1. Evaluating the Influence of Temporal Context on Automatic Mouse Sleep Staging through the Application of Human Models

Abstract

In human sleep staging models, augmenting the temporal context of the input to the range of tens of minutes has recently demonstrated performance improvement. In contrast, the temporal context of mouse sleep staging models is typically in the order of tens of seconds. While long-term time patterns are less clear in mouse sleep, increasing the temporal context further than that of the current mouse sleep staging models might still result in a performance increase, given that the current methods only model very short term patterns. In this study, we examine the influence of increasing the temporal context in mouse sleep staging up to 15 minutes in three mouse cohorts using two recent and high-performing human sleep staging models that account for long-term dependencies. These are compared to two prominent mouse sleep staging models that use a local context of 12 s and 20 s, respectively. An increase in context up to 28 s is observed to have a positive impact on sleep stage classification performance, especially in REM sleep. However, the impact is limited for longer context windows. One of the human sleep scoring models, L-SeqSleepNet, outperforms both mouse models in all cohorts. Abstract

Summary The entorhinal cortex consists of several important cell types including, the grid cells, speed cells, border cells and head-direction cells and is important for memory, spatial navigation and perception of time. Here, we trace in detail the development of the entorhinal cortex. Using single-cell profiling we provide unique transcriptional signatures for glia, excitatory and inhibitory neurons existing in the region, including RELN+ cells in layer (L) II and superficial pyramidal neurons. We identified a sandwich layered cortex, where LII emerges prior to LIII and superficial cells maintain a deep layer molecular identity after birth. Our findings contribute to the understanding of the formation of the brain’s cognitive memory and spatial processing system and provides insight into the transcriptional identity and spatial position of the entorhinal cells.

Abstract

γ‐Aminobutyric acid (GABA) acting through heteropentameric GABAA receptors plays a pivotal role in the sleep‐promoting circuitry. Whereas the role of the different GABAA receptor α‐subunits in sleep regulation and in mediating the effect of benzodiazepines for treatment of insomnia is well‐described, the β‐subunits are less studied. Here we report the first study characterizing sleep in mice lacking the GABAA receptor β1‐subunit (β1−/− mice). We show that β1−/− mice have a distinct and abnormal sleep phenotype characterized by increased delta power in non‐rapid eye movement (NREM) sleep and decreased theta activity in rapid eye movement (REM) sleep compared to β1+/+ mice, without any change in the overall sleep‐wake architecture. From GABAA receptor‐specific autoradiography, it is further demonstrated that functional β1‐subunit‐containing GABAA receptors display the highest binding levels in the hippocampus and frontal cortex. In conclusion, this study suggests that the GABAA receptor β1‐subunit does not play an important role in sleep initiation or maintenance but instead regulates the power spectrum and especially the expression of theta rhythm. This provides new knowledge on the complex role of GABAA receptor subunits in sleep regulation. In addition, β1−/− mice could provide a useful mouse model for future studies of the physiological role of delta and theta rhythms during sleep. [ABSTRACT FROM AUTHOR]

Abstract

Narcolepsy type 1, a neurological sleep disorder strongly associated with Human Leukocyte Antigen (HLA-)DQB1*06:02, is caused by the loss of hypothalamic neurons producing the wake-promoting neuropeptide hypocretin (hcrt, also known as orexin). This loss is believed to be caused by an autoimmune reaction. To test whether hcrt itself could be a possible target in the autoimmune attack, CD4 + T-cell reactivity towards six different 15-mer peptides from prepro-hypocretin with high predicted affinity to the DQA1*01:02/DQB1*06:02 MHC class II dimer was tested using EliSpot in a cohort of 22 narcolepsy patients with low CSF hcrt levels, and 23 DQB1*06:02 positive healthy controls. Our ELISpot assay had a detection limit of 1:10,000 cells. We present data showing that autoreactive CD4 + T-cells targeting epitopes from the hcrt precursor in the context of MHC-DQA1*01:02/DQB1*06:02 are either not present or present in a frequency is < 1:10,000 among peripheral CD4 + T-cells from narcolepsy type 1 patients. Abstract

The sleep disorder narcolepsy with cataplexy is characterized by a highly specific loss of hypocretin (orexin) neurons, leading to the hypothesis that the condition is caused by an immune or autoimmune mechanism. All genetic variants associated with narcolepsy are immune-related. Among these are single nucleotide polymorphisms in the P2RY11-EIF3G locus. It is unknown how these genetic variants affect narcolepsy pathogenesis and whether the effect is directly related to P2Y11 signalling or EIF3G function. Exome sequencing in 18 families with at least two affected narcolepsy with cataplexy subjects revealed non-synonymous mutations in the second exon of P2RY11 in two families, and P2RY11 re-sequencing in 250 non-familial cases and 135 healthy control subjects revealed further six different non-synonymous mutations in the second exon of P2RY11 in seven patients. No mutations were found in healthy controls. Six of the eight narcolepsy-associated P2Y11 mutations resulted in significant functional deficits in P2Y11 signalling through both Ca2+ and cAMP signalling pathways. In conclusion, our data show that decreased P2Y11 signalling plays an important role in the development of narcolepsy with cataplexy. [ABSTRACT FROM AUTHOR]
