2-(3-Phenyl-4,5-dihydro-1H-1,2,4-triazol-5-on-4-yl)-azomethine) benzoic acid molecule has been optimized using the 6-311G(d,p) basis set of DFT(B3LYP) and HF methods. Thus, the most stable geometrical conformer of the compound with different methods was obtained. 1H/ 13C-NMR chemical shifts, vibrational wavenumbers, thermodynamic parameters, molecular geometric optimizations (bond angles, bond lengths), HOMO-LUMO analyses, electronic properties, mulliken charges, dipole moment, total energy of the molecule were studied using Gaussian 09W software package. Gaussian results are drawn graphically with GaussView5.0 program. All quantum chemical computations were performed in the B3LYP functional of DFT method ve HF at the 6-311G(d,p) basis set. 1H/ 13C Nuclear magnetic Resonance spectral values were calculated in gas phase and in DMSO solvent according to GIAO method. Theoretical spectral values of the molecule were compared with experimental data obtained from the literature. In the Veda4f program, vibration types were determined and harmonic vibration frequencies were scaled with certain scale factors of different methods. In addition, thermodynamics properties (heat capacity CV0, entropy S0 and enthalpy H0), electronic properties (electronegativity (χ), electron affinity (A), global hardness (η), softness (σ), ELUMO-EHOMO energy gap (ΔEg) and ionization potential (I) were calculated. Also, the molecular surfaces such as the electron spin potential (ESP), molecular electrostatic potential (MEP), the total density, the electron density, the electrostatic potential of the molecule were visualized., 2-(3-Fenil-4,5-dihidro-1H-1,2,4-triazol-5-on-4-il)-azometin benzoik asit molekülü DFT (B3LYP) ve HF yöntemlerinin 6-311G (d, p) temel seti kullanılarak optimize edilmiştir. Ayrıca, elektron spin potansiyeli (ESP), moleküler elektrostatik potansiyel (MEP), toplam yoğunluk, elektron yoğunluğu, molekülün elektrostatik potansiyeli gibi moleküler yüzeyler görselleştirildi.

Monitoring the malignant tumors via cancer biomarkers is very significant process. Nonetheless, the practical clinical applications need selective and sensitive analytical methods/techniques. In this study, a novel sandwich type immunosensor based on surface-enhanced raman scattering (SERS) was presented including 4-mercaptobenzoic acid labeled MoS2 nanoflowers@Au nanoparticles (MoS2 NFs@Au NPs/ MBA) as CEASERS tag and Fe3O4@Au nanoparticles functionalized delaminated Ti3C2Tx MXene (Fe3O4 NPs@Au NPs/d-Ti3C2TX MXene) as SERS magnetic supporting substrate for carcinoembryonic antigen (CEA) detection. Especially, the determination of single molecule by using SERS method enables early diagnosis of major diseases. In addition, this technique can be utilized for multiplex analyzes owing to narrow well-resolved peaks. The prepared CEASERS tag and SERS magnetic supporting substrate were characterized by scanning electron microscope (SEM), x-ray diffraction (XRD) method, x-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM) and fourier transform infrared spectroscopy (FTIR). A linearity of 0.0001-100.0 ng mL-1 was observed with high sensitivity. Finally, sandwich type immunosensor demonstrated good selectivity and stability for target CEA recognition in plasma sample.

Bu çalışmada, 3-(n-Propil)-4-(4-izopropilbenzilidenamino)-4,5-dihidro-1H-1,2,4-triazol-5-on Gaussian 09W programında Yoğunluk Fonksiyonel Teorisi (DFT) ve Hartee Fock (HF) metotları ile 6-31G(d,p) temel seti kullanılarak incelenmiştir. Bunun için öncelikle, molekülün en kararlı üç boyutlu formu GaussView5.0 programı ile çizilmiştir. Bu optimize olmuş molekülün yapısından yola çıkarak molekülün geometrik özellikleri (bağ açı ve uzunluğu), termodinamik özellikleri entropi (S), termal enerji (E), termal kapasite (CV)), elektronik özellikleri iyonlaşma potansiyeli (I), kimyasal sertlik-yumuşaklık, elektronegatiflik (χ), elektron ilgisi (A), elektrofilik ve nükleofilik indeks), yüksek enerjili dolu moleküler orbital (HOMO) ve düşük enerjili boş moleküler orbital(LUMO) enerjileri ve bu orbitaller arasındaki enerji farkı ΔEg, spektroskopik özellikleri (FT-IR, 13C/1H-NMR) DFT/ HF 6-31G(d,p) ile hesaplanmıştır. FT-IR titreşim frekans değerleri hesaplamasında Veda4f programından faydalanılmıştır. Teorik IR tireşim frekans değerleri ile literatürde kayıtlı olan deneysel IR değerleri karşılaştırılmıştır. 13C/1H-NMR spektral verilerinin teorik hesaplaması gaz ve çözücü (DMSO) fazında yapılmıştır ve deneysel değerler ile karşılaştırılarak regrasyon analizleri yapılmıştır. Bulunan R2 değerleri mukayese edilerekregrasyon grafikleri oluşturulmuştur. Bunlardan başka, molekülün toplam enerjisi, dipol momenti, mulliken atomik yük değerleri, moleküler elektron potansiyel, toplam yoğunluk, molekülün yüzey haritaları belirlenmiştir. First, the most stable three-dimensional shape of the molecule was drawn using the GaussView5.0 program. Based on the structure of this optimized molecule,"the geometric properties (bond angle and length), the thermodynamic properties entropy (S), thermal energy (E), thermal capacity (CV), the electronic properties (ionization potential (I) and chemical hardness softness, electronegativity (χ), electron affinity (A), electrophilic and nucleophilic index), the highest occupied molecular orbital (HOMO) and the lowest unoccupied molecular orbital (LUMO) energies and energy difference between these orbitals ΔEg, spectroscopic properties"(FT-IR, 13C/ 1H-NMR, UV-vis) were calculated with DFT/ HF 6-31G(d,p). FT-IR vibration frequency values were calculated from the Veda4f program. The theoretical IR vibration frequency values were compared with the experimental IR values recorded in the literature. Theoretical calculations of 13C/ 1H-NMR spectral data were performed in gas and solvent (DMSO) phase and regression analyzes were performed by comparison with experimental values. R2 values were compared and regression graphs were created. In addition, the total energy of the molecule, dipole moment, Mulliken atomic charge values, molecular electron potential, total density, surface maps of molecule were determined. The electrophilic and nucleophilic regions of the structure were confirmed.

3-Metil-4-(3-metil-2-tiyenilmetilenamino)-4,5-dihidro-1H-1,2,4-triazol-5-on molekülü HF/ B3LYP6-311G(d,p) temel setleri kullanılarak optimize edilmiştir. Bu çalışmada, beş adet 3-alkil-4-(2-asetoksi-3-metoksibenzilidenamino)-4,5-dihidro-1H-1,2,4-triazol-5-on bileşiğinin teorik özellikleri B3LYP/6-311G(d,p) ve HF/6-311G(d,p) temel setleri kullanılarak incelenmiştir. İlk olarak, her bir molekül B3LYP/6-311G(d,p) ve HF/6-311G(d,p) temel setleri kullanılarak optimize edilmiştir. Gaussian G09W paket programı kullanılarak GIAO metoduna göre moleküllerin 1H-NMR ve 13C-NMR isotropik kayma değerleri hesaplanmıştır. IR verilerinin belirlenmesinde veda4f programı kullanılmıştır. İncelenen her bir molekülün iki farklı temel set ile IR (infrared) titreşim frekans değerleri gaz fazında hesaplanmış ve uygun uyum faktörleri ile çarpılmıştır. Bileşiklerin bağ uzunlukları, mulliken yükleri, en yüksek dolu moleküler orbital enerjisi (HOMO) ve en düşük boş moleküler orbital enerjisi (LUMO), dipol moment değerleri de hesaplanmıştır., In this study, theoretical properties of five 3-alkyl-4-(2-aceoxy-3-methoxybenzylidenamino)-4,5- dihydro-1H-1,2,4- triazol-5-ones were investigated using B3LYP and HF basis sets. Firstly, each molecule were optimized by using the B3LYP/6-311G(d,p) and HF/6-311G(d,p) basis sets. 1H-NMR and 13C-NMR isotropic shift values of molecules were calculated by the method of GIAO using the program package Gaussian G09W. In this study, 1-(morpholine-4-yl-methyl)-3-benzyl-4-(4-isopropylbenzylidenamino)-4,5-dihydro-1H-1,2,4-triazol-5-one (1) was synthesized by the reaction of compound 3-benzyl-4-(4-isopropylbenzylidenamino)-4,5-dihydro-1H-1,2,4-triazol-5-one with morpholine and formaldehyde. The structure of compound 1 was determined by IR, 1H-NMR, 13C-NMR spectral data. Then, the synthesized compound 1 was optimized by using the B3LYP/6-31G (d,p) and HF/6-31G (d,p) basis sets. 1H-NMR and 13C-NMR isotropic shift values, IR absorption frequencies, bond angles, bond lengths, the HOMO-LUMO energy, electronegativity and mulliken charges were calculated theoretically by using the program package Gaussian G09W. In addition, IR, 1H-NMR, 13C-NMR theoretical spectral data were compared with certain experimental data.

Araştırmada farklı pişirme yöntemlerinin (mikrodalgada pişirme, tavada kızartma, fırında pişirme, ısıtıcı plaka üzerinde pişirme ve mangalda pişirme) bazı yağlı (somon, uskumru ve sardalya) ve yağsız (mezgit, alabalık ve levrek) balıklarda çeşitli kalite kriterleri ve heterosiklik aromatik amin (HAA) oluşumu üzerine etkileri incelenmiştir. Isıl işlem görmemiş balıklarda su, yağ içeriği, pH ve TBARS değerleri belirlenmiş, ısıl işlem gören örneklerde ise su, pH, TBARS ve HAA analizleri yapılmıştır. HAA ekstraksiyonu, katı faz ekstraksiyon yöntemine göre gerçekleştirilmiş ve HPLC-DAD ile okuma işlemi yapılmıştır.Araştırma sonucunda pişirme yöntemlerinin; örneklerin su içeriği ve pH değeri üzerine çok önemli (p0,05) belirlenmiştir. Balık türünün; su içeriği, pH ve TBARS değerleri üzerine çok önemli (p0,05) tespit edilmiştir. Öte yandan pişirme yöntemi ve balık türü interaksiyonunun; örneklerin sadece su içeriği üzerine çok önemli (p0,05) belirlenmiştir. Analizi yapılan örneklerin hiçbirinde MeAαC bileşiği tespit edilemezken, örneklerin değişen miktarlarda IQx (0,22 ng/g'a kadar), IQ (0,71 ng/g'a kadar), MeIQx (3,71 ng/g'a kadar), MeIQ (0,42 ng/g'a kadar), 7,8-DiMeIQx (0,06 ng/g'a kadar), 4,8-DiMeIQx (0,11 ng/g'a kadar), PhIP (2,67 ng/g'a kadar) ve AαC (0,03 ng/g'a kadar) içerdiği tespit edilmiştir. Pişirilen örneklerin toplam HAA içeriğinin nd-5,72 ng/g arasında değiştiği ve yapılan istatistiksel değerlendirme neticesinde en yüksek toplam HAA içeriğinin mangalda pişirilmiş örneklere ait olduğu belirlenmiştir. İlaveten tavada kızartılmış somon ve levrek balıklarının toplam HAA içeriğinin, HAA'ların tespit edilme limitinin altında olduğu da belirlenmiştir. In the present study, the effects of different cooking methods (microwave, pan-frying, oven, hot plate and barbecuing) on some quality criteria and the formation of heterocyclic aromatic amines (HCAs) in various fatty (salmon, mackerel and sardine) and lean fishes (whiting, trout and sea bass) were investigated. Water, crude fat, pH and TBARS values were determined in raw samples, while water, pH, TBARS and HCAs analyses were done in cooked samples. HCAs extraction was carried out according to solid phase extraction method and HPLC-DAD was used for the analysis of HCAs in the samples.It was determined that cooking methods had very significant (p0,05) on TBARS value of the samples. Çalışma için gerekli 3-metil-4-amino-4,5-dihidro-1H-1,2,4-triazol-5-on bileşiği asetonitrilden başlanarak sentezlenmiştir. Bu amaçla Pinner Metodu kullanılarak [1] Denklem 1 uyarınca etil imidoasetat hidroklorür (1) elde edilmiş ve bu bileşiğin soğukta mutlak etanollü ortamda etil karbazat ile muamelesinden Denklem 2 uyarınca karşın olan 2 tipi etil asetat etoksi karbonilhidrazon bileşiği sentezlenmiştir. 2 bileşiğinin hidrazin hidrat ile muamelesinden ise 3-metil-4-amino-4,5-dihidro-1H-1,2,4-triazol-5-on (3) bileşiği elde edilmiştir (Denklem 1-3) [2-6]., In this study, firstly3-methyl-4-amino-4,5-dihydro-1H-1,2,4-riazole-5-one requiring for this studywere synthesized.
