Physical exercise represents one of the most effective approaches to anti-aging. The goal of this study was to verify the effects of different modes and intensities of exercise on longevity proteins in the skeletal muscle in midlife. Middle-aged mice were trained in aerobic or resistance exercise for 8 weeks, and the changes in sirtuin 1 (SIRT1), adenosine monophosphate-activated kinase (AMPK), and mammalian target of rapamycin (mTOR) pathways in the skeletal muscle were evaluated by western blotting. Long-term exercise had no effects on skeletal muscle SIRT1 abundance, whereas high-intensity aerobic exercise increased AMPK phosphorylation and peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α). Low-intensity resistance exercise facilitated Akt/mTOR/p70 ribosomal protein kinase S6 (p70S6K) signaling but did not induce muscle hypertrophy. Conversely, high-intensity resistance exercise stimulated muscle hypertrophy without phosphorylation of mTOR signaling-related proteins. These results suggest the importance of setting exercise modes and intensities for anti-aging in midlife.

In operational weather forecasting, it is effective to aggregate information on all members of an ensemble forecast through the cluster analysis. The temporal coherence of ensemble members in each cluster is an important piece of information whether each cluster is well-divided. This information is especially important for forecasts where the target area is city or prefecture, i.e., Eulerian framework, because the members that compose the clusters can change over time due to the small target region. This study provided the temporal coherence of members in clusters by performing principal component analysis and cluster analysis on 3-hourly 500-hPa geopotential height forecasts and linking the clustering results in the time direction. The new method provided a consistently well-divided forecast scenario throughout the forecast period for Eulerian frame forecasts, as well as information on the temporal coherency of the members in the clusters, which was demonstrated effective through the experiment to the pre-select cluster with small errors. The application of the new technique to improve precipitation forecasts was also discussed.

[Purpose] The purpose of this study was to determine a preventive strategy for ankle sprain injury through a questionnaire survey of coaches of junior high school basketball teams. [Participants and Methods] The questionnaire comprised questions concerning coaches’ characteristics, prevention-related content, and collaboration with medical staff. We distributed the questionnaire to 108 coaches, of which 48 (41.7%) responded. [Results] The results revealed that none of the participant coaches had ever collaborated with medical staff. [Conclusion] Additional effort from medical staff is needed to hold seminars and contact coaches of junior high school teams in order to conduct an injury prevention program.

Purpose: Abdominal aortic calcification (AAC) is a well-known risk marker for cardiovascular disease. However, its clinical effect on patients who underwent radical surgery for colorectal cancer (CRC) stages II–III is unclear. This study aimed to analyze the associations between AAC and prognosis of patients with stage II–III CRC. Methods: To evaluate the effect of AAC on clinical outcomes, prognosis, and metastatic patterns of CRC, we analyzed 362 patients who underwent radical surgery for stage II–III CRC between 2010 and 2018. Results: The high AAC group had significantly worse overall survival (OS), cancer-specific survival (CSS), and recurrence-free survival (RFS) after propensity score matching to adjust for differences in baseline characteristics of patients and tumors. A novel synthetic strategy for large macrocyclic molecules using boroxine formation was developed. For this, the threefold intramolecular olefin metathesis of 3,5-bis(alkenyloxy)phenylboroxines with various lengths of alkenyl chains, formed by the dehydration of the corresponding boronic acid substrates, together with treatment with pinacol, was used to produce 39-, 45-, and 51-membered macrocyclic compounds with three boronate units. The boroxine moiety functions as a covalent template but can also be used to postmodify the macrocycle. Boroxine-templated macrocyclization implemented in this way does not require the addition of template molecules and simplifies the synthetic procedure.

Boroxines are well-known six-membered ring compounds and their fundamental properties such as aromaticity, Lewis acidity, photophysical property, interactions with anions, thermal stability, and reactivity have been investigated. Since the 2000s, boroxine formation has been paid much attention as a reversible covalent bond formation. Boronic acids (RB–(OH)2) reversibly trimerize to afford the tripodal structure of boroxines. The equilibrium between boronic acids and boroxines can be controlled simply by the addition or removal of water. The addition of N-donor Lewis bases also promotes the reversible formation of boroxine⋅N-donor Lewis base adducts. To date, by using this unique reversible trimerization, various self-assembled boroxine-based covalent architectures and polymeric materials have been reported. Boroxine-based architectures are categorized into three types as follows. (a) Tripodal structures by monoboroxine formation, (b) network structures by polyboroxine formation including covalent organic frameworks (= COFs) and dynamic polymer materials, and (c) discrete structures by oligoboroxine formation. Compared to mono or polyboroxine-based architectures (a) or (b), oligoboroxine-based discrete architectures (c) containing a precise number of boroxines are an emerging research subject and have the potential to create a novel type of discrete covalent architectures. Thus, in this review, we summarize recent progress including our works in this field.

Background Traumatic pancreatic injury with a main pancreatic duct injury has a high incidence of mortality and requires a prompt and appropriate treatment. However, the best approach, and treatment options, which may be limited, remains controversial especially for the elderly patients. Herein, we present a case of traumatic pancreatic injury in an elderly patient for whom pancreatoduodenectomy was safe and effective. Case presentation An 87-year-old man was diagnosed with a traumatic pancreatic injury with a main pancreatic duct injury. In addition, the horizontal segment of the duodenum was largely eradicated. There were no comorbidities, and his vital signs were stable. A pancreatoduodenectomy was performed. The postoperative course was uneventful, and he was discharged. Pathological examination revealed a primary follicular lymphoma of the duodenum. Conclusions This case demonstrated that pancreatoduodenectomy could be performed safely for a severe pancreatic injury in an elderly patient. However, special attention should be paid to select the optimal surgical procedure. Further, this was a rare case, as initially a primary follicular lymphoma of the duodenum was suspected as a duodenal injury coexisting with a traumatic pancreatic injury because of the increased duodenal thickness.

The post-transplant development of donor-specific antibodies (DSAs) initiates the antibody-mediated rejection, which is associated with an increased rate of graft loss. Therefore, risk prediction of de novo DSA (dnDSA) is important for understanding long-term prognostic implications for kidney transplantation outcomes. Cytotoxic T lymphocyte antigen-4 (CTLA-4), a cell surface molecule, suppresses T cell responses. Single-nucleotide polymorphisms (SNPs) in CTLA-4 are known to be associated with acute rejection; however, their association with dnDSA formation is not established. In the present study, we investigated the impact of CTLA-4 SNPs on dnDSA formation after kidney transplantation (KT) by analyzing three CTLA-4 SNPs (rs231775, rs3087243, and rs5742909) in 88 recipients. Patients with the GG genotype of CTLA-4 SNPs rs231775 and rs3087243 had higher rates of dnDSA formation than patients with the AA genotype or heterozygous genotypes. In conclusion, our findings indicate that CTLA-4 SNPs are predisposing factors for dnDSA formation after KT.

Boronic esters are known to be formed simply by mixing boronic acids and alcohols under neutral conditions, and the equilibrium is in favor of the boronic esters when 1,2- or 1,3-diols are employed as alcohols. By utilizing the dynamic nature of the boronic ester formation, our group successfully constructed unique boron-containing 3D structures, such as ring-shaped macrocycles, cages, and tubes, based on the boronic ester formation of various aromatic di-, tri-, or hexaboronic acids with an originally designed tetrol 1 containing two sets of fixed 1,2-diol units oriented on the same face of an indacene framework. Various functions of the obtained boronates were further pursued to disclose the characteristic features of this system. This personal account describes our self-assembled boronate system using tetrol 1 including synthesis, host-guest chemistry, kinetic connection, characteristic dynamic behaviors, and supramolecular catalysis.

An isolation condenser (IC) is a passive core cooling system in boiling water reactors. The cooling performance of IC is deteriorated when hydrogen generated in the core flows into the IC pipes. In this study, we conducted high pressure experiments using natural circulation loop with non-condensable gas injection, where helium was used to simulate hydrogen effect on the IC. The reaching distance of steam in the heat transfer tube was estimated by observing the region where nucleate boiling occurred on the outer surface of the heat transfer tube, and the heat transfer coefficient was estimated. The heat transfer coefficient hardly changed when helium was injected to the loop that indicates injected helium was not accumulated in the heat transfer tube. The system pressure at quasi-steady state increased with increasing amount of the injected helium. Since the differential pressure at the down comer section increased by helium injection, the injected helium may be accumulated in the section, leading to increment of the system pressure.

Hepatic epithelioid hemangioendothelioma is a rare neoplasm with a variable malignant potential and a high risk of recurrence. No general treatment guidelines have been established. Fortunately, we were able to minimize immunosuppressant after liver transplantation because of a full HLA‐matched case. There was no recurrence 1 year after treatment.

Helical structures were constructed by using π-spacer-bridged dimers of dibenzopyrrolo[1,2-a][1,8]naphthyridine, which has a highly fluorescent L-shaped π-extended skeleton. Three dimers with biphenylene (dimer 1), phenanthrene (dimer 2), and m-phenylene (dimer 3) spacers, as well as a fixed-helical dimer 4 where two quinolone rings were covalently cross-linked, were designed and prepared. 1 H NMR and ROESY spectra revealed that dimers 1 and 2 adopted helical forms in solution, whereas dimer 3 did not. The helical conformation of 1 was strengthened by addition of either polar or nonpolar solvents to the chloroform solution, which suggested that π-π stacking was the main contributor to the stabilization of the helical structure. All of the dimers, including fixed-helical dimer 4, emitted fluorescence with high quantum yields (ϕ=0.79-0.86).

This study extends Bayesian model averaging (BMA) to a form suitable for time series forecasts. BMA is applied to a three-member ensemble for temperature forecasts with a 1-h interval time series at specific stations. The results of such an application typically have a problematic characteristic. BMA weights assigned to ensemble members fluctuate widely within a few hours because BMA optimizations are independent at each lead time, which is incompatible with the spatiotemporal continuity of meteorological phenomena. To ameliorate this issue, a degree of correlation among different lead times is introduced by the extension of latent variables to lead times adjacent to the target lead time for the calculation of BMA weights and variances. This extension approach stabilizes the BMA weights, improving the performance of deterministic and probabilistic forecasts. Also, an investigation of the effects of this extension technique on the shapes of forecasted probability density functions showed that the extension approach offers advantages in bimodal cases. This extension technique may show promise in other applications to improve the performance of forecasts by BMA.

Isolation condensers (ICs) are important passive cooling systems in BWRs. After the Fukushima Daiichi Nuclear Power Station accident, concerns if the IC was able to restart with the inflow of hydrogen were arose. Because ICs lose heat removal ability when non-condensable gas inflow occurs, accurate evaluation of the effect is necessary. To develop analysis methods, as an initial stage, experiments and analyses considering only high-pressure steam and water were conducted. The experiment was done by an isolation condenser simulator which contains an accumulator with heaters inside, and a heat transfer tube. From the experiment, all steam was condensed at the heat transfer tube and the approximate position of complete condensation was confirmed from the temperature distribution and the observation. The experiment provided data such as temperature distribution, natural circulation flow rate, and pressure to compare with the analysis. The analyses were conducted for 4 cases of void fraction values at the heat transfer tube inlet and found that it has a high sensitivity to condensation. The reason is estimated to be the difference in inflow velocity that strongly depends on the void fraction even if the mass flow rate is constant. And the initial condition of the liquid film also affected condensation process. Heat removal at the section before the heat transfer tube should be considered to adjust void fraction at the inlet of heat transfer tube.

Human leukocyte antigen (HLA) molecular mismatch (MM) analysis improves the prediction of clinical outcomes in kidney transplantation compared with prediction via traditional antigen MM. However, it remains unclear whether the level of MM can be used for risk stratification among liver transplantation (LT) recipients. A retrospective observational study of 45 living donor LTs was performed to evaluate eplet MM as a risk factor for both T cell-mediated rejection (TCMR) in the first month and de novo donor-specific antibody (dnDSA) formation. A total of 9 (20%) patients displayed TCMR. HLA-A, HLA-B, HLA-C, and HLA-DRB1 eplet MM numbers were not associated with TCMR. By contrast, HLA-DQB1 eplet MM (DQB1-EpMM) number was significantly high in patients with TCMR. The predicted indirectly recognizable HLA epitopes (PIRCHE-II) score for the HLA-DQB1 locus (DQB1-PIRCHE-II) was also significantly higher in the TCMR group than in the no-TCMR group. There was a high probability for TCMR to occur with either a DQB1-EpMM ≥7 or a DQB1-PIRCHE-II ≥13. Background Hepatectomy for resectable colorectal liver metastasis (CRLM) is recommended. However, the efficacy of upfront hepatectomy without neoadjuvant chemotherapy (NAC) is unclear due to the uncertainty of perioperative systemic chemotherapy. Moreover, it is crucial to predict the prognosis when considering perioperative chemotherapy. This study evaluated the impact of neoadjuvant chemotherapy on the prognosis of patients with resectable CRLM and assessed the usefulness of Beppu’s nomogram for predicting prognosis. Methods This retrospective study identified 88 consecutive inpatients who underwent primary hepatic resection for CRLM; 58 received neoadjuvant chemotherapy and 30 underwent upfront surgery. Factors associated with recurrence-free survival were identified via univariate and multivariate analysis. Furthermore, propensity score analysis using inverse probability of treatment weighting (IPTW) was performed. Results On univariate analysis, poor recurrence-free survival was associated with multiple tumors, advanced primary tumor stage, vascular invasion by the primary tumor, a Beppu’s nomogram score ≥ 6, and neoadjuvant chemotherapy. On multivariate analysis, a Beppu’s nomogram score ≥ 6 and neoadjuvant chemotherapy were independent risk factors for recurrence. Neoadjuvant chemotherapy recipients had a higher incidence of lymph node metastasis and vascular invasion than non-recipients. Propensity score analysis revealed no significant difference in the recurrence-free survival rate between these groups. Conclusions Our results show that upfront hepatectomy without neoadjuvant chemotherapy can be considered for resectable CRLM treatment. Beppu’s nomogram score can be a tool for predicting the prognosis of patients with CRLM.
