Background: We aimed to assess perioperative changes in fibrinogen in the cerebrospinal fluid (CSF), their association with markers of blood–brain barrier breakdown and neuroinflammation, and their association with postoperative delirium severity. Methods: We conducted a secondary analysis of the Interventions for Postoperative Delirium-Biomarker 2 (IPOD-B2, NCT02926417) study, a prospective observational cohort study. We included 24 patients aged >21 yr undergoing aortic aneurysm repair. CSF samples were obtained before (n=24) and after surgery (n=13), with some participants having multiple postoperative samples. Abstract The purpose of this review was to analyze the scientific literature on exocrine pancreatic insufficiency (EPI) in dogs and cats and our own research on porcine model to compare animal‐ and microbial‐derived enzymes in the treatment of animals with this disease. Clinical signs of EPI occur when more than 85% of the pancreatic parenchyma is non‐functional. EPI can be a consequence of various diseases. The insufficient activity or deficiency of pancreatic enzymes leads to impaired digestion and absorption, and consequently, to malnutrition. The primary treatment for enzyme insufficiency is pancreatic enzyme replacement therapy (PERT). PERT in animals with EPI is a lifetime therapy. Most commercially available products are of animal origin (processed pancreata obtained from a slaughter house) and contain lipases, alpha‐amylase, and proteases. Enzymes of microbial and plant origin seem to be a promising alternative to animal‐derived enzymes, but to date there are no registered preparations containing all enzymes simultaneously for use in clinical practice to treat EPI. Results from some previous studies have highlighted the “extra‐digestive” functions of pancreatic enzymes, as well as the actions of pancreatic‐like microbial enzymes. For example, trypsin activates protease‐activated receptor and provokes maturation of enterocytes and enterostatin inhibits fat absorption. It has been postulated that intrapancreatic amylase is the main component of the acini‐islet‐acinar axis—the reflex which down regulates insulin release, while gut and blood amylase exhibit anti‐incretin actions “per se.” Additionally, high but still physiological blood amylase activity coincide with physiological glucose homeostasis and a lack of obesity.

AbstractThere is limited intersectionality-informed, contextually diverse qualitative research on school experiences of LGBTQ+ students and links to mental health. Focusing on racialized, ethnic minority and religious/faith groups, this coproduced UK study examines how: 1) sexual and/or gender diverse students with intersecting minoritized identities experience school; 2) school environments could better support their mental health; and 3) what shapes school inclusion initiatives. In-depth interviews and focus groups were conducted with 22 staff members, 32 students (13–19 years), and nine LGBTQ+ school inclusion training providers.The sample was diverse across gender and sexual identity, race/ethnicity, religion and context. Data were analysed using reflexive thematic analysis, identifying three themes (and subthemes): 1) “What's important is putting the young person first” (‘Minority stress: it's more stressful for me’, ‘We're in this together but who's in charge?’, ‘Prioritising students’ needs and lived reality’), 2) “Move beyond viewing religious beliefs and culture versus LGBTQ+ inclusion as a zero-sum game” (‘Faith clashes?’, ‘Fears about going there maintain distance’), and 3) “We need to talk: engaging with parents and school communities” (‘Credible messengers', ‘Finding ways into the conversation’).Findings emphasize the importance of intersectional perspectives. Also, a need to acknowledge historic and current oppressions shaping current perspectives on gender and sexual diversity; reflexivity and intersubjectivity in communication with parents, students and communities; greater intersectional representation; and raised awareness and disruption of inequity-creating norms. This requires policy-level support, guidance and resourcing to avoid widening inequities on socio-economic grounds and to prepare students to live within a mutually tolerant society.

The study aimed to analyze and evaluate the production and economic performance of a broiler chicken farm in Poland during the years 2020–2023. Production performance was assessed by comparing final body weight (BW), feed intake (FI), feed conversion ratio (FCR), and the European Performance Efficiency Factor (EPEF). Economic results were determined using the sales value, production costs, profit (direct surplus), and profitability index (PI). During the study years, the broiler rearing period lasted on average 40 days, with six production cycles each year. The average BW was found to be 2.51 kg with an average FCR of 1.54 kg/kg. For the four production years, the EPEF was calculated, reaching a high average value of 399. Broiler chicken production in Poland was profitable despite the continuously increasing production costs. In the years studied, the cost of feed accounted for 69.6–76.1% of the total production costs, chicks 15.0–19.7%, and labor costs 1.59–2.39%. In all of the analyzed production cycles, the PI of broiler chicken rearing was above 100, averaging 120.48%, and the average profit per bird was 0.46 EUR. During the study period, several negative events occurred, including the COVID-19 pandemic, the spread of avian influenza outbreaks, and consequent fluctuations in feed and poultry prices. Despite these challenges, namely the consistently rising production costs, broiler production in the study farm remained profitable.

This study aimed to investigate the effects of a Standardized Natural Citrus Extract (SNCE) on broiler chickens’ growth performance, gut health, carcass quality, and welfare. A total of 756 one-day-old Ross 308 males were randomly assigned to two groups: a control group (CTL) fed with a standard diet, and a citrus group (SNCE) fed with the same standard diet supplemented with 250 g/ton of feed of SNCE. Growth performance was recorded weekly until d 35, while mortality was recorded daily. The feed conversion ratio (FCR) and European Efficiency Index (EEI) of broiler chickens were also calculated weekly. At day 35, 10 birds per group were randomly selected for slaughter performance. In parallel, broiler chickens’ welfare was assessed according to the Welfare Quality Assessment Protocol. Caecal digest was also collected post mortem for short-chain fatty acids (SCFA) analyses, and jejunum samples were collected for ex vivo gut permeability assay. SNCE dietary supplementation enhanced broiler chickens’ performance, i.e., final bodyweight and EEI, compared to the CTL group. The carcass weight was also significantly higher in the SNCE group. In addition, the fat percentage was lower in the SNCE group. Regarding broiler chickens’ welfare and gut health parameters, Footpad Dermatitis (FPD) and gate score were also lower in birds supplemented with SNCE. The SCFA measurement showed a lower concentration of iso-butyric acid, iso-valeric acid, and total putrefactive SCFA in the SNCE group. The differences in gut permeability measured as TEER value indicate that using citrus extract lowered the risk of gut inflammation. This study provides valuable insights into the mechanisms of action that may underlie the observed effects of SNCE on performance, as demonstrated in this study and others. These effects could potentially be attributed to the reduction in inflammation and the enhanced utilization of nutrients. Further studies are needed to confirm these results.

The increasing resistance of bacteria to antibiotics has obliged the EU Member States to reduce by 50% the use of antibiotics in animal production by 2030. This study was undertaken with the aim to analyse the use of antibiotics in flocks of broiler turkeys reared in the Warmia and Mazury province in a two-year period.

The objective of this study was primarily to assess the different performance impacts of two ionophore coccidiostats (narasin and salinomycin) used to manage coccidiosis. While both products may be efficacious in controlling disease challenges, previous literature has suggested that some ionophores are less well tolerated by the broiler chickens. In this study, we were particularly interested to know how the use of different coccidiostat programs translates into broiler health and performance, as measured by zootechnical parameters such as the feed conversion ratio, average daily gain, and final body weight. A total of 352 male Ross 308 one-day-old broilers were randomly divided into two treatment groups (T1 and T2). Treatment 1 included a basal diet (BD) + nicarbazin/narasin (Maxiban®, Elanco) at 100 ppm 0–24 days, narasin at 70 ppm 25–42 days, and (2) Treatment 2 included basal diet + nicarbazin/narasin at 100 ppm 0–24 days, salinomycin (Sacox®, Huvepharma) at 70 ppm 25–42 days. Efficacy and performance parameters, slaughter analysis, dry matter (DM) in litter, and intestinal integrity (I2) were measured for the broilers from both treatment groups. The findings demonstrated more favorable results for broilers reared in the group diet fed with narasin (in the finisher phase), including higher daily body weight gain, higher final body weight, lower feed conversion ratio value (improved feed efficiency), and higher European Production Efficiency Factor value, compared with the salinomycin-supplemented group.

Abstract Background Alongside the benefits of Total-Body imaging modalities, such as higher sensitivity, single-bed position, low dose imaging, etc., their final construction cost prevents worldwide utilization. The main aim of this study is to present a simulation-based comparison of the sensitivities of existing and currently developed tomographs to introduce a cost-efficient solution for constructing a Total-Body PET scanner based on plastic scintillators. Methods For the case of this study, eight tomographs based on the uEXPLORER configuration with different scintillator materials (BGO, LYSO), axial field-of-view (97.4 cm and 194.8 cm), and detector configurations (full and sparse) were simulated. In addition, 8 J-PET scanners with different configurations, such as various axial field-of-view (200 cm and 250 cm), different cross sections of plastic scintillator, and multiple numbers of plastic scintillator layers (2, 3, and 4), based on J-PET technology have been simulated by GATE software. Furthermore, Siemens’ Biograph Vision has been simulated to compare the results with standard PET scans. Two types of simulations have been performed. The first one with a centrally located source with a diameter of 1 mm and a length of 250 cm, and the second one with the same source inside a water-filled cylindrical phantom with a diameter of 20 cm and a length of 183 cm. Results With regards to sensitivity, among all the proposed scanners, the ones constructed with BGO crystals give the best performance ( $$\sim$$ ∼ 350 cps/kBq at the center). The utilization of sparse geometry or LYSO crystals significantly lowers the achievable sensitivity of such systems. The J-PET design gives a similar sensitivity to the sparse LYSO crystal-based detectors while having full detector coverage over the body. Moreover, it provides uniform sensitivity over the body with additional gain on its sides and provides the possibility for high-quality brain imaging. Conclusion Taking into account not only the sensitivity but also the price of Total-Body PET tomographs, which till now was one of the main obstacles in their widespread clinical availability, the J-PET tomography system based on plastic scintillators could be a cost-efficient alternative for Total-Body PET scanners.

In positron emission tomography (PET) studies, convolutional neural networks (CNNs) may be applied directly to the reconstructed distribution of radioactive tracers injected into the patient's body, as a pattern recognition tool. Nonetheless, unprocessed PET coincidence data exist in tabular format. This paper develops the transformation of tabular data into -dimensional matrices, as a preparation stage for classification based on CNNs. This method explicitly introduces a nonlinear transformation at the feature engineering stage and then uses principal component analysis to create the images. We apply the proposed methodology to the classification of simulated PET coincidence events originating from NEMA IEC and anthropomorphic XCAT phantom. Comparative studies of neural network architectures, including multilayer perceptron and convolutional networks, were conducted. The developed method increased the initial number of features from 6 to 209 and gave the best precision results (79.8) for all tested neural network architectures; it also showed the smallest decrease when changing the test data to another phantom.

The effects of dietary probiotic supplementation with viable Bacillus subtilis and Bacillus amyloliquefaciens spores on sow performance, immunity, gut functional status and biofilm formation by probiotic bacteria in piglets at weaning were investigated. Ninety-six sows reared in a continuous farrowing system for one full cycle were fed gestation diets during the first 90 d of pregnancy and lactation diets until the end of lactation. The sows were fed a basal diet without probiotics (control; n = 48) or a diet supplemented with viable spores (1.1 × 109 CFU/kg of feed) (probiotic; n = 48). At 7 d of age, sucking piglets (n = 12/group) were provided prestarter creep feed until weaning at 28 d of age. The piglets in the probiotic group were supplemented with the same probiotic and dosage as their dams. Blood and colostrum collected from sows and ileal tissues collected from piglets on the day of weaning were used for analyses. Probiotics increased the weight of piglets (P = 0.077), improved the weaning weight (P = 0.039) and increased both the total creep feed consumption (P = 0.027) and litter gain (P = 0.011). Probiotics also improved the faecal score in the second (P = 0.013) week of life. The immunoglobulin G (IgG) concentrations in sow blood at farrowing and the IgM concentrations in piglet blood at weaning were higher in the probiotic group than in the control group (P = 0.046). The piglets from the probiotic-treated sows showed a higher IgM concentration in the ileal mucosa (P = 0.050) and a lower IgG concentration in the ileal mucosa (P = 0.021) compared with the piglets from control sows. School is a key site for prevention and early intervention in public mental health, with sexual and gender minority students being a priority group for action. Context is important in understanding how school inclusion of sexual and gender minorities shapes mental health and well-being, with rapidly changing social and political forces necessitating ongoing research. This coproduced UK secondary school-based study aimed to understand (a) key components of mentally, socially and emotionally healthy school environments for LGBTQ+ students considerate of intersecting minoritised identities; (b) staff information, skills and capacity needs and (c) factors influencing uptake and implementation. Online interviews and focus groups were conducted with 63 participants (22 staff, 32 students (aged 13-19 years), and 9 training providers), diverse in relation to gender and sexual identity, ethnicity, religious and social context. Data were analysed thematically. One overarching theme captured the need for an intersectionality-informed, contextually adaptable, whole school approach which 'shifts the narrative' away from deficit thinking, challenging prevailing cis/heteronormative and White norms. This underpinned four themes: (1) 'Feeling safe, seen and celebrated: embedding intersectional signs, signals and symbols', (2) 'Everyone's business: the need for collaboration', (3) 'Embedding a culture of change', and (4) 'Re-locating the problem: challenging deficit thinking'. Contextually diverse research is needed which critically addresses ways in which social power enacted interpersonally and structurally serves to hinder schools from enacting LGBTQ+ inclusivity. Evidence to inform and develop implementation strategies for institutional changes and to advocate for wider socio-political support is also key to mitigate the potential for widening inequities linked to inequitable school environments.

Abstract No study has demonstrated the use of dietary Cannabis-derived cannabidiol (CBD) to alter the stress response in chickens or examined its effects on meat volatile compounds (VOCs). Here, we subjected chickens to dysbiosis via C. perfringens infection or Escherichia coli lipopolysaccharide (LPS) treatment and investigated the potential link between meat VOCs and cecal bacterial activity and the ameliorative effect of CBD. The cecal bacterial production of short-chain fatty acids (SCFAs) was closely correlated with meat VOCs. CBD supplementation reduced the formation of breast meat spoilage VOCs, including alcohols, trimethylamine and pentanoic acid, in the challenged birds, partly by decreasing cecal putrefactive SCFA production. Meat VOC/cecal SCFA relationships differed according to the challenge, and CBD attenuated the effects of C. perfringens infection better than the effects of LPS challenge on meat VOCs. These findings provide new insights into the interactions among bioactive agent supplementation, gut microbiota activity and meat properties in birds.

Abstract Selenium is an essential element that is important for many metabolic processes. Feed components used in chicken nutrition, especially cereals, may be deficient in selenium, hence selenium supplementation is necessary. Taking into account the progress in breeding, and thus the higher demand of birds for this element, it seems obvious to investigate an increased selenium dose in the diet of chickens. The aim of the study was to evaluate the effect of feed enriched with different forms of selenium at an increased dose of 0.5 mg/kg feed on the profile and metabolism of fatty acids in the breast muscle and liver of chickens. The study was conducted on 300 Ross 308 chickens reared for 42 days under standard conditions. The control group received feed supplemented with sodium selenite at a dose of 0.3 mg/kg feed. The research groups received different forms of selenium (sodium selenate, selenised yeast, nano-selenium) at an increased dose of 0.5 mg/kg feed. The study showed that the administration of different forms of selenium in the feed affected its concentration in the breast muscle and liver (p ≤ 0.01). Nano-selenium was found to have a high bioavailability, but also a lower risk of toxicity compared to other forms of selenium. Using different forms of selenium (p ≤ 0.01) at a dose of 0.5 mg/kg feed can significantly modify the fatty acid profile, lipid and enzymatic indices of fatty acid metabolism in breast muscle and liver.
