Abstract

Background/Objectives: The mechanisms of action of phosphine are diverse and include neurotoxicity, metabolic inhibition, and oxidative stress; however, its efficacy at low temperatures is unclear. Methods: Comparative metabolomics is suitable for investigating the response of the spotted-wing fly Drosophila suzukii to exposure toward a combination of cold stimuli and fumigant PH3. Results: Under this combined exposure, 52 metabolites exhibiting significant differences in stress were identified and their physiological roles were analyzed in the Drosophila metabolic pathway. Most metabolites were involved in amino acids, TCA cycle, and nucleic acids. In addition, the alteration levels of cell membrane lipids, such as glycerophospholipids, sphingolipids, and glycerolipids, clearly showed changes in the combined treatment compared to PH3 and low temperatures alone. Aconitic acid, a component of the TCA cycle, was completely inhibited by the combined treatment. Conclusions: These results suggest that treatment-specific indicators could be useful biomarkers to indicate the synergistic effects of PH3 and low temperature on energy metabolism. Abstract

Nilaparvata lugens, the brown planthopper (BPH), is a serious pest species. BPHs cause significant damage to rice plants in Korea as well as other countries in East and Southeast Asia. As BPHs cannot survive winter in Korea, they annually migrate into Korea from foreign countries. The BPHs found in Korea are believed to originate from China, but most BPHs in China are also known to originate from Southeast Asia. To understand the origin of Korean BPHs, we conducted a genotyping‐by‐sequencing (GBS) study. We sampled BPHs from five locations in Korea and five countries in Southeast Asia and analyzed the GBS and sequencing results using various methods based on the f statistics and admixture graph analyses. We confirmed that the domestic BPHs shared a greater genetic drift than the BPHs from Southeast Asian populations over several years, implying that a continuous genetic substratum of Korea exists. This genetic substratum is genetically closer to BPHs from the southern part (Thailand and Cambodia) of Southeast Asia than to BPHs from the northern part (Bhutan, Myanmar and Laos) of Southeast Asia. In addition, direct gene flows from Southeast Asia seem possible, so Korea is considered a hotspot where diverse BPH populations mix. Therefore, the origin of Korean BPHs extends beyond China and as far as southern Southeast Asia. This result will help to understand and control the population dynamics of the Korean BPH population. 32. Effects of Electron Beam Irradiation on Acaricide-Resistant and Susceptible Strains of Tetranychus urticae (Acari: Tetranychidae).

Abstract

Simple Summary: A lab-selected strain of the two-spotted spider mite, Tetranychus urticae Koch, exhibited strong resistance to acequinocyl, bifenazate, and etoxazole in comparison with a susceptible strain. This study compared the development and reproduction in acaricide-resistant and -susceptible strains of T. urticae by electron beam irradiation. Both T. urticae strains had similar radiosensitivities and 400 Gy could be used as a phytosanitary treatment for both strains of this species. The objective of our study was to compare the effects of electron beam irradiation on the development and reproduction of Tetranychus urticae strains that were susceptible or resistant to acequinocyl, bifenazate, and etoxazole. Electron beam irradiation (50–400 Gy) was applied to T. urticae eggs, nymphs, and adults, after which the hatching rate, the emergence rate, longevity, and fecundity were analyzed. Eggs of all the T. urticae strains were irradiated with 150 Gy, and none of them hatched. When protonymphs were irradiated with 300 Gy, oviposition and thus F1 hatchability in newly emerged adults were completely suppressed. The F1 hatching rate of irradiated adults began to markedly decrease with a dose of 200 Gy, and all strains experienced 100% inhibition at a dose of 400 Gy. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (Sodium dodecyl sulfate polyacrylamide gel electrophoresis) showed substantial reductions in the levels of a 50 kDa protein in the susceptible and three resistant strains of T. urticae after irradiation with 400 Gy. These results indicate that a dose of 400 Gy was required to inhibit the normal development and reproduction of T. urticae, and susceptible and acaricide-resistant strains of T. urticae had identical radiosensitivities.
