6. In search of biomarkers and the ideotype of barley tolerant to water scarcity

Abstract

In barley plants, water shortage causes many changes on the morphological, physiological and biochemical levels resulting in the reduction of grain yield. In the present study the results of various experiments on the response of the same barley recombinant inbred lines to water shortage, including phenotypic, proteomic and metabolomic traits were integrated. Obtained results suggest that by a multi-omic approach it is possible to indicate proteomic and metabolomic traits important for reaction of barley plants to reduced water availability. Analysis of regression of drought effect (DE) for grain weight per plant on DE of proteomic and metabolomic traits allowed us to suggest ideotype of barley plants tolerant to water shortage. It was shown that grain weight under drought was determined significantly by six proteins in leaves and five in roots, the function of which were connected with defence mechanisms, ion/electron transport, carbon (in leaves) and nitrogen (in roots) metabolism, and in leaves additionally by two proteins of unknown function. Out of numerous metabolites detected in roots only Aspartic and Glutamic acids and one metabolite of unknown function, were found to have significant influence on grain weight per plant. The role of these traits as biomarkers, and especially as suggested targets of ideotype breeding, has to be further studied. One of the direction to be followed is genetic co-localization of proteomic, metabolomic and phenotypic traits in the genetic and physical maps of barley genome that can describe putative functional associations between traits; this is the next step of our analysis that is in progress., Comment: 12 pages, 8 figures, 2 supplementary tables

16. Management and Treatment Outcomes of Hemolytic Disease of the Fetus and Newborn (HDFN)—A Retrospective Cohort Study.

Abstract

Background: Hemolytic disease of the fetus and newborn (HDFN) is caused by maternal antibodies attacking fetal blood cell antigens. Despite routine antenatal anti-D prophylaxis, intrauterine transfusions (IUTs) are still needed in some HDFN cases. Methods: We conducted a retrospective cohort study on newborns with HDFN born in the 1st Department of Obstetrics and Gynecology of the Medical University of Warsaw. We analyzed 274 neonates with HDFN, identifying 46 who required IUT due to fetal anemia and 228 who did not. The laboratory results, management, and outcomes were compared between these groups. Results: Comparative analysis showed that newborns treated with IUT were more likely to have significant anemia, hyperbilirubinemia, and iron overload, indicated by a high ferritin concentration. These neonates more often required top-up transfusions, phototherapy, intravenous immunoglobulin infusions, and exchange transfusions. The length of stay was longer for newborns who received IUT. Conclusions: HDFN requiring IUT is associated with a greater number of complications in the neonatal period and more often requires additional treatment compared to HDFN not requiring IUT. Abstract

Doping high-zinc aluminium alloys with Ti builds in-situ composites reinforced with ternary aluminides Ti (Al,Zn)3 with a significantly grain-refined matrix. In a series of studies, Ti was introduced with Al-6 wt% Ti (AlTi6) and Zn-4 wt% Ti (ZnTi4) master alloys in amount to contribute about 3 wt% Ti in the examined alloys. The alloy microstructure has been studied using light microscopy, SEM / EDS and XRD measurements. The observed significant refinement of the alloys matrix should lead to improvement in ductility, while the in-situ reinforcement should improve tensile strength and wear properties.

Abstract

Renewable energy resources such as biomass are crucial for a sustainable global society. Trees are a major source of lignocellulosic biomass, which can vary in response to different environmental factors owing to epigenetic regulation, such as DNA C‐methylation. To investigate the effects of DNA methylation on plant development and wood formation, and its impacts on gene expression, with a focus on secondary cell wall (SCW)‐associated genes, Salix purpurea plantlets were cloned from buds derived from a single hybrid tree for both treatment and control conditions. For the treatment condition, buds were exposed to 50 μM zebularine in vitro and a combined strategy of whole‐genome bisulfite sequencing (WGBS) and RNA‐seq was employed to examine the methylome and transcriptome profiles of different tissues collected at various time points under both conditions. Transcriptomic and methylome data revealed that most of the promoter and gene body demethylation had no marked effects on the expression profiles of genes. Nevertheless, gene expression tended to decrease with the increased methylation levels of genes with highly methylated promoters. Results indicated that demethylation is less evident in centromeric regions and sex chromosomes. Promoters of secondary cell wall‐associated genes, such as 4‐coumarate‐CoA ligase‐like and Rac‐like GTP‐binding protein RHO, were differentially methylated in the secondary xylem samples collected from two‐month potted treated plants compared to control samples. Our results provide novel insights into DNA methylation and gene expression landscapes and a basis for investigating the epigenetic regulation of wood formation in S. purpurea as a model plant for bioenergy species. Abstract

The basilar bifurcation region is a common site for intracranial aneurysms, as well as it gives rise to a group of perforating arteries that supply the mesencephalon and the thalamus. Complex vascular microanatomy poses a diagnostic and therapeutic challenge for neurosurgeons, neuroradiologists and neurologists. In this paper, we present a previously unreported case of basilar tip fenestration that gave rise to five perforating arteries: the artery of Percheron and four mesencephalic arteries. Due to invaluable clinical significance, the possibility of such a variant must be considered during performing various neurovascular procedures, since e.g. embolization of the fenestration misdiagnosed as an aneurysm would inevitably lead to severe neurological complications (consciousness disturbances, quadriplegia, and sensory loss). Abstract

Hemolytic disease of the fetus and newborn (HDFN) may cause severe cholestasis with direct bilirubin concentrations reaching up to 50 times the upper limit of normal. This case report describes twins whose highest direct bilirubin concentrations were 32.2 mg/dL and 50.2 mg/dL, with no significant signs of hepatic impairment. The index pregnancy was complicated by Rhesus factor immunization with anti-D antibodies present in maternal serum, which caused fetal anemia requiring intrauterine blood transfusions. Complementary tests demonstrated Rhesus D alloimmunization as the sole cause of cholestasis. To the best of our knowledge, this is the first study to describe such elevated direct bilirubin concentrations caused by HDFN. [ABSTRACT FROM AUTHOR]

50. Genome-wide association mapping in elite winter wheat breeding for yield improvement

Abstract

Increased grain yield (GY) is the primary breeding purpose of wheat. In order to identify the main determinants of grain yield we used Genome-Wide Association Study (GWAS) on 168 elite lines of winter wheat from an ongoing breeding program. Sequencing of Diversity Array Technology fragments (DArTseq) resulted in 19,350 Single Nucleotide Polymorphism (SNP) and Presence Absence Variation (PAV) markers. We identified 15 main regions involving ten wheat chromosomes (1B, 2B, 2D, 3A, 3D, 5A, 5B, 6A, 6B, and 7B) that explain from 7.9 to 20.3% of the variation in grain yield and 13.3% of the yield stability. Loci identified in the reduced genepool are important for wheat improvement using marker assisted selection. We first found marker-trait associations of three genes involved in starch biosynthesis with grain yield. Two starch synthase genes, TraesCS2B03G1238800, TraesCS2D03G1048800, and sucrose synthase (TraesCS3D03G0024300) were located in regions of QGY3, QGY6, and QGY9, respectively. The loci and significantly associated SNP markers found in this study can be used for pyramiding favourable alleles in high-yielding varieties or to improve the accuracy of prediction in genomic selection.
