Abstract The duality of Jackiw-Teitelboim (JT) gravity and a double scaled matrix integral has led to studies of the canonical spectral form factor (SFF) in the so called τ−scaled limit of large times, t → ∞, and fixed temperature, in order to demonstrate agreement with universal random matrix theory (RMT). Though this has been established for the unitary case, extensions to other symmetry classes requires the inclusion of unorientable manifolds in the sum over geometries, necessary to address time reversal invariance, and regularization of the corresponding prime geometrical objects, the Weil-Petersson (WP) volumes. We report here how universal signatures of quantum chaos, witnessed by the fidelity to the Gaussian orthogonal ensemble, emerge for the low-energy limit of unorientable JT gravity, i.e. the unorientable Airy model/topological gravity. To this end, we implement the loop equations for the corresponding dual (double-scaled) matrix model and find the generic form of the unorientable Airy WP volumes, supported by calculations using unorientable Kontsevich graphs. In an apparent violation of the gravity/chaos duality, the τ−scaled SFF on the gravity side acquires both logarithmic and power law contributions in t, not manifestly present on the RMT side. We show the expressions can be made to agree by means of bootstrapping-like relations hidden in the asymptotic expansions of generalized hypergeometric functions. Thus, we are able to establish strong evidence of the quantum chaotic nature of unorientable topological gravity.

This work develops a quantum control application of many-body quantum chaos for ultracold bosonic gases trapped in optical lattices. It is long known how to harness exponential sensitivity to changes in initial conditions for control purposes in classically chaotic systems. In the technique known as targeting , instead of a hindrance to control, the instability becomes a resource. Recently, this classical targeting has been generalized to quantum systems either by periodically countering the inevitable quantum state spreading or by introducing a control Hamiltonian, where both enable localized states to be guided along special chaotic trajectories toward any of a broad variety of desired target states. Only strictly unitary dynamics are involved; i.e. it gives a coherent quantum targeting . In this paper, the introduction of a control Hamiltonian is applied to Bose–Hubbard systems in chaotic dynamical regimes. Properly selected unstable mean field solutions can be followed particularly rapidly to states possessing precise phase relationships and occupancies. In essence, the method generates a quantum simulation technique that can access rather special states. The protocol reduces to a time-dependent control of the chemical potentials, opening up the possibility for application in optical lattice experiments. Explicit applications to custom state preparation and stabilization of quantum many-body scars are presented in one- and two-dimensional lattices (three-dimensional applications are similarly possible).

Abstract Computer‐tomography‐guided needle biopsies are useful for diagnosing, staging, and classification of peripheral pulmonary nodules. However, the procedure carries a risk of iatrogenic pneumothorax. This report describes a patient‐case where a woman had undergone a computer‐tomography guided biopsy. Approximately 4 hours following discharge the patient was admitted to the emergency ward with severe chest pain and dyspnea. Chest x‐ray revealed bilateral pneumothorax and subcutaneous emphysema at the biopsy site. Pleural drainage was administered on the patient's right side. Another chest x‐ray following drainage showed regression of pneumothorax on both sides thus indicating communicating pleural cavities. Medical history revealed that the patient had been thymectomized 2 years earlier and a computer tomography visualized that the patient lacked mediastinal separation of the two pleural cavities. It is possible that patients with a history of mediastinal or thoracic surgery should be observed longer following procedures carrying risk of iatrogenic pneumothorax.

Wires made of topological insulators (TI) are a promising platform for searching for Majorana bound states. These states can be probed by analyzing the fractional ac Josephson effect in Josephson junctions with the TI wire as a weak link. An axial magnetic field can be used to tune the system from trivial to topologically nontrivial. Here, we investigate the oscillations of the supercurrent in such wire Josephson junctions as a function of the axial magnetic field strength and different contact transparencies. Although the current flows on average parallel to the magnetic field we observe h/2e, h/4e, and even h/8e periodic oscillations of the supercurrent in samples with lower contact transparencies. Corresponding tight-binding transport simulations using a Bogoliubov–de Gennes model Hamiltonian yield the supercurrent through the Josephson junctions, showing in particular the peculiar h/4e-periodic oscillations observed in experiments. A further semiclassical analysis based on Andreev-reflected trajectories connecting the two superconductors allows us to identify the physical origin of these oscillations. They can be related to flux-enclosing paths winding around the TI nanowire, thereby highlighting the three-dimensional character of the junction geometry compared to common planar junctions.

Abstract The protein HSF-1 is the controlling transcription factor of the heat-shock response (HSR). Its binding to the heat-shock elements (HSEs) induces the strong upregulation of conserved heat-shock proteins, including Hsp70s, Hsp40s and small HSPs. Next to these commonly known HSPs, more than 4000 other HSEs are found in the promoter regions of C. elegans genes. In microarray experiments, few of the HSE-containing genes are specifically upregulated during the heat-shock response. Most of the 4000 HSE-containing genes instead are unaffected by elevated temperatures and coexpress with genes unrelated to the HSR. This is also the case for several genes related to the HSP chaperone system, like dnj-12, dnj-13, and hsp-1. Interestingly, several promoters of the dedicated HSR-genes, like F44E5.4p, hsp-16.48p or hsp-16.2p, contain extended HSEs in their promoter region, composed of four or five HSE-elements instead of the common trimeric HSEs. We here aim at understanding how HSF-1 interacts with the different promoter regions. To this end we purify the nematode HSF-1 DBD and investigate the interaction with DNA sequences containing these regions. EMSA assays suggest that the HSF-1 DBD interacts with most of these HSE-containing dsDNAs, but with different characteristics. We employ sedimentation analytical ultracentrifugation (SV-AUC) to determine stoichiometry, affinity, and cooperativity of HSF-1 DBD binding to these HSEs. Interestingly, most HSEs show cooperative binding of the HSF-1 DBD with up to five DBDs being bound. In most cases binding to the HSEs of inducible promoters is stronger, even though the consensus scores are not always higher. The observed high affinity of HSF-1 DBD to the non-inducible HSEs of dnj-12, suggests that constitutive expression may be supported from some promoter regions, a fact that is evident for this transcription factor, that is essential also under non-stress conditions.

Abstract Background The mechanical drying of wood chips is an innovative method that improves the heating value of sawmill by-products in an energy-efficient continuous process. The liquid that comes out of the wood chips as press water (PW), however, contains a variety of undissolved as well as dissolved organic substances. The disposal of the PW as wastewater would generate additional costs due to its high organic load, offsetting the benefits in energy costs associated with the enhanced heating value of the wood chips. Our research explored if the organic load in PW could be utilized as a substrate by cellulolytic filamentous fungi. Hence, using the industrially relevant Ascomycete Trichoderma reesei RUT-C30 as well as several Basidiomycete wood-rotting fungi, we examined the potential of press water obtained from Douglas-fir wood chips to be used in the growth and enzyme production media. Results The addition of PW supernatant to liquid cultures of T. reesei RUT-C30 resulted in a significant enhancement of the endoglucanase and endoxylanase activities with a substantially shortened lag-phase. A partial replacement of Ca2+, Mg2+, K+, as well as a complete replacement of Fe2+, Mn2+, Zn2+ by supplementing PW of the liquid media was achieved without negative effects on enzyme production. Concentrations of PW above 50% showed no adverse effects regarding the achievable endoglucanase activity but affected the endoxylanase activity to some extent. Exploring the enhancing potential of several individual PW components after chemical analysis revealed that the observed lag-phase reduction of T. reesei RUT-C30 was not caused by the dissolved sugars and ions, nor the wood particles in the PW sediment, suggesting that other, so far non-identified, compounds are responsible. However, also the growth rate of several basidiomycetes was significantly enhanced by the supplementation of raw PW to the agar medium. Moreover, their cultivation in liquid cultures reduced the turbidity of the PW substantially. Conclusions PW was identified as a suitable media supplement for lignocellulolytic fungi, including the cellulase and xylanase producer T. reesei RUT-C30 and several wood-degrading basidiomycetes. The possibility to replace several minerals, trace elements and an equal volume of fresh water in liquid media with PW and the ability of fungal mycelia to filter out the suspended solids is a promising way to combine biological wastewater treatment with value-adding biotechnological applications.

Abstract The molecular chaperones Hsc70 and Hsp90 are required for proteostasis control and specific folding of client proteins in eukaryotic and prokaryotic organisms. Especially in eukaryotes these ATP-driven molecular chaperones are interacting with cofactors that specify the client spectrum and coordinate the ATPase cycles. Here we find that a Hsc70-cofactor of the Hsp40 family from nematodes, DNJ-13, directly interacts with the kinase-specific Hsp90-cofactor CDC-37. The interaction is specific for DNJ-13, while DNJ-12 another DnaJ-like protein of C. elegans, does not bind to CDC-37 in a similar manner. Analytical ultracentrifugation is employed to show that one CDC-37 molecule binds to a dimeric DNJ-13 protein with low micromolar affinity. We perform cross-linking studies with mass spectrometry to identify the interaction site and obtain specific cross-links connecting the N-terminal J-domain of DNJ-13 with the N-terminal domain of CDC-37. Further AUC experiments reveal that both, the N-terminal part of CDC-37 and the C-terminal domain of CDC-37, are required for efficient interaction. Furthermore, the presence of DNJ-13 strengthens the complex formation between CDC-37 and HSP-90 and modulates the nucleotide-dependent effects. These findings on the interaction between Hsp40 proteins and Hsp90-cofactors provide evidence for a more intricate interaction between the two chaperone systems during client processing.

Abstract Nematode development is characterized by progression through several larval stages. Thousands of genes were found in large scale RNAi-experiments to block this development at certain steps, two of which target the molecular chaperone HSP-90 and its cofactor UNC-45. Aiming to define the cause of arrest, we here investigate the status of nematodes after treatment with RNAi against hsp-90 and unc-45 by employing an in-depth transcriptional analysis of the arrested larvae. To identify misregulated transcriptional units, we calculate and validate genome-wide coexpression cliques covering the entire nematode genome. We define 307 coexpression cliques and more than half of these can be related to organismal functions by GO-term enrichment, phenotype enrichment or tissue enrichment analysis. Importantly, hsp-90 and unc-45 RNAi induce or repress many of these cliques in a coordinated manner, and then several specifically regulated cliques are observed. To map the developmental state of the arrested nematodes we define the expression behaviour of each of the cliques during development from embryo to adult nematode. hsp-90 RNAi can be seen to arrest development close to the L4 larval stage with further deviations in daf-16 regulated genes. unc-45 RNAi instead leads to arrested development at young adult stage prior to the programmatic downregulation of sperm-cell specific genes. In both cases processes can be defined to be misregulated upon depletion of the respective chaperone. With most of the defined gene cliques showing concerted behaviour at some stage of development from embryo to late adult, the “clique map” together with the clique-specific GO-terms, tissue and phenotype assignments will be a valuable tool in understanding concerted responses on the genome-wide level in Caenorhabditis elegans.

Abstract The glucocorticoid receptor is a key regulator of essential physiological processes, which under the control of the Hsp90 chaperone machinery, binds to steroid hormones and steroid-like molecules and in a rather complicated and elusive response, regulates a set of glucocorticoid responsive genes. We here examine a human glucocorticoid receptor variant, harboring a point mutation in the last C-terminal residues, L773P, that was associated to Primary Generalized Glucocorticoid Resistance, a condition originating from decreased affinity to hormone, impairing one or multiple aspects of GR action. Using in vitro and in silico methods, we assign the conformational consequences of this mutation to particular GR elements and report on the altered receptor properties regarding its binding to dexamethasone, a NCOA-2 coactivator-derived peptide, DNA, and importantly, its interaction with the chaperone machinery of Hsp90.

Abstract Protein kinases are important regulators in cellular signal transduction. As one major type of Hsp90 client, protein kinases rely on the ATP-dependent molecular chaperone Hsp90, which maintains their structure and supports their activation. Depending on client type, Hsp90 interacts with different cofactors. Here we report that besides the kinase-specific cofactor Cdc37 large PPIases of the Fkbp-type strongly bind to kinase•Hsp90•Cdc37 complexes. We evaluate the nucleotide regulation of these assemblies and identify prominent interaction sites in this quaternary complex. The synergistic interaction between the participating proteins and the conserved nature of the interaction suggests functions of the large PPIases Fkbp51/Fkbp52 and their nematode homolog FKB-6 as contributing factors to the kinase cycle of the Hsp90 machinery.

Abstract Most of the commercial apple cultivars are highly susceptible to fire blight, which is the most devastating bacterial disease affecting pome fruits. Resistance to fire blight is described especially in wild Malus accessions such as M. × robusta 5 (Mr5), but the molecular basis of host resistance response to the pathogen Erwinia amylovora is still largely unknown. The bacterial effector protein AvrRpt2EA was found to be the key determinant of resistance response in Mr5. A wild type E. amylovora strain and the corresponding avrRpt2 EA deletion mutant were used for inoculation of Mr5 to induce resistance or susceptible response, respectively. By comparison of the transcriptome of both responses, 211 differentially expressed genes (DEGs) were identified. We found that heat-shock response including heat-shock proteins (HSPs) and heat-shock transcription factors (HSFs) are activated in apple specifically in the susceptible response, independent of AvrRpt2 EA . Further analysis on the expression progress of 81 DEGs by high-throughput real-time qPCR resulted in the identification of genes that were activated after inoculation with E. amylovora. Hence, a potential role of these genes in the resistance to the pathogen is postulated, including genes coding for enzymes involved in formation of flavonoids and terpenoids, ribosome-inactivating enzymes (RIPs) and a squamosa promoter binding-like (SPL) transcription factor.

Two ways of to enhance the heat insulation of cold-protecting garments are studied using the mathematical model, which describes the coupled transport of temperature, humidity, and bound and condensed water. The model is developed in a one-dimensional formulation. The thermal radiation transport is explicitly considered by the subdivision of the heat flux into radiative and conduction parts. The model is utilized to study the improvement of heat-insulating properties of cold protective garments using aerogel materials and thin infrared reflective textile layers. Special attention is paid to the technological aspects of manufacturing such reflective textiles. The numerical investigations show that the use of infrared reflective textiles is the most effective of the two studied methods. Due to the reflection of the radiant heat flow coming from the human body, the skin temperature rises and the thermal insulation of clothing is significantly improved.

Methylation of a lysine residue in Hsp90 is a recently discovered post-translational modification but the mechanistic effects of this modification have remained unknown so far. Here the authors combine biochemical and biophysical approaches, molecular dynamics (MD) simulations and functional experiments with yeast and show that this lysine is a switch point, which specifically modulates conserved Hsp90 functions including co-chaperone regulation and client activation.

Abstract Background Although the most common path of infection for fire blight, a severe bacterial disease on apple, is via host plant flowers, quantitative trait loci (QTLs) for fire blight resistance to date have exclusively been mapped following shoot inoculation. It is not known whether the same mechanism underlies flower and shoot resistance. Results We report the detection of a fire blight resistance QTL following independent artificial inoculation of flowers and shoots on two F1 segregating populations derived from crossing resistant Malus ×robusta 5 (Mr5) with susceptible ‘Idared’ and ‘Royal Gala’ in experimental orchards in Germany and New Zealand, respectively. QTL mapping of phenotypic datasets from artificial flower inoculation of the ‘Idared’ × Mr5 population with Erwinia amylovora over several years, and of the ‘Royal Gala’ × Mr5 population in a single year, revealed a single major QTL controlling floral fire blight resistance on linkage group 3 (LG3) of Mr5. This QTL corresponds to the QTL on LG3 reported previously for the ‘Idared’ × Mr5 and an ‘M9’ × Mr5 population following shoot inoculation in the glasshouse. Interval mapping of phenotypic data from shoot inoculations of subsets from both flower resistance populations re-confirmed that the resistance QTL is in the same position on LG3 of Mr5 as that for flower inoculation. These results provide strong evidence that fire blight resistance in Mr5 is controlled by a major QTL on LG3, independently of the mode of infection, rootstock and environment. Conclusions This study demonstrates for the first time that resistance to fire blight caused by Erwinia amylovora is independent of the mode of inoculation at least in Malus ×robusta 5.

Besides their role as a storage for neutral lipids and sterols, there is increasing evidence that lipid droplets (LDs) are involved in cellular detoxification. LDs are in close contact to a broad variety of organelles where protein- and lipid exchange is mediated. Mitochondria as a main driver of the aging process produce reactive oxygen species (ROS), which damage several cellular components. LDs as highly dynamic organelles mediate a potent detoxification mechanism by taking up toxic lipids and proteins. A stimulation of LDs induced by the simultaneously overexpression of Lro1p and Dga1p (both encoding acyltransferases) prolongs the chronological as well as the replicative lifespan of yeast cells. The increased number of LDs reduces mitochondrial fragmentation as well as mitochondrial ROS production, both phenotypes that are signs of aging. Strains with an altered LD content or morphology as in the sei1∆ or lro1∆ mutant lead to a reduced replicative lifespan. In a yeast strain defective for the LON protease Pim1p, which showed an enhanced ROS production, increased doubling time and an altered mitochondrial morphology, a LRO1 overexpression resulted in a partially reversion of this “premature aging” phenotype.

Summary: Transcription factors harbor defined regulatory intrinsically disordered regions (IDRs), which raises the question of how they mediate binding to structured co-regulators and modulate their activity. Here, we present a detailed molecular regulatory mechanism of Forkhead box O4 (FOXO4) by the structured transcriptional co-regulator β-catenin. We find that the disordered FOXO4 C-terminal region, which contains its transactivation domain, binds β-catenin through two defined interaction sites, and this is regulated by combined PKB/AKT- and CK1-mediated phosphorylation. Binding of β-catenin competes with the autoinhibitory interaction of the FOXO4 disordered region with its DNA-binding Forkhead domain, and thereby enhances FOXO4 transcriptional activity. Furthermore, we show that binding of the β-catenin inhibitor protein ICAT is compatible with FOXO4 binding to β-catenin, suggesting that ICAT acts as a molecular switch between anti-proliferative FOXO and pro-proliferative Wnt/TCF/LEF signaling. These data illustrate how the interplay of IDRs, post-translational modifications, and co-factor binding contribute to transcription factor function.

Malus ×arnoldiana accession MAL0004 has been found to be resistant to moderately and highly virulent strains of the fire blight causal pathogen – the Gram-negative bacterium, Erwinia amylovora. Genetic analyses with an F1 segregating population derived from crossing the highly susceptible apple cultivar ‘Idared’ and MAL0004 led to the detection and mapping of the fire blight resistance locus of M. ×arnoldiana to linkage group (LG)12 (FB_Mar12). FB_Mar12 mapped at the distal end of LG12 below the apple SSR Hi07f01 in an interval of approximately 6 cM (Centimorgan), where both the fire blight resistance loci of M. floribunda 821 and ‘Evereste’ were located. We fine mapped the region containing FB_Mar12 using 892 progenies. Mining of the region of interest (ROI) on the ‘Golden Delicious’ doubled haploid genome (GDDH13) identified the presence of 2.3 Mb (megabases) in the homologous region. Of 40 primer pairs designed within this region, 20 were polymorphic and nine were mapped, leading to the identification of 24 significant recombinant individuals whose phenotypes were informative in determining the precise position of the locus within a 0.57 cM interval. Analyses of tightly linked marker sequences on the M. baccata draft genome revealed scaffolds of interest putatively harboring the resistance loci of M. ×arnoldiana, a hybrid between M. baccata and M. floribunda. Open reading frame (ORF) analyses led to the prediction of first fire blight resistance candidate genes with serine/threonine kinase and leucine-rich repeat domains, including homologs of previously identified ‘Evereste’ candidate genes. We discuss the implications of these results on breeding for resistance to fire blight.

Priming allows plants to respond faster and stronger to abiotic or biotic stresses. Leaf rust (Puccinia hordei) is an important pathogen of barley (Hordeum vulgare), for which resistance genes are known, but mostly overcome. Therefore, the aims of this study were (i) to establish a priming system in barley, based on bacterial N-acyl homoserine lactone (AHL), and (ii) to get information on the effect of priming on the reaction to leaf rust. Plants were inoculated with bacteria, i.e., Ensifer meliloti with repaired expR copy, producing the oxo-C14-homoserine lactone (AHL) and an E. meliloti strain carrying the attM lactonase gene from Agrobacterium tumefaciens, which cleaves the AHL and acts here as negative control. After three bacterial inoculations, plants were challenged with P. hordei strain I-80 at the three leaves stage. Twelve days after infection, scoring of the leaf area diseased and the infection type was conducted followed by the calculation of the relative susceptibility. First results indicate a significantly (P < 0.001) higher resistance level to P. hordei after inoculation with E. meliloti. Furthermore, significant (P < 0.001) differences were detected between the accessions tested for priming efficiency, which can be the basis to screen a larger set of barley accessions to detect quantitative trait loci or candidate genes involved in priming.

DNA microarrays are highly sensitive tools to evaluate the gene expression status of organismic samples and standardized array formats exist for many different sample types. Differential expression studies usually utilize the strongest up- or downregulated genes to generate networks visualizing the relationships among these genes. To include all yeast genes in one analysis and to get broader information on all cellular responses, we test a priori input of predefined genome-wide expression cliques and subsequent statistical analysis of the expression data. To this end, we generate a set of 72 co-regulation cliques using the information from 3196 microarray experiments. The obtained cliques performed highly significant in gene ontology and transcription factor enrichment analyses. We then tested the clique set on individual microarray experiments reporting on responses to pheromone, glycerol versus glucose based growth and the cellular response to heat. In all cases a highly significant determination of affected expression cliques was possible based on their average expression differences, the positions of their genes within hit rankings (UpRegScore) or the enrichment of the Top200 hits in certain cliques. The 72 cliques were finally used to compare experiments, which reported on the transcriptional response to polyglutamine proteins of different lengths. Using the predefined clique set it is possible to identify with high sensitivity and good significance sample and condition specific changes to gene expression. We thus conclude that an analysis, starting with these 72 preformed expression cliques, can complement traditional microarray analyses by visualizing the entire response on a static genome-wide gene set.

Safety and clinical feasibility of injecting a novel liquid fiducial marker for use in image guided radiotherapy in 15 patients with non-small cell lung cancer are reported. No major safety or toxicity issues were encountered. Markers present at start of radiotherapy remained visible in cone beam computed tomography and fluoroscopy images throughout the treatment course and on computed tomography images during follow-up (0–38 months). Marker volume reduction was seen until 9 months after treatment, after which no further marker breakdown was found. No post-treatment migration or marker related complications were found. The AvrRpt2EA effector protein of Erwinia amylovora is important for pathogen recognition in the fire blight–resistant crabapple Malus × robusta 5; however, little is known about its role in susceptible apples. To study its function in planta, we expressed a plant-optimized version of AvrRpt2EA driven by a heat shock–inducible promoter in transgenic plants of the fire blight–susceptible cultivar Pinova. After induced expression of AvrRpt2EA, transgenic lines showed shoot necrosis and browning of older leaves, with symptoms similar to natural fire blight infections. Transgenic expression of this effector protein resulted in an increase in the expression of the salicylic acid (SA)-responsive PR-1 gene but, also, in the levels of SA and its derivatives, with diverse kinetics in leaves of different ages. In contrast, no increase of expression levels of VSP2 paralogs, used as marker genes for the activation of the jasmonic acid (JA)-dependent defense pathway, could be detected, which is in agreement with metabolic profiling of JA and its derivatives. Our work demonstrates that AvrRpt2EA acts as a virulence factor and induces the formation of SA and SA-dependent systemic acquired resistance.

The Yersinia YaxAB system is a pore-forming toxin of so far unknown structure. Here authors present X-ray and cryo-EM to structures of individual subunits and of the YaxAB pore complex, and find that YaxA binds to membranes first and recruits YaxB for subsequent oligomerization.

Recombinant adeno-associated virus (rAAV) vectors have evolved as one of the most promising technologies for gene therapy due to their good safety profile, high transduction efficacy, and long-term gene expression in nondividing cells. rAAV-based gene therapy holds great promise for treating genetic disorders like inherited blindness, muscular atrophy, or bleeding disorders. There is a high demand for efficient and scalable production and purification methods for rAAVs. This is particularly true for the downstream purification methods. The current standard methods are based on multiple steps of gradient ultracentrifugation, which allow for the purification and enrichment of full rAAV particles, but the scale up of this method is challenging. Here, we explored fast, scalable, and universal liquid chromatography-based strategies for the purification of rAAVs. In contrast to the hydrophobic interaction chromatography (HIC), where a substantial amount of AAV was lost, the cation exchange chromatography (CEX) was performed robustly for multiple tested serotypes and resulted in a mixture of full and empty rAAVs with a good purity profile. For the used affinity chromatography (AC), a serotype dependence was observed. Anion exchange chromatography (AEX) worked well for the AAV8 serotype and achieved high levels of purification and a baseline separation of full and empty rAAVs. Depending on the AAV serotype, a combination of CEX and AEX or AC and AEX is recommended and holds promise for future translational projects that require highly pure and full particle-enriched rAAVs.

Radiation-induced changes may cause a non-malignant high 2-deoxy-2-[18F]fluoro-d-glucose (FDG)-uptake. The 3′-deoxy-3′-[18F]fluorothymidine (FLT)-PET/CT performs better in the differential diagnosis of inflammatory changes and lung lesions with a higher specificity than FDG-PET/CT. We investigated the association between post-radiotherapy FDG-PET-parameters, FLT-PET-parameters, and outcome. Sixty-one patients suspected for having a relapse after definitive radiotherapy for lung cancer were included. All the patients had FDG-PET/CT and FLT-PET/CT. FDG-PET- and FLT-PET-parameters were collected from within the irradiated high-dose volume (HDV) and from recurrent pulmonary lesions. For associations between PET-parameters and relapse status, respectively, the overall survival was analyzed. Thirty patients had a relapse, of these, 16 patients had a relapse within the HDV. FDG-SUVmax and FLT-SUVmax were higher in relapsed HDVs compared with non-relapsed HDVs (median FDG-SUVmax: 12.8 vs. 4.2; p < 0.001; median FLT-SUVmax 3.9 vs. 2.2; p < 0.001). A relapse within HDV had higher FDG-SUVpeak (median FDG-SUVpeak: 7.1 vs. 3.5; p = 0.014) and was larger (median metabolic tumor volume (MTV50%): 2.5 vs. 0.7; 0.014) than the relapsed lesions outside of HDV. The proliferative tumor volume (PTV50%) was prognostic for the overall survival (hazard ratio: 1.07 pr cm3 [1.01–1.13]; p = 0.014) in the univariate analysis, but not in the multivariate analysis. FDG-SUVmax and FLT-SUVmax may be helpful tools for differentiating the relapse from radiation-induced changes, however, they should not be used definitively for relapse detection.

DNA-Microarrays are powerful tools to obtain expression data on the genome-wide scale. We performed microarray experiments to elucidate the transcriptional networks, which are up- or down-regulated in response to the expression of toxic polyglutamine proteins in yeast. Such experiments initially generate hit lists containing differentially expressed genes. To look into transcriptional responses, we constructed networks from these genes. We therefore developed an algorithm, which is capable of dealing with very small numbers of microarrays by clustering the hits based on co-regulatory relationships obtained from the SPELL database. Here, we evaluate this algorithm according to several criteria and further develop its statistical capabilities. Initially, we define how the number of SPELL-derived co-regulated genes and the number of input hits influences the quality of the networks. We then show the ability of our networks to accurately predict further differentially expressed genes. Including these predicted genes into the networks improves the network quality and allows quantifying the predictive strength of the networks based on a newly implemented scoring method. We find that this approach is useful for our own experimental data sets and also for many other data sets which we tested from the SPELL microarray database. Furthermore, the clusters obtained by the described algorithm greatly improve the assignment to biological processes and transcription factors for the individual clusters. Thus, the described clustering approach, which will be available through the ClusterEx web interface, and the evaluation parameters derived from it represent valuable tools for the fast and informative analysis of yeast microarray data.

We identify a (pseudo) relativistic spin-dependent analogue of the celebrated quantum phase transition driven by the formation of a bright soliton in attractive one-dimensional bosonic gases. In this new scenario, due to the simultaneous existence of the linear dispersion and the bosonic nature of the system, special care must be taken with the choice of energy region where the transition takes place. Still, due to a crucial adiabatic separation of scales, and identified through extensive numerical diagonalization, a suitable effective model describing the transition is found. The corresponding mean-field analysis based on this effective model provides accurate predictions for the location of the quantum phase transition when compared against extensive numerical simulations. Furthermore, we numerically investigate the dynamical exponents characterizing the approach from its finite-size precursors to the sharp quantum phase transition in the thermodynamic limit.

Both in Germany and in China, there is strong expertise regarding the different aspects of forest management, as well as forest products management. Nevertheless, forestry in both countries is facing challenges, some of which are regional, but many of which are shared. Therefore, experts from both countries (Technical University of Munich Germany; Northwest A&F University Yangling, China; Forestry Academy of Shaanxi, China; Thünen Institut, Germany; FEDRC GIZ Forest Policy Facility (Forestry Economics Development and Research Center of the Deutsche Gesellschaft für Internationale Zusammenarbeit GmbH), Germany; and Center for Natural Forest Protection in Shaanxi, China) met to share their knowledge and deduce recommendations for future multifunctional forest management for the temperate zone. The workshop, held at the Northwest A&F University in September 2018, included presentations and intensive discussions, as well as a field tour. The results of the workshop that are summarized in this white paper are meant to provide an overview of the multi-faceted nature of the topic for interested scientists and forest practitioners, describe tools that can be used to analyze various aspects of multifunctionality and, in an exemplary fashion, highlight gathered experience from long- and short-term experiments. Included are social demands, economic goals, and scientific baselines. The topics reach from economic evaluations of forest ecosystem services over forest management practices, including afforestation, restoration, and preparations to face climate change, to wood/forest products utilization and participation of local people for poverty reduction. Overall, an optimistic picture emerges, showing that by using adapted forest management practices, which try to embrace the concept of multifunctionality, various use schemes and demands can be integrated at single sites, allowing us to achieve both environmental protection and productive forests, including societal demands, as well as aspects of tradition and national identity.

Oxidative stress in skin plays a major role in the aging process. This is true for intrinsic aging and even more for extrinsic aging. Although the results are quite different in dermis and epidermis, extrinsic aging is driven to a large extent by oxidative stress caused by UV irradiation. In this review the overall effects of oxidative stress are discussed as well as the sources of ROS including the mitochondrial ETC, peroxisomal and ER localized proteins, the Fenton reaction, and such enzymes as cyclooxygenases, lipoxygenases, xanthine oxidases, and NADPH oxidases. Furthermore, the defense mechanisms against oxidative stress ranging from enzymes like superoxide dismutases, catalases, peroxiredoxins, and GSH peroxidases to organic compounds such as L-ascorbate, α-tocopherol, beta-carotene, uric acid, CoQ10, and glutathione are described in more detail. In addition the oxidative stress induced modifications caused to proteins, lipids and DNA are discussed. Finally age-related changes of the skin are also a topic of this review. They include a disruption of the epidermal calcium gradient in old skin with an accompanying change in the composition of the cornified envelope. This modified cornified envelope also leads to an altered anti-oxidative capacity and a reduced barrier function of the epidermis.

The current rationale for target selection in Tourette syndrome revolves around the notion of cortico-basal ganglia circuit involvement in the pathophysiology of the disease. However, despite extensive research, the ideal target for deep brain stimulation (DBS) is still under debate, with many structures being neglected and underexplored. Based on clinical observations and taking into account the prevailing hypotheses of network processing in Tourette syndrome, we chose the fields of Forel, namely field H1, as a target for DBS. The fields of Forel constitute the main link between the striatopallidal system and the thalamocortical network, relaying pallidothalamic projections from core anatomical structures to the thalamic ventral nuclear group. In a retrospective study we investigated two patients suffering from chronic, medically intractable Tourette syndrome who underwent bilateral lead implantation in field H1 of Forel. Clinical scales revealed significant alleviation of tics and comorbid symptoms, namely depression and anxiety, in the postoperative course in both patients.

NADPH oxidases of human cells are not only functional in defense against invading microorganisms and for oxidative reactions needed for specialized biosynthetic pathways but also during the past few years have been established as signaling modules. It has been shown that human Nox4 is expressed in most somatic cell types and produces hydrogen peroxide, which signals to remodel the actin cytoskeleton. This correlates well with the function of Yno1, the only NADPH oxidase of yeast cells. Using two established tumor cell lines, which are derived from hepatic and neuroblastoma tumors, respectively, we are showing here that in both tumor models Nox4 is expressed in the ER (like the yeast NADPH oxidase), where according to published literature, it produces hydrogen peroxide. Reducing this biochemical activity by downregulating Nox4 transcription leads to loss of F-actin stress fibers. This phenotype is reversible by adding hydrogen peroxide to the cells. The effect of the Nox4 silencer RNA is specific for this gene as it does not influence the expression of Nox2. In the case of the SH-SY5Y neuronal cell line, Nox4 inhibition leads to loss of cell mobility as measured in scratch assays. We propose that inhibition of Nox4 (which is known to be strongly expressed in many tumors) could be studied as a new target for cancer treatment, in particular for inhibition of metastasis.

Hsp90 is a molecular chaperone involved in the regulation and maturation of kinases and transcription factors. In Caenorhabditis elegans, it contributes to the development of fertility, maintenance of muscle structure, the regulation of heat-shock response and dauer state. To understand the consequences of Hsp90-depletion, we studied Hsp90 RNAi-treated nematodes by DNA microarrays and mass spectrometry. We find that upon development of phenotypes the levels of chaperones and Hsp90 cofactors are increased, while specific proteins related to the innate immune response are depleted. In microarrays, we further find many differentially expressed genes related to gonad and larval development. These genes form an expression cluster that is regulated independently from the immune response implying separate pathways of Hsp90-involvement. Using fluorescent reporter strains for the differentially expressed immune response genes skr-5, dod-24 and clec-60 we observe that their activity in intestinal tissues is influenced by Hsp90-depletion. Instead, effects on the development are evident in both gonad arms. After Hsp90-depletion, changes can be observed in early embryos and adults containing fluorescence-tagged versions of SEPA-1, CAV-1 or PUD-1, all of which are downregulated after Hsp90-depletion. Our observations identify molecular events for Hsp90-RNAi induced phenotypes during development and immune responses, which may help to separately investigate independent Hsp90-influenced processes that are relevant during the nematode's life and development.

Pear (Pyrus communis) is an economically important fruit crop. Drops in yield and even losses of whole plantations are caused by diseases, most importantly fire blight which is triggered by the bacterial pathogen Erwinia amylovora. In response to the infection, biphenyls and dibenzofurans are formed as phytoalexins, biosynthesis of which is initiated by biphenyl synthase (BIS). Two PcBIS transcripts were cloned from fire blight-infected leaves and the encoded enzymes were characterized regarding substrate specificities and kinetic parameters. Expression of PcBIS1 and PcBIS2 was studied in three pear cultivars after inoculation with E. amylovora. Both PcBIS1 and PcBIS2 were expressed in 'Harrow Sweet', while only PcBIS2 transcripts were detected in 'Alexander Lucas' and 'Conference'. Expression of the PcBIS genes was observed in both leaves and the transition zone of the stem; however, biphenyls and dibenzofurans were only detected in stems. The maximum phytoalexin level (~110 μg/g dry weight) was observed in the transition zone of 'Harrow Sweet', whereas the concentrations were ten times lower in 'Conference' and not even detectable in 'Alexander Lucas'. In 'Harrow Sweet', the accumulation of the maximum phytoalexin level correlated with the halt of migration of the transition zone, whereby the residual part of the shoot survived. In contrast, the transition zones of 'Alexander Lucas' and 'Conference' advanced down to the rootstock, resulting in necrosis of the entire shoots.

Abstract:Over Easter 1903, a large-scale anti-Jewish riot in Kišinev, capital of the Russian governorate of Bessarabia, left dozens of Jews dead and hundreds injured, thus leading to a massive wave of emigration. A product of social discontent and anti-Semitic agitation, the riots of Kišinev became notoriously famous as the onset of a wave of pogroms of hitherto unprecedented brutality, which only subsided after the end of the Russian Revolution of 1905/06. This article analyzes the incidents by emphasizing cultural transfers between Kišinev and Lithuania, using the histoire croisée approach in order to provide for the different ethnic, social and political backgrounds and motivations of the actors. It also compares the disturbances in the rural north of Lithuania and in the Bessarabian industrial city of Kišinev in order to contextualize anti-Jewish violence in Lithuania on the larger scale of the Russian pogroms. When Lithuanian Jews were sometimes threatened to be killed “as in Kišinev” and at other times to be treated “as in Linkuva”, the significance of analyzing cultural transfer while keeping the regional context in mind becomes apparent.Rezumat:În perioada Paştelui anului 1903, o revoltă antievreiască la scară largă izbucnită la Chişinău, capitala guberniei ruseşti Basarabia, lăsa în urma sa zeci de evrei morţi şi sute de răniţi, ceea ce a condus la un val masiv de emigrare. Rezultat al nemulţumirilor sociale şi al agitaţiilor antisemite, revolta de la Chişinău a devenit foarte cunoscută ca marcând debutul unui val de pogromuri, de o brutalitate fără precedent până în acel moment, care s-a diminuat abia după încheierea Revoluţiei Ruse din 1905-1906. Articolul de faţă abordează incidentele prin studierea transferurilor culturale dintre Chişinău şi Lituania, utilizând metodele histoire croisée în scopul de a oferi explicaţii pentru diferenţele ce apar în ceea ce priveşte mediile etnice, sociale şi politice şi motivaţiile actorilor. Acesta compară, de asemenea, tulburările din partea rurală de nord a Lituaniei şi din oraşul industrial basarabean Chişinău, în scopul de a contextualiza violenţa anti-evreiască din Lituania la scara mai largă a pogromurilor ruseşti. Atunci când evreii lituanieni au fost, uneori, ameninţaţi că vor fi ucişi „ca la Chişinău” şi în alte momente că vor trataţi „ca în Linkuva”, semnificaţia analizei transferurilor culturale, păstrând în acelaşi timp în minte contextul regional, devine evidentă.

In previous publications, we could establish that a hallmark of human skin aging is the breakdown of the epidermal calcium gradient. This redistribution of calcium has many implications, including a restructuring of the cornified envelope, a reduced epidermal barrier function, a change in lipid composition, a reduced skin hydration, and an increased skin pH. Especially the age-dependent change in the cornified envelope composition was solely studied in human foreskin samples. The aim of this study was to confirm that this effect is neither restricted to UV-protected skin area nor limited to a specific sex. In addition, we wanted to show that the collapse of the epidermal calcium gradient is not only a hallmark of human skin aging, but is also evolutionarily conserved in mammals. By using such techniques as IHC, Western blot analysis, and RT-PCR, we could demonstrate the following: (1) A change in the epidermal calcium gradient is in fact the most important sign of epidermal aging in mammals (as shown in female human eyelids and mouse skin samples of the external ear-shell); (2) The disturbed calcium homeostasis affects the expression and crosslinking of most cornified-envelope-specific genes such as loricrin and filaggrin. In this way, we could establish that the age-dependent altered composition of the cornified envelope is a typical sign of skin aging not only in humans, but in mice, too. This makes the mouse an important model organism to study these changes.

We propose a method—a quantum time mirror (QTM)—for simulating a partial time-reversal of the free-space motion of a non-relativistic quantum wave packet. The method is based on a short-time spatially homogeneous perturbation to the wave packet dynamics, achieved by adding a nonlinear time-dependent term to the underlying Schrödinger equation. Numerical calculations, supporting our analytical considerations, demonstrate the effectiveness of the proposed QTM for generating a time-reversed echo image of initially localized matter-wave packets in one and two spatial dimensions. We also discuss possible experimental realizations of the proposed QTM.

Analytical ultracentrifugation (AUC) is a first principles based method to determine absolute sedimentation coefficients and buoyant molar masses of macromolecules and their complexes, reporting on their size and shape in free solution. The purpose of this multi-laboratory study was to establish the precision and accuracy of basic data dimensions in AUC and validate previously proposed calibration techniques. Three kits of AUC cell assemblies containing radial and temperature calibration tools and a bovine serum albumin (BSA) reference sample were shared among 67 laboratories, generating 129 comprehensive data sets. These allowed for an assessment of many parameters of instrument performance, including accuracy of the reported scan time after the start of centrifugation, the accuracy of the temperature calibration, and the accuracy of the radial magnification. The range of sedimentation coefficients obtained for BSA monomer in different instruments and using different optical systems was from 3.655 S to 4.949 S, with a mean and standard deviation of (4.304 ± 0.188) S (4.4%). After the combined application of correction factors derived from the external calibration references for elapsed time, scan velocity, temperature, and radial magnification, the range of s-values was reduced 7-fold with a mean of 4.325 S and a 6-fold reduced standard deviation of ± 0.030 S (0.7%). In addition, the large data set provided an opportunity to determine the instrument-to-instrument variation of the absolute radial positions reported in the scan files, the precision of photometric or refractometric signal magnitudes, and the precision of the calculated apparent molar mass of BSA monomer and the fraction of BSA dimers. These results highlight the necessity and effectiveness of independent calibration of basic AUC data dimensions for reliable quantitative studies.

Für die vergleichende Kartierung von Feuerbrandresistenz bei Malus wurden vier Populationen bearbeitet, die als einen Elter einen Donor für Feuerbrandresistenz und als zweiten Elter die anfällige Sorte ‘Idared’ hatten. Als Donoren für Widerstandsfähigkeit wurden definierte Abstammungen der Wildarten M. baccata, M. fusca, M. ×robusta und die widerstandsfähige Pillnitzer Apfelsorte ‘Rewena’ genutzt. Die Phänotypisierung der Populationen erfolgte in unterschiedlichen Jahren durch Inokulation von vege­tativen Trieben mit dem Erwinia amylovora-Isolat Ea222 und anderen definierten Isolaten unter Gewächshaus­bedingungen. Der Befall der Triebe wurde als Befallsrate, dem Verhältnis der befallenen Trieblänge zur Gesamt­trieblänge, ermittelt. Je nach Isolat und Population va­riierte die durchschnittliche Befallsrate zwischen 10 bis 82%. Die Genotypisierung der Nachkommen zur Erstellung genetischer Karten erfolgte mit Mikrosatelliten-, SNP- und DArT-Markern. Die genetischen Kopplungs­karten wurden mittels Joinmap 4.0 und die Resistenz gegenüber Feuerbrand mit MapQTL5.0 kartiert. Major-QTLs konnten für M. ×robusta auf Kopplungsgruppe 3, für M. baccata auf Kopplungsgruppe 12 und für M. fusca auf Kopplungsgruppe 10 detektiert werden. Während die Feuerbrand-QTLs von M. ×robusta 5 und M. fusca je nach Jahr bis zu ca. 87% der phänotypischen Varianz erklären können, erklärt der QTL von M. baccata nur ca. 45%, was die Existenz eines zweiten QTL nahe legt. Für die resistente Sorte ‘Rewena’ konnte kein QTL detektiert werden. Mit den Markern, die im Bereich der detektierten QTLs liegen, kann auf Resistenz gegenüber Feuerbrand selektiert werden. Damit ist eine Kombinierung unterschiedlicher Feuerbrandresistenz-QTLs möglich. Auf dieser Basis können zukünftig Sorten gezüchtet werden, die dauerhaft widerstandsfähig gegenüber Feuerbrand sind, um diese dann dem deutschen Obstbau zur Verfügung stellen zu können. A box-model for the prognosis of tropospheric ozone is used to assess the sensitivity of simulated peak ozone concentrations to local VOC and NOx emissions. Distinction is made between biogenic and anthropogenic emissions. The study is performed for a rural and an urban area in Bavaria in July. Main outcome is that in the urban area much less relative increase in emissions is necessary to produce drastically high ozone concentrations than in the rural area, although the simulated concentrations of ozone were higher in the rural area than in the urban area.

The heat shock response (HSR) is essential to survive acute proteotoxic stress and has been studied extensively in unicellular organisms and tissue culture cells, but to a lesser extent in intact metazoan animals. To identify the regulatory pathways that control the HSR in Caenorhabditis elegans, we performed a genome-wide RNAi screen and identified 59 genes corresponding to 7 positive activators required for the HSR and 52 negative regulators whose knockdown leads to constitutive activation of the HSR. These modifiers function in specific steps of gene expression, protein synthesis, protein folding, trafficking, and protein clearance, and comprise the metazoan heat shock regulatory network (HSN). Whereas the positive regulators function in all tissues of C. elegans, nearly all of the negative regulators exhibited tissue-selective effects. Knockdown of the subunits of the proteasome strongly induces HS reporter expression only in the intestine and spermatheca but not in muscle cells, while knockdown of subunits of the TRiC/CCT chaperonin induces HS reporter expression only in muscle cells. Yet, both the proteasome and TRiC/CCT chaperonin are ubiquitously expressed and are required for clearance and folding in all tissues. We propose that the HSN identifies a key subset of the proteostasis machinery that regulates the HSR according to the unique functional requirements of each tissue.

As we have shown previously, yeast Mmi1 protein translocates from the cytoplasm to the outer surface of mitochondria when vegetatively growing yeast cells are exposed to oxidative stress. Here we analyzed the effect of heat stress on Mmi1 distribution. We performed domain analyses and found that binding of Mmi1 to mitochondria is mediated by its central alpha-helical domain (V-domain) under all conditions tested. In contrast, the isolated N-terminal flexible loop domain of the protein always displays nuclear localization. Using immunoelectron microscopy we confirmed re-location of Mmi1 to the nucleus and showed association of Mmi1 with intact and heat shock-altered mitochondria. We also show here that mmi1Δ mutant strains are resistant to robust heat shock with respect to clonogenicity of the cells. To elucidate this phenotype we found that the cytosolic Mmi1 holoprotein re-localized to the nucleus even in cells heat-shocked at 40°C. Upon robust heat shock at 46°C, Mmi1 partly co-localized with the proteasome marker Rpn1 in the nuclear region as well as with the cytoplasmic stress granules defined by Rpg1 (eIF3a). We co-localized Mmi1 also with Bre5, Ubp3 and Cdc48 which are involved in the protein de-ubiquitination machinery, protecting protein substrates from proteasomal degradation. A comparison of proteolytic activities of wild type and mmi1Δ cells revealed that Mmi1 appears to be an inhibitor of the proteasome. We conclude that one of the physiological functions of the multifunctional protein module, Mmi1, is likely in regulating degradation and/or protection of proteins thereby indirectly regulating the pathways leading to cell death in stressed cells.

BACKGROUND AND AIMS: High frequency electrosurgery has a key role in the broadening application of liver surgery. Its molecular signature, i.e. the metabolites evolving from electrocauterization which may inhibit hepatic wound healing, have not been systematically studied. METHODS: Human liver samples were thus obtained during surgery before and after electrosurgical dissection and subjected to a two-stage metabolomic screening experiment (discovery sample: N = 18, replication sample: N = 20) using gas chromatography/mass spectrometry. RESULTS: In a set of 208 chemically defined metabolites, electrosurgical dissection lead to a distinct metabolic signature resulting in a separation in the first two dimensions of a principal components analysis. The idea of having to liberate the Christian peasants from the harmful economic and moral influence of Jewish merchants was an essential element of the political agenda of both the secular intelligentsia and the Catholic clergy. Activists of both political camps started founding cooperative shops, which were seen as the most promising tool to “emancipate” the peasants and the founding of which became legally possible after a streak of reforms shortly before the Russian Revolution of 1905/06. The article thus poses the questions of what role antisemitism played in the cooperatives, what tasks these cooperatives were supposed to fulfill and whether they were a success or not. The article comes to the conclusion that after the Revolution, there was a significant dissent between the two groups mentioned: While priests argued that cooperatives needed to be antisemitic in order to be successful, the liberal intelligentsia countered that antisemitism deterred cooperative shops from being economically successful. Both groups, however, celebrated the founding of such shops as a means for Lithuanians to gain foot in the Jewish dominated towns.

Hsc70 is a conserved ATP-dependent molecular chaperone, which utilizes the energy of ATP hydrolysis to alter the folding state of its client proteins. In contrast to the Hsc70 systems of bacteria, yeast and humans, the Hsc70 system of C. elegans (CeHsc70) has not been studied to date.We find that CeHsc70 is characterized by a high ATP turnover rate and limited by post-hydrolysis nucleotide exchange. This rate-limiting step is defined by the helical lid domain at the C-terminus. A certain truncation in this domain (CeHsc70-Δ545) reduces the turnover rate and renders the hydrolysis step rate-limiting. The helical lid domain also affects cofactor affinities as the lidless mutant CeHsc70-Δ512 binds more strongly to DNJ-13, forming large protein complexes in the presence of ATP. Despite preserving the ability to hydrolyze ATP and interact with its cofactors DNJ-13 and BAG-1, the truncation of the helical lid domain leads to the loss of all protein folding activity, highlighting the requirement of this domain for the functionality of the nematode's Hsc70 protein.

The ATP-dependent molecular chaperone Hsp90 is required for the activation of a variety of client proteins involved in various cellular processes. Despite the abundance of known client proteins, functions of Hsp90 in the organismal context are not fully explored. In Caenorhabditis elegans, Hsp90 (DAF-21) has been implicated in the regulation of the stress-resistant dauer state, in chemosensing and in gonad formation. In a C. elegans strain carrying a DAF-21 mutation with a lower ATP turnover, we observed motility defects. Similarly, a reduction of DAF-21 levels in wild type nematodes leads to reduced motility and induction of the muscular stress response. Furthermore, aggregates of the myosin MYO-3 are visible in muscle cells, if DAF-21 is depleted, implying a role of Hsp90 in the maintenance of muscle cell functionality. Similar defects can also be observed upon knockdown of the Hsp90-cochaperone UNC-45. In life nematodes YFP-DAF-21 localizes to the I-band and the M-line of the muscular ultrastructure, but the protein is not stably attached there. The Hsp90-cofactor UNC-45-CFP contrarily can be found in all bands of the nematode muscle ultrastructure and stably associates with the UNC-54 containing A-band. Thus, despite the physical interaction between DAF-21 and UNC-45, apparently the two proteins are not always localized to the same muscular structures. While UNC-45 can stably bind to myofilaments in the muscular ultrastructure, Hsp90 (DAF-21) appears to participate in the maintenance of muscle structures as a transiently associated diffusible factor.
