Dreifach negativer Brustkrebs (TNBC) stellt die aggressivste Brustkrebs-Untergruppe dar und wird häufig mit Mutationen in den Tumorsuppressorgenen BRCA1/2 in Verbindung gebracht. Patientinnen mit mutierten BRCA1/2-Genen können mit zugelassenen Poly (ADP-Ribose)-Polymerase – Inhibitoren (PARPi) behandelt werden. Eine lange nichtkodierende RNA (lncRNA) die durch DNA-Schäden aktiviert wird (NORAD), ist an molekularen Mechanismen der Krebsentstehung beteiligt. Das Ziel dieser Arbeit war es, die Funktionen von NORAD und seine Rolle gegenüber PARP-Inhibitoren (PARPi) in dreifach negativem Brustkrebs zu untersuchen. Auf Grund kürzlich publizierter Studien stellten wir die Hypothese auf, dass die verminderte Expression von NORAD und die gleichzeitige Behandlung mit PARP-Inhibitoren zu einem vermehrten Zelltod in TNBC-Zellen führen würde. Im Tumorgewebe von TNBC-Patientinnen ist die lncRNA NORAD, im Vergleich zu gesundem Gewebe, signifikant hochreguliert und geht mit einer schlechten Prognose einher. Um die Rolle von NORAD bei der malignen Entartung der Brust zu untersuchen, nutzten wir in drei voneinander unabhängigen TNBC-Zelllinien einen Gapmer-vermittelten silencing-Ansatz und ein Plasmid für die NORAD-Überexpression. Dadurch konnte ein verringertes Zellwachstum in Zellen mit hoch- als auch mit runterregulierten NORAD-Expressionslevels gezeigt werden. Eine Kombination von NORAD-Knockdown und dem PARP-Inhibitor Olaparib führte zu keiner gesteigerten genomischen Instabilität und folglich zu keinem vermehrten Zelltod. Es konnte auch nicht nachgewiesen werden, dass die Herabregulation von NORAD einen Einfluss auf die PARPi Sensitivität hat. Konträr dazu, konnte gezeigt werden, dass die Überexpression von NORAD einen negativen Einfluss auf das Ansprechen gegenüber Olaparib hat. Mit einem induzierbaren CRISPR/Cas9-System wurden BRCA1/2-mutierte TNBC-Zelllinien erzeugt, um die Auswirkungen der NORAD-Expression zu analysieren. Es wurde jedoch kein Hinweis darauf gefunden, dass der NORAD-knockdown einen Einfluss auf das Zellwachstum in BRCA1/2-KO-Zelllinien hat. Diese Daten deuten darauf hin, dass NORAD eine Bedeutung in TNBC hat und dabei helfen könnte, das Verständnis der Resistenzmechanismen gegen PARP-Inhibitoren bei TNBC zu verbessern. Weitere Untersuchungen sind erforderlich um NORAD als potenziellen Biomarker bei dreifach negativen Brustkrebspatientinnen klinisch implementieren zu können. Triple-negative breast cancer (TNBC) is the most aggressive breast cancer subtype and is frequently associated with mutations in BRCA1/2 genes. Patients with mutated BRCA1/2 genes can be treated with approved Poly (ADP-ribose) polymerase (PARP) inhibitors (PARPi). The long non-coding RNA Activated by DNA damage (NORAD) has been implicated in molecular mechanisms associated with carcinogenesis. The master thesis aimed to investigate the functions of NORAD in triple-negative breast cancer and its role in PARP inhibitor sensitivity. Based on previously reported findings, we hypothesized that combining NORAD knock-down and PARP inhibitor treatment would render TNBC cells more sensitive to PARPi. Overall, I found that the lncRNA NORAD is significantly up-regulated in cancerous tissue of TNBC patients compared to healthy breast tissue and is associated with a poor outcome for patients. To clarify the role of NORAD in breast carcinogenesis, we used Gapmer-mediated silencing in three independent TNBC cell lines and a plasmid for NORAD overexpression to find reduced cellular growth in cells with up- as well as down-regulated NORAD expression levels. A combination of NORAD knock-down and the PARP inhibitor olaparib did not show synthetic lethality, nor was it found that NORAD knock-down impacts PARPi sensitivity. Conversely, the overexpression of NORAD has been shown to negatively affect PARP inhibitor sensitivity. Additionally, an inducible CRISPR/Cas9 system was used to generate BRCA1/2-mutated TNBC cell lines to analyze the effects of NORAD expression on cancer cell features. However, no evidence was found that NORAD silencing influences cellular growth in BRCA1/2 KO cell lines. These data suggest the relevance of NORAD in TNBC and may improve the understanding of resistance mechanisms against PARP inhibitors in TNBC. Further investigations are required to validate whether NORAD could serve as a promising biomarker in triple negative breast cancer patients. Uvođenje ISDN-a u hrvatsku telefonsku mrežu osnovna je tema ovoga rada. Opisana je telefonska mreža kao osnova za ISDN te aspekti ISDN-a. Navedeni su primjeri primjene ISDN usluga te koju je ulogu imao ISDN u povezivanju Interneta i javne telefonske mreže. Dani su kvalitativni prikazi ukupnog prometa i posebno prometa prema Internetu na pristupnim i tranzitnim centralama. Kao slijedeći korak u razvoju telekomunikacijske mreže, predstavljena je konvergencija dviju mreža i usluga: telefonske i podatkovne. Predstavljen je mogući koncept mreže nove generacije. Kao nezaobilazna funkcija u mreži opisano je usmjeravanje prometa te moguća unaprijeđenja prema dinamičkom i adaptivnom usmjeravanju. Na već izrađenom modelu za testiranje metoda usmjeravanja (LAG) napravljeni su eksperimenti koji pokazuju usporedne rezultate simulacije fiksnog usmjeravanja i usmjeravanja na osnovu automata sa svojstvom učenja. The basic subject of this paper is the implementation of ISDN to the Croatian PSTN network. Public Switched Telephony Network is described as a basis for ISDN. The implementation of ISDN services is presented through all relevant aspects. Further, the examples of ISDN usage and the impact of ISDN on connection of Internet and PSTN are mentioned. Description of the network and ISDN implementation are followed by qualitative presentations of the total and Internet traffic on the local and transit exchanges. The next step in network development is presented through the convergence of the PSTN and data networks and services. Possible concept of the next generation network is described. As traffic routing is an important function in the network, its improvement towards dynamic and adaptive routing is presented. Simulations of fixed routing and the routing based on learning automata are performed on the already designed mogul (LAG). The results of simulations that are presented serve for the comparison of routing methods.

Following the concept of Intelligent Network and PSTN evolution trends, Application Programming Interfaces are developed. Creation of service logic that is independent of the underlying technologies is done through usage of middleware distributed object programming technologies. TINA-C Business Model is used for identification of key business areas in services providing process. Parlay work is related to creation of capabilities to open and secure access to telecommunication network for service developers and providers.
