Imaging infections in patients is challenging using conventional methods, motivating the development of positron emission tomography (PET) radiotracers targeting bacteria-specific metabolic pathways. Numerous techniques have focused on the bacterial cell wall, although peptidoglycan-targeted PET tracers have been generally limited to the short-lived carbon-11 radioisotope (t1/2 = 20.4 min). In this article, we developed and tested new tools for infection imaging using an amino sugar component of peptidoglycan, namely, derivatives of N-acetyl muramic acid (NAM) labeled with the longer-lived fluorine-18 (t1/2 = 109.6 min) radioisotope. Muramic acid was reacted directly with 4-nitrophenyl 2-[18F]fluoropropionate ([18F]NFP) to afford the enantiomeric NAM derivatives (S)-[18F]FMA and (R)-[18F]FMA. Both diastereomers were easily isolated and showed robust accumulation by human pathogens in vitro and in vivo, including Staphylococcus aureus. These results form the basis for future clinical studies using fluorine-18-labeled NAM-derived PET radiotracers.

Pepper (Capsicum annuum) is an important vegetable crop that has been subjected to intensive breeding, resulting in limited genetic diversity, especially for sweet peppers. Previous studies have reported pepper draft genome assemblies using short read sequencing, but their capture of the extent of large structural variants (SVs), such as presence-absence variants (PAVs), inversions, and copy-number variants (CNVs) in the complex pepper genome falls short. In this study, we sequenced the genomes of representative sweet and hot pepper accessions by long-read and/or linked-read methods and advanced scaffolding technologies. First, we developed a high-quality reference genome for the sweet pepper cultivar Dempsey and then used the reference genome to identify SVs in 11 other pepper accessions and constructed a graph-based pan-genome for pepper. We annotated an average of 42 972 gene families in each pepper accession, defining a set of 19 662 core and 23 115 non-core gene families. The new pepper pan-genome includes informative variants, 222 159 PAVs, 12 322 CNVs, and 16 032 inversions. Pan-genome analysis revealed PAVs associated with important agricultural traits, including potyvirus resistance, fruit color, pungency, and pepper fruit orientation. Comparatively, a large number of genes are affected by PAVs, which is positively correlated with the high frequency of transposable elements (TEs), indicating TEs play a key role in shaping the genomic landscape of peppers. The datasets presented herein provide a powerful new genomic resource for genetic analysis and genome-assisted breeding for pepper improvement.

High‐performance 0D–2D hybrid photodetectors integrated with a crosslinker for direct pattering of quantum dots on the large‐scale synthesized MoS2 layer are reported. In the patterned hybrid structure, QD layers are patterned with a resolution of up to 2 µm, ensuring high precision. Enhanced charge transfer from QDs to 2D materials is confirmed using PL quenching, TR‐PL, and UPS analysis. As a result, the QD/2D hybrid photodetectors with crosslinker‐assisted direct patterning demonstrated a remarkable photoresponsivity of ≈105 A W−1 and a specific detectivity of over 1011 Jones, attributed to the difference in built‐in potential. The crosslinker patterning of QDs opens up potential applications for the photodetectors in highly integrated image sensors and can be further extended to high‐resolution display industries, eliminating unnecessary fabrication processes. Distant metastasis, the leading cause of cancer death, is efficiently kept in check by immune surveillance. Studies have uncovered peripheral natural killer (NK) cells as key antimetastatic effectors and their dysregulation during metastasis. However, the molecular mechanism governing NK cell dysfunction links to metastasis remains elusive. Herein, MAP4K1 encoding HPK1 is aberrantly overexpressed in dysfunctional NK cells in the periphery and the metastatic site. Conditional HPK1 overexpression in NK cells suffices to exacerbate melanoma lung metastasis but not primary tumor growth. Conversely, MAP4K1‐deficient mice are resistant to metastasis and further protected by combined immune‐checkpoint inhibitors. Mechanistically, HPK1 restrains NK cell cytotoxicity and expansion via activating receptors. Likewise, HPK1 limits human NK cell activation and associates with melanoma NK cell dysfunction couples to TGF‐β1 and patient response to immune checkpoint therapy. Thus, HPK1 is an intracellular checkpoint controlling NK‐target cell responses, which is dysregulated and hijacked by tumors during metastatic progression. [ABSTRACT FROM AUTHOR]

A major goal of education is to help students retain as much meaningful information as possible for long periods of time. The learning process by which students acquire new knowledge and behaviors is closely related to memory, which is the core of cognition. In a previous study, we developed virtual reality visualization model (VRVM) that can be applied to address difficulties with learning medical biochemistry. Validation and evaluation studies are necessary to assess the usefulness of VRVM. The purpose of the present study is to apply VRVM and evaluate its effectiveness in a real educational environment. We modeled and verified the concept of spatiotemporality of information obtained through VRVM, which augments memories in linkage with hippocampal-centered networks while helping students to transition to higher-order thinking. The effectiveness of VRVM of the tricarboxylic acid (TCA) cycle, an onerous topic for medical students, was compared with that of traditional educational methods. The experimental group used VRVM to learn the TCA cycle, while the control group learned from PowerPoint (Microsoft Co., Redmond, WA, USA) presentations using 2-dimensional (2D) diagrams. Evaluations of short-term and long-term memory indicated that learning using VRVM was more effective than learning through traditional lecture-based methods, and that VRVM resulted in better recall compared to traditional methods. Our findings suggest that VRVM can be applied when teaching students about the complicated TCA cycle, and that it helps medical students to improve their recall of carbohydrate metabolism in the context of medical biochemistry studies.
