18. Tumor Microenvironment Prognostic Risk and Its Association With MUC5AC in Ampullary Carcinoma

Abstract

Context.--The tumor-host interaction in the tumor microenvironment (TME) affects the prognosis of patients with malignant tumors. TME assessed via tumor budding (BD) and tumor-infiltrating lymphocyte (TIL) had a prognostic impact in patients with nonampullary small intestinal and colorectal carcinomas. In ampullary carcinoma (AC), MUC5AC was recently revealed as a significant prognosticator, but studies about the TME have not been conducted. Objective.--To assess TME-based prognostic risk in AC. Design.--We generated a collective TME risk index based on high-grade BD at the invasive front (BD3) and high density of stromal-TIL (>5%) in 64 surgically resected ACs. We evaluated its predictive values for overall survival (OS) and recurrence-free survival (RFS). We also investigated the relationship of TME to MUC5AC expression. Results.--TME prognostic risk index was classified into low-risk ([BD.sup.Low]/[TIL.sup.High]; 26 of 64; 41%), intermediate-risk ([BD.sup.Low]/[TIL.sup.Low] or [BD.sup.High]/[TIL.sup.High]; 23; 36%), and high-risk ([BD.sup.High]/[TIL.sup.Low]; 15; 23%) groups. Higher TME prognostic risk was associated with higher tumor grade (P = .03), lymphovascular invasion (P = .05), and MUC5AC immunopositivity (P = .02). TME prognostic risk index displayed better predictive ability for both OS (53.9 versus 46.1 versus 42.2) and RFS (24.8 versus 16.9 versus 15.3) than BD or TIL alone. In multivariate analysis, TME prognostic risk index was an independent prognosticator for OS (P = .003) and RFS (P = .03). Conclusions.--TME risk index in combination with BD and TIL was a stronger predictor of prognostic risk stratification than either BD or TIL alone for both OS and RFS in patients with AC. Abstract

Magnetic skyrmions are swirling magnetic textures with novel characteristics suitable for future spintronic and topological applications. Recent studies confirmed the room-temperature stabilization of skyrmions in ultrathin ferromagnets. However, such ferromagnetic skyrmions show undesirable topological effect, the skyrmion Hall effect, which leads to their current-driven motion towards device edges, where skyrmions could easily be annihilated by topographic defects. Recent theoretical studies have predicted enhanced current-driven behaviour for antiferromagnetically exchange-coupled skyrmions. Here we present the stabilization of these skyrmions and their current-driven dynamics in ferrimagnetic GdFeCo films. By utilizing element-specific X-ray imaging, we find that the skyrmions in the Gd and FeCo sublayers are antiferromagnetically exchange-coupled. We further confirm that ferrimagnetic skyrmions can move at a velocity of ~50 m s-1 with reduced skyrmion Hall angle, {\theta}SkHE ~20{\deg}. Abstract

In this study, a 4D trajectory weather (4DT-Wx) prototype system was developed and evaluated for effective weather information integration in trajectory-based operation (TBO) environments. The system has two key distinguishing features: multi-model-based trajectory services and buffer zone information provision. We constructed a distributed processing system using Apache Spark, enabling the efficient processing of large-scale weather data. The performance evaluation demonstrated excellent scalability and efficiency in processing large-scale data. An analysis of the buffer configurations highlighted that buffer zone information is valuable in decision-making processes and has the potential to enhance the system performance. The system's practical applicability is presented through visualizations of the extracted weather information. This system is expected to enhance aviation safety and operational efficiency, providing a foundation for addressing increasingly complex weather conditions and flight scenarios in the future. The approach presented in this study marks a significant step toward effective TBO implementation and the advancement of future air traffic management. The evaluation of the 4DT-Wx system analyzed the accuracy of weather data processing and the performance of distributed processing, finding that the temperature (T) estimation had the highest accuracy, and that the parallel processing using Apache Spark was most effectively modeled by Ahmed et al.'s model. The findings suggest the potential for further optimization in integrating various weather models and developing algorithms to enhance their utilization. [ABSTRACT FROM AUTHOR]
