1. Chronic Astrocytic TNFα Production in the Preoptic-Basal Forebrain Causes Aging-like Sleep-Wake Disturbances in Young Mice.

Abstract

Sleep disruption is a frequent problem of advancing age, often accompanied by low-grade chronic central and peripheral inflammation. We examined whether chronic neuroinflammation in the preoptic and basal forebrain area (POA-BF), a critical sleep-wake regulatory structure, contributes to this disruption. We developed a targeted viral vector designed to overexpress tumor necrosis factor-alpha (TNFα), specifically in astrocytes (AAV5-GFAP-TNFα-mCherry), and injected it into the POA of young mice to induce heightened neuroinflammation within the POA-BF. Compared to the control (treated with AAV5-GFAP-mCherry), mice with astrocytic TNFα overproduction within the POA-BF exhibited signs of increased microglia activation, indicating a heightened local inflammatory milieu. These mice also exhibited aging-like changes in sleep-wake organization and physical performance, including (a) impaired sleep-wake functions characterized by disruptions in sleep and waking during light and dark phases, respectively, and a reduced ability to compensate for sleep loss; (b) dysfunctional VLPO sleep-active neurons, indicated by fewer neurons expressing c-fos after suvorexant-induced sleep; and (c) compromised physical performance as demonstrated by a decline in grip strength. These findings suggest that inflammation-induced dysfunction of sleep- and wake-regulatory mechanisms within the POA-BF may be a critical component of sleep-wake disturbances in aging.

Abstract

Driven by life-science applications, mega-electron-volt Scanning Transmission Electron Microscope (MeV-STEM) has been proposed to image thick biological samples. The high penetration of inelastic scattering signals of MeV electrons could make the MeV-STEM an appropriate microscope for biological samples as thick as 10 {\mu}m or more with a nanoscale resolution, considering the effect of electron energy, beam broadening and low-dose limit on resolution. The best resolution is inversely related to the sample thickness and changes from 6 nm to 24 nm when the sample thickness increases from 1 {\mu}m to 10 {\mu}m. To achieve such a resolution in STEM, the imaging electrons must be focused on the specimen with a nm size and a mrad semi-convergence angle. This requires an electron beam emittance of a few picometer, which is ~1,000 times smaller than the presently achieved nm emittance, in conjunction with less than 10-4 energy spread and 1 nA current. We numerically simulated two different approaches that are potentially applicable to build a compact MeV-STEM instrument: 1) DC gun, aperture, Superconducting radio frequency (SRF) cavities, and STEM column; 2) SRF gun, aperture, SRF cavities, and STEM column. Beam dynamic simulations show promising results, which meet the needs of an MeV-STEM, a few picometer emittance, less than 10-4 energy spread, and 0.1-1 nA current from both options. Also, we designed a compact STEM column based on permanent quadrupole quintuplet not only to demagnify the beam size from 1 {\mu}m at the source point to 2 nm at the specimen, but also to provide the freedom of changing the magnifications at the specimen and a scanning system to raster the electron beam across the sample with a step size of 2 nm and the repetition rate of 1 MHz. This makes it possible to build a compact MeV-STEM and use it to study thick, large-volume samples in cell biology.

Abstract

Traveling wave SRF cavity is a new technology and requires a multi-stage process for development. Conceptual designs have been proposed to adopt TW resonance in an SRF cavity The early stages of developments have been funded by several SBIR grants to Euclid Techlabs which were completed in collaboration with Fermilab. A 3-cell proof-of-principle TW cavity was fabricated as part of that and demonstrated the TW resonance excitation at room temperature. A TW resonance control tuner for the 3-cell was also fabricated and the preliminary tests were performed. 15. Organic carbon as a factor of iodine differentiation of the soil cover in the Bryansk and Oryol regions by iodine availability

Abstract

The distribution of stable iodine and humus in the upper horizon (0–20 cm) of various types of soils of the Bryansk and Oryol regions used as pastures has been studied. Soil sampling was carried out with a hand stick drill in layers in the depth interval: 0–5 cm, 5-10 cm and 10–20 cm. Iodine content was determined by kinetic rhodanide-nitrite method. Corg content was determined by the bichromate method of Tyurin. The work includes data from both long-term studies of the iodine content in the soil cover of the Bryansk region (2008–2020) and new data from expeditionary studies in the Oryol region (2022). It is shown that the soils of the Bryansk pastures are depleted of iodine and organic carbon (OC) in comparison with the Oryol region, which corresponds to the change of soil cover composition from west to east. The humus content in soils was determined and statistically significant correlation between iodine and C org. content in pasture soils were obtained (from r = 0.28 in Chernozems to r = 0.49 in Haplic Albeluvisols; in addition, for the whole sample (n = 48) the correlation coefficient ranged from 0.31 in the 0–5 cm layer to 0.43 in the 10–20 cm layer). The dependence of iodine accumulation on the OC content in different types of soils (Chernozems, Grey-Luvic Phaeozems, Umbric Albeluvisols, Haplic Albeluvisols) and their hydromorphism has been established. The identified differences in soil iodine fixation should be considered as they may influence iodine intake levels in pasture animal milk and local rations.

Abstract

Abstract Identifying high-affinity antibodies in human serum is challenging due to extremely low number of circulating B cells specific to the desired antigens. Delays caused by a lack of information on the immunogenic proteins of viral origin hamper the development of therapeutic antibodies. We propose an efficient approach allowing for enrichment of high-affinity antibodies against pathogen proteins with simultaneous epitope mapping, even in the absence of structural information about the pathogenic immunogens. To screen therapeutic antibodies from blood of recovered donors, only pathogen transcriptome is required to design an antigen polypeptide library, representing pathogen proteins, exposed on the bacteriophage surface. We developed a two-dimensional screening approach enriching lentiviral immunoglobulin libraries from the convalescent or vaccinated donors against bacteriophage library expressing the overlapping set of polypeptides covering the spike protein of SARS-CoV-2. This platform is suitable for pathogen-specific immunoglobulin enrichment and allows high-throughput selection of therapeutic human antibodies.

Abstract

Combinatorial optimization problems require an exhaustive search to find the optimal solution. A convenient approach to solving combinatorial optimization tasks in the form of Mixed Integer Linear Programs is Branch-and-Bound. Branch-and-Bound solver splits a task into two parts dividing the domain of an integer variable, then it solves them recursively, producing a tree of nested sub-tasks. The efficiency of the solver depends on the branchning heuristic used to select a variable for splitting. In the present work, we propose a reinforcement learning method that can efficiently learn the branching heuristic. We view the variable selection task as a tree Markov Decision Process, prove that the Bellman operator adapted for the tree Markov Decision Process is contracting in mean, and propose a modified learning objective for the reinforcement learning agent. Our agent requires less training data and produces smaller trees compared to previous reinforcement learning methods., Comment: Submitted to NeurIPS 2023

Abstract

Muon neutrino and antineutrino disappearance probabilities are identical in the standard three-flavor neutrino oscillation framework, but CPT violation and non-standard interactions can violate this symmetry. In this work we report the measurements of $\sin^{2} \theta_{23}$ and $\Delta m_{32}^2$ independently for neutrinos and antineutrinos. The aforementioned symmetry violation would manifest as an inconsistency in the neutrino and antineutrino oscillation parameters. The analysis discussed here uses a total of 1.97$\times$10$^{21}$ and 1.63$\times$10$^{21}$ protons on target taken with a neutrino and antineutrino beam respectively, and benefits from improved flux and cross-section models, new near detector samples and more than double the data reducing the overall uncertainty of the result. No significant deviation is observed, consistent with the standard neutrino oscillation picture.

19. Optimal Motions of an Elastic Structure under Finite-Dimensional Distributed Control

Abstract

An optimal control problem for longitudinal motions of a thin elastic rod is considered. We suppose that a normal force, which changes piecewise constantly along the rod's length, is applied to the cross-section so that the positions of force jumps are equidistantly placed along the length. Additionally, external loads act at the rod ends. These distributed force and boundary loads are considered as control functions of the dynamic system. Given initial and terminal states at fixed time instants, the problem is to minimize the mean mechanical energy stored in the rod during its motion. We replace the classical wave equation with a variational problem solved via traveling waves defined on a special time-space mesh. For a uniform rod, the shortest admissible time horizon is estimated exactly, and the exact optimal control law is symbolically found in a recurrent way.

Abstract

This paper reports the first measurement of muon neutrino charged-current interactions without pions in the final state using multiple detectors with correlated energy spectra at T2K. The data was collected on hydrocarbon targets using the off-axis T2K near detector (ND280) and the on-axis T2K near detector (INGRID) with neutrino energy spectra peaked at 0.6 GeV and 1.1 GeV respectively. The correlated neutrino flux presents an opportunity to reduce the impact of the flux uncertainty and to study the energy dependence of neutrino interactions. The extracted double-differential cross sections are compared to several Monte Carlo neutrino-nucleus interaction event generators showing the agreement between both detectors individually and with the correlated result., Comment: Updated discussion in Sec. Abstract

In this paper, we study the problem of recovering two unknown signals from their convolution, which is commonly referred to as blind deconvolution. Reformulation of blind deconvolution as a low-rank recovery problem has led to multiple theoretical recovery guarantees in the past decade due to the success of the nuclear norm minimization heuristic. In particular, in the absence of noise, exact recovery has been established for sufficiently incoherent signals contained in lower-dimensional subspaces. However, if the convolution is corrupted by additive bounded noise, the stability of the recovery problem remains much less understood. In particular, existing reconstruction bounds involve large dimension factors and therefore fail to explain the empirical evidence for dimension-independent robustness of nuclear norm minimization. Recently, theoretical evidence has emerged for ill-posed behavior of low-rank matrix recovery for sufficiently small noise levels. In this work, we develop improved recovery guarantees for blind deconvolution with adversarial noise which exhibit square-root scaling in the noise level. Hence, our results are consistent with existing counterexamples which speak against linear scaling in the noise level as demonstrated for related low-rank matrix recovery problems.

Abstract

The T2K experiment presents new measurements of neutrino oscillation parameters using $19.7(16.3)\times10^{20}$ protons on target (POT) in (anti-)neutrino mode at the far detector (FD). Compared to the previous analysis, an additional $4.7\times10^{20}$ POT neutrino data was collected at the FD. Significant improvements were made to the analysis methodology, with the near-detector analysis introducing new selections and using more than double the data. Additionally, this is the first T2K oscillation analysis to use NA61/SHINE data on a replica of the T2K target to tune the neutrino flux model, and the neutrino interaction model was improved to include new nuclear effects and calculations. Frequentist and Bayesian analyses are presented, including results on $\sin^2\theta_{13}$ and the impact of priors on the $\delta_\mathrm{CP}$ measurement. Both analyses prefer the normal mass ordering and upper octant of $\sin^2\theta_{23}$ with a nearly maximally CP-violating phase. Assuming the normal ordering and using the constraint on $\sin^2\theta_{13}$ from reactors, $\sin^2\theta_{23}=0.561^{+0.021}_{-0.032}$ using Feldman--Cousins corrected intervals, and $\Delta{}m^2_{32}=2.494_{-0.058}^{+0.041}\times10^{-3}~\mathrm{eV^2}$ using constant $\Delta\chi^{2}$ intervals. The CP-violating phase is constrained to $\delta_\mathrm{CP}=-1.97_{-0.70}^{+0.97}$ using Feldman--Cousins corrected intervals, and $\delta_\mathrm{CP}=0,\pi$ is excluded at more than 90% confidence level. A Jarlskog invariant of zero is excluded at more than $2\sigma$ credible level using a flat prior in $\delta_\mathrm{CP}$, and just below $2\sigma$ using a flat prior in $\sin\delta_\mathrm{CP}$. When the external constraint on $\sin^2\theta_{13}$ is removed, $\sin^2\theta_{13}=28.0^{+2.8}_{-6.5}\times10^{-3}$, in agreement with measurements from reactor experiments. These results are consistent with previous T2K analyses.

29. Cartograms in oncology: analysis of the regional situation

Abstract

Relevance. Over the past few years, there has been a worldwide trend towards an increase in cancer morbidity and mortality. Health officials and epidemiology researchers often use disease maps to identify potential disease groups. Cartograms are cartographic images of various territories that contain information not about the area of the occupied territory, but about a variable of interest to the reader, for example, the population or a specific disease. This tool has been used for data visualization for more than a century, but right now it is becoming increasingly popular to display patterns and directions of changes in the world around us. The introduction of cartograms into the healthcare sector is gradual, depending on the results obtained in the development of various types of cartograms. It is difficult to fully assess the importance of collecting and analyzing information about the course of cancer. However, with the improvement of this system, good results can be achieved in preventing the occurrence of oncological diseases and increasing the level of cancer care provided. The purpose of this study is to analyze existing information about cartograms used in oncology. This publication discusses issues related to the need to use cartograms in oncoepidemiology, the causes of oncology, the advantages of using cartograms to consider a variety of diseases from simply infectious to HIV and technical issues related to geographical visualization, related problems and ways to solve them. Cartograms are a tool that clearly demonstrates the changes taking place in a small area, for example, in a certain area, within a large territory, for example, an entire republic in a format that can be perceived by all users and “readers” of cartograms. Conclusion. Oncology is a disease that spreads through the population by leaps and bounds. In this regard, it is necessary to collect as much statistical data as possible for more accurate predictions of the outcome of the disease, treatment of oncology and, most importantly, the preparation of preventive measures for this disease. Digitalization of healthcare in the form of digital cartograms will undoubtedly help in this.

Abstract

Relevance. Healthcare today faces the inevitability of change as it faces a wide range of challenges in the face of complete uncertainty. Through effective change management, success can be achieved in states, globalization and complementarity. The scientific community recommends developing approaches to changes in management that focus on coordinated work in the field of healthcare of personnel, continuous training, exchange of experience, and professional development of medical personnel not only as specialists in the field of oncology, but also as successful managers. It is also necessary not to ignore social factors, namely the so-called “power of compassion”, which is expressed in the introduction and improvement of palliative care. The purpose of this article is to analyze the structure of management and organization of cancer care and proposals for improving the quality of treatment and care for cancer patients. This publication is devoted to a comprehensive analysis of the problems and prospects for the development of cancer care. Oncology is considered by us as one of the largest health problems. In this regard, ideas have been proposed on the need to modernize and optimize the processes of providing oncological care. According to the literature, important aspects in this process are the coordination of the actions of medical personnel, both doctors and nurses, and patients; their qualifications as oncology specialists and managers; provision and implementation of palliative care everywhere; quality control of provided medical services carried out directly by medical personnel. However, all of these components of the cancer care structure face the reality of a lack of actual time for medical personnel to train management skills, a lack of resources for regional health structures to implement palliative care and train staff in management skills. A positive aspect in this situation is the desire of the scientific community to improve itself and improve the skills of both oncologists and managers. Conclusion.The creation of specialized courses on management in the field of oncology and continuous international training through distance education and telemedicine is an important milestone in resolving these issues.

Abstract

The ventrolateral preoptic area (VLPO) contains GABAergic sleep-active neurons. However, the extent to which these neurons are involved in expressing spontaneous sleep and homeostatic sleep regulatory demands is not fully understood. We used calcium (Ca2+) imaging to characterize the activity dynamics of VLPO neurons, especially those expressing the vesicular GABA transporter (VGAT) across spontaneous sleep-waking and in response to homeostatic sleep demands. The VLPOs of wild-type and VGAT-Cre mice were transfected with GCaMP6, and the Ca2+ fluorescence of unidentified (UNID) and VGAT cells was recorded during spontaneous sleep-waking and 3 h of sleep deprivation (SD) followed by 1 h of recovery sleep. Although both VGAT and UNID neurons exhibited heterogeneous Ca2+ fluorescence across sleep-waking, the majority of VLPO neurons displayed increased activity during nonREM/REM (VGAT, 120/303; UNID, 39/106) and REM sleep (VGAT, 32/303; UNID, 19/106). Compared to the baseline waking, VLPO sleep-active neurons (n = 91) exhibited higher activity with increasing SD that remained elevated during the recovery period. These neurons also exhibited increased Ca2+ fluorescence during nonREM sleep, marked by increased slow-wave activity and REM sleep during recovery after SD. These findings support the notion that VLPO sleep-active neurons, including GABAergic neurons, are components of neuronal circuitry that mediate spontaneous sleep and homeostatic responses to sustained wakefulness.

Abstract

We analyze the synchronization dynamics of the thermodynamically large systems of globally coupled phase oscillators under Cauchy noise forcings with bimodal distribution of frequencies and asymmetry between two distribution components. The systems with the Cauchy noise admit the application of the Ott-Antonsen ansatz, which has allowed us to study analytically synchronization transitions both in the symmetric and asymmetric cases. The dynamics and the transitions between various synchronous and asynchronous regimes are shown to be very sensitive to the asymmetry degree whereas the scenario of the symmetry breaking is universal and does not depend on the particular way to introduce asymmetry, be it the unequal populations of modes in bimodal distribution, the phase delay of the Kuramoto-Sakaguchi model, the different values of the coupling constants, or the unequal noise levels in two modes. In particular, we found that even small asymmetry may stabilize the stationary partially synchronized state, and this may happen even for arbitrarily large frequency difference between two distribution modes (oscillator subgroups). This effect also results in the new type of bistability between two stationary partially synchronized states: one with large level of global synchronization and synchronization parity between two subgroups and another with lower synchronization where the one subgroup is dominant, having higher internal (subgroup) synchronization level and enforcing its oscillation frequency on the second subgroup. For the four asymmetry types, the critical values of asymmetry parameters were found analytically above which the bistability between incoherent and partially synchronized states is no longer possible.

The research note presents results of coupled RF and thermal simulation of a cryocooler conduction cooled 650 MHz SRF cavity made of bulk niobium and coated with Nb3Sn on the RF surface. The cavity is part of a particle accelerator design capable of producing 10 MeV, 1000 kW electron beam for application in wastewater treatment. Abstract

The T2K experiment widely uses plastic scintillator as a target for neutrino interactions and an active medium for the measurement of charged particles produced in neutrino interactions at its near detector complex. Over 10 years of operation the measured light yield recorded by the scintillator based subsystems has been observed to degrade by 0.9--2.2\% per year. Extrapolation of the degradation rate through to 2040 indicates the recorded light yield should remain above the lower threshold used by the current reconstruction algorithms for all subsystems. This will allow the near detectors to continue contributing to important physics measurements during the T2K-II and Hyper-Kamiokande eras. Additionally, work to disentangle the degradation of the plastic scintillator and wavelength shifting fibres shows that the reduction in light yield can be attributed to the ageing of the plastic scintillator., Comment: 29 pages, 18 figures.
