The Kentucky distilling industry ranks as one of the state's largest industries and continues to expand. In 2017, the Kentucky distilling industry was responsible for approximately $235 million in state and local tax revenues (Coomes and Kornstein, 2019). Rye (Secale cereale L.) grains are a vital component for production of some distilled spirits. Although winter rye is produced on relatively few hectares in Kentucky currently, a recent initiative has supported expanding production to help meet the growing demand of local distilleries. University of Kentucky winter rye research field trials were visited in Caldwell and Logan Counties, KY in May 2018, and in Fayette County, KY in May 2019. Leaves were collected that had dark brown, oval to irregular-shaped lesions with definite margins and yellow halos. Symptoms were present on approximately 50% to 80% of the flag leaves, with severity ranging from 5% to 30% of the flag leaf area affected. Leaves were surface-disinfested by soaking in a 2% NaOCl solution for 1 min and rinsed twice in sterilized water and then placed in a humidity chamber (plastic bag with moist paper towels) at room temperature (approximately 24°C) to induce fungal sporulation. Seventeen single-spore isolates were obtained and stored at -80°C in 15% glycerol solution. Isolates were grown on potato dextrose agar under 12 h cycles of white light/darkness for 5 days. Colonies were gray to black. Conidia that formed were mostly straight or slightly curved, dark olivaceous brown, 3-7 septate, and 41.0-90.4 × 15.2-29.3 µm. Based on the symptoms observed on the collected leaves and these morphological characteristics similar to those described by Chang and Hwang (2000) and Manamgoda et al. (2014), the fungus was tentatively identified as Bipolaris sorokiniana (Sorokin) Shoemaker. The sequence of internal transcribed spacer regions (ITS) and glyceraldehyde 3-phosphate dehydrogenase (GAPDH) were used to identify three isolates (18Bs004, 18Bs111 and 19Bs064) using primer ITS1/ITS4 (White et al. 1990) and GPD1/GPD2 (Berbee et al. 1999), respectively. The sequences were deposited in GenBank with accession numbers MT457817, MT457818 and MZ066635 for ITS sequences and MZ073644 to MZ073646 for GAPDH sequences. BLAST searches with ITS and GAPDH sequences matched 100% identity (344/344 bp and 515/515 bp for ITS and GAPDH sequences, respectively) to B. sorokiniana (GenBank accession No. MT254731 and MH844813, respectively). To prove pathogenicity, a conidial suspension (1 × 105 conidia/ml) was used to inoculate 15-day-old cultivar 'Serafino' winter rye plants in the greenhouse. Leaves of 8 plants were inoculated with 50 ml of the conidial suspension using a spray bottle. Plants were covered with a transparent plastic bag for 48 h, and symptoms were observed 10 days after inoculation. Leaf lesions, similar to those described above, were present on all inoculated plants, but no symptoms were observed on non-inoculated control plants. Bipolaris sorokiniana was reisolated from symptomatic leaves and the identity of the pathogen was confirmed based on the morphology previously described. To our knowledge, this is the first report of spot blotch caused by B. sorokiniana on winter rye in Kentucky, but B. sorokiniana has been reported on rye in the neighboring state of Virginia (Roane 2009). Kentucky produces approximately 150,000 and 4,000 ha of winter wheat (Triticum aestivum) and winter barley (Hordeum vulgare) annually, respectively, which are both known hosts of B. sorokiniana (Kumar et al. 2002). An isolate of B. sorokiniana from rye was reported by Ghazvini and Tekauz (2007) to be less virulent on barley differential lines. Further research is needed to better understand spot blotch distribution, susceptibility in winter rye cultivars, and potential yield and quality loss implications in winter rye production and end use. It is unknown how susceptible various winter rye cultivars grown in Kentucky are to spot blotch.

Fusarium graminearum , the major causal agent of Fusarium head blight (FHB) of wheat ( Triticum aestivum ) in the U.S., can produce mycotoxins, such as deoxynivalenol (DON), during infection. Contamination of wheat grain with DON is a major concern for wheat producers and millers, and the U.S. Food and Drug Administration (FDA) has set advisory levels for DON in finished wheat products for human and animal consumption. Practices utilized to manage FHB and DON contamination include planting wheat cultivars with moderate resistance to FHB and applying efficacious fungicides at the beginning of anthesis. Under severe epidemics, DON contamination can exceed FDA advisory levels despite implementation of these measures. Additionally, fungicide efficacy can be limited when anthesis is not uniform among plants in the field, which can occur when planting is delayed or if there is non-uniform seedling establishment. The objectives of this study were to evaluate the effect of (1) in-furrow phosphorus application at planting and seeding rate on heading and anthesis uniformity, FHB symptomology, DON contamination, grain yield, yield components, and test weight; and (2) harvesting at different grain moisture concentrations on FHB symptomology, DON contamination, grain yield and test weight. Field trials were established in Princeton, Kentucky, from 2017 to 2019, to evaluate in-furrow phosphorus application at planting (0 kg P 2 O 5 ha -1 and 47 kg P 2 O 5 ha -1 ); seeding rate (377 live seeds m -2 and 603 live seeds m -2 ); and grain moisture at harvest (20 to 22% and 13 to 15%). In-furrow phosphorus increased grain yield and spikes m -2 , but had no effect on heading and anthesis uniformity or DON contamination. The 603 live seeds m -2 seeding rate decreased the number of days to Zadoks 60 for the November planted wheat, and decreased FHB incidence, but did not decrease DON contamination. Harvesting at 20 to 22% grain moisture decreased Fusarium damaged kernel ratings and percent kernel infection but increased DON contamination in the harvested grain. Although in-furrow phosphorus, seeding rate, and harvesting 20 to 22% grain moisture did not decrease DON contamination, there is potential for these treatments to alleviate negative effects of late planted wheat grown in stressful environments., (Copyright © 2020 Rod, Bradley, Van Sanford and Knott.)

The time course of persistence of Mycobacterium tuberculosis as measured by detection of rRNA, acid-fast bacillus (AFB) smear, and culture was determined for pulmonary tuberculosis patients during antimicrobial therapy. Twenty-three patients who were initially AFB smear positive and who subsequently completed a course of antimicrobial therapy were selected for the study. Sequential specimens were tested by AFB smear, culture, and rRNA amplification (Gen-Probe Amplified Mycobacterium Tuberculosis Direct Test [MTD]). The initial diagnostic specimens of all patients were positive by culture; those of 22 patients (96%) also were positive by MTD. Overall, MTD results remained positive longer than both smear and culture results. The median times to the last positive test result were 9 days for AFB smear, 26 days for culture, and 30 days for MTD. The last positive test result was the AFB smear result in 4% of cases, the culture result in 22%, and the MTD result in 52%. Fifty-six percent of patients had a period of shedding of noncultivable M. tuberculosis which was detected by MTD after culture results had converted to negative. This noncultivable period lasted 7 to 245 days. All three tests became reproducibly negative before the end of therapy and remained negative during follow-up for up to 1 year. These results indicate that during successful antimicrobial therapy, M. tuberculosis is eliminated in sputum samples as measured by amplification of rRNA, as well as by AFB smear and culture. No long-term rRNA carrier state was detected. While the time course of clearance of M. tuberculosis measured by rRNA overall was longer than with the two traditional tests, the rRNA test results allow sensitive and precise measurement of the clearance of noncultivable M. tuberculosis from respiratory specimens. This attribute may allow rRNA testing to be useful in clarifying patient response to antimicrobial therapy.

During previous cooperative numerical taxonomic studies of slowly growing mycobacteria, the International Working Group on Mycobacterial Taxonomy described a number of strains whose taxonomic status was ambiguous. A new study of DNA, RNA, and proteins from 66 of these organisms was performed to correlate their properties with phenotypic clustering behavior; the results of this study permitted 51 of the strains studied to be assigned to known species. The methods used to characterize the semantides included nucleotide sequencing and assessment of levels of semantide relatedness by affinity binding techniques, including whole DNA-DNA hybridization, probe hybridization, and antibody binding. There was good overall agreement between the phenotypic and chemotaxonomic clusters and the groups of organisms identified by semantide analyses. Our results supported the conclusion that we should continue to rely on polyphasic taxonomy to provide satisfactory systematic resolution of members of the genus Mycobacterium. We identified no single 16S rRNA interstrain nucleotide sequence difference value that unequivocally defined species boundaries. DNA-DNA hybridization remains the gold standard, but common resources are needed to permit DNA-DNA hybridization analyses to be made available to laboratories that are not prepared to use this technology. One of the large novel clusters which we studied corresponds to the recently described species Mycobacterium interjectum, a pathogen that resembles the nonpathogen Mycobacterium gordonae phenotypically. We also identified strains that appear to represent ribovars of Mycobacterium intracellulare which do not react with the commercial diagnostic probes that are currently used for identification of this species. Other branches or clusters consisted of too few strains to permit a decision about their taxonomic status to be made.

Seven hundred fifty-eight processed sputum sediments received for the diagnosis of tuberculosis or other mycobacterial infections were tested by utilizing a rRNA target amplification assay and traditional culture techniques. The results from the rRNA target amplification assay (Gen-Probe Amplified Mycobacterium Tuberculosis Direct Test), available in 5 h, were compared with the results from standard culture techniques held for 6 weeks. A total of 119 specimens (16%) were culture positive for Mycobacterium tuberculosis. Overall sensitivity, specificity, positive predictive value, and negative predictive value were 82, 99, 97, and 96%, respectively, for the Gen-Probe assay; 88, 100, 100, and 97%, respectively, for culture; and 53, 99.8, 99.6, and 91%, respectively, for fluorochrome stain. The Gen-Probe assay employs the isothermal enzymatic amplification of M. tuberculosis complex rRNA followed by detection of the amplicon with an acridinium ester-labeled DNA probe. This assay has the potential of reducing the time for diagnosis of tuberculosis to 1 day.

Synopsis Greasy hair is a common problem in Europe. The first step in developing anti-grease hair products must be to establish a sensitive protocol for measuring any changes in perceived hair greasiness. Sensitive clinical trials and in vivo evaluation methods of determining hair greasiness have been developed that show significant differences in the perceived hair greasiness following the use of different shampoos. Products tested in two clinical trials for efficacy as anti-grease shampoos were an anti-grease shampoo containing 2% 3,4-thiolanediol as the active ingredient, and a baby shampoo based on mild surfactants. The same placebo shampoo based on ether sulphates used in typical European shampoo formulations was used in both tests. A shampoo containing 2% zinc pyrithione was included in the trials as a control, since there were indications that this would increase the amount of hair greasiness. Neither of the test products were shown to be effective anti-grease shampoos. The shampoo containing 2% zinc pyrithione was shown to produce significantly more greasiness than both the placebo and the test shampoos. Since significant differences could be shown between the different shampoos, the protocol and in vivo evaluation techniques used in these clinical trials are considered to be validated.
