19. Identification of Staphylococcus and Micrococcus species with the STAPHYtest system.

Abstract

A collection of 216 well-characterized strains of Staphylococcus, Micrococcus and Stomatococcus was examined by a commercially available STAPHYtest system ( Lachema, Brno, Czechoslovakia). The results of STAPHYtest agreed with those of conventional tests. The STAPHYtest permitted a clear-cut separation of Staphylococcus from Micrococcus and Stomatococcus strains and correctly identified 104 of 145 (72%) Staphylococcus strains after 24 h of incubation. However, it allowed the identification only of 19 of 29 validly published Staphylococcus species. The STAPHYtest proved to be a simple and rapid system for the separation of staphylococci from micrococci and for the identification of most frequent clinically significant staphylococci. Abstract

The guanine+cytosine content (GC) of DNAs of 9 strains of pediococci and 6 strains of aerococci was determined using different physicochemical methods. It was found that the GC content of DNA of the pediococci ranged from 37.8 to 41.2%, while that of the aerococci varied from 42.0 to 43.2%. The results obtained show that genetically related species are involved whose mutual position in the system is determined by only slight differences in the phenotype.

Bacteriol. 90:808–816. 1965.—Ultrathin sections of Sporosarcina ureaecells were studied by means of electron microscopy. The cell wall consists of several layers and is 340 A thick. The cytoplasm is of globular structure and includes ribosomelike structures, occasional mesosomes, and inclusions not precisely identifiable. The nuclear area has various shapes and is formed by filaments 10 to 20 A thick which proceed in various directions. Cell division occurs similarly to that of sarcinate. Both synchronic and asynchronic cell division was observed. The spores of S. ureaeconsist of an outer coat having several layers, a cortex, a spore wall, and cytoplasm. The results of the present investigation substantiate our previous suggestion that S. ureaeshould be transferred from the family Micrococcaceae to the family Bacillaceae.

Abstract

A study of ultrathin sections ofMicrococcus denitrificansandM.halodenitrificanshas shown similar cell structures. The cell wall consists of several layers corresponding to those of the cell wall of gram-negative bacteria. The thickness of the cell wall is 250 – 350 Å; that of the cytoplasmic membrane 70 Å. The cytoplasm in both species contains ribosomes and inclusions of polymetaphosphate. Comparison with ultrathin sections ofThiobacillus novellusshows too much difference to consider the former two species to be identical with the latter one. Abstract

SUMMARY: Staphylococcus flavocyaneus produces at least two pigments on glucose yeast-extract agar; a violet diffusible pigment and a yellow non-diffusing pigment. On some media the production of violet diffusible pigment is inhibited. In broth cultures of this organism a mutation takes place giving rise to yellow-pigmented mutants which have lost the ability of producing the violet pigment. A considerable accumulation of these mutants occurs in cultures on both solid and liquid media. No reversion to the violet-pigmented state or other change in pigmentation in the mutants has been observed. The yellow-pigmented mutants do not differ from strains of Micrococcus luteus with the exception of some characteristics which are variable within that species. This supports the conclusions of Kocur & Martinec (1963) that Staphylococcus flavocyaneus Knaysi should be regarded as a strain of Micrococcus luteus (Schroeter) Cohn.
