appropriate conditions on the drift, which is an extension of authors' previous works done in the whole space into the case of bounded domains. Abstract

Obesity is a major public health problem related to various chronic health conditions. Lactobacillus species has been reported in obese individuals; however, its role is unknown. We compared the abundance and composition of Lactobacillus species by analyzing feces from 64 healthy control subjects and 88 obese subjects. We isolated one Lactobacillus strain from the feces of a subject with obesity and further analyzed its genetic and molecular features. We found that an increased abundance and higher prevalence of Lactobacillus sakei distinguished the fecal microbiota of the obese group from that of healthy subjects and that it was related to the increased levels of reactive oxygen species (ROS) induced by higher fat intake. The L. sakei ob4.1 strain, isolated from the feces of a subject with obesity, showed high catalase activity, which was regulated by oxidative stress at the gene transcription level. L. sakei ob4.1 maintained colon epithelial cell adhesion ability under ROS stimulation, and treatment with saturated fatty acid increased colon epithelial ROS levels in a dose-dependent manner; however, L. sakei ob4.1 did not change the level of fat-induced colon epithelial ROS. Exposing mice to a high-fat diet revealed that high-fat-diet-induced colon ROS was associated with the increased colonization of L. sakei ob4.1 through catalase activity. Four-week supplementation with this strain in mice fed a high-fat diet did not change their body weights or ROS levels. A high-fat diet induces changes in the colon environment by increasing ROS levels, which provides a colonization benefit to an L. sakei strain with high catalase activity. IMPORTANCELactobacillus provides many health benefits; its various species are widely used as probiotics. However, an increased abundance of Lactobacillus has been reported in obesity, and the role of Lactobacillus strains in obesity remains unknown. We found a high abundance of the Lactobacillus sakei species in a group of obese subjects and examined its relationship with a high-fat diet and reactive oxygen species (ROS) in the feces. To find the underlying mechanism, we analyzed and characterized an L. sakei strain isolated from a severely obese individual. We found that higher gut oxidative stress could link high-fat-diet-induced obesity and L. sakei. This translational research identifies the roles of the host gut environment in the colonization and survival of L. sakei.

Abstract

We are concerned with large-time behaviors of solutions for Vlasov--Navier--Stokes equations in two dimensions and Vlasov-Stokes system in three dimensions including the effect of velocity alignment/misalignment. We first revisit the large-time behavior estimate for our main system and refine assumptions on the dimensions and a communication weight function. In particular, this allows us to take into account the effect of the misalignment interactions between particles. We then use a sharp heat kernel estimate to obtain the exponential time decay of fluid velocity to its average in $L^\infty$-norm. For the kinetic part, by employing a certain type of Sobolev norm weighted by modulations of averaged particle velocity, we prove the exponential time decay of the particle distribution, provided that local particle distribution function is uniformly bounded. Moreover, we show that the support of particle distribution function in velocity shrinks to a point, which is the mean of averaged initial particle and fluid velocities, exponentially fast as time goes to infinity. This also provides that for any $p \in [1,\infty]$, the $p$-Wasserstein distance between the particle distribution function and the tensor product of the local particle distributions and Dirac measure at that point in velocity converges exponentially fast to zero as time goes to infinity.

Abstract

Cancer cells utilize epigenetic alterations to acquire autonomous capabilities for tumor maintenance. Here, we show that pancreatic ductal adenocarcinoma (PDA) cells utilize super-enhancers (SEs) to activate the transcription factor EVI1 (ecotropic viral integration site 1) gene, resulting in activation of an EVI1-dependent transcription program conferring PDA tumorigenesis. Our data indicate that SE is the vital cis-acting element to maintain aberrant EVI1 transcription in PDA cells. Consistent with disease progression and inferior survival outcomes of PDA patients, we further show that EVI1 upregulation is a major cause of aggressive tumor phenotypes. Specifically, EVI1 promotes anchorage-independent growth and motility in vitro and enhances tumor propagation in vivo. Mechanistically, EVI1-dependent activation of tumor-promoting gene expression programs through the stepwise configuration of the active enhancer chromatin attributes to these phenotypes. In sum, our findings support the premise that EVI1 is a crucial driver of oncogenic transcription programs in PDA cells. Further, we emphasize the instructive role of epigenetic aberrancy in establishing PDA tumorigenesis.
