Der Zweck dieser Arbeit ist die Untersuchung des Faktors Vertrauen im B2B-Verkaufsbereich. Dabei liegt der Fokus auf den unterschiedlichen Wahrnehmungen hinsichtlich des Vertrauens seitens des Verkaufsbereichs und jenen seitens des Einkaufsbereich. Untersucht wird, wie Vertrauen im Verkaufsgespräch entsteht, aufrechterhalten werden kann und welche Faktoren vertrauensbildend wirken. Im Vordergrund steht dabei, der Vergleich der Wahrnehmung von vertrauensbildenden Maßnahmen von Vertriebsmitarbeiter/innen und Einkäufer/innen und die daraus resultierenden Unterschiede. Zu Beginn werden der Begriff Vertrauen und seine theoretische Fundierung umfassend beleuchtet und die wichtigsten Erfolgs- und Einflussfaktoren vertrauensbildender Kundenbeziehungen erläutert. Danach erfolgen Interviews mit ausgewählten Expert/innen aus beiden Bereichen, deren Ergebnisse mit Hilfe der qualitativen Inhaltsanalyse untersucht werden. Diese zeigen, dass Einkäufer/innen viel Wert auf aktives Zuhören in einer vertrauten Kundenbeziehung legen, während Vertriebsmitarbeiter/innen diese Fähigkeit nicht mit Vertrauen in Verbindung bringen. Zum Schluss finden eine ausführliche Analyse und Interpretation weiterer festgestellter Wahrnehmungsunterschiede bezüglich des Vertrauens statt und es werden Vorschläge für weitere Untersuchungen unterbreitet.*****The purpose of this work is to study the factor of trust in the B2B sales area. The focus lies on the different perceptions of trust on the part of the seller and that of the buyer. The aim is to examine how trust develops in sales conversation, how it can be maintained and which factors have a trust-building effect. The focus is on the comparison of the perception of trust-building measures by sales staff and buyers and the resulting differences. With continuing advances in the resolving power of super-resolution microscopy, the inefficient labeling of proteins with suitable fluorophores becomes a limiting factor. For example, the low labeling density achieved with antibodies or small molecule tags limits attempts to reveal local protein nano-architecture of cellular compartments. On the other hand, high laser intensities cause photobleaching within and nearby an imaged region, thereby further reducing labeling density and impairing multi-plane whole-cell 3D super-resolution imaging. Here, we show that both labeling density and photobleaching can be addressed by repetitive application of trisNTA-fluorophore conjugates reversibly binding to a histidine-tagged protein by a novel approach called single-epitope repetitive imaging (SERI). For single-plane super-resolution microscopy, we demonstrate that, after multiple rounds of labeling and imaging, the signal density is increased. Using the same approach of repetitive imaging, washing and re-labeling, we demonstrate whole-cell 3D super-resolution imaging compensated for photobleaching above or below the imaging plane. This proof-of-principle study demonstrates that repetitive labeling of histidine-tagged proteins provides a versatile solution to break the ‘labeling barrier’ and to bypass photobleaching in multi-plane, whole-cell 3D experiments.
