Deregulated energy markets enable various actors to benefit from energy, either by selling it for profit or buying it at low prices. A market is a complex system where multiple actors and systems interact with each other. As a result, electricity prices when viewed as time series contain complex features such as high volatility, non-stationarity, non-seasonal effects, extreme values of Market Clearing Price (MCP). Therefore, this renders electricity price a demanding task. A MCP forecast for a given future time interval is a tantamount process for every actor, since it can assist decisions associated with financial or energy portfolio management. For the purpose of MCP forecasting, a methodology of low runtime complexity and low demand in resources is needed in order to provide the user sufficient time to test multiple scenarios or deploy it on a system which lacks scalability or high computational resources. For this reason the concept of Extreme Learning Machine (ELM) is utilized for this thesis. ELM has a low runtime complexity but in many cases can provide poor generalization abilities, the hyperparameters when altered slightly produce different matrix weights that significantly affect forecasting accuracy. In order to enhance accuracy and robustness ELM is modified by bootstrapping, bagging, and hyperparameters optimization via genetic algorithm. The MCP time series used for this work was taken from the Australian energy market Operator (AEMO). It is concluded that multiple clusters with moderate number of samples sharing similar features, lead to well-trained ELM learners with satisfactory generalization ability. Thus, this changes the forecast activity in forecasting clusters first and once the cluster is determined forecasting within this cluster is implemented. The Australian Market was chosen for training, validation and testing due to its high volatility of prices. The analysis conducted indicates that the ELM can be deployed as a forecasting methodology when accompanied with additional methods, for increased robustness of forecasts. Οι αποκεντρωμένες αγορές ενέργειας επιτρέπουν σε διάφορους παίκτες/συμμετέχοντες να επωφεληθούν από διάφορα προϊόντα ενέργειας, είτε πουλώντας τα με σκοπό το κέρδος είτε αγοράζοντας τα σε χαμηλές τιμές για κατ’ ιδίαν χρήση. Μία αγορά ενέργειας είναι ένα πολύπλοκο σύστημα όπου πολλοί παράγοντες και συστήματα αλληλοεπιδρούν μεταξύ τους. Ως αποτέλεσμα, οι τιμές ηλεκτρικής ενέργειας όταν αντιμετωπίζονται ως χρονοσειρές περιέχουν πολύπλοκα χαρακτηριστικά όπως υψηλή μεταβλητότητα, έλλειψη σταθερότητας, εποχικές διακυμάνσεις και ακραίες τιμές της τιμής εκκαθάρισης αγοράς (MCP). Επομένως η πρόβλεψη της τιμής αγοράς σε μελλοντικούς χρόνους καθίσταται μια απαιτητική εργασία. Μια πρόβλεψη MCP για ένα δεδομένο μελλοντικό χρονικό διάστημα είναι μια χρήσιμη για τους παίκτες πράξη, καθώς μπορεί να βοηθήσει στη λήψη αποφάσεων που σχετίζονται με τη διαχείριση ενός χρηματοοικονομικού ή ενεργειακού χαρτοφυλακίου. Επίσης οι διάφοροι παίκτες θα πρέπει να είναι σε θέση να τρέξουν τον αλγόριθμο σε ένα σύστημα που δεν διαθέτει επιλογές επεκτασιμότητα ή απαιτήσεις για υψηλούς υπολογιστικούς πόρους. Για το λόγο αυτό η έννοια του Extreme Learning Machine (ELM) χρησιμοποιείται για την παρούσα μεταπτυχιακή εργασία. Το ELM έχει χαμηλή πολυπλοκότητα χρόνου εκτέλεσης, αλλά σε πολλές περιπτώσεις μπορεί να παρέχει μη ικανοποιητικές ικανότητες γενίκευσης, οι παραμέτροι όταν τροποποιούνται ελαφρώς παράγουν διαφορετικά βάρη μήτρας που επηρεάζουν την ακρίβεια πρόβλεψης. Προκειμένου να βελτιωθεί η ακρίβεια και η ευρωστία, το ELM τροποποιείται με την εισαγωγή μεθόδων βελτιστοποίησης όπως bootstrapping, bagging και ρύθμιση των παραμέτρων του ELM μέσω γενετικού αλγόριθμου. Η χρονοσειρά MCP που χρησιμοποιήθηκε για αυτήν την εργασία λήφθηκε από τον Διαχειριστή της Αυστραλιανής αγοράς ενέργειας (AEMO). Εξάγεται το συμπέρασμα ότι πολλαπλές κλάσεις με μέτριο αριθμό δειγμάτων που μοιράζονται παρόμοια χαρακτηριστικά, οδηγούν σε καλά εκπαιδευμένα ELM αντικείμενα, με ικανοποιητική ικανότητα γενίκευσης. Για τον λόγο αυτό πρώτα γίνεται πρόβλεψη της κλάσης που ανήκει η επιθυμητή μέρα και μόλις καθοριστεί η κλάση, η εκπαίδευση και πρόβλεψη εκτελούνται. Η αγορά της Αυστραλίας επιλέχθηκε για εκπαίδευση, επικύρωση και δοκιμή λόγω της υψηλής μεταβλητότητας των τιμών. Η ανάλυση που πραγματοποιήθηκε δείχνει ότι το ELM μπορεί να χρησιμοποιηθεί ως μεθοδολογία πρόβλεψης όταν συνοδεύεται από πρόσθετες μεθόδους, για αυξημένη ευρωστία των προβλέψεων.

Aging, genomic stress, and mitochondrial dysfunction are risk factors for neurodegenerative pathologies, such as Parkinson disease (PD). Although genomic instability is associated with aging and mitochondrial impairment, the underlyingmechanisms are poorly understood. Here, we show that base excision repair generates genomic stress, promoting age-related neurodegeneration in a Caenorhabditis elegans PD model. A physiological level of NTH-1 DNA glycosylase mediates mitochondrial and nuclear genomic instability, which promote degeneration of dopaminergic neurons in older nematodes. Conversely, NTH-1 deficiency protects against a-synuclein-induced neurotoxicity, maintaining neuronal function with age. This apparent paradox is caused by modulation of mitochondrial transcription in NTH-1-deficient cells, and this modulation activates LMD-3, JNK-1, and SKN-1 and induces mitohormesis. The dependance of neuroprotection on mitochondrial transcription highlights the integration of BER and transcription regulation during physiological aging. Finally, whole-exome sequencing of genomic DNA from patients with idiopathic PD suggests that base excision repair might modulate susceptibility to PD in humans.

Η αύξηση του αριθμού των ηλεκτρικών οχημάτων (HO) θα επιφέρει μεγάλη αύξηση στη ζήτηση της ηλεκτρικής ενέργειας. Η έξυπνη φόρτιση των ΗΟ έχει πολλά οφέλη όπως την μείωση του κόστος φόρτισης, την αποφυγή μεγάλων επενδύσεων στο δίκτυο διανομής κ.ά. Ο συνδυασμός της έξυπνης φόρτισης των ΗΟ με υπηρεσίες όπως Vehicle-to-Grid (V2G) (μεταφορά ενέργειας από τα ΗΟ στο δίκτυο διανομής) μπορεί να αποδώσει παραπάνω οφέλη στον δίκτυο της ηλεκτρικής ενέργειας καθώς και στον ιδιοκτήτη του ΗΟ. Στην παρούσα εργασία, προτείνεται μια αποτελεσματική μέθοδος φόρτισης των ΗΟ σε ένα πάρκινγκ χρησιμοποιώντας V2G λειτουργία και λαμβάνοντας υπόψη τους περιορισμούς σε επίπεδο ΗΟ, του τοπικού μετασχηματιστή του πάρκινγκ και του δικτύου διανομής. Ο κύριος στόχος της προτεινόμενης μεθόδου φόρτισης είναι η μείωση του κόστους φόρτισης σε επίπεδο πάρκινγκ και ΗΟ. Χρησιμοποιήθηκε ο αλγόριθμος Βελτιστοποίησης Σμήνους Σωματιδίων (Particle Swarm Optimization, PSO) για τον προγραμματισμό της φόρτισης των ΗΟ. Δυο συμβατικοί μέθοδοι φόρτισης των ΗΟ χρησιμοποιήθηκαν για την σύγκριση των αποτελεσμάτων: 1) τα ΗΟ φορτίζουν με την μέγιστη ισχύ που τους επιτρέπει ο σταθμός φόρτισης και ο on board μετατροπέας ισχύος του κάθε ΗΟ και 2) τα ΗΟ φορτίζουν με την απαιτούμενη σταθερή ισχύ που χρειάζεται για να επιτύχουν το επιθυμητό επίπεδο φόρτισης. Επιπλέον, έγινε σύγκριση των αποτελεσμάτων της προτεινόμενης μεθόδου με ισοδύναμο σωρευτικό μοντέλο του πάρκινγκ όπου το σύνολο των ΗΟ ισοδυναμούσε με μια μπαταρία. Χρησιμοποιήθηκαν πραγματικά δεδομένα για τη τιμή της ηλεκτρικής ενέργειας, του φορτίου του δικτύου διανομής και της δραστηριότητας του πάρκινγκ (αφίξεις/αναχωρήσεις). Η αποτελεσματικότητα της προτεινόμενης μεθόδου αποδεικνύεται από τα αποτελέσματα που προέκυψαν από τα ενδεικτικά σενάρια λειτουργίας. Τέλος, αναφέρεται ότι για την εφαρμογή της μεθόδου φόρτισης δεν απαιτούνται εξελιγμένα συστήματα υπολογισμού, μέτρησης και επικοινωνίας., The increase of the number of electric vehicles will lead to a large increase in electricity demand. Smart charging of electric vehicles (EVs) has many benefits such as reduction of charging costs, limitation of large investments in the distribution network, etc. The combination of smart charging of EVs with ancillary services such as Vehicle-to-Grid (V2G) (energy transfer from EVs to the distribution network) can provide more benefits to the electricity network as well as to the owner of the EV. In this work, a method for the efficient charging of electric vehicles (EVs) at the parking lot (PL) level, including V2G operation and taking into account lifetime of EV batteries, distribution network and local transformer loading, is proposed. The main goal of the proposed charging method is to reduce the cost of charging at the levels of PL and EV. The Particle Swarm Optimization (PSO) algorithm was used to derive the charging schedule of the EVs. The proposed method is compared with two conventional charging strategies: 1) the EVs charge with the maximum power allowed by the charging station and the on-board power converter of each EV and 2) the EVs charge with the constant power required to achieve the desired level of charge at the end of the charging period. In addition, the results of the proposed method were compared with an equivalent aggregation model of the EVs parking lot where all EVs’ batteries were aggregated to one battery. Real-world data were used for the price of electricity, the load of the distribution network and the activity of the PL (arrivals / departures of the hosted EVs). The effectiveness of the proposed method is demonstrated by the results obtained by the indicative operation scenarios. Finally, it is noted that the application of the proposed charging method does not require advanced calculation, measurement and communication systems.

Phenotypic screening for autophagy inhibitors followed by target identification resulted in the discovery of aumitin, a new autophagy inhibitor that inhibits mitochondrial respiration by targeting complex I., Macroautophagy is a conserved eukaryotic process for degradation of cellular components in response to lack of nutrients. It is involved in the development of diseases, notably cancer and neurological disorders including Parkinson's disease. Small molecule autophagy modulators have proven to be valuable tools to dissect and interrogate this crucial metabolic pathway and are in high demand. Phenotypic screening for autophagy inhibitors led to the discovery of the novel autophagy inhibitor aumitin. Target identification and confirmation revealed that aumitin inhibits mitochondrial respiration by targeting complex I. We show that inhibition of autophagy by impairment of mitochondrial respiration is general for several mitochondrial inhibitors that target different mitochondrial complexes. Our findings highlight the importance of mitochondrial respiration for autophagy regulation.

Rule-based systems play a critical role in health and safety, where policies created by experts are usually formalised as rules. When dealing with increasingly large and dynamic sources of data, as in the case of Internet of Things (IoT) applications, it becomes important not only to efficiently apply rules, but also to reason about their applicability on datasets confined by a certain schema. In this paper we define the notion of a triplestore schema which models a set of RDF graphs. Given a set of rules and such a schema as input we propose a method to determine rule applicability and produce output schemas. Output schemas model the graphs that would be obtained by running the rules on the graph models of the input schema. We present two approaches: one based on computing a canonical (critical) instance of the schema, and a novel approach based on query rewriting.
