3. Transmission IR Microscopy for the Quantitation of Biomolecular Mass In Live Cells

Abstract

Absolute quantity imaging of biomolecules on a single cell level is critical for measurement assurance in biosciences and bioindustries. While infrared (IR) transmission microscopy is a powerful label-free imaging modality capable of chemical quantification, its applicability to hydrated biological samples remains challenging due to the strong water absorption. We overcome this challenge by applying a solvent absorption compensation (SAC) technique to a home-built quantum cascade laser IR microscope. SAC-IR microscopy improves the chemical sensitivity considerably by adjusting the incident light intensity to pre-compensate the IR absorption by water while retaining the full dynamic range. We demonstrate the label-free chemical imaging of key biomolecules of a cell, such as protein, fatty acid, and nucleic acid, with sub-cellular spatial resolution. By imaging live fibroblast cells over twelve hours, we monitor the mass change of the three molecular species of single cells at various phases, including cell division. While the current live-cell imaging demonstration involved three wavenumbers, more wavenumber images could measure more biomolecules in live cells with higher accuracy. As a label-free method to measure absolute quantities of various molecules in a cell, SAC-IR microscopy can potentially become a standard chemical characterization tool for live cells in biology, medicine, and biotechnology., Comment: Body: 19 pages, 5 figures. Abstract

The behavior of airport precast concrete pavement (APCP) involving new design and construction concepts was experimentally analyzed under environmental and moving airplane loads, and the long-term performance of the APCP was evaluated using fatigue failure analysis. The strain characteristics and curling behavior of the APCP under environmental loads were comprehensively analyzed. The APCP slabs exhibited a pronounced curling phenomenon similar to conventional concrete pavement slabs. The dynamic response of the APCP subjected to impact loads was analyzed by performing heavy weight deflectometer tests. The test results confirmed that the vertical deformation of the APCP was small and within the typical range of vertical deformation of conventional concrete pavement. The dynamic strain response of the APCP under moving airplane loads was then analyzed and the strain variation during day and night times was compared. The strains during the day were found to be significantly larger than those at night under airplane loads because of the curling phenomenon of the APCP slabs. Finally, the long-term performance of the APCP was evaluated using fatigue failure analysis based on the obtained behavior. Even using the most conservative fatigue failure prediction model, the service life of the APCP was ascertained to be more than 30 years. Based on the overall results of this study, it is concluded that the APCP, which is designed to reduce slab thickness by placing reinforcing bars in the slabs via reinforced concrete structural design, exhibits typical behavior of concrete pavements and can be successfully applied to airport pavement rehabilitation. Abstract

Implantable electrical neurostimulators offer a promising avenue for treating neurological disorders. However, their dependency on a finite battery life limits their long‐term utility. Emerging transcutaneous ultrasound‐driven triboelectric nanogenerator (TENG) techniques provide solutions for converting external ultrasound waves into internal electricity. This study proposes an implantable ultrasound‐driven TENG (IU‐TENG) using polyether ether ketone (PEEK) for its exceptional stability inside a human body and acoustic impedance compatibility with human tissues. This IU‐TENG remarkably surpasses traditional titanium‐based encapsulation, resulting in a 99.94% efficiency in ultrasound transmission. In addition, PEEK contains numerous electron‐donating functional groups, making it suitable for TENG applications, particularly as a positive triboelectric layer. The device exhibits robust voltage outputs, reaching up to 11.50 and 8.75 V in water and in vivo, respectively, under body‐safe ultrasound intensities. Moreover, its ability to sustain a stable electrical output for over 300 min emphasizes the durability and mechanical resilience of PEEK. In vivo mouse models and ex vivo porcine tissue trials demonstrate the effectiveness of the IU‐TENG in nerve stimulation, showing its potential in medical treatments, enhancing the functionality and longevity of implantable medical devices. Abstract

In electron microscopy, charging of non-conductive biological samples by focused electron beams hinders their high-resolution imaging. Gold or platinum coatings have been commonly used to prevent such sample charging, but it disables further quantitative and qualitative chemical analyses by energy dispersive spectroscopy (EDS). Here we report that graphene-coating on biological samples enables non-destructive high-resolution imaging by scanning electron microscopy (SEM) as well as chemical analysis by EDS, utilizing graphene's transparency to electron beams, high conductivity, outstanding mechanical strength, and flexibility. We believe that the graphene-coated imaging and analysis would provide us a new opportunity to explore various biological phenomena unseen before due to the limitation in sample preparation and image resolution, which will broaden our understanding on the life mechanism of various living organisms.

Abstract

Background: The health‐related quality of life instrument with 8 items (HINT‐8) was developed to measure health‐related quality of life (HRQoL) in Korea. However, the HINT‐8 has not yet been validated among the family caregivers of people with dementia (PwD). Design: A cross‐sectional pilot study. Objective: The study aimed to examine the convergent and discriminant validity of the HINT‐8 among family caregivers of individuals with dementia. Sample: Forty‐seven family caregivers of PwD. Measurements: HINT‐8 was compared with the 5‐level EQ‐5D (EQ‐5D‐5L) to assess its convergent and discriminant validity. Additionally, the association between the two instruments assessing HRQoL was examined using the short‐form Bédard‐Zarit Burden Interview (SZBI). Results: The HINT‐8 was a promising and valid HRQoL instrument for family caregivers of PwD. There was a significantly high correlation between the overall HINT‐8 and EQ‐5D‐5L indices (r = 0.85, p <.001). The HINT‐8 had acceptable psychometric properties compared to the commonly used EQ‐5D‐5L, as indicated by the subdomains associated with family caregiversʼ burden measured by the SZBI. Conclusion: Future studies should compare the HINT‐8 with existing dementia carer‐specific QoL instruments among a larger study sample to enhance its statistical power and confirm its reliability and structural validity.
