3. Effect of expansion of human umbilical cord blood CD34 + cells on neurotrophic and angiogenic factor expression and function

Abstract

AbstractThe use of CD34 + cell-based therapies has largely been focused on haematological conditions. However, there is increasing evidence that umbilical cord blood (UCB) CD34 + -derived cells have neuroregenerative properties. Due to low cell numbers of CD34 + cells present in UCB, expansion is required to produce sufficient cells for therapeutic purposes, especially in adults or when frequent applications are required. However, it is not known whether expansion of CD34 + cells has an impact on their function and neuroregenerative capacity. We addressed this knowledge gap in this study, via expansion of UCB-derived CD34 + cells using combinations of LDL, UM171 and SR-1 to yield large numbers of cells and then tested their functionality. CD34 + cells expanded for 14 days in media containing UM171 and SR-1 resulted in over 1000-fold expansion. The expanded cells showed an up-regulation of the neurotrophic factor genes BDNF, GDNF, NTF-3 and NTF-4, as well as the angiogenic factors VEGF and ANG. In vitro functionality testing showed that these expanded cells promoted angiogenesis and, in brain glial cells, promoted cell proliferation and reduced production of reactive oxygen species (ROS) during oxidative stress. Collectively, this study showed that our 14-day expansion protocol provided a robust expansion that could produce enough cells for therapeutic purposes. These expanded cells, when tested in in vitro, maintained functionality as demonstrated through promotion of cell proliferation, attenuation of ROS production caused by oxidative stress and promotion of angiogenesis.

Abstract

Recent spinal cord literature abounds with descriptions of genetic preprogramming and the molecular control of circuit formation. In this paper, we explore to what extent circuit formation based on learning rather than preprogramming could explain the selective formation of the monosynaptic projections between muscle spindle primary afferents and homonymous motoneurons. We adjusted the initially randomized gains in the neural network according to a Hebbian plasticity rule while exercising the model system with spontaneous muscle activity patterns similar to those observed during early fetal development. Normal connectivity patterns developed only when we modeled β motoneurons, which are known to innervate both intrafusal and extrafusal muscle fibers in vertebrate muscles but were not considered in previous literature regarding selective formation of these synapses in animals with paralyzed muscles. It was also helpful to correctly model the greatly reduced contractility of extrafusal muscle fibers during early development. Stronger and more coordinated muscle activity patterns such as observed later during neonatal locomotion impaired projection selectivity. These findings imply a generic functionality of a musculoskeletal system to imprint important aspects of its mechanical dynamics onto a neural network, without specific preprogramming other than setting a critical period for the formation and maturation of this general pattern of connectivity. Such functionality would facilitate the successful evolution of new species with altered musculoskeletal anatomy, and it may help to explain patterns of connectivity and associated reflexes that appear during abnormal development.

10. Collagen immunoassay as a method to optimise surface functionalisation

Abstract

Traditional methods of assessing surface functionalisation, including spectroscopy and chemical labelling, often involve significant error and conjecture about bonds. Proteins that improve cell attachment have specific pKa's and optimum binding requirements that may differ from the conditions required for chemical labelling. The utility of collagen ELISA to optimise acetaldehyde glow discharge polymerisation reactor parameters was tested. Accurate stepwise increases in collagen conjugation strength were demonstrated by incubating specimens in 8 M urea for 5–8 days followed by ELISA to test for residual surface collagen. Surface modifications also were assessed by XPS. The results indicated that ELISA after bond‐stressing with urea may suffice for optimising surface functionalisation and that traditional methods of analysis may be superfluous if protein conjugation is the aim of functionalisation.

Abstract

The electrical and magnetic properties of light rare-earth metals and trans-plutonium actinide metals are of interest to study the f-shell delocalization phenomenon under high compressions. Using designer diamond anvil technology, sensitive electrical four-probe measurements were performed on light rare-earth metal praseodymium to pressures of 179 GPa at room temperature. We document an average drop in resistivity of 53% at a pressure of 20 GPa in a series of high-pressure experiments. This large drop in resistivity provides the strongest experimental evidence yet for the view that the 20 GPa phase transition is indeed associated with f-electron delocalization. Our results show that the precise electrical measurements are ideally suited for f-delocalization studies, especially where structural data do not provide clear evidence of this transition.

16. ORP3 splice variants and their expression in human tissues and hematopoietic cells

Abstract

ORP3 is a member of the newly described family of oxysterol-binding protein (OSBP)-related proteins (ORPs). We previously demonstrated that this gene is highly expressed in CD34+ hematopoietic progenitor cells, and deduced that the "full-length" ORP3 gene comprises 23 exons and encodes a predicted protein of 887 amino acids with a C-terminal OSBP domain and an N-terminal pleckstrin homology domain. To further characterize the gene, we cloned ORP3 cDNA from PCR products and identified multiple splice variants. A total of eight isoforms were demonstrated with alternative splicing of exons 9, 12, and 15. Isoforms with an extension to exon 15 truncate the OSBP domain of the predicted protein sequence. In human tissues there was specific isoform distribution, with most tissues expressing varied levels of isoforms with the complete OSBP domain; while only whole brain, kidney, spleen, thymus, and thyroid expressed high levels of the isoforms associated with the truncated OSBP domain. Interestingly, the expression in cerebellum, heart, and liver of most isoforms was negligible. These data suggest that differential mRNA splicing may have resulted in functionally distinct forms of the ORP3 gene.

42. Ellipsometric Study of Monodisperse Silica Particles at an Oil−Water Interface

Abstract

Results are reported for ellipsometric measurements of hydrophobized monodisperse silica particles, with a diameter of about 25 nm, spread at the toluene−water interface. Theoretical values for the ellipsometric parameters are derived by treating the particles as a core−shell model and performing integrations of the refractive index profile through the interface using Drude's equations. With justifiable choices of the fixed parameters for the system, the agreement is good between measured and calculated values for the ellipsometric parameter Δ as a function of the amount of silica particles added to the interface. However, the results at high particle concentration at the interface are consistent either with coverage greater than a close-packed monolayer or with a monolayer with corrugations whose amplitude is less than the radius of the particles. The results show that this is not a suitable method for the determination of the contact angle of the particles at the oil−water interface.

The Pivot IBM-compatible portable computer from Morrow Designs features fully-loaded, 512K of RAM, two 5.25-inch floppy disk drives, an internal 300 baud direct-connect modem, a 16-line LCD screen, and Centronics and RS-232 ports. It also features built-in software which includes calculator, communications and calendar programs. It runs MS-DOS, weighs ten pounds, and is battery-operated. Pivot comes bundled with a WordStar clone called NewWord. The Pivot package lists for $2,895. It is an impressive computer that runs a lot of IBM PC software without any modification. Its few limitations include its display, which Morrow is improving, and its expansion capabilities. Three photographs of the computer are included. A table summarizes specifications.
