Bu tez çalışmasında gıda endüstrisindeki temel sorunlardan biri olan, muz, elma ve yer elması meyvelerinde bulunan polifenol oksidaz enziminin neden olduğu esmerleşme reaksiyonunun inhibisyonu incelenmiştir. Literatürde gerçekleştirilen tek substratlı inhibisyon kinetiği çalışmalarının aksine, endüstriyel üretimde meyvenin kendi substratları ve eklenen inhibitörün bulunduğu ortamla karşılaşılacağından çok substratlı inhibisyon çalışması gerçekleştirilmiştir. Katı ve sıvı faz olarak iki ana hatta yapılan çalışmada inhibitör olarak gallik asit veya kuersetin kullanılmıştır. Seçilen inhibitörlerin aynı zamanda antimikrobiyal özelliğe sahip olmalarına dayanarak tez çalışmasında katı fazda mikrobiyal büyüme inhibisyonu da çalışılmıştır. Katı faz inhibisyon deneylerinde hem meyvenin kendine has renginin korunması hem de mikrobiyal bozulmanın engellenmesine neden olabilecek konsantrasyonların belirlenmesi gerçekleştirilmiştir. Sıvı fazda ise spektrofotometrik ve kolorimetrik analizlerle ortamdaki renk değişimleri üzerine odaklanılmış ve meyve ortamlarının doğal renginin korunmasını sağlayacak inhibitör derişimleri ve ortam koşulları belirlenmiştir. Ayrıca olası reaksiyon mekanizmaları da ortaya konulmuştur. Düşük, orta ve yüksek derişimlerde kullanılan inhibitör türlerinden, kuersetinin kararma reaksiyonunun, gallik asitin ise genellikle mikrobiyal inhibisyonun sağlanmasında daha etkin rol oynadığı belirlenmiştir. Seçilen tüm meyvelerde yüksek derişimde kuersetin kullanımının pürelerin doğal rengini koruyabildiği belirlenmiştir. Seçilen inhibitör türü ve derişimlerinin tamamının, bir gün süreyle mikrobiyal inhibisyon sağladığı tespit edilmiştir. Düşük derişimde gallik asit kullanımı elma pürelerinde %90, yüksek derişimde kullanımı ise yer elması pürelerinde %68 inhibisyona neden olmuştur. Muzda ise maksimum mikrobiyal inhibisyon yüksek derişimde kuersetin kullanımıyla %96 oranında gerçekleşmiştir.Sıvı faz inhibisyon deneylerinde spektrofotometrik analiz bulguları, hem enzim kaynağının hem de meyvenin substrat bileşimlerinin farklı olmasından kaynaklı olarak değişen konsantrasyonlarda da olsa, spesifik bir derişim değerine kadarki bölgede gerek gallik asitin, gerekse kuersetinin PFO enzimini inhibe ettiğini göstermiştir. Bu bölgede artan gallik asit derişiminin inhibisyon oranını arttırması yarışmalı inhibisyonun varlığını işaret etmektedir. Spesifik değerlerin üstündeki belli bir aralıktaki derişim değerlerinde giderek azalan bir inhibisyon, bu aralık da aşıldığında ise ortamda ikinci bir reaksiyonun gerçekleştiği tespit edilmiştir. Bu durum, elmada "eğilim ve renk farklılığı", muzda sadece "eğilim farklılığı" şeklinde belirmekte, yer elmasında ise spesifik aralık bölgesinde kalındığından bu farklılık gözlenmemektedir. Elma ve muz ortamında gözlenen bu ikinci reaksiyon oksijenden bağımsızken, yer elması ortamında oksijenin de bu reaksiyonun substratlarından biri olduğu belirlenmiştir. Kuersetin inhibisyonunun spektrofotometrik analiz bulgularında da farklı konsantrasyon değerleriyle benzer sonuçlar elde edilmiştir. Ancak, artan kuersetin derişimlerinin oluşturduğu renklerin benzer tonlarda olması, kullanılan derişimlerin spesifik aralık bölgesi içerisinde kaldığını göstermektedir. En yüksek inhibisyon oluşumunu sağlayan derişimlerin elma ve yer elması için 100 µl, muz için ise 50 µl ve 100 µl olduğu sonucuna varılmıştır. Sıvı ortamdaki gallik asit ve kuersetin inhibisyonu karşılaştırıldığında, elma ve muz gibi nisbeten daha yavaş ve oksijenden bağımsız reaksiyon ortamlarında daha az miktarda kuersetin kullanılarak etkili bir inhibisyonun sağlanabileceği tespit edilmiştir. Yer elmasında olduğu gibi oksijen bağımlı reaksiyon ortamlarında ise gallik asit seçilecekse düşük, kuersetin seçilecekse nisbeten daha yüksek derişimlerin vakum ortamıyla desteklenmesi durumunda, ortamların kendi renginin korunabileceği sonucuna varılmıştır. Sonuç olarak, seçilen meyvelerdeki mevcut polifenoller ve PFO enzimini içeren ortamlara farklı konsantrasyonlarda inhibitör eklenmesinin neden olduğu reaksiyonlar için iki olası reaksiyon mekanizması öngörülmektedir. İlk mekanizmada, inhibitörsüz ortamda doğal substratı olan polifenolleri oksitleyen PFO kahverengi renkte ürün oluşturmakta, ortama kritik bir konsantrasyon değerine kadar inhibitör eklenmesi durumunda ise eklenen inhibitörle polifenoller arasında PFO enziminin aktif bölgesine bağlanmak adına bir yarış gerçekleşmekte ve inhibitör bağlanması durumunda kahverengi renkteki ürün oluşamamaktadır. Ancak ortamdaki inhibitör derişimi belli bir aralığın da üstüne ulaştığında PFO-inhibitör kompleksinin muhtemelen farklı aktif bölgelerine inhibitörlerin de bağlanmasıyla ikinci bir reaksiyon gerçekleşmekte ve yeşil renkte bir ürün ortaya çıkmaktadır. İkinci mekanizma ise, substrat ve inhibitörün enzimin aktif bölgelerine bağlanma sırasıyla değişen reaksiyonların gözlendiği bir ping-pong mekanizmasıdır. Dolayısıyla ortamda bir renk karışımı oluşmakta ve baskın olan reaksiyonun rengi gözle görünür boyuta ulaşmaktadır. Çalışmada, kolorimetrik analiz bulgularıyla üretilen altıncı derece polinom fonksiyonların üretilen reaksiyon mekanizmalarını desteklediği, ancak ortamda oluşan rengin öngörülmesinde yetersiz kaldığı tespit edilmiştir. Özellikle renk saçılması ve toplam renk değişimine ait polinomlarla üretilen bulguların, ortamdaki renk oluşumunu ifade etme ve spektrofotometrik bulguları desteklemedeki başarısı, diğer renk ifadelerinden daha yüksektir. Bu nedenle, çoklu substrat ve inhibitör varlığındaki enzim reaksiyonlarında kolorimetrik analiz bulgularıyla üretilen polinomlarda bu ifadelerin tercih edilmesinin gerekliliği ortaya konulmuştur. Yapılan diğer iki ve üç boyutlu grafik incelemelerinde de kolorimetrik sonuçların spektrofotometrik bulgular ve ortam görselleri ile kısmen uyumlu veya tamamen uyumsuz bulgular üretmesinden yola çıkarak, spektrofotometrik incelemeler yapılması, hem ortamın rengini ortaya koyan görseller, hem de enzim mekanizmasını açıklama açısından kolorimetrik analizden çok daha başarılı bulunmuştur. Anahtar Kelimeler: Polifenol oksidaz enzimi, inhibisyon, tekli ve çoklu substrat, gallik asit ve kuersetin., In this thesis, the inhibition of browning reaction caused by polyphenol oxidase enzyme found in banana, apple and jerusalem artichoke, which is one of the main problems in food industry, was investigated. In contrast to the single substrate inhibition kinetic studies conducted in the literature, the investigation of inhibition via multiple substrate and/or inhibitor was carried out, since the fruit's own substrates and added inhibitor in the medium will be encountered in industrial production. In the study conducted in two main lines as solid and liquid phase, gallic acid or quercetin was used as inhibitors. Based on the fact that the selected inhibitors also have antimicrobial properties, inhibition of microbial growth in solid phase was also studied in the thesis. In the solid phase inhibition experiments, the determination of the concentrations that could cause both the preservation of the natural color of the fruit and the prevention of microbial spoilage were carried out. In the liquid phase, spectrophotometric and colorimetric analysis focused on the color changes in the medium, and the inhibitor concentrations and ambient conditions that will ensure the preservation of the natural color of the fruit environment were determined. Possible reaction mechanisms were also revealed. It has been determined that among the inhibitor types used in low, medium and high concentrations, quercetin plays a more effective role in providing the blackening reaction and gallic acid generally plays a more effective role in providing microbial inhibition. It has been determined that the use of high concentrations of quercetin in all selected fruits can preserve the natural color of purees. All of the selected inhibitor types and concentrations were found to provide microbial inhibition for one day. The use of gallic acid at low concentration caused 90% inhibition in apple puree, while its use in high concentration caused an inhibition of 68% in jerusalem artichoke puree. In bananas, the maximum microbial inhibition was 96% with the use of high concentration quercetin.Spectrophotometric analysis findings in liquid phase inhibition experiments showed that both gallic acid and quercetin inhibited the PFO enzyme in the region up to a specific concentration value, albeit at varying concentrations due to the different enzyme source and the substrate composition of the fruit. Increasing the inhibition rate of increased gallic acid concentration in this region indicates the presence of competitive inhibition. It was determined that there was a gradually decreasing inhibition at concentration values in a certain range above the specific values, and when this range was exceeded, a second reaction took place in the environment. This situation appears as "trend and color difference" in apple and only as "trend difference" in banana, whereas in jerusalem artichoke, this difference is not observed since it is in the specific range region. While this second reaction observed in apple and banana environment is independent of oxygen, it has been determined that oxygen in Jerusalem artichoke environment is one of the substrates of this reaction. Similar results were obtained with different concentration values in spectrophotometric analysis findings of quercetin inhibition. However, the fact that the colors created by the increasing concentrations of quercetin are similar tones, indicating that the concentrations used remain within the specific range. It was concluded that the concentrations providing the highest inhibition were 100µl for apple and Jerusalem artichoke, 50 µl and 100 µl for banana. When the inhibition of gallic acid and quercetin in liquid medium was compared, it was found that an effective inhibition could be achieved by using less amount of quercetin in relatively slow and oxygen-free reaction media such as apple and banana. It has been concluded that if gallic acid is selected, relatively higher concentrations of gallic acid are selected, and relatively higher concentrations are supported by a vacuum environment in oxygen-dependent reaction environments such as jerusalem artichoke, it is concluded that the color of the environments can be preserved. As a result, two possible reaction mechanisms are predicted for reactions caused by adding different concentrations of inhibitor to media containing PFO enzyme and existing polyphenols in selected fruits. In the first mechanism, PFO, which oxidizes the natural substrate polyphenols in the environment without inhibitor, creates a brown colored product, and if inhibitors are added to the environment up to a critical concentration value, a race takes place between the added inhibitor and the polyphenols to bind to the active site of the PFO enzyme, and if the inhibitor is attached, the brown colored product cannot be formed. However, when the inhibitor concentration in the environment reaches a certain range, a second reaction takes place, possibly by binding the inhibitors to different active sites of the PFO-inhibitor complex, resulting in a green product. The second mechanism is a ping-pong mechanism in which changes in the binding of substrate and inhibitor to active sites of the enzyme are observed. Therefore, a mixture of colors is formed in the environment and the color of the dominant reaction reaches a visible dimension. In the study, it was determined that sixth order polynomial functions produced by the colorimetric analysis findings support the reaction mechanisms produced, but are insufficient in predicting the color formed in the environment. The success of the findings produced by polynomials of color scattering and total color change in expressing the color formation in the environment and supporting spectrophotometric findings is higher than other color expressions. Therefore, the necessity of using these expressions in polynomials produced by colorimetric analysis findings in enzyme reactions in the presence of multiple substrates and inhibitors has been revealed. In other two and three dimensional graphic examinations, based on the fact that the colorimetric results produce findings that are partially or completely incompatible with spectrophotometric findings and ambient images, spectrophotometric examinations have been found to be much more successful than colorimetric analysis in terms of both visuals revealing the color of the environment and explaining the enzyme mechanism.
