Abstract

Cardiac experimental electrophysiology is in need of a well-defined Minimum Information Standard for recording, annotating, and reporting experimental data. As a step towards establishing this, we present a draft standard, called Minimum Information about a Cardiac Electrophysiology Experiment (MICEE). The ultimate goal is to develop a useful tool for cardiac electrophysiologists which facilitates and improves dissemination of the minimum information necessary for reproduction of cardiac electrophysiology research, allowing for easier comparison and utilisation of findings by others. It is hoped that this will enhance the integration of individual results into experimental, computational, and conceptual models. In its present form, this draft is intended for assessment and development by the research community. We invite the reader to join this effort, and, if deemed productive, implement the Minimum Information about a Cardiac Electrophysiology Experiment standard in their own work.

Abstract

An experiment was conducted to investigate the role of the transient outward current (Ito) in the sinoatrial node by blocking the current with 4-aminopyridine in small ball-like tissue preparations from different regions of the rabbit sinoatrial node. It was shown for the first time ever that 4-aminopyridine-sensitive current plays a significant part in the electrical activity of the sinoatrial node and that this function differs from the periphery to the center and from the superior part of the sinoatrial node to the inferior part.

Abstract

The action potential characteristics of the central and peripheral rabbit sinoatrial (SA) node was analyzed to characterize the role of Ca2+ and Na+ currents on pacemaker activity. Analysis of spontaneous action potentials in the isolated rabbit right atrium in the presence of nifedipine and tetrodotoxin indicated the role of the L-type Ca2+ current inhibitor in attenuating central SA node action potential. On the other hand, the inhibition of Na+ currents by tetrodotoxin attenuated the action potentials of the peripheral SA node.

Abstract

The effects of UL-FS-49 or zatebradine, ZD-7288 and Cs+ on the spontaneous action potentials of the sinoatrial (SA) node were analyzed in isolated rabbit right atrial preparations. The administration of Cs+, UL-FS-49 and ZD-7288 induced significant reductions in pacemaker slope in the peripheral regions of the SA node tissues. Furthermore, the highest percent decrease in pacemaker slope was induced by Cs+ compared to UL-FS-49 and ZD-7288.

Item does not contain fulltext, AIM: In healthy hearts, ventricular gap junctions are mainly composed by connexin43 (Cx43) and localize in the intercalated disc, enabling appropriate electrical coupling. In diseased hearts, Cx43 is heterogeneously down-regulated, whereas activity of calmodulin/calcium-calmodulin protein kinase II (CaM/CaMKII) signalling increases. It is unclear if CaM/CaMKII affects Cx43 expression/localization or impulse propagation. We analysed different models to assess this. METHODS AND RESULTS: AC3-I mice with CaMKII genetically inhibited were subjected to pressure overload (16 weeks, TAC vs. sham). Optical and epicardial mapping was performed on Langendorff-perfused rabbit and AC3-I hearts, respectively. Cx43 subcellular distribution from rabbit/mouse ventricles was evaluated by immunoblot after Triton X-100-based fractionation. In mice with constitutively reduced CaMKII activity (AC3-I), conduction velocity (CV) was augmented (n = 11, P < 0.01 vs. WT); in AC3-I, CV was preserved after TAC, in contrast to a reduction seen in TAC-WT mice (-20%). Cx43 expression was preserved after TAC in AC3-I mice, though arrhythmias and fibrosis were still present. In rabbits, W7 (CaM inhibitor, 10 microM) increased CV (6-13%, n= 6, P< 0.05), while susceptibility to arrhythmias decreased. Immunoconfocal microscopy revealed enlarged Cx43 cluster sizes at intercalated discs of those hearts. Total Cx43 did not change by W7 (n= 4), whereas Triton X-100 insoluble Cx43 increased (+21%, n= 4, P< 0.01). Similar findings were obtained in AC3-I mouse hearts when compared with control, and in cultured dog cardiomyocytes. Functional implication was shown through increased intercellular coupling in cultured neonatal rat cardiomyocytes. CONCLUSION: Both acute and chronic CaM/CaMKII inhibition improves conduction characteristics and enhances localization of Cx43 in the intercalated disc. In the absence of fibrosis, this reduced the susceptibility for arrhythmias.

Abstract

Mathematical models of action potentials in the periphery and center of the rabbit sinoatrial node. Am J Physiol Heart Circ Physiol 279: H397-H421, 2000.--Mathematical models of the action potential in the periphery and center of the rabbit sinoatrial (SA) node have been developed on the basis of published experimental data. Simulated action potentials are consistent with those recorded experimentally: the model-generated peripheral action potential has a more negative takeoff potential, faster upstroke, more positive peak value, prominent phase 1 repolarization, greater amplitude, shorter duration, and more negative maximum diastolic potential than the model-generated central action potential. In addition, the model peripheral cell shows faster pacemaking. The models behave qualitatively the same as tissue from the periphery and center of the SA node in response to block of tetrodotoxinsensitive [Na.sup.+] current, L- and T-type [Ca.sup.2+] currents, 4-aminopyridine-sensitive transient outward current, rapid and slow delayed rectifying [K.sup.+] currents, and hyperpolarizationactivated current. A one-dimensional model of a string of SA node tissue, incorporating regional heterogeneity, coupled to a string of atrial tissue has been constructed to simulate the behavior of the intact SA node. In the one-dimensional model, the spontaneous action potential initiated in the center propagates to the periphery at ~0.06 m/s and then into the atrial muscle at 0.62 m/s. heart; pacemaking; regional differences; computer modeling

Abstract

To develop basic technologies for the maritime transport of hydrogen energy, a liquid hydrogen experiment facility (LHEF) with system enabling observation under horizontal vibration has been designed and constructed. The LHEF consists of a liquid hydrogen optical cryostat, a gas-handling system, vacuum pumps, and apparatus for generating horizontal vibrations. The liquid hydrogen optical cryostat, which is 1200 mm in height and 300 mm in diameter, includes a vacuum jacket, a liquid nitrogen space (10.0 liters), a liquid hydrogen space (13.6 liters), a sample space (3.8 liters), optical windows, and a needle valve. The results of performance tests show that the heat leak in the liquid hydrogen space is sufficiently small. Using the LHEF under horizontal vibration, the damped oscillation of the liquid hydrogen surface is successfully observed and analyzed on the basis of a simple model. [ABSTRACT FROM AUTHOR]
